Abstract

A visible-light-driven α-hydroxymethylation of ketones to generate the corresponding alcohols was achieved under continuous-flow conditions. MeOH was used as a green and renewable C1 source and solvent to enable the α-C(sp3)–H functionalization of ketones under irradiation by white LEDs. A flow microreactor operated under optimized conditions permitted this oxidation to proceed with a higher efficiency and a shortened reaction time of 215 minutes, which was improved ten times compared with the batch parallel reaction (36 h). Abstract

The selective oxidation of C(sp3)–H bonds is of uncontested utility in organic synthesis. Here, we report a novel and sustainable divergent protocol using O2 as a green oxidant enabled by disulfide in batch and flow reactions. In particular, by simply adjusting the wavelength of the light source, the chemoselectivity of amine oxygenation can be significantly switched from oxidative homo-coupling to oxidative deamination without a concomitant loss in photocatalytic activity. Furthermore, a wide range of aromatic hydrocarbons participates in the reaction with high efficiency, offering a sustainable alternative for the late-stage oxidative functionalization of benzylic C–H bonds. Mechanistic investigations revealed that the disulfide served as an excellent photoinitiator. The key C(sp3)–H bond activation step is believed to proceed via hydrogen atom abstraction affected by a benzothiazole-substituted thiyl radical for further oxidation processes. The reaction can be significantly improved in a microflow reactor, enabling the safe use of oxygen and enhanced irradiation of the reaction mixture. We anticipate that a broad scope with wide functional group compatibility, operational simplicity, and scalability in a flow reaction would bring useful and practical applications of this catalysis strategy to various fields. 28. Selective Oxidation of Benzylic sp3C–H Bonds using Molecular Oxygen in a Continuous-Flow Microreactor

Abstract

Selective aerobic oxidation of benzylic sp3C–H bonds to generate the corresponding ketones was achieved under continuous-flow conditions. The catalysts N-hydroxyphthalimide (NHPI) and tert-butyl nitrite (TBN) as the precursor of the radical under aerobic conditions motivated this process. Flow microreactors operating under optimized conditions enabled this oxidation with higher efficiency and a shortened reaction time of 54 s (total time was 10 min), which was improved 466 times compared with the batch parallel reaction (7.0 h). Notably, the catalyst and solvent recycling (92.6 and 94.5%) and scale-up experiments (0.87 g h–1in 28 h) demonstrated the practicability of the protocol. The high product selectivity and functional group tolerance of the process allowed the production of ketones in yields of 41.2 to 90.3%. To reveal the versatility and applicability of this protocol, the late-stage modification of an antiepileptic drug to obtain oxcarbazepine was further conducted.
