2. Effect of Preharvest Spraying of Diethyl Aminoethyl Hexanoate on Membrane Lipid Metabolism of Grapes during Storage

In order to investigate the effect of preharvest spraying of diethyl aminoethyl hexanoate (DA-6) on membrane lipid metabolism in ‘Kyoho’ grapes during postharvest storage, distilled water (control) and 50 mg/L DA-6 were used to spray grapes at the veraison stage. The grapes were harvested when they were ripe, stored at (0 ± 1) ℃ and relative humidity of 65%–70%, and evaluated for cell membrane related indicators of grape skin after 0, 20, 30, 40, 50, and 60 days. The results showed that compared with the control group, DA-6 treatment effectively inhibited the increase in the relative permeability of cell membrane during postharvest storage, maintained the activities of lipoxygense (LOX), lipase and phospolipase D (PLD), inhibited the decrease of phosphatidylcholine and phosphatidylinositol content and the increase of phosphatidyl acid content, and maintained a high relative content of unsaturated acids such as linoleic acid and linolenic acid as well as saturated fatty acids such as stearic acid, behenic acid, arachidic acid and palmitoleic acid, consequently maintaining the degree of unsaturation of fatty acids at a high level. Additionally, DA-6 treatment suppressed the expression of the LOX, Lipase, and PLD genes. In summary, preharvest spraying of 50 mg/L DA-6 can delay the senescence and prolong the storage period of ‘Kyoho’ grapes by effectively relieving the membrane lipid metabolism during storage.

The vigorous metabolism of reactive oxygen species (ROS) in postharvest ‘Kyoho’ grapes shortens the storage period, and reduce the commercial value. In this study, different concentrations (0, 10, 30, 50 and 70 mg/L) of diethyl aminoethyl hexanoate (DA-6) were sprayed on table grapes at veraison to investigate the effects of DA-6 treatment on postharvest fruit quality and ROS metabolism in ‘Kyoho’ grapes. By measuring the physiological indexes related to fruit quality, it was found that DA-6 treatment significantly reduced fruit mass loss, and delayed the decrease in fruit firmness and total soluble solid content, and this effect was most significant at a concentration of 50 mg/L. More importantly, 50 mg/L DA-6 treatment significantly increased the activity of superoxide dismutase, ascorbate peroxidase, catalase, glutathione reductase, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging capacity and total reducing capacity, and suppressed the decrease in reduced ascorbic acid and glutathione contents and the accumulation of superoxide anion radical and malondialdehyde. In conclusion, pre-harvest spray of DA-6 maintained the stability of the ROS scavenging system in postharvest table grapes, kept ROS metabolism in a relatively balanced state, reduced oxidative damage, delayed fruit senescence, and prolonged the shelf life of ‘Kyoho’ grapes.

Sulfur dioxide (SO
2 ) is the most effective preservative for table grapes as it reduces the respiratory intensity of berries and inhibits mold growth. However, excessive SO2 causes berry abscission during storage, resulting in an economic loss postharvest. In this study, grapes were exogenously treated with SO2 , SO2 + 1.5% chitosan, SO2 + 1.5% eugenol, and SO2 + eugenol-loaded chitosan nanoparticles (SN). In comparison to SO2 treatment, SN treatment reduced the berries' abscission rate by 74% while maintaining the quality of the berries. Among the treatments, SN treatment most effectively inhibited berry abscission and maintained berry quality. RNA-sequencing (RNA-seq) revealed that SN treatment promoted the expression of genes related to cell wall metabolism. Among these genes, VlCOMT was detected as the central gene, playing a key role in mediating the effects of SN. Dual luciferase and yeast one-hybrid (Y1H) assays demonstrated that VlbZIP14 directly activated VlCOMT by binding to the G-box motif in the latter's promoter, which then participated in lignin synthesis. Our results provide key insights into the molecular mechanisms underlying the SN-mediated inhibition of berry abscission and could be used to improve the commercial value of SO2 -treated postharvest table grapes. 14. PpGATA21 Enhances the Expression of PpGA2ox7 to Regulate the Mechanism of Cerasus humilis Rootstock-Mediated Dwarf in Peach Trees.

Dwarfing rootstocks enhance planting density, lower tree height, and reduce both labor in peach production. Cerasus humilis is distinguished by its dwarf stature, rapid growth, and robust fruiting capabilities, presenting substantial potential for further development. In this study, Ruipan 4 was used as the scion and grafted onto Amygdalus persica and Cerasus humilis, respectively. The results indicate that compared to grafting combination R/M (Ruipan 4/Amygdalus persica), grafting combination R/O (Ruipan 4/Cerasus humilis) plants show a significant reduction in height and a significant increase in flower buds. RNA-seq indicates that genes related to gibberellin (GA) and auxin metabolism are involved in the dwarfing process of scions mediated by C. humilis. The expression levels of the GA metabolism-related gene PpGA2ox7 significantly increased in R/O and are strongly correlated with plant height, branch length, and internode length. Furthermore, GA levels were significantly reduced in R/O. The transcription factor PpGATA21 was identified through yeast one-hybrid screening of the PpGA2ox7 promoter. Yeast one-hybrid (Y1H) and dual-luciferase reporter (DLR) demonstrate that PpGATA21 can bind to the promoter of PpGA2ox7 and activate its expression. Overall, PpGATA21 activates the expression of the GA-related gene PpGA2ox7, resulting in reduced GA levels and consequent dwarfing of plants mediated by C. humilis. This study provides new insights into the mechanisms of C. humilis and offers a scientific foundation for the dwarfing and high-density cultivation of peach trees. [ABSTRACT FROM AUTHOR]

21. Alternative splicing of the PECTINESTERASEgene encoding a cell wall-degrading enzyme affects postharvest softening in grape

- Abstract
The firmness of table grape berries is a crucial quality parameter. Despite extensive research on postharvest fruit softening, its precise molecular mechanisms remain elusive. To enhance our comprehension of the underlying molecular factors, we initially identified differentially expressed genes (DEGs) by comparing the transcriptomes of folic acid (FA)-treated and water-treated (CK) berries at different time points. We then analyzed the sequences to detect alternatively spliced (AS) genes associated with postharvest softening. A total of 2,559 DEGs were identified and categorized into four subclusters based on their expression patterns, with subcluster-4 genes exhibiting higher expression in the CK group compared with the FA treatment group. There were 1,045 AS-associated genes specific to FA-treated berries and 1,042 in the CK-treated berries, respectively. Gene Ontology (GO) annotation indicated that the AS-associated genes in CK-treated berries were predominantly enriched in cell wall metabolic processes, particularly cell wall degradation processes. Through a comparison between treatment-associated AS genes and subcluster-4 DEGs, we identified eight genes, including Pectinesterase 2(VvPE2, Vitvi15g00704), which encodes a cell wall-degrading enzyme and was predicted to undergo an A3SS event. The reverse transcription polymerase chain reaction further confirmed the presence of a truncated transcript variant of VvPE2in the FA-treated berries. Our study provides a comprehensive analysis of AS events in postharvest grape berries using transcriptome sequencing and underscores the pivotal role of VvPE2during the postharvest storage of grape berries.

26. Low-Power, High-Speed, and Area-Efficient Multiplier Based on the PTL Logic Style

- Abstract
With the development of Very Large-Scale Integration (VLSI) technology, circuit designers are increasingly focused on achieving low power, high speed, and small area. Among various circuit components, multiplier circuits play an important role in improving the overall system performance due to their significant power consumption and impact on circuit speed. In this brief, we propose a novel 8-bit signed multiplier based on the Pass Transistor Logic (PTL) that outperforms existing designs. The post-layout simulation results show that the proposed design reduces area, delay, and power by 13.45%, 9.72%, and 15.19%, respectively, compared to the multiplier synthesized using Synopsys Design Compiler (DC). Compared to other 8-bit multipliers proposed in references, our design also shows at least a 27.66% improvement in power-delay product (PDP). Additionally, the proposed circuits exhibit superior performance at different operating voltages and process corners.

In statistics, samples are drawn from a population in a data- generating process (DGP). Standard errors measure the uncer- tainty in sample estimates of population parameters. In sci- ence, evidence is generated to test hypotheses in an evidence- generating process (EGP). We claim that EGP variation across researchers adds uncertainty: non-standard errors. To study them, we let 164 teams test six hypotheses on the same sam- ple. We find that non-standard errors are sizeable, on par with standard errors. Their size (i) co-varies only weakly with team merits, reproducibility, or peer rating, (ii) declines significantly after peer-feedback, and (iii) is underestimated by participants.

- Abstract
In statistics, samples are drawn from a population in a data-generating process (DGP). Standard errors measure the uncertainty in sample estimates of population parameters. In science, evidence is generated to test hypotheses in an evidence-generating process (EGP). We claim that EGP variation across researchers adds uncertainty: non-standard errors. To study them, we let 164 teams test six hypotheses on the same sample. We find that non-standard errors are sizeable, on par with standard errors. Their size (i) co-varies only weakly with team merits, reproducibility, or peer rating, (ii) declines significantly after peer-feedback, and (iii) is underestimated by participants.

- Abstract
Fruit set is a key stage in determining yield potential and guaranteeing quality formation and regulation. N-(2-chloro-4-pyridyl)-N′-phenylurea (CPPU) has been widely applied in grape production, the most iconic of which is the promotion of grape fruit set. However, current studies still lack the molecular mechanism of CPPU-induced grape fruit set. Here, the dynamic, high-resolution stage-specific transcriptome profiles were generated based on two different treatments and five developmental periods during fruit set in ‘Kyoho’ grape (Vitis viniferaL. × V. labruscaL.). Pairwise comparison and functional category analysis showed that phytohormone action cytokinin was significantly enriched during the CPPU-induced grape fruit set, but not the natural one. Value differentially expressed gene (VDEG) was a newly proposed analysis strategy for mining genes related to the grape fruit set. Notably, the cytokinin metabolic process was significantly enriched among up-regulated VDEGs. Of importance, a key VDEG VlCKX4related to the cytokinin metabolic process was identified as related to the grape fruit set. Overexpression of VlCKX4gene promoted the Arabidopsis plants that produce more and heavier siliques. The transcription factor VlMYB59 directly bound to the promoter of VlCKX4and activated its expression. Moreover, overexpression of VlMYB59gene also promoted the Arabidopsis fruit set. Overall, VlMYB59 responded to CPPU treatment and directly activated the expression of VlCKX4, thus promoting the fruit set. A regulatory pathway of the VlMYB59-VlCKX4module in the fruit set was uncovered, which provides important insights into the molecular mechanisms of the fruit set and good genetic resources for high fruit set rate breeding.
