Abstract

Uncovering early gene network changes of human hematopoietic stem cells (HSCs) leading to differentiation induction is of utmost importance for therapeutic manipulation. We employed single cell proteo-transcriptomic sequencing to FACS-enriched bone marrow hematopoietic stem and progenitor cells (HSPCs) from 15 healthy donors. Pseudotime analysis reveals four major differentiation trajectories, which remain consistent upon aging, with an early branching point into megakaryocyte-erythroid progenitors. However, young donors suggest a more productive differentiation from HSPCs to committed progenitors of all lineages. tradeSeq analysis depicts continuous changes in gene expression of HSPC-related genes (DLK1, ADGRG6), and provides a roadmap of gene expression at the earliest branching points. We identify CD273/PD-L2 to be highly expressed in a subfraction of immature multipotent HSPCs with enhanced quiescence. Functional experiments confirm the immune-modulatory function of CD273/PD-L2 on HSPCs in regulating T-cell activation and cytokine release. Here, we present a molecular map of early HSPC differentiation across human life. Therapeutic application of hematopoietic stem cells requires a thorough understanding of their in situ differentiation pathways. Abstract

Diminishing soil fertility and crop productivity due to traditional intensive cultivation has prompted the use of straw and manure to improve soil health in Northeast China. However, few comparative studies have explored the influence of varying straw and manure incorporation depths on crop growth. A field experiment in the rainfed black soil regions of Gongzhuling and Keshan assessed the effects of deep (30 cm) and shallow (15 cm) incorporations of straw and manure on soil fertility, maize root growth, and maize productivity. Deep incorporations, via subsoiling tillage (DST) and deep-plow (DDT) tillage, enhanced soil water storage of 30–100 cm soil layer during periods of low rainfall, improved the availability of nutrients (nitrogen, phosphorus, and potassium) and soil organic matter content, especially in deeper soil, compared to shallow incorporation using rotary tillage (SRT). Both DST and DDT induced a larger rooting depth and a higher fine root (diameter class 0–0.5 mm) length density by 31.0% and 28.9%, respectively, accompanied by reduced root turnover. Furthermore, the sub-surface foraging strategies of roots under the DST and DDT treatments boosted the total nitrogen, phosphorus, and potassium uptake (6.5–17.9%) and achieved a higher dry mass accumulation during the later growth period, thus leading to notable improvements in the 100-kernel weight and yield (16.1–19.7%) and enhancing water- and nutrient-use efficiencies by 2.5–20.5%. Overall, compared to shallow incorporation, deep incorporation of straw and manure significantly enhances root growth and spatial distribution of soil water and nutrients, which has great potential for increasing maize yield in rainfed agricultural areas. 30. The functional and molecular role of transglutaminase 2 in hematopoietic stem and progenitor cells

Abstract

Long-term repopulating hematopoietic stem cells (LT-HSCs) that reside in the bone marrow (BM) give rise to all blood cell types including erythrocytes, leukocytes and platelets. LT-HSCs are mainly quiescent during steady state hematopoiesis. LT-HSCs can process self-renewal to expand and maintain stemness, or commit to differentiation into short-term (ST) repopulating HSC and multipotent progenitors (MPPs). MPPs differentiate into oligopotent lineagerestricted progenitors which eventually produce all mature blood cell lineages, and thereby regenerate hematopoietic system. Previous studies have shown in transcription profiles and quantitative PCR (qPCR) analysis that transglutaminase 2 (Tgm2) is one of the most upregulated genes in quiescent LT-HSCs in comparison to active HSCs, mobilized HSCs, ST-HSCs, MPPs, as well as leukemic stem cells (LSC). However, the reason why Tgm2 is strongly upregulated in dormant mouse LTHSCs and what the role of Tgm2 is in LT-HSCs has not been investigated yet. Tgm2, encoded by the Tgm2 gene, is a multi-functional protein within the transglutaminase family. It has been found to be widely expressed inside and outside the cells. It consists of four domains and two functionally exclusive forms that are regulated by the Ca2+ and GTP concentration. Besides the most well-known transglutaminase enzymatic activity for transamidation, deamidation and crosslinking, Tgm2 acts also as a GTPase/ATPase, kinase, adhesion/scaffold protein, as well as disulfide isomerase. The role of Tgm2 in hematopoiesis remains elusive. Accordingly, the aim of this dissertation is to investigate the role of Tgm2 in murine hematopoiesis, especially in murine LT-HSCs. 42. Tensor-Based Angle Estimation for Bistatic MIMO Radar Systems With Multislot Gain-Phase Error

Abstract

In this article, a tensor-based angle estimation algorithm is proposed for bistatic multiple-input multiple-output (MIMO) radar systems with a multislot gain-phase error (MSGPE). First, the received signals are constructed into a complex-valued third-order parallel factor model that contains information, such as transmit MSGPE, receive MSGPE, directions of departure (DODs), and directions of arrival (DOAs). Then, a two-stage tensor-based angle algorithm is proposed for antenna arrays with the MSGPE. In the first stage, the MSGPE information part and angle information part are separated by the least-squares Khatri–Rao factorization. In the second stage, the DODs and DOAs are obtained jointly based on spatial smoothing and real-valued processing techniques under eliminating the influence of MSGPE. Compared with the existing competitive algorithms, the proposed algorithm achieves an improved performance and is applicable for both the coherent and noncoherent targets. Simulation results show the superiority of the proposed tensor-based angle estimation algorithm.
