9. Ultrasensitive detection of uveal melanoma using [18F]AlF-NOTA-PRGD2 PET imaging.

Abstract

Background: Uveal melanoma (UM) is the most common primary intraocular tumor in adults, and early detection is critical to improve the clinical outcome of this disease. In this study, the diagnostic effectiveness of [18F]AlF-NOTA-PRGD2 (an investigational medicinal product) positron emission tomography (PET) imaging in UM xenografts and UM patients were evaluated. The cell uptake, cell binding ability and in vitro stability of [18F]AlF-NOTA-PRGD2 were evaluated in 92-1 UM cell line. MicroPET imaging and biodistribution study of [18F]AlF-NOTA-PRGD2 were conducted in 92-1 UM xenografts. Then, UM patients were further recruited for evaluating the diagnostic effectiveness of [18F]AlF-NOTA-PRGD2 PET imaging (approval no. NCT02441972 in clinicaltrials.gov). In addition, comparison of [18F]AlF-NOTA-PRGD2 and 18F-labelled fluorodeoxyglucose ([18F]FDG) PET imaging in UM xenografts and UM patients were conducted. Results: The in vitro data showed that [18F]AlF-NOTA-PRGD2 had a high cell uptake, cell binding ability and in vitro stability in 92-1 UM cell line. The in vivo data indicated that 92-1 UM tumors were clearly visualized with the [18F]AlF-NOTA-PRGD2 tracer in the subcutaneous and ocular primary UM xenografts model at 60 min post-injection. And the tumor uptake of the tracer was 2.55 ± 0.44%ID/g and 1.73 ± 0.15%ID/g at these two tissue locations respectively, at 7 days after animal model construction. The clinical data showed that tumors in UM patients were clearly visualized with the [18F]AlF-NOTA-PRGD2 tracer at 60 min post-injection. In addition, [18F]AlF-NOTA-PRGD2 tracer showed higher sensitivity and specificity for PET imaging in UM xenografts and UM patients compared to [18F]FDG tracer. Conclusion: [18F]AlF-NOTA-PRGD2 PET imaging may be a more preferred approach in the diagnosis of primary UM compared to [18F]FDG PET imaging. Additionally, due to the high tumor-to-background ratio, [18F]AlF-NOTA-PRGD2 PET imaging seems also to be applicable for the diagnosis of UM patients with liver metastasis. 16. A Subpixel Calibration Strategy for Micro-Raman Spectrometer in Subvisible Particles Traceable Detection

Abstract

Subvisible particles are a major cause of adverse clinical infusion reactions and can lead to serious health problems. Raman spectroscopy is a rapid, nondestructive method for traceable detection of subvisible particles; however, the pixel-level spectral resolution limits the accuracy of the existing micro-Raman spectrometers. Consequently, we propose a subpixel calibration strategy to improve the detection accuracy of micro-Raman spectrometers. First, a combined light source with higher stability and uniformity is selected to improve the standard wavelength coverage of the charge-coupled device (CCD) pixels and the stability of the calibration. Subsequently, a wavelet-based automatic peak localization method is designed to obtain the pixel-integer values of the peak points corresponding to the standard wavelengths. Based on the spectral distribution characteristics, we can obtain a subpixel value corresponding to the peak center of the wavelength by fitting the Gaussian spectral peak curve. Then, a higher resolution subpixel-level calibration is achieved by establishing a mapping function between wavelengths and peak centers. We compare the standard sample detection shift accuracy and stability by using subpixel and pixel calibration. The subpixel-level calibration reduces the accuracy error from 4.3 to 1.3 cm-1 and the stability error from 4.8% to 2.4% compared to the pixel-level calibration, improving the micro-Raman performance indicator from Level III to Level II. Additionally, comparing the results of traceable detection of eight common injectable particles, the average similarity of micro-Raman using subpixel calibration was 96.6%, which is 7.5% higher than the matching accuracy of pixel calibration. These results demonstrate that the improved calibration of micro-Raman enables more accurate and stable detection of particles, meeting the requirements of high-accuracy engineering applications in particle traceable detection for pharmaceutical companies.

Abstract

PdSe2, a star photosensitive functional material, has been successfully used in photodetectors based on sensing mechanisms of photogating, photoconductive, and photovoltaic effects. Here, a photothermoelectric (PTE) effect is observed in photodetectors based on PdSe2 flakes grown by chemical vapor deposition. The unique photoresponse arises from an electron temperature gradient instead of electron–hole separation. Direct evidence of the PTE effect is confirmed by a nonlocal photoresponse under zero bias. Moreover, the PdSe2 photodetector shows high performance in terms of ultrafast response speed (4 µs), high air‐stability, broadband spectrum photodetection, reasonable responsivity, and anisotropic optical response. Abstract

In order to realize the benefit maximization of power generation, doubly fed induction generator (DFIG) actually do not participate in reactive power regulation. Meanwhile, the wind power fluctuation leads to a great variety of reactive power shortage of wind farms. Therefore it is hard to satisfy the requirements of reactive compensation by using the reactive compensation device fixed in each wind farm independently. Currently reactive power compensation strategies of wind farms in a wind farms cluster are mainly controlled by every farm independently, and there is a lack of coordinated controlling between them. In view of all these, this thesis presents a reactive power coordinated control strategy which is suitable for multiple wind farms region. To improve voltage quality and realize the economical operation, we establish a multi-objective optimization model according to the operation information of power grid, and take a research on period-decoupled compensation technology as wind power fluctuate strongly. In this model, PSO algorithm is introduced to calculate the minimization problem of the reactive compensation. Finally, the simulation system testing by actual production data, show the control strategy is available and has perfect accuracy.
