Research has been conducted on Aspergillus parasiticus. The construction of A. parasiticus strain with deleted alfR gene has been carried out in determining the ability of A. sojae alfR coding region to fuse to the inducible amylase gene promoter and the results are reported.

Abstract

Aflatoxin pathway regulatory protein (AFLR), a DNA-binding protein of 444 amino acids, transactivates the expression of aflatoxin biosynthesis genes in Aspergillus parasiticus and Aspergillus flavus, as well as the sterigmatocystin synthesis genes in Aspergillus nidulans. By fusion of various aflR gene coding regions to the GAL4 DNA-binding coding region, it was found that the AFLR carboxyl terminus contained a region that activated GAL1::lacZ gene expression in Saccharomyces cerevisiae and that the AFLR internal region was required for the activation activity.

Abstract

A study was conducted to clone the ordA and ordA1 genes of the aflatoxigenic strain Aspergillus parasiticus SRRC 143 and SRRC 2043, respectively, and to evaluate the function of the ordA gene in the formation of aflatoxins. Point mutations were introduced into the ordA gene using polymerase chain reaction-based site-directed mutagenesis while mycelia were obtained by filtration. Results indicated that cytochrome P-450 type enzymes were involved in the synthesis of different primary and secondary metabolites in filamentous fungi.

Abstract

The avnA gene from the filamentous fungi Aspergillus flavus, corresponding to one of the two transcripts found between the ver-1 and omtA genes on the gene cluster involved in the aflatoxin biosynthetic pathway, was sequenced. Its nucleotide sequence has a coding region for a protein of 495 amino acids with a molecular mass of 56.3 kDa. The gene is composed of three exons and two introns. Disruption of the avnA gene in the wild-type aflatoxigenic A. parasiticus strain produced a nonaflatoxigenic mutant which accumulated a bright yellow pigment.

Abstract

cDNA that encodes O-methylsterigmatocystin (OMT-1) from an Aspergillus parasiticus SRRC 143 was isolated. The isolation of the 1,460-bp full length cDNA sequence of the OMT-1 gene was also conducted. This gene encodes the 46-kDa native OMT-1 found in Escherichia coli which reveals a higher level of enzymatic activity than the 42-kDa enzyme obtained from fungal mycelia.
