Abstract

The molecularly cloned virus known as SIVmac239/YEnef causes extensive lymphocyte activation in unstimulated peripheral mononuclear cell cultures and induces an acute disease syndrome in macaque monkeys. Here we describe the histopathological and immunophenotypic changes and viral localization in peripheral lymph nodes, spleen, and gastrointestinal tract (including the gut-associated lymphoid tissue (GALT) in rhesus monkeys inoculated with SIVmac239/YEnet beginning at day 3 postinoculation (pi). The findings are compared with those of rhesus monkeys inoculated with the same dose of parental SIVmac239. Histopathological examination of peripheral lymphoid tissue and GALT demonstrated marked hyperplasia of T-cell-dependent regions and involution of germinal centers as early as day 7 pi. The most striking lesions were multifocal areas of lymphohistiocytic gastroenteritis and colitis. Cellular infiltrates peaked between day 7 and 14 pi and were composed primarily of CD3+ T lymphocytes and HAM-56+ monocyte/macrophages. Many of these inflammatory cells were also strongly immunoreactive for teh nuclear proliferation antigen Ki-67. Despite the presence of severe gastrointestinal pathology by day 7 pi, no significant difference in the numbers of virus-positive cells in the gastrointestinal tract was observed between these animals and SIVmac239-infected animals examined at the same time point. However, the distribution of virus in the gastrointestinal tract was markedly different, with virus localized to lymphoid nodules of GALT in SIVmac239-infected animals and restricted to areas of lymphohistiocytic gastroenteritis and colitis in animals infected with SIVmac239/YEnef. Our data indicate that the acute disease syndrome induced by SIVmac239/YEnef is not simply related to increased viral replication in the gastrointestinal tract but is likely due to inappropriate virus-induced T lymphocyte activation and proliferation in GALT and subsequent mucosal destruction.

13. Plasma lactic dehydrogenase activities in men during bed rest with exercise training

Abstract

Peak oxygen uptake and the activity of lactic dehydrogenase (LDH-T) and its five isoenzymes were measured by spectrophotometer in seven men before, during, and after bed rest and exercise training. Exercise training consisted of isometric leg exercises of 250 kcal/hr for a period of one hour per day. It is found that LDH-T was reduced by 0.05 percent in all three regimens by day 10 of bed rest, and that the decrease occurred at different rates. The earliest reduction in LDH-T activity in the no-exercise regimen was associated with a decrease in peak oxygen uptake of 12.3 percent. It is concluded that isometric (aerobic) muscular strength training appear to maintain skeletal muscle integrity better during bed rest than isotonic exercise training. Reduced hydrostatic pressure during bed rest, however, ultimately counteracts the effects of both moderate isometric and isotonic exercise training, and may result in decreased LDH-T activity.

16. Effects of exercise on fluid exchange and body composition in man during 14-day bed rest

Abstract

A description is presented of an investigation in which body composition, fluid intake, and fluid and electrolyte losses were measured in seven normal, healthy men during three 2-wk bed-rest periods, separated by two 3-wk recovery periods. During bed rest the subjects remained in the horizontal position continuously. During the dietary control periods, body mass decreased significantly with all three regimens, including no exercise, isometric exercise, and isotonic excercise. During bed rest, body mass was essentially unchanged with no exercise, but decreased significantly with isotonic and isometric exercise. With one exception, there were no statistically significant changes in body density, lean body mass, or body fat content by the end of each of the three bed-rest periods.

Abstract

It is difficult to separate the effects of reduction in hydrostatic pressure from that of reduced energy expenditure when investigating the confinement deconditioning problem. Experiments were conducted on seven healthy young men aged 19-21 yr with the purpose of separating these two factors by using isotonic physical exercise during bed rest to provide a daily energy expenditure greater than normal ambulatory levels. Fluid and electrolyte shifts were measured during three two-week bed rest periods, each of which being separated by a three-week ambulatory recovery period. During two of the three bed rest periods they performed isometric and isotonic exercises to compare their effects on fluid and electrolyte shifts during bed rest. It is shown that during bed rest, preservation of the extracellular volume takes precedence over maintenance of the plasma volume and that this mechanism is independent of the effects of isometric or isotonic exercise.

Abstract

Measurement of the reduction in centrifugation tolerance after two weeks of bedrest with moderate daily exercise, with an attempt to determine if rehydration improves +Gz tolerance. There were significant reductions in +Gz tolerance during bedrest periods at three acceleration levels. Rehydration resulted in a significant increase in tolerance at 2.1G but it did not restore tolerance to control levels. Rehydration did not affect tolerance at 3.2G and 3.8G.

27. Body water compartments during bed rest: Evaluation of analytical methods

Abstract

Nine healthy young men were studied to determine the reproducibility and interchangeability of the use of radio-iodinated human serum albumin and Evans Blue dye for estimating plasma volume, sodium bromide for extracellular fluid volume, and deuterium oxide for total body water volume. All subjects were tested in a semibasal condition and allowed to rest for at least 30 min. after arriving at the laboratory. The results indicate that there was uniform distribution of I131 and Evans Blue dye 10 min. after injection and of NaBr and D2O 3 hours after oral ingestion; the buildup of residual tracer did not interfere appreciably with the measurement of either or Evans Blue spaces when they are administered at equal intervals, and the buildup of background tracer after ingestion of NaBr and D2O once per week for three consecutive weeks did not affect the accuracy of the measurement. It was found that I131 and Evans Blue may be used interchangeably for estimating plasma volume; for estimating bromide and D2O spaces, one 3-hour equilibrium blood sample gives results similar to the extrapolation of multiple samples.

43. Medical education by satellite: The EuroTransMed experience

Abstract

Continuing medical education is a key feature of ensuring quality delivery of health care by doctors. In a rapidly changing technological environment it is important that medical education is undertaken in an effective and efficient manner. Euro-TransMed, set up as an experiment to investigate the use of satellite-delivered continuing medical education, now supplies regular programming to receive sites in 20 European countries. Programmes are broadcast live and are interactive, enabling doctors to have access to experts and opinion leaders and to question and challenge them. This review paper highlights the development and technical problems which have taken place during the initial establishment of EuroTransMed. The current status and future prospects are identified.

A water‐budget model was developed to aid the study of transport of nutrients in surface and subsurface flows across a marsh. The model is based on field studies of Wingra marsh adjacent to Lake Wingra in Madison, Wis., and was used to calculate monthly totals of actual evapotranspiration, surface runoff, and subsurface drainage from April 1970 through August 1976. About 90% of the water entering the marsh comes from urban storm runoff. The remainder is precipitation (8%) and deep ground‐water inflow (2%). Water leaves the marsh through evapotranspiration (7%), shallow ground‐water discharge (8%), and surface outflow (85%). The model was used to estimate the water‐table elevation at an observation well where direct measurements were taken (November 1975 through October 1976). The observed depth to the water table ranged from 0 to 150 cm below the surface and the simulated value usually was within ± 10 cm. This suggests that the model adequately computes the water budget for the marsh and provides a sound basis for calculating dissolved and suspended material loads transported through the marsh into Lake Wingra.

Abstract

Under “nongratuitous” inducing conditions, in Pseudomonas saccharophila, d-and l-valine and l-isoleucine inhibit net protein synthesis. At a concentration of 0.5 μmole or greater of valine per mg of bacterial protein, net protein synthesis declined approximately 70%. The inhibitory effect of valine is proportional to the exogenous valine concentration. Studies of 14C amino acid incorporation and 14C amino acid release from prelabeled cells indicate that valine stimulates protein turnover.

Abstract

The energetic salts NF4AsF6 and N2F4.AsF5 were used in the preparation of fluorine generator grains. Based on thermochemical calculations, blends of these salts with a fuel and fluorocarbon binder were formulated and pressed into solid grains. Stability, compatibility, and sensitivity data were gathered. Solid grains were successfully combusted. Mechanical properties were demonstrated from inert specimen data. Precombustor concepts and insulation materials were evaluated for application to HF/DF laser systems.
