Background and aim: Few studies have reported hepatitis B surface antigen (HBsAg) kinetics after nucleos(t)ide analog (NA) discontinuation in patients with noncirrhotic chronic hepatitis B (CHB). The study specifically investigated long-term HBsAg kinetics after NA discontinuation. Methods: Between January 2014 to January 2024, this study prospectively enrolled 106 outpatients with noncirrhotic CHB who met the discontinuation criteria after NA consolidation treatment. Demographic, clinical, and laboratory data were collected and analyzed after NA discontinuation. Results: Ninety-six patients who finished 5 years of follow-up were included. HBsAg remained undetectable in 29 patients with end of treatment (EOT) HBsAg negativity. Among 67 patients with EOT HBsAg positivity, HBsAg seroclearance occurred in 12 (17.9%) patients with an estimated annual incidence of HBsAg seroclearance of 3.6%. Patients with EOT HBsAg levels of ≤1000 IU/mL had a higher HBsAg seroclearance rate than those with EOT HBsAg levels of >1000 IU/mL (33.3% vs. 5.4%). The proportion of patients with HBsAg ≤1000 IU/mL increased during follow-up. Logistic regression analysis indicated that the EOT HBsAg level was an independent factor for HBsAg seroclearance and an HBsAg level decline exceeding 1 log10 IU/mL. The optimal EOT HBsAg cutoff for both HBsAg seroclearance and an HBsAg level decline exceeding 1 log10 IU/mL was 359 IU/mL. Conclusions: Patients with EOT HBsAg negativity experienced no relapse and maintained HBsAg seroclearance during 5 years of follow-up after NA discontinuation. A higher HBsAg seroclearance rate can be obtained in patients with EOT HBsAg levels of ≤1000 IU/mL during 5 years of follow-up after NA discontinuation. Close monitoring and proper NA retreatment are recommended to guarantee the safety of NA discontinuation. Clinical trial number: Clinicaltrials.gov number NCT02883647.

Abstract The early-life organ development and maturation shape the fundamental blueprint for later-life phenotype. However, a multi-organ proteome atlas from infancy to adulthood is currently not available. Herein, we present a comprehensive proteomic analysis of ten mouse organs (brain, heart, lung, liver, kidney, spleen, stomach, intestine, muscle and skin) at three crucial developmental stages (1-, 4- and 8-weeks after birth) acquired using data-independent acquisition mass spectrometry. We detect and quantify 11,533 protein groups across the ten organs and obtain 115 age-related differentially expressed protein groups that are co-expressed in all organs from infancy to adulthood. We find that spliceosome proteins prevalently play crucial regulatory roles in the early-life development of multiple organs, and detect organ-specific expression patterns and sexual dimorphism. This multi-organ proteome atlas provides a fundamental resource for understanding the molecular mechanisms underlying early-life organ development and maturation.

Abstract Background The immunoglobulin superfamily protein Trem2 (triggering receptor expressed on myeloid cells 2) is primarily expressed on myeloid cells where it functions to regulate macrophage‐related immune response induction. While macrophages are essential mediators of diabetic wound healing, the specific regulatory role that Trem2 plays in this setting remains to be established. Objective This study was developed to explore the potential importance of Trem2 signalling in diabetic wound healing and to clarify the underlying mechanisms through which it functions. Methods and results Following wound induction, diabetic model mice exhibited pronounced upregulation of Trem2 expression, which was primarily evident in macrophages. No cutaneous defects were evident in mice bearing a macrophage‐specific knockout of Trem2 (T2‐cKO), but they induced more pronounced inflammatory responses and failed to effectively repair cutaneous wounds, with lower levels of neovascularization, slower rates of wound closure, decreased collagen deposition following wounding. Mechanistically, we showed that interleukin (IL)‐4 binds directly to Trem2, inactivating MAPK/AP‐1 signalling to suppress the expression of inflammatory and chemoattractant factors. Co‐culture of fibroblasts and macrophages showed that macrophages from T2‐cKO mice suppressed the in vitro activation and proliferation of dermal fibroblasts through upregulation of leukaemia inhibitory factor (Lif). Injecting soluble Trem2 in vivo was also sufficient to significantly curtail inflammatory responses and to promote diabetic wound healing. Conclusions These analyses offer novel insight into the role of IL‐4/Trem2 signalling as a mediator of myeloid cell‐fibroblast crosstalk that may represent a viable therapeutic target for efforts to enhance diabetic wound healing.

The burden of fungal infections on human health is increasing worldwide. Aspergillus, Candida, and Cryptococcus are the top three human pathogenic fungi that are responsible for over 90% of infection-related deaths. Moreover, effective antifungal therapeutics are lacking, primarily due to host toxicity, pathogen resistance, and immunodeficiency. In recent years, nanomaterials have proved not only to be more efficient antifungal therapeutic agents but also to overcome resistance against fungal medication. This review will examine the limitations of standard antifungal therapy as well as focus on the development of nanomaterials.

Background and aims: There is currently no single model for predicting Wilson's disease (WD). We aimed to create a nomogram using daily clinical parameters to improve the accuracy of WD diagnosis in patients with abnormal liver function. Methods: Between July 2016 and December 2020, we identified 90 WD patients with abnormal liver function who had homozygous or compound heterozygous mutations in the ATP7B gene. The control group included 128 patients with similar liver function but no WD during the same time period. To create a nomogram, we screened potential predictive variables using the least absolute shrinkage and selection operator model and multivariate logistic regression. Results: We developed a nomogram for screening for WD based on six predictive factors: serum copper, direct bilirubin, uric acid, cholinesterase, prealbumin, and reticulocyte percentage. In the training cohort, the area under curve (AUC) of the nomogram reached 0.967 (95% confidence interval (CI) 0.946–0.988), while the area under the precision-recall curve was 0.961. Based on the optimal cutpoint of 213.55, our nomogram performed well, with a sensitivity of 96% and a specificity of 87%. In the validation cohort, the AUC of the nomogram was as high as 0.991 (95% CI 0.970–1.000). Conclusions: We developed a nomogram that can predict the risk of WD prior to the detection of serum ceruloplasmin or urinary copper, greatly increasing screening efficiency for patients with abnormal liver function.

Abstract Non-small cell lung cancer (NSCLC) is one of the most threatening malignancies to human health and life. In most cases, patients with NSCLC are already at an advanced stage when they are diagnosed. In recent years, lung cancer has made great progress in precision therapy, but the efficacy of immunotherapy is unstable, and its response rate varies from patient to patient. Several biomarkers have been proposed to predict the outcomes of immunotherapy, such as programmed cell death-ligand 1 (PD-L1) expression and tumor mutational burden (TMB). Nevertheless, the detection assays are invasive and demanding on tumor tissue. To effectively predict the outcomes of immunotherapy, novel biomarkers are needed to improve the performance of conventional biomarkers. Liquid biopsy is to capture and detect circulating tumor cells (CTCs), circulating tumor DNA (ctDNA) and exosomes in body fluids, such as blood, saliva, urine, pleural fluid and cerebrospinal fluid as samples from patients, so as to make analysis and diagnosis of cancer and other diseases. The application of liquid biopsy provides a new possible solution, as it has several advantages such as non-invasive, real-time dynamic monitoring, and overcoming tumor heterogeneity. Liquid biopsy has shown predictive value in immunotherapy, significantly improving the precision treatment of lung cancer patients. Herein, we review the application of liquid biopsy in predicting the outcomes of immunotherapy in NSCLC patients, and discuss the challenges and future directions in this field.

We show by constructing explicit homotopy operators that the Hochschild (co)homology of an $ A_{\infty} $-algebra of Stasheff admits a differential calculus structure. As an application, we reproduce a result of Tradler which says that the Hochschild cohomology of a cyclic $ A_\infty $-algebra admits a Batalin-Vilkovisky algebra structure.

Abstract Background Neutrophilic inflammation in the airway is a hallmark of bronchiectasis. Neutrophil extracellular traps (NETs) have been reported to play an important role in the occurrence and development of bronchiectasis. Neutrophil side fluorescence is one of the characteristics of neutrophils that can reflect the activation of neutrophils and the formation of NETs. Objective To explore the relationship between the values of neutrophil side fluorescence (NEUT-SFL) in the peripheral blood of bronchiectasis patients, and the severity of the disease. Methods 82 patients with bronchiectasis from the Department of Respiratory and Critical Medicine, at the Third Affiliated Hospital of Southern Medical University and were scored with Bronchiectasis Severity Index (BSI) (2019–2021). The clinical data such as the value of NEUT-SFL, neutrophil count, C-reactive protein, and procalcitonin levels were collected and retrospectively analyzed. NEUT-SFL values neutrophil count from 28 healthy subjects were also used to ascertain cut-off values. Results Based on the BSI scores, patients were divided into three categories as mild (32%), moderate (29%), and severe (39%). Our results showed that the values of NEUT-SFL were higher in bronchiectasis patients compared to healthy controls. The levels of NEUT-SFL positively correlated with the high BSI scores in patients (P = 0.037, r = 0.23) and negatively correlated with the lung function in these patients (r = − 0.35, P = 0.001). The area under the ROC curve was 0.813, the best cut-off was 42.145, indicating that NEUT-SFL values > 42.145 can potentially predict the severity of bronchiectasis. Conclusions The values of NEUT-SFL in the peripheral blood can be used for predicting the severity of bronchiectasis.

The current study was performed to assess the effects of dietary administration of ferulic acid (FA) on digestive enzyme activities, antioxidant capacity, morphology, immunity, and microbiota in the gut of hybrid grouper (Epinephelus fuscoguttatus♀ × Epinephelus polyphekadion♂). A total of 630 fish (9.51 ± 0.01 g) were allocated randomly to seven dietary treatments, each with three replicates. Experiment diets supplemented with FA at the levels of 0, 40, 80, 160, 320, 640, and 1280 mg kg−1 (FA0 (control), FA40, FA80, FA160, FA320, FA640, and FA1280), were fed to fish for 70 days. The results showed that dietary 80–160 mg kg−1 FA intake considerably augmented the enzymes activities of digestion (lipase, trypsin, amylase, and pepsin) and antioxidation (superoxide dismutase, glutathione peroxidase, and catalase) and the values of immune parameters (Immunoglobulin, C3 and lysozyme), as well as reduced malondialdehyde content, with respect to the control group (P

At the end of 2019, the outbreak of the COVID-19 has a huge impact on China and the world economy, highlighting the vulnerability of the economy in the face of major shocks. The capability to resist pressure and risk of economic resilience can significantly improve the capability of the economy to resist external interference and recover its development. Kunming is located in the “Belt and Road” Economic Zone and is a bridgehead that radiates South Asia and Southeast Asia. Its economic development is particularly affected by the epidemic. How to improve the resilience of the regional economy will be of great strategic significance to achieve high-quality economic development. Taking Kunming as an example, this paper identifies the influencing factors of regional economic development through literature research and expert interviews and constructs a hypothesis model of influencing factors of economic resilience. From the three dimensions of “defense, absorption and learning” of resilience theory, data are collected through questionnaires, and the mechanism between resilience theory and economic development is explored by using the structural equation model(SEM). According to the main factors identified by the model, the paper figures out countermeasures to prompt the development of the regional economic better and faster.

This paper focused on the impact of window shades’ physical characteristics and opening modes on annual air conditioning energy consumption of residential buildings in China. Three building models with different window layouts were proposed initially, and then validated by the dimensionless indicator annual energy performance (AEP) to determine the final building model adopted in this study. Harbin, Beijing, Guangzhou, Changsha and Kunming were selected to represent five different climate zones in China. The shading model has taken six physical characteristics of window shades, viz. outer emissivity, inner emissivity, solar transmittance, solar reflectance, thermal infrared transmittance, conductivity and two opening modes, viz. up & down mode as well as left & right mode, into consideration. Besides, the window shades operation schedule was summarized through 949 valid questionnaires. The annual air conditioning energy consumption was calculated via EnergyPlus software, and then the impact of the above different opening modes and parameters on energy use were investigated. The analysis results indicated that the top & bottom opening mode is better than the left & right mode when used in Harbin, and the effect of two different modes in air conditioning energy consumption in Kunming is similar. But in the other three cities, the left and right mode is preferred. Besides, the analysis results of the impact of different physical characteristics on air conditioning energy use in different climate zones could help improve the energy performance of window shades.

The golden pompano (Trachinotus blochii) is a warm water fish that is widely distributed in tropical and subtropical waters. T. blochii is one of the most important economic species in China where it is produced in marine cage culture; however, few studies have been performed on the mechanism of sex determination and differentiation, and there are no obvious morphological differences between males and females. To further understand the role of long non-coding RNAs (lncRNAs) in sex determination and gonadal development of T. blochii, we conducted a tran-scriptome analysis of T. blochii ovaries and testes. In this study, a total of 530,128,262 clean reads were produced from six cDNA libraries, after stringent filtering. The RNA-seq analysis yielded 13,509 novel lncRNAs, 2621 novel mRNAs, and 18,683 known mRNA transcripts with 4909 and 10,410 differentially expressed lncRNAs and mRNAs. The Gene Ontology (GO) enrichment patterns between the lncRNAs and mRNAs were different. Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis confirmed that some of the differentially expressed mRNAs and target genes of the lncRNAs were mainly associated with several sex determination and gonadal development signaling pathways, including the GnRH signaling pathway, the Wnt signaling pathway, and the insulin signaling pathway. The lncRNA-mRNA co-expression network analysis showed that the lncRNA TCONS_00083175 may play a key role in the T. blochii sex differentiation. The findings broaden our understanding of lncRNAs and gonad genetics, and provide new insight into the function of lncRNAs in sex differentiation and highlight a key lncRNA for future studies.

Golden Pompano (Trachinotus blochii) has rapidly developed into the one of the main valuable fish species in Chinese marine aquaculture. Due to its rapid growth, active metabolism, and high oxygen consumption, hypoxia will increase its mortality and cause serious economic losses. We constructed two experimental groups of fish with different degrees of tolerance to hypoxia, used BSR-Seq analysis based on genome and genetic linkage groups to locate SNPs and genes that were related to the differences in hypoxia tolerance. The results showed that hypoxia tolerance SNPs of golden pompano may be jointly determined by multiple linkage groups, especially linkage groups 18 and 22. There were 768 and 348 candidate genes located in the candidate regions of the brain and liver, respectively. These genes were mainly involved in anaerobic energy metabolism, stress response, immune response, waste discharge, and cell death. The prostaglandin-endoperoxide synthase 2 (PTGS2) on LG8, which is involved in the metabolism of arachidonic acid, has a G/A nonsynonymous mutation at position 20641628, and the encoded amino acid was changed from hydrophobic aspartic acid to asparaginate. The specific pathway of the RIG-I-like receptor signaling pathway in the liver may mediate the metabolic system and the immune system, linking glucose metabolism with immune regulation. The death of the hypoxia-intolerant group may be due to the accumulation of lactic acid caused by the activation of anaerobic glycolysis during the early stage of hypoxia stress, and the activation of type I interferon was inhibited, which resulted in decreased immunity. Among the genes involved in the RIG-I-like receptor signaling pathway, the CYLD Lysine 63 Deubiquitinase (CYLD) located on LG16 had a G/T nonsynonymous mutation at position 13629651, and the encoded amino acid was changed from alanine acid to valine. The interferon induced with helicase C domain 1 (Ifih1) located on LG18 has a G/C nonsynonymous mutation at position 16153700, and the encoded hydrophilic glycine was changed to hydrophobic alanine. Our findings suggest these SNPs may assist in the molecular breeding of hypoxia-tolerant golden pompano, and speculate that the balance of glucose and lipid metabolism plays a key role in Trachinotus blochii under acute hypoxia.

Rationale: Oxidative stress plays a critical role in the development of cardiac remodeling and heart failure. Lutein, the predominant nonvitamin A carotenoid, has been shown to have profound effects on oxidative stress. However, the effect of lutein on angiotensin II (Ang II)-induced cardiac remodeling and heart failure remains unknown. Objective: The aim of this study was to determine whether lutein is involved in cardiac remodeling and to elucidate the underlying molecular mechanisms. Methods and results: In vitro experiments with isolated neonatal rat cardiomyocytes (NRCMs) and cardiac fibroblasts (CFs) revealed that lutein significantly attenuated Ang II-induced collagen expression in CFs, and cardiomyocyte hypertrophy. The Ang II-induced increases in superoxide generation, inflammation and apoptosis in cultured CFs were strikingly prevented by lutein. In vivo, fibrosis, hypertrophic cardiomyocyte and superoxide generation were analyzed, and lutein was demonstrated to confer resistance to Ang II-induced cardiac remodeling in mice. Mechanistically, RNA sequencing revealed that interleukin-11 (IL-11) expression was significantly upregulated in mouse hearts in response to Ang II infusion and was significantly suppressed in the hearts of lutein-treated mice. Furthermore, IL-11 overexpression blocked the effects of lutein on fibrosis and oxidative stress in CFs and impaired the protective effect of lutein on cardiac remodeling. Notably, we discovered that lutein could reduce Ang II-induced IL-11 expression, at least partly through the regulation of activator protein (AP)-1 expression and activity. Conclusions: Lutein has potential as a treatment for cardiac remodeling and heart failure via the suppression of IL-11 expression.

Face recognition plays an important role in many robotic and human–computer interaction systems. To this end, in recent years, sparse-representation-based classification and its variants have drawn extensive attention in compress sensing and pattern recognition. For image classification, one key to the success of a sparse-representation-based approach is to extract consistent image feature representations for the images of the same subject captured under a wide spectrum of appearance variations, for example, in pose, expression and illumination. These variations can be categorized into two main types: geometric and textural variations. To eliminate the difficulties posed by different appearance variations, the article presents a new collaborative-representation-based face classification approach using deep aligned neural network features. To be more specific, we first apply a facial landmark detection network to an input face image to obtain its fine-grained geometric information in the form of a set of 2D facial landmarks. These facial landmarks are then used to perform 2D geometric alignment across different face images. Second, we apply a deep neural network for facial image feature extraction due to the robustness of deep image features to a variety of appearance variations. We use the term deep aligned features for this two-step feature extraction approach. Last, a new collaborative-representation-based classification method is used to perform face classification. Specifically, we propose a group dictionary selection method for representation-based face classification to further boost the performance and reduce the uncertainty in decision-making. Experimental results obtained on several facial landmark detection and face classification data sets validate the effectiveness of the proposed method.

OBJECTIVE: To the best of our knowledge, the association of insulin resistance (IR) with chronic kidney disease (CKD) has not been well studied in normal-weight individuals. The aim of this study is to examine whether IR is associated with CKD in non-diabetic subjects with normal weight. We also examine whether the presence of obesity modifies the association of IR with CKD. METHODS: Data were drawn from a cross-sectional survey in China. Both estimated glomerular filtration rate and urinary albumin to creatinine ratio were used as markers of CKD. Logistic regression models and the quartiles of homeostatic model assessment of insulin resistance were used to explore the associations of IR with CKD in entire cohort, normal-weight and overweight/obese subpopulations. RESULTS: In normal-weight subpopulation, the prevalence of IR and metabolic syndrome were 11.11% and 8.99%, respectively. In the entire cohort, the highest quartile HOMA-insulin resistance had a 70% increased risk for CKD (RR 1.70, 95% CI 1.07, 2.71, P=0.03, comparing the highest to the lowest quartile). However, when adding obesity to the model, the association was abolished. IR was associated with CKD in overweight/obese subpopulation but not in normal-weight subpopulation. CONCLUSION: IR and MetS in normal-weight individuals is common in the Chinese population. IR is associated with CKD in overweight/obese subpopulation but not in normal-weight subpopulation and the presence of obesity modifies the association of IR with CKD.

BACKGROUND: High serum levels of lipopolysaccharide (LPS) with LPS-MD-2/TLR4 complex activated NF-kb and cytokine cause hepatic necrosis in animal models. We investigated the dynamic changes of LPS levels in patients with acute on chronic hepatitis B liver failure (ACHBLF). METHODS: We enrolled ACHBLF patients for a 12-week study. Patients' LPS levels were measured along with 10 healthy controls. Patients on supportive care and recovered without intervention(s) were analyzed. Patients' LPS levels during the disease progression phase, peak phase, and remission phase were compared with healthy controls. RESULTS: Among 30 patients enrolled, 25 who received interventions or expired during the study period were excluded from the analysis, five patients on supportive care who completed the study were analyzed. RNA 5-methylcytosine (m5C) is an abundant chemical modification in mammalian RNAs and plays crucial roles in regulating vital physiological and pathological processes, especially in cancer. However, the dysregulation of m5C and its underlying mechanisms in non-small cell lung cancer (NSCLC) remain unclear. Here we identified that NSUN2, a key RNA m5C methyltransferase, is highly expressed in NSCLC tumor tissue. We found elevated NSUN2 expression levels strongly correlate with tumor grade and size, predicting poor outcomes for NSCLC patients. Furthermore, RNA-seq and subsequent confirmation studies revealed the antioxidant-promoting transcription factor NRF2 is a target of NSUN2, and depleting NSUN2 decreases the expression of NRF2 and increases the sensitivity of NSCLC cells to ferroptosis activators both in vitro and in vivo. Intriguingly, the methylated-RIP-qPCR assay results indicated that NRF2 mRNA has a higher m5C level when NSUN2 is overexpressed in NSCLC cells but shows no significant changes in the NSUN2 methyltransferase-deficient group. Mechanistically, we confirmed that NSUN2 upregulates the expression of NRF2 by enhancing the stability of NRF2 mRNA through the m5C modification within its 50UTR region recognized by the specific m5C reader protein YBX1, rather than influencing its translation. In subsequent rescue experiments, we show knocking down NRF2 diminished the proliferation, migration, and ferroptosis tolerance mediated by NSUN2 overexpression. In conclusion, our study unveils a novel regulatory mechanism in which NSUN2 sustains NRF2 expression through an m5C--YBX1--axis, suggesting that targeting NSUN2 and its regulated ferroptosis pathway might offer promising therapeutic strategies for NSCLC patients. Background. As a tumor suppressor, p16 can competitively block the cyclin D1-CDK4/6 complex to arrest the cell cycle in the G1 phase. Lack of p16 gene expression can lead to infinite cell proliferation and malignant transformation. Objectives. To determine whether the hepatitis B virus X protein (HBx) regulates the methylation and expression of the p16 gene. Materials and methods. We constructed a eukaryotic expression vector carrying the HBx gene and green fluorescent protein (GFP), and transfected it into HepG2 cells to build cell lines stably expressing GFP and GFP-HBx. The p16 protein level and p16 gene methylation were measured in these cell lines. We further detected the mRNA expression of DNA methyltransferases (DNMTs) 1, 3A and 3B. The DNMT1, DNMT3A and DNMT3B gene promoter sequences were inserted into a reporter vector (pGL3-Basic) to build recombinant vectors, which were then transiently transfected into different cell lines. After 48 h, the luciferase activity was measured. Results. The level of p16 protein in GFP-HBx/HepG2 cells was significantly lower than in HepG2 and GFP/HepG2 cells. The CpG methylation was present in the p16 gene promoter region of GFP-HBx/HepG2 cells. The DNMT1 and DNMT3A mRNA levels in GFP-HBx/HepG2 cells were significantly elevated compared to that in the HepG2 cells (p = 0.0495). The luciferase activity in GFP-HBx/HepG2 cells transfected with the pGL3-DNMT1/3A pro plasmid was significantly higher than in HepG2 and GFP/HepG2 cells (both p < 0.05). Conclusions. The HBx can induce p16 gene promoter methylation and inhibit the expression of p16 in HepG2 cells. This occurs due to HBx activation of DNMT1 and DNMT3A promoters and the induction of p16 promoter methylation, which downregulates the expression of p16 protein. [ABSTRACT FROM AUTHOR]

In this study, carbon fiber-graphite flake/copper composites were prepared by powder metallurgy. The impact of the alloying elements Ti and Zr on the composites' properties was investigated, and the influence of varying Zr contents on the phase structure and thermal conductivity of the composites was also explored. The results indicate that composites containing Zr exhibit better flexural strength and thermal conductivity compared to those containing Ti. When the volume fraction of carbon fiber is 2% and 2 wt% Zr is added to copper matrix, the carbide layer at the composite interface is uniform and has a thickness of 0.36 µm. The thermal conductivity of the composite material is up to 597.5 W/(m·K). The mechanical properties of the composites are enhanced by the synergistic effect of adding carbon fiber and alloying matrix and the composite shows a remarkable flexural strength of 150.5 MPa, which is 58.3% higher compared to composites without carbon fiber.
