ABSTRACT Background The primary cause of cancer‐related fatalities globally is lung cancer. Although the chemotherapy drug cisplatin (DDP) has brought certain benefits to patients, the rapid development of drug resistance has greatly hindered treatment success. Methods We used the lung squamous cell carcinoma (LUSC) mRNA data set to explore the differentially expressed gene (RND1) in LUSC and detected RND1 expression in LUSC cells and DDP‐resistant cells by qRT‐PCR. Meanwhile, we performed abnormal expression treatment on RND1 and conducted CCK8, colony formation, and flow cytometry to evaluate the impact of RND1 expression on cell proliferation, apoptosis, and DDP resistance. In addition, we analyzed metabolism pathways involving RND1 using GSEA. We also used online tools such as hTFtarget and JASPAR to screen for the upstream transcription factor FOXA2 of RND1 and verified their relationship through CHIP and dual luciferase experiments. Finally, we validated the role of FOXA2‐RND1 in DDP resistance in LUSC through the above experiments. Results RND1 was downregulated in LUSC, and overexpression of RND1 repressed proliferation and DDP resistance of LUSC cells and facilitated cell apoptosis. RND1 modulated the arachidonic acid (AA) metabolism pathway, and FOXA2 positively manipulated RND1 expression. By activating FOXA2, stabilizing RND1, and regulating AA levels, the sensitivity of LUSC cells to DDP could be enhanced. Conclusion Our study suggested that FOXA2 positively modulated the RND1‐AA pathway, which repressed the resistance of LUSC cells to DDP.

The coupling of railway power conditioner (RPC), network, and train will change the impedance characteristics of the existing traction power supply systems, which will lead to the unclear law of high-frequency harmonic resonance. This paper aims to study the resonance mechanism of the RPC-network train coupling system, and then solve the resonance problem of the existing traction power supply system from the perspective of reshaping the high-frequency impedance of the system. Firstly, the RPC-network-train coupling system’s dynamic node admittance matrix is established. After that, a comprehensive resonance evaluation method is proposed to obtain the resonance impact factors (RIFs) of multiple virtual impedances in the control strategy and the filtering parameters, which can realize the comparison and selection of five existing impedance reshaping strategies. To ensure the safe and stable operation of the RPC-network-train coupling system, a multi-objective sorting optimization strategy (MOSOS) considering resonance suppression, harmonics filtering, filter cost, and control system stability is proposed for the RPC based on the RIFs. Finally, the effectiveness of the proposed method is verified by the simulation result in MATLAB/Simulink and the experiment result in the StarSim hardware-in-the-loop (HIL) platform respectively.

Abstract Preeclampsia (PE) is considered as a disease of placental origin. However, the specific mechanism of placental abnormalities remains elusive. This study identified thrombospondin‐1 (THBS1) is downregulated in preeclamptic placentae and negatively correlated with blood pressure. Functional studies show that THBS1 knockdown inhibits proliferation, migration, and invasion and increases the cycle arrest and apoptosis rate of HTR8/SVneo cells. Importantly, THBS1 silencing induces necroptosis in HTR8/SVneo cells, accompanied by the release of damage‐associated molecular patterns (DAMPs). Necroptosis inhibitors necrostatin‐1 and GSK′872 restore the trophoblast survival while pan‐caspase inhibitor Z‐VAD‐FMK has no effect. Mechanistically, the results show that THBS1 interacts with transforming growth factor B‐activated kinase 1 (TAK1), which is a central modulator of necroptosis quiescence and affects its stability. Moreover, THBS1 silencing up‐regulates the expression of neuronal precursor cell‐expressed developmentally down‐regulated 4 (NEDD4), which acts as an E3 ligase of TAK1 and catalyzes K48‐linked ubiquitination of TAK1 in HTR8/SVneo cells. Besides, THBS1 attenuates PE phenotypes and improves the placental necroptosis in vivo. Taken together, the down‐regulation of THBS1 destabilizes TAK1 by activating NEDD4‐mediated, K48‐linked TAK1 ubiquitination and promotes necroptosis and DAMPs release in trophoblast cells, thus participating in the pathogenesis of PE.

Background Among those with heart failure (HF), women are more likely to develop depression than men. Few studies have focused on the outcomes of female patients with HF with depressive symptoms. Methods and Results A total of 506 female patients with HF with preserved ejection fraction were included in this secondary analysis from the TOPCAT (Treatment of Preserved Cardiac Function Heart Failure With an Aldosterone Antagonist) cohort, and 439 female patients with HF with reduced ejection fraction were included from the HF‐ACTION (Heart Failure: A Controlled Trial Investigating Outcomes of Exercise Training) cohort. Depressive symptoms were measured using the Patient Health Questionnaire‐9 and Beck Depression Inventory‐II. The depression class was categorized by severity, and the change in clinical depression class was defined as aggravated (1‐grade increase) or improved (1‐grade decrease). The prognostic value of depressive symptoms was determined by using multivariable Cox proportional hazards models. Female patients with improved depressive symptoms had worse depressive status at baseline and lower baseline Kansas City Cardiomyopathy Questionnaire scores. Depression class at the 12‐month visit and depression class change were the dominant prognostic factors for cardiovascular death in female patients with HF with preserved ejection fraction (hazard ratio [HR], 1.43 [95% CI, 1.02–2.01], P=0.036; HR, 1.71 [95% CI, 1.14–2.55], P=0.009). BackgroundTetanus is a rare surgical infectious disease with a high reported relevant mortality. It still remains a serious problem in public health, particularly in low-income and middle-income countries. The purpose of this study was to investigate the management and prognosis of adult generalized tetanus in our hospital.MethodsA total of 20 adult generalized tetanus patients were recruited in this retrospective observational study. Patients were retrieved from the hospital data base via discharge diagnosis. Patients were divided into two groups (Severe or Non-severe tetanus group) based on the severity of tetanus by using the Ablett classification. The differences between the two groups were compared.ResultsThe study included 11 males (55%) and 9 females (45%). All tetanus patients recovered. The median age was 53.5 years [IQR: 19–78]. There were 1 mild (Grade 1) case (5%),5 moderate (Grade 2) cases (25%), 2 severe (Grade 3) cases (10%), and 12 very severe (Grade 4) cases (60%). Nineteen patients (95%) did not have tetanus immunization before. The majority of patients were farmers (60%), and came from rural areas (60%). Thirteen (65%) patients had a history of puncture injury. The rate of wound debridement after admission was 60% overall. Thirteen (65%) patients required mechanical ventilation for a median of 21 [IQR:12–41] days. Autonomic instability occurred in 13 (65%) patients. Pulmonary infections occurred in 12 (60%) patients. Median duration of hospital stay was 29.5 [IQR:12–68] days. ObjectivesTo explore the preoperative high-risk clinical factors for contralateral medium-volume central lymph node metastasis (conMVCLNM) in unilateral papillary thyroid carcinoma (uPTC) and the indications for dissection of contralateral central lymph nodes (conCLN).MethodsClinical and pathological data of 204 uPTC patients who underwent thyroid surgery at the Hangzhou First People’s Hospital from September 2010 to October 2022 were collected. Univariate and multivariate logistic regression analyses were conducted to determine the independent risk factors for contralateral central lymph node metastasis (conCLNM) and conMVCLNM in uPTC patients based on the preoperative clinical data. Predictive models for conCLNM and conMVCLNM were constructed using logistic regression analyses and validated using receiver operating characteristic (ROC) curves, concordance index (C-index), calibration curves, and decision curve analysis (DCA).ResultsUnivariate and multivariate logistic regression analyses showed that gender (P < 0.001), age (P < 0.001), tumor diameter (P < 0.001), and multifocality (P = 0.008) were independent risk factors for conCLNM in uPTC patients. Gender(P= 0.026), age (P = 0.010), platelet-to-lymphocyte ratio (PLR) (P =0.003), and tumor diameter (P = 0.036) were independent risk factors for conMVCLNM in uPTC patients. A predictive model was established to assess the risk of conCLNM and conMVCLNM, with ROC curve areas of 0.836 and 0.845, respectively. The C-index, the calibration curve, and DCA demonstrated that the model had good diagnostic value.ConclusionGender, age, tumor diameter, and multifocality are high-risk factors for conCLNM in uPTC patients. Gender, age, tumor diameter, and PLR are high-risk factors for conMVCLNM in uPTC patients, and preventive conCLN dissection should be performed.

Background: Systemic lupus erythematosus (SLE) is a chronic autoimmune disease involving multi-system and multi-organ dysfunction, and is easily misdiagnosed early in the disease course. We aimed to accurately predict early SLE nomogram to provide a reference basis for the early clinical diagnosis of SLE. Methods: We retrospectively analyzed 167 patients who were first diagnosed with SLE at Fengxian District Central Hospital, Shanghai, between March 2017 and October 2022. Three groups of 129 physically healthy subjects, 67 patients with rheumatoid arthritis, and 40 patients with rashes were selected as controls during the same period. Patients with SLE and control group were randomly divided into training (n = 217) and validation (n = 141) group. Univariate and multivariate analyses were used to identify independent risk factors for early SLE diagnosis. The independent risk factors for diagnosis were used to construct a nomogram to predict early SLE. Results: Based on the training group, three variables were identified as independently influencing early SLE: platelets (odds ratio OR = 0.993, P = 0.047), albumin (OR = 0.833, P = 0.007), and complement component 1q (OR = 0.956, P = 0.000). The precision of the nomogram was assessed using C-index values and calibration plot diagrams. The C-index values were 0.929 for training group and 0.898 for validation group. Both the training group and validation group calibration curves showed good predicted outcomes. Conclusion: The construction of a nomogram can accurately predict the risk of early SLE. The model showed good discriminatory power and calibration for use in the diagnosis of SLE, providing a visual tool and reference basis for the early diagnosis of SLE.

Abstract Background Substantial studies have demonstrated that oxidative stress placenta and endothelial injury are considered to inextricably critical events in the pathogenesis of preeclampsia (PE). Systemic inflammatory response and endothelial dysfunction are induced by the circulating factors released from oxidative stress placentae. As a novel biomarker of oxidative stress, advanced oxidation protein products (AOPPs) levels are strongly correlated with PE characteristics. Nevertheless, the molecular mechanism underlying the effect of factors is still largely unknown. Methods With the exponential knowledge on the importance of placenta-derived extracellular vesicles (pEVs), we carried out lncRNA transcriptome profiling on small EVs (sEVs) secreted from AOPPs-treated trophoblast cells and identified upregulated lncRNA TDRKH-AS1 as a potentially causative factor for PE. We isolated and characterized sEVs from plasma and trophoblast cells by transmission electron microscopy (TEM), nanoparticle tracking analysis (NTA) and western blotting. The expression and correlation of lncRNA TDRKH-AS1 were evaluated using qRT-PCR in plasmatic sEVs and placentae from patients. Pregnant mice injected with TDRKH-AS1-riched trophoblast sEVs was performed to detect the TDRKH-AS1 function in vivo. To investigate the potential effect of sEVs-derived TDRKH-AS1 on endothelial function in vitro, transcriptome sequencing, scanning electron Microscopy (SEM), immunofluorescence, ELISA and western blotting were conducted in HUVECs. RNA pulldown, mass spectrometry, RNA immunoprecipitation (RIP), chromatin isolation by RNA purification (ChIRP) and coimmunoprecipitation (Co-IP) were used to reveal the latent mechanism of TDRKH-AS1 on endothelial injury. Results The expression level of TDRKH-AS1 was significantly increased in plasmatic sEVs and placentae from patients, and elevated TDRKH-AS1 in plasmatic sEVs was positively correlated with clinical severity of the patients. Moreover, pregnant mice injected with TDRKH-AS1-riched trophoblast sEVs exhibited a hallmark feature of PE with increased blood pressure and systemic inflammatory responses. Pyroptosis, an inflammatory form of programmed cell death, is involved in the development of PE. Indeed, our in vitro study indicated that sEVs-derived TDRKH-AS1 secreted from AOPPs-induced trophoblast elevated DDIT4 expression levels to trigger inflammatory response of pyroptosis in endothelial cells through interacting with PDIA4. Conclusions Herein, results in the present study supported that TDRKH-AS1 in sEVs isolated from oxidative stress trophoblast may be implicated in the pathogenesis of PE via inducing pyroptosis and aggravating endothelial dysfunction.

The antioxidant properties of butterfly pea flower (BF), which is rich in natural anthocyanins, have garnered significant attention. The impact of digestion and metabolism on BF extracts and evaluate their subsequent antioxidant activities in vivo were explored in the present study. After in vitro digestion, 42.03 ± 2.74% of total anthocyanins from BF extracts remained, indicating a negative influence of the digestion process on the bioaccessibility of phenolic compounds derived from BF. Furthermore, UPLC-LTQ-Orbitrap-MS2 analysis identified a total of four prototypes and twenty-seven metabolites in rat plasma or urine samples following the intake of BF extracts. The kinetics of key metabolites including delphinidin 3-glucoside (D3G), cyanidin-3-glucoside (C3G), and 4-hydroxybenzoic acid were subsequently determined in blood, and the Cmax values were 69.034 ± 8.05 nM and 51.65 ± 3.205 nM. These key metabolites derived from BF anthocyanins, including C3G and D3G, and flavonoid quercetin exhibited main antioxidant attributes that improved the plasmic and hepatic activities of various antioxidant enzymes and the total antioxidant capacity (T-AOC) and malondialdehyde (MDA) in a D-galactose-induced rat model. These findings provide insights into the bioaccessibility and bioavailability of bioactive constitutes derived from BF extracts, which are crucial for determining the actual efficacy of BF as well as developing functional foods based on BF.

Urban parks are important for improving the quality of living environments. Although the impact of parks on housing prices has been well documented, the effects of the COVID-19 pandemic remain vague. This paper analyzes the housing prices of neighborhoods around Meixi Lake park in Changsha, at the initial stage of the COVID-19 pandemic in June 2020 and the stable stage of the recovery period in June 2023, which demonstrates the impacts of urban parks on property pricing and housing choice based on residential transaction data. The results indicate that urban parks are given a high priority in determining people’s choices. In June 2020, the risk of epidemic transmission and noise interference lowered the price of property that is close to public parks and recreational facilities. However, good management and social services in residential areas increase housing prices. However, when the pandemic ended, the strong demand for outdoor activities led to a price rise in the properties near Meixi Lake park. People are most likely to choose houses in a neighborhood characterized by good educational facilities and a high-quality living environment. More specially, the houses with a short distance to parks and where residents can enjoy the view of a lake or mountains are preferable to any others. A residential area adjacent to a super large park paired with a small park is the most valuable consideration for property developers and housing consumers.

IntroductionThe COVID-19 pandemic, which began in the last quarter of 2019, has had a significant impact on urban transportation. With increasing demand for urban transport, the internal roads and public spaces of university campuses play an important role in facilitating commuting and communication between various functional zones. While considerable research has been conducted on route planning, pedestrian-vehicle segregation, and safety management in the internal transportation environment of university campuses, empirical investigations exploring barrier-free inclusive campus environment design and the subjective evaluation of road and public space users in the aftermath of the COVID-19 pandemic are lacking. Recent developments in travel behavior models and positive psychology have led to an increased focus on the correlation among subjective perceptions, attitudes, emotions, and commuting satisfaction in urban transportation and planning design.MethodsTo elucidate this relationship, a study was conducted on the new campus of Central South University in Changsha, Hunan Province, China. Using 312 valid samples, a structural equation model was constructed to analyse the relationship between commuting satisfaction and the barrier-free environment perception of university students regarding the internal transportation environment of the campus.ResultsThe results revealed that individuals' instantaneous barrier-free environment perceptions and long-term established positive emotions had a significant positive effect on commuting satisfaction. Furthermore, positive emotions were found to mediate the relationship between commuting attitudes induced by COVID-19, barrier-free environment perceptions, and commuting satisfaction.DiscussionThe results of this study provide a theoretical basis for the necessity of accessibility design in the post-COVID era. In addition, this study considers the perspective of users to provide ideas for the planning and construction of barrier-free campus environments that are based on convenient and inclusive design.

Objective: To evaluate the clinical effect of subcapsular saline injection (SCASI) after total thyroidectomy. Methods: A total of 77 patients who underwent total thyroidectomy in our hospital from January 2020 to December 2021 were selected and divided into the SCASI group (n = 43) and the non-SCASI group (n = 34). The general clinical data of the patients were collected, and serum parathyroid hormone (PTH) and serum calcium levels were determined preoperatively, on the 1st postoperative day, and at 1 and 6 months after the operation. These data were compared between groups. Results: There was no significant difference in postoperative complications between the two groups. IntroductionThe effects of restoration and inspiration in the therapeutic landscape of natural environments on visitors during the COVID-19 pandemic have been well-documented. However, less attention has been paid to the heterogeneity of visitor perceptions of health and the potential impacts of experiences in wetland parks with green and blue spaces on visitors’ overall perceived health. In this study, we investigate the impact of the restorative landscapes of wetland parks on visitors’ health perceptions in the context of the COVID-19 pandemic.MethodsIn our survey, 582 respondents participated in an online questionnaire. We analyzed the respondents’ health perceptions in terms of latent class analysis, used multinomial logistic regression to determine the factors influencing the potential categorization of health perceptions, and used structural equation modeling to validate the relationships between health perceptions of different groups and landscape perceptions of wetland parks, restorative experiences, and personality optimistic tendencies.ResultsThe results identified three latent classes of health perceptions. Gender, marital status, education, occupation, income, distance, frequency of activities, and intensity of activities were significant predictors of potential classes of perceived health impacts among wetland park visitors.DiscussionThis study revealed the nature and strength of the relationships between health perception and landscape perception, restorative experience, and dispositional optimism tendencies in wetland parks. These findings can be targeted not only to improve visitor health recovery but also to provide effective references and recommendations for wetland park design, planning, and management practices during and after an epidemic.

Abstract A novel three-phase traction power supply system is proposed to eliminate the adverse effects caused by electric phase separation in catenary and accomplish a unifying manner of traction power supply for rail transit. With the application of two-stage three-phase continuous power supply structure, the electrical characteristics exhibit new features differing from the existing traction system. In this work, the principle for voltage levels determining two-stage network is dissected in accordance with the requirements of traction network and electric locomotive. The equivalent model of three-phase traction system is built for deducing the formula of current distribution and voltage losses. Based on the chain network model of the traction network, a simulation model is established to analyze the electrical characteristics such as traction current distribution, voltage losses, system equivalent impedance, voltage distribution, voltage unbalance and regenerative energy utilization. In a few words, quite a lot traction current of about 99% is undertaken by long-section cable network. The proportion of system voltage losses is small attributed to the two-stage three-phase power supply structure, and the voltage unbalance caused by impedance asymmetry of traction network is less than 1‰. In addition, the utilization rate of regenerative energy for locomotive achieves a significant promotion of over 97%.

Abstract Hair conditions, such as hair loss and graying, are prevalent human conditions. But they are often poorly controlled due to our insufficient understanding of human scalp hair follicle (hsHF) in health and disease. Here we describe a comprehensive single-cell RNA-seq (scRNA-seq) analysis on highly purified black and early-stage graying hsHFs. Based on these, a concise single-cell atlas for hsHF and its early graying changes is generated and verified using samples from multiple independent individuals. These data reveal the lineage trajectory of hsHF in unprecedented detail and uncover its multiple unexpected features not found in mouse HFs, including the presence of an innerbulge like compartment in the growing phase, lack of a discrete companion layer, and enrichment of EMT features in HF stem cells (HFSCs). Moreover, we demonstrate that besides melanocyte depletion, early-stage human hair graying is also associated with specific depletion of matrix hair progenitors but not HFSCs. The hair progenitors’ depletion is accompanied by their P53 pathway activation whose pharmaceutical blockade can ameliorate hair graying in mice, enlightening a promising therapeutic avenue for this prevalent hair condition.

Linguistic terms have proven to be more effective for DMs when defining complex and imprecise cognition. Consequently, a variety of novel linguistic models such as fuzzy linguistic approach, hesitant fuzzy linguistic term sets (HFLTSs), and hesitant intuitionistic fuzzy linguistic term sets (HIFLTSs) have been successively proposed. As HIFLTSs provide an effective and symmetrical technique for expressing qualitative decisions by decision-makers, they have attracted substantial scholarly interest. This paper proposes a series of new operational laws to address the current limitations and enhance the applicability and methodology of HIFLTSs. Additionally, we introduce a new score function and an accuracy function that both consider the degree of hesitancy, thus addressing the current shortcomings. Furthermore, the correlation measures and correlation coefficients are established to enrich the framework of the HIFLTSs theory. Finally, a MULTIMOORA approach in the HIFLTS environment is presented, which results in more robustness, simplicity, and effectiveness. A practical example is employed to demonstrate the effectiveness of the proposed approach, and a comparison with related works highlights the advantages and novelty of the proposed model.

During an investigation of Xylariaceae from 2019 to 2020, isolates representing eight Nemania (Xylariacese) species were collected from Yunnan, Guizhou and Hainan Provinces in China. Morphological and multi-gene phylogenetic analyses, based on combined ITS, α-actin, rpb2 and β-tubulin sequences, confirmed that six of them are new to science, viz. Nemania camelliae, N. changningensis, N. cyclobalanopsina, N. feicuiensis, N. lishuicola and N. rubi; one is a new record (N. caries) for China and one is a known species (N. diffusa). Morphological descriptions and illustrations of all species are detailed. In addition, the characteristics of Nemania are summarised and prevailing contradictions in generic concepts are discussed.

Daidzein (D1) has been proved to be of great benefit to human health. More and more attention was paid to the metabolic process of D1. Most studies focused on the metabolites of D1 and analogs were determined through the excretion of animals and humans by traditional HPLC-MS, while their in situ distribution and metabolism in organs in vivo has not been reported. In our group, novel daidzein sulfonate derivatives were synthesized and confirmed to have excellent pharmaceutical properties. They exhibited good anti-inflammatory, inhibitory activities on human vascular smooth muscle cell proliferation and other bioactivities. Compared with traditional analytical methods, matrix-assisted laser desorption ionization time-of-flight mass spectrometry imaging (MALDI-TOF MSI) can directly analyze the distribution of compounds in tissues and organs. In this study, we investigate the in situ distribution and metabolism of D1 and its derivatives (DD2, DD3) in the organs of mice based on MALDI-TOF MSI for the first time. Trace prototype compounds were detected in the plasma 4 h after the intravenous injection of D1, DD2, and DD3. Seven phase I metabolites and seven phase II metabolites were detected. D1 sulfates were found in the plasma and in organs except the heart. The presence of D1 and DD3 monosulfates in the brain indicated that they could penetrate the blood–brain barrier. DD2 and DD3 could be hydrolyzed into D1 and their metabolic pathways were similar to those of D1. In addition, a ligand-receptor docking of D1 and DD2 with mitogen-activated protein kinase 8 (JNK1) was performed because of their significant anti-inflammatory activities through the JNK signaling pathway. It showed that the binding energy of DD2 with JNK1 was obviously lower than that of D1 which was consistent with their anti-inflammatory activities. It provided a theoretical basis for further validation of their anti-inflammatory mechanism at the protein level. In summary, the research will provide beneficial guidance for further pharmacological, toxicological studies and the clinical-use research of these compounds.

Green space around the university campus is of paramount importance for emotional and psychological restorations in students. Positive emotions in students can be aroused when immersed in green space and naturalness. However, to what extent can perceived naturalness influence students' positive emotion remains unclear, especially in the context of COVID-19 countermeasures. This study, therefore, attempts to investigate in-depth the nature and strength of the relationships between students' positive emotion and their perceived naturalness, place attachment, and landscape preference, which are potentially varying across universities in different social and environmental contexts and different restrictions policies regarding the COVID-19 pandemic. A course of questionnaire-based surveys was administered on two university campuses in Heilongjiang and Hunan Provinces, China, resulting in 474 effective samples. Structural equation modeling was used to explore the hypothetical conceptual framework of latent variables and the indicators. The findings indicate that the higher students' perceived naturalness results in greater positive emotion. Students' perceived naturalness in green spaces of campus has a positive effect on their place attachment and landscape preference. Moreover, the difference between mediate effects of place attachment and landscape preference were addressed, which verifies the contextual influences.

To achieve sustainable urban development, it is essential to gain insight into the spatial and temporal differentiation characteristics and the driving mechanisms of the urban thermal environment (UTE). As urbanization continues to accelerate, human activity and landscape configuration and composition interact to complicate the UTE. However, the differences in UTE-driven mechanisms at different stages of urbanization remain unclear. In this study, the UTE of Shenyang was measured quantitatively by using the land surface temperature (LST). The spatial and temporal differentiation characteristics were chronologically studied using the standard deviation ellipse (SDE) and hotspot analysis (Getis–Ord Gi*). Then, the relationship between human activities, landscape composition and landscape configuration and LST was explored in a hierarchical manner by applying the geographical detector. The results show that the UTE in Shenyang continues to deteriorate with rapid urbanization, with significant spatial and temporal differentiation characteristics. The class-level landscape configuration is more important than that at the landscape level when studying UTE-driven mechanisms. At the class level, the increased area and abundance of cropland can effectively reduce LST, while those of impervious surfaces can increase LST. At the landscape level, LST is mainly influenced by landscape composition and human activities. Due to rapid urbanization, the nonlinear relationship between most drivers and LST shifts to near-linear. In the later stage of urbanization, more attention needs to be paid to the effect of the interaction of drivers on LST. At the class level, the interaction between landscape configuration indices for impervious surfaces, cropland and water significantly influenced LST. At the landscape level, the interactions among the normalized difference building index (NDBI) and other selected factors are significant. The findings of this study can contribute to the development of urban planning strategies to optimize the UTE for different stages of urbanization.

Objective Prenatal infection and postpartum inflammatory experiences have been associated with increased risk of schizophrenia. To explore whether prenatal immune activation and prepubertal inflammation have cumulative impact on vulnerability to schizophrenia. Methods Polyinosinin-polycytidylic acid was injected into pregnant mice to establish maternal immune activation models on gestation day 9.5. Offspring were treated with poly I:C during puberty or adulthood as the second hit. 1)Using ELISA and quantitative real-time PCR to detect inflammatory response after administration of poly Ⅰ:C. 2)The effects of single or combined exposure to prenatal immune activation and prepubertal inflammation were examined by prepulse inhibition and locomotor reactions to dizocilpine(MK-801). Behavioral assessment took place when the animals reached adulthood. Results 1)Offspring from poly Ⅰ:C -treated mothers showed sensorimotor gating deficiency, as well as in the precipitation of behavioral hypersensitivity to the drugs dizocilpine, which were similar with schizophrenia. The phenotype of female was more pronounced than male. 2)However, double-hit model's sensorimotor gating deficient level was lower than that of prenatal immune activation model, and the phenotype of schizophrenia was relieved after second hit. Conclusions There is no synergistic pathological effect of prenatal immune activation plus pre-pubertal or adulthood inflammation.

A growing number of studies have demonstrated that N6 methyladenine (m6A) acts as an important role in the pathogenesis of reproductive diseases. Therefore, it is essential to profile the genome-wide m6A modifications such as in spontaneous abortion. In this study, due to the trace of human villi during early pregnancy, we performed high-throughput sequencing in villous tissues from spontaneous abortion (SA group) and controls with induced abortion (normal group) in the first trimester. Based on meRIP-seq data, 18,568 m6A peaks were identified. These m6A peaks were mainly located in the coding region near the stop codon and were mainly characterized by AUGGAC and UGGACG motif. Compared with normal group, the SA group had 2,159 significantly upregulated m6A peaks and 281 downregulated m6A peaks. Biological function analyses revealed that differential m6A-modified genes were mainly involved in the Hippo and Wnt signaling pathways. Based on the conjoint analysis of meRIP-seq and RNA-seq data, we identified thirty-five genes with differentially methylated m6A peaks and synchronously differential expression. And these genes were mainly involved in the Wnt signaling pathway, phosphatase activity regulation, protein phosphatase inhibitor activity, and transcription inhibitor activity. This study is the first to profile the transcriptome-wide m6A methylome in spontaneous abortion during early pregnancy, which provide novel insights into the pathogenesis and treatment of spontaneous abortion in the first trimester.

Outdoor physical activities can promote public health and they are largely influenced by the built environment in different urban settings. Understanding the association between outdoor physical activities and the built environment is important for promoting a high quality of life. Existing studies typically focus on one type of outdoor activity using interview-based small samples and are often lack of systematic understanding of the activities' intensity and frequency. In this study, we intend to gain deeper insight into how the built environment influences physical activities using the data extracted from individual's wearables and other open data sources for integrated analysis. Multi-linear regression with logarithm transformation is applied to perform the analysis using the data from Changsha, China. We found that built environment impacts on outdoor physical activities in Changsha are not always consistent with similar studies' results in other cities. The most effective measures to promote outdoor physical activities are the provision of good arterial and secondary road networks, community parks, among others in Changsha. The results shed light on future urban planning practices in terms of promoting public health.

Long noncoding RNAs (lncRNAs) play a significant role in cancer biology. This study aimed to determine the roles of lncRNAs in establishing the differences in clinical features between patients with papillary thyroid carcinoma (PTC) without Hashimoto’s thyroiditis (HT) and patients with PTC and HT. In the present study, we detected the differentially expressed lncRNAs between tumor tissues of patients with PTC with or without HT through lncRNA microarrays. The data were verified and analyzed through qRT-PCR, cell viability, cell cycle and bioinformatics analyses. We found that 1031 lncRNAs and 1338 mRNAs were abnormally expressed in 5 tissue samples of PTC complicated with HT [PTC/HT (+)] compared with 5 samples of PTC without HT [PTC/HT (-)]. Gene Ontology and pathway analyses of the mRNAs suggested that several biological processes and pathways, particularly immune system processes, were induced in the PTC/HT (+) tissues. Twenty lncRNAs were verified in 31 PTC/HT (+) and 64 PTC/HT (-) specimens by qRT-PCR, and the results were consistent with the microarray data. Specifically, ENST00000452578, a downregulated lncRNA in PTC/HT(+), was negatively correlated with the tumor size. Cell viability assays revealed that ENST00000452578 could inhibit cell proliferation. Our results indicate that lncRNAs and mRNAs play an important role in establishing the different clinical characteristics between patients with PTC/HT(+) and patients with PTC/HT(-), and might provide new insights from the perspective of RNA for obtaining a further understanding of the clinical features related to PTC with HT.

Background. Ferroptosis plays a key role in cardiomyopathy. Atorvastatin (ATV) has a protective effect on ischemia-reperfusion (I/R) cardiomyopathy. The purpose of this study is to elucidate the mechanism of ATV in I/R injury. Methods. H9C2 cells and cardiomyopathy rats were induced by hypoxia/reoxygenation (H/R) and I/R to construct in vitro and in vivo models. Cell viability was determined by CCK8. Cardiac histopathology was observed by HE staining. Transmission electron microscope (TEM) was used to observe the mitochondrial morphology. The reactive oxygen species (ROS) content in cells was analyzed by the biochemical method. ELISA was conducted to calculate the concentrations of total iron/Fe2+ and hepcidin. The expression of ferroptosis and SMAD pathway-related genes were detected by qPCR. Western blot was performed to detect the expression levels of ferroptosis and SMAD pathway-related proteins. Results. In H9C2 cells, ATV reversed the decline in cell viability, mitochondrial shrinkage, and ROS elevation induced by erastin or H/R. The concentration of total iron and Fe2+ in H/R-induced H9C2 cells increased, and the protein expression of FPN1 decreased. After ATV treatment, the concentration of total iron and Fe2+ decreased, and the protein expression of FPN1 increased. The expression of the SMAD7 gene in H/R-induced H9C2 cells decreased, and the expression of the hepcidin gene increased, which were reversed by ATV. When SMAD7 was knocked down, ATV treatment failed to produce the above effect. ATV also improved ferroptosis in I/R rat myocardium through the SMAD7/hepcidin pathway. Conclusions. ATV reversed the decline in H9C2 cell viability, mitochondrial shrinkage, and ROS elevation, and improved the myocardium ferroptosis through the SMAD7/hepcidin pathway in I/R rat.

Accurate measurement of the minimum distance between bony structures of the humeral head and the acromion or coracoid helps advance a better understanding of the shoulder anatomical features. Our goal was to precisely determine the minimum acromiohumeral distance (AHD), coracohumeral distance (CHD), and glenohumeral distance (GHD) in a sample of the Chinese population as an in vivo anatomical analysis. We retrospectively included 146 patients who underwent supine computed tomography (CT) examination of the shoulder joint. The minimum AHD, CHD, and GHD values were quantitatively measured using three-dimensional (3D) CT reconstruction techniques. The correlation between minimum AHD, CHD, and GHD value and age with different sexes was evaluated using Pearson Correlation Coefficient. The mean value of minimum AHD in males was greater than that in females (male 7.62 ± 0.98 mm versus female 7.27 ± 0.86 mm, p = 0.046). The CHD among different sexes differed significantly (male 10.75 ± 2.40 mm versus female 8.76 ± 1.38 mm, p < 0.001). However, we found no statistical differences in GHD with different sexes (male 2.00 ± 0.31 mm versus female 1.96 ± 0.36 mm, p > 0.05). In terms of age correlation, a negative curve correlation existed between age and AHD among the different sexes (male R2 = 0.124, p = 0.030, female R2 = 0.112, p = 0.005). A negative linear correlation was found in CHD among the different sexes (male R2 = 0.164, p < 0.001, female R2 = 0.122, p = 0.005). There were no differences between age and minimum GHD in both sexes. The 3D CT reconstruction model can accurately measure the minimum AHD, CHD, and GHD value in vivo and is worthy of further investigation for standard clinical anatomical assessment. Aging may correlate with AHD and CHD narrowing for both sexes.

Background: No effective therapeutic agents for calcific aortic valve disease (CAVD) are available currently. Dietary supplementation has been proposed as a novel treatment modality for various diseases. As a flavanone, hesperetin is widely abundant in citrus fruits and has been proven to exert protective effects in multiple diseases. However, the role of hesperetin in CAVD remains unclear. Methods: Human aortic valve interstitial cells (VICs) were isolated from aortic valve leaflets. A mouse model of aortic valve stenosis was constructed by direct wire injury (DWI). Immunoblotting, immunofluorescence staining, and flow cytometry were used to investigate the roles of sirtuin 7 (Sirt7) and nuclear factor erythroid 2-related factor 2 (Nrf2) in hesperetin-mediated protective effects in VICs. Results: Hesperetin supplementation protected the mice from wire-injury-induced aortic valve stenosis; in vitro, hesperetin inhibited the lipopolysaccharide (LPS)-induced activation of NF-κB inflammatory cytokine secretion and osteogenic factors expression, reduced ROS production and apoptosis, and abrogated LPS-mediated injury to the mitochondrial membrane potential and the decline in the antioxidant levels in VICs. These benefits of hesperetin may have been obtained by activating Nrf2–ARE signaling, which corrected the dysfunctional mitochondria. Furthermore, we found that hesperetin could directly bind to Sirt7 and that the silencing of Sirt7 decreased the effects of hesperetin in VICs and potently abolished the ability of hesperetin to increase Nrf2 transcriptional activation. Conclusions: Our work demonstrates that hesperetin plays protective roles in the aortic valve through the Sirt7–Nrf2–ARE axis; thus, hesperetin might be a potential dietary supplement that could prevent the development of CAVD.

ObjectiveOur goal was to investigate the correlation between papillary thyroid carcinoma (PTC) characteristics on ultrasonography and metastases of lymph nodes posterior to the right recurrent laryngeal nerve (LN-prRLN). There is still no good method for clinicians to judge whether a patient needs LN-prRLN resection before surgery, and we also wanted to establish a new scoring system to determine whether patients with papillary thyroid carcinoma require LN-prRLN resection before surgery.Patients and MethodsThere were 482 patients with right or bilateral PTC who underwent thyroid gland resection from December 2015 to December 2017 recruited as study subjects. The relationship between the PTC characteristics on ultrasonography and the metastases of LN-prRLN was analyzed by univariate and logistic regression analyses. Based on the risk factors identified in univariate and logistic regression analysis, a nomogram-based LN-prRLN prediction model was established.ResultLN-prRLN were removed from all patients, of which 79 had LN-prRLN metastasis, with a metastasis rate of 16.39%. Multivariate logistic regression analysis revealed that LN-prRLN metastasis was closely related to sex, age, blood supply, larger tumors (> 1 cm) and capsular invasion. A risk prediction model has been established and fully verified. The calibration curve used to evaluate the nomogram shows that the consistency index was 0.75 ± 0.065.ConclusionPreoperative clinical data, such as sex, age, abundant blood supply, larger tumor (> 1 cm) and capsular invasion, are positively correlated with LN-prRLN metastasis. Our scoring system can help surgeons non-invasively determine which patients should undergo LN-prRLN resection before surgery. We recommend that LN-prRLN resection should be performed when the score is above 103.1.

In view of the problems of poor reliability of information transmission and low fusion degree of heterogeneous data for coal mine safety monitoring system, a multi-protocol fusion gateway for underground coal mine was designed. The gateway can be connected to sensors with different protocol types at the same time and transmit multi-source data to remote server in the form of Ethernet protocol data for information fusion. In the gateway, physical layer receives data information of the terminal through different protocols (RS232, RS485, CAN, ZigBee) transmission medium and interface. Protocol layer de-archive different data formats to obtain original data of the end user. System layer completes access of multiple types of devices through task management and communication scheduling. Application layer designs user function and calls the kernel space network layer protocol for data encapsulation to obtain the data in the Ethernet protocol format, the data can be transmitted to the upper node without processing, so as to complete conversion and transmission of multi-type protocol data to the Ethernet protocol data of the gateway. The test results show that the gateway can realize transparent transmission of RS232, RS485, CAN, ZigBee protocol data to Ethernet protocol data, and the transmission delay is concentrated within 15 ms, the packet loss of transmission data is extremely low, so the gateway has a strong communication reliability.

The accuracy of the acoustic signal prediction model for wood–plastic composites milling has an important influence on the condition monitoring of the cutting process and the improvement of the machining environment. To establish a high-precision prediction model of sound signal in the high-speed milling of wood–plastic composites, high-speed milling experiments on self-developed wood–plastic composites were carried out with cemented carbide tools. A mathematical model of the relationship of the four milling parameters, including axial cutting depth, radial cutting depth, feed rate and cutting speed, and the sound signal of wood–plastic composites milling, was established by using the full-factor test method. The experimental data obtained by the orthogonal test method were used as the test samples in the mathematical model. Test results show that the prediction accuracy of the mathematical model of the sound signal in the milling of wood–plastic composites exceeds 95.4%. To further improve the prediction accuracy of the sound signal in the milling of wood–plastic composites, a prediction model was established using back propagation (BP) neural network. Then, the particle swarm optimization (PSO) algorithm was used to optimize the BP neural network, obtaining the PSO–BP neural network prediction model. The test results show that the prediction accuracy of the PSO–BP prediction model for the sound signal in the high-speed milling of wood–plastic composites exceeds 97.5%. The PSO–BP model has a better global approximation ability and higher prediction accuracy than the BP model. The research results can provide a reference basis for sound signal prediction in the high-speed milling of wood–plastic composites.

High-speed cutting technology has become a development trend in the material processing industry. However, high-intensity noise generated during high-speed cutting exerts a potential effect on the processing efficiency, processing accuracy, and product quality of the workpiece; it may even cause hidden safety hazards. To conduct an in-depth study of noise in high-speed cutting machining, this work reviews noise sources, noise collection and numerical recognition, noise control, and condition monitoring based on acoustic signals. First, this article introduces noise sources, noise signal acquisition equipment, and analysis software. It is pointed out that how to accurately classify and recognize the target signal in the complex high-speed machining environment is one of the focuses of scholars’ research. Then, it points out that a computer achieves high accuracy and practicability in signal analysis, processing, and result display. Second, in the aspect of noise signal processing, the characteristics of noise signals are analyzed. It is pointed out that accurately analyzing the characteristics of different noise source signals and adopting appropriate methods for identification and processing are the necessary conditions for effectively controlling and reducing the noise in the process of high-speed cutting. The advantages and applicable fields of artificial intelligence algorithms in processing mixed noise source signals with different frequency characteristics are compared, providing ideas for studying the mechanism of noise generation and the identification of noise sources. Third, in terms of noise control, a detailed overview is provided from the aspects of the treatment of the noise source that contributes the most to the overall noise, the improvement of the tool structure, the optimization of cutting parameters, and the analysis of contact factors between the tool and the workpiece. It provides an effective way for noise control in the process of high-speed cutting. In addition, the application of acoustic signals to condition monitoring is also thoroughly analyzed. The practical application value of condition monitoring based on acoustic signals in high-speed machining is highlighted. Finally, this paper summarizes the positive significance of noise research in high-speed machining and identifies key problems and possible research methods that require further study in the future.

Novel daidzein napsylates (DD4 and DD5) were synthesized by microwave irradiation, according to structural modification of daidzein (DAI) using the principle of pharmacokinetic transformation. The pharmacological properties of DD4 and DD5 were evaluated via high performance liquid chromatography (HPLC) and calculated based on the drug design software ChemAxon 16.1.18. The cell uptake changes of DD4 and DD5 were investigated to analyse the structure–property relationship. The metabolisms of DD4 and DD5 were analysed by HPLC-mass spectrometry in human aortic vascular smooth muscle cells (HAVSMCs) and their possible metabolic pathways were inferred in vivo. The results showed that the solubility of DD4 and DD5 was increased by 2.79 × 105 and 2.16 × 105 times compared to that of DAI, separately, in ethyl acetate. The maximum absorption rates of DD4 and DD5 were enhanced by 4.3–4.5 times relative to DAI. Preliminary studies on metabolites of DD4 and DD5 in HAVSMCs showed that DD4 and DD5 were hydrolysed into DAI under the action of intracellular hydrolase in two ways, ester hydrolysis or ether hydrolysis. Then, DAI was combined with glucuronic acid to form daidzein monoglucuronate under the action of uridine diphosphate (UDP)-glucuronidase. Meanwhile, it was also found that metabolite M5 of DD5 could undergo glucuronidation under the action of UDP-glucuronosyltransferase and competitive sulphation under the action of sulphotransferase to produce its sulfate conjugate M7. Analysis of structure–property relationships indicated that the absorption and utilization of drugs is closely relative to the physical properties and could be improved by adjusting the liposolubility. The pharmaceutical properties were optimized comprehensively after DAI was modified by naphthalene sulphonate esterification. This indicates that this kind of derivatives may have relatively good absorption and transport characteristics and biological activities in vivo. The research on biological activities of the new derivatives (DD4 and DD5) is ongoing in our laboratory.

For problems that mine personnel location system based on traditional received signal strength indication (RSSI) fingerprint location algorithm had heavy workload in fingerprint database collection during off-line stage and was easily influenced by underground environment, and the location system based on pedestrian dead reckoning (PDR) algorithm had accumulative error, a mine personnel fusion location system was proposed which was based on an improved RSSI fingerprint location algorithm and PDR algorithm. The system uses intelligent terminals consisting of GS1011 controller and MPU9150 inertial sensor to send the data of inertial sensor, RSSI and timestamp to location server on the ground through underground WiFi network. The location server fuses location information of RSSI fingerprinting location algorithm and PDR algorithm by use of extended Kalman filter, so as to realize underground personnel location. The test result shows that the average location error of the system is 1.79 m and smaller than the one of RSSI fingerprint location algorithm or PDR algorithm, and location accuracy satisfies underground personnel location requirement.

Widely varied compounds, including certain plasticizers, hypolipidemic drugs (e.g., ciprofibrate, fenofibrate, WY-14643, and clofibrate), agrochemicals, and environmental pollutants, are peroxisome proliferators (PPs). Appropriate dose of PPs causes a moderate increase in the number and size of peroxisomes and the expression of genes encoding peroxisomal lipid-metabolizing enzymes. However, high-dose PPs cause varied harmful effects. Chronic administration of PPs to mice and rats results in hepatomegaly and ultimately carcinogenesis. Nuclear receptor protein peroxisome proliferator-activated receptor-α (Pparα) was shown to be required for this process. However, biological adaptations to minimize this risk are poorly understood. In this study, we found that miR-181a2 expression was induced by the Pparα agonist WY-14643. Moreover, exogenous expression of miR-181a-5p dramatically alleviated the cell toxicity caused by overactivation of Pparα. Further studies showed that miR-181a-5p directly targeted the Pparα 3′ untranslated region and depressed the Pparα protein level. This study identified a feedback loop between miR-181a-5p and Pparα, which allows biological systems to approach a balance when Pparα is overactivated.

To improve the activity of the isoflavone daidzein (1) and to explore its structure-activity relationship, new daidzein sulfonic acid ester derivatives (2–4) were synthesized. All three derivatives showed significantly enhanced anti-inflammatory activities by 100–10,000-fold over that of the parent daidzein in TNF-α-stimulated Caco-2 cells. Daidzein derivatives 2 and 3 showed the same anti-inflammatory effect at 1 μM and derivative 4 had at 0.01 μM as 100 μM daidzein. Compound 3 also showed significant anti-oxidative effect at 100 μM in H2O2-treated caco-2 cells. Treatment with daidzein and its derivatives significantly inhibited TNF-α-induced phosphorylation of JNK. Results suggest that sulfonic acid esterification at 4′- and/or 7- position of daidzein can significantly improve its physicochemical and pharmaceutical properties, consequently the cellular uptake or absorption and ultimately the biological activities.

User identification is very helpful for building a better profile of a user. Some works have been devoted to this issue. However, the existing works with a good performance are mainly based on the rich online data and do not consider the cost of online data acquisition. In this paper, we aim to address this issue with a lower cost of data acquisition. A machine learning-based solution is proposed solely based on the user's display names. It consists of three key steps: we first analyze the users' unique naming patterns that lead to information redundancies across sites; second, we construct features that exploit information redundancies; afterward, we employ machine learning method for user identification. The experiment shows that the proposed solution can provide excellent performance with F1 score reaching 96.24%, 92.49%, and 90.68% on three real different data sets, respectively. This paper shows the possibility of user identification with a lower cost of data acquisition.

Multi-attribute group decision-making (MAGDM) is widely applied to various areas for solving real-life problems, including technology selection, credit assessment, strategic planning evaluation, supplier selection, etc. To describe the complex and imprecise cognition, it is more convenient to provide the decision-making information in linguistic terms rather than concrete numerical values. Thus, several linguistic models, such as the fuzzy linguistic approach (FLA), hesitant fuzzy linguistic term sets (HFLTSs), hesitant intuitionistic fuzzy linguistic term sets (HIFLTSs), and probabilistic linguistic term sets (PLTS) have been proposed successively. Due to the flexibility and comprehensiveness of PLTS, it has aroused growing concern. However, it also has a big limitation of requiring the membership degree to be 1 by default, and it does not consider the degree of non-membership and hesitancy of a linguistic variable. Therefore, the probabilistic hesitant intuitionistic fuzzy linguistic term sets (PHIFLTSs) have been presented to extend the PLTS by combining the membership and non-membership in symmetry to depict the evaluation of the experts. To overcome the existing shortcomings and enrich the methodology framework of PHIFLTSs, some novel operational laws are defined to extend the applicability and methodology of the PHIFLTSs in MAGDM. Furthermore, the distance and correlation measures for the PHIFLTSs are improved to make up the shortage of the current distance measures. In addition, the unbalanced linguistic terms are taken into account to represent the cognitive complex information of experts. At last, a MAGDM model based on the multiplicative multi-objective optimization by ratio analysis (MULTIMOORA) approach with the use of the developed novel operational laws and correlation measures is presented, which results in more accuracy and effectiveness. A real-word application example is presented to demonstrate the working of the proposed methodology. Moreover, a thorough comparison is done with related existing works in order to show the validity of this methodology.

The antioxidant and anti-inflammatory activities of the novel pyranoanthocyanins and other polyphenols from staghorn sumac (Rhus hirta L.) were examined. The results not only confirmed the previously found antioxidant activity by chemical based assays, but more importantly strong anti-inflammatory activity for the first time in cell-based assays. These unique compounds significantly inhibited the production of pro-inflammatory cytokine IL-8 production stimulated by H2O2 and TNF-α. It was also found that these activities of the crude extract were primarily from the individual polyphenols, particularly the unique pyranoanthocyanins and 7-methoxyanthocyanins. Pyranoanthocyanin 1, in particular, showed a markedly strong anti-inflammatory activity at 1 M, followed by pyranoanthocyanin 2 and a 7-methoxyanthocyanin 5. Further mechanism investigation showed that the anti-inflammatory activity was likely mediated through inhibition of NF-κB activation via phosphorylation of IκBα. Results from the present study warrant further investigation into the in vivo anti-inflammatory effect and health benefits using animal models or human subjects.

How to improve the accuracy of population spatialization by using downscaling technology has always been a difficult issue in academic research. The population spatialization model constructed from the global or local perspective alone has its own limitations that cannot capture the local and global characteristics of the population distribution. Based on the counties of Chongqing municipality in 2010, this paper uses the two steps of “removing-rough” rasterizationof partitioned multivariate statistical regression and the “getting-accuracy” of super-resolution convolutional neural network to construct a coupling model of population spatialization to complete global and local Feature learning and compare and analyze with other four schemes. The results show that the mean square error and root mean square error of the coupled model of partitioned multivariate statistical regression and super-resolution convolutional neural network are the lowest, especially in densely populated areas. Studies have shown that although super-resolution convolutional neural network has a good ability to downscale learning, it still does not reflect the heterogeneity of population spatial patterns well, and the coupling of multilevel global feature learning models and super-resolution convolutional neural network models can make up for this to a certain extent.

Triple negative breast cancer (TNBC) is one of the most aggressive breast cancers without effective targeted therapies. Numerous studies have implied that KLF5 plays an important roles in TNBC. How is KLF5 regulated by microRNAs has not been well studied. Here, we demonstrated that miR-217 down-regulates the expression of KLF5 and KLF5's downstream target gene FGF-BP and Cyclin D1 in TNBC cell lines HCC1806 and HCC1937. Consequently, miR-217 suppresses TNBC cell growth, migration, and invasion. MiR-217 suppresses TNBC, at least partially, through down-regulating the KLF5 expression. These results suggest that the miR-217-KLF5 axis might serve as a potential target for treatment of TNBC.

This study screened microRNAs (miRNAs) that are abnormally expressed in papillary thyroid carcinoma (PTC) tissues to identify PTC and nodular goiter and the degree of PTC malignancy. A total of 51 thyroid tumor tissue specimens paired with adjacent normal thyroid tissues were obtained from the Department of Surgical Oncology of Hangzhou First People’s Hospital from June-December 2011. miRNA expression profiles were examined by microarrays and validated by quantitative real-time PCR (qRT-PCR). Expression levels of the miRNAs were analyzed to assess if they were associated with selected clinicopathological features. Eleven miRNAs were significantly differentially expressed between nodular goiter and PTC and between highly invasive and low invasive PTC. miR-199b-5p and miR-30a-3p were significantly differentially expressed among the three groups. miR-30a-3p, miR-122-5p, miR-136-5p, miR-146b-5p and miR-199b-5p were selected for further study by qRT-PCR and miR-146b-5p, miR-199b-5p and miR-30a-3p were different between the PTC and nodular goiter groups. miR-199b-5p was over-expressed in PTC patients with extrathyroidal invasion and cervical lymph node metastasis. In conclusion miR-146b-5p, miR-30a-3p, and miR-199b-5p may serve as biomarkers for the diagnosis of PTC and miR-199b-5p is associated with PTC invasiveness.

This research discusses the Internet service provider (ISP) bandwidth allocation and pricing problems for a duopoly bandwidth market with two competitive ISPs. According to the contracts between Internet subscribers and ISPs, Internet subscribers can enjoy their services up to their contracted bandwidth limits. However, in reality, many subscribers may experience the facts that their on-line requests are denied or their connection speeds are far below their contracted speed limits. One of the reasons is that ISPs accept too many subscribers as their subscribers. To avoid this problem, ISPs can set limits for their subscribers to enhance their service qualities. This paper develops constrained nonlinear programming to deal with this problem for two competitive ISPs. The condition for reaching the equilibrium between the two competitive firms is derived. The market equilibrium price and bandwidth resource allocations are derived as closed form solutions.
