2. Incident Angle Dependent Formation of Ion Tracks in Quartz Crystal with C60+ Ions: Big Ions in Small Channels

Quartz (SiO2) crystals possess intrinsic columnar pores perpendicular to (0001) surfaces, consisting of three- and six-membered ring (3MR and 6MR) structures of Si and O atoms. The diameters of the larger pores, i.e., 6 MRs, are ~0.49 nm, while the diameters of fullerene (C60) ions are 0.7 nm, i.e., larger than either type of the pores. Transmission electron microscopy observation evidenced approximately two times longer ion tracks in the channeling condition, i.e., 0° incidence to (0001) surface, than an off-channeling condition, i.e., 7° incidence in this case, under 6 MeV C60 ion injection. The track length at the 0° incidence decreases more steeply than that at the 7° incidence with decreasing the energy from 6 MeV to 1 MeV. Finally, the track lengths at the 0° and 7° incidences become comparable, i.e., the channeling-like effect disappears at 1 MeV irradiation. This study experimentally indicates that the channeling-like effect of C60 ions is induced in quartz crystals, while the sizes of the channels are smaller than the C60 ions.

3. Proton-Cluster-Beam Lethality and Mutagenicity in Bacillus subtilis Spores

The unique energy transfer characteristics of swift cluster ions have attracted the attention of many researchers working on the analysis or processing of material surfaces, but the effects on living organisms remain unclear. We irradiated B. subtilis spores with monomer and cluster proton beams and examined their lethality; the 2 MeV H2+ shows a clearly lower lethality than 340 keV H+, even though both have a comparable linear energy transfer. The 2 MeV H2+ dissociates into a pair of 1 MeV H+ by losing the bonding electrons at the target surface. The estimated internuclear distance and the radial dose distribution suggest that the spread of deposited total energy over two areas separated by just several nanometers greatly diminishes beam lethality and that the energy density in the very center of the trajectory, possibly within a 1 nm radius, has a great impact on lethality. We also performed a whole genome resequencing of the surviving colonies to compare the molecular nature of mutations but failed to find a clear difference in overall characteristics. Our results suggest that cluster beams may be a useful tool for understanding biological effects of high linear energy transfer radiation.

4. Novel Approaches for Intensifying Negative C60 Ion Beams Using Conventional Ion Sources Installed on a Tandem Accelerator

We developed novel methods for producing negative C60 ion beams at the accelerator facility Takasaki Ion Accelerators for Advanced Radiation Application (TIARA) to increase the current intensity of swift C60 ion beams accelerated to the MeV energy region using a tandem accelerator. We produced negative C60 ion beams with an intensity of 1.3 µA, which is several tens of thousands of times greater than the intensity of beams produced using conventional methods based on the Cs sputtering process. These beams were obtained by temporarily adding an ionization function based on electron attachment to an existing ion source that is widely used in tandem accelerators. The high-intensity swift C60 ion beams can be made available relatively easily to institutes that have tandem accelerators and ion sources of the type used at TIARA because there is no need to change existing ion sources or install new ones.

5. Experimental Study on the Biological Effect of Cluster Ion Beams in Bacillus subtilis Spores

Cluster ion beams have unique features in energy deposition, but their biological effects are yet to be examined. In this study, we employed bacterial spores as a model organism, established an irradiation method, and examined the lethal effect of 2 MeV C, 4 MeV C2, and 6 MeV C3 ion beams. The lethal effect per particle (per number of molecular ions) was not significantly different between cluster and monomer ion beams. The relative biological effectiveness and inactivation cross section as a function of linear energy transfer (LET) suggested that the single atoms of 2 MeV C deposited enough energy to kill the spores, and, therefore, there was no significant difference between the cluster and monomer ion beams in the cell killing effect under this experimental condition. We also considered the behavior of the atoms of cluster ions in the spores after the dissociation of cluster ions into monomer ions by losing bonding electrons through inelastic collisions with atoms on the surface. To the best of our knowledge, this is the first report to provide a basis for examining the biological effect of cluster ions.

量子科学技術研究開発機構(QST)高崎量子応用研究所のイオン照射施設(TIARA)には4台の加速器が設置されており、主にバイオ技術や材料開発の研究分野へ様々なイオン種のビームを幅広いエネルギー範囲で提供している。AVFサイクロトロン(K110)、3MVタンデム加速器、3MVシングルエンド加速器、400kVイオン注入装置の2020年度運転時間は,それぞれ1879.8h,1629.0h,1637.8h,1555.1hであり,コロナ禍による施設の利用停止措置のため例年よりも短い運転となった。年間を通じて大きな故障はなく,コロナ禍による実験キャンセルを除けば,計画した照射実験はすべて実施した。主な保守・整備としては,サイクロトロンの共振器へ電力を供給するRFアンプの前段増幅部分について,800W真空管アンプから1kWトランジスタアンプへの変更を行った。これまで30年近く使用してきた真空管が製造中止になり,入手不能となっためである。主な技術開発として,タンデム加速器においてC60フラーレン専用イオン源の整備やフラーレン・マイクロビーム形成実験を実施した。本発表では,上記に加え,その他加速器の保守・整備及び技術開発,施設の利用状況について報告する。

Cluster ion beams show unique irradiation effects that cannot be achieved by monomer ion beams. This is largely attributed to the incidence of multiple atoms that are in close proximity, although their biological effects are largely unknown. We have developed an experimental method in order to examine the cluster ion-specific biological effects using bacterial spores as a model organism. Here, we considered the lethality and energy deposition profile of proton cluster beams in the spores.

9. Morphological changes of nanostructures on silicon induced by C60-ion irradiation

We study morphological changes on the Si surface induced by the C60 ion beam under various irradiation conditions. The fluence and incident energy of the C60 ion beam was 1 × 1016–1 × 1019 ions/m2 and 50 keV–9 MeV, respectively. The beam’s incident angle to the surface normal was 0° or 60°. As a result of the changes in these variables, three types of nanostructures were observed: concave and convex, stringlike, and ripple structures. Structure shapes or sizes varied with changes in the parameters. Almost all cases showed concave and convex structures at an incident angle of 0°. At a 60° incident angle, stringlike or ripple structures were formed. Stringlike structures were formed at higher incident energy, while ripple structures were formed at lower incident energy. We found that both stringlike and ripple structures can form from sputtering and thermal effects. The formation of string structure requires a thicker amorphous layer and the proper balance of sputtering and annealing.

ポリメチルメタクリレート(PMMA)標的に0.12-0.54MeVのC60イオンを照射した際の二次負イオン放出特性を分析した結果を報告する。C60イオンパルスビームと飛行時間測定二次イオン質量分析法にイベント毎の測定モードを組み合わせることにより、C60イオン1個によって放出される二次負イオン数の確率分布及び特定の二次負イオン種について相対収量のエネルギー依存性を測定した。これらの分析により明らかになった二次負イオンの放出特性は、サブMeV級C60イオンによって標的中に形成されるイオントラックの構造(直径、長さ)によって説明できることがわかった。

12. TIARA静電加速器の現状

量子科学技術研究開発機構(QST)高崎量子応用研究所のイオン照射施設(TIARA)には3 MVタンデム加速器(9SDH-2、NEC)、3 MVシングルエンド加速器(NC3000B、日新ハイボルテージ)、400 kVイオン注入装置(NH40SR、日新電機)の3台の静電加速器が稼働している。令和2年度の年間計画に対して、3台ともトラブル等なく100 %の稼働率を達成した。 タンデム加速器では、新しく増設したフラーレン専用イオン源を実験に供した。イオン源で生成されたフラーレン負イオンは新たに設計した静電デフレクターで偏向され、既存のタンデム加速器入射ラインに輸送される。従来の磁場による偏向と比較し静電場は時間変動が小さいため、ビーム軌道の揺らぎが改善された。また、より高エネルギーのフラーレン負イオンでも輸送可能となり、末端のファラデーカップまでの透過率も向上された。さらにタンデム加速器では、高速フラーレンイオンのマイクロビーム化を目指すため静電四重極レンズを備えたマイクロビームラインを設計・設置し、フラーレンイオンのマイクロビーム形成実験を行った。静電四重極レンズによって収束された高速フラーレンイオンビームのビームサイズは、電顕用メッシュからの2次イオン像で評価された。 イオン注入装置では新ビームとしてフタロシアニンを生成した。フタロシアニンは窒素を多く含んだ構造であり、ダイヤモンド中の窒素原子と空孔のペアで構成された量子ビット(NVセンター)の多量子ビット化を目指した研究に用いられる。 講演では上記の詳細に加え、TIARA静電加速器の運転状況、各加速器での整備状況等を報告する。, 第33回タンデム加速器及びその周辺技術の研究会

15. MeV級C60イオンを用いたアミノ酸薄膜の透過型SIMSイメージング

- Abstract
我々のグループで開発した、MeV級C60イオンを一次イオンとする透過型二次イオン質量分析(SIMS)イメージングは、サブµmの横方向分解能で生体試料を含む有機材料の分子イメージングが可能であるが、試料が導電性の薄膜で、かつ良い二次電子放出体である必要がある。そこで、本手法に適した試料の構成を探るために、一次イオン(9 MeV C60++)が入射する薄膜試料の上流側に形成する帯電防止膜の構成が異なる試料について透過型SIMSイメージング実験を行った。アルミニウムの単層膜を堆積した試料は一次イオンを照射したときに帯電が起こり、二次電子収率も低かったため、効率が悪くぼやけたイメージしか得られなかった。それに対して、最表面に極薄の金を蒸着したアルミニウムとの二層膜を堆積した試料では帯電が抑制され、明るくシャープなイメージが得られた。

16. MeV C60イオン照射による媒質に埋め込まれた金属ナノ粒子の楕円変形とSi中でのイオントラック形成

- Abstract
1-6 MeVのC60イオン照射によってシリコン結晶中に形成されたイオントラックについて報告する。先行研究の結果より、17 MeVより低いエネルギーのC60イオン照射ではシリコン結晶中にイオントラックは形成されないと信じられてきた。しかし、我々は6 MeV以下のC60イオン照射によって直径~10 nmの大きなイオントラックが形成されることを観測した。これは、40 MeVのC60イオンによって形成されるものに匹敵し、20 MeVの場合よりも大きい。

A vapor-deposited films of PDI-Cl4 were irradiated with 1.5 ~ 6.0 MeV Aln (n = 1, 2, 3, 4) cluster ions at the fluence of 1.0 × 1011 cm–2, and the irradiated film was treated with a vacuum condition (~10–2 Pa) at high temperature. The non-irradiated area was sublimated by this treatment, yielding vertically aligned polymerized nanowires whose number density corresponds to the irradiation fluence. The radii of nanowires became thicker by increasing number of cluster ion resulting in clear and rigid standing nanowires.

- Abstract
A swift C60-ion microbeam apparatus for molecular imaging analysis of organic polymeric materials has been developed. A highly-intense negative C60 ion source has been installed on the 3-MV Tandem accelerator of TIARA, QST/Takasaki for this purpose as well as an electrostatic deflector. The microbeam with a size of one μm is successfully formed. Molecular images of a test sample are demonstrated. In addition, nonlinear effect on Au sputtering yields by Cn-ion (n =60 and 70) bombardment is discussed.

MeVエネルギーC_60イオンと物質との相互作用を調べる一環として、C60イオンの直径(0.7nm)より小さなチャンネル径(0.49nm)を持つSiO2結晶にチャネリング条件と非チャネリング条件で、C60イオンを照射し、形成されるイオントラック長を比較した。その結果、チャネリング条件の方が長いトラック長となることが観測され、チャネリング径より入射イオンの直径が大きいにもかかわらず、チャネリング様効果が現れることがわかった。

A radiochromic film, Gafchromic HD-V2, is applied to a possible method of measuring a two-dimensional (2D) spatial profile of MeV cluster ion beams. The coloration responses of the HD-V2 film to MeV carbon and gold cluster ion beams are experimentally investigated since some cluster effect may appear. The degree of the film coloration is quantified as a change in optical density (OD) by reading the films with an image scanner for high-resolution measurement of the 2D beam profile. The OD response of HD-V2 is characterized as a function of the ion and atom fluence for comparison. The dependences of the OD response on the cluster size, kinetic energy, and ion species are discussed. It is found that the sensitivity of the OD change is reduced when the cluster size is large. The beam profile of MeV cluster ion beams delivered from the tandem accelerator in TIARA is characterized from the measurement result using HD-V2 films. The present results show that the use of the Gafchromic HD-V2 film is suitable for the detail beam profile measurement of MeV cluster ions, especially C 60 ions, whose available intensity is rather low in comparison with that of monatomic ion beams.

クラスターイオンは、複数の原子がnm距離に近接した状態で物質に入射することから、材料表面の改質や分析では単原子イオンに比べて特異なエネルギー付与を示すことが知られている。しかしながら、生物試料における研究は実施されていなかった。我々は、枯草菌の胞子を生物試料のモデルとし、クラスターイオンの生物効果の特徴について明らかにすることを目的とした研究を行っている。原子あたり2MeVの炭素イオンの致死効果を評価した結果、LETが非常に高いために1原子のヒットで致死するため、致死効果に関するクラスター化の効果は確認されないことがわかった。そこで、炭素よりも低いLETを持つプロトンのクラスターイオンの致死効果を評価した。2 MeV H2は1MeV Hに比べて原子あたりの致死効果が低く、負のクラスター効果を示す可能性が示唆された。これは、2つのプロトンが近接して通過するために、同じ遺伝子に影響する確率が高いことによると解釈できる。340 keV Hは2MeV H2と粒子あたりで同じLETを持つが、致死効果は340 keV Hの方が明らかに高かった。これは、イオントラック中心付近でのエネルギー密度が致死効果に大きく影響することによると考えられる。

電子的阻止能が大きな高エネルギーイオンを標的に照射したときにその飛跡に沿ってダメージによるイオントラックが形成されることが知られているが、炭素原子60個からなるC_60クラスターイオンの場合、これまで17MeV以下ではSi中にトラックが形成されないと考えられていた。今回、照射実験により1MeVのC_60照射でもSi中にトラックが形成されることを確認した。これはこれまでの電子的阻止能のエネルギー付与のみによる形成メカニズムでは説明することができず、新たに核的阻止能を加えたシナジー効果を示唆するものである。

タンデム加速器によるMeVエネルギーのC60イオンビームの高強度化を目指し、高崎量子応用研究所放射線高度利用施設(TIARA)において革新的なC60負イオン生成法を開発した。本生成法により、従来のセシウムスパッター方式による生成法に比べ数万倍、1.3µAのC60負イオンビームの生成に成功した。この高強度ビームは、タンデム加速器において広く用いられる既存のイオン源に電子付着に基づくイオン化機構を一時的に付加することで得られる。そのため、TIARAと同様なタンデム加速器とイオン源を有する加速器施設において、イオン源の増設や置き換えをすることなく、比較的容易にMeV級の高強度C60イオンビームを利用することができる。

The anesthetic effect of a combination of medetomidine, midazolam and butorphanol (Me-Mi-Bu) was evaluated in healthy cynomolgus monkeys. The Me-Mi-Bu combination was intramuscularly administered as follows: Dose 1, Me 0.015 mg/kg-Mi 0.1 mg/kg-Bu 0.15 mg/kg; Dose 2, Me 0.02 mg/kg-Mi 0.15 mg/kg-Bu 0.2 mg/kg; and Dose 3, Me 0.04 mg/kg-Mi 0.3 mg/kg-Bu 0.4 mg/kg. The combination rapidly induced immobilization, and lateral recumbency was reached within 15 min. The duration of anesthesia for each dose administered was follows: Dose 1, 47 ± 27 min; Dose 2, 113 ± 31 min; and Dose 3, 190 ± 24 min. The anesthetic effect of the combination was abolished by the α2-adrenoceptor antagonist atipamezole. No marked changes in the levels of hematologic or serum biochemical parameters were noted in cynomolgus monkeys administered the combination plus atipamezole. Taken together, these results suggest that the Me-Mi-Bu combination exhibits reversible anesthetic effect and may be useful for studies involving cynomolgus monkeys.

SiO2中に形成された金属ナノ粒子が高エネルギー重イオン(200MeV, Xe)を照射するとビーム方向に延びることが知られており、異方性光学素子等への応用が期待されている。今回、高崎研で開発したタンデム加速器用負C60イオン源からの大電流C60イオンを用いて、電子的阻止能が上記Xeイオンと同程度になる1MeV-6MeVのC60イオン照射でも同様のナノ粒子の変形を起こすことを見出した。本成果は台数の少ない高エネルギー加速器に限定されていた大きな電子的阻止能を利用した材料改質研究を電圧が1MVから3MV程度の多く普及している静電加速器で実施可能なことを意味し、その普及に大きく貢献するものである。

- Abstract
Quantum information processing requires quantum registers based on coherently interacting quantum bits. The dipolar couplings between nitrogen vacancy (NV) centres with nanometre separation makes them a potential platform for room-temperature quantum registers. The fabrication of quantum registers that consist of NV centre arrays has not advanced beyond NV pairs for several years. Further scaling up of coupled NV centres by using nitrogen implantation through nanoholes has been hampered because the shortening of the separation distance is limited by the nanohole size and ion straggling. Here, we demonstrate the implantation of C5N4Hn from an adenine ion source to achieve further scaling. Because the C5N4Hn ion may be regarded as an ideal point source, the separation distance is solely determined by straggling. We successfully demonstrate the fabrication of strongly coupled triple NV centres. Our method may be extended to fabricate small quantum registers that can perform quantum information processing at room temperature.

Cluster ion beams have unique features in energy deposition, but their biological effects are yet to be examined. In this study, we employed bacterial spores as a model organism, established an irradiation method, and examined the lethal effect of 2 MeV C, 4 MeV C2, and 6 MeV C3 ion beams. The lethal effect per particle (per number of molecular ions) was not significantly different between cluster and monomer ion beams. The relative biological effectiveness and inactivation cross section as a function of linear energy transfer (LET) suggested that the single atoms of 2 MeV C deposited enough energy to kill the spores, and, therefore, there was no significant difference between the cluster and monomer ion beams in the cell killing effect under this experimental condition. We also considered the behavior of the atoms of cluster ions in the spores after the dissociation of cluster ions into monomer ions by losing bonding electrons through inelastic collisions with atoms on the surface. To the best of our knowledge, this is the first report to provide a basis for examining the biological effect of cluster ions.

Here, to determine the effects of transport stress on blood parameters in dogs, we investigated the changes in hematologic and serum chemical parameters in healthy beagle dogs transported from Beijing, China, to Osaka, Japan, to obtain the background data. Only the activity of serum alkaline phosphatase increased clearly upon arrival, a change attributed to transport stress, but the activity gradually reduced afterward. No marked changes in levels of other blood parameters were noted. Our findings here suggest that alkaline phosphatase is a useful tool for studying transport stress.

The tobacco industry has promoted certain cigarette products with claims that their use may be less harmful to the smoker as they purportedly deliver lower amounts of toxic chemicals compared to conventional cigarettes. This study was designed to compare the relative antigen presenting cellular toxicity of Eclipse, a presumed reduced exposure product (PREP) cigarette, when compared with the reference research 3R4F cigarettes (Kentucky University). Utilizing a murine macrophage cell line, murine bone marrow derived dendritic cells (DCs) and human monocyte-derived DCs incubated with extracts generated from Eclipse and Kentucky reference 3R4F cigarettes, we determined the relative toxic effects of the different cigarette smoke extracts on cellular viability, oxidative stress, T-helper-1 (Th-1) polarizing cytokine production and general gene expression. Eclipse and 3R4F cigarette smoke extracts induced equivalent oxidatively-mediated cellular heme oxygenase-1 (HO-1) protein levels in macrophages and DCs. Cellular viability determination demonstrated greater induction of cell death by apoptosis and necrosis by Eclipse extracts in DCs. The production of the key Th-1 polarizing cytokine interleukin-12 (IL-12) by activated DCs or macrophages was suppressed to an equivalent or greater extent by Eclipse extracts. Microarray studies performed on bone marrow derived murine DCs incubated with Eclispe or 3R4F cigarette extracts showed identical genotoxic profiles. These studies imply that presumed reduced harm Eclipse cigarettes induce equivalent or greater antigen presenting cell dysfunction relative to 3R4F cigarettes and illustrate the importance of independent validation and testing of similar products claimed to be associated with reduced toxicity relative to other cigarettes.

The interaction which stabilizes the intermediate state of the protein folding and/or unfolding is important for understanding the structure formation mechanism of proteins. The partitioning of a hydrophobic fluorescence probe, pyrene, into the core of a 'molten globule' structure of bovine carbonic anhydrase B was measured, revealing a partition coefficient of about 10(4). The result leads to the conclusion that the compact structure of the molten-globule state is formed by the hydrophobic interaction, as detergent micelles are formed by the same interaction.
