To examine the effect of the arginine-vasopressin fragment, [pGlu4,Cyt6]AVP(4–9) (AVP4–9), on group II metabotropic glutamate receptor (mGluR2/3) agonist and antagonist induced impairment of passive avoidance (PA) task performance, AVP4–9 or phorbol 12-myristate 13-acetate (PMA) was administered in the presence of mGluR2/3-related drugs that induced the impairment of the step-through-type PA task performance. The PA task performance was evaluated in terms of the latency (the time that elapsed prior to entry into the dark compartment) at 24 h after the electrical stimulation. The subcutaneous injection of AVP4–9 at 1 μg/kg had the greatest facilitative effect on the performance, and the facilitative effect of AVP4–9 was inhibited by NPC-15437, a specific protein kinase C (PKC) inhibitor. The injection of AVP4–9 ameliorated PA task performance impairment induced by DCG-IV, an mGluR2/3 agonist. Intracisternal injection of PMA, a PKC activator, also ameliorated the DCG-IV-induced impairment. High doses of AVP4–9 exacerbated the PA task performance impairment induced by LY341495 (an mGluR2/3 antagonist), and PMA injection (1 μg) also exacerbated the impairment induced by the antagonist. These results suggest that an increase in the activity of the PKC-signaling pathway may not always facilitate PA task performance; therefore, AVP4–9 can either enhance or inhibit memory performance in mice. In order to determine the mechanism of action of a new AVP(4-9) analog, NC-1900, on memory processes, memory retention and retrieval tests were conducted in a step-through passive avoidance (PA) task in mice. The administration of NC-1900 facilitated memory retention and retrieval in the PA task through vasopressin1A (V1A) receptors but not V2 receptors. The effect of NC-1900 on memory retention test performance appeared to be due to activation of the protein kinase C (PKC) signaling pathway via V1A receptors; however, the modulation of PKC was not essential for the facilitative effect of the new peptide in the retrieval test. The facilitation of memory retrieval by NC-1900 may also be mediated by other non-PKC-dependent signaling pathways, such as the phospholipase C-inositol trisphosphate pathway.

In the present study, we investigated the facilitative effect of NC-1900, a new arginine vasopressin (AVP(1-9)) fragment analog, on memory performance in eight-arm radial maze or passive avoidance (PA) tasks in nonamnesic and amnesic (PA tasks only) mice. In the radial maze, all injections (subcutaneous) were given daily 60 min before each trail. NC-1900 (1 ng/kg)-treated animals showed enhancement of performance, and AVP(4-9) (1 microg/kg), an AVP(1-9) fragment, had similar effects, although the effective dose was 1000-fold higher. In the PA task, all drugs were administrated subcutaneously 60 min before the acquisition trial (Acq.), and the amnesic mice were exposed to CO(2) just after the Acq. NC-1900 (1 ng/kg) enhanced the memory performance of nonamnesic mice and ameliorated CO(2)-induced amnesia. AVP(4-9) (1 microg/kg) had a similar effect, although only at higher doses, while AVP(1-9) (0.1-1 microg/kg) had no effect. The facilitating effect of NC-1900 on nonamnesic mice was inhibited by coinjection [Pmp(1)-Tyr(Me)(2)]-AVP (Pmp,Tyr-AVP; 1 microg/kg), a V(1A) antagonist, but not by OPC-31260, a vasopressin(2) (V(2)) antagonist. The effect of NC-1900 on CO(2)-induced amnesia was also decreased by coinjection of Pmp,Tyr-AVP or [deamino-Pen(1), Me-Tyr(2)]-AVP (10 microg/kg), both of which are V(1) antagonists. These results suggested that NC-1900 has a more potent effect on facilitation of memory via the V(1A) receptor than AVP(4-9) in non- and CO(2)-amnesic conditions.

Inhibition of sodium-potassium adenosine 5'-triphosphatase ((Na(+), K(+))-ATPase) activity causes edema and cell death in the central nervous system, and impairment of learning and memory. Several sex steroid hormones have a protective effect against neuronal cell damage and the hypofunction of learning and memory. To examine the possible roles and mechanism of action of steroid hormones against amnesia induced by ouabain, an inhibitor of (Na(+), K(+))-ATPase, gonadectomized male mice were administrated ouabain (0.1 microg per mouse) intracisternally (i.cist.), and the learning and memory abilities of the mice were assessed by a step-through passive avoidance task. Subcutaneous (s.c.) administration of 17beta-estradiol (betaE2; 10 microg kg(-1)) or testosterone (TES; 1 mg kg(-1)) improved the memory impairment induced by ouabain, while administration of dihydrotestosterone (1 mg kg(-1)) or corticosterone (COR) (1 mg kg(-1)) did not. Treatment with the estradiol receptor antagonists, tamoxifen (TAM) (10 mg kg(-1); s.c. or 0.1 microg; i.cist.) and 4-hydroxytamoxifen (10 mg kg(-1); s.c.), or the androgen receptor antagonist, cyproterone (10 mg kg(-1); s.c. or 1 microg; i.cist.), did not influence the protective effect of betaE2 or TES on ouabain-induced amnesia. Moreover, we studied the effects of several free radical scavengers-17alpha-estradiol (10 microg kg(-1); s.c.), alpha-tocopherol (VE: 200 mg kg(-1); per os (p.o.), ascorbic acid (VC: 200 mg kg(-1); p.o.), or VE+VC (200 mg kg(-1) each; p.o.)-on ouabain-induced amnesia, and compared those effects with that of betaE2. The administration of free radical scavengers had no significant effect on memory impairment. These results indicate that betaE2 and TES ameliorate the amnesia induced by inhibition of (Na(+), K(+))-ATPase activity, and that the protective effect of betaE2 is caused by a non-genomic, rather than a genomic effect or a radical scavenging action. Additionally, the ameliorative effect of TES does not appear to involve free radical scavenging, but its aromatization to estrogen could contribute to the non-genomic action of betaE2.

There is increasing evidence that estrogen and calcium ion are involved in learning and memory. In the present study, to examine the effect of estrogen deficiency and low-calcium diet on learning and memory, middle-aged female Wistar rats (50 weeks old) were fed either a low-calcium (0.02% Ca) or a normal-calcium (1.25% Ca) diet throughout the experiment. Rats were ovariectomized (OVX) or sham-operated (Sham). These animals were divided into four groups: 1) Sham group with normal-calcium diet [Sham-normal Ca group], 2) OVX group with normal-calcium diet [OVX-low Ca group], 3) Sham group with low-calcium diet [Sham-low Ca group], 4) OVX group with low-calcium diet [OVX-low Ca group]. Seventy-seven days after the OVX or Sham operation, the learning and memory abilities in the female rats were examined by using a radial maze task according to the method of Olton and Samuelson (regular trials) and using a delay-interposed task following regular trials. During regular trials and delay-interposed tasks, the OVX-low Ca group was inferior to all the other groups in accuracy of choice behavior. Both Sham-normal Ca and Sham-low Ca groups showed more accurate choices than the OVX-low Ca group, but were less accurate than the Sham-normal Ca group. In addition, there was no significant difference in locomotor activity between any of the groups. These results suggest that OVX or low-calcium diet may impair learning and memory, and that the combination of these factors impaired more markedly when the rats were tested in the eight-arm radial maze. These results may also imply the possibility that a woman in menopause or post-menopause suffers impairment of learning and/or memory when intakes low-calcium diet.

We examined the effect of the acetylcholinesterase (ACHE) inhibitor, donepezil hydrocloride (DONP), on group II metabotropic glutamate (mGlu) receptor agonist- or antagonist-induced amnesia in the step-through passive avoidance task in male mice. DCG-IV, a group II mGlu receptor agonist, at dose of 50 ng and LY341495, a group II mGlu receptor antagonist, at dose of 300 ng, significantly attenuated the latency on the step-through task. The subcutaneous injection of DONP at dose of 1 mg/kg 1 hour before passive avoidance performance ameliorated the amnesia induced by DCG-IV and LY341495, whereas donepezil alone did not affect task latency. The results suggest that activation of group II mGlu receptors and disinhibition of the cAMP/PKA signaling pathway (caused by group II mGlu receptor antagonist) have a negative action on step-through passive avoidance memory performance, and that group II mGlu receptors and ACh interact to modulate learning and memory function.

Fullerene is known to effectively produce mainly singlet oxygen by absorbing light energy, and therefore may be useful as a virucidal photosensitizer. This study was designed to investigate the virucidal activity of a water-soluble fullerene derivative, which is conjugated with methoxy polyethylene glycol amine to enhance its water solubility. Vesicular stomatitis virus (VSV) was inactivated upon illumination with this water-soluble fullerene, in a concentration- or dose-dependent manner. The titer of VSV was reduced by5log 10 in the presence of 10 mg/ml (400 microM) fullerene derivative with 120 J/cm2 white light irradiation. VSV inactivation was inhibited by oxygen removal or by the addition of sodium azide, a known singlet oxygen scavenger. The substitution of H2O by D2O, which is known to prolong the lifetime of singlet oxygen, promoted the virucidal activity. These results indicate that singlet oxygen may play a major role in VSV photoinactivation by the water-soluble fullerene derivative. The concentration needed for virus inactivation is higher than that of other sensitizers such as methylene blue.

In the CNS, reactive oxygen species (ROS) have been implicated in a wide range of degenerative processes including amyotrophic lateral sclerosis, ischemia-reperfusion injury, Alzheimer disease, Parkinson disease and aging. However, the exact mechanism is unknown, and there is little information on possible roles of ROS in cell injury and the process on recovery of astrocytes, the most abundant glial cells in the brain. We examined hydrogen peroxide (H2O2)-induced DNA fragmentation and thymidine incorporation into cultured astrocytes as an indicator of the process of recovery from astrocytic DNA injury. Astrocytes were isolated from cerebral cortices of 0-day-old rats and treated with 1 mM dibutyryl cyclic AMP for 4 days. H2O2 of 100 microM stimulated thymidine incorporation into astrocytes. Caffeine, ryanodine, cyclic ADP-ribose (endogenous ryanodine receptor agonist) and beta-NAD+ (precursor of cyclic ADP-ribose) suppressed partially the stimulatory effect of H2O2. Ruthenium red (ryanodine receptor antagonist) facilitated further the stimulatory effect of H2O2. The facilitated effect of ruthenium red on H2O2-induced thymidine incorporation was suppressed by caffeine, ryanodine, cyclic ADP-ribose and beta-NAD+. H2O2-induced DNA fragmentation and astrocytic death were suppressed by ruthenium red. These findings suggest that the process of recovery from astrocytic DNA injury by H2O2 may be regulated by Ca2+ efflux from ryanodine-sensitive intracellular Ca2+ stores.

To decrease the risk of virus infection by blood transfusion, we investigated a virus photoinactivation method which employs methylene blue (MB) in combination with red light irradiation on human plasma. Irradiation equipment containing light emitting diodes (LED) has been developed. The LED irradiation spectrum overlaps well the MB absorption spectrum. Vesicular stomatitis virus (VSV) 4.1 log10 was inactivated with 1uM MB at 2.4J/cm2 irradiation. Similarly, 4.1 log10 infectivity of human immunodeficiency virus type 1 (HIV-1) was reduced to below the detection limit (0.5 log10) with 1uM MB at 1.4J/cm2. Under conditions in which HIV-1 was completely killed, the activities of coagulation factor VIII and fibrinogen were maintained at more than 90% of pretreatment level. Antithrombin III activity was not affected by phototreatment. Prothrombin time and activated partial thromboplastin time were slightly prolonged. These data suggest that MB phototreatment is an effective method of inactivating viruses in plasma and, hence, of ensuring the supply of safer plasma products.

The alloreactive cytotoxic T lymphocytes (CTL) were generated by coculturing peripheral blood mononuclear cells (PBMC) with allogeneic Sa cells (an Epstein-Barr virus [EBV]-transformed B-cell line). The CTL did not proliferate in response to UV-B-irradiated Sa cells unless exogenous interleukin-2 (IL-2) was present, although they could kill the UV-B-irradiated Sa cells. The results indicate that UV-B-irradiated Sa cells do not provide sufficient signals for the proliferation of the CTL while they can be recognized by CTL and induce high-affinity IL-2 receptor (IL-2R) expression on them. The alloreactive CTL could be rendered anergic by previous exposure to UV-B-irradiated Sa cells. The alloreactive CTL previously stimulated with UV-B-irradiated Sa cells failed to proliferate in response to nontreated Sa cells. Proliferation of the anergic CTL could not be restored by Sa cells and exogenous IL-2 but by the combination of phorbol 12-myristate 13-acetate (PMA) and calcium ionophore (A23187). The anergic CTL showed a considerably low cytotoxic activity against Sa target cells. The expression of TCR on the anergic CTL was downregulated while expression of high-affinity IL- 2R was upregulated. Their CD28 and CD8 expression were unchanged. In addition, the proliferative response and cytotoxicity of the anergic CTL to Sa cells could be restored after the cells had been rested for 7 days to allow reexpression of TCR. These results suggest that downregulation of T-cell receptor (TCR) and impairment in the post-IL- 2/IL-2R signaling pathway are relevant to the clonal anergy induced in the alloreactive CTL by stimulation of UV-B-irradiated Sa cells.

Inhibitory effects of UV-B irradiation on allosensitization were analysed in vitro. In mixed lymphocyte reaction (MLR), UV-B irradiated cells could neither stimulate allogeneic responder cells nor induce cytokine production (IL-1β and IL-2) in the supernatants. ConA induced proliferation and cytokine production were also inhibited by UV-B irradiation while mitomycin C (MMC)-treated cells produced the cytokines by ConA stimulation.Inactivation of lymphocytes by UV-B irradiation cannot be attributed soley to low viability or reduced expression of cell surface molecules in UV-B irradiated cells. Viability of UV-B irradiated cells were comparable to MMC or 60Co treated cells. UV-B irradiation induced a time-dependent reduction of HLA-class I, -DR and CD54 (ICAM-1) expression on the cells. However, a similar reduction of the cell surface antigens were observed in MMC-treated cells.By UV-B irradiation, capping of HLA-class I, -DR and CD54 (ICAM-1) was markedly inhibited, while it remained unaffected by MMC or 60Co treatment. UV-B irradiation may induce certain damages to cell membrane or subcellular cytoskeleton that are detected as capping inhibition and impair the rearrangement of cell surface molecules that is required for cell adhesion.

The cAMP/protein kinase A signaling pathway is negatively modulated by group II metabotropic glutamate receptors (mGluRs), and the cross-talk that occurs between these receptors may modulate learning and memory. To examine the relationship among cAMP/PKA-signaling pathway activity, group II mGluRs, and learning and memory, mice were trained to perform a step-through-type passive avoidance task, and 10 min before each avoidance trial the following drugs were injected intracisternally (i.cist.): vehicle (0.05% dimethylsulfoxide); a specific group II mGluR agonist, DCG-IV (1-50 ng/mouse); a specific group II mGluR antagonist, LY341495 (10-300 ng); a selective inhibitor of cAMP-specific phosphodiesterase, rolipram (100-1000 ng); an activator of adenylyl cyclase, forskolin (25-250 ng); a specific inhibitor of PKA, H-89 (150 or 300 ng) or; an activator of protein kinase C, phorbol 12-myristate 13-acetate (PMA 200 ng). DCG-IV (25 and 50 ng) or LY341495 (150 and 300 ng) reduced the latency in the avoidance task. The reduction of latency by DCG-IV was not observed in mice coinjected with DCG-IV (50 ng) together with rolipram (500 ng) or forskolin (25 ng). Conversely, coinjection of LY341495 with 100 or 1000 ng rolipram, or with 25 or 250 ng forskolin tended to potentiate the LY341495-induced shortening of latency. In addition, the reduction of latency by DCG-IV (50 ng) was not observed in mice coinjected with DCG-IV and PMA together. However, the reduction of latency by LY341495 (300 ng) was potentiated when the drug was coadministered with PMA. These results suggest that changes in the cAMP/PKA-signaling pathway, mediated by group II mGluRs, influence memory in the passive avoidance task, and that both the excessive activation and deactivation of this pathway may induce the impairment of learning and memory.

There is increasing evidence that ovarian steroids and calcium ions are involved in learning and memory. To examine the effect of ovarian steroids on learning and memory under a low-calcium condition, middle-aged female rats were fed either a low-calcium (0.02% Ca) or a normal-calcium (1.25% Ca) diet. All rats were ovariectomized (OVX), and these animals were divided into eight groups: 1) an OVX group with a normal-calcium diet (OVX-normal-Ca group), 2) an OVX group with 17beta-estradiol treatment and a normal-calcium diet (E2 group), 3) an OVX with progesterone treatment and a normal-calcium diet (P4 group), 4) an OVX with 17beta-estradiol and progesterone treatments and a normal-calcium diet (E2 + P4 group), 5) an OVX group with a low-calcium diet (OVX-low-Ca group), 6) an OVX group with 17beta-estradiol treatment and a low-calcium diet (LE2 group), 7) an OVX group with progesterone treatment and a low-calcium diet (LP4 group), and 8) an OVX group with 17beta-estradiol and progesterone treatments and a low-calcium diet (LE2 + LP4). Seventy-seven days after the OVX operation, the learning and memory abilities of the rats were examined by using an eight-arm radial maze task. E2 and E2 + P4 groups learned in fewer trials, and performed better in the radial maze and the working memory task than the other groups under the normal-calcium condition. Rats in the LE2 group learned in fewer trials, and performed better in the maze and working memory task than the other low-calcium groups, but in combination with progesterone under the low-calcium condition (LE2 + LP4 group), the facilitative effect of estradiol in all the tasks was inhibited. Treatment with progesterone alone did not inhibit the learning and memory task performance. These results suggest the possibility that treatment with estradiol under low-calcium conditions cannot improve impaired learning and memory when progesterone is applied simultaneously, and that the intake of adequate calcium may be necessary and effective for patients with learning and memory hypofunction receiving hormone replacement therapy.

The effects of a novel vasopressin (AVP) fragment analog NC-1900 (pGlu-Asn-Ser-Pro-Arg-Gly-NH2 acetate) were studied on the performance of memory retention and retrieval in mice. NC-1900 of one time application of 1 hr after the acquired trial (electric shock) extended the latent period of passive avoidance task 21 days after the acquired trial. Though the extended response was also recognized with AVP4-9, the potency was approx. 1/1000 of NC-1900. The potentiation wasn't recognized with vasopressin. NC-1900 showed a significantly high correct answer after 21 days after the last trial in the search task. While, V1 antagonist Pmp1-Tyr (Me)2-AVP shortened the latent period of passive avoidance task. On the other hand, NC-1900 extended the reaction latency 21 days after the acquired trial by the application 1 hr before the retention trial. Though this improvement of memory retrieval was recognized with vasopressin and AVP4-9, the potency was 1/100-1/1000. NC-1900 improved the retrieval 24 h after the CO2 exposure. V1 antagonists Pmp1-Tyr-Me2-AVP or Deamino-Pen1, O-Me-Tyr2-AVP, and PMA had no effects on the retrieval 21 days after the acquired trial. These results suggest that NC-1900 may have the memory retention and retrieval potentiating action, and that phospholipase C-protein kinase C system may be involved in the former action, and the latter action not be involved.

The effects of a novel vasopressin fragment analog NC-1900 (pGlu-Asn-Ser-Pro-Arg-Gly-NH2 acetate) were studied on learning and/or memory impairment in passive avoidance task and on cell damage of cultured cerebro-cortical neurocytes induced by glutamic acid. A small dose of NC-1900 (1 ng/kg, s.c.) ameliorated impairments of learning and/or memory induced by intracisternal injection of 467.6 micrograms of 10 microliters glutamic acid. NC-1900 also ameliorated the impairments induced by intracisternal NMDA, AMPA-antagonist CNQX and by metabotropic receptor (mGluR1) agonist 3,5-dihydroxyphenylglycine but not by kainate agonist domoic acid nor MK-801 in mice. NC-1900 (100 pM, 1nM) ameliorated the cell damage of cultured rat cerebro-cortical neurocytes induced by 100 and 1000 microM of glutamic acid. These results suggest that NC-1900 may serve as a remedies in various patients with certain brain disorders induced by excess glutamic acid.

We investigated the photoinactivation of virus infectivity by hypocrellin A and its mechanism. The titers of vesicular stomatitis virus (VSV) and human immunodeficiency virus type 1 (HIV-1), both of which are enveloped viruses, were reduced upon illumination with hypocrellin A in a concentration-dependent manner, whereas canine parvovirus, a nonenveloped virus, was not killed. The removal of oxygen or addition of sodium azide or beta-carotene both inhibited VSV inactivation. Mannitol and superoxide dismutase had no effect on VSV inactivation. These results indicate that singlet oxygen was involved in the process of VSV inactivation. Of the three major VSV membrane proteins, peripheral membrane protein M was most damaged by the hypocrellin A phototreatment.

The effects of ultraviolet (UV) irradiation on cell-mediated cytolysis were examined in order to clarify the inhibitory mechanisms of allosensitization by UV irradiation. UV-B-irradiated target cells (Sa; an Epstein-Barr virus-transformed B cell line) exhibited more resistance against alloreactive cytotoxic T lymphocytes (CTL) than mitomycin C (MMC)-treated target cells. In the conjugate formation assay, UV-B-irradiated target cells showed a considerably lower binding to alloreactive CTL than MMC-treated target cells. UV-B irradiation induced a reduction of HLA-class I, -DR, CD54 (ICAM-1) and CD58 (LFA-3) expression on target cells. However, it does not seem to contribute to the inhibition of cell adhesion induced by UV-B irradiation because a similar reduction of cell surface antigens was observed in MMC-treated target cells. Number of cells capped with anti-HLA-class I, -DR, CD54 or CD58 monoclonal antibody were markedly reduced by UV-B irradiation compared to that by MMC treatment. These findings suggest the possibility that the inhibition of cell adhesion between UV-B-irradiated Sa target cells and alloreactive CTL is due to the impaired mobility of cell surface antigens which will affect the early process of cell-mediated cytolysis.

The study of chemical forms and physiological mobility of elements found in plants is always essential to establishing the roles of elements in plant life. Such information in combination with the results of growth experiments will lead to more. reliable conclusions on the physiological functions of elements.

As seen in the previous paper1) the use of HF-etching method has added much information to the mode of silicon deposition in rice tissues. The results obtained cover a wide range from root to husk. However, the importance of silicon deposition in epidermis in relation to the resistance of plants to diseases or insect pests makes it desirable to continue further studies of the mode of silicon deposition in outer region of epidermis. The study of microstructure of epidermis in connection with silicon deposition will provide valuable information to the mechanism of resistance rice plant to blast disease, because penetration of the rice leaf by piricularia oryzae is ordinarily through the cuticle rather than via the stomata. It will be of importance also to the study of a possible role of silicon in water economy of rice plant as already suggested by the present authors.
