Abstract

Introduction Vitamin D plays a critical role in bone and mineral metabolism and also controls other physiologic and pathophysiologic processes, including skeletal muscle function, reproductive health, cancer risk and progression, [...], The renal actions of parathyroid hormone (PTH) promote 1,25-vitamin D generation; however, the signaling mechanisms that control PTH-dependent vitamin D activation remain unknown. Here, we demonstrated that salt-inducible kinases (SIKs) orchestrated renal 1,25-vitamin D production downstream of PTH signaling. PTH inhibited SIK cellular activity by cAMP-dependent PKA phosphorylation. Whole-tissue and single-cell transcriptomics demonstrated that both PTH and pharmacologic SIK inhibitors regulated a vitamin D gene module in the proximal tubule. SIK inhibitors increased 1,25-vitamin D production and renal Cyp27b1 mRNA expression in mice and in human embryonic stem cell-derived kidney organoids. Global- and kidney-specific Sik2/Sik3 mutant mice showed Cyp27b1 upregulation, elevated serum 1,25- vitamin D, and PTH-independent hypercalcemia. The SIK substrate CRTC2 showed PTH and SIK inhibitor-inducible binding to key Cyp27b1 regulatory enhancers in the kidney, which were also required for SIK inhibitors to increase Cyp27b1 in vivo. Finally, in a podocyte injury model of chronic kidney disease-mineral bone disorder (CKD-MBD), SIK inhibitor treatment stimulated renal Cyp27b1 expression and 1,25-vitamin D production. Together, these results demonstrated a PTH/SIK/CRTC signaling axis in the kidney that controls Cyp27b1 expression and 1,25-vitamin D synthesis. These findings indicate that SIK inhibitors might be helpful for stimulation of 1,25-vitamin D production in CKD-MBD.

7. The question of the beginning and the ending of the so-called history of David's rise : a methodological reflection and its implications

Abstract

The thesis argues that we can maintain that the so-called History of David’s Rise (HDR) existed independently before the deuteronomistic work, by identifying its beginning in I Samuel 16. 14 and ending in II Samuel 5. 3. Additionally, the thesis proposes that the source was first composed during Hezekiah’s reign with a view to persuading the northerners to embrace Hezekiah’s one Israel policy, and then went through two major redactions – one in the late exilic period and the other in the post-exilic period. These later redactions were prompted not only by the political situations of the time, but also by the literary milieu. In other words, a growing interest in narratives and the emergence of the ‘Jewish novelistic impulse’ in the Neo-Babylonian and Persian period triggered the creation of more extensive narratives about Saul and David. These historical-critical arguments are preceded in the thesis by a methodological argument that a traditio-historical issue is inevitably related to a literary understanding of the larger whole. The background for this two-foci research is the wild disagreement on the issue, and the confusion around the methodology that has been aggravated by an unnecessary tension between different approaches. The thesis therefore discusses the methodological issues as carefully as possible, so that it might be transparent what actually happens when one does biblical criticism. This gives the thesis the features of a case study, but the thesis also hopes to present a satisfactory and attractive view on a particular traditio-historical issue in its own right. The study hopes to be an experiment of self-reflective biblical criticism that is serious but open. Since the thesis has two different but essentially related theses, the conclusion is established in two stages – methodological and historical. Chapter 1 shows that a literary understanding of the whole is foundational to traditio-historical discussions, and Chapter 2 demonstrates that literary understanding is always open to revision, and so are historical answers, as the latter are inevitably related to the former. Chapter 3 asks what is the most appropriate understanding of the whole HDR at this point, and the answer provides the last two chapters with the foundation by which various evidences can be measured. Chapter 4 revisits the initial question, and provides a provisional answer. And Chapter 5, after discussing the relationship between the materials in the books of Samuel, confirms the conclusion reached in the previous chapter, and elaborates further implications.

La revista Luvina y la Editorial de la Universidad de Guadalajara reúnen estos cien Cuentos de Asia, Europa y América para celebrar el número 100 de Luvina y sus veinticinco años ininterrumpidos de publicación. Son textos representativos de la literatura universal contemporánea que Luvina ha publicado, especialmente en los números dedicados al país invitado de honor de la Feria Internacional del Libro de Guadalajara. Es una muestra formada por distintas literaturas, en el afán de nombrar nuevos y originales mundos, en una dimensión habitada en su totalidad por la lengua. Cada uno de los cuentos permite internarse en la singularidad humana al mismo tiempo que en los entramados de los relatos, cuya belleza encierra vitalidad y resignificación de lo real gracias al encuentro de la creatividad y la imaginación del autor, la obra y el lector.

Abstract

The purpose of this study was to investigate the differences in fashion lifestyles, shopping orientation, the perception of time constraints and shopping cost among two consumer groups(day and night-time internet shopping mall users). 1. Fashion lifestyles depend on fashion leadership, the importance of fashion, fashion imitation and fashion indifference. Compared to night-time users, day-time users showed relatively low interest in fashion while night-time users showed a high degree of fashion persuit. 2. Shopping orientation was summarized as six factors; Enjoyment shopping orientation, Fashion orientation, Brand orientation, Convenience/time saving, Price conscious orientation, Practical orientation. Night-time users were also noted to have a high interest in fashion and tendencies to pursue brand names and trends. 3. 48. Selective addition of CXCR3+CCR4-CD4+Th1 cells enhances generation of cytotoxic T cells by dendritic cells in vitro

Abstract

Increasing importance is being given to the stimulation of Th1 response in cancer immunotherapy because its presence can shift the direction of adaptive immune responses toward protective immunity. Based on chemokine receptor expression, CXCR3+CCR4-CD4+T cells as Th1-type cells were investigated its capacity in monocyte-derived dendritic cell (DC) maturation and polarization, and induction of antigen specific cytotoxic T lymphocytes (CTL) in vitro. The levels of IL-4, IL-5 and IL-10 were decreased to the basal level compared with high production of IFN-?, TNF-a, and IL-2 in CXCR3+CCR4-CD4+T cells stimulated with anti-CD3 and anti-CD28 antibodies. Co-incubation of activated CD4+or CXCR3+CCR4-CD4+T cells with DC (CD4+/DC or CXCR3+CD4+/DC, respectively) particularly up-regulated IL-12 and CD80 expression compared with DC matured with TNF-a and LPS (mDC). Although there was no significant difference between the effects of the CXCR3+CCR4-CD4+and CD4+T cells on DC phenotype expression, CXCR3+CD4+/DC in CTL culture were able to expand number of CD8+T cells and increased frequencies of IFN-? secreting cells and overall cytolytic activity against tumor antigen WT-1. These results demonstrated that the selective addition of CXCR3+CCR4-CD4+T cells to CTL cultures could enhance the induction of CTLs by DC in vitro, and implicated on a novel strategy for adoptive T cell therapy.
