AbstractShiitake mushroom (Lentinula edodes) hold high nutritional and medicinal value as they contain an abundance of health-promoting compounds. However, the effect of long-term postharvest storage on the variation in the levels of health-promoting compounds has not been extensively studied. In this study, we investigated the changes in the levels of phenolic compounds, antioxidants, eritadenine, and ergothioneine in shiitake mushrooms stored at three different temperatures (1, 3, and 5 °C) for 4 weeks. Compared to mushrooms stored at lower temperatures, those stored at 5 °C exhibited a higher level of total phenolics in their pileus after 2 weeks of storage; however, storage at 5 °C also increased the deterioration of the fruiting body of these mushrooms. In mushrooms stored at all temperatures, the eritadenine content in the pilei tended to increase up to 2 weeks of storage. In contrast, the ergothioneine content in the pileus decreased during storage, with a significantly lower level detected in mushrooms stored at 5 °C for 4 weeks. Together, these results suggest that the mechanisms underlying the accumulation of phenolics and eritadenine may be related to mushroom deterioration during storage. Our findings indicate that the levels of health-promoting compounds in shiitake mushrooms are influenced by storage temperature, suggesting the potential to control adjustments of specific bioactive compounds by regulating storage conditions.

A cold storage system is useful for maintaining the quality of hardy kiwifruit. However, extended cold storage periods inevitably result in cold stress, leading to lower fruit marketability; the severity of chilling injury depends on fruit types and cultivars. In this study, the impact of cold storage conditions on the physicochemical properties and antioxidant capacity of two phenotypically different hardy kiwifruit cultivars—‘Cheongsan’ (large type) and ‘Daebo’ (small type)—stored at low (L; 3 °C, relative humidity [RH]; 85–90%) and moderate-low (ML; 5 °C, RH; 85–90%) temperatures was determined. Significant differences in fruit firmness and titratable acidity between treatments L and ML were observed in both cultivars during the experimental storage period. Meanwhile, the browning and pitting rates of the ‘Cheongsan’ fruits in treatment L increased for 8 weeks compared with those of the ‘Daebo’ fruits in treatments L and ML; nonetheless, fruit decay was observed in the ‘Daebo’ fruits in treatment ML after 6 weeks. The total chlorophyll, carotenoid, flavonoid, and ascorbic acid concentrations as well as the antioxidant activities of both the cultivars significantly differed between treatments L and ML. After 2 weeks of storage, the ‘Cheongsan’ fruits in treatment L had lower antioxidant activities and ascorbic acid content than those in treatment ML. These results demonstrate that the quality attributes and antioxidant activity of hardy kiwifruit are influenced by the low-temperature storage conditions and the specific kiwifruit cultivars. Our findings suggest that optimal cold storage conditions, specific to each hardy kiwifruit cultivar, promise to maintain fruit quality, including their health-promoting compounds, during long-term storage.

Circulating tumor cells (CTCs) are some of the key culprits that cause cancer metastasis and metastasis-related deaths. These cells exist in a dynamic microenvironment where they experience fluid shear stress (FSS), and the CTCs that survive FSS are considered to be highly metastatic and stem cell-like. Biophysical stresses such as FSS are also known to cause the production of extracellular vesicles (EVs) that can facilitate cell–cell communication by carrying biomolecular cargos such as microRNAs. Here, we hypothesized that physiological FSS will impact the yield of EV production, and that these EVs will have biomolecules that transform the recipient cells. The EVs were isolated using direct flow filtration with and without FSS from the MDA-MB-231 cancer cell line, and the expression of key stemness-related genes and microRNAs was characterized. There was a significantly increased yield of EVs under FSS. These EVs also contained significantly increased levels of miR-21, which was previously implicated to promote metastatic progression and chemotherapeutic resistance. When these EVs from FSS were introduced to MCF-7 cancer cells, the recipient cells had a significant increase in their stem-like gene expression and CD44+/CD24− cancer stem cell-like subpopulation. There was also a correlated increased proliferation along with an increased ATP production. Together, these findings indicate that the presence of physiological FSS can directly influence the EVs’ production and their contents, and that the EV-mediated transfer of miR-21 can have an important role in FSS-existing contexts, such as in cancer metastasis.

Cirsium setidens is commonly used as a food ingredient, and it is typically stored and distributed in a dried form to prolong its shelf life. In a previous study, a micro-oil-sprayed thermal air (MOTA) technique was developed, which effectively enhanced the rehydration properties and improved the color characteristics of Cirsium setidens after processing. Here, we investigated the relationship between the color characteristics and taste of MOTA-processed C. setidens and the effect of NaCl pretreatment, prior to processing, on the final quality of dried C. setidens. NaCl pretreatment, whether combined with the MOTA technique or not, showed improved color characteristics, in which MOTA-and NaCl+ MOTA-processed C. setidens manifested equal color characteristics. In contrast, NaCl + MOTA-processed C. setidens resulted in significantly higher values of sourness and saltiness than MOTA-processed C. setidens when the taste of the rehydrated C. setidens was examined using an electronic tongue (Astree II; Alpha MOS, Toulouse, France). Pearson correlation coefficient analysis showed that sourness and saltness were negatively correlated with Hunter a* values and positively correlated with Hunter L* and Hunter b* values, indicating that the color characteristics of dried and rehydrated C. setidens could be indicators of taste. Furthermore, the amounts of total phenol and chlorophyll were better preserved in C. setidens processed by the MOTA technique with NaCl than by the MOTA technique alone. Our data revealed that the color characteristics of dried plants are associated with the taste of processed C. setidens, and that the MOTA technique with NaCl pretreatment is a useful method for preserving health-promoting compounds during processing.

Elevated CO2 (eCO2; 1000 ppm) influences developing rice leaf formation, reducing leaf blade length and width as compared to rice grown under ambient CO2 (aCO2; 400 ppm). Since micro RNAs (miRNAs) are known to play multiple roles in plant development, we hypothesized that miRNAs might be involved in modulating leaf size under eCO2 conditions. To identify miRNAs responding to eCO2, we profiled miRNA levels in developing rice leaves (P4; plastochron number of the fourth-youngest leaf) under eCO2 using small RNA-seq. We detected 18 mature miRNA sequences for which expression levels varied more than two-fold between the eCO2 and aCO2 conditions. Among them, only miR396e and miR396f significantly differed between the two conditions. Additionally, the expression of growth-regulating factors (GRFs), potential target mRNA of miR396s, were repressed under the eCO2 condition. We used an antisense oligonucleotide approach to confirm that single-strand DNA corresponding to the miR396e sequence effectively downregulated GRF expression in developing leaves, reducing the leaf blade length, such as for rice grown under eCO2. These results suggest that the miR396–GRF module is crucially relevant to controlling rice leaf blade length in eCO2 environments.

Following the launch of Rare Isotope Science Project in December 2011, a heavy ion accelerator complex in South Korea, named RAON, has since been designed. It includes a muon facility for muon spin rotation, relaxation, and resonance. The facility will be provided with 600 MeV and 100 kW (one-fourth of the maximum power) proton beam. In this study, the graphite target in RAON was designed to have a rotating disk shape and was cooled by radiative heat transfer. This cool-down process has the following advantages: a low-temperature gradient in the target and the absence of a liquid coolant cooling system. Monte Carlo simulations and ANSYS calculations were performed to optimize the target system in a thermally stable condition when the 100 kW proton beam collided with the target. A comparison between the simulation and experimental data was also included in the design process to obtain reliable results. The final design of the target system will be completed within 2020, and its manufacturing is in progress. The manufactured target system will be installed at the RAON in the Sindong area near Daejeon-city in 2021 to carry out verification experiments.

Protic ruthenium complexes using the dihydroxybipyridine (dhbp) ligand combined with a spectator ligand (N,N = bpy, phen, dop, Bphen) have been studied for their potential activity vs. cancer cells and their photophysical luminescent properties. These complexes vary in the extent of π expansion and the use of proximal (6,6′-dhbp) or distal (4,4′-dhbp) hydroxy groups. Eight complexes are studied herein as the acidic (OH bearing) form, [(N,N)2Ru(n,n′-dhbp)]Cl2, or as the doubly deprotonated (O− bearing) form. Thus, the presence of these two protonation states gives 16 complexes that have been isolated and studied. Complex 7A, [(dop)2Ru(4,4′-dhbp)]Cl2, has been recently synthesized and characterized spectroscopically and by X-ray crystallography. The deprotonated forms of three complexes are also reported herein for the first time. The other complexes studied have been synthesized previously. Three complexes are light-activated and exhibit photocytotoxicity. The log(Do/w) values of the complexes are used herein to correlate photocytotoxicity with improved cellular uptake. For Ru complexes 1–4 bearing the 6,6′-dhbp ligand, photoluminescence studies (all in deaerated acetonitrile) have revealed that steric strain leads to photodissociation which tends to reduce photoluminescent lifetimes and quantum yields in both protonation states. For Ru complexes 5–8 bearing the 4,4′-dhbp ligand, the deprotonated Ru complexes (5B–8B) have low photoluminescent lifetimes and quantum yields due to quenching that is proposed to involve the 3LLCT excited state and charge transfer from the [O2-bpy]2− ligand to the N,N spectator ligand. The protonated OH bearing 4,4′-dhbp Ru complexes (5A–8A) have long luminescence lifetimes which increase with increasing π expansion on the N,N spectator ligand. The Bphen complex, 8A, has the longest lifetime of the series at 3.45 μs and a photoluminescence quantum yield of 18.7%. This Ru complex also exhibits the best photocytotoxicity of the series. A long luminescence lifetime is correlated with greater singlet oxygen quantum yields because the triplet excited state is presumably long-lived enough to interact with 3O2 to yield 1O2.

It is essential to build a practical environment of the training/test site for cyber training and weapon system test evaluation. In a military environment, cyber training sites should be continuously developed according to the characteristics of the military. Weapons with cyber security capabilities should be deployed through cyber security certification. Recently, each military has been building its own cyber range that simulates its battlefield environment. However, since the actual battlefield is an integrated operation environment, the cyber range built does not reflect the integrated battlefield environment that is interconnected. This paper proposes a configuration plan and operation function to construct a multi-cyber range reflecting the characteristics of each military to overcome this situation. In order to test the multi-cyber range, which has scenario authoring and operation functions, and can faithfully reflect reality, the impact of DDoS attacks is tested. It is a key to real-world mission-based test evaluation to ensure interoperability between military systems. As a result of the experiment, it was concluded that if a DDoS attack occurs due to the infiltration of malicious code into the military network, it may have a serious impact on securing message interoperability between systems in the military network. Cyber range construction technology is being developed not only in the military, but also in school education and businesses. The proposed technology can also be applied to the construction of cyber ranges in industries where cyber-physical systems are emphasized. In addition, it is a field that is continuously developing with the development of technology, such as being applied as an experimental site for learning machine learning systems.

Abstract Neuropathic pain is a complex, chronic pain state characterized by hyperalgesia, allodynia, and spontaneous pain. Accumulating evidence has indicated that the microglial Toll-like receptor 4 (TLR4) and autophagy are implicated in neurodegenerative diseases, but their relationship and role in neuropathic pain remain unclear. In this study, we examined TLR4 and its association with autophagic activity using a chronic constriction injury (CCI)-induced neuropathic pain model in wild-type (WT) and TLR4-knockout (KO) mice. The mice were assigned into four groups: WT-Contralateral (Contra), WT-Ipsilateral (Ipsi), TLR4 KO-Contra, and TLR4 KO-Ipsi. Behavioral and mechanical allodynia tests and biochemical analysis of spinal cord tissue were conducted following CCI to the sciatic nerve. Compared with the Contra group, mechanical allodynia in both the WT- and TLR4 KO-Ipsi groups was significantly increased, and a marked decrease of allodynia was observed in the TLR4 KO-Ipsi group. Although glial cells were upregulated in the WT-Ipsi group, no significant change was observed in the TLR4 KO groups. Moreover, protein expression and immunoreactive cell regulation of autophagy (Beclin 1, p62) were significantly increased in the neurons, but not microglia, of WT-Ipsi group compared with the WT-Contra group. The level of PINK1, a marker for mitophagy was increased in the neurons of WT, but not in TLR4 KO mice. Together, these results show that TLR4-mediated p62 autophagic impairment plays an important role in the occurrence and development of neuropathic pain. And what is more, microglial TLR4-mediated microglial activation might be indirectly coupled to neuronal autophage.

Nine percent nickel steel has excellent properties in a cryogenic environment, so it has recently been used as a tank material for most LNG fuel-powered ships. However, 9% nickel steel causes arc deflection due to its tendency of magnetization during manual FCAW welding and the currently used filler metal is 10–25 times more expensive as a base metal compared to other materials, depending on manufacturers. Furthermore, the properties of its filler metal cause limitation in the welding position. To overcome these disadvantages, in this study, the tendency of penetration shape was analyzed through a fiber laser Bead on Plate (BOP) welding for 9% nickel steel with a thickness of 6 mm and a range of welding conditions for 1-pass laser butt welding of 6 mm thick 9% nickel steel with I-Groove were derived. Through this study, basic data capable of deriving optimal conditions for laser butt welding of 9% nickel steel with a thickness of 6 mm were obtained.

Purpose Smoking cessation intervention is one of the key components of successful lung cancer screening program. We investigated the effectiveness and related factors of smoking cessation services provided to the participants in a population-based lung cancer screening trial. Materials and Methods The Korean Lung Cancer Screening Project (K-LUCAS) is a nationwide, multi-center lung cancer screening trial that evaluates the feasibility of implementing population-based lung cancer screening. All 5,144 current smokers who participated in the K-LUCAS received a mandatory smoking cessation counseling. Changes in smoking status were followed up using a telephone survey in 6 months after lung cancer screening participation. The lung cancer screening's impact on smoking cessation is analyzed by variations in the smoking cessation interventions provided in screening units. Results Among 4,136 survey responders, participant's motivation to quit smoking increased by 9.4% on average after lung cancer screening. After 6 months from the initial screening, 24.3% of participants stopped smoking, and 10.6% of participants had not smoked continuously for at least 6 months after screening. Over 80% of quitters stated that participation in lung cancer screening motivated them to quit smoking. Low-cost public smoking cessation program combined with lung cancer screening increased the abstinence rates. The smokers were three times more likely to quit smoking when the smoking cessation counseling was provided simultaneously with low-dose computed tomography screening results than when provided separately. Conclusion A mandatory smoking cessation intervention integrated with screening result counselling by a physician after participation in lung cancer screening could be effective for increasing smoking cessation attempts.
