Background: Acupuncture has been wildly applied for cerebral ischemia treatment in China for thousands of years, while the specific mechanism remains uncertain. Recently, many studies have shown that acupuncture promotes angiogenesis after ischemia occurs. Here, we examined the effect of electro-acupuncture (EA) exosomes on angiogenesis in hypoxia-induced human umbilical vein endothelial cells (HUVECs). Objective: To investigate whether EA exosomal miR-210 promotes angiogenesis in the hypoxiainduced HUVECs via the HIF-1α/VEGF/Notch 1 signal pathway. Methods: The middle cerebral artery occlusion (MCAO) model was established and treated with EA therapy. Then, exosomes were identified and isolated from rats' plasma in the MCAO+EA group by transmission electron microscopy (TEM), surface markers expressions, and PKH26 reagent. MiR- 210 mimic, miR-210 inhibitor, and HIF-1α were transfected. Flow cytometry, CCK-8 assay, and Transwell assay were conducted to assess the migration, apoptosis, and proliferation of each group of cells. Western blot and quantitative PCR were performed to detect the CD34, HIF-1α, VEGF, Notch 1, and miR-210 expression levels in each group. Results: MiR-210 was significantly upregulated in exosomes of the MCAO plasma, and further enhanced by EA therapy. EA-EXOs and miR-210 mimic inhibited cell apoptosis, promoted cell proliferation and cell migration in hypoxia-induced HUVECs. However, the miR-210 inhibitor reversed the proliferation and migration number induced by EA-EXOs. Besides, EA-EXOs and miR- 210 mimic further enhanced those HIF-1α, VEGF, and Notch 1 levels compared to the hypoxia treatment only. Silencing HIF-1α or miR-210 reversed the high expressions of those three angiogenic factors induced by hypoxia and EA-EXO. qPCR showed similar trends with their relative mRNAs. To analyze these associations quantificationally, Spearman's rank correlation coefficient was calculated. As revealed by results, the expression of proteins and mRNA were highly correlative with each other. Conclusions: These results indicated that EA-EXO miR-210 promotes angiogenesis in hypoxia conditions via HIF-1α/VEGF/Notch 1 signal pathway.

Background: Acupuncture has been recommended as an alternative and complementary therapy for preventing and treating cerebral ischemia by the World Health Organization (WHO) for years. However, the mechanisms remain unclear. Accumulating evidence has shown that acupuncture can promote angiogenesis to attenuate brain damage after ischemic stroke. In recent years, exosome- carried microRNAs (miRNAs) activated by acupuncture have proven effective in regulating pathological changes. We, therefore, investigated whether electro-acupuncture (EA) enhanced angiogenesis in cerebral stroke via exosome-carried miR-210. Methods: We extracted and identified the exosomes from the serum of MCAO with EA treatment and injected them into MCAO rats for further observation. Simultaneously, miR-120 siRNA and HIF-1α inhibitor were transfected. Then, we evaluated the volume of infarction, pathological changes, and expression levels of angiogenic related factors of each group of rats by TTC and HE staining, transmission electron microscope (TEM), western blot, and quantitative PCR (qPCR). Results: Compared with the MCAO group, EA-Exosome (EA-EXO) treatment significantly decreased the infarct volume and the pathological damage, but miR-210 siRNA or HIF-1α inhibitor reversed the protective outcomes induced by EA-EXO. Moreover, EA-EXO treatment upregulated miR-210 and increased CD34, HIF-1α, VEGF, Notch1 protein, and mRNA expressions compared to the MCAO group. MiR-210 siRNA or HIF-1α inhibitor treatments both down-regulated those angiogenic related proteins and mRNAs. Conclusion: EA treatment could activate the HIF-1α/VEGF/Notch 1 signal pathway to facilitate angiogenesis after ischemic stroke via exosomal miR-210.

To observe the effect of different intensities of electroacupuncture (EA) preconditioning on car-diac function and polarization state of macrophages in mice with acute myocardial ischemia (AMI), so as to explore its possible mechanism underlying improvement of AMI.A total of 50 male C57BL/6J mice were randomly divided into sham ope-ration, AMI model, and EA pretreatment groups (0.5 mA, 1 mA, 3 mA subgroups), with 10 mice in each group/subgroup. The mice in the EA pretreatment groups were subjected to EA stimulation of bilateral "Neiguan"(PC6) with 0.5, 1.0 and 3 mA respectively and frequency of 2 Hz/15 Hz for 20 min, once a day, for 3 days. The acute myocardial ischemia model was established by ligating the anterior descending branch (ADB) of the left coronary artery, while the sham operation only had a surgical suture trans-passed below the ADB but without ligation. The myocardial infarction area was measured after TTC staining, and the cardiac function ［left ventricular ejection fraction (EF), short-axis contraction rate (FS)］ was detected by using echocardiography. The M1 macrophages were labeled with CD11b+F480+CD206Compared with the sham operation group, the model group had a significant increase in the infarction area (EA preconditioning with electrical current intensities of 0.5 mA, 1 mA and 3 mA can effectively reduce myocardial infarction size, improve cardiac function in mice with AMI, which may be related with its effects in reducing the number of cardiac macrophages and down-regulating the expression of myocardial IL-1β, TNF-α and TLR4 proteins. The therapeutic effect of 1 mA is better than that of 0.5 and 3 mA.

Acupuncture has been widely used for treating diseases since the ancient days in China, but the mechanism by which acupuncture exerts such powerful roles is unclear. Epigenetics, including DNA methylation, histone modification, and post-transcriptional regulation of miRNAs, is the study of heritable changes in gene expression that do not include DNA sequence alterations. Epigenetics has become a new strategy for the basic and clinical research of acupuncture in the last decade. Some investigators have been trying to illustrate the mechanism of acupuncture from an epigenetics perspective, which has shed new lights on the mechanisms and applications of acupuncture. Moreover, the introduction of epigenetics into the regulatory mechanism in acupuncture treatment has provided more objective and scientific support for acupuncture theories and brought new opportunities for the improvement of acupuncture studies. In this paper, we reviewed the literatures that has demonstrated that acupuncture could directly or indirectly affect epigenetics, in order to highlight the progress of acupuncture studies correlated to epigenetic regulations. We do have to disclose that the current evidence in this review is not enough to cover all the complex interactions between multiple epigenetic modifications and their regulations. However, the up-to-date results can help us to better understand acupuncture's clinical applications and laboratory research.

Objective: To systematically evaluate the clinical efficacy and analgesic effect of acupuncture and moxibustion in the treatment of herpes zoster by comparing western medicine.Methods: Randomized controlled trials on acupuncture and western medicine for herpes zoster were searched from CNKI, Wanfang Database, VIP, CMB, PubMed, Embase and Cochrane library databases, and the retrieval time was from January 2010 to July 2020.RevMan5.3 software was used for Meta analysis. Results: A total of 30 randomized controlled studies were included, involving a total of 4292 patients. Subgroup analysis was conducted according to different intervention measures in the experimental group. The results showed that: (1) The total effective rate of the subgroup analysis showed that the combined effect of acupuncture treatment alone RR=1.10, 95% CI[1.05,1.15], P < 0.0001.Results: RR=1.16, 95%CI[0.86,1.57], P=0.33. It suggests that the therapeutic effect of acupuncture and moxibustion on herpes zoster is superior to that of western medicine, but acupuncture and western medicine treatment cannot improve the clinical efficacy of western medicine. However, due to the unstable meta-analysis of acupuncture and western medicine treatment in the subgroup, the results need to be treated with caution and further clinical verification is still needed.(2) VAS score improvement analysis showed that the combined effect of acupuncture alone RR=2.20, 95%CI[1.53,2.87], P < 0.00001.RR=1.12, 95%CI[0.39,1.85], P=0.002, suggesting that acupuncture is significantly better than western medicine in improving the pain of shingles, and western medicine combined with acupuncture can improve the analgesic effect.(3) The subgroup analysis of sequelae incidence showed that the combined effect amount of acupuncture alone on sequelae incidence at 30 days was RR=0.34, 95%CI[0.25,0.48], P < 0.00001, and the combined effect amount of acupuncture combined with western medicine was RR=0.18, 95%CI[0.05, 0.69], P=0.01.Incidence of sequelae at 60 days RR=0.38, 95%CI [0.23, 0.62], P =0.0001;The incidence of post-herpetic neuralgia treated with acupuncture alone at 90 days RR=0.39, 95%CI [0.20, 0.74], P =0.004, suggesting that the incidence of post-herpetic neuralgia treated with acupuncture is significantly lower than that treated with western medicine, and western medicine combined with acupuncture can reduce the incidence of post-herpetic neuralgia.(4) The time analysis of the onset of pain relief showed that the combined effect of acupuncture alone RR=-2.32, 95%CI[-2.48,-2.17], P < 0.00001.RR=-2.39, 95%CI[-2.70,-2.09], P < 0.00001, suggesting that the onset time of acupuncture and moxibustion was shorter than that of western medicine, and western medicine combined with acupuncture and moxibustion could accelerate the onset time of analgesia.(5) The time analysis of pain relief showed that the combined effect of acupuncture alone RR=-10.80, 95%CI[-12.49,-9.01], P < 0.00001.RR=-5.98, 95%CI[- 11.55,-0.41], P=0.04, suggesting that acupuncture treatment of herpes zoster can significantly reduce the pain time, combined with western medicine can shorten the pain time. Conclusion: Acupuncture and moxibustion treatment of herpes zoster in the total effective rate, pain relief and the incidence of post-neurosis is significantly better than western medicine;Acupuncture and moxibustion combined with western medicine can improve the analgesic effect and reduce the incidence of post-neurosis. In view of the low overall quality of the literature included in this study, further clinical validation is needed for this conclusion.

At present, intestinal flora has attracted more and more attention from scholars in China and foreign countries, and its association with ischemic stroke (IS) has gradually become a new research hotspot in the field of stroke. Studies also showed that intestinal flora may be a risk factor which directly or indirectly affects the occurrence and development of IS through bacterial metabolites and immune activities. In the present paper, we review the positive effect of acupuncture and moxibustion in alleviating the symptoms of limb locomotor, speech, swallowing dysfunction, cognition, etc. to improve the IS patients' daily life ability and in strengthening the cellular immune function of the body. In addition, acupuncture and moxibustion have a positive effect in regulating intestinal flora and immune inflammation. Hence, in the present paper, we discuss their relationship and the possibility of application of acupuncture and moxibustion therapies to the treatment of IS according to the theory of "intestinal flora-immune response". It is thus reasonable to speculate that acupuncture and moxibustion can be used to promote the recovery of brain tissue injury and neurological function after stroke via correcting intestinal flora disturbance and reducing immune inflammatory response. In-depth exploration of the role of "intestinal flora-immune response" in the treatment of IS and the specific regulatory function of acupuncture and moxibustion will provide new ideas and research approaches to reveal their mechanisms in the treatment of stroke from a new perspective.

The applied technology in effectiveness of weapons systems is the ability of weapons to complete the task in the offensive and defensive battle field environment. In this paper, ADC method is proposed to assess weapons systems. The assessment method of the probability of service, hit probability, mobility and protection for a weapon is studied. Firstly, after the components level division of weapons systems, the solution process of system availability row vector is analyzed based on reliability and maintenance-related. Secondly, the characteristics of laser range finder is analyzed, the whole process of search target is simulated. At last, simulation statistics according to the process of antiaircraft gun burst firing to obtain the burst firing kill probabilities is proposed.

Based on the designing of information and communication networking training needs analysis simulation system, this paper studies some key technology about needs analysis simulation system’s and some practical problems during designing, combining with the primary work done by self. At first, based on the summary of the basic concepts of information and communication networking training needs analysis simulation system and domestic and overseas development status, needs analysis simulation system and its modeling, the technique of simulation is studied, and the basic concept and model frame of needs analysis simulation system’s are summarized. In succession, the primary project of simulation training system is demonstrated and designed, including analyzing the requirement of the system function, putting forward the principle of system design, confirming the framework of hardware and software and the configuration of computer, as well as doing some top design on the software system of simulation training such as module partition and function describing.

In order to research on applied technology in assessing the application effectiveness of command information system (CIS) based on digital command of CIS, the article sticks tightly the army operations during wartime, setting up the index system of the effectiveness assessment. The paper putts forward the whole assessment index architecture. By computing index gray white clustering coefficient and weight vector and setting up multilayers gray clustering effectiveness valuation mode, the thesis uses the judgment matrix of effectiveness valuation, assessing the effectiveness of the application of CIS under the " based on digital command mode" and " based on audio command mode". Based on the valuation conclusion, the article explains these important factors to the system effectiveness and provides a research basis for making some scientific improvement measures.

Objective To investigate the factors influencing the therapeutic effect in acupuncture treatment of apoplectic pseudobulbar palsy (PBP).

To observe the effect of electroacupuncture (EA) of "Tianshu" (ST 25) on colonic smooth muscle structure and interstitial cells of Cajal (ICC) in slow transit constipation (STC) rats, so as to investigate its mechanism underlying improving STC.Wistar rats (n = 56) were randomly allocated to control (n = 20), model and EA groups (n = 18/ group). STC model was established by feeding the rats with forage containing compound Diphenoxylate. EA (0.8 - 1.3 mA, 2 Hz/15 Hz) was applied to bilateral "Tianshu" (ST 25) for 15 min, once daily for 14 days. The treatment was given beginning from the next day on after the rat's first black stool excretion was found. The colonic smooth muscle structure was observed under microscope after sectioning and HE staining. The immunoactivity (average optical density, OD) of intestinal immuno-reaction (IR) po-sitive ICC product was detected by immunohistochemistry.(1) Microscopic observation displayed that after modeling, the rat's intestinal mucosa gland showed atrophy with reduction in the acinar number, being sparse in distribution; and the smooth muscle thickness was obviously thinner compared with the normal control group (P0.05). Whereas this situation of the colonic smooth muscle in the EA group was close to that of the normal control group. Compared with the model group, the thickness of the intestinal smooth muscle in the EA group was significantly bigger (P0.05). (2) Immunohistochemical results showed that the average OD values and number of IR-positive colonic ICC in the model group were obviously decreased in comparison with the normal control group (P0.05). Compared with the model group, the average OD values and the number of IR-positive coIonic ICC in the EA group were upregulated considerably (P0.05).EA of bilateral "Tianshu"(ST 25) is able to improve STC-induced structural changes of the colonic smooth muscle, and upregulate expression of colonic ICC in STC rats, which may be responsible for its efficacy in improving STC.

To study the effect of electroacupuncture (EA) on changes of aquaporin-4 (AQP4) expression in rats with cerebral ischemia/reperfusion(CI/R).One hundred and seventy male Sprauge-Dawley rats were randomized into control (n = 50), CI/R model (n = 60) and EA (n = 60) groups, and the later 2 groups were further divided into 6 h, 12 h, 24 h, 48 h and 72 h subgroups, with 12 cases in each. CI/R model was established by occlusion of the middle cerebral artery (MCAO). EA (3.85/6.25 Hz, 0.8 - 1.0 mA) was applied to "Shuigou" (GV 26) and "Baihui" (GV 20) for 30 min. Transcardial perfusion was conducted with normal saline + paraformaldehyde in rats of different groups for taking brain tissue samples. The degree of brain edema was detected by cresyl violet stain method, permeability of the blood-brain barrier (BBB) reflected with IgG value in brain tissue was displayed by immunohistochemistry, and AQP4 protein and mRNA expression of corpora striata (CS) were detected by using immunohistochemistry (ABC) and RT-PCR techniques respectively.Twelve hours after CI/R, the hemisphere of brain began to get swelling significantly and the exosmose state of IgG became evidently; compared with the corresponding phases of model group, the swell rates in 24 h, 48 h and 72 h subgroups of EA group were significantly lower (P0.05, 0.01), and nearly no IgG immunoreaction positive products were found in different phases of EA group. The expression of AQP4 protein and mRNA increased significantly from 12 h and 24 h on separately in model group, while the expression in EA group was markedly weaker. The ratios of AQP4 mRNA and beta-actin mRNA of CS on the affected side of the brain from 24 h to 72 h of EA group were significantly lower than those of model group (P0.05, 0.01).EA can reduce the degree of brain edema induced by CI/R and improve the degree of BBB injury, which may be related to its effect in down-regulating AQP4 expression.
