To investigate the effect of baicalin on osteogenic differentiation of stem cells from human apical papilla(SCAPs).The third-passage SCAPs with stable passage ability were selected. After culturing for 24 hours in a medium containing 0, 10, 20, 50, 100 μmol/L of baicalin, cell viability in each group was measured by live/dead fluorescence staining. Then the SCAPs were divided into normal medium group(DMEM) and osteogenic induction medium group(ODM), and the experimental groups were further divided into 5 subgroups according to different concentrations of baicalin, i.e. 0, 10, 20, 50 and 100 μmol/L. Alkaline phosphatase(ALP) staining and ALP quantitative assay were used to detect the effects of different concentrations of baicalin on early osteogenic differentiation of SCAPs for 7 and 14 days; the effect of different concentrations of baicalin on osteogenic mineralization of SCAPs was determined by alizarin red staining after 21 days of culturing. MTT assay was used to detect the effects of baicalin at different concentrations on the growth of SCAPs. SPSS 20.0 software package was used for statistical analysis of the data.The results of live/dead cell fluorescence staining showed that SCAPs were vigorous after treatment in each concentration group, but when the concentration of baicalin was higher than 50 μmol/L, the number of dead cell increased. ALP quantitative results showed that ALP activity in the 20 μmol/L concentration group was significantly higher than that in the control group on the 14th day after induced differentiation in ODM group (P0.05), and ALP activity in 100 μmol/L concentration group was significantly lower than that in the control group(P0.05). After 14 days of DMEM culture, ALP activity of 20 μmol/L group was also significantly higher than that of the control group(P0.05). Quantitative analysis of alizarin red showed that the A value of the 100 μmol/L group was significantly lower than the control group in ODM. 20 μmol/L baicalin was the optimum concentration to promote growth of SCAPs.Baicalin with a concentration higher than 50 μmol/L can significantly inhibit osteogenic differentiation of SCAPs, while low concentration of baicalin, especially 20 μmol/L of baicalin, has a promoting effect on SCAPs osteogenesis differentiation and proliferation.

A series of ( E)- N-aryl-4 H-thiochromen-4-imines has been conveniently obtained through a cascade reaction between o-bromobenzothioamides and terminal alkynes. This novel approach probably involved an initial generation of benzothietane-2-imine intermidates via an intramolecular Ullmann reaction under CuI/L-proline cocatalysis and alkaline conditions followed by imine alkynylation, ring opening, and cyclization sequences to provide the unexpected 4 H-thiochromen-4-imines rather than isothiochromans.

Human bone marrow-derived mesenchymal stromal cells (hBMSCs) have been revealed to be beneficial for the regeneration of tissues and cells in several diseases. The present study aimed to elucidate the mechanisms underlying the effect of hBMSC transplantation on neuron regeneration in a rat model of middle cerebral artery occlusion (MCAO). The hBMSCs were isolated, cultured and identified. A rat model of MCAO was induced via the modified Longa method. Neurological severity scores (NSS) were adopted for the evaluation of neuronal function in the model rats after cell transplantation. Next, the expression levels of nestin, β-III-tubulin (β-III-Tub), glial fibrillary acidic protein (GFAP), HNA and neuronal nuclear antigen (NeuN) were examined, as well as the positive expression rates of human neutrophil alloantigen (HNA), nestin, NeuN, β-III-Tub and GFAP. The NSS, as well as the mRNA and protein expression of nestin, decreased at the 1st, 2nd, 4 and 8th weeks, while the mRNA and protein expression of NeuN, β-III-Tub and GFAP increased with time. In addition, after treatment, the MCAO rats showed decreased NSS and mRNA and protein expression of nestin, but elevated mRNA and protein expression of NeuN, β-III-Tub and GFAP at the 2nd, 4 and 8th weeks, and decreased positive expression of HNA and nestin with enhanced expression of NeuN, β-III-Tub and GFAP. Therefore, the present findings demonstrated that hBMSC transplantation triggered the formation of nerve cells and enhanced neuronal function in a rat model of MCAO.

Background and objective Angiographic staging progression in the operated hemisphere of patient with moyamoya disease (MMD) is a common phenomenon that neurosurgeons may encounter. However, few studies have been carried out to demonstrate the correlation between the operation and angiographic staging progression. This study aimed to reveal whether cerebral revascularization would affect the stage progression in patient with MMD. Methods A total of 20 pediatric patients with bilateral MMD were included in this study. All enrolled patients were at the similar Suzuki angiographic staging on bilateral hemispheres and cerebral revascularization was performed in unilateral hemisphere. Angiographic examination was performed in the first year after the operation to evaluate the progression of the lesions. Results Cerebral revascularizations including direct bypass or encephaloduroarteriosynangiosis were performed in 20 hemispheres. During the follow-up, the incidence of stage progression in the operated hemisphere was significantly higher than in the contralateral side (80.0% versus 20.0%, respectively; P = 0.036) and the interval of disease progression was significantly shorter in the operated side than in the contralateral side (mean interval time, 7.3 versus 10 months). Earlier Suzuki stage on hemisphere represented faster pace of stage progression compared with server Suzuki stage (OR = -0.612, P = 0.004), while neoformative collaterals had not significant correlation with the progression pace. The leptomeningeal collaterals from the posterior circulation decreased in almost all operated hemisphere, while gradually increasing in the contralateral hemisphere. Symptoms were improved in all patients after operation. Conclusion Cerebral revascularization would significantly accelerate the angiographic staging progression on the treated hemisphere, while it would not cause the deterioration of the patient's condition. The progression pace was associated with Suzuki stage.

The domino reaction of 4-halogenated isatins and 2-amino-N′-arylbenzohydrazides catalyzed by CuI in the presence of Cs2CO3 is described. A possible reaction mechanism containing the consecutive condensation, cyclization, C–N bond Ullmann coupling, ring-opening and decarboxylation is put forward according to the control experiments. This procedure provides an efficient method for the synthesis of fused polycyclic 1-amino-5-arylindazolo[3,2-b]quinazolin-7(5H)-ones derivatives in good yields.

Consecutive Knoevenagel condensation, Michael addition, cyclization, and Ullmann Csp2-N coupling reactions took place in the Domino reaction of o-halogenated benzaldehydes, tert-butyl 2,4-dioxopiperidine-1-carboxylate, and 1H-indazol-6-amine or 3H-benzo[d]imidazol-5-amine catalyzed by CuI. It gave series of fused hexacyclic heterocycles containing naphthyridine, acridine, and pyrazole (imidazole) moieties in one-pot under ligand-free conditions.

Purpose To explore the possible surgical factors related with nonunion in femoral shaft fracture following intramedullary nailing. Methods We retrospectively analyzed totally 425 patients with femoral shaft fracture in level I urban trauma center, including 254 males and 171 females, with an average age of 37.6 (ranging from 21 to 56) years old. The inclusion criteria included: (1) traumatically closed fracture of femoral shaft, with preoperative films showing non-comminuted fracture, such as transverse fracture, oblique fracture or spiral fracture; (2) closed reduction and fixation with interlocking intramedullary nail at 3–7 days after trauma; (3) complete follow-up data available. The relationship between the following factors (fracture site, reduction degree, direction of nail insertion and nail size) and nonunion was studied. Results The incidence of femoral nonunion was 2.8% in patients with closed simple fracture undergoing interlocking intrameduallary nailing, including 11 cases of hypertrophic nonunion. Nonunion was related significantly to distal fracture, unsatisfactory reduction and unreamed nail (p 0.05). Conclusions Nonunion in femoral shaft facture following interlocking intramedullary nailing is related to fracture site, fracture reduction and nail diameter. The choice of reamed nails or unreamed nails depends on the fracture site and reduction degree.

The iodine-catalyzed reaction of 2-(2-aminophenyl)quinazolin-4(3H)-ones and benzaldehydes was treated in ionic liquids, giving a series of 6-aryl-5H-quinazolino[4,3-b]quinazolin-8(6H)-one derivatives in high yields. This novel procedure has the advantages of high yields, being metal-free and having a mild reaction condition.

A consecutive Sonogashira coupling reaction, acetylene hydroamination cyclization of 2-(2-bromophenyl)quinazolin-4(3H)-ones and terminal alkynes, is described catalyzed by CuI/l-proline in the presence of Cs2CO3. This procedure provided a facile method for the synthesis of isoindolo[1,2-b]quinazolin-10(12H)-one derivatives in good yields.

There has been a lot of research on traffic flow density detection in recent years. In this paper, we propose a simple and practical method for traffic flow density detection through using the multiple pairs of infrared photoelectric sensors and STC15F2K60S2 as the main controller. This method can be used for the multiple cars two-way or multi-way side by side traffic flow density detection. The circuit is simple, accurate and efficient. The proposed method is economical and practical. Thus it can promote the progress of urban traffic management.

A total of 168 courtship sequences from 12 male and 18 female adult captive Sacalia quadriocellata were recorded on video. Thirty male and four female discrete motor patterns were defined and described. The behavioural sequences were summarized in an intra-individual dyadic transition matrix and analyzed using chi-square and kappa analyses. Courtship behaviour models and a flow diagram were constructed for this species. The male display patterns involved tactical and visual signals to induce female receptivity to mating. In response, females may emit olfactory signals for gender recognition. Female rejection of male suitors resulted in a male success rate of 4.17%. Only seven copulations were observed. Copulation only occurred when a female became quiescent and relaxed her tail for coition.

This study aims to investigate the preventive role and potential mechanisms of blocking extracellular HMGB1 function on doxorubicin induced cardiac injury. Mice were treated with HMGB1 blocker glycyrrhizin 1 h before and one time every day (intraperitoneal, 10 mg per mouse) after doxorubicin injection, and sacrificed on the day 14 after doxorubicin challenge. Cardiac function was evaluated by echocardiography and hemodynamic measurement. Myocardial inflammation and collagen deposition were analyzed by immunohistochemistry and picrosirius red staining. The interaction of HMGB1 and TLR2 was assessed by co-immunoprecipitation and confocal microscopy. The protein contents of HMGB1, MyD88, p65NF-kappaB and phospho-p65NF-kappaB were measured by Immunoblot. Compared with mice treated with saline, doxorubicin treatment led to an upregulation in HMGB1 expression. Blocking HMGB1 activity with glycyrrhizin protected mice against cardiac dysfunction, inflammatory response, and cardiac fibrosis induced by doxorubicin challenge. Glycyrrhizin inhibited the interaction of HMGB1 and TLR2, and blocked the downstream signaling of TLR2. In conclusion, blocking HMGB1 protected against doxorubicin induced cardiac injury by inhibiting TLR2 signaling pathway.

Purpose : To evaluate the growth of the composite corium (constructed with fibroblast cells and gelatinco- Bletillastriata gelatin/ Salvia miltiorrhiza materials) on rats. Methods : The composite artificial corium was constructed by culturing fibroblast cells in gelatin-co- Bletillastriata gelatin/ Salvia miltiorrhiza materials. Full-thickness area of skin was excised from the mice and subsequently, the composite corium was transplanted on the wound. Thereafter, the growth difference of the composite artificial corium and natural corium were compared. In addition, real-time fluorogenic reverse transcription polymerase chain reaction (qRT-PCR) and western blot were performed to determine vascular endothelial growth factor (VEGF) expression at gene and protein levels. Results : The composite artificial corium showed significant repair promoting effect on the skin, and the structure of the repaired skin was similar to that of natural corium. Interestingly, PCR and western blot results showed that the expressions of VEGF were higher in composite artificial corium than in natural corium on days 3 and 7 post-transplantation. Conclusion : The composite artificial corium has some clinical prospects for use in the treatment of wounds on large areas of skin. To study the effect of surface treating methods on the shear strength of Panavia F luting cements to Cercon zirconia.Forty sample disc of Cercon zirconia with 20 mm in diameter and 4 mm in height were prepared. Another 40 sample discs with 5 mm in diameter and 4 mm in height were also prepared Both of the two types of samples were randomly devided into four groups A, B, C, and D in which different surface treatments were delivered. In group A samples was treated with 600# sand paper, and in group Bwith sand blasting, and silanization in group C, and sand blasting plus silanization in group D. All samples were bonded with Panavia F luting cement under the aid of glass mould. The value of shear strength was measured and statistically analysed.The shear strength of four groups of samples were (21.50 ± 1.98), (23.68 ± 2.31), (20.69 ± 1.55), (24.01 ± 2.19) MPa respectively. The population mean was not equal. There was no significant difference between 600# sand paper treated group and silanization group, nor between sand blasting group and sand blasting plus silanization group.Sand blasting is a effective means to increase the shear strength, and 600# sand paper treatment and silanization can't increase the shear strength.

To introduce the characteristics and its clinical effects of Hennipen external rotation procedure for reduction of acute anterior shoulder dislocation.From March 2007 to March 2010, 72 patients with acute anterior shoulder dislocation were treated with manipulative reduction in our department. All the patients were divided into two groups: the Hippocratic group with 44 patients receiving regular Hippocratic procedure, and the Hennipen group with 28 patients receiving Hennipen external rotation procedure. Clinical manifestation and radiology were used to diagnose. After reduction procedures, external fixation was applied for another 3 to 4 weeks. The reduction course, complication and function recovery at follow-up were recorded and compared. The student test was used to analyze the difference between groups.Thirty-two patients in the Hippocratic group and 3 in Hennipen received intravenous anesthesia. Manipulation time of the Hippocratic group ranged from 2 to 5 minutes with a mean of (3.9 +/- 1.2) minutes, obviously different from that of Hennipen,which ranged from 0.5 to 2 minutes with a mean of (1.3 +/- 0.7) minutes. Complications occurred in 2 cases of the Hippocratic group and none of the Hennipen group. The UCLA scores of shoulder joint at follow-up were 34.2 +/- 2.1 and 33.8 +/- 1.0 respectively in two groups, with no significant difference.Compared with Hippocratic procedure, Hennipen external rotation procedure is easier to manipulate, bring less complications and the same effect. It is worth further clinical applications.

To investigate the influence of post-core background color on chromatic value of four all-ceramic system core materials at clinically appropriate thicknesses.Disc specimens of 15 mm in diameter and 0.80 mm in thickness (Empress II: Group A), and 0.50 mm in thickness (In-Ceram Zirconia core: Group B; Cercon base color zirconia core: Group C; Cercon base zirconia core: Group D) were fabricated, five in each group. Au-Pt alloy, Ni-Cr alloy and visible light cured dental composite resin (A2 color) background were prepared. Samples were put on different background and their chromatic values were measured with colorimeter (CIE-1976-L(*)a(*)b(*)). Color differences of each specimen on different background material were calculated.The color differences among specimens of Group A on different background material were more than 1.5 (2.83 ± 0.70) which meant it could be noticeable to eyes. Those of zirconia were less than 1.5 [Group B: (0.14 ± 0.08); Group C: (0.90 ± 0.20); Group D: (0.99 ± 0.09)].The influence of background color on Group A was noticeable to human eyes, and as a result, tooth-colored post should be used for this all-ceramic system. For the other three kinds of zirconia core materia1 system, the color differences among specimens on different background material were unnoticeable. Therefore the three all-ceramic systems have excellent color masking ability and can be used on all color background.

Aims Innate and adaptive immune responses are associated with the development of hypertension-induced myocardial hypertrophy and fibrosis. As a result, we investigated whether heat shock protein (HSP) 70, which is a molecule of damage-associated molecular patterns, could induce inflammation in the myocardium and promote the development of hypertension-induced cardiac hypertrophy and fibrosis. Methods and results We found that HSP70 serum levels, as well as the amount of HSP70 translocation to the cardiomyocyte membranes and the interstitial space, were elevated in the hypertensive mice caused by abdominal aortic constriction (AAC). Transcriptional inhibition of HSP70 expression by a specific heat shock transcript factor inhibitor, KNK437, reduced the serum level, and the re-distribution of HSP70. It promoted myocardial hypertrophy and cardiac dysfunctions although it protected animals from AAC-induced cardiac fibrosis. On the other hand, the functional antagonism of HSP70 by an anti-HSP70 antibody attenuated AAC-induced cardiac hypertrophy and fibrosis without adverse haemodynamic effects. The cardioprotective effect of the anti-HSP70 antibody was largely attributed to its ability to block AAC-activated immune response in the heart, as was indicated by suppressing the hypertension-enhanced conjugation of HSP70 with toll-like receptor 4, reducing heart-infiltrating macrophages, decreasing the expression of pro-inflammatory factor monocyte chemoattractant protein-1 and profibrotic factor transforming growth factor beta 1, and attenuating pro-hypertrophy signal MAPK P38 and ERK. Conclusion These results indicate that intracellular and extracellular HSP70 have different roles in the regulation of cardiac remodelling and function in response to hypertension. Extracellular HSP70 is a potential therapeutic target against cardiac hypertrophy and fibrosis.

In this article, canonical correlation analysis was used to explore the relationship between the toxicity-attenuating effect and the variation of chemical contents in rhubarb caused by processing. With quasi-acute toxicity test, the difference of hepatic and renal toxicity to mice with the processed materials of rhubarb was researched. The chemical contents of anthraquinones and tannins in rhubarb were measured by UV-vis spectrophotometry and high performance liquid chromatography. The results showed that there were toxic effects to liver and kidney in mice after repeated intragastric administration of rhubarb and its processed materials for 14 days at a dosage of 76 g x kg(-1). The toxic effect of processed materials was much lower than crude drug. With canonical correlation analysis, the sequence of the hepatic and renal toxicity of chemical contents in rhubarb were found as follows: total anthraquinone glycosides (AQGs)tannins (Tns)total anthraquinones (AQs); aloe-emodin (AE)physcione (Ph)rhein (Rn)emodin (Ed)chrysophanol (Ch) and AEGPhGChGEdGRnG of glycosyl-anthraquinone. It could be concluded that processing would attenuate the toxicity of crude drug of rhubarb. The toxicity-attenuating effect might be correlated to the decline of the contents of both anthraquinone glycosides and tannins, especially the aloe-emodin glycoside and physcione glycoside. The results also suggested that the serum alanine aminotransferase (ALT) and creatine (CREA) would be useful to monitor the hepatic and renal toxicity of rhubarb.

To set up the HPLC fingerprints of different samples of processed Rhizoma Coptidi (RC) in order to offer evidence for identifying different processed RC and preliminary discussion on processing mechanism.Seven different samples of processed RC were determined by HPLC and the results were analyzed by Hierarchical clustering and similarity evaluation system for chromatographic fingerprint of TCM.The characteristic of common peaks for fingerprints of different samples of processed RC is distinct, but there are differences among various processed RC. The different samples of processed RC were classified into two and the result of hierarchical clustering is correlated with the property of traditional Chinese drug.It is proved that the method which is convenient, rapid and repeat well can be used to identify different samples of processed RC. The change of property of different processed RC is related with processing adjuvant, the possible mechanism is that new components are produced or the contents of known components have changed due to the adjuvant during processing.

The adverse drug reaction (ADR) of traditional Chinese medicine (TCM) has been concerned nowadays. Some bans were published to limit their use abroad. There wasn't a feasible criterion to reply to the ADR of TCM yet in China. The ADR of 6 kinds of typical TCM which state attached importance to was chosen to discuss. Based on the retrospect investigation, the objectivity of ADR was explored in this study.

To observe the different extracts from Radix Isatidis on multiplication of mice lymphocytes.Lymphocytes were separated and cultured. Immunological activities of different extracts from Radix Isatidis were studied by thermodynamics and the results were tested by the conventional pharmacological experiments.The results showed that the water extract and residue had significant immunological effects while organic solvent extracts had immunological activity to some extent.The comparison of immunological activity among the extracts from Radix Isatidis were as follows: residue after extractinggeneral extractnBuOH extractEtOAc extractCHCl3 extractP E extract.

To detect the concentration of monocyte chemoattractant protein-1 (MCP-1) in the serum of patients with incomplete spinal cord injury and evaluate its relation with the pathologic classification of the spinal cord injury.MCP-1 concentration in the serum of patients with incomplete spinal cord injury (iSCI), single spine compression and healthy subjects were detected by ELISA, respectively in the present study and the magnetic resonance imaging data of these patients were studied at the same time on a blind base.Serum level of MCP-1 in iSCI patients was 428 pg/ml +/- 11 pg/ml by ELISA, which was higher than both that of the patients with single spine compression and of controls, with the concentration of 184 pg/ml +/- 21 pg/ml and 124 pg/ml +/- 15 pg/ml, respectively. There was significant difference between any two groups (P0.01). iSCI patients with normal MRI showed a lower serum level of MCP-1 as 312 pg/ml+/- 30 pg/ml. Pathological classification of spinal cord edema and hematoma corresponded to 390 pg/ml +/- 16 pg/ml and 508 pg/ml+/- 24 pg/ml in the concentration of MCP-1.MCP-1 may induce secondary inflammatory response by recruiting inflammatory cells to the injury site and thus affect the prognosis of spinal cord injury.
