Abstract Background The ambitious expansion of social health insurance in China has played a crucial role in preventing and alleviating poverty caused by illness. However, there is no government-sponsored health insurance program specifically for younger children and inequities are more pronounced in healthcare utilization, medical expenditure, and satisfaction in some households with severely ill children. This study assessed the effectiveness of child health insurance in terms of alleviating poverty caused by illness. Methods Data were collected from two rounds of follow-up surveys using the China Family Panel Studies 2016 and 2018 child questionnaires to investigate the relationship between child health insurance and household medical impoverishment (MI). Impoverishing health expenditure (IHE) and catastrophic health expenditure (CHE) were measured to quantify “poverty due to illness” in terms of absolute and relative poverty, respectively. Propensity score matching with the difference-in-differences (PSM-DID) method, robustness tests, and heterogeneity analysis were conducted to address endogeneity issues. Results Social health insurance for children significantly reduced household impoverishment due to illness. Under the shock of illness, the incidences of IHE and CHE were significantly lower in households with insured children. The poverty alleviation mechanism transmitted by children enrolled in social health insurance was primarily driven by hospitalization reimbursements and the proportion of out-of-pocket medical payments among the total medical expenditure for children. Conclusions Children’s possession of social health insurance significantly reduced the likelihood of household poverty due to illness. The poverty-reducing effect of social medical insurance is most significant in rural areas, low-income families, no-left-behind children, and infants. Targeted poverty alleviation strategies for marginalized groups and areas would ensure the equity and efficiency of health system reforms, contributing to the goal of universal health insurance coverage in China.

Abstract Background The development of the human vermiform appendix at the cellular level, as well as its function, is not well understood. Appendicitis in preschool children, although uncommon, is associated with a high perforation rate and increased morbidity. Methods We performed single-cell RNA sequencing (scRNA-seq) on the human appendix during fetal and pediatric stages as well as preschool-age inflammatory appendices. Transcriptional features of each cell compartment were discussed in the developing appendix. Cellular interactions and differentiation trajectories were also investigated. We compared scRNA-seq profiles from preschool appendicitis to those of matched healthy controls to reveal disease-associated changes. Bulk transcriptomic data, immunohistochemistry, and real-time quantitative PCR were used to validate the findings. Results Our analysis identified 76 cell types in total and described the cellular atlas of the developing appendix. We discovered the potential role of the BMP signaling pathway in appendiceal epithelium development and identified HOXC8 and PITX2 as the specific regulons of appendix goblet cells. Higher pericyte coverage, endothelial angiogenesis, and goblet mucus scores together with lower epithelial and endothelial tight junction scores were found in the preschool appendix, which possibly contribute to the clinical features of preschool appendicitis. Preschool appendicitis scRNA-seq profiles revealed that the interleukin-17 signaling pathway may participate in the inflammation process. Conclusions Our study provides new insights into the development of the appendix and deepens the understanding of appendicitis in preschool children. Graphical Abstract

The experimental process of rebound testing of rubber material was simulated using finite element analysis software ABAQUS, and the energy changes during the rebound process were discussed, laying the foundation for further research on viscoelasticity of rubber materials. The results showed that the characterization results of the new method were basically consistent with the actual test results, proving that the fitted intrinsic equation was consistent with the properties of the material itself, and at the same time characterizing the accurate energy changes during the rebound process. During the impact process, the kinetic energy of the pendulum was mainly transformed into the elastic potential energy and dissipation energy of the specimen. In addition, by analyzing the changes in the temperature field of the sample model, it was more intuitive to observe the temperature rise due to the energy loss during rubber deformation, indicating that part of the lost kinetic energy was converted into thermal energy and dissipated into the environment.

Southern Xinjiang is an important soybean production region in China. However, the short growing season and the cultivation of winter crops (such as wheat) in the region limit the expansion of soybean planting areas. An increased planting density can compensate for the loss in yield due to delayed sowing. To identify the quantitative relationship between increased density and delayed days, a two-year field experiment was conducted at the Tarim University Agronomy Experiment Station. Two sowing dates (April 7 (S1) and May 7 (S2)) and three planting densities of 206,800 plants·ha−1 (D1), 308,600 plants·ha−1 (D2), and 510,200 plants·ha−1 (D3) were used to compare various plant growth parameters and canopy characteristics. Late sowing and a high planting density significantly increased the plant height (S2 was 37.3% higher than S1, and D3 was 17.6% and 8.8% higher than D1 and D2), main stem internode, petiole length, and the mean tilt angle of the leaves (S2 was 22.5% higher than S1, and D3 was 11.7% higher than D2) but reduced the stem diameter (D3 was 28.6% and 12.5% lower than D1 and D2), branch number (S2 was 26.7% lower than S1, and D2 was 75% lower than D1), canopy light transmittance (S2 was 49.2% lower than S1, and D3 was 36.7% and 20.8% lower than D1 and D2), photosynthetic rate, and dry matter. The highest yield was achieved at S1D1, but the lowest yield was found for S2D1. Overall, the results suggest that earlier sowing and a lower planting density contribute to achieving an optimum canopy structure and higher yield. Our conclusions provide a reference for soybean production in southern Xinjiang.

Soybean, an important cash crop, is often affected by soil salinity, which is one of the important types of abiotic stress that affects its growth. Poly (acrylic) acid coated Mn3O4 (PMO) has been reported to play a vital role in defending against a variety of abiotic stresses in plants. To date, the effects of PMOs on soybean have not been reported; this study explored the mechanism of PMO-enhanced soybean germination under salt stress. In this experiment, 100 mg/L PMO was used as an immersion agent with a salt treatment of 150 mM NaCl. The results showed that when compared with the PMO treatment, salt stress significantly decreased the germination rate, fresh weight, carbohydrate content, and antioxidant enzyme activity of soybean and significantly increased the contents of reactive oxygen species, malondialdehyde, and osmoregulatory substances. However, PMO treatment enhanced the antioxidant defense system and significantly reduced the malondialdehyde content of soybean. Moreover, the activities of H+-ATPase and Ca2+-ATPase were significantly higher in treated soybean than in the control, and the content of ATP was also higher in treated soybean than in the control. Generally, PMO regulates the homeostasis of reactive oxygen species and reduces ATP consumption, thereby improving the ability of soybeans to germinate under salt stress. This study provides new insights into how nanomaterials improve plant salt tolerance.

Machining-induced residual stress (MIRS) in thin-walled components affects their machining accuracy, especially for large-size thin-walled components. This study focuses on the bodies of marine diesel engines, exploring the distribution of MIRS and distortion caused by the gravity-coupled machining residual stress during the boring process of the main bearing hole. The research obtained the distribution of MIRS and the machining distortion based on the finite element method and the mapping method. It examined the influence rules of various parameters, such as the cutting speed, feed, and depth of cut, on MIRS and machining distortion. The study shows that cutting speed, feed, and depth of cut are vital factors affecting MIRS and machining distortion. For the machining distortion of large-size and thin-walled components, their own weight is an essential factor that cannot be ignored. By optimizing the wall thickness, the distortion range can be effectively controlled, supporting the lightweight design of the structure.

Background: The introduction of endovascular repair provides an alternative to traditional open repair of thoracoabdominal aortic aneurysms (TAAA). Its utility is not well defined, however. Using a national database, we studied the treatment patterns and outcomes of TAAA to gain insight into its contemporary surgical practice in the United States. Methods: Records of TAAA patients who received endovascular and open repair were retrieved from the 2002 to 2018 National Inpatient Sample database. Each cohort was stratified into 4 age groups: ≤50, 51 to 60, 61 to 70, and >70 years. Patient characteristics and in-hospital outcomes were compared between the 2 repair modalities. Temporal trends were investigated. Results: Endovascular repair use increased steadily, whereas open repair volume remained stable until 2012, before declining by 50% by 2018. This appears to be associated with a declining number of open repairs in patients age >60 years. Patients who underwent endovascular repair were older and had a higher Charlson Comorbidity Index (mean, 2.8 ± 1.7 vs 2.5 ± 1.5; P 60 years but not for patients age ≤60 years. Conclusions: There has been a shift in the treatment of TAAA in the United States from open repair–dominant to endovascular repair–dominant. It has increased surgical access for older and more comorbid patients and has led to a decline in the use of open repair while lowering in-hospital mortality.

Existing intrusion detection techniques often require numerous malicious samples for model training.However, in real-world scenarios, only a small number of intrusion traffic samples can be obtained, which belong to few-shot scenarios.To address this challenge, a network intrusion detection method designed for few-shot scenarios was proposed.The method comprised two main parts: a packet sampling module and a meta-learning module.The packet sampling module was used for filtering, segmenting, and recombining raw network data, while the meta-learning module was used for feature extraction and result classification.Experimental results based on three few-shot datasets constructed from real network traffic data sources show that the method exhibits good applicability and fast convergence and effectively reduces the occurrence of outliers.In the case of 10 training samples, the maximum achievable detection rate is 99.29%, while the accuracy rate can reach a maximum of 97.93%.These findings demonstrate a noticeable improvement of 0.12% and 0.37% respectively, in comparison to existing algorithms.

Machining quality prediction is the critical link of quality control in parts machining. With the advent of the Industry 4.0 era, intelligent manufacturing and data-driven technologies bring new ideas for quality control in complex machining processes. Quality control is complicated for multi-process, multi-condition, small-batch, and high-precision parts processing requirements. To solve this problem, this paper proposes a machining quality prediction method based on the machining error transfer network and the grey neural network. Initially, by constructing a processing error transfer network, the error transfer law in part processing is described, and the PageRank algorithm and the influence degree of the nodes are used to determine the critical quality features. Additionally, the problem of low prediction accuracy due to small sample data and multiple coupling relationships is solved using the grey neural network algorithm, and a high accuracy prediction of critical quality features is achieved. Finally, the effectiveness and reliability of the method are verified by the case of medium-speed marine diesel engine fuselage processing. The results indicate that this method not only effectively identifies critical quality features in the machining process of complex parts, but it also maintains a high predictive accuracy for these features, even with small samples and limited data.

To address the problem of predicting machining quality for critical features in the manufacturing process of mechanical products, a method that combines information entropy and XGBoost (version 2.1.1) hyperparameter optimization is proposed. Initially, machining data of mechanical products are analyzed based on information entropy theory to identify critical quality characteristics. Subsequently, a quality prediction model for these critical features is established using the XGBoost machine learning framework. The model’s hyperparameters are then optimized through Bayesian optimization. This method is applied as a case study to a medium-speed marine diesel engine piston. After the critical quality characteristics in the machining process are identified, the machining quality of these vital characteristics is predicted, and the results are compared with those obtained from a machine learning model without hyperparameter optimization. The findings demonstrate that the proposed method effectively predicts the machining quality of mechanical products.

Aiming at the difficulty in effectively identifying critical quality features in the complex machining process, this paper proposes a critical quality feature recognition method based on a machining process network. Firstly, the machining process network model is constructed based on the complex network theory. The LeaderRank algorithm is used to identify the critical processes in the machining process. Secondly, the Entropy-CRITIC method is used to calculate the weight of the quality features of the critical processes, and the critical quality features of the critical processes are determined according to weight ranking results. Finally, the feasibility and effectiveness of the method are verified by taking the medium-speed marine diesel engine coupling rod machining as an example. The results show that the method can still effectively identify the critical quality features in the case of small sample data and provide support for machining process optimization and quality control, thus improving product consistency, reliability, and machining efficiency.

Aiming at the difficulty of quantitatively evaluating the critical processes in the manufacturing process of complex mechanical products, a critical process identification method based on the PageRank algorithm is proposed with the goal of identifying key processes in the machining process. Based on the complex network theory, the error transfer network model of the machining process is established in this paper. Adopting the actual machining process as the data set of the complex network, the weights of the machining feature nodes are calculated by the PageRank ranking algorithm, and the nodes are ranked according to the weight values to assess the influence and importance of the nodes in the network model. Finally, taking the connecting rod machining process of a medium-speed marine diesel engine as an example, the results show that the method can quickly and effectively identify the key processes in the machining process.

Background: Aidi injection, a classic traditional Chinese medicine (TCM) formula, has been used on a broader scale in treating a variety of cancers. In this study, we aimed to explore the potential anti-tumor effects of Aidi injection in the treatment of neuroblastoma (NB) using network pharmacology (NP).Methods: To elucidate the anti-NB mechanism of Aidi injection, an NP-based approach and molecular docking validation were employed. The compounds and target genes were collected from the Traditional Chinese Medicine Systems Pharmacology (TCMSP) database and Bioinformatics Analysis Tool for Molecular mechANism of Traditional Chinese Medicine (BATMAN-TCM) database. The protein–protein interaction network was constructed using the STRING database. clusterProfiler (R package) was utilized to annotate the bioinformatics of hub target genes. The gene survival analysis was performed on R2, a web-based genomic analysis application. iGEMDOCK was used for molecular docking validation, and GROMACS was utilized to validate molecular docking results. Furthermore, we investigated the anticancer effects of gomisin B and ginsenoside Rh2 on human NB cells using a cell viability assay. The Western blot assay was used to validate the protein levels of target genes in gomisin B- and ginsenoside Rh2-treated NB cells.Results: A total of 2 critical compounds with 16 hub target genes were identified for treating NB. All 16 hub genes could potentially influence the survival of NB patients. The top three genes (EGFR, ESR1, and MAPK1) were considered the central hub genes from the drug–compound–hub target gene–pathway network. The endocrine resistance and estrogen signaling pathways were identified as the therapeutic pathways using the Kyoto Encyclopedia of Genes and Genomes (KEGG) analysis. Gomisin B and ginsenoside Rh2 showed a good binding ability to the target protein in molecular docking. The results of cell experiments showed the anti-NB effect of gomisin B and ginsenoside Rh2. In addition, the administration of gomisin B over-regulated the expression of ESR1 protein in MYCN-amplified NB cells.Conclusion: In the present study, we investigated the potential pharmacological mechanisms of Aidi against NB and revealed the anti-NB effect of gomisin B, providing clinical evidence of Aidi in treating NB and establishing baselines for further research.

Background Pediatric medullary thyroid cancer (MTC) is one of the rare pediatric endocrine neoplasms. Derived from C cells of thyroid glands, MTC is more aggressive and more prompt to metastasis than other types of pediatric thyroid cancer. The mechanism remains unclear. Methods We performed single-cell transcriptome sequencing on the samples of the primary tumor and metastases lymph nodes from one patient diagnosed with MTC, and it is the first single-cell transcriptome sequencing data of pediatric MTC. In addition, whole exome sequencing was performed and peripheral blood was regarded as a normal reference. All cells that passed quality control were merged and analyzed in R to discover the association between tumor cells and their microenvironment as well as tumor pathogenesis. Results We first described the landscape of the single-cell atlas of MTC and studied the interaction between the tumor cell and its microenvironment. C cells, identified as tumor cells, and T cells, as the dominant participant in the tumor microenvironment, were particularly discussed in their development and interactions. In addition, the WES signature of tumor cells and their microenvironment were also described. Actively immune interactions were found, indicating B cells, T cells and myeloid cells were all actively participating in immune reaction in MTC. T cells, as the major components of the tumor microenvironment, proliferated in MTC and could be divided into clusters that expressed proliferation, immune effectiveness, and naive markers separately.

Objectives: This study aimed to explore the distribution and differences in the health status of migrant workers in China by gender and age. In addition, it investigated the causes of health inequalities among them.Methods: This paper analyzes the differences in health status across age groups for migrant workers of different genders based on the data from the China Migrant Dynamic Survey in 2018. It also empirically assesses how education level and health insurance impact gender-related health inequalities.Results: The results suggest that female migrant workers in China have significantly lower health levels than males. Furthermore, these differences in health are exacerbated with age. This disparity may be attributed to lower participation in social insurance participation and less educational attainment among female migrant workers than their male counterparts.Conclusion: The government should take effective practical measures to increase the social insurance participation rate of female migrant workers. Moreover, investing in female education to reduce health inequality among migrant workers is essential.

To realize automatic management via digitization and intellectualization has become a key technology for thousands of industries to reduce their operation and maintenance costs.As an enabler of digitization and intellectualization, 5G mobile communication system has evolved from manual operating network to human-assisted automation network.Based on the study of automatic technologies of 5G network, the L0～L5 autonomous network levels proposed by 3GPP were analyzed, several key technologies for intent-driven 5G autonomous network including intent life-cycle maintenance and solutions for intent conflict were discussed.Last but not least, a practice of intent-driven 5G autonomous network was proposed: solution of intent-driven application experience assurance.

Background Aortic dissection (AD) during pregnancy and puerperium is a rare catastrophe with devastating consequences for both parent and fetus. Population‐level incidence trends and outcomes remain relatively undetermined. Methods and Results We queried a US population‐based health care database, the National Inpatient Sample, and identified all patients with a pregnancy‐related AD hospitalization from 2002 to 2017. In total, 472 pregnancy‐related AD hospitalizations (mean age, 30.9±0.6 years) were identified from 68 514 000 pregnancy‐related hospitalizations (0.69 per 100 000 pregnancy‐related hospitalizations), with 107 (22.7%) being type A and 365 (77.3%) being type B. The incidence of AD appeared to increase over the 16‐year study period but was not statistically significant (P for trend >0.05). Marfan syndrome, primary hypertension, and preeclampsia/eclampsia were found in 21.9%, 14.4%, and 11.5%, respectively. Abstract Objective To explore the analgesic effects of different concentrations of ropivacaine in transversalis fascia plane (TFP) block during laparotomy. Methods Ninety patients who underwent laparotomy admitted to our hospital from March 2019 to March 2020 were selected as the study subjects and were divided equally into a low concentration group, a medium concentration group, and a high concentration group according to the randomized grouping method. The low concentration group adopted 0.4% ropivacaine 40 ml, the medium concentration group was given 0.5% ropivacaine 40 ml, and the high concentration group was given 0.6% ropivacaine 40 ml. The hemodynamic indexes and the incidence of adverse reactions in the two groups were compared. The Numerical Rating Scale (NRS) was used to assess the postoperative pain in the three groups, the Bruggrmann comfort scale (BCS) was used to assess the comfort level in the three groups, and the Mini-mental State Examination (MMSE) was used to evaluate the postoperative cognitive function of the three groups of patients. Abstract Background Hirschsprung's disease (HSCR) is a relatively common congenital disability. Accumulating extracellular matrix (ECM) prompts intestinal fibrosis remodelling in the aganglionic segments of HSCR. The contributions of various cellular subsets in the fibrogenesis of HSCR segments are poorly understood. Methods Single‐cell transcriptomics from 8 aganglionic segments and 5 normal segments of 7 HSCR subjects and 26 healthy segments of seven healthy donors were analysed. Fibrotic phenotype and alterations were explored using differential expression analysis and single‐cell trajectory analysis. Fibrosis‐related transcription factors were inferred through single‐cell regulatory network inference. Bulk transcriptomic data, proteomic data, immunohistochemistry (IHC) and real‐time polymerase chain reaction were used to validate the alterations in the HSCR intestine. Results Various collagen, fibronectin and laminin protein‐coding genes expression were up‐regulated in the stromal and glial cells of the HSCR intestine. The number of fibroblasts and myofibroblasts in the aganglionic segments increased, and more myofibroblasts were activated at an earlier stage in HSCR segments, which infers that there is an intestinal fibrosis phenotype in HSCR segments. The fibrotic regulators POSTN, ANXA1 and HSP70 were highly expressed in the ECM‐related cellular subsets in the transitional segments and aganglionic segments. The transcription factor regulatory network revealed that fibrosis‐related and megacolon‐related NR2F1 in the fibroblasts and glial subsets was up‐regulated in the aganglionic segment. Conclusions This work identifies intestinal fibrosis and related regulators in aganglionic segments of HSCR; hence, anti‐fibrotic therapy may be considered to prevent HSCR‐associated enterocolitis (HAEC), relieve intestinal stricture and improve cell therapy.

Abstract As a clean and renewable energy, wave energy has abundant reserves, and its reasonable development and utilization can effectively alleviate the problems of energy shortage and environmental pollution. This paper proposes a mechanical rectification wave energy converter. The reciprocating linear motion of the buoy is directly converted into the rotary motion through the ball screw. The magnetic coupling force transmission system is adopted to ensure the sealing of the device. Using mechanical rectification mechanism to ensure the service life of the generator. In addition, a dynamic model of the mechanical rectification wave energy converter is established. Through simulation analysis, the curve of power with load resistance is obtained. And there is an optimal load resistance value corresponding to the maximum power in different wave conditions. In order to improve the power generation efficiency, the maximum power point tracking control algorithm is designed based on the admittance differentiation method. By controlling the duty cycle of the switch tube in the Buck‐Boost chopper circuit, the variable load is realized, and then the maximum power point tracking control is realized. Finally, simulation analysis verifies the feasibility of the algorithm under different wave conditions.

Abstract Background Biliary atresia (BA) is a devastating inflammatory and fibrosing cholangiopathy of neonates with unknown aetiology. We aim to investigate the relationship between these two main characteristics. Methods Single‐cell RNA sequencing and spatial transcriptomics were performed on liver samples from a cohort of 14 objects (BA: n = 6; control: n = 8). We conducted data integration and cell‐type annotation based on gene expression profiling. Furthermore, we identified fibrosis‐related immune cells according to their spatial locations, GO and KEGG analysis. Finally, SPOTlight and CIBERSORTx were used to deconvolute ST data and microarray data of the GSE46960 cohorts, respectively. Results Immune subpopulations inhabiting the ‘fibrotic niche’ (areas of scarring), comprising ‘intermediate’ CD14++CD16+ monocytes, scar‐associated macrophages, natural killer T cells, transitional B cells and FCN3+ neutrophils were identified. GO and KEGG analyses showed that pathways including ‘positive regulation of smooth muscle cell/fibroblast proliferation’ and ‘positive regulation of/response to VEGFR/VEGF/EGFR/FGF’ were enriched in these cell types. Interactions analysis showed that communication among ‘FGF_FGFR’, ‘RPS19‐C5AR1’, ‘CD74_COPA/MIF/APP’ and ‘TNFRSF1A/B_GRN’ was extensive. Finally, the results of deconvolution for ST data and microarray data validated that the proportions of certain identified fibrosis‐related cell types we identified were increased in BA. Discussion Fibrosis is an important feature of BA, in which the immune system plays an important role. Our work reveals the subpopulations of immune cells enriched in the fibrotic niche of BA liver, as well as key related pathways and molecules; some are highlighted for the first time in liver fibrosis. These newly identified interactions might partly explain why the rate of liver fibrosis occurs much faster in BA than in other liver diseases. Conclusion Our study revealed the molecular, cellular and spatial immune microenvironment of the fibrotic niche of BA.

As a neuroendocrine tumor derived from the neural crest, neuroblastoma (NB) is the most common extracranial solid tumor in children. The prognosis in patients with low- and intermediate-risk NB is favorable while that in high-risk patients is often detrimental. However, the management of the considerably large proportion of high-risk patients remains challenging in clinical practice. Among various new approaches, oncolytic virus (OV) therapy offers great advantages in tumor treatment, especially for high-risk NB. Genetic modified OVs can target NB specifically without affecting normal tissue and avoid the widespread drug resistance issue in anticancer monotherapy. Meanwhile, its safety profile provides great potential in combination therapy with chemo-, radio-, and immunotherapy. The therapeutic efficacy of OV for NB is impressive from bench to bedside. The effectiveness and safety of OVs have been demonstrated and reported in studies on children with NB. Furthermore, clinical trials on some OVs (Celyvir, Pexa-Vec (JX-594) and Seneca Valley Virus (NTX-010)) have reported great results. This review summarizes the latest evidence in the therapeutic application of OVs in NB, including those generated in cell lines, animal models and clinical trials.

Sialoblastoma (SBL) is an infrequent embryonal malignant tumor originating from the salivary gland, resembling primitive salivary gland anlage, whereas hepatoblastoma (HB) is the most common pediatric liver malignancy. The simultaneous occurrence of both tumors is extremely rare. Here we reported a case of a 6-month-old infant diagnosed with synchronous SBL and HB. The patient received neoadjuvant chemotherapy followed by surgical resection. Fresh tissues of both tumors were collected before and after chemotherapy, which were further profiled by whole exome sequencing (WES) and single-cell RNA sequencing (scRNA-seq). WES analysis revealed potential somatic driver mutation PIK3CA p.Glu454Lys for SBL and canonical mutation CTNNB1 p.Ser45Pro for HB. No shared somatic variants or common copy number alterations were found between SBL and HB primary tumor samples. Though scRNA-seq, single-cell atlases were constructed for both tumors. SBL may recapitulate a pre-acinar stage in the development of salivary gland, including basaloid, duct-like, myoepithelial-like, and cycling phenotypes. In the meantime, HB was composed of tumor cells resembling different stages of the liver, including hepatocyte-like, hepatic progenitor-like, and hepatoblast-like cells. After chemotherapy, both tumors were induced into a more mature phenotype. In terms of transcriptional signatures, SBL and HB showed enhanced expression of epithelial markers KRT8, KRT18, and essential embryo development genes SDC1, MDK, indicating the disruption of normal embryo epithelium development. Finally, heterozygous deleterious germline mutation BLM and FANCI were identified which could predispose the patient to higher cancer risk. It partially explained the reason for the co-occurrence of SBL and HB. Taken together, we provided valuable resources for deciphering cellular heterogeneity and adaptive change of tumor cells after chemotherapy for synchronous SBL and HB, providing insights into the mechanisms leading to synchronous pediatric tumors.

During the rough marine environment, heave compensation is used to offset the heave motion of the vessel when a marine crane lifts and lands the load. Thus, load motion and vessel motion are realized decoupled. In previous studies, the interference items such as hydraulic cylinder friction, underwater drag force and nonlinear friction in the active heave compensation system of a marine hydraulic crane are compensated as a concentrated interference force to be estimated. In this paper, we disassembled the interference items; the disturbance observer and adaptive rate are designed to estimate unmodeled disturbance force and system uncertain parameters, respectively; and we designed an active heave compensator with the adaptive nonlinear cascade controller which has the disturbance observer (DOB-ANCC). For the heave compensation of load displacement, this paper derived the control law of the nonlinear system model based on the backstepping method. The outer loop control is displacement control and the inner loop control is pressure control. The simulation verifies the effectiveness of the control strategy proposed in this paper and the availability of heave displacement compensation for a marine crane hoisting load. The compensation efficiency of the designed controller (DOB-ANCC) for the heave motion of the load can reach more than 95%, and the maximum displacement tracking error of the controller can reach ± 0.035 m.

Abstract Background Transaxillary access (TAx) has shown promise as an excellent alternative TAVR option, but data on the Edwards SAPIEN 3 in TAx-TAVR is limited. We sought to study the safety and efficacy of TAx-TAVR using this current-generation balloon-expandable valve. Methods A retrospective study of our first 24 TAx and 20 transthoracic (TT) TAVR patients treated with the SAPIEN 3 valve was performed, and the patients’ preoperative characteristics, procedural outcomes, and clinical outcomes were compared to our first 100 transfemoral (TF) patients using the SAPIEN 3 device. Results There were no statistical differences observed for outcomes between the TAx and TF groups, despite the TAx patients having more comorbidities (STS-PROM 11.3 ± 7.6 versus 7.3 ± 5.2, p = 0.042). In addition, no significant difference was found in the fluoroscopy time and contrast amount between the two groups. The patients’ baseline characteristics were similar between the TAx and TT groups. Their procedural and clinical outcomes were comparable, but there was a trend towards lower incidence of acute kidney injury (13.0% versus 23.5%), new-onset atrial fibrillation (5.6% versus 33.3%), shorter median length of stay postoperatively (4 versus 6 days), fewer discharges to rehabilitation (16.7% versus 35.0%), and a lower rate of readmission within 30-days (8.3% versus 35.0%), all favoring TAx access. Conclusions TAx-TAVR with the SAPIEN 3 valve is a safe alternative to TF access. It offers advantages of improved recovery over TT access, and appears to be a superior alternative-access option for TAVR. TAx access could be preferred when TF access is not feasible.

The reliability of the machining process is a vital indicator in the manufacturing process of mechanical parts, and directly related to the machining quality of mechanical products. In order to effectively utilize machining process information and machining quality inspection data to evaluate the reliability of the machining process, this paper extends the relevant concepts of information theory to the field of mechanical manufacturing error analysis and establishes a reliability analysis model of the machining process. The model introduces information entropy into the analysis of machining process reliability by defining vertex entropy to measure the uncertainty of the machining quality of each process, and thus realizing the determination of critical processes using a fully quantitative method. Meanwhile, the transfer coefficient defined by mutual information is used to measure the transfer of machining error between two processes. Through combining vertex entropy and the transfer coefficient, the quantitative analysis of machining process reliability is achieved. The model proposed in this paper addresses the issue that the subjective factors of researchers in other related studies have an impact on the analysis results. Finally, the correctness of the proposed model is verified using Monte Carlo simulations.

An important task in the construction of China’s ecological civilization, the selection and implementation of policy instruments fully reflect the actual effectiveness of the government’s efforts to control air pollution. Based on the content analysis method, this study examines the changing process of air pollution control policy instruments in southwest China from 2010 to 2021 in terms of implementation, synergy, and integration of policy instruments. The results show that, in terms of the degree of mandatory, the frequency of using policy instruments generally increased with time, but the overall balance of the instrument portfolio was poor. In terms of the degree of synergy, a gradual shift occurred from government-led to government-society governance. However, the concept and modes of inter-governmental linkage and cross-regional collaborative governance need to be improved. As for the degree of systemic, a clear trend of instrument integration and more frequent information interaction was found. Emergency-oriented characteristics appear strong, but a regular governance mechanism is lacking. Therefore, this paper provides policy suggestions and academic considerations for further improving the effectiveness of air pollution management in southwest China from three aspects: optimizing the policy tool system, deepening the regional joint prevention and control mechanism of air pollution, and promoting intelligent air pollution management.

Abstract As major environment factors, drought or high salinity affect crop growth, development and yield. Transgenic approach is an effective way to improve abiotic stress tolerance of crops. In this study, we comparatively analyzed gene structures, genome location, and the evolution of syntaxin proteins containing late embryogenesis abundant (LEA2) domain. GmSYP24 was identified as a dehydration-responsive gene. Our study showed that the GmSYP24 protein was located on the cell membrane. The overexpression of GmSYP24 (GmSYP24ox) in soybean and heteroexpression of GmSYP24 (GmSYP24hx) in Arabidopsis exhibited insensitivity to osmotic/drought and high salinity. However, wild type soybean, Arabidopsis, and the mutant of GmSYP24 homologous gene of Arabidopsis were sensitive to the stresses. Under the abiotic stresses, transgenic soybean plants had greater water content and higher activities of POD, SOD compared with non-transgenic controls. And the leaf stomatal density and opening were reduced in transgenic Arabidopsis. The sensitivity to ABA was decreased during seed germination of GmSYP24ox and GmSYP24hx. GmSYP24hx induced up-regulation of ABA-responsive genes. GmSYP24ox alters the expression of some aquaporins under osmotic/drought, salt, or ABA treatment. These results demonstrated that GmSYP24 played an important role in osmotic/drought or salt tolerance in ABA signal pathway.

Background Transcatheter aortic valve replacement (TAVR) has become the preferred treatment for symptomatic patients with aortic stenosis and elevated procedural risk. Many deaths following TAVR are because of noncardiac causes and comorbid disease burden may be a major determinant of postprocedure outcomes. The prevalence of comorbid conditions and associations with outcomes after TAVR has not been studied. Methods and Results This was a retrospective single‐center study of patients treated with TAVR from January 2015 to October 2018. The association between 21 chronic conditions and short‐ and medium‐term outcomes was assessed. A total of 341 patients underwent TAVR and had 1‐year follow‐up. The mean age was 81.4 (SD 8.0) years with a mean Society of Thoracic Surgeons predicted risk of mortality score of 6.7% (SD 4.8). Two hundred twenty (65%) patients had ≥4 chronic conditions present at the time of TAVR. Background: Effective anticancer therapy can be achieved by identifying novel tumor-specific drug targets and screening of new drugs. Recently, TMEM16A has been identified to be overexpressed in lung adenocarcinoma, and inhibitors of TMEM16A showed obvious antitumor efficacy.Methods: YFP fluorescence quenching and whole-cell patch clamp experiments were used to explore the inhibitory effect of silibinin on TMEM16A. Molecular docking and site-directed mutagenesis were performed to confirm the binding sites of silibinin and TMEM16A. MTT assay, wound healing assay, and annexin-V assay were used to detect the effect of silibinin on cancer cell proliferation, migration, and apoptosis. shRNA was transfected into LA795 cells to knock down the expression of endogenous TMEM16A. Tumor xenograft mice combined with Western blot experiments reveal the inhibitory effect and mechanism of silibinin in vivo.Results: Silibinin concentration dependently inhibited the whole-cell current of TMEM16A with an IC50 of 30.90 ± 2.10 μM. The putative binding sites of silibinin in TMEM16A were K384, R515, and R535. The proliferation and migration of LA795 cells were downregulated by silibinin, and the inhibition effect can be abolished by knockdown of the endogenous TMEM16A. Further, silibinin was injected to tumor xenograft mice which exhibited significant antitumor activity without weight loss. Finally, Western blotting results showed the mechanism of silibinin inhibiting lung adenocarcinoma was through apoptosis and downregulation of cyclin D1.Conclusion: Silibinin is a novel TMEM16A inhibitor, and it can be used as a lead compound for the development of lung adenocarcinoma therapy drugs.

In this paper, the passive heave compensator for the strand jack lifting system is studied. An analytical model is developed, which considers the nonlinear characteristic of the compensator stiffness thus as to predict its response under different parameters accurately. This analytical model helps to find the feasible gas volume of the compensator. The comparative analysis is carried out to analyze the influence of key design parameters on the dynamic response of the compensator. In order to evaluate the effectiveness of the compensator, a coupling model of the strand jack lifting system is derived. The compensator efficiency is evaluated in terms of the lifted structure displacement and the strand dynamic tension. The numerical simulations are performed to evaluate the effectiveness of the compensator. Numerical results show that the compensator is able to significantly decrease the tension variation in the strands and the motion of the lifted structure.

A wave compensation platform based on 3-SPR parallel platform is designed for marine ships with a dynamic positioning system. It can compensate for the heave, rolling, and pitching movement of a vessel under level 4 sea state. The forward kinematics of the mechanism is used to draw the central point position workspace and the attitude workspace of the moving deck of the compensation platform. The compensation effects of the 3-RPS parallel compensation platform and the 3-SPR parallel compensation platform are compared, and the feasibility and superiority of the compensation scheme using the 3-SPR parallel compensation platform are proved. To lower the working height of the upper deck of the compensation platform and reduce the extension range of the support legs, the structure of the compensation platform is optimized, and a novel 3-SPR parallel platform is designed. Finally, a simulation model was established. Using the inverse kinematic model as a compensation movement solver which can online calculate the length of branch legs based on the measured heaving, rolling, and pitching values of vessels, the compensation effect of the new structure under a certain sea state is simulated. The result demonstrated the efficiency of the ship motion decoupling movement of the newly designed compensation platform and proved the competence of it.

Wave energy is a renewable energy source that is green, clean and has huge reserves. In order to develop wave energy resources, an oscillating buoy Wave Energy Converter (WEC) device based on the artificial breakwater is presented in this paper. In order to effectively vent the gas in the hydraulic PTO and to improve the active control capability of the PTO system to guarantee the safety performance of the system under high sea conditions, a hydraulic PTO with an active control circuit is designed. Additionally, for the Power Take-Off (PTO) system, there is a optimal damping point under different sea conditions for PTO system, so the PTO can be controlled by the Maximum-Power-Point-Tracking (MPPT) control algorithms to improve the generated power of the system. At present, the MPPT control algorithms for wave energy are mainly used to control the load of generator. However, a fixed-load storage battery is used for the load of the generator in this paper. Additionally, an MPPT control taken at a hydraulic PTO system is executed to improve the power generated by hydraulic PTO under different sea conditions effectively in this paper. The MPPT control based on the hydraulic system is conducted by controlling the displacement of hydraulic motor to achieve the optimal damping point tracking control. The control flow of the MPPT algorithm is provided. The variable step hill-climbing method is used in MPPT control algorithm in which the big step can reduce the time of tracking and the small step can increase the accuracy of MPPT control algorithm. Due to the slow stability of the hydraulic system, a filter method for hydraulic PTO power is used. In addition, the hydraulic PTO system and MPPT control are verified to be feasible with the simulation. Additionally, MPPT control based on hydraulic variable motor is easier to carry out in practical applications than the traditional control of resistance. Finally, the simulation results demonstrate that it is an effective power control strategy for hydraulic PTO system to improve the generated power.

Calcium ions bind at the gating ring which triggers the gating of BK channels. However, the allosteric mechanism by which Ca2+ regulates the gating of BK channels remains obscure. Here, we applied Molecular Dynamics (MD) and Targeted MD to the integrated gating ring of BK channels, and achieved the transition from the closed state to a half-open state. Our date show that the distances of the diagonal subunits increase from 41.0 Å at closed state to 45.7Å or 46.4 Å at a half-open state. It is the rotatory motion and flower-opening like motion of the gating rings which are thought to pull the bundle crossing gate to open ultimately. Compared with the 'Ca2+ bowl' at RCK2, the RCK1 Ca2+ sites make more contribution to opening the channel. The allosteric motions of the gating ring are regulated by three group of interactions. The first weakened group is thought to stabilize the close state; the second strengthened group is thought to stabilize the open state; the third group thought to lead AC region forming the CTD pore to coordinated motion, which exquisitely regulates the conformational changes during the opening of BK channels by Ca2+.

Soybean (Glycine max (L.) Merr.) isoflavone is important for human health and plant defense system. To identify novel quantitative trait loci (QTL) and epistatic QTL underlying isoflavone content in soybean, F5:6, F5:7 and F5:8 populations of 130 recombinant inbred (RI) lines, derived from the cross of soybean cultivar 'Zhong Dou 27' (high isoflavone) and 'Jiu Nong 20' (low isoflavone), were analyzed with 95 new SSR markers. A new linkage map including 194 SSR markers and covering 2,312 cM with mean distance of about 12 cM between markers was constructed. Thirty four QTL for both individual and total seed isoflavone contents of soybean were identified. Six, seven, ten and eleven QTL were associated with daidzein (DZ), glycitein (GC), genistein (GT) and total isoflavone (TI), respectively. Of them 23 QTL were newly identified. The qTIF_1 between Satt423 and Satt569 shared the same marker Satt569 with qDZF_2, qGTF_1 and qTIF_2. The qGTD2_1 between Satt186 and Satt226 was detected in four environments and explained 3.41%-10.98% of the phenotypic variation. The qGTA2_1, overlapped with qGCA2_1 and detected in four environments, was close to the previously identified major QTL for GT, which were responsible for large a effects. QTL (qDZF_2, qGTF_1 and qTIF_2) between Satt144-Satt569 were either clustered or pleiotropic. The qGCM_1, qGTM_1 and qTIM_1 between Satt540-Sat_244 explained 2.02%-9.12% of the phenotypic variation over six environments. Moreover, the qGCE_1 overlapped with qGTE_1 and qTIE_1, the qTIH_2 overlapped with qGTH_1, qGCI_1 overlapped with qDZI_1, qTIL_1 overlapped with qGTL_1, and qTIO_1 overlapped with qGTO_1. In this study, some of unstable QTL were detected in different environments, which were due to weak expression of QTL, QTL by environment interaction in the opposite direction to a effects, and/or epistasis. The markers identified in multi-environments in this study could be applied in the selection of soybean cultivars for higher isoflavone content and in the map-based gene cloning.

Kir channels display voltage-dependent block by cytosolic cations such as Mg2+ and polyamines that causes inward rectification. In fact, cations can regulate K channel activity from both the extracellular and intracellular sides. Previous studies have provided insight into the up-regulation of Kir channel activity by extracellular K+ concentration. In contrast, extracellular Mg2+ has been found to reduce the amplitude of the single-channel current at milimolar concentrations. However, little is known about the molecular mechanism of Kir channel blockade by external Mg2+ and the relationship between the Mg2+ blockade and activity potentiation by permeant K+ ions. In this study, we applied an interactive approach between theory and experiment. Electrophysiological recordings on Kir2.2 and its mutants were performed by heterologous expression in Xenopus laevis oocytes. Our results confirmed that extracellular Mg2+ could reduce heterologously expressed WT Kir2.2 currents in a voltage dependent manner. The kinetics of inhibition and recovery of Mg2+ exhibit a 3∼4s time constant. Molecular dynamics simulation results revealed a Mg2+ binding site located at the extracellular mouth of Kir2.2 that showed voltage-dependent Mg2+ binding. The mutants, G119D, Q126E and H128D, increased the number of permeant K+ ions and reduced the voltage-dependent blockade of Kir2.2 by extracellular Mg2+.

Photoperiod and sucrose levels play a key role in the control of flowering. GmRAV reflected a diurnal rhythm with the highest expression at 4 h after the beginning of a dark period in soybean leaves, and was highly up-regulated under short-day (SD) conditions, despite of not following a diurnal pattern under long-day (LD) conditions. GmRAV-i (GmRAV-inhibition) transgenic soybean exhibited early flowering phenotype. Two of the FT Arabidopsis homologs, GmFT2a and GmFT5a, were highly expressed in the leaves of soybeans with inhibition (-i) of GmRAV under SD conditions. Moreover, the transcript levels of the two FT homologs in GmRAV-i soybeans were more sensitive to SD conditions than LD conditions compared to the WT plant. GmRAV-i soybeans and Arabidopsis rav mutants showed more sensitive hypocotyl elongation responses when compared with wild-type seedlings, and GmRAV-ox overevpressed in tobacco revealed no sensitive changes in hypocotyl length. These indicated that GmRAV was a novel negative regulator of SD-mediated flowering and hypocotyl elongation. Although sucrose has been suggested to promote flowering induction in many plant species, high concentration of sucrose (4% [w/v]) applied into media defer flowering time in Arabidopsis wild-type and rav mutant. This delayed flowering stage might be caused by reduction of LEAFY expression. Furthermore, Arabidopsis rav mutants and GmRAV-i soybean plants were less sensitive to sucrose by the inhibition assays of hypocotyls and roots growth. In contrast, transgenic GmRAV overexpressing (-ox) tobacco plants displayed more sensitivity to sucrose. In conclusion, GmRAV was inferred to have a fundamental function in photoperiod, darkness, and sucrose signaling responses to regulate plant development and flowering induction.

The treated distillate aromatic extract (TDAE) is widely used in tread compounds. However, polycyclic aromatic hydrocarbons (PAHs) contained in TDAE is serious harm to the environment and human. In this paper, liquid polybutadiene (LPB) of different brands was used to replace TDAE to plasticize solution polymerized styrene-butadiene rubber/cis-1,4-polybutadiene rubber (SSBR/BR). The effects of liquid polybutadiene instead of TDAE on crosslinking network and mechanical properties of matrix rubber were studied. LPB could participate in vulcanization reaction (different from TDAE). The results showed that LPB effectively reduced the critical strain amplitude of Payne effect (ameliorated filler dispersion) compared with TDAE plasticizing SSBR/BR. The degree of crosslinking decreased slightly, but there was no significant difference in rolling resistance and wet skid resistance at the same sulfur content. Changing the sulfur content, the wet skid resistance of tread and tensile strength of SSBR/BR vulcanizate plasticized by LPB were significantly improved, and the effect of LPB with high 1,2-vinyl content was better. In a word, LPB as a new environmental protection plasticizer can be used to effectively replace the TDAE plasticizing SSBR/BR. This study delves into the transformative potential of low-carbon leisure education (LCLE) in promoting sustainable development within rural lake regions, focusing particularly on its impact on youth well-being and regional economic vitality. Utilizing a mixed-methods approach, we surveyed 650 participants and conducted interviews with experts, visitors, and locals to gauge the effectiveness of LCLE initiatives. The findings indicate a significant positive correlation between LCLE engagements and improvements in the leisure environment, economic development, and ecological conservation. However, the study reveals a youth welfare divide, whereby younger individuals are unable to fully benefit from LCLE programs due to gaps in environmental consciousness and accessibility. The paper proposes a holistic approach to LCLE that includes enhancing environmental awareness among youth, making participation in low-carbon leisure activities more accessible and affordable, and fostering a culture of sustainable enjoyment. The expected outcomes suggest strengthened rural economies, enriched leisure and ecological environments, and a healthier, more sustainable lifestyle for the younger population. This research underscores the importance of integrating educational initiatives into rural development strategies, thereby contributing to the knowledge economy through innovative approaches to sustainability and youth engagement in rural settings.
