Objective. To investigate synergic therapeutic effects of combined injection of intralesional mesenchymal stem cells derived from human umbilical cord blood (UCB-MSCs) and polydeoxyribonucleotide (PDRN) combined with microcurrent therapy (MIC) on full thickness rotator cuff tendon tear (FTRCTT) in rabbit models. Methods. Thirty-two rabbit models were assigned to 4 different groups. FTRCTT in the supraspinatus tendon was created. After 6 weeks, 4 types of procedures (0.2 mL normal saline injection, group 1 (G1-NS); 0.2 mL SC injection, group 2 (G2-MSC); 0.2 mL SC and weekly four injections of 0.2 mL PDRN with sham MIC, group 3 (G3-MSC+PDRN+sham MIC); and 0.2 mL SC and weekly four injections of 0.2 mL PDRN with MIC for four weeks, group 4 (G4-MSC+PDRN+MIC)) were performed in FTRCTT. Gross morphologic and histological changes of proliferating cell nuclear antigen (PCNA), vascular endothelial growth factor (VEGF) and platelet endothelial cell adhesion molecule (PECAM-1) and motion analysis were performed. Results. There was a significant difference in gross morphologic changes between baseline and week 4 posttreatment in group 4 compared to the other three groups ( p = 0.01 ). In groups 3 and 4, all parameters of histochemical and motion analysis have been found to be significantly greater than the ones in groups 1 and 2 ( p < 0.05 ). In group 4, PCNA-, VEGF-, and PECAM-1-stained cells, as well as walking distance, were significantly greater than the ones in group 3 ( p < 0.05 ). Conclusion. The treatment with UCB-MSCs and PDRN combined with MIC might be the most effective in rabbit models’ traumatic FRTCTT.

Objectives The purpose of this animal research was to compare bone regeneration in augmented rabbit maxillary sinuses treated with demineralized particulate human-tooth graft and anorganic bovine bone by immunohistochemical analysis. Materials and methods Piezoelectric bilateral sinus augmentation was performed in eight adult rabbits. In the control group, anorganic bovine was grafted in the maxillary sinus following elevation of the sinus membrane. In the experimental group, demineralized human particulate tooth bone was grafted in the sinus. Bone regeneration in augmented sinuses was evaluated by immunohistochemical analysis using various markers of osteoprogenitor cells. Results The number of bromodeoxyuridine-labeled cells was significantly higher in the experimental group than in the control group at eight weeks. The immunoreactivity of proliferating-cell nuclear antigen was increased slightly in the experimental group relative to the control group at eight weeks. Other bone markers were expressed equally in the two groups. Conclusion In the rabbit maxillary sinus, higher osteoinduction was correlated with demineralized human particulate tooth bone grafting than with anorganic bovine grafting.

Background: Chronic full-thickness rotator cuff tears (FTRCTs) represent a major clinical concern because they show highly compromised healing capacity. Purpose: To evaluate the efficacy of using a 3-dimensional (3D) bioprinted scaffold with human umbilical cord blood (hUCB)–mesenchymal stem cells (MSCs) for regeneration of chronic FTRCTs in a rabbit model. Study Design: Controlled laboratory study. Methods: A total of 32 rabbits were randomly assigned to 4 treatment groups (n = 8 per group) at 6 weeks after a 5-mm FTRCT was created on the supraspinatus tendon. Group 1 (G1-SAL) was transplanted with normal saline. Group 2 (G2-MSC) was transplanted with hUCB-MSCs (0.2 mL, 1 × 106) into FTRCTs. Group 3 (G3-3D) was transplanted with a 3D bioprinted construct without MSCs, and group 4 (G4-3D+MSC) was transplanted with a 3D bioprinted construct containing hUCB-MSCs (0.2 mL, 1 × 106 cells) into FTRCTs. All 32 rabbits were euthanized at 4 weeks after treatment. Examination of gross morphologic changes and histologic results was performed on all rabbits after sacrifice. Motion analysis was also performed before and after treatment. Results: In G4-3D+MSC, newly regenerated collagen type 1 fibers, walking distance, fast walking time, and mean walking speed were greater than those in G2-MSC based on histochemical and motion analyses. In addition, when compared with G3-3D, G4-3D+MSC showed more prominent regenerated tendon fibers and better parameters of motion analysis. However, there was no significant difference in gross tear size among G2-MSC, G3-3D, and G4-3D+MSC, although these groups showed significant decreases in tear size as compared with the control group (G1-SAL). Conclusion: Findings of this study show that a tissue engineering strategy based on a 3D bioprinted scaffold filled with hUCB-MSCs can improve the microenvironment for regenerative processes of FTRCT without any surgical repair. Clinical Relevance: In the case of rotator cuff tear, the cell loss of the external MSCs can be increased by exposure to synovial fluid. Therefore, a 3D bioprinted scaffold in combination with MSCs without surgical repair may be effective in increasing cell retention in FTRCT.

This study aimed to examine the synergic effects of polydeoxyribonucleotide (PDRN) through extracorporeal shock wave therapy (ESWT) on atrophied calf muscles in cast-immobilized rabbit models.Twenty male New Zealand rabbits (aged 12 weeks) were allocated into four groups. Four types of procedures [0.7 mL normal saline to Group 1 (G1-NS); 0.7 mL PDRN to Group 2 (G2-PDRN); ESWT to Group 3 (G3-ESWT); and 0.7 mL PDRN with ESWT to Group 4 (G4-PDRN + ESWT)] were injected to the atrophied calf muscles of the rabbits after two weeks of cast immobilization. Background. The aim of this study was to evaluate how polydeoxyribonucleotide (PDRN) and microcurrent therapy (MT) functioned synergistically in a cast-immobilized rabbit model with an atrophied calf muscle. Methods. At the age of 12 weeks, 32 male New Zealand rabbits were enrolled in four groups. After 2 weeks of cast-immobilization, 4 procedures were performed on atrophied calf muscle [weekly two injections normal saline 0.2 ml injection group 1 (G1-NS), weekly two injections 0.2 ml PDRN injection group 2 (G2-PDRN), MT group 3 (G3-MT), and 0.2 ml PDRN injection with MT group 4 (G4-PDRN+MT)]. For 2 weeks, MT was used for 60 minutes each day. The calf circumference (CC), the thickness of gastrocnemius muscle (TGCM), and the tibial nerve compound muscle action potential (CMAP) were evaluated using ultrasound before and after 2 weeks of treatment. Proliferating cell nuclear antigen (PCNA), vascular endothelial growth factor, and platelet endothelial cell adhesion molecule-1 (PECAM-1) of GCM fibers (type I, type II, and total) were measured. Statistical analyses were performed using ANOVA. Results. The mean atrophic alterations of right CC, CMAP, and TGCM (medial/lateral) were substantially lower in G4-PDRN+MT than in the G1-NS, G2-PDRN, and G3-MT, respectively (p<0.05). Furthermore, mean CSAs (type I, type II, and total) of medial and lateral GCM muscle fibers in G4-PDRN+MT were significantly higher when compared to other three groups (p<0.05). In terms of the PCNA-, VEGF-, and PECAM-1-positive cell ratio of medial and lateral GCM muscle fibers, G4-PDRN+MT was considerably higher than G1-NS, G2-PDRN, and G3-MT (p<0.05). Conclusions. On the atrophied calf muscle of the rabbit model, PDRN injection combined with MT was more effective than PDRN injection alone, MT alone, and normal saline injection separately.

PURPOSE The aim of this animal study was to evaluate new bone formation in human dentin block grafted on rabbit calvaria according to a comparison of histologic analysis. MATERIALS AND METHODS Human teeth were prepared according to four different types of dentin blocks: group 1, demineralized and microperforated dentin block; group 2, demineralized dentin block; group 3, undemineralized and microperforated dentin block; group 4, undemineralized dentin block. These four different dentin blocks were grafted on nine rabbit calvaria, and animals were sacrificed at 2, 4, and 8 weeks after the surgical procedure for histologic evaluation. RESULTS In group 1, histologically, new bone formation was initiated at the interface between demineralized and microperforated dentin block and host bone and microholes at 2 weeks, and mature bone was observed at 8 weeks. In group 3, new bone formation was observed at 8 weeks in the undemineralized and microperforated dentin block bottom and microholes. The bone formation ratio of group 1 was significantly higher at 2, 4, and 8 weeks compared with groups 2, 3, and 4 (P < .05). The bone formation ratio in microholes at 2 and 8 weeks in group 1 was significantly greater than in group 3 (P < .05). CONCLUSION New bone formation of the demineralized dentin block began more rapidly than the undemineralized dentin block, and perforated dentin block was more effective in bone formation than dentin block without microholes.

While therapies using mesenchymal stem cells (MSCs) to treat rotator cuff tendon tear (RCTT) have yielded some promising preliminary results, MSCs therapy has not yet completely regenerated full-thickness RCTT (FTRCTT). It has recently been reported that polydeoxyribonucleotide (PDRN) is effective in the treatment of chronic rotator cuff disease. We hypothesized that local injection of human umbilical cord blood-derived (UCB)–MSCs with PDRN would be more effective in regenerating tendon tear than UCB-MSCs alone. The purpose of this study was to evaluate the effects of UCB–MSCs combined with different doses of PDRN on the regeneration of RCTT in a chronic RCTT model by using a rabbit model. New Zealand white rabbits ( n = 24) with FTRCTT were allocated randomly into three groups (8 rabbits per group). Three different injectants (G1-S, 0.2 mL UCB-MSCs; G2-P1, 0.2 mL UCB-MSCs with one injection of 0.2 mL PDRN; G3-P4, 0.2 mL UCB-MSCs, and four injections of 0.2 mL PDRN per week) were injected into FTRCTT under US-guidance. After the rabbits were euthanized, we evaluated ross morphological and histological change. Motion analysis was also performed. There were significant differences in gross morphological changes between before, and at 4 weeks after injection, in all three groups, but no differences were found among the three groups. Masson’s trichrome (MT) or anti-type 1 collagen antibody (COL-1)-positive cell densities in G2-P1 and G3-P4 were improved significantly compared with those in G1-S, but showed no significant difference between G2-P1 and G3-P4. On motion analysis, walking distance and fast walking time in G2-P1 and G3-P4 were significantly longer/higher than those in G1-S, but showed no significant differences between G2-P1 and G3-P4. These results demonstrated that there was no significant difference in the gross morphologic change of tendon tear between UCB-MSCs only and combination with PDRN injection in rabbit model of chronic traumatic FTRCTT. The purpose of this study was to investigate the intensity-specific regenerative effects of microcurrent therapy on gastrocnemius muscle atrophy induced by cast-immobilization in rabbits. Fifteen rabbits were randomly allocated to 3 groups after cast removal: cast-immobilization and sham microcurrent therapy for 2 weeks (group 1); cast-immobilization and microcurrent therapy (25 μA) for 2 weeks (group 2); cast-immobilization and microcurrent therapy (5,000 μA) for 2 weeks (group 3). Clinical parameters [calf circumference, compound muscle action potential (CMAP) of the tibial nerve, thickness of gastrocnemius muscle], cross sectional area of gastrocnemius muscle fibres, and immunohistochemistry was evaluated. The clinical parameters representing mean atrophic changes in group 2 were significantly lower than those in group 3. The cross sectional area of the gastrocnemius muscle fibres and immunohistochemical parameters in group 2 were significantly greater than those in group 3. The results showed that low-intensity microcurrent therapy can more effectively promote regeneration in atrophied gastrocnemius muscle than high-intensity microcurrent therapy.

After preparation of replaceable bony windows on the lateral wall of the nasal cavity with a piezoelectric surgical device, deproteinized bovine graft (Bio-Oss) was grafted in the new compartment of the maxillary sinus after elevation of the sinus membrane in the control group. In the experimental group, the demineralized human particulate tooth bone was grafted in the sinus. The replaceable bony window was repositioned over the bone graft in both groups. Animals were killed at 2 and 8 weeks after the surgical procedure. The augmented sinuses were evaluated by histomorphometric analysis using hematoxylin-eosin and Masson trichrome stains. Results Histologically, new bone was revealed along the elevated sinus membrane and both bone grafts. In the control group, the new bone area at 8 weeks was not significantly different than that at 2 weeks. In the experimental group, the new bone area at 8 weeks was significantly greater than that at 2 weeks. Conclusion Significant higher new bone formation was revealed in the experimental group than in the control group.

Background Rotator cuff tendon tears (RCTTs) are common adult injuries. We hypothesized that a local injection of polydeoxyribonucleotide (PDRN) and microcurrent therapy (MIC) would be more effective in regenerating a tendon tear than PDRN administration alone. Objectives To evaluate the effect of PDRN combined with MIC on the regeneration of RCTTs in a rabbit subscapularis tendon chronic RCTT model. Methods Rabbits (n = 24) were allocated to 3 groups at 6 weeks after full-thickness RCTT (FTRCTT): 0.2 mL normal saline (G1-SAL); 0.2 mL PDRN with Sham MIC (G2-PDRN + Sham MIC); and 0.2 mL PDRN with MIC (G3-PDRN + MIC). All treatments were performed under ultrasound guidance. PDRN was injected weekly for 4 weeks and sham MIC or MIC was applied daily for 4 weeks after the first PDRN injection. Results In the G3-PDRN + MIC group, the mean (SD) subscapularis tendon tear size was continuously reduced from 1 week post-treatment to 4 weeks and was significantly decreased as compared with the other 2 groups [6.0 (1.5) vs. G1: 11.5 (1.8) and G2: 9.1 (1.6) mm2; G3 vs. G1, P Conclusions On gross morphologic, histological, and motion analysis, combined PDRN with MIC therapy was more effective than PDRN alone treating a rabbit model of chronic traumatic FTRCTT.

The purpose of this study was to investigate the structure and secretory function of the von Ebner’s gland in parasympathetic or sympathetic nerve innervation. Sprague Dawley rats were sacrificed 3, 7, 10, 14, and 21 days after bilateral glossopharyngeal or hypoglossal nerve axotomy, respectively. The circumvallate papilla portion of the tongue was dissected and we observed morphological changes in the von Ebner’s gland. The properties of glycoconjugate in the von Ebner’s gland were investigated using nine biotinylated lectins (PSA, UEA I, GSL I B4, ECL, DBA, SBA, HPA, SJA, or sWGA). Compared with the control group, cytoplasmic vacuoles appeared in the serous acini of the von Ebner’s gland in the 3-day group, and the serous acini were significantly vacuolized and degenerated in the 10-day group after glossopharyngeal nerve axotomy. However, the structure of the von Ebner’s gland did not change after hypoglossal nerve axotomy. In the control group, the von Ebner’s glands secreted glycoconjugates containing α-D-galactose, N-acetyl-D-galactosamine, and N-acetyl-D-glucosamine oligomer, and the amount of the secretion decreased significantly in the 10-day group after glossopharyngeal nerve axotomy. However, the amount of the glycoconjugate secretion did not change after hypoglossal nerve axotomy. Therefore, the results of this study suggest that the glossopharyngeal nerve containing parasympathetic nerve fibers is important for maintaining the structure of and secretory function in the von Ebner’s gland in rats.

The genioglossus muscle plays an important role in maintaining upper airway patency during inspiration; if this muscle does not contract normally, breathing disorders occur due to closing of the upper airway. These occur because of disorders of synaptic input to the genioglossus motoneurons, however, little is known about it. In this study, the distribution of GABA-, glycine-, and glutamate-like immunoreactivity in axon terminals on dendrites of the rat genioglossus motoneurons, stained intracellularly with horseradish peroxidase (HRP), was examined by using postembedding immunogold histochemistry in serial ultrathin sections. The motoneurons were divided into four compartments: the soma, and primary (Pd), intermediate (Id), and distal dendrites (Dd). Quantitative analysis of 157, 188, 181, and 96 boutons synapsing on 3 soma, 14 Pd, 35 Id, and 28 Dd, respectively, was performed. 71.9% of the total number of studied boutons had immunoreactivity for at least one of the three amino acids. 32.8% of the total number of studied boutons were immunopositive for GABA and/or glycine and 39.1% for glutamate. Among the former, 14.2% showed glycine immunoreactivity only and 13.3% were immunoreactive to both glycine and GABA. The remainder (5.3%) showed immunoreactivity for GABA only. Most boutons immunoreactive to inhibitory amino acids contained a mixture of flattened, oval, and round synaptic vesicles. Most boutons immunoreactive to excitatory amino acids contained clear and spherical synaptic vesicles with a few dense-cored vesicles. When comparisons of the inhibitory and excitatory boutons were made between the soma and three dendritic segments, the proportion of the inhibitory to the excitatory boutons was high in the Dd (23.9% vs. 43.8%) but somewhat low in the soma (35.7% vs. 38.2%), Pd (34.6% vs. 37.8%) and Id (33.1% vs. 38.7%). The percentage of synaptic covering of the inhibitory synaptic boutons decreased in the order of soma, Pd, Id, and Dd, but this trend was not applicable to the excitatory boutons. The present study provides possible evidence that the spatial distribution patterns of inhibitory and excitatory synapses are different in the soma and dendritic tree of the rat genioglussus motoneurons.

Rationale Carpal tunnel syndrome (CTS) is the most common peripheral nerve neuropathy resulting from compression of the median nerve as it traverses the carpal tunnel. The pathophysiology of this condition is multifactorial, and majority of cases of CTS are idiopathic. We report cases of CTS caused by synovial osteochondromatosis (SOC), which has rarely been reported. Patient concerns A 45-year-old female was admitted to the clinic due to right hand tingling sensation for 4 months. On physical examination, the patient's symptoms and signs corresponded to the median nerve entrapment at wrist. However, there is mild swelling and tenderness around the second metacarpal bone. Pain was aggravated during wrist and finger flexion. Diagnoses An electrodiagnostic study revealed CTS. She was advised to begin splinting the hand using a wrist brace and to undergo physiotherapy. After 2 weeks, the tingling sensation decreased slightly. However mild swelling and tenderness around the second metacarpal bone did not improve. Ultrasonography showed multiple echogenic foci. Magnetic resonance imaging (MRI) revealed a nodule at the proximal metacarpal level with synovial thickening, enhancement, and a calcified shadow close to the flexor tendon. After confirming the presence of an osseous nodule with synovial thickening, the patient underwent surgery INTERVENTIONS:: Carpal tunnel release and mass excision with synovectomy of the adjacent structures were performed. Histologically, the lesion was compatible with a diagnosis of SOC. Outcome The symptoms have improved. Lessons CTS due to SOC on finger flexor tendon is rare but should be considered for possible etiology. Appropriate clinical examination, plain radiography, ultrasonography, and MRI will help physicians to diagnose this condition. In this paper, we report the successful diagnosis and treatment of CTS caused by SOC within the finger flexor tendon.

In the group 1, blood clots were filled; group 2, anorganic bovine graft (Bio-Oss) was grafted; group 3, β-TCP was grafted; group 4, DTD was grafted, and covered by replaceable bony windows. Animals were sacrificed at 2, 4, and 8 weeks after surgical procedure. The augmented sinuses were evaluated by histomorphometric analysis using hematoxylin and eosin and Masson's trichrome stains. Histologically, new bone formation was revealed along the elevated sinus membrane and all graft materials. PEG1/MEST gene has been known to be an imprinting gene, which is associated with growth of mesodermal origin cells. Its expression was also reported to be increased in leiomyoma. Several reports showed that loss of imprinting is associated with carcinogenesis in some types of cancer. The purpose of this study was to investigate whether overexpression of PEG1/MEST gene in leiomyoma is associated with loss of imprinting of the gene (biallelic), or whether the overexpression occurs while maintaining the imprinting (monoallelic). We investigated the expression and the imprinting status of PEG1/MEST and its isoforms in samples from 25 patients with uterine leiomyomas as well as in matched normal myometrial tissue. The isoform 1 transcripts were found to be more increased in uterine leiomyomas, compared to myometrium. However, there was no difference in the mRNA levels of isoform 2 between normal myometrium and leiomyoma. All normal myometrial tissues and 19 of 20 leiomyomas showed monoallelic expression of PEG1/MEST. Thus, these data demonstrated that tumorigenesis of leiomyoma is associated with overexpression of isoform 1 of PEG1/MEST gene, but not with loss of imprinting of the gene.

In experimental groups, β-TCP was grafted and covered by replaceable bony windows. The rabbits were killed at 1, 2, 4, and 8 weeks postoperatively. The augmented sinuses were stained with hematoxylin-eosin and Masson trichrome stains and examined, under light microscopy, for newly formed bone and soft tissue changes in the maxillary sinuses. Results Histologically, significantly higher and faster new bone formation was observed in the augmented sinuses of the experimental groups, receiving homologous replaceable bony windows than in those of the control group receiving collagen membranes. Conclusion This study demonstrates that, for augmentation, the use of the homologous replaceable bony window over bone graft material on the maxillary sinus accelerates bone regeneration.

The present study was conducted to elucidate whether the expression level of poly(ADP‐ribose) polymerase (PARP) is related to the ultraviolet radiation (UV)‐induced apoptosis. After treatment of the mammalian cell lines HeLa S3 and Chinese hamster ovary (CHO) with 50 J/m2 UV, induction of apoptosis was determined by several means during 24 h post‐incubation. Incidence of apoptosis was much lower in CHO than HeLa S3 cells based on the percentage of apoptotic cells in terms of morphological changes in nucleus or direct counting of viable cells and qualitative or quantitative DNA fragmentation. Interestingly, when the expression level of PARP was measured by western blotting, the amounts of PARP that was retained at each time point inversely correlated with the incidences of apoptosis in these cells. Background Bee venom has been used to relieve pain and to treat inflammatory diseases, including rheumatoid arthritis, in humans. To better understand the mechanisms of the anti-inflammatory and anti-atherosclerosis effect of bee venom, gel electrophoresis and mass spectrometry were used to identify proteins whose expression was altered in human Vascular Smooth Muscle Cells (hVSMCs) stimulated by tumor necrosis factor alpha after 12 h in the presence of melittin. Results To obtain valuable insights into the anti-inflammatory and anti-atherosclerosis mechanisms of melittin, two-dimensional (2-D) gel electrophoresis and MALDI-TOF/TOF were used. The proteome study, we showed 33 significant proteins that were differentially expressed in the cells treated with tumor necrosis factor alpha and melittin. Thirteen proteins were significantly increased in the cells treated with tumor necrosis factor alpha, and those proteins were reduced in the cells treated with melittin. Five of the proteins that showed increased expression in the cells treated with tumor necrosis factor alpha are involved in cell migration, including calreticulin, an essential factor of development that plays a role in transcription regulation. The proteins involved in cell migration were reduced in the melittin treated cells. The observed changes in the expression of GRP75, prohibitin, and a select group of other proteins were validated with reverse transcribed-PCR. It was confirmed that the observed change in the protein levels reflected a change in the genes level. In addition, the phosphorylation of EGFR and ERK was validated by analyzing the protein pathway. Conclusion Taken together, these data established that the expression of some proteins was significantly changed by melittin treatment in tumor necrosis factor alpha stimulated the cells and provided insights into the mechanism of the melittin function for its potential use as an anti-inflammatory agent.

Non-random simple chromosomal aberrations in various malignancies provide important insights into the molecular pathogenesis of human cancer. Although extensive data exist on recurring chromosomal abnormalities in hematological cancer, data on individual solid tumor types remain limited. Here we present the case of a patient with ovarian cancer with a specific chromosomal abnormality.Cytogenetic analysis utilized a G-banding technique, which was performed with direct culture of the surgically removed cancer cells from a 23-year-old woman with grade II ovarian serous cystadenocarcinoma. The patient had no family history of ovarian cancer.We report a novel der(16)t(3;16)(p25;q24) accompanied by terminal deletion of 3p25 as the simple chromosomal aberration in this case.To the best of our knowledge, no such translocation has been previously reported. The present study supports the possible role of both del(3)(p25) and the translocation t(3;16)(p25;q24) in ovarian cancer; nevertheless, the significance of these chromosomal changes in the development of ovarian cancer remains unknown. The significance of this finding and its role in the pathogenesis of ovarian cancer requires further clarification.

The aim of this animal study was to evaluate, through immunochemical analysis, new bone formation in rabbit maxillary sinuses with and without bone grafts.Bilateral sinus augmentation procedures were performed in rabbits. Bony windows were created with a piezoelectric surgical device with a saw insert. In the first group, the bony window was repositioned after careful elevation of the sinus mucosa without bone grafting. A miniscrew was inserted into the bony window to support the elevated sinus membrane. In the second group, anorganic bovine graft (Bio-Oss) was grafted after very careful elevation of the sinus membrane. Collagen membrane was placed over the bone graft. Rabbits were sacrificed after 1, 2, 4, 6, and 8 weeks. The augmented sinuses were evaluated by immunochemical analysis of proliferating cell nuclear antigen (PCNA), type I collagen, and osteocalcin content.Immunochemically positive cells for PCNA were present along the floor of the replaced bony window and the elevated sinus membrane from 1 to 4 weeks in the graftless group. In the grafted group, immunochemically positive cells for PCNA were not present after 1 week. In the ungrafted group, the expression of type I collagen was present from week 1. However, the expression of type I collagen was present after 2 weeks in the grafted group. Osteocalcin was observed in both groups after 1 week. Osteocalcin was observed along the floor of the replaced bony window in the graftless group but was not observed along the collagen membrane in the grafted group.According to this immunochemical study, faster and greater new bone formation was observed in sites that received no grafting material. The repositioned bony window may accelerate new bone formation earlier during healing versus the placement of a collagen membrane grafting material in the sinus.

Fibrosis is induced by the excessive and abnormal deposition of extracellular matrix (ECM) with various growth factors in tissues. Transforming growth factor-β1 (TGF-β1), the growth factor involved in fibrosis, modulates ECM synthesis and accumulation. TGF-β1 enhances the production of stimulators of ECM synthesis such as plasminogen activator inhibitor type 1 (PAI-1). As such, PAI-1 expression directly influences the proteolysis, invasion, and accumulation of ECM. It was shown in this study that ascochlorin, a prenylpenl antiobiotic, prevents the expression of profibrotic factors, such as PAI-1 and collagen type I, and that the TGF-β1-induced PAI-1 promoter activity is inhibited by ascochlorin. Ascochlorin abolishes the phosphorylation of the EGFR-MEK-ERK signaling pathway to regulate the TGF-β1-induced expression of PAI-1 without the inhibition of TβRII phosphorylation. Furthermore, the MEK inhibitor and EGFR siRNA block PAI-1 expression, and the Raf-1, MEK, and ERK signaling pathways for the regulation of PAI-1 expression. Ascochlorin suppresses the matrix metalloproteinases (MMPs) activity to activate the heparin-binding EGF-like growth factor (HB-EGF), to induce the phosphorylation of EGFR, and the MMPs inhibitor suppresses EGFR phosphorylation and the PAI-1 mRNA levels. These results suggest that ascochlorin prevents the expression of PAI-1 via the inhibition of an EGFR-dependent signal transduction pathway activated by MMPs.

This study was undertaken to evaluate the incidence of bifurcated distal biceps tendons and the tendon's insertional footprint on the radial tuberosity.Twenty-five embalmed cadaveric specimens were dissected. The relationships and orientation of the muscle bellies and distal biceps tendon were examined. The insertional length, width, and footprint area of the distal biceps tendon on the radial tuberosity were evaluated.In 12 specimens (48%), the distal biceps tendon was in 2 distinct, easily separated parts. The average footprint length, width, and area of the tendon's insertion on the radial tuberosity were 20.5 mm ± 2.0 mm, 9.7 mm ± 1.3 mm, and 156.3 mm(2) ± 29.4 mm(2), respectively. We calculated that the tendon's insertion occupied approximately 35.9% of the area of the radial tuberosity. In the specimens with a bifurcated distal biceps tendon, the long head of the tendon inserted at the posterosuperior portion of the radial tuberosity, and the average area was 71.4 mm(2) ± 11.3 mm(2). The short head of the distal biceps tendon inserted at the anteroinferior portion, and the average area was 88.3 mm(2) ± 24.1 mm(2).This study confirmed that bifurcated distal biceps tendon insertion is not a rare anatomical variation, showed by recent investigations, and found that the short head of the distal biceps tendon was inserted more anteriorly than the long head on the radial tuberosity. These findings may allow functional independence and isolated rupture of each portion. It can make correct diagnosis possible and allow for a more anatomical orientation of the tendon during surgical repair.

PEG1/MEST gene has been known to be an imprinting gene, which is associated with growth of mesodermal origin cells. Its expression was also reported to be increased in leiomyoma. Several reports showed that loss of imprinting is associated with carcinogenesis in some types of cancer. The purpose of this study was to investigate whether overexpression of PEG1/MEST gene in leiomyoma is associated with loss of imprinting of the gene (biallelic), or whether the overexpression occurs while maintaining the imprinting (monoallelic). We investigated the expression and the imprinting status of PEG1/MEST and its isoforms in samples from 25 patients with uterine leiomyomas as well as in matched normal myometrial tissue. The isoform 1 transcripts were found to be more increased in uterine leiomyomas, compared to myometrium. However, there was no difference in the mRNA levels of isoform 2 between normal myometrium and leiomyoma. All normal myometrial tissues and 19 of 20 leiomyomas showed monoallelic expression of PEG1/MEST. Thus, these data demonstrated that tumorigenesis of leiomyoma is associated with overexpression of isoform 1 of PEG1/MEST gene, but not with loss of imprinting of the gene.

Cancer cells, characterized by local invasion and distant metastasis, are very much dependent on the extracellular matrix. The expression of matrix metalloproteinases (MMPs) has been implicated in the invasion and metastasis of cancer cells. In this study, we reported the effects of disulfiram, a clinically used anti-alcoholism drug, on tumor invasion suppression, as well as its effects on the activity of MMP-2 and MMP-9 in human osteosarcoma cells (U2OS). Disulfiram has been used for alcohol aversion therapy. However, recent reports have shown that disulfiram may have potential in the treatment of human cancers. Herewith, we showed that the anti-tumor effects of disulfiram, in an invasion assay using U2OS cells and that disulfiram has a type IV collagenase inhibitory activity that inhibits expression of genes and proteins responsible for both cell and non-cell mediated invasion on pathways. In conclusion, disulfiram inhibited expression of MMP-2 and MMP-9 and it regulated the invasion of human osteosarcoma cells. These observations raise the possibility of disulfiram being used clinical for the inhibition of cancer invasion.

To explore the effects of bears' bile on tumor cell p53 protein expression with different gene properties.The effects of bears' bile on the expression of p53 protein in 6 cancer cell strains were determined by Western blot and reverse transcription-polymerase chain reaction (RT-PCR) analysis. Results Western blot analysis showed that the expression of p53 protein in HaCaT, KUMA3, KUMA4 and KUMA6 cell strains with gene mutation were increased, but no change was found in HCT116 and KUMA5 cell strains without gene mutation. We investigated the effects of halogenated aromatic compounds (HACs) including naturally occurring ones (L-thyroxine, 3-chloro-L-tyrosine, 5-chloroindole, 2-chlorophenol, 4-chlorophenol and chlorobenzene) on polychlorinated biphenyl (PCB) dechlorination in sediment cultures. A PCB-dechlorinating enrichment culture of sediment microorganisms from the St. Lawrence River was used as an initial inoculum. When the culture was inoculated into Aroclor 1248 sediments amended with each of the six HACs, the extent of dechlorination was not enhanced by amendment with HACs. The dechlorination patterns in the HAC-amended sediments were nearly identical to that of the HAC-free sediments except the 3-chloro-L-tyrosine-amended ones where no dechlorination activity was observed. When these sediment cultures were transferred into fresh sediments with the same HACs, the dechlorination specificities remained the same as those of the initial inoculations. Thus, in the present study, the substrate range of the highly selected enrichment culture could not be broadened by the HACs. It appears that HACs affect PCB dechlorination mainly through population selection rather than enzyme induction of single population.

The expression of aquaporin-1 (AQP1) protein, a water channel, in the choroid plexus epithelium was examined in the rat to determine the role of AQP1 in the pathophysiology of systemic hyponatremia. Systemic hyponatremia was produced by coadministration of 30 ml hypotonic dextrose solution (140 mM) intraperitoneally and 3 microg 1-deamino-8-d-arginine vasopressin (dDAVP) subcutaneously. This hydration protocol produced severe systemic hyponatremia: 2h after drug administration, normonatremic serum osmolarity and Na(+) concentration (296+/-5mOsm/L and 140+/-5 mM) were reduced to 252+/-5mOsm/L and 117+/-2 mM, respectively. AQP1 expression in the third ventricle choroid plexus epithelium was determined by using immunohistochemistry and quantitative immunoblot analysis. After 2h of systemic hyponatremia, AQP1 expression was significantly increased by 28.2+/-2.7% (n=6, P0.05), whereas it was attenuated to 15.7+/-2.2% (n=6, P0.05) after 6h of systemic hyponatremia. In addition, TUNEL staining revealed apoptotic granule cells in the dentate gyrus (DG) adjacent to the third ventricle with a peak measured 2h after the start of the systemic hyponatremia protocol, while virtually no apoptotic granule cells left 6h after the start of the hyponatremic treatment. These findings suggest that AQP1-mediated excessive water influx into the brain through the blood-cerebrospinal fluid barrier during systemic hyponatremia may elevate the intracranial pressure and result in the apoptotic brain cell death. Thus, AQP1 in the choroid plexus may play an important role in the pathogenesis of systemic hyponatremia.

To support the increasing requirement for additional services demanding very high bit rates and higher system capacity, high speed downlink packet access (HSDPA) is provided in the UMTS standard. Orthogonal frequency division multiplexing (OFDM) is a technology that has been shown to be well suited to HSDPA for high data rate transmission. Selective transmit diversity is a well-known technique to combat the detrimental effects of multipath fading and obtain diversity gain from multiple antennas. We consider transmit selection diversity applied to the OFDM-HSDPA system. The proposed OFDM architecture adopts antenna selection diversity and a novel groupwise subcarrier allocation algorithm in closed-loop mode. In the conventional closed-loop mode, user equipment (UE) should feed back channel quality information (CQI) bits for each associated subcarrier, and there may be a lot of feedback information. We try to reduce the feedback bandwidth compared to previous allocation methods while the system performance should not be degraded by grouping adjacent subcarriers. Through simulation results, we found that the throughput, capacity, coverage and reliability can be dramatically increased by employing the proposed scheme.

The delay estimation algorithms of a synchronous DS-CDMA system are considered under multipath fading channels. A novel maximum-likelihood delay estimation algorithm is addressed for resolving closely spaced delays in indoor wireless communications. The observation space can be partitioned into a signal and a noise subspace by eigenvector decomposition of the time-averaged correlation matrix generated from the output of a sliding correlator without prior knowledge of the signal vectors. Exploiting known statistical properties of the estimated subspaces to maximize the likelihood of the projections forms the maximum-likelihood delay estimation. The simulation results show that the proposed delay estimation algorithm is superior to the conventional sliding correlator and MUSIC-type algorithm.

The oblique projection supports the framework to resolve a signal space into desired signal and interference subspaces. This paper presents subspace-based detection methods using the oblique projection for the CDMA channel. For the synchronous case, it is shown that this detector represents the geometrical form of the decorrelating detector, and performs a complete rejection of interfering signals. This paper also suggests the approach of combining the subspace-based detection with the MUSIC algorithm for an asynchronous CDMA channel. It is shown that the BER performance of this detection approach, which depends on the accuracy of the code timing acquisition, is better than that of the blind adaptive demodulation technique.

This paper presents a robust detection approach in the presence of transmission delay estimation errors in asynchronous CDMA frequency selective Rayleigh fading channels. The transmission delays in asynchronous channels need to be estimated in practice and are subject to estimation errors, which may cause severe performance degradation for most detectors assuming perfect synchronization. As the fundamental framework of the robust detector, an asynchronous subspace detection scheme based on oblique projection is introduced and examined in comparison with the conventional RAKE receiver and the multipath decorrelator. The performance degradation of the subspace detector due to delay estimation errors is also investigated. The robust detector shows reliable performance even when the errors are much larger than those of delay estimation algorithms like MUSIC, and it is not affected by the near-far problem.
