Genetic markers for acne are being studied to create personalized treatments based on an individual’s genes, and the field is benefiting from the application of artificial intelligence (AI) techniques. One such AI tool, the Q-learning algorithm, is increasingly being utilized by medical researchers to delve into the genetics of acne. In contrast to previous methods, our research introduces a Q-learning model that is adaptable to diverse sample groups. This innovative approach involves preprocessing data by identifying differentially expressed genes and constructing gene-gene connectivity networks. The key advantage of using the Q-learning model lies in its ability to transform acne gene data into Markovian domains, which are essential for selecting relevant genetic markers. Performance evaluations of our Q-learning model have shown high accuracy and specificity, although there may be some sensitivity variations. Notably, this research has identified specific genes, such as CD86, AGPAT3, TMPRSS11D, DSG3, TNFRSF1B, PI3, C5AR1, and KRT16, as being acne-related through biological verification and text data mining. These findings underscore the potential of AI-driven Q-learning models to revolutionize the study of acne genetics. In conclusion, our Q-learning model offers a promising approach for the selection of acne-related genetic markers, despite minor sensitivity fluctuations. This research highlights the transformative potential of Q-learning in advancing our understanding of the genetics underlying acne, paving the way for more personalized and effective treatments in the future.

The tiger pufferfish (Takifugu rubripes), also known as fugu, has recently suffered from severe C. irritans infections under aquaculture environment, yet the underlying immune mechanisms against the parasite remain poorly understood. In this study, we conducted a comprehensive transcriptome analysis of the gill tissue from infected and uninfected fish using PacBio long-read (one pooled sample each for seriously infected and healthy individuals, respectively) and Illumina short-read (three pools for mildly infected, seriously infected, and healthy individuals, respectively) RNA sequencing technologies. After aligning sequence data to fugu’s reference genome, 47,307 and 34,413 known full-length transcripts were identified and profiled in healthy and infected fish, respectively. Similarly, we identified and profiled 1126 and 803 novel genes that were obtained from healthy and infected fish, respectively. Interestingly, we found a decrease in the number of alternative splicing (AS) events and long non-coding RNAs (lncRNAs) after infection with C. irritans, suggesting that they may be involved in the regulation of the immune response in fugu. There were 687 and 1535 differentially expressed genes (DEGs) in moderately and heavily infected fish, respectively, compared to uninfected fish. Kyoto Encyclopedia of Genes and Genomes (KEGG) analyses showed that immune-related DEGs in the two comparison groups were mainly enriched in cytokine-cytokine receptor interactions, ECM-receptor interactions, T-cell receptor signaling pathways, Th1 and Th2 cell differentiation, and Th17 cell differentiation pathways. Further analysis revealed that a large number of immune-related genes were downregulated in infected fish relative to uninfected ones, such as CCR7, IL7R, TNFRSF21, CD4, COL2A1, FOXP3B, and ITGA8. Our study suggests that C. irritans is potentially a highly efficient parasite that may disrupt the defense mechanisms of fugu against it. In addition, in combination of short-read RNA sequencing and previous genome-wide association analyses, we identified five key genes (NDUFB6, PRELID1, SMOX, SLC25A4, and DENND1B) that might be closely associated with C. irritans resistance. This study not only provides valuable resources of novel genic transcripts for further research, but also provides new insights into the immune mechanisms underlying C. irritans infection response in farmed fugu.

Objective: Assessing carotid and radial pulses is an essential sphygmology method in traditional Chinese medicine (TCM). It is believed in TCM and by doctors that pulse force is markedly linked with the physiological state of a human, including body mass index (BMI) and temperature. However, comprehensive analysis elucidating these potential correlations remains undetermined. Therefore, this investigation aimed to assess the association of pulse strength with human metabolism. Materials and Methods: The mean blood flow velocity (MBFV) of the carotid artery and pulse force of 122 healthy adults included in this investigation were assessed using ultrasound and manual palpation. Their thermal texture map was also acquired. Results: No substantial variation was observed in the carotid arterial MBFV among normal individuals (P > 0.05) regardless of body side and gender. However, young individuals had higher blood velocity than middle-aged individuals (P < 0.05). Furthermore, it was identified that MBFV was negatively associated with BMI and torso temperature; however, it was positively linked with hand temperature. Conclusions: This investigation revealed that carotid arterial MBFV could indicate the physical state of humans, including BMI and temperature, and therefore, is valuable for elucidating the effect of TCM on yin–yang. The data validated some TCM beliefs and supported the implementation of sphygmology in TCM.

BackgroundAlcohol dependence (AD) is a complex addictive disorder with a high relapse rate. Previous studies have shown that both repetitive transcranial magnetic stimulation (rTMS) and cognitive behavioral therapy (CBT) may be effective for AD, and we aim to explore more effective treatment options to reduce relapse rates for AD.Materials and methodsA total of 263 AD patients were recruited. They were divided into six groups according to the location and the type of rTMS: left dorsolateral prefrontal cortex (DLPFC), right DLPFC, sham stimulation, and whether they received CBT treatment: with a fixed schedule (C1) and without a fixed plan (C0). There were included in sham rTMS + C0 group (n = 50), sham rTMS + C1 group (n = 37), right rTMS + C0 group (n = 45), right rTMS + C1 group (n = 42), left rTMS + C0 group (n = 49), left rTMS + C1 group (n = 40). We used obsessive compulsive drinking scale (OCDS), visual analogue scale (VAS), alcohol dependence scale (ADS), montreal cognitive assessment (MoCA), generalized anxiety disorder-7 (GAD-7), patient health questionnaire-9 items (PHQ-9), and Pittsburgh sleep quality index (PSQI) to assess alcohol cravings, alcohol dependence, cognition, anxiety, depression, and sleep quality. They were followed up and evaluated for relapse.ResultsThe sham rTMS + C0 group relapse rate was significantly higher than the right rTMS + C1 group (P = 0.006), the left rTMS + C0 group (P = 0.031), the left rTMS + C1 group (P = 0.043). The right rTMS + C0 group showed significantly higher relapse rate compared to the right rTMS + C1 group (P = 0.046). There was no significant difference in relapse rates between other groups. The repeated-measures ANOVA showed an interaction effect between group and time was significant in the rate of patient health questionnaire-9 items (PHQ-9) scale reduction (P = 0.020). Logistic analysis indicated that smoking and alcohol consumption were independent determinants of relapse (P < 0.05). At 24 weeks of follow-up, Kaplan–Meier survival analysis reveal that there is statistically significant relapse rate between six groups (P = 0.025), left rTMS + C1 group has the best treatment effect for alcohol dependent patients. Cox regression analysis confirmed that current smoking, total cholesterol, and total bilirubin (TBIL) level were risk factors of relapse (P < 0.05).ConclusionThis study is the first to suggest that the combination of rTMS and CBT may be a potentially effective treatment for reducing relapse.

Ciliates are unique in having nuclear dimorphism (i.e., possessing both a germline micronucleus and a somatic macronucleus in one cell), and undergo dramatic changes during their sexual process (known as conjugation). Here we investigate the nuclear events and morphogenetic processes during conjugation in the freshwater species Euplotes aediculatus. In addition, three conjugation-related pheromone genes were sequenced and analyzed. The results indicate that: (i) the 65-hour-long conjugation process includes five nuclear divisions, i.e., three prezygotic divisions (mitosis, meiosis I and II) and two postzygotic divisions; (ii) the ciliature reorganizes twice during conjugation with the frontoventral-transverse cirral anlagen regenerating during both morphogenetic events and breaking apart in a 3:3:3:2:2 pattern; and (iii) each of three pheromone genes contains a 444 bp coding region and encodes 147 amino acid polypeptides. These results suggest a broadly shared morphogenetic pattern during conjugation in Euplotes species and provide new information for studies on the evolutionary history of pheromone genes.

Species of the ciliate genus Spirostomum Ehrenberg, 1834 are distributed worldwide and have a research history spanning more than two centuries. However, species delimitation and phylogenetic relationships within this genus are still uncertain due to the paucity of stable morphologic characters for species separation and the unavailability of accompanying morphological data for most molecular sequences in public databases. In the present study, S. yagiui Shigenaka, 1959 (three populations) and S. caudatum (Müller, 1786) Delphy, 1939 (one population) were investigated using morphological and molecular methods for the first time in China. Detailed morphological data for the two species were documented, and improved diagnoses were supplied based on a combination of previous studies and the current work. It should be highlighted that there were three different atypical morphotypes identified in a Ningbo population of S. yagiui which may represent various stages in conjugative reproduction. Molecular phylogenies based on 18S, ITS1-5.8S-ITS2, and 28S rRNA gene sequences show that the genus Spirostomum is monophyletic, however, the internal relationships inferred from different genes were poorly resolved but suggest that the species with a moniliform macronucleus comprise an early-diverging clade within this genus. Finally, the global distribution of Spirostomum is summarized based on previous and present studies.

BackgroundLong-term excessive use of morphine leads to addictive diseases and affects cognitive function. Cognitive performance is associated with genetic characteristics.MiR-124 plays a critical regulatory role in neurogenesis, synaptic development, brain plasticity, and the use of addictive substances. As a scaffold protein, IQGAP1 affects learning and memory dose-dependent. However, the role of miR-124 and its target protein as potential addiction biomarkers and the impact on cognitive function have not been fully explored.MethodA total of 40 patients with morphine dependence and 40 cases of healthy people were recruited. We collected basic and clinical information about the two groups. The Generalized Anxiety Disorder Scale (GAD-7), Patient Health Questionnaire-9(PHQ-9), Montreal Cognition Assessment Scale (MoCA), Pittsburgh Sleep Quality Index (PSQI) were used to assess the severity of depression, anxiety, depressive symptoms, cognitive dysfunction, and sleep quality.ResultsCompared to the control group, the morphine-dependent group had higher GAD-7, PHQ-9, PSQI scores, and more elevated miR-124 levels but lower MOCA scores and IQGAP1 levels. MiR-124, IQGAP1, the average intake last year were related to OASI scores.MiR-124, IQGAP1, PHQ-9 were associated with MOCA scores. In the multiple regression model, the levels of miR-124 and IQGAP1 were independent factors influencing the severity of morphine dependence. The level of miR-124 was an independent factor influencing the severity of cognitive impairment in patients with morphine dependence. In addition, the luciferase report confirmed that IQGAP1 mRNA is the direct target of miR-124.ConclusionMiR-124 and its target protein IQGAP1 are involved in the regulation of addiction and cognitive function in patients with morphine dependence.

Abstract Background Heterotrichous ciliates are common members of microeukaryote communities which play important roles in both the transfer of material and the flow of energy in aquatic food webs. This group has been known for over two centuries due to their large body size and cosmopolitan distribution. Nevertheless, species identification and phylogenetic relationships of heterotrichs remain challenging due to the lack of accurate morphological information and insufficient molecular data. Results The morphology and phylogeny of two heterotrichous ciliates, namely Gruberia foissneri spec. nov. and Linostomella vorticella (Ehrenberg, 1833) Aescht in Foissner et al., 1999, were studied using rigorous methods (living morphology, stained preparations, and small subunit rDNA sequence data). Gruberia foissneri spec. nov. is morphologically very similar to G. uninucleata Kahl, 1932, however, it can be distinguished from the latter by having more ciliary rows (about 32 vs. about 20) and macronuclear shape (sausage-shaped vs. ellipsoid). Based on a combination of previous and present studies, an improved diagnosis of L. vorticella is supplied and several taxonomic anomalies are clarified. In addition, phylogenetic analyses based on SSU rDNA sequence data support the generic assignment of these two species. Conclusions Modern ciliate taxonomy should be performed by means of detailed living observation, stained preparations and molecular information. For those species that have been reported in previous studies, it is necessary to provide as much useful information as possible using state-of-the-art methods in order to resolve taxonomic anomalies.

Members of the genus Paradileptus are apex predators in microbial food webs. They are often encountered in freshwater biotopes and have been used in research on water quality monitoring and ecology. Nevertheless, our understanding of the biodiversity of Paradileptus, especially its ecological and genetic diversities, is very poor which hinders our ability to understand the ecosystem services it provides. The present study gives a detailed account of two Chinese populations of Paradileptus elephantinus and P. conicus including their living morphology, infraciliature, and molecular phylogenies based on 18S, 5.8S, and ITS ribosomal DNA sequences. The phylogenetic relationships between these two species and other rhynchostomatians are investigated. We also explore the potential contribution of differentiation of the proboscis (e.g., extrusomes, dorsal brush, and differentiated kineties) to niche partitioning and speciation in Paradileptus. The global distribution of Paradileptus is summarized based on published data. Finally, a key to the identification of the valid species of Paradileptus is provided.

During an investigation on freshwater peritrichs, a new colonial sessilid ciliate, Campanella sinica n. sp., was isolated from aquatic plants in an artificial freshwater pond in Qingdao, China. Specimen observations of this species were performed both in vivo and using silver staining. C. sinica n. sp. is characterized by the appearance of the mature colony, which is up to 2 cm high and contains more than 1,000 zooids, the asymmetric horn-shaped zooids, strongly everted and multi-layered peristomial lip, the slightly convex peristomial disc, and the well-developed haplokinety and polykinety, which make more than four circuits of the peristome before descending into the infundibulum. The small subunit ribosomal DNA (SSU rDNA), 5.8s rDNA and its flank internal transcribed spacers (ITS1-5.8s rDNA-ITS2), and large subunit ribosomal DNA (LSU rDNA) are sequenced and used for phylogenetic analyses which reveal that the family Epistylididae Kahl, 1933 is non-monophyletic whereas the genus Campanella is monophyletic and nests within the basal clade of the sessilids. The integrative results support the assertion that the genus Campanella represents a separate lineage from other epistylidids, suggesting a further revision of the family Epistylididae is needed. We revise Campanella including the transfer into this genus of a taxon formerly assigned to Epistylis, which we raise to species rank, i.e., Campanella ovata (Nenninger, 1948) n. grad. & n. comb. (original combination Epistylis purneri f. ovataNenninger, 1948). In addition, we provide a key to the identification of the species of Campanella.

A long-standing dilemma for microbial analyses is how to handle and store samples, as it is widely assumed that the microbial diversity and community patterns would be affected by sample storage conditions. However, it is quite challenging to maintain consistency in field sampling, especially for water sample collection and storage. To obtain a comprehensive understanding of how sample storage conditions impact microbial community analyses and the magnitude of the potential storage effects, freshwater samples were collected and stored in bottles with lid closed and without lid at room temperature for up to 6 days. We revealed the dynamics of prokaryotic and eukaryotic microbial communities under different storage conditions over time. The eukaryotic microbial communities changed at a faster rate than the prokaryotic microbial communities during storage. The alpha diversity of the eukaryotic microbial communities was not substantially influenced by container status or storage time for up to 12 h, but the beta diversity differed significantly between the control and all treatment samples. By contrast, no significant changes of either the alpha or beta diversity of the prokaryotic microbial communities were observed within 12 h of room-temperature storage, regardless of the container status. The potential interactions between microbial taxa were more complex when samples were stored in sealed bottles, and the deterministic processes played an increasingly important role in shaping the freshwater microbial communities with storage time. Our results suggest that water samples collected and stored without refrigeration for no more than 12 h may still be useful for downstream analyses of prokaryotic microbial communities. If the eukaryotic microbial communities are desired, storage of water samples should be limited to 3 h at room temperature.

Morphogenesis is an important process that widely occurs in almost all the organisms, including the ciliated protists. Ciliates are a large group of single-celled eukaryotes that can reproduce asexually (e.g., binary fission) and perform sexual process (e.g., conjugation). Morphogenesis happens in both asexual reproduction and sexual process in ciliates and the reorganization during conjugation is more complex. However, studies of morphogenesis focusing on conjugation are very limited. Here we studied the morphogenetic process during conjugation in the marine species Euplotes raikovi Agamaliev, 1966. The results indicate that: (1) the ciliature in the ventral side reorganizes twice during sexual process, i.e., conjugational and postconjugational reorganization; (2) the adoral zone of membranelles (AZM) is generated de novo in a pouch beneath the cortex during both reorganizations, with the anterior part generated during the first reorganization, while the posterior part formed during the second reorganization; (3) the frontoventral-transverse (FVT) cirri anlagen are formed de novo in both processes with the fragmentation pattern of 2:2:3:3:2; (4) one left marginal cirrus is generated de novo during both reorganizations; and (5) the dorsal ciliature remains intact during the whole process, except that the two caudal cirri originate from the end of the right-most two dorsal kineties during both reorganizations. Comparisons of the morphogenetic process during conjugation demonstrate a considerably stable pattern within Euplotes while the patterns vary dramatically among different ciliate groups.

RAD54 stimulates activity of the RAD51 recombinase and catalyzes branch migration of Holliday junctions during DNA repair and recombination. Here the authors show that the N-terminal domain of RAD54 mediates RAD54 oligomerization to promote branch migration, and is the target of phosphorylation that inhibits oligomerization and branch migration but not RAD51 stimulation.

Ultraviolet (UV) radiation is a carcinogen that generates DNA lesions. Here, we demonstrate an unexpected role for DGCR8, an RNA binding protein that canonically functions with Drosha to mediate microRNA processing, in the repair of UV-induced DNA lesions. Treatment with UV induced phosphorylation on serine 153 (S153) of DGCR8 in both human and murine cells. S153 phosphorylation was critical for cellular resistance to UV, the removal of UV-induced DNA lesions, and the recovery of RNA synthesis after UV exposure but not for microRNA expression. The RNA-binding and Drosha-binding activities of DGCR8 were not critical for UV resistance. DGCR8 depletion was epistatic to defects in XPA, CSA, and CSB for UV sensitivity. DGCR8 physically interacted with CSB and RNA polymerase II. JNKs were involved in the UV-induced S153 phosphorylation. These findings suggest that UV-induced S153 phosphorylation mediates transcription-coupled nucleotide excision repair of UV-induced DNA lesions in a manner independent of microRNA processing.

Effects of metal salt catalysts and gasification temperature on the gasification characteristics of combustible solid waste were studied, based on the source-classified waste in a fluidized bed gasifier, to provide guidance for utilizing the source-classified waste effectively. The results showed that the gasification characteristics of combustible solid waste, such as paper and sawdust, improved noticeably after adding NaCl, K2CO3, or sodium dodecyl benzene sulfonate (LAS). Adding NaCl to sawdust increased the yield of CO and CH4, while the gasification was inhibited gradually with increasing addition. Adding NaCl promoted the generation of H2 in paper gasification. NaCl played a catalytic role only when it exceeded a certain value. Adding K2CO3 increased the yield of H2 noticeably in sawdust and paper gasification. The catalytic effect of K2CO3 on sawdust was better than that on paper. Similar results were obtained for LAS in producing H2 in gasification. The carbon conversion efficiency and the gasification efficiency were increased with additional LAS. Moreover, the catalytic effect of K2CO3 was superior to that of LAS by comparison. As the temperature rises, the activity of the metal salt catalyst is enhanced, but it is inhibited if the temperature is too high.

Catalpol, a natural iridoid glycoside, has potential therapeutic benefits, including anti‐inflammatory and neuroprotective effects. Investigating catalpol's role in angiogenesis is critical for understanding its potential therapeutic applications, particularly in diseases where modulating angiogenesis is beneficial. This study investigates catalpol's influence on angiogenesis and its mechanisms, combining network pharmacology and in vitro experiments. The target genes corresponding to the catalpol were analyzed by SwissTargetPrediction. Then angiogenesis‐related targets were acquired from databases like GeneCards. Subsequently, the Database for Annotation, Visualization and Integrated Discovery was employed for Gene Ontology and pathway analysis, while Cytoscape visualized protein interactions. The effect of catalpol on viability and angiogenesis of HUVECs was further examined using Cell Counting Kit‐8 and angiogenesis assays. RT‐qPCR and western blot were applied to check the expression of angiogenesis‐related proteins. Totally, 312 target genes of catalpol and 823 angiogenesis‐related targets were obtained with 56 common targets leading to PPI network analysis, highlighting hub genes (AKT1, EGFR, STAT3, MAPK3, and CASP3). These hub genes were mainly enriched in lipid and atherosclerosis pathway and EGFR‐related pathway. The in vitro experimental results showed that catalpol achieved a concentration‐dependent increase in HUVECs viability. Catalpol also promoted the migration and angiogenesis of HUVECs and up‐regulated the expression of EGFR. EGFR knockdown inhibited the effect of catalpol on HUVECs. Catalpol promotes angiogenesis in HUVECs by upregulating EGFR and angiogenesis‐related proteins, indicating its potential therapeutic application in vascular‐related diseases. [ABSTRACT FROM AUTHOR]

Placenta interception is extremely rare, and herein, we report the case of a 33-year-old woman with placenta interception during cesarean section caused by carbetocin, which was intravenously injected immediately after delivery of the infant to prevent postpartum hemorrhage. There was no sign of placental separation in the third stage of labor. A protuberance with gross subserous blood vessels in the left fundus of the uterus was detected and was misdiagnosed as placental accreta. The patient was transferred to the district referral hospital to manage the protuberance after stitching up the incision. On opening the original incision in the tertiary hospital, the protuberance disappeared, and the placenta was found in the lower segment of the uterus cavity. The intercepted placenta showed a spontaneous resolution from the uterine wall and was removed intact. This case report details the typical presentation of placenta interception to make obstetricians aware of the possibility that carbetocin might lead to this adverse event. [ABSTRACT FROM AUTHOR]
