Abstract A complete goat (Capra hircus) reference genome enhances analyses of genetic variation, thus providing insights into domestication and selection in goats and related species. Here, we assemble a telomere-to-telomere (T2T) gap-free genome (2.86 Gb) from a cashmere goat (T2T-goat1.0), including a Y chromosome of 20.96 Mb. With a base accuracy of >99.999%, T2T-goat1.0 corrects numerous genome-wide structural and base errors in previous assemblies and adds 288.5 Mb of previously unresolved regions and 446 newly assembled genes to the reference genome. We sequence the genomes of five representative goat breeds for PacBio reads, and use T2T-goat1.0 as a reference to identify a total of 63,417 structural variations (SVs) with up to 4711 (7.42%) in the previously unresolved regions. T2T-goat1.0 was applied in population analyses of global wild and domestic goats, which revealed 32,419 SVs and 25,397,794 SNPs, including 870 SVs and 545,026 SNPs in the previously unresolved regions. Also, our analyses reveal a set of selective variants and genes associated with domestication (e.g., NKG2D and ABCC4) and cashmere traits (e.g., ABCC4 and ASIP).

Abstract Herein we have reported a fluorescent probe (MB-M) based on MB derivative for Cu2+ ions detection. The probe was well characterized by 1H NMR, 13C NMR and HR-MS spectrum. Probe MB-M showed naked-eyes recognition to Cu2+ as color change from colorless to indigo. The probe exhibited promising features such as high fluorescence and UV–vis selectivity, fast response (5 mint), workable at pH 2–7, and low limit of detection (LOD = 0.33 µM). Probe MB-M was also used for Cu2+ ions imaging in HepG-2 cells and detection in daily life (Test Strip and lake water). Moreover, non-covalent interaction (NCI) and quantum theory of atoms in molecules (QTAIM) analysis were used to study the interaction between MB-M and Cu2+ ions. By examining the electronic characteristics of the complex using natural bond orbital (NBO), electron density difference (EDD), and frontier molecular orbital (FMO) analysis, the sensitivity of MB-M towards Cu2+ ions were investigated. The results illustrated that the interactions between MB-M and Cu2+ ions involved chemisorption.

ObjectiveTo investigate the efficacy and potential adverse events of Tuina therapy for idiopathic constipation in children with cerebral palsy (CP).MethodsA total of 60 CP children with idiopathic constipation were enrolled and randomly divided into Tuina and control groups. The control group was treated with basic treatment and 12 sessions of lactulose oral solution, whereas the Tuina group received basic treatment and 12 sessions of infantile Tuina treatment. The following parameters were compared: the Bristol stool form scale (BSFS), the Constipation Assessment Scale (CAS) and the improvement in constipation. In addition, adverse effects were recorded.ResultsAt 4 weeks after the final treatment, the percentage of infants whose constipation improved was 23 (76.7%) in the Tuina group and 21 (70.0%) in the control group (P = 0.771). Initially, the CAS score, weekly bowel movements and proportion of infants with bowel evacuation ≥2 h were comparable between the two groups (P > 0.05). ABSTRACTKlebsiella pneumoniae is a common causative pathogen of intra-abdominal infection with concomitant bacteraemia, leading to a significant mortality risk. The time to positivity (TTP) of blood culture is postulated to be a prognostic factor in bacteraemia caused by other species. Therefore, this study aimed to investigate the prognostic value of TTP in these patients. The single-centred, retrospective, observational cohort study was conducted between 1 July 2016 and 30 June 2021. All adult emergency department patients with diagnosis of intra-abdominal infection and underwent blood culture collection which yield K. pneumoniae during this period were enrolled. A total of 196 patients were included in the study. The overall 30-day mortality rate was 12.2% (24/196), and the median TTP of the studied cohort was 12.3 h (10.5–15.8 h). TTP revealed a moderate 30-day mortality discriminative ability (area under the curve 0.73, p 12 h, N = 109), patients in the early TTP (≤12 h, N = 87) group had a significantly higher risk of 30-day morality (21.8% vs. 4.6%, p

Public servants have faced substantial accountability pressure during the public emergency. It was evident during the pandemic. However, the factors that influence accountability pressure remain to be examined. In this study, we attempt to demonstrate that the Robbins pressure theory provides a useful conceptual framework on how factors affect accountability pressure, and a regression method using primary survey data was applied to investigate how external pressure, the public servants’ capacity, accountability awareness, and communication between superiors and subordinates influence accountability pressure in the fact of a pandemic emergency. The findings show that external pressure had a positive impact on accountability pressure, the public servants’ capacity and communication between superiors and subordinates had a negative impact on accountability pressure, and accountability awareness played a negative moderating role in the effect of external pressure on accountability pressure. This study contributes toward the limited research base, and offers the suggestion that accountability pressure would be alleviated through optimized training and learning.

Abstract Hybrid mapping is a powerful approach to efficiently identify and characterize genes regulated through mechanisms in cis. In this study, using reciprocal crosses of the phenotypically divergent Duroc and Lulai pig breeds, we perform a comprehensive multi-omic characterization of regulatory variation across the brain, liver, muscle, and placenta through four developmental stages. We produce one of the largest multi-omic datasets in pigs to date, including 16 whole genome sequenced individuals, as well as 48 whole genome bisulfite sequencing, 168 ATAC-Seq and 168 RNA-Seq samples. We develop a read count-based method to reliably assess allele-specific methylation, chromatin accessibility, and RNA expression. We show that tissue specificity was much stronger than developmental stage specificity in all of DNA methylation, chromatin accessibility, and gene expression. We identify 573 genes showing allele specific expression, including those influenced by parent-of-origin as well as allele genotype effects. We integrate methylation, chromatin accessibility, and gene expression data to show that allele specific expression can be explained in great part by allele specific methylation and/or chromatin accessibility. This study provides a comprehensive characterization of regulatory variation across multiple tissues and developmental stages in pigs.

Soil temperature is one of the important environmental factors for the underground parts of plants. It is important to detect soil temperature in agricultural production. Acoustic waves serve as effective carriers of soil information, providing a reliable means to detect soil physical properties. In order to detect the temperature of sandy loam soil based on acoustic technology, this study extends the application of Wyllie model to the temperature measurement of sandy loam soil. The relationship between soil temperature and acoustic velocity is explored. A model of soil moisture content-soil temperature-acoustic velocity (MTV) for temperature measurement in sandy loam soil is proposed by extending the temperature–velocity of sound relationship with the introduction of a variable empirical coefficient related to soil moisture β(θ). In order to determine the parameters of proposed model, a pulsed acoustic velocity detection system was built in this paper. The influence of temperature on the acoustic velocity in sandy loam soil was analyzed through experiments. Based on the experimental results, the key parameters of the MTV model are refined. Finally, a validation experiment was carried out on sandy loam soil. The maximum estimation error of soil temperature based on the MTV model for sandy loam soil temperature estimation is 8.55 %. The results indicate that the MTV model proposed in this study can be used for temperature estimation in sandy loam soil, providing a theoretical basis for the development of acoustic soil physical information detection sensors.

Abstract Purpose To identify the key infection processes and risk factors in Computed Tomography (CT) examination process within the standard prevention and control measures for the COVID-19 epidemic, aiming to mitigate cross-infection occurrences in the hospital. Method The case hospital has assembled a team of 30 experts specialized in CT examination. Based on the CT examination process, the potential failure modes were assessed from the perspective of severity (S), occurrence probability (O), and detectability (D); they were then combined with corresponding risk prevention measures. Finally, key infection processes and risk factors were identified according to the risk priority number (RPN) and expert analysis. Results Through the application of RPN and further analysis, four key potential infection processes were identified, including “CT request form (A 1),” “during the scan of CT patient (B 2),” “CT room and objects disposal (C 2),” and “medical waste (garbage) disposal (C 3)”. In addition, eight key risk factors were also identified, including “cleaning personnel does not wear masks normatively (C 32),” “nurse does not select the vein well, resulting in extravasation of the peripheral vein for enhanced CT (B 25),” “patient cannot find the CT room (A 13),” “patient has obtained a CT request form but does not know the procedure (A 12),” “patient is too unwell to continue with the CT scan (B 24),” “auxiliary staff (or technician) does not have a good grasp of the sterilization and disinfection standards (C 21),” “auxiliary staff (or technician) does not sterilize the CT machine thoroughly (C 22),” and “cleaning personnel lacks of knowledge of COVID-19 prevention and control (C 33)”. Conclusion Hospitals can publicize the precautions regarding CT examination through various channels, reducing the incidence of CT examination failure. Hospitals’ cleaning services are usually outsourced, and the educational background of the staff employed in these services is generally not high. Therefore, during training and communication, it is more necessary to provide a series of scope and training programs that are aligned with their understanding level. The model developed in this study effectively identifies the key infection prevention process and critical risk factors, enhancing the safety of medical staff and patients. This has significant research implications for the potential epidemic of major infectious diseases.

Abstract Background This study assessed the impact of pressure-controlled ventilation (PCV) focusing on end-inspiratory flow rate on the incidence of postoperative pulmonary complications (PPCs) and inflammation levels in patients undergoing spinal surgery in the prone position. Methods A total of 187 patients who underwent posterior spinal surgery were enrolled and randomly divided into 3 groups: 61 in the volume-controlled ventilation (VCV) group (group V), 62 in the PCV-volume-guaranteed (VG) group (group P1), and 64 in the PCV-VG end-expiratory zero flow rate group (group P2). Indicators including tidal volume (VT), peak airway pressure (Ppeak), and dynamic lung compliance (Cdyn) were recorded. Abstract The unprecedented growth in voice assistants (VAs) provided with artificial intelligence (AI) challenges managers aiming to harness various new technologies to enhance the competitiveness of their products. This article thus investigates how VAs can more effectively improve the user experience by focusing on the attributes of service contexts, matching a utilitarian-dominant (hedonic-dominant) context with concrete (abstract) language in VA–human interactions. Through such matching, VA companies can potentially create a beneficial congruity effect, leading to more favorable evaluations. The results of three studies therefore suggest that users prefer VAs with abstract language in a hedonic-dominant service context, but that VAs with concrete language are more competitive in a utilitarian-dominant service context. Furthermore, the perception of processing fluency mediates this effect. Accordingly, these findings provide a better understanding of AI–human interactions and open a straightforward path for managers or technology providers to enhance users’ continuous usage intention.

The Aux/IAA (auxin/indole-3-acetic acid) gene family plays a crucial role in regulating various aspects of plant growth, development, and abiotic tolerance in the auxin transduction pathway. However, limited information is available about the Aux/IAA family in Osmanthus fragrans. This study aims to comprehensively analyze the Aux/IAA gene family on a genome-wide scale. A total of 39 OfIAA genes containing four conserved domains were identified. These genes were unevenly distributed across 19 chromosomes and grouped into six clades based on phylogenetic analysis, showing conserved gene structure and motif composition. The expansion of OfIAA genes in the O. fragrans genome was partially due to segmental duplication events. Analysis of cis-regulatory elements (CREs) in the promoters of the OfIAA genes revealed the presence of many CREs related to different hormones and abiotic stresses. Through transcriptome and expression pattern analysis, we found that the majority of OfIAA genes were expressed in the stem tissue. Moreover, during the flower opening process, 18 OfIAA genes exhibited differential expression, while three and 11 OfIAA genes, respectively, showed altered expression patterns after salt and drought treatments. These differentially expressed genes are likely involved in the regulation of flower opening and abiotic stress response. This study provides new insights into the potential roles of OfIAAs and contributes to a better understanding of the regulatory mechanisms of flower opening and abiotic stress tolerance in O. fragrans.

Abstract To investigate the occurrence and 90-day mortality of cancer patients following unplanned admission to the intensive care unit (ICU), as well as to develop a risk prediction model for their 90-day prognosis. We prospectively analyzed data from cancer patients who were admitted to the ICU without prior planning within the past 7 days, specifically between May 12, 2021, and July 12, 2021. The patients were grouped based on their 90-day survival status, and the aim was to identify the risk factors influencing their survival status. A total of 1488 cases were included in the study, with an average age of 63.2 ± 12.4 years. The most common reason for ICU admission was sepsis (n = 940, 63.2%). During their ICU stay, 29.7% of patients required vasoactive drug support (n = 442), 39.8% needed invasive mechanical ventilation support (n = 592), and 82 patients (5.5%) received renal replacement therapy. We conducted a multivariate COX proportional hazards model analysis, which revealed that BMI and a history of hypertension were protective factors. On the other hand, antitumor treatment within the 3 months prior to admission, transfer from the emergency department, general ward, or external hospital, high APACHE score, diagnosis of shock and respiratory failure, receiving invasive ventilation, and experiencing acute kidney injury (AKI) were identified as risk factors for poor prognosis within 90 days after ICU admission. The average length of stay in the ICU was 4 days, while the hospital stay duration was 18 days. A total of 415 patients died within 90 days after ICU admission, resulting in a mortality rate of 27.9%. We selected 8 indicators to construct the predictive model, which demonstrated good discrimination and calibration. The prognosis of cancer patients who are unplanned transferred to the ICU is generally poor. Assessing the risk factors and developing a risk prediction model for these patients can play a significant role in evaluating their prognosis.

Abstract Background The establishment of a robust gut microbiota in piglets during their early developmental stage holds the potential for long-term advantageous effects. However, the optimal timeframe for introducing probiotics to achieve this outcome remains uncertain. Results In the context of this investigation, we conducted a longitudinal assessment of the fecal microbiota of 63 piglets at three distinct pre-weaning time points. Simultaneously, we gathered vaginal and fecal samples from 23 sows. Employing 16S rRNA gene and metagenomic sequencing methodologies, we conducted a comprehensive analysis of the fluctuation patterns in microbial composition, functional capacity, interaction networks, and colonization resistance within the gut microbiota of piglets. As the piglets progressed in age, discernible modifications in intestinal microbial diversity, composition, and function were observed. A source-tracking analysis unveiled the pivotal role of fecal and vaginal microbiota derived from sows in populating the gut microbiota of neonatal piglets. By D21, the microbial interaction network displayed a more concise and efficient configuration, accompanied by enhanced colonization resistance relative to the other two time points. Moreover, we identified three strains of Ruminococcus sp. at D10 as potential candidates for improving piglets' weight gain during the weaning phase. Conclusions The findings of this study propose that D10 represents the most opportune juncture for the introduction of external probiotic interventions during the early stages of piglet development. This investigation augments our comprehension of the microbiota dynamics in early-life of piglets and offers valuable insights for guiding forthcoming probiotic interventions.

Salted duck egg yolk, a key ingredient in various specialty foods in China, frequently contains broken eggshell fragments embedded in the yolk due to high-speed shell-breaking processes, which pose significant food safety risks. This paper presents an online detection method, YOLOv7-SEY-DeepSORT (salted egg yolk, SEY), designed to integrate an enhanced YOLOv7 with DeepSORT for real-time and accurate identification of salted egg yolks with impurities on production lines. The proposed method utilizes YOLOv7 as the core network, incorporating multiple Coordinate Attention (CA) modules in its Neck section to enhance the extraction of subtle eggshell impurities. To address the impact of imbalanced sample proportions on detection accuracy, the Focal-EIoU loss function is employed, adaptively adjusting bounding box loss values to ensure precise localization of yolks with impurities in images. The backbone network is replaced with the lightweight MobileOne neural network to reduce model parameters and improve real-time detection performance. DeepSORT is used for matching and tracking yolk targets across frames, accommodating rotational variations. Experimental results demonstrate that YOLOv7-SEY-DeepSORT achieves a mean average precision (mAP) of 0.931, reflecting a 0.53% improvement over the original YOLOv7. The method also shows enhanced tracking performance, with Multiple Object Tracking Accuracy (MOTA) and Multiple Object Tracking Precision (MOTP) scores of 87.9% and 73.8%, respectively, representing increases of 17.0% and 9.8% over SORT and 2.9% and 4.7% over Tracktor. Overall, the proposed method balances high detection accuracy with real-time performance, surpassing other mainstream object detection methods in comprehensive performance. Thus, it provides a robust solution for the rapid and accurate detection of defective salted egg yolks and offers a technical foundation and reference for future research on the automated and safe processing of egg products.

Objective: To establish the method for content determination of the five phenolic acids ingredients of Phyllanthus emblica L. in different harvest time, and to investigate its variation rules so as to determine the best harvest time. Methods: The contents of Epigallocatechin, Corilagin, Scorpion Biphenyl acid, Ellagic acid and Scorpion acid of Phyllanthus emblica L. in different harvest time were determined by HPLC, and the total amount of accumulation of the five active ingredients was calculated. The chromatographic column was Inertsil ODS-3 C18, acetonitrile−0.1% phosphoric acid aqueous solution as mobile phase for gradient elution. A diode array detector was used with full-wavelength scanning. Flow rate was 1.0 mL·min−1. Injection was 10 μL. Used SPSS 24.0 software for principal component analysis, three principal components were selected and the comprehensive score was calculated. Results: The above five ingredients had a good linear relationship, and the correlation coefficients was more than 0.998, while the average recovery rates were 98.30%~101.22% and RSD were 0.43%~2.66%. There were some differences in the contents of the five phenolic acids ingredients in the leaves of Phyllanthus emblica L. during different harvesting periods. The contents of above five ingredients were 0.2317~1.5074, 1.9517~4.6593, 2.5105~6.3325, 2.2440~5.3386, 13.5570~39.1897 mg·g−1, respectively. The change in content of the five phenolic acids ingredients in different harvest time was: July>May>August>June>January>September>April>February>October>December>March>November. The total content was the highest in July, which was (55.158±0.83) mg/g and showed a dynamic trend of decreasing first and then increasing. Principal component analysis results showed that the month with the highest comprehensive score was July. Conclusion: Established method can be used to determine the contents of the five phenolic acids ingredients of Phyllanthus emblica L. in different harvest time. July was the best harvest time for Phyllanthus emblica L.

PurposeWhether the diagnosis of non-alcoholic fatty liver disease or metabolic dysfunction-associated fatty disease has a different impact on liver transplant recipients with hepatocellular carcinoma is not yet clear.MethodsData from a two-center retrospective cohort study were collected to compare and investigate the differences between non-alcoholic fatty liver disease and metabolic dysfunction-associated fatty liver disease in clinicopathologic parameters and prognosis among liver transplant recipients with hepatocellular carcinoma.ResultsA total of 268 liver transplant recipients with hepatocellular carcinoma were included. The prevalence among pre- and post-transplant metabolic dysfunction-associated fatty liver disease was 10.82% and 30.22%, while for non-alcoholic fatty liver disease, it was 7.09% and 26.87%, respectively. The clinicopathological parameters were similar between the two pre-transplant groups. In contrast, the post-transplant group with metabolic dysfunction-associated fatty liver disease exhibited a higher prevalence of diabetes mellitus and a greater body mass index. However, the other parameters were similar between the two post-transplant groups (p > 0.05). Factors such as the largest tumor size > 4 cm, microvascular invasion, lack of tumor capsule, post-transplant metabolic dysfunction-associated fatty liver disease, and decreased post-transplant lymphocyte percentage were related to an increased risk of recurrence.ConclusionIn patients undergone liver transplantation for hepatocellular carcinoma, the diagnosis of metabolic dysfunction-associated fatty disease is more strongly associated with metabolic abnormalities than the diagnosis of non-alcoholic fatty liver disease and is an independent predictor of hepatocellular carcinoma recurrence.

Objective Loin muscle area (LMA) is an important target trait of pig breeding. This study aimed to identify single nucleotide polymorphisms (SNPs) and genes associated with LMA in the Duroc×(Landrace×Yorkshire) crossbred pigs (DLY). Methods A genome-wide association study was performed using the Illumina 50K chip to map the genetic marker and genes associated with LMA in 511 DLY pigs (255 boars and 256 sows). Results After quality control, we detected 35,426 SNPs, including six SNPs significantly associated with LMA in pigs, with MARC0094338 and ASGA0072817 being the two key SNPs responsible for 1.77% and 2.48% of the phenotypic variance of LMA, respectively. Based on previous research, we determined two candidate genes (growth hormone receptor [GHR] and 3-oxoacid Co A-transferase 1 [OXCT1]) that are associated with fat deposition and muscle growth and found further additional genes (MYOCD, ARHGAP44, ELAC2, MAP2K4, FBXO4, FBLL1, RARS1, SLIT3, and RANK3) that are presumed to have an effect on LMA. Conclusion This study contributes to the identification of the mutation that underlies quantitative trait loci associated with LMA and to future pig breeding programs based on marker-assisted selection. Further studies are needed to elucidate the role of the identified candidate genes in the physiological processes involved in LMA regulation.

As a crucial member of the gene family involved in the biosynthesis of strigolactones, D27 plays an important regulatory role in plant branching and root development, which is essential for field management and yield increase in peppers (Capsicum annuum L.). To comprehensively understand the characteristics of the pepper D27 gene family, we identified three CaD27 genes. By analyzing their physicochemical properties, phylogenetic relationships, gene structures, promoters, and expression patterns in different tissues, the characteristics of the CaD27 gene family were revealed. The research results showed that these three CaD27 genes are located in three different chromosomes. Evolutionary analysis divided the members of CaD27 into three groups, and gene collinearity analysis did not find any duplicates, indicating the diversity and non-redundancy of the CaD27 gene family members. In addition, we identified and classified cis-elements in the promoter regions of CaD27 genes, with a relatively high proportion related to light and plant hormone responses. Expression pattern analysis showed that CaD27.1 is expressed in leaves, while CaD27.2 is expressed in roots, indicating tissue specificity. Furthermore, protein interaction predictions revealed an interaction between D27.2 and CCD7. This study provided important insights into the function and regulatory mechanisms of the CaD27 gene family and the role of strigolactones in plant growth and development.

Background: Liver cancer had become the sixth most common cancer. Nitidine chloride (NC) has demonstrated promising anti-HCC properties; however, further elucidation of its mechanism of action is necessary. Methods: The anti-HCC targets of NC were identified through the utilization of multiple databases and ChIPs data analysis. The GO and KEGG analyses to determine the specific pathway affected by NC. The Huh 7 and Hep G2 cells were subjected to a 24-h treatment with NC, followed by evaluating the impact of NC on cell proliferation and cell cycle. The involvement of the p53/14-3-3 Sigma/CDK1 axis in HCC cells was confirmed by qPCR and WB analysis of the corresponding genes and proteins. Results: The GO and KEGG analysis showed the targets were related to cell cycle and p53 signaling pathways. In vitro experiments showed that NC significantly inhibited the proliferation of HCC cells and induced G2/M phase arrest. In addition, qPCR and WB experiments showed that the expression of p53 in HCC cells increased after NC intervention, while the expression of 14-3-3 Sigma and CDK1 decreased. Conclusion: NC can inhibit the proliferation of HCC cells and induce G2/M cell cycle arrest, potentially by regulating the p53/14-3-3 Sigma/CDK1 axis.

Nowadays, there are many researches on flexible humidity sensors based on different composites, such as polymer with graphene oxide, carbon nanotubes as conductive fillers. Herein, a flexible wearable device used for humidity detection and breath monitoring was fabricated based on the molecular designable thermoplastic polyamide elastomer (TPAE). TPAE is block copolymers consisting of the polyamide hard segments and the polyether soft segments. Among them, the hard segments (PA1212 or PA6) form the crosslinking network providing resilience, and the soft segments composed of PPO and PEO endow TPAE with flexibility and solvent-responsiveness. The resistance of TPAE-based humidity sensor decreases with the increase of relative humidity (RH), exhibiting negative humidity sensitivity. Besides, it has a wide humidity monitoring range (11 %RH ∼ 91 %RH) and good fitting degree of R2 > 0.99 which is hardly influenced by bending deformation. When the humidity sensor was exposed to organic vapor (e.g. ethanol), it exhibited a faster response of astonishing 4 s. This novel block copolymer not only shows good humidity sensing performance in many aspects, but also provides a novel strategy and approach for the preparation of the humidity sensing materials afterwards.

Context Viruses have the characteristics of rapid transmission and high mortality. At present, western medicines still lack an ideal antiviral. As natural products, many traditional Chinese medicines (TCM) have certain inhibitory effects on viruses, which has become the hotspot of medical research in recent years.Objective The antiviral active ingredients and mechanisms of TCM against viral diseases was studied in combination with the pathogenesis of viral diseases and antiviral effects.Materials and methods English and Chinese literature from 1999 to 2021 was collected from databases including Web of Science, PubMed, Elsevier, Chinese Pharmacopoeia 2020 (CP), and CNKI (Chinese). Traditional Chinese medicines (TCM), active ingredients, antiviral, mechanism of action, and anti-inflammatory effect were used as the key words.Results The antiviral activity of TCM is clarified to put forward a strategy for discovering active compounds against viruses, and provide reference for screening antivirus drugs from TCM. TCM can not only directly kill viruses and inhibit the proliferation of viruses in cells, but also prevent viruses from infecting cells and causing cytophilia. It can also regulate the human immune system, enhance human immunity, and play an indirect antiviral role.Discussion and conclusion Based on the experimental study and antiviral mechanism of TCM, this paper can provide analytical evidence that supports the effectiveness of TCM in treating virus infections, as well as their mechanisms against viruses. It could be helpful to provide reference for the research and development of innovative TCMs with multiple components, multiple targets and low toxicity.

Abstract Objectives Patients are classified according to the severity of their condition and graded according to the diagnosis and treatment capacity of medical institutions. This study aims to correctly assign patients to medical institutions for treatment and develop patient allocation and medical resource expansion schemes among hospitals in the medical network. Methods Illness severity, hospital level, allocation matching benefit, distance traveled, and emergency medical resource fairness were considered. A multi-objective planning method was used to construct a patient allocation model during major epidemics. A simulation study was carried out in two scenarios to test the proposed method. Results (1) The single-objective model obtains an unbalanced solution in contrast to the multi-objective model. The proposed model considers multi-objective problems and balances the degree of patient allocation matching, distance traveled, and fairness. (2) The non-hierarchical model has crowded resources, and the hierarchical model assigns patients to matched medical institutions. (3) In the “demand exceeds supply” situation, the patient allocation model identified additional resources needed by each hospital. Conclusion Results verify the maneuverability and effectiveness of the proposed model. It can generate schemes for specific patient allocation and medical resource amplification and can serve as a quantitative decision-making tool in the context of major epidemics.

This study develops a conceptual framework based on the unified theory of acceptance and use of technology, the network theory perspective, and metaverse characteristics. Data were gathered through questionnaires from 209 Chinese manufacturers, and covariance-based structural equation modelling was the main approach used. The results indicate that performance expectancy, facilitating conditions, and establishing initial trust amongst supply chain partners can drive behavioural intention with respect to metaverse adoption for knowledge sharing to improve supply chain resilience. In addition, sensory feedback is an important characteristic of the metaverse and has a critical influence on adoption behaviour. Moreover, it has a complementary partial mediation effect on the relationship between establishing initial trust amongst supply chain partners and behavioural intention as well as a full mediating effect on the relationship between the initial trust of supply chain partners and behavioural expectations. Finally, sensory feedback moderates the relationship between behavioural intention and expectancy. Our findings contribute to the literature on metaverse adoption in the context of supply chain resilience.

Water quality in rivers usually exhibits markedly spatiotemporal variations as affected by urbanization, while the magnitude of these effects remain unclear. This study aimed to investigate the spatiotemporal water quality variations in the urbanizing Chongqing reach of Jialing River and its tributary using a 6-yr multisite dataset (January 2010–December 2015). Water quality variables including pH, dissolved oxygen (DO), permanganate index (CODMn), ammonia nitrogen (NH3-N) and total phosphorus (TP) were deciphered. Results showed that the trunk river displayed respectively 1.2 and 39.8% higher pH and DO concentration but 66.9, 94.7 and 85.2% lower CODMn, NH3-N and TP concentrations relative to the tributary (P < 0.05), due largely to the dilution effects occurring in the trunk river. The dry season presented respectively 1.3, 18.2, 102.8 and 32.5% higher pH, DO, NH3-N and TP concentrations than those in the wet season (P < 0.05). DO concentration showed significant inter-annual variations (P < 0.05), and CODMn, NH3-N and TP concentrations all presented markedly declining trends from 2010 to 2015 (P < 0.05). Significant relationships among the study variables were found in different spatiotemporal scales (P < 0.05). Our results are valuable to optimize strategies for sustainable water quality management in rivers experiencing urbanization worldwide. HIGHLIGHTS Spatiotemporal water quality variations in the lower Jialing River are examined.; Trunk Jialing River shows better water quality condition than its tributary river.; Lower river pollutant concentrations are found in the wet season rather than the dry season.; Pollutant concentrations decline in the lower Jialing River from 2010 to 2015.; Water quality variables display marked links across various spatiotemporal scales.;

The evaluation of the shear performance of stone panel–panel joints (SPPJs) in stone cladding has important engineering significance, as it plays a crucial role in stone cladding failure. The purpose of this paper is to analyze and predict the influence of the dimension and the Young’s modulus of sealant on the shear performance of SPPJs. Based on monotonic and cyclic loading tests, the effects of Young’s modulus and the dimension of sealant on the failure characteristics, stress–strain characteristics, stiffness degradation, and energy dissipation capacity of an SPPJs were investigated. According to finite element analysis, the strain distribution of an SPPJ under monotonic loading was analyzed for different sealant widths and number of sealant layers. The results indicate that the failure modes of SPPJs change with the variation of sealant amount. As the Young’s modulus of the sealant increases, the shear failure strength and shear yield strain of SPPJs increase. The increase in sealant thickness reduces the shear failure strength and stiffness of SPPJs. Based on the same shear strain, the increase in the sealant thickness enhances the cumulative energy consumption of SPPJs. The strain concentration zone of the specimens with two sealant layers in unilateral SPPJs becomes larger with the increase in sealant width.

Reversible chemical covalency provides a path to materials that can degrade and recombine with appropriate stimuli and which can be used for tissue regeneration and repair. However, designing and preparing efficient and quickly self-healing materials has always been a challenge. The preparation strategies of photoresponsive gels attract a lot of attention due to their precise spatial and temporal control and their predetermined response to light stimulation. In this work, the linear copolymer PAC was synthesized via precipitation polymerization of acrylic acid and 7-(2-acrylate-ethoxylated)-4-methylcoumarin. The coumarin groups on the copolymer PAC side chains provide a reversible chemical cross-linking via photostimulation, which achieves reversible regulation of the gel network structure. The concentration of 18 wt% PAC solution produces gelation under irradiation with 365 nm. In contrast, PAC gel is restored to soluble copolymers under irradiation with 254 nm. Meanwhile, the mechanical and self-healing properties of the gel were also explored. It is demonstrated that the cracks of the gel can be repaired simply, quickly, and efficiently. Furthermore, the PAC copolymer shows an excellent adhesion property based on the reversible sol–gel transition. Thus, the PAC gel has considerable potential for applications in engineering and biomedical materials.

A fresh censored δ shock model is investigated. The arrival of random shocks follows a generalized Pólya process, and the failure mechanism of the system occurs based on the censored δ shock model. The generalized Pólya process is used for modeling because the generalized Pólya process has excellent properties, including the homogeneous Poisson process, the non-homogeneous Poisson process, and the Pólya process. Thus far, the lifetime properties of the censored δ shock model under the generalized Pólya process have not been studied. Therefore, for the established generalized Pólya censored δ shock model, the corresponding reliability function, the upper bound of the reliability function, the mean lifetime, the failure rate, and the class of life distribution are obtained. In addition, a replacement strategy N, based on the number of failures of the system, is considered using a geometric process. We determined the optimal replacement policy N* by objective function minimization. Finally, a numerical example is presented to verify the rationality of the model.

While cycloid pin-wheel precision reducers (referred to as RV reducers) are widely used in industrial robots, a widely accepted design standard or verification method of their test platforms is not available. In this study, a comprehensive sliding-separation test platform of RV reducers was developed. The test platform can test various measurement items such as transmission error, static measurement of lost motion, dynamic measurement of lost motion, torsional rigidity, no-load running torque, starting torque, backdriving torque, and transmission efficiency of the RV reducer for robots. The principle and method of dynamic measurement of lost motion tests based on the two-way transmission error method were studied and this test function was successfully integrated with the comprehensive test platform in order to increase the test items of the dynamic performance parameters of RV reducers. The measurement results of the no-load running torque of the RV reducer were consistent with the Stribeck curve. Based on the concept of optimal measurement speed, a decomposition test method of the geometric component of the dynamic measurement of lost motion and the elastic component of the dynamic measurement of lost motion was proposed in the dynamic measurement test of lost motion. Through precision calibration, function test and repeatability test, the results were compared with the data of enterprise’s samples. The consistent results have proved that the test platform met engineering requirements and measurement accuracy requirements. Based on the new test principle, the developed platform can test more parameters of RV reducers with high precision and display the comprehensive test performance.

This paper presents an experimental study on the flexural behavior of reinforced concrete (RC) beams strengthened with prestressed steel wire ropes-polymer mortar composite (PSWR-PM). The effects of reinforcement ratio of steel wire ropes, strengthening procedures, and unbonded length of the steel wire rope-to-polymer mortar interface were studied with large-scale bending tests on RC beams strengthened by PSWR-PM. The test results showed that the proposed strengthening technique could substantially increase the load carrying capacity of the RC beams. Both strengthening procedures were effective in enhancing the flexural behaviour of the RC beams, and strengthening procedure II could even restrain the debonding failure. The unbonded length of the steel wire rope-to-polymer mortar interface had a minor impact on the ultimate capacity when the anchoring length of the steel wire rope in the polymer mortar was sufficient. Moreover, a theoretical model was proposed to predict the flexural strength of RC beams strengthened with the PSWR-PM technique and the predictions showed good agreement with the experimental results.

ObjectiveTo detect the effect and safety of massage therapy on infants with congenital muscular torticollis.MethodsA total of 56 infants with unilateral congenital muscular torticollis were enrolled in this retrospective comparative study. The subjects were divided in two groups, namely, the control group and the massage group. The control group (n = 28) received the treatment of sternocleidomastoid muscle (SCM) stretching, while the massage group (n = 28) received massage therapy combined with SCM stretching. The following parameters were compared: the cervical range of motion (ROM) and functional level (muscle function scale and ratio of muscle function scale scores). Complications, if any, were also recorded.ResultsOf the 56 infants, 7 infants (12.5%) underwent surgery with little functional improvement. The total effective rate of conservative treatment was 87.5%. No significance was found in terms of the surgery rate between both groups (14.29 vs. 10.71%, P = 0.693). During the process of pork production, the carcasses of pigs are divided and sold, which provides better economic benefits and market competitiveness for pork production than selling the carcass as a whole. Due to the significant cost of post-slaughter phenotypic measurement, the genetic architecture of tenderloin weight (TLNW) and rib weight (RIBW)—important components of pig carcass economic value—remain unknown. In this study, we conducted genome-wide association studies (GWAS) for TLNW and RIBW traits in a population of 431 Duroc × Landrace × Yorkshire (DLY) pigs. In our study, the most significant single nucleotide polymorphism (SNP) associated with TLNW was identified as ASGA0085853 (3.28 Mb) on Sus scrofa chromosome 12 (SSC12), while for RIBW, it was Affx-1115046258 (172.45 Mb) on SSC13. Through haplotype block analysis, we discovered a novel quantitative trait locus (QTL) associated with TLNW, spanning a 5 kb region on SSC12, and a novel RIBW-associated QTL spanning 1.42 Mb on SSC13. Furthermore, we hypothesized that three candidate genes, TIMP2 and EML1, and SMN1, are associated with TLNW and RIBW, respectively. Our research not only addresses the knowledge gap regarding TLNW, but also serves as a valuable reference for studying RIBW. The identified SNP loci strongly associated with TLNW and RIBW may prove useful for marker-assisted selection in pig breeding programs.

Concrete-filled stainless steel tubes (CFSST) could be used as structural members in corrosion-prone environments. A detailed numerical investigation of the mechanical performance and calculation method of CFSST members under eccentric tension is carried out in this paper. A finite element analysis (FEA) model that adopts three-dimensional elements is established, and related experimental results of CFSST and conventional concrete-filled carbon steel tubes (CFST) subjected to tension are used to validate the FEA model. Then, the calibrated FEA model is used to investigate the performance of CFSST eccentrically tensile members, especially the composite actions and stress distribution laws between the stainless steel tube and the concrete core, which play a key role in the load-carrying capacity of the composite member. To quantitatively determine the influence of different parameters on the load-carrying capacity of CFSST tensile members, a wide-range parametric analysis is performed. Finally, a calculation model is proposed to be used to predict the ultimate tensile strength of CFSST members subjected to eccentric tension, and the model-predicted values show good agreement with the FEA-computed results.

In order to survey the possible periodic, uncertainty and common features in runoff with multi-temporal scales, the empirical mode decomposition (EMD) method combined with the set pair analysis (SPA) method was applied, with data observed at Zhangjiashan hydrological station. The results showed that the flood season and annual runoff time series consisted of four intrinsic mode function (IMF) components, and the non-flood season time series exhibited three IMF components. Moreover, based on the different coupled set pairs from the time series, the identity, discrepancy, and contrary of different periods at multi-temporal scales were determined by the SPA method. The degree of connection μ between the flood season and annual runoff periods were the highest, with 0.94, 0.77, 0.7 and 0.73, respectively, and the μ between the flood periods and the non-flood periods were the lowest, with 0.66, 0.46, 0.24 and 0.24, respectively. Third, the maximum μ of each SPA appeared in the first mode function. In general, the different extractive periods decomposed by the EMD method reflected the average state of Jinghe River. Results also verified that runoff suffered from seasonal and periodic fluctuations, and fluctuations in the short-term corresponded to the most important variable. Therefore, the conclusions drawn in this study can improve water resources regulation and planning. BackgroundBreast cancer (BC) is one of the most common malignant tumors in women and poses a serious threat to their health. Compound Kushen injection (CKI) has shown therapeutic effects on a variety of cancers, including BC, and it can significantly improve the lives of patients. However, the underlying mechanism remains unclear and needs to be fully elucidated.MethodsThe active constituents of CKI were identified through a literature review, and the anti-BC targets of CKI were determined using multiple databases and a ChIP data analysis. Subsequently, the target was analyzed on the DAVID database through GO and KEGG to identify the key pathway that CKI affects to exhibit anti-BC activity. In addition, MCF-7 and MDA-MB-231 cells were treated with CKI for 24 and 48 hours at five concentrations, and the effects of CKI on cell proliferation and apoptosis were measured using MTT and annexin V/propidium iodide staining assays, respectively. The genes and protein identified to be involved in this pathway were verified using real-time quantitative PCR (qPCR) and western blot(WB) in BC cells.ResultsTwelve CKI anti-BC targets were obtained by a comprehensive analysis of the targets collected in the databases and results from the ChIP analysis. Bioinformatics analysis was performed for 12 targets. KEGG analysis showed that the 12 targets were mainly related to the VEGF, ErbB, and TNF signaling pathways. We focused our study on the VEGF signaling pathway as the p-value for the VEGF signaling pathway was the lowest among the three pathways. In vitro experiments showed that CKI significantly inhibited the proliferation of BC cells and induced apoptosis. Furthermore, qPCR and WB experiments showed that the expression of VEGF signaling pathway genes PIK3CA and NOS3 were significantly increased meanwhile SRC was significantly decreased after CKI intervention.ConclusionCKI significantly inhibited the proliferation of BC cells and induced apoptosis. The main mechanism for the anti-BC effect of CKI may be that it regulates the VEGF signaling pathway by increasing the expression of PIK3CA, SRC, and NOS3. Macrozamin and lamprolobine may be the main active components of CKI against BC.

Abstract Background Improving feed efficiency is economically and environmentally beneficial in the pig industry. A deeper understanding of feed efficiency is essential on many levels for its highly complex nature. The aim of this project is to explore the relationship between fecal metabolites and feed efficiency-related traits, thereby identifying metabolites that may assist in the screening of the feed efficiency of pigs. Results We performed fecal metabolomics analysis on 50 individuals selected from 225 Duroc x (Landrace x Yorkshire) (DLY) commercial pigs, 25 with an extremely high feed efficiency and 25 with an extremely low feed efficiency. A total of 6749 and 5644 m/z features were detected in positive and negative ionization modes by liquid chromatography-mass spectrometry (LC/MS). Regrettably, the PCA could not classify the the samples accurately. To improve the classification, OPLS-DA was introduced. However, the predictive ability of the OPLS-DA model did not perform well. Then, through weighted coexpression network analysis (WGCNA), we found that one module in each positive and negative mode was related to residual feed intake (RFI), and six and three metabolites were further identified. The nine metabolites were found to be involved in multiple metabolic pathways, including lipid metabolism (primary bile acid synthesis, linoleic acid metabolism), vitamin D, glucose metabolism, and others. Then, Lasso regression analysis was used to evaluate the importance of nine metabolites obtained by the annotation process. Conclusions Altogether, this study provides new insights for the subsequent evaluation of commercial pig feed efficiency through small molecule metabolites, but also provide a reference for the development of new feed additives.

Abstract Introduction Cirrhotic patients with septic shock have a poorer prognosis compared with the general population. Our study aimed to investigate the survival benefit of the implementation of hour-1 bundle proposed by Surviving Sepsis Campaign, and to analyze the predictors associated with short-term mortality of these patients. Methods A single-center, retrospective case-control study was conducted among adult patients who visited the emergency department between January 1, 2018 and December 31, 2019. All patients with a diagnosis of liver cirrhosis and septic shock were enrolled. Their baseline characteristics, laboratory results, source of sepsis, and sepsis bundle management were recorded. We further divided the patients into survivor and non-survivor groups to identify independent prognostic factors. Results A total of 88 patients were eligible for this study. The overall 30-day mortality rate was 53.4% (47/88). The proportion of hour-1 bundle achievement was 30.7% (27/88). There were no significant mortality differences between the hour-1 bundle achievement and non-achievement groups (44.4% vs. 57.4%, p = 0.35). Compared with the patients in the survivor group, patients in the non-survivor group had significantly more advanced stage of cirrhosis and a lower proportion of receiving source control (4.3% vs. 22.0%, p = 0.02).
