Abstract: The recent development of biological sensors has extended marine plankton studies from conducting laboratory bench work to in vivo and real-time observations. Flow cytometry (FCM) has shed new light on marine microorganisms since the 1980s through its single-cell approach and robust detection of the smallest cells. FCM records valuable optical properties of light scattering and fluorescence from cells passing in a single file in front of a narrow-collimated light source, recording tens of thousands of cells within a few minutes. Depending on the instrument settings, the sampling strategy, and the automation level, it resolves the spatial and temporal distribution of microbial marine prokaryotes and eukaryotes. Cells are usually classified and grouped on cytograms by experts and are still lacking standards, reducing data sharing capacities. Therefore, the need to make FCM data sets FAIR (Findability, Accessibility, Interoperability, and Reusability of digital assets) is becoming critical. In this paper, we present a consensus vocabulary for the 13 most common marine microbial groups observed with FCM using blue and red-light excitation. The authors designed a common layout on two-dimensional log-transformed cytograms reinforced by a decision tree that facilitates the characterization of groups. The proposed vocabulary aims at standardising data analysis and definitions, to promote harmonisation and comparison of data between users and instruments. This represents a much-needed step towards FAIRification of flow cytometric data collected in various marine environments.

Abstract: Along with their important diversity, coastal ecosystems receive various amounts of nutrients, principally arising from the continent and from the related human activities (mainly industrial and agricultural activities). During the 20th century, nutrients loads have increased following the increase of both the global population and need of services. Alongside, climate change including temperature increase or atmospheric circulation change has occurred. These processes, Ecosystem state changes are hard to monitor and predict. To study the long-term changes of nutrients concentrations in coastal ecosystems, eleven French coastal ecosystems were studied over 20 years as they encompass large climatic and land pressures, representative of temperate ecosystems, over a rather small geographical area. Both univariate (time series decomposition) and multivariate (relationships between ecosystems and drivers) statistical analyses were used to determine ecosystem trajectories as well as typologies of ecosystem trajectories. It appeared that most of the French coastal ecosystems exhibited trajectories towards a decrease in nutrients concentrations. Differences in trajectories mainly depended on continental and human influences, as well as on climatic regimes. One single ecosystem exhibited very different trajectories, the Arcachon Bay with an increase in nutrients concentrations. Ecosystem trajectories based on ordination techniques were proven to be useful tools to monitor ecosystem changes. This study highlighted the importance of local environments and the need to couple uni- and multi-ecosystem studies. Although the studied ecosystems were influenced by both local and large-scale climate, by anthropogenic activities loads, and that their trajectories were mostly similar based on their continental influence, non-negligible variations resulted from their internal functioning.

Abstract: Adaptation of cell populations to environmental changes is mediated by phenotypic variability at the single-cell level. Enzyme activity is a key factor in cell phenotype and the expression of the alkaline phosphatase activity (APA) is a fundamental phytoplankton strategy for maintaining growth under phosphate-limited conditions. Our aim was to compare the APA among cells and species revived from sediments of the Bay of Brest (Brittany, France), corresponding to a pre-eutrophication period (1940’s) and a beginning of a post-eutrophication period (1990’s) during which phosphate concentrations have undergone substantial variations. Both toxic marine dinoflagellate Alexandrium minutum and the non-toxic dinoflagellate Scrippsiella acuminata were revived from ancient sediments. Using microfluidics, we measured the kinetics of APA at the single-cell level. Our results indicate that all S. acuminata strains had significantly higher APA than A. minutum strains. For both species, the APA in the 1990’s decade was significantly lower than in the 1940’s. For the first time, our results reveal both inter and intraspecific variabilities of dinoflagellate APA and suggest that, at a half-century timescale, two different species of dinoflagellate may have undergone similar adaptative evolution to face environmental changes and acquire ecological advantages.

Abstract: International audience; Marine mucilage has been described as worldwide phenomena occurring sporadically, or frequently in certain coastal areas. They are transitory phenomena that can remain in the photic zone for several days or weeks. Their occurrence has been more frequent and persistent, and their magnitude has increased during the last decades. Their formation typically reflects an imbalance in the marine microbial communities, and has frequently be linked to global changes. Recurrent marine mucilage events have been observed in recent years in the Southern Bay of Biscay near the French coast, mainly south of the Adour estuary. In this study, we used a fingerprinting method (T-RFLP) targeting 16S and 18S rRNA genes to investigate marine microbial community composition and dynamics over an annual cycle across the three domains of life in coastal water at three depths, in the Adour River upstream of the mouth and in the marine mucilage. In line with studies conducted in marine environments we highlighted that the dynamics of marine microbial communities in the Bay of Biscay coincided with two environmental contexts: winter and spring on one hand and summer and autumn on the other hand. More interestingly, this dynamics affects also the marine pelagic mucilage that was observed during the sampling campaigns. Thus, the composition of the marine mucilage that appears in winter/spring context diverges from the composition of the marine mucilage that occurred during the summer/autumn context. We highlighted also that marine microbial communities were partly different from marine mucilage communities with an enrichment of some microorganisms suggesting that marine mucilage behaves as a microhabitat in seawater and possesses distinct microbial assemblages from the surrounding communities.

Abstract: International audience; In coastal systems, particulate organic matter (POM) originates from various autochthonous and allochthonous organic matter sources. Also, some coastal systems are characterized by the occurrence of large amounts of mucilaginous material of biologic origin (i.e. phytoplankton, bacteria), which aggregates and potentially traps other organisms and particles present in the water column. This study focuses on POM origin and spatio-temporal dynamics in the South-East coast of the Bay of Biscay, an area subject to mucilage occurrence. In order to investigate POM quantitative and qualitative (C and N elemental and isotopic ratios) characteristics, sampling was performed over an annual cycle at two sites experiencing different mucilage occurrence and river influence. Contribution of phytoplankton, terrestrial POM and anthropogenic POM to coastal-POM composition was calculated using a three-sources mixing model. Overall, phytoplankton dominated the coastal-POM composition at all seasons, sites and most of the depths (71.6 ± 24.2%). Terrestrial-POM contribution was moderate (22.7 ± 21.8%) and anthropogenic-POM contribution was usually negligible (5.7 ± 7.4%). Both sites mainly exhibited similar vertical and temporal variations in terms of POM origin and dynamics: terrestrial-POM contribution increased with depth and was higher in winter at all depths and in autumn in bottom waters, compared to other seasons. The main differences between both sites were related to the vertical dynamics of the terrestrial contribution to the coastal POM. Horizontal, vertical and temporal variation of POM composition was linked to processes driving the sedimentary hydrodynamics: the river flow, the direction of the river plume and events of sediment resuspension/deposition. During the study period, the mucilage occurred only as flocs (small aggregates). The mucilage was of autochthonous origin and did not trap detectable amount of allochthonous material. © 2016 Elsevier B.V.

Abstract: Plankton produces numerous chemical compounds used in cosmetics and functional foods. They also play a key role in the carbon budget on the Earth. In a context of global change, it becomes important to understand the physiological response of these microorganisms to changing environmental conditions. Their adaptations and the response to specific environmental conditions are often restricted to a few active cells or individuals in large populations. Using analytical capabilities at the subnanoliter scale, microfluidic technology has also demonstrated a high potential in biological assays. Here, we review recent advances in microfluidic technologies to overcome the current challenges in high content analysis both at population and the single cell level.

Abstract: International audience; In costal systems, particulate organic matter (POM) results from a multiplicity of sources having their respective dynamics in terms of production, decomposition, transport and burial. The POM pool experiences thus considerable spatial and temporal variability. In order to better understand this variability, the present study employs statistical multivariate analyses to investigate links between POM composition and environmental forcings for a panel of twelve coastal systems distributed along the three maritime regions of France and monitored weekly to monthly for 1 to 8 years.At multi-system scale, two main gradients of POC composition have been identified: a 'Continent-Ocean' gradient associated with hydrodynamics, sedimentary dynamics and depth of the water column, and a gradient of trophic status related to nutrient availability. At local scale, seasonality of POC composition appears to be station-specific but still related to part of the above-mentioned forcings. A typology of systems was established by coupling spatial and temporal variability of POC composition. Four groups were highlighted: (1) the estuarine stations where POC composition is dominated by terrestrial POM and driven by hydrodynamics and sedimentary processes, (2) the oligotrophic systems, characterized by the contribution of diazotrophs due to low nutrient availability, and the marine meso/eutroph systems whose POC composition is (3) either deeply dominated by phytoplankton or (4) dominated by phytoplankton but where the contribution of continental and benthic POC is not negligible and is driven by hydrodynamics, sedimentary processes and the height of the water column.Finally, the present study provides several insights into the different forcings to POM composition and dynamics in temperate coastal systems at local and multi-system scales. This work also presents a methodological approach that establishes statistical links between forcings and POM composition, helping to gain more objectively insight of forcings.

Abstract: International audience; One way for phytoplankton to survive orthophosphate depletion is to utilize dissolved organic phosphorus by expressing alkaline phosphatase. The actual methods to assay alkaline phosphate activity—either in bulk or as a presence/absence of enzyme activity—fail to provide information on individual living cells. In this context, we develop a new microfluidic method to compartmentalize cells in 0.5 nL water-in-oil droplets and measure alkaline phosphatase activity at the single-cell level. We use enzyme-labeled fluorescence (ELF), which is based on the hydrolysis of ELF-P substrate, to monitor in real time and at the single-cell level both qualitative and quantitative information on cell physiology (i.e., localization and number of active enzyme sites and alkaline phosphatase kinetics). We assay the alkaline phosphatase activity of Tetraselmis sp. as a function of the dissolved inorganic phosphorus concentration and show that the time scale of the kinetics spans 1 order of magnitude. The advantages of subnanoliter-scale compartmentalization in droplet-based microfluidics provide a precise characterization of a population with single-cell resolution. Our results highlight the key role of cell physiology to efficiently access dissolved organic phosphorus.

Abstract: International audience; In coastal systems, the multiplicity of sources fueling the pool of particulate organic matter (POM) leads to divergent estimations of POM composition. Eleven systems (two littoral systems, eight embayments and semi-enclosed systems and one estuary) distributed along the three maritime façades of France were studied for two to eight years in order to quantify the relative contribution of organic matter sources to the surface-water POM pool in coastal systems. This study was based on carbon and nitrogen elemental and isotopic ratios, used for running mixing models. The POM of the estuary is dominated by terrestrial material (93% on average), whereas the POM of the other systems is dominated by phytoplankton (84% on average). Nevertheless, for the latter systems, the POM composition varies in space, with 1) systems where POM is highly composed of phytoplankton (≥ 93%), 2) systems characterized by a non-negligible contribution of benthic (8 to 19%) and/or riverine (7 to 19%) sources, and 3) the Mediterranean systems characterized by the contribution of diazotroph organisms (ca. 14%). A continent-to-ocean gradient of riverine and/or benthic POM contribution is observed. Finally, time series reveal 1) seasonal variations of POM composition, 2) differences in seasonality between systems, and 3) an inshore-offshore gradient of seasonality within each system that were sampled at several stations. Spatial and seasonal patterns of POM composition are mainly due to local to regional processes such as hydrodynamics and sedimentary hydrodynamic (e.g. resuspension processes, changes in river flows, wind patterns influencing along-shore currents) but also due to the geomorphology of the systems (depth of the water column, distance to the shore). Future studies investigating the link between these forcings and POM composition would help to better understand the dynamics of POM composition in coastal systems.

Abstract: Microalgae worldwide regularly cause harmful effects, considered from the human perspective, in that they cause health problems and economic damage to fisheries and tourism. Cyanobacteria cause similar problems in freshwaters. These episodes encompass a broad range of phenomena collectively referred to as "harmful algal blooms" (HABs). For adequate management of these phenomena, monitoring of microalgae is required. However, effective monitoring is time-consuming because cell morphology as determined by light microscopy may be insufficient to give definitive species and toxin attribution. In the European Union FP7 project MIDTAL (Microarrays for the Detection of Toxic Algae), we achieved rapid species identification using rRNA genes as the target. These regions can be targeted for probe design to recognise species or even strains. We also included antibody reactions to specific toxins produced by these microalgae because, even when cell numbers are low, toxins can be present and can accumulate in the shellfish. Microarrays are the state-of-the-art technology in molecular biology for the processing of bulk samples for detection of target RNA/DNA sequences. After 36 months, we have completed RNA-cell number-signal intensity calibration curves for 18 HAB species and the analysis of monthly field samples from five locations from year 1. Results from one location, Arcachon Bay (France), are reported here and compared favourably with cell counts in most cases. In general, the microarray was more sensitive than the cell counts, and this is likely a reflection in the difference in water volume analysed with the volume filtered for the microarray an order of magnitude greater.

17. Variability of nutrients and phytoplankton primary production in a shallow macrotidal coastal ecosystem (Arcachon Bay, France)

Abstract: Seasonal and spatial variations of phytoplankton primary production were studied using a high frequency sampling strategy in the external (ENW) and internal (INW) part of Arcachon Bay, during 2002 and 2003. In order to better assess the availability of nutrients and their relationship with phytoplankton primary production, nutrient variability was studied in relation to environmental conditions and phytoplankton production. During winter, when primary production rates were the lowest, nutrient concentrations were maximal but did not show excessive levels compared to highly urbanised areas. Seasonal and spatial variations of nutrient concentrations (especially DIN-nitrate + nitrite + ammonium- and Si) were largely influenced by Leyre River loads coupled with high tidal exchange with the Atlantic Ocean creating a nutrient gradient between the INW and ENW. By February, diatom growth leads to an early severe nutrient depletion in the entire bay. Examination of nutrient ratios showed that the potential limiting nutrient during spring was P in 2003, and Si in 2002. During summer 2003, N and Si concentrations reached their lowest values, and nutrient ratios revealed a N-deficient environment, more pronounced in the INW. The high Si:N ratios during this period might be explained by (1) important N-uptake by all autotroph communities and (2) benthic-pelagic coupling with high Si regeneration. This study shows that nutrient levels in Arcachon Bay seem to play an important role in the control of phytoplankton primary production rates during the productive period and explain their spatial, seasonal and inter-annual variability. Our estimates of annual integrated phytoplankton primary production (103 g C m−2 y−1) place this bay within the low to moderate phytoplankton primary production systems.

Abstract: Phytoplankton dynamics were assessed in the macrotidal ecosystem of Arcachon Bay through high-frequency surveys over a 5-year period in order to characterize typology of environmental conditions at the onset of the productive period. Temporal variations of hydrological and biological parameters were examined in external and internal waters of the lagoon, during the winter-spring periods from 1999 to 2003. An additional survey was performed during winter-spring 2005 in order to study the vertical structure of the water column. The occurrence of winter phytoplankton blooms between January and March emerged as a recurrent event. The early onset of the productive period is influenced by the biological functioning of adjacent Bay of Biscay oceanic waters. It is hypothesized that under a propitious hydrodynamic regime, phytoplankton inocula from the Bay of Biscay enter in the Arcachon Bay where cells presumably find favourable conditions for their fast development. The timing of the onset of those winter blooms in Arcachon Bay seems to be mainly influenced by the presence of anticyclonic weather conditions (associated with an increase in incident irradiance) during late winter (i.e. by February), while the water column does not show any particular stabilization nor stratification liable to facilitate the onset of these blooms. Moreover, these winter blooms dominated by diatoms led to an early nutrient depletion which could have inevitable consequences on the structuration of the food web during spring and summer.

19. THE CHEMICAL FORM OF DISSOLVED SI TAKEN UP BY MARINE DIATOMS

Abstract: Results of past studies of the pH-dependent Si uptake kinetics of Phaeodactylum tricornutum Bohlin suggested that the anion SiO(OH) is the chemical form of dissolved Si taken up by marine diatoms. We determined the chemical form of Si taken up by three other marine diatom species and P. tricornutum by examining the kinetics of Si use under two dramatically different SiO(OH):Si(OH)4 ratios in seawater by varying pH from ≈8 to ≈9.6. Uptake rates were determined using a precise and sensitive 32Si tracer methodology. The pH-dependent uptake kinetics obtained for all species except P. tricornutum suggest that marine diatoms transport Si(OH)4. The half-saturation constant (Km) varies strongly as a function of pH for all species when the substrate of transport is assumed to be SiO(OH). Kinetic curves for Thalassiosira pseudonana (Hustedt) Hasle et Heimdal, Thalassiosira weissflogii (Grunow) G. Fryxell et Hasle, and Cylindrotheca fusiformis Reimann et Lewin have statistically identical values of Km at each pH when the substrate for transport is assumed to be Si(OH)4 (T. pseudonana and T. weissflogii) or total dissolved silicon (C. fusiformis). In contrast, P. tricornutum exhibits unusual biphasic uptake kinetics: uptake conforms to Michaelis–Menten kinetics up to 15 to 25 μM, above which uptake increases linearly. This enigmatic response may have biased conclusions drawn from past experiments using this species. However, based on the consistency of the results for the three other species, a new model of Si transport in marine diatoms is proposed on the basis of the direct formation of a complex between the Si-transport protein and Si(OH)4.

20. Resistance of a coastal ecosystem to increasing eutrophic conditions: the Bay of Brest (France), a semi-enclosed zone of Western Europe

Abstract: The Bay of Brest is a semi-enclosed coastal ecosystem receiving high nutrients loading from freshwater inputs. In order to analyse the response of phytoplankton stocks to increasing eutrophic conditions, a survey of the annual cycle of hydrographic properties, nutrients and chlorophyll a concentrations, and carbon uptake rates was performed at four stations in 1993. This database has been compared to earlier measurements performed during several comparable surveys within the last 20 years. As compared to the seventies, a doubled nitrate loading is now entering this ecosystem, which is related to increased agricultural activities on the drainage basins, while the geographical origin of the nitrate input has been modified. As a result of these anthropogenic modifications, summer averaged Si/N stoichiometric balance has decreased during the two last decades but, contrary to what has been observed in other coastal ecosystems, phytoplankton stocks have not increased. Several ecological factors have hindered eutrophication: the high hydrodynamic mixing with adjacent marine waters, caused by the macrotidal regime, induces important nutrients losses, temperature and mostly light limit primary production while Si and P high recycling maintain nitrogen limitation in this ecosystem. Conjunction of these non-anthropogenic factors explains the global stability of phytoplankton stocks.

We examined the differences in interactions between these compartments in two French Atlantic bays. Microbes, considered here as central actors of the planktonic food web, were first monitored seasonally in Arcachon (2005) and Marennes-Oleron (2006) bays. Their dynamics were evaluated to categorize trophic periods using the models of Legendre and Rassoulzadegan as a reference framework. Microbial interactions were then compared through 48 h batch culture experiments performed during the phytoplankton spring bloom, identified as herbivorous in Marennes and multivorous in Arcachon. Marennes was spatially homogeneous compared with Arcachon. The former was potentially more productive, featuring a large number of heterotrophic pathways, while autotrophic mechanisms dominated in Arcachon. A link was found between viruses and phytoplankton in Marennes, suggesting a role of virus in the regulation of autotroph biomass. Moreover, the virus–bacteria relation was weaker in Marennes, with a bacterial lysis potential of 2.6% compared with 39% in Arcachon. The batch experiments (based on size-fractionation and viral enrichment) revealed different microbial interactions that corresponded to the spring-bloom trophic interactions in each bay. In Arcachon, where there is a multivorous web, flagellate predation and viral lysis acted in an opposite way on picophytoplankton. When together they both reduced viral production. Conversely, in Marennes (herbivorous web), flagellates and viruses together increased viral production. Differences in the composition of the bacterial community composition explained the combined flagellate-virus effects on viral production in the two bays.

Abstract: Measurement of biogenic silica (BSi) in the suspended matter of estuarine and coastal waters is not trivial because of the interference of lithogenic silica (LSi) that can represent up to 90% or more of the total particulate silica within the water column. Till date, no method has provided a satisfying way to correct this mineral interference when the most common wet-alkaline digestions are used. The most recently published method, Ragueneau and Tréguer (Mar. Chem., 45 (1994) 43-51), presents the disadvantage to be site-specific. In this paper, we present a robust method to measure BSi in all estuarine and coastal waters and correction for LSi interference. It consists of a double wet-alkaline digestion where the filter sample is submitted to a first digestion (0.2 M NaOH, pH 13.3) at 100 °C for 40 min. At the end of this first leach, all the BSi and part of the LSi have been converted into Si(OH)4. Si and Al concentrations ([Si]1 and [Al]1) in the supernatant are analyzed. After rinsing and drying, the filter is submitted to a second digestion, exactly identical to the first one, leading to the determination of the (Si:Al)2 ratio that is characteristic of the silicate minerals present in the sample. The corrected biogenic silica concentration is thus given by [BSi]=[Si]1-[Al]1(Si:Al)2. There are three basic assumptions behind the proposed method: (1) all the Al measured during the first leach is derived from silicate minerals, (2) all the biogenic silica has been digested during the first leach, so the second leach addresses the Si:Al ratio of silicate minerals only, and (3) silicate minerals dissolve during the second digestion exactly as during the first digestion, i.e. with a similar Si:Al ratio. Theoretical considerations and field experiments demonstrate that the above assumptions are accurate. Blanks and repeatability tests indicate that the detection limit ranges from 0.6 to 1.5 nmol of BSi present on the filter and that the uncertainty is close to 10%. This method was successfully applied to more than 200 samples from five different coastal areas, demonstrating the importance of this correction, which can represent up to 92% of the apparent BSi concentration determined after the first digestion.

Abstract: Background: Biological silica production has drawn intense attention and several molecules involved in biosilicification have been identified. Cellular mechanisms, however, remain unknown mainly due to the lack of probes required for obtaining information on live specimens. Results: The fluorescence spectra of the compound 2-(4-pyridyl)-5-((4-(2-dimethylaminoethylaminocarbamoyl)methoxy)phenyl)oxazole (PDMPO) are affected by the presence of >3.2 mM silicic acid. Increase in intensity and shift in the fluorescence coincide with the polymerization of Si. The unique PDMPO–silica fluorescence is explored here to visualize Si deposition in living diatoms. The fluorophore is selectively incorporated and co-deposited with Si into the newly synthesized frustules (the outer silica shells) showing an intense green fluorescence. Conclusions: We suggest that a fluorescence shift is due to an interaction between PDMPO and polymeric silicic acid. PDMPO is an excellent probe for imaging newly deposited silica in living cells and has also a potential for a wide range of applications in various Si-related disciplines, including biology of living organisms as diatoms, sponges, and higher plants, clinical research (e.g. lung fibrosis and cancer, bone development, artificial bone implantation), and chemistry and physics of materials research.

Abstract: The chemical factors (inorganic nitrogen, phosphate, silicic acid) that potentially or actually control primary production were determined for the Bay of Brest, France, a macrotidal ecosystem submitted to high-nitrate-loaded freshwater inputs (winter nitrate freshwater concentrations >700 mu M, Si:N molar ratio as low as 0.2, i.e. among the lowest ever published). Intensive data collection and observations were carried out from February 1993 to March 1994 to determine the variations of physical [salinity, temperature, photosynthetically active radiation (PAR), freshwater discharges] and chemical (oxygen and nutrients) parameters and their impacts on the phytoplankton cycle (fluorescence, pigments, primary production). With insufficient PAR the winter stocks of nutrients were almost nonutilized and the nitrate excess was exported to the adjacent ocean, due to rapid tidal exchange. By early April, a diatom-dominated spring bloom developed (chlorophyll a maximum = 7.7 mu g l(-1); primary production maximum = 2.34 g C m(-2) d(-1)) under high initial nutrient concentrations. Silicic acid was rapidly exhausted over the whole water column; it is inferred to be the primary limiting factor responsible for the collapse of the spring bloom by mid-May. Successive phytoplankton developments characterized the period of secondary blooms during summer and fall (successive surface chlorophyll a maxima = 3.5, 1.6, 1.8 and 1.0 mu g l(-1); primary production = 1.24, 1.18 and 0.35 g C m(-2) d(-1)). Those secondary blooms developed under lower nutrient concentrations, mostly originating from nutrient recycling. Until August, Si and P most likely limited primary production, whereas the last stage of the productive period in September seemed to be N limited instead, this being a period of total nitrate depletion in almost the whole water column. Si limitation of spring blooms has become a common feature in coastal ecosystems that receive freshwater inputs with Si:N molar ratios

Abstract: Diatoms, one of the most productive phytoplankton groups in the ocean, have an essential requirement for silicic acid for the construction of their opaline shells. In seawater silicic acid is present in three chemical species (H4SiO4, H3SiO4-, H2SiO42-). The silicic acid molecule actually taken up by diatoms is still under discussion. Algal cells are surrounded by a diffusive boundary layer (DBL) which has an effective thickness of the order of the cell radius. The nutrient transport through this layer is by diffusion only and thus may limit the supply of silicic acid to the cell. Due to uptake of one of the species of silicic acid by the cell, the chemical system in the DBL is out of equilibrium. We have developed a diffusion-reaction model for the components H4SiO4, H3SiO4-, H2SiO42-, OH-, and H+ in the DBL which allows us to calculate maximum silicic acid supply rates as a function of the total concentration of dissolved silicon, pH, algal radius, and silicic acid species taken up by the cell. In addition, analytical solutions for the simplified diffusion-reaction system are presented. Model calculations of the silicic acid maximum uptake rates are compared with uptake data for Thalassiosira weissflogii from recent laboratory experiments. Results indicate that the supply of H4SiO4 to T. weissflogii by diffusion-reaction processes is suffcient to sustain the observed growth rates. Thus, the H4SiO4 uptake of T. weissffogii is not diffusion-limited. This does not hold true for H3SiO4-.

Abstract: Phytoplankton dynamics were assessed in the macrotidal ecosystem of Arcachon Bay through high-frequency surveys over a 5-year period in order to characterize typology of environmental conditions at the onset of the productive period. Temporal variations of hydrological and biological parameters were examined in external and internal waters of the lagoon, during the winterâspring periods from 1999 to 2003. An additional survey was performed during winterâspring 2005 in order to study the vertical structure of the water column. The occurrence of winter phytoplankton blooms between January and March emerged as a recurrent event. The early onset of the productive period is influenced by the biological functioning of adjacent Bay of Biscay oceanic waters. It is hypothesized that under a propitious hydrodynamic regime, phytoplankton inocula from the Bay of Biscay enter in the Arcachon Bay where cells presumably find favourable conditions for their fast development. The timing of the onset of those winter blooms in Arcachon Bay seems to be mainly influenced by the presence of anticyclonic weather conditions (associated with an increase in incident irradiance) during late winter (i.e. by February), while the water column does not show any particular stabilization nor stratification liable to facilitate the onset of these blooms. Moreover, these winter blooms dominated by diatoms led to an early nutrient depletion which could have inevitable consequences on the structuration of the food web during spring and summer.
