Abstract

A probe of a catalytic antibody for a multistep Norrish type II photochemical reaction was conducted to determine the fate of a biradical intermediate as well as reaction pathways and product distributions, and to evaluate the effects of chemical catalysis in the antibody microenvironment. The alpha-ketoamide was utilized as both the hapten and substrate. Results showed that the mechanism in the antibody catalyst under review involved binding of the alpha-ketoamide and the routing of a biradical intermediate to form a single, unique product through the interaction of conformational control and chemical catalysis.

Abstract

A new approach for producing Diels-Alder catalytic antibodies shows that ferrocene is an acceptable mimic of the stereoelectronic features in the transition state of the Diels-Alder reaction. It is conformational of flexible haptenic design of freely rotating eta squared-cyclopentadienyl iron complex.

Abstract

The catalytic monoclonal antibodies, (mAbs), VTT1E3 and VTT35C8 accelerate the rate of cis to trans isomerization like the peptidyl-prolyl isomerase. The hapten evokes antibody combining sites that are complementary to alpha-ketoamide group. The mechanism involves transfer of the tripeptide of hapten from aqueous medium to non-polar cavity of antibody and stabilization by a rotation about prolyl-amide bond. The geometry of the twisted di-carbonyl unit are also essential for the antibody activity.

Abstract

The binding of therapeutically relevant synthetic retinoid derivatives to bovine and reindeer beta-lactoglobulin (betaLG) is demonstrated using fluorescence quenching and ultrafiltration/HPLC methods. Furthermore, synthesis of methyl (E)-3-[4-[(E)-2-(2,6,6-trimethylcyclohex-1-enyl)vinyl]phenyl]-acrylate 4 and (E)-3-[4-[(E)-2-(2,6,6-trimethylcyclohex-1-enyl)vinyl]phenyl]acrylic acid 5 is described. All studied compounds bind to both betaLG homologues with nanomolar K(d) values, and the interaction diminishes the pH-dependent aggregation of retinoids. Thus, betaLG may show benefits in improving the bioavailability of retinoid derivatives.
