Abstract Computer-aided diagnosis has been slow to develop in the field of oral ulcers. One of the major reasons for this is the lack of publicly available datasets. However, oral ulcers have cancerous lesions and their mortality rate is high. The ability to recognize oral ulcers at an early stage in a timely and effective manner is a very critical issue. In recent years, although there exists a small group of researchers working on these, the datasets are private. Therefore to address this challenge, in this paper a multi-tasking oral ulcer dataset (Autooral) containing two major tasks of lesion segmentation and classification is proposed and made publicly available. To the best of our knowledge, we are the first team to make publicly available an oral ulcer dataset with multi-tasking. In addition, we propose a novel modeling framework, HF-UNet, for segmenting oral ulcer lesion regions. Specifically, the proposed high-order focus interaction module (HFblock) performs acquisition of global properties and focus for acquisition of local properties through high-order attention. The proposed lesion localization module (LL-M) employs a novel hybrid sobel filter, which improves the recognition of ulcer edges. Experimental results on the proposed Autooral dataset show that our proposed HF-UNet segmentation of oral ulcers achieves a DSC value of about 0.80 and the inference memory occupies only 2029 MB. The proposed method guarantees a low running load while maintaining a high-performance segmentation capability. Apple replant disease (ARD) negatively affects plant growth and reduces yields in replanted orchards. In this study, biochar was applied to apple replant soil with Fusarium oxysporum. Our aim was to investigate whether biochar could promote plant growth and alleviate apple replant disease by reducing the growth of harmful soil microorganisms, changing soil microbial community structure and improving the soil environment. This experiment included five treatments: apple replant soil (CK), methyl bromide fumigation apple replant soil (FM), replant soil with biochar addition (2%), replant soil with F. oxysporum spore solution (8 × 107 spores · mL−1), and replant soil with biochar and F. oxysporum spore solution addition. Seedling biomass, the activity of antioxidant enzymes in the leaves and roots, and soil environmental variables were measured. Microbial community composition and community structure were analyzed using 16S rDNA and ITS2 gene sequencing. Biochar significantly reduced the abundance of F. oxysporum and increased soil microbial diversity and richness. Biochar also increased the soil enzyme activities (urease, invertase, neutral phosphatase, and catalase), the biomass (plant height, fresh weight, dry weight) and the activity of antioxidant enzymes (superoxide dismutase, peroxidase, and catalase). The root indexes of apple seedlings was also increased in replant soil by biochar. In sum, biochar promoted the growth of plants, improved the replant soil environment, and alleviated apple replant disease.

Biochar and humic acid also increased the soil enzymes activity (urease, invertase, neutral phosphatase, and catalase), the plant height, fresh weight, dry weight, the activity of antioxidant enzymes (superoxide dismutase, peroxidase, and catalase), and root indexes of apple seedlings increased in replant soil. In sum, We can use biochar combined with humic acid to alleviate apple replant disease.

Abstract In order to understand whether corn straw (CS) and straw-derived biochar (CB) can be used as soil amendments to alleviate soil contaminated with metal oxide nanoparticles, CS and CB prepared by pyrolysis at 300 °C (with a weight ratio of 2% and 5%) were added to two types of soil contaminated with copper oxide nanoparticles (CuO NPs) to study the effects of CS and CB application on soil properties, wheat seedling growth, and metal transport. Our results showed that both 2% and 5% additions of CS and CB enhanced the nitrogen, phosphorus, and potassium fertility of both soils. In red earth (acidic), the addition of 2% and 5% of CB significantly increased soil pH (9.24% ~ 18.4%), wheat growth (12.2% ~ 14.9%), chlorophyll content (significantly increased by 32.3% for 2% CB treatment), and decreased antioxidant enzyme activity and membrane lipid peroxidation. In fluvo-aquic soil (alkaline), CS and CB did not increase soil pH, only 5% of CB significantly promoted wheat growth (by 11.0%), and decreased antioxidant enzyme activity, whereas 2% of CB reduced chlorophyll content (by 22.2%). Furthermore, 2% and 5% of CS and CB affected the transport of Cu in wheat, decreasing the translocation factor in red earth but increasing it in fluvo-aquic soil. Principal component analysis showed that 5% of CB had the best alleviating effect on CuO NPs toxicity in red earth, compared to CS. Taken together, CB obtained by pyrolysis at 300 °C appears to be an effective soil amendment specifically for red earth. Abstract Pneumothorax is a condition involving a collapsed lung, which requires accurate segmentation of computed tomography (CT) images for effective clinical decision-making. Numerous convolutional neural network-based methods for medical image segmentation have been proposed, but they often struggle to balance model complexity with performance. To address this, we introduce the Efficient Feature Alignment Network (EFA-Net), a novel medical image segmentation network designed specifically for pneumothorax CT segmentation. EFA-Net uses EfficientNet as an encoder to extract features and a Feature Alignment (FA) module as a decoder to align features in both the spatial and channel dimensions. This design allows EFA-Net to achieve superior segmentation performance with reduced model complexity. In our dataset, our method outperforms various state-of-the-art methods in terms of accuracy and efficiency, achieving a Dice coefficient of 90.03%, an Intersection over Union (IOU) of 81.80%, and a sensitivity of 88.94%. Notably, EFA-Net has significantly lower FLOPs (1.549G) and parameters (0.432M), offering better robustness and facilitating easier deployment. Future work will explore the integration of downstream applications to enhance EFA-Net’s utility for clinicians and patients in real-world diagnostic scenarios. The purpose of the present study was to investigate the effect of additives on the fermentation properties of ensiled mixed alfalfa and perennial ryegrass silage in the karst terrain of Southwest China. A mixture of alfalfa and perennial ryegrass was ensiled at a ratio of 3:7 using three experimental treatments: (1) CK (without additives) and distilled water (5 mL kg−1 fresh weight (FW)); (2) FA and formic acid (88%) (5 mL kg−1 FW); and (3) LAB combined with the application of Lactiplantibacillus plantarum and Lentilactobacillus buchneri (2 × 107 cfu/g FW). All samples were packed manually into polyethylene bags, and three polyethylene bags from each treatment were sampled on days 7, 15, and 45. The findings demonstrated that the pH values of all the mixed silages gradually decreased during ensiling. The lactic acid (LA) and acetic acid (AA) contents increased gradually with ensiling time and peaked after 45 days of ensiling. After 45 days of ensiling, the FA and LAB groups effectively preserved the nutrient content of the mixed silage, which presented a reduced neutral detergent fiber and acid detergent fiber content (p < 0.05) and higher water-soluble carbohydrate content (p < 0.05) than the CK group. The fermentation quality of the mixed silages in the FA and LAB groups improved, as indicated by higher (p < 0.05) LA contents and lower (p < 0.05) pH and ammoniacal nitrogen contents after 45 days of ensiling compared to those in the CK group. As fermentation progressed, the abundance of harmful microorganisms (Hafnia obesumbacterium, Enterobacteriaceae, and Sphingomonas) and beneficial microorganisms (Lactiplantibacillus and Lentilactobacillus) decreased and increased, respectively. In addition, compared to those in the CK group, the FA group had higher abundances of “lipid metabolism” and “biosynthesis of antibiotics” and lower abundances of “membrane transport”. Briefly, the results of this study suggest that the incorporation of FA and LAB additives could improve the quality of fermented mixed silage, and that FA is better than LAB. This information is useful for combining forage resources to satisfy the requirements for high-protein feed and for manufacturing ruminant feed annually.

Increasing soil salinization can severely restrict local agricultural production. Planting alfalfa is considered an effective measure to ameliorate saline–alkali soil. However, it remains unclear how alfalfa planting years affect the sustained impact on soil and rhizosphere microecology. This study analyzed the effects of alfalfa planted 3, 6, and 8 years ago on soil physicochemical properties and key soil enzyme activities and investigated the rhizosphere microbial community structure and diversity. The results indicate that cultivating alfalfa plants for six years can improve soil physicochemical properties and enhance soil fertility to a certain extent. This is attributed to a higher abundance of plant growth-promoting bacteria, such as Bradyrhizobium and Allorhizobium, as well as degradation bacteria, such as Flavobacterium, Stenotrophomonas, Brevundimonas, and Massilia, in the rhizosphere of alfalfa plants. These microorganisms promote alfalfa growth, improve soil quality, and inhibit the accumulation of autotoxins. This not only maintains high alfalfa yields but also optimizes soil physicochemical properties and enzyme activity, facilitating more effective nutrient cycling and metabolic processes in the soil. However, extending plant growth to 8 years is not beneficial.

Summary: The generation of neurons from quiescent radial-glia-like neural stem cells (RGLs) in adult brain goes hand in hand with the modulation of cellular metabolism. However, it is still unclear how the exact metabolic program governs the balance between quiescent and activated RGLs. Here, we find that loss of mitochondrial D-2-hydroxyglutarate dehydrogenase (D2HGDH) leads to aberrant accumulation of D-2-hydroxyglutarate (D-2-HG) and impaired RGL activation. Mechanistically, accumulated D-2-HG bonds directly to ATP-citrate lyase and competitively inhibits its enzymatic activity, thereby reducing acetyl-CoA production and diminishing histone acetylation. However, administration of acetate restores the acetyl-CoA levels via acetyl-CoA synthetase-mediated catabolism and rescues the deficiencies in histone acetylation and RGL activation caused by loss of D2HGDH. Therefore, our findings define the role of cross talk between mitochondria and the nucleus via a mitochondrial metabolite, D-2-HG, the aberrant accumulation of which hinders the regulation of histone acetylation in RGL activation and attenuates continuous neurogenesis in adult mammalian brain.

Alfalfa cultivated in saline-alkaline soils exhibits a high buffering capacity but low carbohydrate content, posing challenges in the production of high-quality silage feed. This study investigated alfalfa plants grown under varying conditions: mild (QE, salt content 1–2%), moderate (ZE, salt content 2–3%), severe (HE, salt content 3–4%), and non-saline-alkaline (CON, salt content < 1%). Employing a two-factor experimental design, we analyzed the fluctuations in nutritional quality, microbial abundance, and community composition of different salt–alkaline alfalfa materials and silage feeds. Furthermore, we elucidated the fermentation mechanism involved in salt–alkaline alfalfa ensiling. Following a 60-day ensiling period, the ZE and HE treatments led to substantial reductions in pH, acetic acids (AA), branched-chain butyric acids (BA) content, facultative anaerobic bacteria, and Escherichia coli populations (p < 0.05). Conversely, the ZE and HE treatments increased lactic acid (LA) content and the population of lactic acid bacteria (LAB) (p < 0.05). Additionally, these treatments significantly mitigated protein losses in both raw alfalfa and silage feeds (p < 0.05), while remarkably augmenting the water-soluble carbohydrates (WSC), Na+, and K+ content of alfalfa materials. Sodium ions were found to exert a considerable influence on bacterial community composition during salt–alkaline alfalfa ensiling, with Enterococcus, Lactococcus, and Lactobacillus identified as the predominant fermentative microorganisms. Overall, moderately salt-alkaline alfalfa materials displayed optimal nutritional and fermentation quality, ensuring favorable nutritional attributes and fermentation outcomes under such conditions.

High-quality alfalfa is an indispensable resource for animal husbandry and sustainable development. Its nutritional quality changes dramatically during its life cycle and, at present, no molecular mechanisms for nutrient metabolic variation in alfalfa leaves at different growth stages have been clearly reported. We have used correlation and network analyses of the alfalfa leaf metabolome, proteome, and transcriptome to explore chlorophyll, flavonoid, and amino acid content at two development stages: budding stage (BS) and full-bloom stage (FBS). A high correlation between the expression of biosynthetic genes and their metabolites revealed significant reductions in metabolite content as the plant matured from BS to FBS. l-Glutamate, the first molecule of chlorophyll biosynthesis, decreased, and the expression of HemA, which controls the transformation of glutamyl-tRNA to glutamate 1-semialdehyde, was down-regulated, leading to a reduction in leaf chlorophyll content. Flavonoids also decreased, driven at least in part by increased expression of the gene encoding CYP75B1: flavonoid 3′-monooxygenase, which catalyzes the hydroxylation of dihydroflavonols and flavonols, resulting in degradation of flavonoids. Expression of NITRILASE 2 (NIT2) and Methyltransferase B (metB), which regulate amino acid metabolism and influence the expression of genes of the glycolysis-TCA pathway, were down-regulated, causing amino acid content in alfalfa leaves to decrease at FBS. This study provides new insights into the complex regulatory network governing the content and decrease of chlorophyll, amino acids, flavonoids, and other nutrients in alfalfa leaves during maturation. These results further provide a theoretical basis for the generation of alfalfa varieties exhibiting higher nutritional quality, high-yield cultivation, and a timely harvest.

ABSTRACT Purpose: This study aimed to develop a minimally invasive surgical procedure for laminar lift and posterior cervical laminoplasty via the intermuscular approach using a canine model. Methods: Six Alaskan dogs were used for developing the surgical approach. The bilateral laminae of C3-7 were cut with an ultrasonic osteotome and fixed with bilateral plates to maintain the lamina lifting and reshape a wider spinal canal. The important structures, such as ligaments, supraspinous ligaments, interspinous ligaments, and ligamentum flavum were preserved. The therapeutic effect was evaluated by preoperative and postoperative imaging results and neck mobility. Results: The surgical procedures were all successfully performed in the 6 animals. All the dogs survived well within 1 year of postoperative follow-up. The postoperative neck mobility was as good as the preoperative one. Computed tomography results showed that the anteroposterior diameter of the spinal canal was successfully enlarged and maintained well. Conclusions: The minimally invasive surgical procedure for laminar lift and posterior cervical laminoplasty via the intermuscular approach was feasible in a canine model, which might be applied in clinical practice.

Subculture, includes animation, comics, games (ACG), and idol fan culture, is popular among young generations in Japan. Previous studies have shown different psychological outcomes within different subcultural groups; however, underlying mechanisms remain unclear. This study proposes that subcultural identity may play a crucial role in mental health outcomes by interweaving social support and reputation. We examined the interplay between subcultural identity, social support, reputation, and different psychological outcomes through structural equation modeling (N = 300). Furthermore, we trained neural networks (NN) by applying a deep learning algorithm to predict psychological outcomes of different subcultures. The results suggest identity related to anime, idol, and hip-hop was positively associated with anxiety, aggression, depressive symptoms, and suicidal tendencies. By contrast, fashion and sports identities associated with no adverse or positive psychological outcomes. Perceived reputation mediates adverse psychological outcomes in the ACG, idol, and hip-hop groups. The highest accuracy in our NN reached 71%, indicating that NN could be an effective tool for predicting mental problems. Our work points up an urgent need to improve the mental health of the young generation by de-stigmatizing discriminated cultural groups.

Alfalfa cultivated in salt–alkali soil was used for fermentation, to which an inoculum of Lactobacillus plantarum YQM48 was added, to assess its effect on the feed quality and the microbial community structure of the fermented silage. A control was included without inoculum. The nutritional components, fermentation quality, pH, and microbial community of the silage were measured after 30 and 60 days of anaerobic fermentation. The results showed that after 30 and 60 days of fermentation in the presence of the inoculum, the content of water-soluble carbohydrates, crude protein content, and dry matter were all significantly higher than those of the control silage, the pH and butyric acid content were lower, and the content of lactic acid and acetic acid were higher (p < 0.05 for all). There was no significant difference in pH and butyric acid content between 30 and 60 days of fermentation in the presence of the inoculum (p > 0.05), while the lactic acid and acetic acid contents were significantly lower in the 60-day silage compared to the 30-day silage (p < 0.05). Fermentation reduced the abundance of Cyanobacteria and Proteobacteria (the abundant phyla in the fresh alfalfa), while the abundance of Firmicutes increased, reaching 92.3% after 60 days of fermentation with inoculum. The dominant genus in that sample was Lactobacillus (70.0%), followed by Enterococcus (12.9%), while fermentation of 60 days without inoculum resulted in only 29.2% Lactobacillus, together with 27.8% Cyanobacteria, and 12.2% Enterococcus. In summary, the addition of L. plantarum YQM48 can improve the nutritional components and fermentation quality of alfalfa silage cultivated on a salt-alkali soil.

Abstract Background Alfalfa (Medicago sativa L.) is one of the most important forage resources in the world due to its high nutritive value. However, its nutritional quality decreases during the transition from budding to flowering. Previous research revealed a decreased crude protein content and increased fibre content in alfalfa forage harvested at later maturity stages, leading to a reduction in nutritional quality. However, the reasons for this phenomenon have not been explained at the molecular level. Results In this study, leaves from the WL319HQ alfalfa cultivar were harvested at two developmental stages (budding and mid-flowering). The leaves were used to test the variable expression of proteins and metabolites during these stages. TMT-based quantitative proteomics and LC-MS/MS-based untargeted metabolomics methods were employed in this study. A total of 415 proteins and 49 metabolites showed at least a 1.2-fold difference in abundance during these stages. Most of the differentially expressed proteins and metabolites were involved in metabolic processes, including carbohydrate metabolism, starch and sucrose metabolism, phenylpropanoid biosynthesis, and biosynthesis of amino acids. Alfalfa leaves in mid-flowering contain less crude protein due to the decrease in L-glutamic acid content. Carbohydrate metabolism provides the raw material for the synthesis of hemicellulose, resulting in an increase in the hemicellulose content of the alfalfa leaves, leading to an increase in the NDF content. In addition, the increase in L-phenylalanine content could have provided the conditions necessary for lignin synthesis. These are the main factors leading to reductions in alfalfa relative feed value (RFV) and quality. Conclusions This study used joint proteomic and metabolomic analyses to elucidate the relationship between the reduction in the nutritional value of alfalfa and complex biological processes. This provides a theoretical basis for producing high-quality alfalfa hay and sets the stage for further research.

Abstract Background It is predicted that climate change may increase the risk of local droughts, with severe consequences for agricultural practices. Methods Here we report the influence of drought on alfalfa yields and nutritional composition, based on artificially induced drought conditions during two field experiments. Two types of alfalfa cultivars were compared, Gold Queen and Suntory. The severity and timing of drought periods were varied, and the crop was harvested either early during flowering, or late at full bloom. Results The obtained dry mass yields of Gold Queen were higher than Suntory, and the first was also more resistant to drought. Early harvest resulted in higher yields. Decreases in yields due to water shortage were observed with both cultivars, and the fraction of crude protein (CP) decreased as a result of drought stress; this fraction was higher in Gold Queen than in Suntory and higher in early harvest compared to late harvest. Severe drought late in spring had the highest effect on CP content. The fraction of fibre, split up into neutral detergent fibre (NDF) and acid detergent fibre (ADF) increased as a result of drought and was lower in early compared to late harvested plants. Suntory alfalfa produced higher fibre fractions than Gold Queen. The fraction of water-soluble carbohydrates (WSC) was least affected by drought. It was consistently higher in Gold Queen compared to Suntory alfalfa, and late harvest resulted in higher WSC content. Conclusions In combination, these results suggest that the nutritive value of alfalfa will likely decrease after a period of drought. These effects can be partly overcome by choosing the Gold Queen cultivar over Suntory, by targeted irrigation, in particular in late spring, and by harvesting at an earlier time.

The prediction of human mobility can facilitate resolving many kinds of urban problems, such as reducing traffic congestion, and promote commercial activities, such as targeted advertising. However, the requisite personal GPS data face privacy issues. Related organizations can only collect limited data and they experience difficulties in sharing them. These data are in “isolated islands” and cannot collectively contribute to improving the performance of applications. Thus, the method of federated learning (FL) can be adopted, in which multiple entities collaborate to train a collective model with their raw data stored locally and, therefore, not exchanged or transferred. However, to predict long-term human mobility, the performance and practicality would be impaired if only some models were simply combined with FL, due to the irregularity and complexity of long-term mobility data. Therefore, we explored the optimized construction method based on the high-efficient gradient-boosting decision tree (GBDT) model with FL and propose the novel federated voting (FedVoting) mechanism, which aggregates the ensemble of differential privacy (DP)-protected GBDTs by the multiple training, cross-validation and voting processes to generate the optimal model and can achieve both good performance and privacy protection. The experiments show the great accuracy in long-term predictions of special event attendance and point-of-interest visits. Compared with training the model independently for each silo (organization) and state-of-art baselines, the FedVoting method achieves a significant accuracy improvement, almost comparable to the centralized training, at a negligible expense of privacy exposure.

This study investigated the impact of endogenous sodium and potassium ions in plants on the quality of alfalfa silage, as well as the stability of bacterial communities during fermentation. Silage was produced from the fermented alfalfa, and the chemical composition, fermentation characteristics, and microbiome were analyzed to understand their interplay and impact on silage fermentation quality. The alfalfa was cultivated under salt stress with the following: (a) soil content of <1‰ (CK); (b) 1‰-2‰ (LP); (c) 2‰-3‰ (MP); (d) 3‰-4‰ (HP). The results revealed that the pH of silage was negatively correlated with the lactic acid content. With the increase of lactic acid (LA) content increased (26.3-51.0 g/kg DM), the pH value decreased (4.9-5.3). With the increase of salt stress, the content of Na+ in silage increased (2.2-5.4 g/kg DM). The presence of endogenous Na+ and K+ ions in plants significantly affected the quality of alfalfa silage and the dynamics of bacterial communities during fermentation. Increased salt stress led to changes in microbial composition, with Lactococcus and Pantoea showing a gradual increase in abundance, especially under high salt stress. Low pH inhibited the growth of certain bacterial genera, such as Pantoea and Pediococcus. The abundance of Escherichia-Shigella and Comamonas negatively correlated with crude protein (CP) content, while Enterococcus and Lactococcus exhibited a positive correlation. Furthermore, the accumulation of endogenous Na+ in alfalfa under salt stress suppressed bacterial proliferation, thereby reducing protein degradation during fermentation. The pH of the silage was high, and the LA content was also high. Silages from alfalfa under higher salt stress had higher Na+ content. The alpha diversity of bacterial communities in alfalfa silages showed distinct patterns. Desirable genera like Lactococcus and Lactobacillus predominated in silages produced from alfalfa under salt stress, resulting in better fermentation quality. [ABSTRACT FROM AUTHOR]
