Abstract Background Cereal/legume intercropping can enhance crop productivity and improve soil health in dryland farming. However, little is known about soil biological health under maize/peanut intercropping. The aim of this study was to assess soil biological health based on nematode communities in a maize/peanut intercropping system. Methods We conducted a field experiment with different planting patterns, including monoculture maize (M), monoculture peanut (P), and maize intercropped with peanut (IM, intercropped maize; IP, intercropped peanut) to determine the influence on soil biological health. We measured soil physicochemical properties and nematode communities, and employed exploratory factor analysis combined with cumulative normal distribution curve scoring to identify potential soil biological health traits. Results The intercropped maize gave the highest plant parasitic nematode abundance, trophic diversity index, evenness index, and structure index. The monoculture peanut gave the highest enrichment index and least plant parasitic nematode abundance, trophic diversity index, Shannon diversity index, evenness index, structure index, and channel index. We identified four soil biological health traits, including basic nutrients and biodiversity, food web complexity, slow energy channel, and fast energy channel, mainly represented by soil ammonium nitrogen and Shannon diversity index, structural index and omnivore-predator nematode abundance, fungivorous nematode abundance and plant parasitic nematode abundance, microbial biomass carbon and bacterivorous nematode abundance, respectively. The intercropping systems improved the comprehensive score of soil biological health, especially maize intercropping soil. Intercropping maize and intercropping peanut significantly improved oil biological health traits representing the food web complexity compared with the corresponding monoculture soils. Conclusions Our results indicate that soil nematode and physicochemical indicators reflect different soil biological health traits. Among those traits, the improvement of basic nutrients and biodiversity and the complexity of the food web were the main reasons for improving soil biological health through the intercropping system.

Abstract Melia azedarach is a species of enormous value of pharmaceutical industries. Although the chloroplast genome of M. azedarach has been explored, the information of mitochondrial genome (Mt genome) remains surprisingly limited. In this study, we used a hybrid assembly strategy of BGI short-reads and Nanopore long-reads to assemble the Mt genome of M. azedarach. The Mt genome of M. azedarach is characterized by two circular chromosomes with 350,142 bp and 290,387 bp in length, respectively, which encodes 35 protein-coding genes (PCGs), 23 tRNA genes, and 3 rRNA genes. A pair of direct repeats (R1 and R2) were associated with genome recombination, resulting in two conformations based on the Sanger sequencing and Oxford Nanopore sequencing. Comparative analysis identified 19 homologous fragments between Mt and chloroplast genome, with the longest fragment of 12,142 bp. The phylogenetic analysis based on PCGs were consist with the latest classification of the Angiosperm Phylogeny Group. Notably, a total of 356 potential RNA editing sites were predicted based on 35 PCGs, and the editing events lead to the formation of the stop codon in the rps10 gene and the start codons in the nad4L and atp9 genes, which were verified by PCR amplification and Sanger sequencing. Taken together, the exploration of M. azedarach gap-free Mt genome provides a new insight into the evolution research and complex mitogenome architecture.

Long-term continuous cropping can lead to the deterioration of soil environment and the decrease of soil productivity. However, the biological mechanism on the negative effects of long-term continuous cropping has not been extensively explored. Soil nematode food web with multiple trophic levels play critical roles in nutrient cycling and energy flowing in the agroecosystem. Quantifying the carbon flux through different trophic channels within the nematode food web can indicate how continuous cropping influences carbon cycling in the agroecosystem by altering soil biota communities. Therefore, the effects of continuous peanut cropping with different years (1, 5, 20, and 30 years) on soil properties, soil nematode community composition, carbon flux within nematode food web and crop yields were investigated. Results showed that soil pH significantly decreased with increasing continuous cropping years. Differently, soil organic carbon and total nitrogen were the highest in 20-year, and decreased in 30-year of continuous cropping. As continuous cropping years increased, the relative abundance of microbivorous nematodes decreased and that of plant parasites reached as high as 76.11 % and 68.22 % in 20- and 30-year, respectively. Pearson correlation analysis revealed the nematode diversity, the carbon flux uniformity and peanut yield had a significant negative correlation with continuous cropping years. The random forest model indicated that the soil pH and the relative abundance of plant parasites were the key influence factor of the carbon flux uniformity within soil nematode food web. Outbreaks of plant parasites lead to the disruption of carbon flux uniformity within soil nematode food web, which can increase the risk of peanut yield decline after long-term continuous cropping. In conclusion, continuous peanut cropping changed soil properties, reduced soil nematode diversity, and disturbed the multitrophic carbon flux complementarity and uniformity in soil nematode food web, ultimately limiting the crop productivity. This study enhances our understanding of the importance of the resource transfers among soil food web in maintaining sustainable agroecosystem productivity.

Abstract The objective of this study was to explore the positive influence and potential mechanism of Erianin on the recovery of brain cells following a traumatic brain injury (TBI). TBI rat models were prepared and treated with Erianin injection via tail vein. The assessment included evaluating the rats' levels of oxidative stress, inflammation, neuronal damage, mitochondrial damage, neuronal regeneration, transformation of pro-inflammatory microglial cells, activation status of the ERK signal pathway, and the functionality of their learning and memory. After administering Erianin, there was a suppression of oxidative stress, inflammation, nerve cell damage, and mitochondrial damage in the TBI rats. Additionally, there was an increase in neuronal regeneration in the cortex and hippocampus, inhibition of pro-inflammatory microglial cell transformation in the cortex, improvement in learning and memory function in TBI rats, and simultaneous inhibition of the activation of the ERK1/c-Jun signal pathway. The findings suggest that Erianin has the potential to reduce oxidative stress and inflammatory reaction in rats with TBI, safeguard nerve cells against apoptosis, stimulate the growth of new neural cells, ultimately enhancing the cognitive abilities and memory function of the rats. The inhibition of the ERK signaling pathway could be closely associated with these effects.

Objective The Hetaoba area in Inner Mongolia has superior uranium metallogenic conditions.Grea tore-prospecting results have been achieved in the eastern part of the study area.However, breakthrough has not been made in the western area. Uranium mineralization is significantly controlled by faults and fractured zones. To identify the distributions of fault framework, structural fracture and alteration zones, and further to infer the favorable metallogenesis areas, geophysical data are provided to explore the uranium metallogenic potential. Methods Through the comprehensive applications of three geophysical methods, including audiomagnetotelluric surveys, soil radon measurements, and ground high-precision magnetic measurements, soil radon anomalies are delineated in the study area. The northwest fault F13 shows a northeast- and northwest-trending with a Chinese word "Tian" grid shape, while the southeast fault F12 presents a northeast-trending grid shape. The faults of F13 and F12 present nearly north-south and northwest trendings in the pinched zones.There are six structural alterated and fractured zones to be inferred. Results Based on the characteristics of uranium mineralization, it is inferred that the structural alteration and fracture zones of S1, S2, S3 and S4 are located at the intersection and pinched zone of faults, with a large scale. Multiple soil radon anomalies are distributed with an anomaly amplitude up to 35 000 Bq/m3, which are the favorable areas for metallogenesis. Conclusion After verification of the drilling results, the structural alteration, fracture zones and thick uranium ore bodies are discovered in the deep of borehole ZKH24. Many industrial uranium ore holes are discovered in the S2 structural alteration zone. Borehole ZKH7-1 has good ore findings, confirming the origin place of the uranium ore. The study indicates that the comprehensive application of the three geophysical methods has effective results in uranium ore exploration and can be promoted to uranium ore prospecting.

Coronavirus infectious disease 2019 (COVID-19), caused by severe acute respiratory virus type 2 (SARS-CoV-2), has caused a global public health crisis. As an RNA virus, the high gene mutability of SARS-CoV-2 poses significant challenges to the development of broad-spectrum vaccines and antiviral therapeutics. There remains a lack of specific therapeutics directly targeting SARS-CoV-2. With the ability to efficiently inhibit the expression of target genes in a sequence-specific way, small interfering RNA (siRNA) therapy has exhibited significant potential in antiviral and other disease treatments. In this work, we presented a highly effective self-assembled siRNA nanoparticle targeting multiple highly conserved regions of SARS-CoV-2. The siRNA sequences targeting viral conserved regions were first screened and evaluated by their thermodynamic features, off-target effects, and secondary structure toxicities. RNA motifs including siRNA sequences were then designed and self-assembled into siRNA nanoparticles. These siRNA nanoparticles demonstrated remarkable uniformity and stability and efficiently entered cells directly through cellular endocytic pathways. Moreover, these nanoparticles effectively inhibited the replication of SARS-CoV-2, exhibiting a superior inhibitory effect compared to free siRNA. These results demonstrated that these self-assembled siRNA nanoparticles targeting highly conserved regions of SARS-CoV-2 represent highly effective antiviral candidates for the treatment of infections, and are promisingly effective against current and future viral variants.

Abstract Background Cases of multiple cerebral aneurysms are rare. In this case report, we describe a male patient with multiple, enlarging, and ruptured aneurysms. The two aneurysms were believed to be dissecting aneurysms. Case description A 47-year-old man presented with left limb paralysis. Magnetic resonance imaging revealed a cerebral infarction. Digital subtraction angiography (DSA) identified an aneurysm and occlusion in the right middle cerebral artery (MCA). The MCA aneurysm was remarkably enlarged on the eighth day after cerebral ischemia and was treated using endovascular techniques. Two weeks after the endovascular treatment, the patient experienced a severe headache and became comatose, and a subarachnoid re-hemorrhage was confirmed. The fourth DSA revealed an enlarging dissecting aneurysm in the posterior cerebral artery. The patient died without further treatment. Conclusion Some dissecting aneurysms rapidly enlarge and rupture.

Aglaia odorata, native to Guangdong, Guangxi, and Hainan provinces in China, has long been utilized as an herbal remedy in ancient China. In this study, we assembled and annotated the complete mitochondrial genome (mitogenome) of A. odorata, which spans a total length of 537,321 bp. Conformation of the A. odorata recombination was verified through PCR experiments and Sanger sequencing. We identified and annotated 35 protein-coding genes (PCGs), 22 tRNA genes, and 3 rRNA genes within the mitogenome. Analysis of repeated elements revealed the presence of 192 SSRs, 29 pairs of tandem repeats, and 333 pairs of dispersed repeats in the A. odorata mitogenome. Additionally, we analyzed codon usage and mitochondrial plastid DNAs (MTPTs). Twelve MTPTs between the plastome and mitogenome of A. odorata were identified, with a combined length of 2,501 bp, accounting for 0.47% of the mitogenome. Furthermore, 359 high-confidence C to U RNA editing sites were predicted on PCGs, and four selected RNA editing sites were specially examined to verify the creation of start and/or stop codons. Extensive genomic rearrangement was observed between A. odorata and related mitogenomes. Phylogenetic analysis based on mitochondrial PCGs were conducted to elucidate the evolutionary relationships between A. odorata and other angiosperms.

We theoretically demonstrate a scheme for efficiently generating near-circularly polarized isolated attosecond pulses from mixed gases driven by non-integer frequency ratio two-color multicycle laser fields. It is shown that isolated attosecond pulses with a duration of 310 as can be generated, exhibiting non-zero ellipticity with values reaching as high as 0.9. Further analysis reveals that the ellipticities of these isolated attosecond pulses are attributed to the interference of high-order harmonics generated from different components of the mixture. When the components in the mixture have opposite orbital parity, constructive and destructive interference will occur for left- and right-circularly polarized harmonic components, or vice versa. Additionally, the temporal profile of the isolated attosecond pulse depends solely on the driving fields, while the polarization is determined by the gas mixture. The obtained elliptically polarized isolated attosecond pulses may serve as a promising tool for manipulating ultrafast dynamics in magnetic materials and chiral media.

The current therapies used to treat sepsis are ineffective and sepsis-related mortality is high. Therefore, it is important to find new treatment options that reduce mortality from that disease. In the present study, the effects of probiotic pretreatment in septic rats were studied on morality, hematological parameters, oxidative stress, antioxidant capacity, and inflammation. After preparation of rats and administration of probiotic VSL # 3, cecal ligation and puncture (CLP) surgery was used to induce sepsis. Then, the animals were divided into four groups: sham, sham + VSL # 3, septic, and septic + VSL # 3. Sepsis-related mortality, hematological parameters, and biochemical factors were assessed. Also, the expression of inflammatory cytokines IL-6, TNF-α and IL-10 in serum were studied using enzyme-linked immunosorbent assay (ELISA). Then, the NF-κB gene and protein expression in intestinal tissue was studied by RT-qPCR and western blot techniques, respectively. Daily pretreatment with VSL # 3 in septic rats resulted in a 20 % reduction in mortality over 10 days and a significant modulation in hematological parameters. Evaluation of oxidant and antioxidant indices showed a decrease in oxidative stress and an increase in antioxidant capacity in septic rats receiving VSL # 3. Also, decreased expressions of inflammatory cytokines IL-6, TNF-α, IL-10, and NF-κB were observed in septic rats receiving VSL # 3. In general, it can be concluded that the administration of probiotic VSL # 3, which resulted in the improved survival of septic rats and better antioxidant and anti-inflammatory conditions, can be considered as a new adjutant therapeutic approach in the treatment of sepsis.

Abstract There are currently two main treatment strategies mainly for high-risk patients: percutaneous transluminal angioplasty and stenting (PTAS) and aggressive medical management (AMM). However, the choice between PTAS or AMM remains controversial for patients with stroke or intracranial atherosclerotic stenosis (ICAS). The investigators searched the PubMed, Web of Science, Embase, Scopus, and Cochrane library databases. Randomized controlled trial (RCT) comparing PTAS and AMM for patients with stroke or ICAS were selected. RevMan 5.3 was used to analyze the results and assess risk of bias. The primary endpoints are stroke and death within 30 days after enrollment, or ischemic stroke in the territory of the qualifying artery beyond 30 days, and entire follow-up endpoints. The secondary outcomes were the disabling or fatal stroke, and incidence of death within 3 years. Four studies, 989 patients were included in this article. The AMM group was superior in the entire follow-up endpoint (OR 0.56; 95% CI 0.40, 0.79). The AMM also better in primary endpoint within 30 days (OR 0.32; 95% CI 0.17, 0.61). There was no significant difference beyond 30 days (OR 1.08; 95% CI 0.63, 1.86). The remaining outcomes, such as stroke and death, were not significantly different (P > 0.05). This meta-analysis shows AMM is significantly more effective than PTAS in subjects with ICAS due to the high rate of periprocedural stroke (OR 0.32; 95% CI 0.17, 0.61) and stroke during the entire follow-up (OR 0.56; 95% CI 0.40, 0.79) associated with PTAS. Furthermore, PTAS offers no additional benefits over AMM beyond 30 days (OR 1.08; 95% CI 0.63, 1.86).

Alpinia oxyphylla is a traditional Chinese medicinal plant with a medicinal history of more than 1700 years. Ring leaf blight (RLB) disease, caused by pestalotioid species, is an important disease of A. oxyphylla, seriously affecting the yield and quality of its fruits. The causal agent of RLB disease has not been systematically identified or characterized yet. In this study, thirty-six pestalotioid strains were isolated from the leaves and stems of A. oxyphylla that was collected from six cities of Hainan province, China. Based on the multi-locus phylogeny (ITS, tef-1α and tub2) and morphological characteristic analyses, seventeen species belonging to three genera (Neopestalotiopsis, Pestalotiopsis and Pseudopestalotiopsis) were identified, and six new species (N. baotingensis, N. oblatespora, N. olivaceous, N. oxyphylla, N. wuzhishanensis and N. yongxunensis) were described. Pathogenicity tests revealed that strains of Neopestalotiopsis species caused more severe ring leaf blight on A. oxyphylla than strains of Pestalotiopsis and Pseudopestalotiopsis under wounded inoculation conditions.

Abstract Inoculation with soil from different ecosystems can induce changes in plant and soil communities and promote the restoration of degraded ecosystems. However, it is unknown how such inoculations influence the plant and soil communities, how much inoculum is needed, and whether inocula collected from similar ecosystems will steer soil and plant communities in different directions. We conducted a three-year soil inoculation experiment at a degraded grassland and used two different soil inocula both from grasslands with three inoculation rates. We measured the development of the soil and plant communities over a period of three years. Our results show that soil inoculation steers the soil microbiome and plant communities at the inoculated site into different directions and these effects were stronger with higher amount of soil used to inoculate. Network analyses showed that inoculation with upland meadow soil introduced more genera occupying the central position in the biotic network and resulted in more complex networks in the soil than inoculation with meadow steppe soil. Our findings emphasize that there are specific effects of donor soil on soil microbiomes as well as plant communities and that the direction and speed of development depend on the origin and the amount of soil inoculum used. Our findings have important implications for the restoration of biodiversity and ecosystem functioning in degraded grassland ecosystems.

Plants have evolved two layers of protection against biotic stress: PAMP-triggered immunity (PTI) and effector-triggered immunity (ETI). The primary mechanism of ETI involves nucleotide-binding leucine-rich repeat immune receptors (NLRs). Although NLR genes have been studied in several plant species, a comprehensive database of NLRs across a diverse array of species is still lacking. Here, we present a thorough analysis of NLR genes across 100 high-quality plant genomes (PlantNLRatlas). The PlantNLRatlas includes a total of 68,452 NLRs, of which 3,689 are full-length and 64,763 are partial-length NLRs. The majority of NLR groups were phyletically clustered. In addition, the domain sequences were found to be highly conserved within each NLR group. Our PlantNLRatlas dataset is complementary to RefPlantNLR, a collection of NLR genes which have been experimentally confirmed. The PlantNLRatlas should prove helpful for comparative investigations of NLRs across a range of plant groups, including understudied taxa. Finally, the PlantNLRatlas resource is intended to help the field move past a monolithic understanding of NLR structure and function.

Abstract Background and purpose Cerebral ischemia‐reperfusion (I/R) injury is a major factor underlying the high mortality and morbidity rates in stroke patients. Our previous study found that the combination of Astragalus membranaceus extract and ligustrazine (Ast+Lig) treatment could protect brain tissues against inflammation in rats with thrombolytic cerebral ischemia. Activation of N‐methyl‐D‐aspartate receptors (NMDAR) is implicated in brain damage induced by cerebral I/R injury. Methods We used in vivo and in vitro models of cerebral I/R injury for middle cerebral artery occlusion/reperfusion in mice and oxygen‐glucose deprivation/reoxygenation in primary rat cerebral cortical neurons to evaluate the protective effects of Ast+Lig on cerebral I/R injury, and whether the protective mechanism was related to the regulation of NMDAR‐ERK/CREB signaling. Results Treatment with Ast+Lig, or MK‐801 (an inhibitor of NMDAR) significantly ameliorated neurological deficits, decreased infarct volumes, suppressed neuronal damage and Ca2+ influx, and maintained the mitochondrial membrane potential in vivo and in vitro following cerebral I/R injury based on 2,3,5‐triphenyl tetrazolium chloride staining, immunohistochemistry, and immunofluorescent staining. Furthermore, treatment with Ast+Lig evidently prevented the upregulation of NR2B, but not NR2A, in vivo and in vitro following cerebral I/R injury based on western blotting and reverse transcription‐quantitative PCR analyses. Moreover, treatment with Ast+Lig significantly increased the phosphorylation of ERK and CREB, as well as increasing their mRNA expression levels in vivo and in vitro following cerebral I/R injury. Conclusions The overall results thus suggest that the Ast+Lig combination conferred neuroprotective properties against cerebral I/R injury via regulation of the NR2B‐ERK/CREB signaling pathway.

Crop diversification can restore land fertility and increase smallholder farmers’ resistance to climate change and market fluctuations, particularly for female farmers who are more vulnerable to the impact of these risks. To understand planting situation, motivation and influencing factors of crop diversification of smallholder farmers based on gender differences, we conducted a detailed survey of 262 smallholder farmers (118 female and 144 male) in the Shaanxi Plain of China. On average, male-headed households planted 2.27 species on 0.47 ha of land, whereas female-headed households planted 1.57 species on 0.37 ha of land. The main factors motivating smallholder farmers to diversify their crops are increased income and reduced economic loss of market fluctuation; the main obstacle is farmer reliance on traditional planting methods. We found significant gender differences following the expanded theory of planned behavior model. Attitude (standardized path coefficient (SPC) = 0.32), subjective norms (SPC = 0.26), perceived behavior control (SPC = 0.32) and environmental knowledge (SPC = 0.27) significantly impact male-headed households’ willingness of crop diversification. In contrast, female-headed households are most significantly influenced by perceived behavior control (SPC = 0.42), followed by attitude (SPC = 0.34) and subjective norms (SPC = 0.21), whereas environmental knowledge has no significant effect on their willingness to diversify their crops. Female-headed households are sensitive to climate change, but their low education, less access to agricultural extension and weak family decision-making power contribute to their unwillingness and inability to reduce risks by adopting crop diversification. Agricultural credit, crop diversification training, and efficient agricultural extension services are recommended for increasing farmer’s adoption of crop diversification and risk alleviation.

In view of the limitations of the current code based on the equivalent beam-column method with the “rod mode” instead of the “arch mode” for the calculation of concrete-filled steel tube arch bridges, this paper takes the real bearing mechanism of the arch as the starting point and analyzes the different bearing mechanisms of the arch and eccentric pressurized column. The concrete-filled steel tube arch model test was carried out to analyze the deformation state and damage mode, and the geometric non-linear bending moment of the measured arch was compared with the bending moment value calculated by the eccentricity increase coefficient of the “rod mode.” The results showed that the transfer of internal force is from the axial force to the arch axis, causing the vertical reaction force and horizontal thrust. However, the eccentric compression column only produced the vertical force at the bottom and combines with the lateral deformation indirectly generated by the eccentric distance. In addition, the deformation stage of the arch is basically the same as that of the eccentric compression column. The final failure mode of the arch is 4-hinge damage, and the final failure mode of the eccentric compression column is single-hinge damage. The preliminary geometric non-linear bending moment value obtained by the two modes accords well. Therefore, the main factors for the difference in the bearing mechanism between the two modes are different force structures, force transmission routes, and sources of deformation. Due to the difference in the bearing mechanism, the final failure mode is different, and the deformation ability of the arch is weakened by using the “rod mode” instead of the “arch mode.” The geometric non-linear bending moment of the control section calculated by the eccentricity increase coefficient is conservative, but the influence of the geometric non-linearity of other sections is not considered enough.

PurposeThe lung is the most common distant metastatic organ in patients with endometrial cancer (EC) but is rarely reported. This study examines the association between clinical characteristics and overall survival (OS) in EC with lung metastasis.MethodsPatients with EC who had accompanying lung metastasis were selected from the Surveillance, Epidemiology, and End Results (SEER) database between 2010 and 2017. Univariate and multivariate Cox regression were used to estimate hazard ratios (HRs) and 95% confidence intervals (95% CIs) and assess OS outcomes related to EC with lung metastasis. A Cox proportional hazards nomogram model for OS was constructed and validated. The calibration plot, receiver operating characteristic (ROC) curve and decision curve analysis (DCA) were used to evaluate the discriminative ability and clinical benefit of the novel nomogram. Kaplan–Meier curves and scatter diagram analysis were used to investigate the risk stratifications of the nomogram.ResultsOverall, 1542 EC patients with lung metastasis between 2010 and 2017 were included and randomly divided into training and validation cohorts. A nomogram model was constructed using the clinical characteristics of tumor grade, histological type, surgery, adjuvant chemotherapy, adjuvant radiation, brain metastasis and liver metastasis. The concordance indexes (C-indexes) were 0.750 (95% CI, 0.732-0.767) and 0.743 (95% CI, 0.719-0.767) for the training cohort and validation cohort, respectively. Calibration plots and DCA showed good clinical applicability of the nomogram. The areas under the curves (AUCs) were 0.803 and 0.766 for 1-year and 3-year OS, respectively, indicating that the nomogram model had a stable discriminative ability. An online calculator of our nomogram is available on the internet at https://endometrialcancer.shinyapps.io/DynNomapp/. Additionally, patients in the high-risk group had a significantly worse OS than those in the low-risk group.ConclusionAn easy-to-use, highly accurate nomogram was developed for predicting the prognosis of EC patients with lung metastasis.

The interactions between non-host roots and pathogens may be key to the inhibition of soilborne pathogens in intercropping systems. Fennel (Foeniculum vulgare) can be intercropped with a wide range of other plants to inhibit soilborne pathogens in biodiversity cultivation. However, the key compounds of fennel root exudates involved in the interactions between fennel roots and pathogens are still unknown. Here, a greenhouse experiment confirmed that intercropping with fennel suppressed pepper (Capsicum annuum) blight disease caused by Phytophthora capsici. Experimentally, the roots and root exudates of fennel can effectively interfere with the infection process of P. capsici at rhizosphere soil concentrations by attracting zoospores and inhibiting the motility of the zoospores and germination of the cystospores. Five terpene compounds (D-limonene, estragole, anethole, gamma-terpenes, and beta-myrcene) that were identified in the fennel rhizosphere soil and root exudates were found to interfere with P. capsica infection. D-limonene was associated with positive chemotaxis with zoospores, and a mixture of the five terpene compounds showed a strong synergistic effect on the infection process of P. capsici, especially for zoospore rupture. Furthermore, the five terpene compounds can induce the accumulation of reactive oxygen species (ROS), especially anethole, in hyphae. ROS accumulation may be one of the antimicrobial mechanisms of terpene compounds. Above all, we proposed that terpene compounds secreted from fennel root play a key role in Phytophthora disease suppression in this intercropping system.

Abstract Grasslands provide a number of ecosystem services for human society. Degradation of grasslands results in the loss of biodiversity and leads to the deterioration of ecosystem functions. In order to accurately assess the influence of grassland degradation on belowground ecosystems, we conducted experiments on a temperate steppe with different levels of degradation and investigated the influence of degradation on soil quality and soil biotic communities. Our results showed that grassland degradation significantly decreased soil quality, with lower values of soil quality index (SQI) observed in the degraded grassland than the meadow steppe and the grassland from the forest-steppe ecotone. Changes in the SQI along the grassland degradation gradient were positively correlated with soil carbon stock and the aboveground biomass, and negatively correlated with the root shoot ratio. Nematode trophic diversity and the ratio of fungal to bacterial PLFA were lower in the degraded grassland than the grassland from the forest-steppe ecotone. The dissimilarities in soil microbial and nematode community composition increased with the changes in soil quality index. Our results indicate that soil quality index based on the minimum data sets could effectively assess the influence of grassland degradation on soil biodiversity and ecosystem function. In order to effectively restore degraded grasslands, the key contributors to the soil quality, such as soil carbon, should be taken on priority basis for revitalizing the soil biodiversity and ecosystem function.

Background and PurposeAbout 20.1% of intracranial aneurysms (IAs) carriers are multiple intracranial aneurysms (MIAs) patients with higher rupture risk and worse prognosis. A prediction model may bring some potential benefits. This study attempted to develop and externally validate a dynamic nomogram to assess the rupture risk of each IA among patients with MIA.MethodWe retrospectively analyzed the data of 262 patients with 611 IAs admitted to the Hunan Provincial People's Hospital between November 2015 and November 2021. Multivariable logistic regression (MLR) was applied to select the risk factors and derive a nomogram model for the assessment of IA rupture risk in MIA patients. To externally validate the nomogram, data of 35 patients with 78 IAs were collected from another independent center between December 2009 and May 2021. The performance of the nomogram was assessed in terms of discrimination, calibration, and clinical utility.ResultSize, location, irregular shape, diabetes history, and neck width were independently associated with IA rupture. The nomogram showed a good discriminative ability for ruptured and unruptured IAs in the derivation cohort (AUC = 0.81; 95% CI, 0.774–0.847) and was successfully generalized in the external validation cohort (AUC = 0.744; 95% CI, 0.627–0.862). The nomogram was calibrated well, and the decision curve analysis showed that it would generate more net benefit in identifying IA rupture than the “treat all” or “treat none” strategies at the threshold probabilities ranging from 10 to 60% both in the derivation and external validation set. Size, neck width, and location are the primary risk factors for ruptured IAs.

We theoretically investigated frustrated tunneling ionization (FTI) driven by spatially inhomogeneous strong laser fields induced by surface plasmon resonance within a bow-tie metal nanostructure. The results show that the FTI probability and the principal quantum number distribution exhibit similar oscillatory behavior as a function of the pulse duration. Our analysis reveals that the periodic defocusing and refocusing of the electron spatial distribution due to the inhomogeneous laser field is responsible for the oscillatory structures. In addition, the initial tunneling coordinates and the angular momentum distributions of the FTI events and theirs pulse duration dependence are also explored. Moreover, our results show that the frequency of the oscillatory structures depends sensitively on the electron quiver amplitude and the inhomogeneity strength. Thus, the electron quiver amplitude and the size of the gap between bow-tie nanostructure are useful and efficient knobs for controlling the yield and properties of exited Rydberg states.

During metabolic reprogramming, glioma cells and their initiating cells efficiently utilized carbohydrates, lipids and amino acids in the hypoxic lesions, which not only ensured sufficient energy for rapid growth and improved the migration to normal brain tissues, but also altered the role of immune cells in tumor microenvironment. Glioma cells secreted interferential metabolites or depriving nutrients to injure the tumor recognition, phagocytosis and lysis of glioma-associated microglia/macrophages (GAMs), cytotoxic T lymphocytes, natural killer cells and dendritic cells, promoted the expansion and infiltration of immunosuppressive regulatory T cells and myeloid-derived suppressor cells, and conferred immune silencing phenotypes on GAMs and dendritic cells. The overexpressed metabolic enzymes also increased the secretion of chemokines to attract neutrophils, regulatory T cells, GAMs, and dendritic cells, while weakening the recruitment of cytotoxic T lymphocytes and natural killer cells, which activated anti-inflammatory and tolerant mechanisms and hindered anti-tumor responses. Therefore, brain-targeted metabolic therapy may improve glioma immunity. This review will clarify the metabolic properties of glioma cells and their interactions with tumor microenvironment immunity, and discuss the application strategies of metabolic therapy in glioma immune silence and escape.

Ceratocystis manginecans causes mango wilt with significant economic losses. In the infection court, cerato-platanin (CP) family proteins (CPPs) are believed to involve in pathogenesis but has not been determined in C. manginecans. To confirm this function, a CP protein (CmCP) of C. manginecans was characterized in this study. A protoplast of C. manginecans was prepared by treating its mycelia with driselase and lysing enzymes. The cmcp gene was edited using CRISPR/Cas-U6-1 expression vectors in 60% PEG and 50 μg/mL hygromycin B in the medium, resulting in mutants with cmcp deletion (Δcmcp). A complemented mutant (Δcmcp-C) was obtained by transforming cmcp to Δcmcp. Both Δcmcp and Δcmcp-C were characterized by comparing them with a wild-type strain on morphology, mycelial growth, conidial production and pathogenicity. Additionally, cmcp was transformed and expressed in Pichia pastoris, and the derived recombinant protein CmCP caused a severe necrosis on Nicotiana tabacum leaves. CmCP-treated plant leaves showed symptoms of hypersensitive response including electrolyte leakage, reactive oxygen species generation and overexpression of defense-related genes PR-1, PAD3, ERF1, HSR203J, and HIN1. All those results suggested that cmcp gene was required for the growth development of C. manginecans and functioned as a major pathogenicity factor in mango infection.

Anthropogenic input of reactive nitrogen (N) is an environmental problem that threatens the diversity and stability of belowground ecosystems. Soil nematodes are abundant in soil and are occurring at multiple trophic levels in the soil food web. However, how N deposition affects the composition and stability of soil nematode community is largely unknown. Here, we investigated the response of soil nematode community composition to N deposition at different sampling seasons and also estimated the stability of nematode community in a semiarid grassland in northern China. We found that the addition of N not only reduced the diversity of the nematode community, but also reduced the temporal stability of nematode community. The stability of different nematode trophic groups had different responses to N addition, and the community of plant parasites was more stable than the other trophic groups at a relatively higher N addition level. Moreover, soil pH was closely correlated with the stability of bacterivores, fungivores and predators-omnivores and the diversity of nematode community under different N additions. Our results highlight that N additions indirectly influence the synchrony and the stability of nematode community through change in soil nematode abundance and richness, and the variations of nematode community stability under different N additions are closely related to soil pH. These changes in nematode community composition and stability will eventually influence soil ecosystem function and nutrient cycling through biotic interactions in the soil food web.

Allyl isothiocyanate (AITC) is a natural product used as a food additive. Due to its strong volatility and broad biological activity, AITC is considered as a bio-fumigant to control soil-borne fungal diseases in agriculture, creating an urgent need for evaluation of the antifungal activity of AITC. Here we study the effect of AITC on Fusarium solani growth and explore the molecular mechanisms. The results indicated that AITC causes rapid inhibition of F. solani after 5 min, hyphal deformity, and electrolyte leakage. A yeast-like vacuolar transient receptor potential channel regulator (FsYvc1, a STRPC family member) was identified in F. solani that seems to play a role in this fungi AITC sensitivity. Genetic evidence suggests the gene FsYvc1 is involved in F. solani growth, development, and pathogenicity. Loss of FsYvc1 resulted in hypersensitivity of F. solani to AITC and induced reactive oxygen species (ROS) accumulation ∼ 1.3 to 1.45- folds that of the wild type (WT), and no difference responses to CaCl2, NaCl, KCl, SDS, and Congo red when compared with WT. In addition, ΔFsYvc1-17 showed significantly reduced (∼ 1-fold) glutathione-S-transferase (GST) expression compared with the WT without AITC induction. Upon exposure to 4.8 μg/mL AITC for 3 h, the relative expression levels were ∼ 12–30 fold higher in both the WT and ΔFsYvc1-17. Nevertheless, no difference in GST expression level was observed between the WT and ΔFsYvc1-17. The current study provides novel insights into the toxicity mechanisms of AITC. Considering our results that show the key role of FsYvc1, we propose that it could act as a new molecular target for future fungicide development.

Background/Aims: Assistance with tumor-associated vascularization is needed for the growth and invasion of non-small cell lung cancer (NSCLC). Recently, it was shown that placental growth factor (PLGF) expressed by NSCLC cells had a critical role in promoting the metastasis of NSCLC cells. However, the underlying molecular mechanisms remain elusive. Methods: Here, we first established a NSCLC model in mice that allows us not only to isolate tumor cells from non-tumor cells in the tumor, but also to trace tumor cells in living animals. Levels of PLGF, its unique receptor Flt-1, as well as transforming growth factor β1 (TGFβ1) was examined in tumor cells and tumor-associated macrophages (TAM) by RT-qPCR. A transwell well co-culture system and HUVEC assay were applied to study the crosstalk between NSCLC cells and TAM. Results: NSCLC cells produced and secreted PLGF to signal to tumor-associated macrophages (TAM) through surface expression of Flt-1 on macrophages. In a transwell co-culture system, PLGF secreted by NSCLC cells triggered macrophage polarization to a TAM subtype that promote growth of NSCLC cells. Moreover, polarized TAM seemed to secrete TGFβ1 to enhance the growth of endothelial cells in a HUVEC assay. Conclusion: The cross-talk between TAM and NSCLC cells via PLGF/Flt-1 and TGFβ receptor signaling may promote the growth and vascularization of NSCLC.

Abstract. Background:. Biliary tract carcinoma (BTC) is relatively rare and comprises a spectrum of invasive tumors arising from the biliary tree. The prognosis is extremely poor. The incidence of BTC is relatively high in Asian countries, and a high number of cases are diagnosed annually in China owing to the large population. Therefore, it is necessary to clarify the epidemiology and high-risk factors for BTC in China. The signs associated with BTC are complex, often require collaborative treatment from surgeons, endoscopists, oncologists, and radiation therapists. Thus, it is necessary to develop a comprehensive Chinese guideline for BTC. Methods:. This clinical practice guideline (CPG) was developed following the process recommended by the World Health Organization. The Grading of Recommendations Assessment, Development, and Evaluation approach was used to assess the certainty of evidence and make recommendations. The full CPG report was reviewed by external guideline methodologists and clinicians with no direct involvement in the development of this CPG. Two guideline reporting checklists have been adhered to: Appraisal of Guidelines for Research and Evaluation (AGREE) and Reporting Items for practice Guidelines in Healthcare (RIGHT). Results:. The guideline development group, which comprised 85 multidisciplinary clinical experts across China. After a controversies conference, 17 clinical questions concerning the prevention, diagnosis, and treatment of BTC were proposed. Additionally, detailed descriptions of the surgical principles, perioperative management, chemotherapy, immunotherapy, targeted therapy, radiotherapy, and endoscopic management were proposed. Conclusions:. The guideline development group created a comprehensive Chinese guideline for the diagnosis and treatment of BTC, covering various aspects of epidemiology, diagnosis, and treatment. The 17 clinical questions have important reference value for the management of BTC.
