Abstract Background Non-coding RNA is a key epigenetic regulation factor during skeletal muscle development and postnatal growth, and miR-542-3p was reported to be conserved and highly expressed in the skeletal muscle among different species. However, its exact functions in the proliferation of muscle stem cells and myogenesis remain to be determined. Methods Transfection of proliferative and differentiated C2C12 cells used miR-542-3p mimic and inhibitor. RT-qPCR, EdU staining, immunofluorescence staining, cell counting kit 8 (CCK-8), and Western blot were used to evaluate the proliferation and myogenic differentiation caused by miR-542-3p. The dual luciferase reporter analysis and rescued experiment of the target gene were used to reveal the molecular mechanism. Results The data shows overexpression of miR-542-3p downregulation of mRNA and protein levels of proliferation marker genes, reduction of EdU+ cells, and cellular vitality. Additionally, knocking it down promoted the aforementioned phenotypes. For differentiation, the miR-542-3p gain-of-function reduced both mRNA and protein levels of myogenic genes, including MYOG, MYOD1, et al. Furthermore, immunofluorescence staining immunized by MYHC antibody showed that the myotube number, fluorescence intensity, differentiation index, and myotube fusion index all decreased in the miR-542-3p mimic group, compared with the control group. Conversely, these phenotypes exhibited an increased trend in the miR-542-3p inhibitor group. Mechanistically, phosphatase and tensin homolog (Pten) was identified as the bona fide target gene of miR-542-3p by dual luciferase reporter gene assay, si-Pten combined with miR-542-3p inhibitor treatments totally rescued the promotion of proliferation by loss-function of miR-542-3p. Conclusions This study indicates that miR-542-3p inhibits the proliferation and differentiation of myoblast and Pten is a dependent target gene of miR-542-3p in myoblast proliferation, but not in differentiation.

Abstract Lipid synthesis increases during the cell cycle to ensure sufficient membrane mass, but how insufficient synthesis restricts cell-cycle entry is not understood. Here, we identify a lipid checkpoint in G1 phase of the mammalian cell cycle by using live single-cell imaging, lipidome, and transcriptome analysis of a non-transformed cell. We show that synthesis of fatty acids in G1 not only increases lipid mass but extensively shifts the lipid composition to unsaturated phospholipids and neutral lipids. Strikingly, acute lowering of lipid synthesis rapidly activates the PERK/ATF4 endoplasmic reticulum (ER) stress pathway that blocks cell-cycle entry by increasing p21 levels, decreasing Cyclin D levels, and suppressing Retinoblastoma protein phosphorylation. Together, our study identifies a rapid anticipatory ER lipid checkpoint in G1 that prevents cells from starting the cell cycle as long as lipid synthesis is low, thereby preventing mitotic defects, which are triggered by low lipid synthesis much later in mitosis.

The occurrence of benign prostate hyperplasia (BPH) was related to disrupted sex steroid hormones, and metformin (Met) had a clinical response to sex steroid hormone-related gynaecological disease. However, whether Met exerts an antiproliferative effect on BPH via sex steroid hormones remains unclear. Here, our clinical study showed that along with prostatic epithelial cell (PEC) proliferation, sex steroid hormones were dysregulated in the serum and prostate of BPH patients. As the major contributor to dysregulated sex steroid hormones, elevated dihydrotestosterone (DHT) had a significant positive relationship with the clinical characteristics of BPH patients. Activation of adenosine 5'-monophosphate (AMP)-activated protein kinase (AMPK) by Met restored dysregulated sex steroid hormone homeostasis and exerted antiproliferative effects against DHT-induced proliferation by inhibiting the formation of androgen receptor (AR)-mediated Yes-associated protein (YAP1)-TEA domain transcription factor (TEAD4) heterodimers. Met’s anti-proliferative effects were blocked by AMPK inhibitor or YAP1 overexpression in DHT-cultured BPH-1 cells. Our findings indicated that Met would be a promising clinical therapeutic approach for BPH by inhibiting dysregulated steroid hormone-induced PEC proliferation.

This study aimed to analyse the influence of adding Lycium ruthenicum Murr. fruit (LRF) on physicochemical, sensory characteristics and antioxidant capacity in goat milk yoghurt (GMY) during 21 days of refrigerated storage. Three different yoghurt formulations were processed using varying amounts of added LRF (1.0, 1.5, and 2.0%). LRF supplementation improved some of the physicochemical characteristics and sensory characteristics of GMY. With the increase of LRF addition, the pH of GMY showed a downward trend, and the titrated acidity showed an upward trend. The corresponding number of colonies in the storage period was higher than that of yoghurt without LRF. The addition of a concentration of 1.0-2.0% LRF played a role in the proliferation of lactic acid bacteria, and viscosity and hardness increased with increasing LRF addition. Although the susceptibility to syneresis of GMY showed a downward trend with increasing LRF addition, the scavenging rate of DPPH (2,2-diphenyl-1-picrylhydrazyl), *OH (hydroxyl free radicals), and the reducing power of Fe3+ of the corresponding concentration of GMY increased. In general, GMY with LRF addition can combine the two nutrients and have higher nutritional value, which is a good supplement to the same types of products in the market.

BackgroundPolygonatum sibiricum (PS) is a traditional Chinese medicine (TCM) first recorded in Mingyi Bielu. The book documents that PS can nourish five internal organs, be taken for a long time, relax the body and prolong lifespan. Presently, PS is widely used in TCM to prevent premature graying of hair. Based on TCM theory and clinical trials, the wine steaming processed product from PS provides a better effect. However, no published study has elucidated the anti-aging mechanism.PurposeThe study aim was to investigate the anti-aging mechanism of PS and its wine steaming processed product in mice, specifically focusing on the effect of D-galactose (D-gal) surrounding the intestinal flora and the Kelch-like ECH-associated protein 1-nuclear factor erythroid 2-related factor 2-antioxidant response elements (Keap1/Nrf2/ARE) pathway.MethodsThe chemical components in Raw PS (RPS) and Wine-steamed PS (WPS) were identified by ultra-performance liquid chromatography–hybrid quadrupole-Orbitrap high-resolution mass spectrometry (UPLC-Q-Orbitrap HRMS). An aging model using Kunming mice was established through intraperitoneally injected D-gal. Concentrations of RPS and WPS at 5, 10, or 15 g/kg/day levels were administered intragastrically, respectively. The body weight, liver and spleen indexes, superoxide dismutase (SOD), glutathione peroxidase (GSH-PX), and malondialdehyde (MDA) activities in serum and brain tissue were recorded. Hematoxylin and eosin (HE) stained brain tissue was histopathologically examined. The expressions of Keap1, Nrf2 and heme oxygenase 1 (HO-1) in the brain tissue at the mRNA and protein levels were respectively detected by reverse transcription-polymerase chain reaction (RT-PCR) and western blot (WB). Moreover, an Illumina Hiseq platform was used for 16S ribosomal RNA (16S rRNA) high-throughput sequencing to evaluate the proportions of intestinal flora in aging mice.ResultsThe proportions of saccharides, flavonoids, and triterpene acids were different between RPS and WPS. In the aging model mice, WPS outperformed RPS in improving body weight and mental state by increasing the spleen index, SOD and GSH-PX activities, decreasing the liver index and MDA activities, and restoring the histopathological morphology in D-gal-induced aging mice. At the mRNA levels, RPS and WPS significantly reduced the expression of Keap1 and increased the expressions of Nrf2 and HO-1. The trend in protein expressions was similar to that of the mRNA results, and WPS had a stronger effect than RPS. Fecal microbiota analysis showed that RPS and WPS restored intestinal microbiota proportions to normal levels.ConclusionThe results demonstrated that PS and its WPS had a positive effect in relieving oxidative stress in aging mice. WPS outperformed RPS, which might be related to the activation of the Keap1/Nrf2/ARE pathway and regulation of intestinal flora.

ABSTRACTForest Stock Volume (FSV) is one of the key indicators in forestry resource investigation and management on local, regional, and national scales. Limited by the saturation problems of optical satellite remote-sensing imagery in the retrieving of stock volume, and the high cost of Light Detection And Ranging (LiDAR) data, it is still challenging to estimate FSV in a large area using single-sensor remote-sensing data. In this paper, a method integrated multispectral satellite imagery and LiDAR data was developed to map stock volume in a large area. A random forest model was adopted to estimate the stock volume of larch forest in China based on the training samples from the Airborne Laser Scanning (ALS)-derived stock volume and corresponding Sentinel-2 imagery. Validation using National Forest Inventory (NFI) data, ALS-derived stock volume and ground investigation data demonstrated that the estimated stock volume had a high accuracy (R2 = 0.59, RMSE = 59.69 m3/ha, MD = 39.96 m3/ha when validated with NFI data; R2 ranged from 0.77 to 0.85, RMSE ranged from 38.68 m3/ha to 67.38 m3/ha, MD ranged from 24.90 m3/ha to 37.27 m3/ha when validated with ALS stock volume; R2 = 0.42, RMSE = 79.10 m3/ha, MD = 62.06 m3/ha when validated with field investigation data). Results of this paper indicated the applicability of estimating stock volume of larch forest in a large area by combining Sentinel-2 data and airborne LiDAR data.

Gas–oil–water three-phase slug flows in pipes commonly exist in the oil and gas industry as oil fields are becoming mature and water production is becoming inevitable. Although studies on multiphase flows in pipes have been ongoing for decades, most previous research has focused on gas–liquid or oil–water two-phase flows, with limited studies on gas–liquid–liquid flows. This leads to limited modeling studies on gas–liquid–liquid flows. One factor contributing to the complexity of the gas–liquid–liquid flow is the mixing between the oil and water phases, which have closer fluid properties and low interfacial tension. Restrictions or piping components play a crucial role in altering phase mixing. Unfortunately, modeling studies that consider the effects of these restrictions are limited due to the scarcity of experimental research. To address this gap, we conducted experimental studies on a gas–liquid–liquid flow downstream of a restriction and developed a new mechanistic modeling approach to predict the pressure gradient. Our model focuses on the flow pattern where the oil and water phases are partially mixed. This work emphasizes the modeling approach. The model evaluation results show that the model outperforms other existing models, with an average absolute relative error of 6.71%. Additionally, the parametric study shows that the new modeling approach effectively captures the effects of restriction size, water cut, and gas and liquid flow rates on the three-phase slug flow pressure gradient in horizontal pipes. Most previous slug flow modeling work assumes either a stratified flow or fully dispersed flow between the oil and water phases. This work provides a novel perspective in modeling a three-phase slug flow in which the oil and water phases are partially mixed. In addition, this novel approach to modeling the restriction effects on the pressure gradient paves the way for future modeling for different types of piping components or restrictions.

This work presents an experimental study on oil–water flow downstream of a restriction. The flow pattern, volumetric phase distribution, and their impacts on pressure drop are discussed. We employed two techniques to visualize the oil–water flow patterns, a high-speed camera and an Electrical Capacitance Volume Tomography (ECVT) system. The ECVT system is a non-intrusive device that measures the volumetric phase distribution at the pipe cross-section with time, which plays a critical role in determining the continuous phase in the oil–water flow, and therefore the oil–water flow pattern. In this study, we delved into the oil–water flow pattern and volumetric phase distribution for different valve openings, flow rates, and water cuts, and how they impact the pressure drop. The experimental results have demonstrated a strong relationship between the oil–water flow pattern and the pressure gradient, while the oil–water flow pattern is significantly influenced by the flowing conditions and the valve openings. The impacts of water cuts on the oil–water flow pattern are more obvious for smaller valve openings. For large valve openings, the oil and water phases tend to be more separated. This results in a moderate variation in the pressure gradient as a function of water cuts. However, it becomes more complex as the valve opening decreases. The pressure gradient generally increases with decreasing valve openings until the flow pattern becomes an oil-in-water dispersed flow. The impact of the valve on the pressure gradient is more pronounced in water-dominated flow when the water cut is above the inversion point, while it seems to be most obvious for medium water cut conditions.

Understanding droplet sizes is crucial for modeling gas–liquid annular flow and has many applications across various industries. This paper presents a comprehensive review and analysis of experimental data on droplet-size characteristics in gas–liquid annular flow from the open-source literature. The existing models for droplet-size predictions are evaluated against experimental data. Based on the data analysis, we propose two new correlations to predict the maximum droplet diameter and Sauter mean droplet diameter. Model evaluation and parametric study show that the new correlations capture the effects of fluid properties and flowing conditions on the droplet diameters well, outperforming other existing models. The average absolute relative errors are reduced to 14.8% and 29% for the maximum droplet diameter and the Sauter mean droplet diameter, respectively.

This article discusses the use of distributed acoustic sensing (DAS) for monitoring gas–liquid two-phase slug flow in horizontal pipes, using standard telecommunication fiber optics connected to a DAS integrator for data acquisition. The experiments were performed in a 14 m long, 5 cm diameter transparent PVC pipe with a fiber cable helically wrapped around the pipe. Using mineral oil and compressed air, the system captured various flow rates and gas–oil ratios. New algorithms were developed to characterize slug flow using DAS data, including slug frequency, translational velocity, and the lengths of slug body, slug unit, and the liquid film region that had never been discussed previously. This study employed a high-speed camera next to the fiber cable sensing section for validation purposes and achieved a good correlation among the measurements under all conditions tested. Compared to traditional multiphase flow sensors, this technology is non-intrusive and offers continuous, real-time measurement across long distances and in harsh environments, such as subsurface or downhole conditions. It is cost-effective, particularly where multiple measurement points are required. Characterizing slug flow in real time is crucial to many industries that suffer slug-flow-related issues. This research demonstrated the DAS’s potential to characterize slug flow quantitively. It will offer the industry a more optimal solution for facility design and operation and ensure safer operational practices.

This work presents an integrated multiphase flow model for downhole pressure predictions with the purpose of producing relatively more accurate predictions under wide-flowing conditions while maintaining a simple form. As a component of the integrated model, an improved two-fluid model for segregated flow is proposed. The new two-fluid model was developed by improving the modeling of the wetted perimeters and the liquid wall shear stress. It outperformed five other existing state-of-the-art models in predicting the liquid holdup and pressure gradient of 11 experimental datasets from the literature, with an average relative error of 37.6% for liquid holdup predictions and 24.0% for pressure gradient predictions. The model succeeds in capturing the effects of inclination angle, pressure or gas density, and liquid and gas superficial velocities on liquid holdup and pressure gradient. The integrated model incorporates the state-of-the-art onset of liquid loading predictive model and classifies the flow into two major categories based on the onset of liquid loading that are modeled by two different approaches, respectively. It outperformed the other multiphase flow models in predicting the downhole pressure of 313 field data points, especially the wells that have both segregated and intermittent (mixed) flows. The errors were reduced to 4.4% for the field cases with mixed flow regions and 5.1% for all the field data points.

This paper presents a detailed literature review on the thermal management issue faced by electronic devices, particularly concerning uneven heating and overheating problems. Special focus is given to the design and structural optimization of heat sinks for efficient single-phase liquid cooling. Firstly, the paper highlights the common presence and detrimental consequences of electronics overheating resulting from multiple heat sources, supported by various illustrative examples. Subsequently, the emphasis is placed on single-phase liquid cooling as one of the effective thermal management technologies for power electronics, as well as on the enhancement of heat transfer in micro/mini channel heat sinks. Various studies on the design and structural optimization of heat sinks are then analyzed and categorized into five main areas: (1) optimization of channel cross-section shape, (2) optimization of channel flow passage, (3) flow distribution optimization for parallel straight channel heat sinks, (4) optimization of pin-fin shape and arrangement, and (5) topology optimization of global flow configuration. After presenting a broad and complete overview of the state of the art, the paper concludes with a critical analysis of the methods and results from the literature and highlights the research perspectives and challenges in the field. It is shown that the issue of uneven and overheating caused by multiple heat sources, which is commonly observed in modern electronics, has received less attention in the literature compared to uniform or single-peak heating. While several design and structural optimization techniques have been implemented to enhance the cooling performance of heat sinks, topology optimization has experienced significant advancements in recent years and appears to be the most promising technology due to its highest degree of freedom to treat the uneven heating problem. This paper can serve as an essential reference contributing to the development of liquid-cooling heat sinks for efficient thermal management of electronics.

Text classification is an important application of machine learning. This paper proposes a communication-efficient federated text classification method based on parameter pruning. In the federated learning architecture, the data distribution of different participants is not independent and identically distributed; a federated word embedding model FedW2V is proposed. Then the TextCNN model is extended to the federated architecture. To reduce the communication cost of the federated TextCNN model, a parameter pruning algorithm called FedInitPrune is proposed, which reduces the amount of communication data both in the uplink and downlink during the parameter transmission phase. The algorithms are tested on real-world datasets. The experimental results show that when the text classification model accuracy reduces by less than 2%, the amount of federated learning communication parameters can be reduced by 74.26%.

How normal cells proliferate without CDK4 and CDK6, two cancer-driving kinases, remains unclear. Here, the authors show that without CDK4/6 activity, cells start the cell cycle with a different signaling order and commitment point, revealing unexpected flexibility in cell-cycle entry mechanisms.

The emergence of e-commerce has rejuvenated backward and poor remote villages. This paper presents a revelatory case study of three remote villages in China to depict how information and communication technology (ICT, specifically e-commerce) can empower remote areas to thrive. With the concept of affordance as basis, this work identifies critical actors in the development of rural e-commerce (i.e., Taobao Village) and illustrates how they use ICT. Eight deterministic affordances of the same ICT used in the evolution of rural e-commerce are specified, and three evolution paths about how ICT empowers the formation and development of rural e-commerce are depicted. This paper also describes the challenges of the current development of rural e-commerce. The findings of the study are discussed, and suggestions on how to capitalize on ICT for rural e-commerce development are offered.

Summary: Contact inhibition of cell proliferation regulates tissue size and prevents uncontrolled cell expansion. When cell density increases, contact inhibition can force proliferating cells into quiescence. Here we show that the variable memory of local cell density experienced by a mother cell controls the levels of the cyclin-dependent kinase (CDK) activator cyclin D1 and inhibitor p27 in newborn daughters, which direct cells to proliferation or quiescence. Much of this regulation can be explained by rapid suppression of ERK activity by high cell density in mothers, which leads to lower cyclin D1 and higher p27 levels in daughters. Strikingly, cell density and mitogen signals compete by shifting the ratio of cyclin D1/p27 levels below or above a single sharp threshold that controls the proliferation decision. Thus, the history of competing cell density and mitogen signals experienced by mothers is funneled into a precise activator-inhibitor balance that decides the fate of daughter cells.

Mammalian cells typically start the cell-cycle entry program by activating cyclin-dependent protein kinase 4/6 (CDK4/6). CDK4/6 activity is clinically relevant as mutations, deletions, and amplifications that increase CDK4/6 activity contribute to the progression of many cancers. However, when CDK4/6 is activated relative to CDK2 remained incompletely understood. Here, we developed a reporter system to simultaneously monitor CDK4/6 and CDK2 activities in single cells and found that CDK4/6 activity increases rapidly before CDK2 activity gradually increases, and that CDK4/6 activity can be active after mitosis or inactive for variable time periods. Markedly, stress signals in G1 can rapidly inactivate CDK4/6 to return cells to quiescence but with reduced probability as cells approach S phase. Together, our study reveals a regulation of G1 length by temporary inactivation of CDK4/6 activity after mitosis, and a progressively increasing persistence in CDK4/6 activity that restricts cells from returning to quiescence as cells approach S phase.

Slug flow is one of the most common flow types encountered in surface facilities, pipelines, and wellbores. The intermittent gas phase, in the form of a Taylor bubble, followed by the liquid phase can be destructive to equipment. However, commonly used point flow sensors have significant limitations for flow analysis. Distributed acoustic sensing (DAS) can turn optical fibers into an array of distributed strain rate sensors and provide substantial insights into flow characterization. We built a 10 m vertical laboratory flow loop equipped with wrapped fiber optic cables to study the DAS response of rising Taylor bubbles. Low-passed DAS data allow for velocity tracking of Taylor bubbles of different sizes and water velocities. Moreover, we measured the velocity of the wake region following the Taylor bubble and explored the process of Taylor bubbles merging. The amplitude analysis of DAS data allows for the estimation of Taylor bubble size. We conclude that DAS is a promising tool for understanding Taylor bubble properties in a laboratory environment and monitoring destructive flow in facilities across different industries to ensure operations are safe and cost-effective.

Thermal energy storage based on phase change materials (PCMs) can improve the efficiency of energy utilization by eliminating the mismatch between energy supply and demand. It has become a hot research topic in recent years, especially for cold thermal energy storage (CTES), such as free cooling of buildings, food transportation, electronic cooling, telecommunications cooling, etc. This paper summarizes the latest research progress of the PCMs-based CTES. Firstly, the classification of PCMs for low temperature storage is introduced; the thermal physical properties (e.g., phase change temperature (PCT) and latent heat) of suitable PCM candidates (−97 to 30 °C) for CTES are summarized as well. Secondly, the techniques proposed to enhance the thermal properties of PCMs are presented, including the addition of nanomaterials, the microencapsulation and the shape stabilization. Finally, several representative applications (−97 to 65 °C) of PCMs in different CTES systems are discussed. The present review provides a comprehensive summary, systematical analysis, and comparison for the PCMs-based CTES systems, which can be helpful for the future development in this field.

Thermal energy storage (TES) system plays an essential role in the utilization and exploitation of renewable energy sources. Over the last two decades, single-tank thermocline technology has received much attention due to its high cost-effectiveness compared to the conventional two-tank storage systems. The present paper focuses on clarifying the performance indicators and the effects of different influencing factors for the thermocline TES systems. We collect the various performance indicators used in the existing literature, and classify them into three categories: (1) ones directly reflecting the quantity or quality of the stored thermal energy; (2) ones describing the thermal stratification level of the hot and cold regions; (3) ones characterizing the thermo-hydrodynamic features within the thermocline tanks. The detailed analyses on these three categories of indicators are conducted. Moreover, the relevant influencing factors, including injecting flow rate of heat transfer fluid, working temperature, flow distributor, and inlet/outlet location, are discussed systematically. The comprehensive summary, detailed analyses and comparison provided by this work will be an important reference for the future study of thermocline TES systems.

Segregated flow, including stratified and annular flows, is commonly encountered in several practical applications such as chemical, nuclear, refrigeration, and oil and gas industries. Accurate prediction of liquid holdup and the pressure gradient is of great importance in terms of system design and optimization. The current most widely accepted model for segregated flow is a physics-based two-fluid model that treats gas and liquid phases separately by incorporating mass and momentum conservation equations. It requires empirically derived closure relationships that have the limitation of being applicable only under a narrow range of input parameters under which they were developed. In this paper, we proposed a more generalized machine learning augmented two-fluid model, using a database that spans the range of various flowing conditions and fluid properties. Machine learning algorithms such as random forest, neural networks, and gradient boosting were tested for the best performing data-driven predictive model. The new model proposed in this work successfully captures the complex, dynamic, and non-linear relationships between the friction factor and flowing conditions. A comprehensive model evaluation against nineteen existing correlations shows the best results from the proposed model.

The columnar jointed rock mass (CJR), composed of polygonal cross-sectional columns cut by several groups of joints in various directions, was exposed during the excavations of the Baihetan hydropower station, China. In order to investigate the unloading performances and the stability conditions during excavation of the columns, an experimental field study was performed. Firstly, on-site investigations indicated that the geotechnical problems, including rock relaxation, cracking and collapse, were the most prominent for the CJR Class I that contains intensive joint network and the smallest column sizes. Comprehensive field tests, including deformation measurement by multi-point extensometers, ultrasonic wave testing, borehole television observation and stress monitoring of rock anchors, revealed that the time-dependent relaxation of the CJRs was marked. The practical excavation experiences for the Baihetan columnar jointed rock masses, such as blasting scheme, supporting time of shotcrete and rock bolts, were presented in the excavations of the diversion tunnels. These detailed investigations and practical construction experiences can provide helpful information for similar geotechnical works in jointed rock mass.

Chalcidoidea (Hymenoptera) are minute wasps which can attack immature and adult stages of virtually all insect orders. Here, we sequenced and annotated the mitochondrial genome (mitogenome) of Chalcidoidea sp. This mitogenome was 15,152 bp long and encoded 13 protein-coding genes (PCGs), 20 transfer RNA genes (tRNAs), and 2 ribosomal RNA unit genes (rRNAs). All 13 PCGs were initiated by the ATN (ATG, ATT, ATA, and ATC) codon. All PCGs terminate with the stop codons TAA or TAG except for nad4 which ended with the incomplete codon T−. Phylogenetic analysis showed that Chalcidoidea sp. got together with the species Encyrtus infelix and Eurytoma sp., and species in Chalcidoidea formed a sister group to other Cynipoidea and Proctotrupoidea species.

This article presents technical data for concentrated solar power (CSP) plants in operation, under construction and in project all over the world in the form of tables. Liver failure is often associated with hepatic encephalopathy, due to dyshomeostasis of the central nervous system (CNS). Under physiological conditions, the CNS homeostasis is precisely regulated by the blood-brain barrier (BBB). The BBB consists of brain microvessel endothelial cells connected with a junctional complex by the adherens junctions and tight junctions. Its main function is to maintain brain homoeostasis via limiting the entry of drugs/toxins to brain. The brain microvessel endothelial cells are characterized by minimal pinocytotic activity, absent fenestrations, and highly expressions of ATP-binding cassette (ABC) family transporters (such as P-glycoprotein, breast cancer resistance protein and multidrug resistance-associated proteins). These ABC transporters prevent brain from toxin accumulation by pumping toxins out of brain. Accumulating evidences demonstrates that liver failure diseases altered the expression and function of ABC transporters at The BBB, indicating that the alterations subsequently affect drugs’ brain distribution and CNS activity/neurotoxicity. ABC transporters also mediate the transport of endogenous substrates across the BBB, inferring that ABC transporters are also implicated in some physiological processes and the development of hepatic encephalopathy. This paper focuses on the alteration in the BBB permeability, the expression and function of ABC transporters at the BBB under liver failure status and their clinical significances.

BACKGROUND: The oriental fruit fly, Bactrocera dorsalis Hendel, causes serious losses to fruit production and is one of the most economically important pests in many countries, including China, Spalangia endius Walker is a pupal parasitoid of various dipteran hosts, and may be considered a potentially important ectoparasitic pupal parasitoid of B. dorsalis. However, lack of genetic information on this organism is an obstacle to understanding the mechanisms behind its interaction with this host. Analysis of the S. endius transcriptome is essential to extend the resources of genetic information on this species and, to support studies on S. endius on the host B. dorsalis. METHODOLOGY/PRINCIPAL FINDINGS: We performed de novo assembly RNA-seq of S. endius. We obtained nearly 10 Gbp of data using a HiSeq platform, and 36319 high-quality transcripts using Trinity software. A total of 22443 (61.79%) unigenes were aligned to homologous sequences in the jewel wasp and honeybee (Apis florae) protein set from public databases. A total of 10037 protein domains were identified in 7892 S. endius transcripts using HMMER3 software. We identified expression of six gustatory receptor and 21 odorant receptor genes in the sample, with only one gene having a high expression level in each family. The other genes had a low expression level, including two genes regulated by splicing. This result may be due to the wasps being kept under laboratory conditions. Additionally, a total of 3727 SSR markers were predicted, which could facilitate the identification of polymorphisms and functional genes within wasp populations. CONCLUSION/SIGNIFICANCE: This transcriptome greatly improves our genetic understanding of S. endius and provides a large number of gene sequences for further study.

Our understanding of cell-cell interactions has been significantly improved in the past years with the help of Total Internal Reflection Fluorescence Microscope (TIRFM) in combination with an antigen presenting system supported by planar lipid bilayer (PLB) membranes, which are used to mimic the extensive receptor and ligand interactions within cell-cell contact interface. In TIRFM experiments, it is a challenge to uniformly present ligand molecules in monomeric format on the surface of PLB membranes. Here, we introduce a new and robust method of tethering IgG surrogate antigen ligands on the surface of Ni(2+)-containing PLB membranes. In this method, we use a modified D domain from staphylococcal protein A molecule that is fused with an N-terminus polyhistidine tag (H12-D-domain) to tether IgG surrogate antigens on Ni(2+)-containing PLB membranes. We systematically assessed the specificity and capability of H12-D-domain construct to capture IgG molecules from different species through live cell and single molecule TIRFM imaging. We find that these IgG surrogate antigens tethered by H12-D-domain show better lateral mobility and are more uniformly distributed on PLB membranes than the ones tethered by streptavidin. Neither IgM molecules, nor Fab or F(ab')2 fragments of IgG molecules can be tethered on PLB membranes by H12-D-domain construct. These tethered IgG surrogate antigens strongly induce the formation and accumulation of signaling active antigen receptor microclusters within the immunological synapse in B or T lymphocyte cells. Thus our method provides a new and robust method to tether IgG surrogate antigens or other molecules fused with IgG Fc portion on PLB membranes for TIRFM based molecule imaging experiments.

Background Hydrogen sulphide (H2S) is considered as the third member of the gasotransmitter family, along with nitric oxide (NO) and carbon monoxide. H2S has been reported to induce angiogenesis by promoting the growth, migration and tube-like structure formation of endothelial cells. Those studies were conducted in conditions of cell culture, mouse Matrigel plug assay model, rat wound healing model or rat hindlimb ischaemia model. Recent in vivo studies showed the physiological importance of H2S in muscle angiogenesis. However, the importance of endogenous H2S for brain angiogenesis during development remains unknown. We therefore aimed at determining the role of H2S in brain vascular development.Methods and results Both knockdown and knockout of H2S-producing enzymes, cystathionine β-synthase (cbs) and cystathionine γ-lyase (cth), using morpholino oligonucleotides and clustered regularly interspaced short palindromic repeats/Cas9-mediated mutation, impaired brain vascular development of larval zebrafish. Incubation with the slow-releasing H2S donor GYY4137 alleviated the defects of brain vascular development in cbs and cth morphants. Quantitative analysis of the midbrain vascular network showed that H2S enhances angiogenesis without affecting the topological structure of the brain vasculature. Mechanically, nitric oxide synthase 2a (nos2a) expression and NO production were decreased in both cbs and cth morphants. Overexpression of nos2a by coinjection of cbs or cth MO with full-length zebrafish nos2a mRNA alleviated the brain vascular developmental defects in cbs and cth morphants.Conclusion We conclude that H2S promotes brain developmental angiogenesis via the NOS/NO pathway in zebrafish.

Abstract This literature review synthesizes the role of soil moisture in regulating carbon sequestration and greenhouse gas emissions (CS-GHG). Soil moisture directly affects photosynthesis, respiration, microbial activity, and soil organic matter dynamics, with optimal levels enhancing carbon storage while extremes, such as drought and flooding, disrupt these processes. A quantitative analysis is provided on the effects of soil moisture on CS-GHG across various ecosystems and climatic conditions, highlighting a “Peak and Decline” pattern for CO₂ emissions at 40% water-filled pore space (WFPS), while CH₄ and N₂O emissions peak at higher levels (60–80% and around 80% WFPS, respectively). The review also examines ecosystem models, discussing how soil moisture dynamics are incorporated to simulate photosynthesis, microbial activity, and nutrient cycling. Sustainable soil moisture management practices, including conservation agriculture, agroforestry, and optimized water management, prove effective in enhancing carbon sequestration and mitigating GHG emissions by maintaining ideal soil moisture levels. The review further emphasizes the importance of advancing multiscale observations and feedback modeling through high-resolution remote sensing and ground-based data integration, as well as hybrid modeling frameworks. The interactive model-experiment framework emerges as a promising approach for linking experimental data with model refinement, enabling continuous improvement of CS-GHG predictions. From a policy perspective, shifting focus from short-term agricultural productivity to long-term carbon sequestration is crucial. Achieving this shift will require financial incentives, robust monitoring systems, and collaboration among stakeholders to ensure sustainable practices effectively contribute to climate mitigation goals.

The ReaxFF can describe the properties of energetic materials (EMs) at equilibrium state, but does not work properly in simulating high-energy particle irradiation process because of its weak short-range interaction. In this paper, a modification was made for such a potential by connecting Ziegler-Biersack-Littmark (ZBL) potential to ReaxFF-lg through comparing to Density Functional Theory (DFT) results to accurately describe short-range interactions. After modification, the newly fitted ReaxFF-lg/ZBL potential predicts better the equation of state for EMs In displacement cascade simulations, comparing to results from ab initio molecular dynamics (AIMD), ReaxFF-lg/ZBL presented the similar transferred energy from a primary knock-on atom to surrounding atoms, better than the original ReaxFF-lg potential. Further large-scale displacement cascade simulations indicated ReaxFF-lg/ZBL could be applied for cascade simulations with PKA energy from less than 1 keV to high energy (e.g. 35 keV) cases, which is suitable for effectively simulating high-energy displacement cascades in EMs using molecular dynamics method.

Abstract Hepatocellular carcinoma (HCC) mainly depends on liver fibrosis/cirrhosis, which is regulated by tumor cells and the tumor microenvironment (TME), and is a crucial factor in tumor progression. This study aimed to identify abnormally expressed miR‐500a‐3p in the hepatitis‐cirrhosis‐HCC pathway and explored the roles of miR‐500a‐3p in HCC progression. A clinical cohort of patients with HCC is studied retrospectively. Subsequently, the role of miR‐500a‐3p transported by HCC exosomes in hepatic stellate cell (HSC) activation, hepatoma growth and invasion, and immune cell differentiation is determined by in vitro and in vivo experiments. In clinical tissues, miR‐500a‐3p is significantly enriched in HCC and cirrhosis tissues, and co‐expression of the immune marker CD4 or PD‐L1 significantly correlates with low survival rates in patients. Extracellular miR‐500a‐3p is taken up by HSC and PBMC, which promotes the secretion of the cytokines TGF‐β1 and IL‐10, increases PD‐L1 expression in HSC, and stabilizes PD‐1 expression in PBMC to affect the TME. Moreover, miR‐500a‐3p is associated with CD4+ T‐cell exhaustion and Treg differentiation and is significantly associated with increased tumorigenicity in in situ mouse HCC models. Mechanistically, HCC‐derived exosomal miR‐500a‐3p directly influences SOCS2 to regulate the JAK3/STAT5A/STAT5B signaling pathway. MiR‐500a‐3p promotes the growth and migration of HCC through the SOCS2/JAK3/STAT5A/STAT5B axis.
