This study is aimed to investigate the effects of Hypericum perforatum olive oil extract on the cytotoxic and metastatic properties of human colorectal cancer cells and human bone marrow-derived mesenchymal stem cells. In addition, ALDH3A1 and Vimentin expressions were evaluated by qRT-PCR and western blot analysis. Total phenolic and flavonoid contents and antioxidant activity of methanol extracts prepared with oil enrichment were measured using spectrophotometry-based methods. The cytotoxic effects of the extracts on SW-480 and bone marrow-derived mesenchymal stem cells were evaluated by MTT assay, resulting in IC50 values of 4.8 mg/ml and 4.9 mg/ml, respectively. It was determined that cell migration and colony formation were significantly reduced at the IC50 values determined for SW-480 and human bone marrow-derived mesenchymal stem cells. Background: There is limited information on the immediate effects of whole-body vibration (WBV) on the upper limb. This study aims to determine the immediate effects of WBV on reaction speed and proprioception in young adult students' upper extremities. Methods: In total, 62 students participated in the study. WBV was applied to the participants, and its immediate effects on proprioception and upper extremity reaction speed were examined. Participants' proprioception and perception of joint position at 30-60 degrees of shoulder flexion, shoulder abduction, and elbow flexion angles were measured with absolute error degrees. Reaction rates were evaluated with the Ruler-Drop Test and the mobile application SWAY. Results: A decrease was observed in the absolute error level of the participants' joint position perception at 30-60 elbow and shoulder position degrees, measured after immediate WBV application (p <0.05). After the RDT application, a decrease in the length of catching the target was observed (p <0.05). The SWAY test determined that they moved the smartphone in a shorter time (p <0.05). Right and left RDT scores showed that the distance to catch the ruler was significantly lower in male individuals before the application. In comparison, the distance to catch the ruler was lower after the application (right/left p<0.05). Conclusions: The study found that applying WBV improved upper extremity proprioception perception and reaction speed in young adults. This information can guide clinicians in applying WBV to healthy individuals and those with symptoms. [ABSTRACT FROM AUTHOR]

We investigated the perceptions of students with disabilities (SWD) and their parents about the education offered in private special education institutions in Turkey, called "Special Education and Rehabilitation Centers" (SERCs). Data were collected from a survey administered to 453 SWD and 479 parents and interviews that were conducted with 38 parents. Survey findings indicated both satisfaction and, largely, dissatisfaction with the quality of special education offered in SERCs. Findings indicated that (i) parents were satisfied with the opportunities provided for their children to receive free special education and (ii) there was discontent about the use of government-funded resources, the quality of teaching, environmental arrangements, attitudes and beliefs toward SWD, inspection and supervision, and the way policy was designed and implemented. Findings demonstrate the need to reconsider the quality of education offered in private special education centers and the ways to improve the quality level.

Aim: This study aims to compare the efficacy of ultrasonography-guided corticosteroid injection to the glenohumeral joint and oral steroid therapy on clinical response and inferior axillary pouch (AR) thickness measured by ultrasonography (USG) in patients with adhesive capsulitis (AC). Materials and Methods: Our prospective randomized study included 48 patients (18-65 years) with a diagnosis of AC. The first group (n=24) underwent USG-guided corticosteroid injection to glenohumeral joint. In the second group (n=24), oral prednisone was started at 0.5 mg/kg dose with the dosing schedule determined by decreasing and continued for 6 weeks. Both groups were also included in a standard physical therapy program (electrotherapy+exercises). The questionnaire, active and passive shoulder joint range of motion (ROM) measurements, Shoulder Pain and Disability Index (SPADI), evaluation of the joint with USG, and AR thickness measurements were performed. Results: There was a statistically significant increase in passive-active ROM values in all directions before and after the treatment (p<0.05) and decrease in AR values after the treatment (p<0.001) in both groups. However, there was no significant difference between the two groups. In addition, the decrease in inferior pouch thickness and the decrease in SPADI scores were positively correlated. Conclusion: Steroid injection into the glenohumeral joint and oral steroid use significantly reduced shoulder pain and disability and increased range of motion in AC patients. Moreover, both oral steroid and steroid injection treatments improved radiologic findings (by reducing AR), consistent with clinical response. Background: Chewing difficulty, poor oral health, inadequate and imbalanced nutrition are serious health problems in individuals with intellectual disabilities. The participants' chewing abilities, oral health and nutritional status were analysed in this study. Methods: Forty‐five adult participants with intellectual disabilities were included. Anthropometric measurements, oral health assessments, chewing ability evaluations and dietary intake analyses were conducted. Results: A 56.8% of the participants were classified as overweight or obese. Teeth grinding was reported in 33.3% of the participants, while 40.0% experienced drooling. All participants with Down syndrome and 58.6% of the participants with developmental delay had chewing difficulties. Inadequate nutrient intake was observed and the fibre, vitamins B1, B3, B9, sodium, phosphorus and iron intakes were significantly lower than reference values in those with chewing difficulty (p <.05). Conclusions: Chewing difficulties were associated with lower intake of certain nutrients, highlighting the importance of addressing oral health and dietary counselling in this population. Carbonic anhydrase IX (CAIX) is a hypoxia-regulated gene. It is over expressed in a variety of cancers, including hepatocellular cancer. Transforming growth factor β (TGFβ) is considered to have an impact on cancer biology due to its important roles in cell proliferation and differentiation. The effect of the TGFβ on CAIX expression under hypoxia and the mechanism underlying the role of the hypoxia response element (HRE) on this expression are unknown. In this study, we demonstrate that TGFβ upregulates CAIX expression under hypoxic conditions in the Hep3B hepatoma cell line, indicating that the mitogen-activated protein kinase (MAPK)- and phosphatidylinositol-4,5-bisphosphate 3-kinase (PI3K)-signaling pathways might be responsible for this response. Site-directed mutagenesis of the HRE region in CAIX promoter reduced the TGFβ-induced CAIX promoter activity, pointing to the significance of HRE for this response. Up regulation of TGFβ-stimulated CAIX expression was consistent with the up regulation of promoter activity of five different truncated constructs of the CAIX promoter under hypoxia. Our findings show that the HRE region is critical for TGFβ-induced CAIX expression, which is mainly controlled by MAPK and PI3K pathways.

Background: This study aims to examine the relationship between post-earthquake anxiety status with sleep problems, low back and neck pain in victims of the Kahramanmaraş-centered earthquakes. Materials and Methods: The present study included 291 victims of the Kahramanmaraş-centered earthquakes between the ages of 18-65 y ears. T he s tudy i ndividuals w ere evaluated w ith t he B eck Anxiety Inventory (BAI), the Oswestry Disability Index (ODI), the Neck Disability Index (NDI), and the Pittsburgh Sleep Quality Index (PSQI). Results: Of the individuals, 67% experienced mild, moderate, and severe anxiety, and all individuals had poor sleep quality (6.9±3.8). While 57% of the individuals sheltered in their own homes had the mean PSQI score of 5 and above, 75% of those sheltered outside their homes had such a score. The ODI score percentage was above 20% in 27% of the individuals sheltered in their own homes and in 37% of those sheltered outside their own homes. A statistically moderate correlation was determined between the BAI score and the ODI score, the NDI score, and the PSQI score (r=0.511, r=0.604, r=0.539, respectively). Conclusions: Post-earthquake anxiety was revealed to be associated with sleep problems and low back and neck pain. Considering the relationship between anxiety, sleep problems, and low back and neck pain in the support approaches to be adopted for earthquake victims, approaches involving these parameters should be planned. Objective: Propolis has been found to have various effects, including antioxidant and anti-inflammatory properties, according to studies. In this recent research, we discovered that reducing allergenic compounds in propolis through biotransformation using specific Lactobacillus plantarum strains enhanced its anti-inflammatory qualities. The study aimed to identify the extraction methods and solvents that had the most significant anti-inflammatory effects and assess how L. plantarum strains biotransformation of propolis affected these qualities in THP-1 cell line cultures. Materials and Methods: Propolis samples were biotransformed with different concentrations (1.5%, 2.5%, 3.5%) of several L. plantarum strains (ISLG-2, ATCC®8014, visbyvac) before extraction using various solvents (ethanol, polyethylene glycol-PEG, water) and ultrasound treatments (300 W/40 Hz for 5, 10, 15 min). Liquid chromatography-mass spectrometer/mass spectrometry was used for phenolic analysis of the samples. ELISA test kits were employed to assess NF-kβ, IL-1α, IL-1β, IL-6, IL-10, TNF-α, IFN-γ, COX-1 in the cell culture supernatant. Results: Results showed that, except for NF-kβ, all cytokine levels decreased in four separate propolis samples. Caffeic acid, kaempferol, ferulic acid, quercetin, pelargonin, and naringenin were the key physiologically active components associated with the anti-inflammatory activity of propolis. The biotransformation process to reduce allergen compounds did not alter propolis's anti-inflammatory properties. Conclusion: In samples that were dissolved in water, dissolved in ethanol+biotransformed with L. plantarum ATCC®8014, dissolved in water+biotransformed with L. plantarum ATCC®8014, and dissolved in water+sonicated for 15 min and biotransformed with L. plantarum ATCC®8014, the maximum anti-inflammatory effect of propolis was assessed.
