Soil acidification poses a significant environmental challenge in China’s southern red soil regions, impacting the abundance of soil microbes and their capacity for carbon fixation. The effect of vegetation types on soil’s biological and abiotic components under acidification, and their regulatory role on the CO2 fixation mechanisms of soil autotrophic microorganisms, is difficult to examine. This gap in understanding constrains the assessment of the carbon fixation potential of red soils. To address this, indoor cultivation coupled with 13C stable isotope labeling was employed to evaluate the disparate abilities of autotrophic microorganisms to assimilate and store CO2 across five vegetation soils from the Qianyanzhou acidic red soil experimental station in China. Findings indicate that carbon fixation rates in these soils spanned from 4.25 to 18.15 mg C kg−1 soil d−1, with paddy field soils demonstrating superior carbon fixation capabilities compared to orchard, coniferous forest, broad-leaved forest, and wasteland soils. The 13C fixation rate in the 0–10 cm soil stratum surpassed that of the 10–30 cm layer across all vegetation types. High-throughput sequencing of 16S rRNA, following cbbL gene purification and amplification, identified Bradyrhizobium, Azospirillum, Burkholderia, Paraburkholderia, and Thermomonospora as the predominant autotrophic carbon-fixing microbial genera in the soil. PERMANOVA analysis attributed 65.72% of the variance in microbial community composition to vegetation type, while soil depth accounted for a mere 8.58%. Network analysis of microbial co-occurrence suggested the soil microbial interactions and network complexity changed with the change of vegetation types. Additionally, multiple linear regression analysis pinpointed the Shannon index and soil organic carbon (SOC) content as primary influencers of carbon fixation rates. Structural equation modeling suggested that iron enrichment and acidification indirectly modulated carbon fixation rates by altering SOC and autotrophic bacterial diversity. This investigation shows the spatial dynamics and mechanisms underpinning microbial carbon fixation across varying vegetation types in southern China’s red soil regions.

Abstract The interaction between programmed cell death ligand 1 (PD-L1), which is expressed on the surface of tumor cells, and programmed cell death 1 (PD-1), which is expressed on T cells, impedes the effective activation of tumor antigen-specific T cells, resulting in the evasion of tumor cells from immune-mediated killing. Blocking the PD-1/PD-L1 signaling pathway has been shown to be effective in preventing tumor immune evasion. PD-1/PD-L1 blocking antibodies have garnered significant attention in recent years within the field of tumor treatments, given the aforementioned mechanism. Furthermore, clinical research has substantiated the efficacy and safety of this immunotherapy across various tumors, offering renewed optimism for patients. However, challenges persist in anti-PD-1/PD-L1 therapies, marked by limited indications and the emergence of drug resistance. Consequently, identifying additional regulatory pathways and molecules associated with PD-1/PD-L1 and implementing judicious combined treatments are imperative for addressing the intricacies of tumor immune mechanisms. This review briefly outlines the structure of the PD-1/PD-L1 molecule, emphasizing the posttranslational modification regulatory mechanisms and related targets. Additionally, a comprehensive overview on the clinical research landscape concerning PD-1/PD-L1 post-translational modifications combined with PD-1/PD-L1 blocking antibodies to enhance outcomes for a broader spectrum of patients is presented based on foundational research.

Abstract Osteosarcoma (OS) is a prevalent bone solid malignancy that primarily affects adolescents, particularly boys aged 14–19. This aggressive form of cancer often leads to deadly lung cancer due to its high migration ability. Experimental evidence suggests that programmed cell death (PCD) plays a crucial role in the development of osteosarcoma. Various forms of PCD, including apoptosis, ferroptosis, autophagy, necroptosis, and pyroptosis, contribute significantly to the progression of osteosarcoma. Additionally, different signaling pathways such as STAT3/c‐Myc signal pathway, JNK signl pathway, PI3k/AKT/mTOR signal pathway, WNT/β‐catenin signal pathway, and RhoA signal pathway can influence the development of osteosarcoma by regulating PCD in osteosarcoma cell. Therefore, targeting PCD and the associated signaling pathways could offer a promising therapeutic approach for treating osteosarcoma.

Abstract Triple-negative breast cancer (TNBC) is an aggressive subtype of breast cancer that has a poor prognosis. TOP2A is a key enzyme in DNA replication and is a therapeutic target for breast and other cancers. TOP2A-specific Th1-promoting epitopes with optimal binding affinity to MHC II were identified using a combined scoring system. The multi-peptide TOP2A vaccine elicited a robust immunologic response in immunized mice, as demonstrated by the significant production of Th1 cytokines from immunized animals’ splenocytes stimulated in vitro with TOP2A peptides. Anti-tumor efficacy of the TOP2A vaccine was demonstrated in a syngeneic TNBC mouse model, in which pre-graft preventive vaccination was associated with significantly decreased tumor growth as compared to adjuvant control. In a genetically engineered mouse (GEM) model of TNBC, vaccinated animals demonstrated a significant reduction in tumor incidence and average tumor volume compared to adjuvant control. Finally, we examined TCR sequences in CD4 tumor Infiltrating lymphocytes (TIL) from vaccinated mice and found that the TIL contained TCR sequences specific to the three vaccine peptides. These data indicate that our newly developed multi-peptide TOP2A vaccine is highly immunogenic, elicits TILs with vaccine specific TCRs, and is highly effective in preventing and intercepting TNBC development and progression in vivo.

Abstract The International Workshop on Seismic Design and Assessment for Resilience, Robustness and Sustainability of Slope Engineering was held on 13–15 January 2023 on line, focusing on the theme of “Seismic resilience of slope engineering and the concept of resilience-based seismic design (RBSD) for geological disaster prevention and control”. In this workshop, a number of keynote and invited lectures provided an international exchange platform for researchers, industrial engineers and students to share their research, engineering practice and exchange novel ideas on seismic resilience for slope engineering in a way of online. At the same time, during this workshop, technical committee of the ICGdR-TC1 also took this opportunity to hold a working meeting on cutting-edge and strategic issues, and released the Shanghai Declaration on slope engineering.

Highlights The intimate NG@SiC heterostructure has been constructed via a direct thermal decomposition method. The NG@SiC heterostructure anode delivers enhanced capacity and cycling stability both in the half-cell and in the full cell. DFT analysis reveals that this NG@SiC anode possesses lower lithium-ion adsorption energy and higher charge and discharge rates.

Abstract Triple-negative breast cancer (TNBC) is the most lethal subtype of breast cancer, with limited therapeutic options readily available. Immunotherapy such as immune checkpoint inhibition has been investigated in TNBC but still encounters low overall response. Neutrophils, the most abundant leukocytes in the body, are increasingly recognized as an active cancer-modulating entity. In the bloodstream, neutrophils escort circulating tumor cells to promote their survival and stimulate their proliferation and metastasis. In the tumor microenvironment, neutrophils modulate the immune milieu through polarization between the anti-tumor and the pro-tumor phenotypes. Through a comprehensive review of recently published literature, it is evident that neutrophils are an important player in TNBC immunobiology and can be used as an important prognostic marker of TNBC. Particularly, in their pro-tumor form, neutrophils facilitate TNBC metastasis through formation of neutrophil extracellular traps and the pre-metastatic niche. These findings will help advance the potential utilization of neutrophils as a therapeutic target in TNBC.

The herb Prunella vulgaris has shown significant immune-stimulatory and anti-inflammatory effects in mouse models. Here, the effects of a novel Prunella vulgaris-containing herbal mixture, PV-1, were examined in several mouse models for cancer, including chemically induced models of lung and oral cancers as well as syngraft models for lung cancer and melanoma. PV-1, consisting of extracts from Prunella vulgaris, Polygonum bistorta, Sonchus brachyotus and Dictamnus dasycarpus, exhibited no toxicity in a dose escalation study in A/J mice. PV-1 significantly inhibited mouse lung tumor development induced by the lung carcinogens vinyl carbamate and benzo[a]pyrene. PV-1 also hindered the induction of oral squamous cell carcinomas in C57BL/6 mice caused by 4-nitroquinoline-1-oxide. Flow cytometry analysis showed that PV-1 increased the numbers of CD8+ tumor-infiltrating lymphocytes (TILs) and increased the production of granzyme B, TNF-α, and IFN-γ by CD8+ TILs. PV-1 also suppressed granulocytic myeloid-derived suppressor cell numbers (g-MDSCs) and improved the anti‐cancer activity of anti-PD‐1 immunotherapy. These results indicate that PV-1 remodels the tumor immune microenvironment by selectively inhibiting g‐MDSCs and increasing CD8+ TILs within tumors, resulting in decreased immune suppression and enhanced cancer chemopreventive efficacy.

With the combined effects of climate change and anthropogenic disturbance, landslide hazards have progressively increased and emerged as one of the most significant natural threats to socio-economic safety and human life. Synthetic aperture radar interferometry (InSAR) can measure subtle ground displacement and thus has immense potential for detecting active landslides. However, the operational application of InSAR for landslide detection and inventory update in wide-area is still hindered by the high labor and time costs for visual interpretation and manual editing of InSAR results. Aiming at this problem, we developed a novel method using InSAR and convolutional neural network (CNN) for automated identification of active landslides over wide areas. It first performs InSAR analysis to produce a surface displacement velocity map of the target region and then employs an improved Faster RCNN based on attended ResNet-34 and Feature Pyramid Networks (FPN) to detect active landslides from the velocity map. Taking the Guizhou province in southwest China as a case study, we processed 1168 scenes of Sentinel-1 images and 473 scenes of PALSAR-2 images to derive the surface displacement and identified 1627 active landslides, including 326 manually labeled landslides and 1301 landslides automatically detected by Faster RCNN. The improved Faster RCNN achieved good recall, precision, F1 score, and average precision (AP) at 91.49%, 91.33%, 0.914, and 0.940, respectively. Further experiments indicated that the trained Faster RCNN showed satisfactory applicability and result accuracy for different test areas and various InSAR techniques. Therefore, the proposed approach has great potential applications for building inventories of active landslides over wide areas and regularly updating the records, which is crucial for preventing landslide disasters and mitigating losses.

In this study, we analyzed the relationship between the refrigerant distribution of a multiparallel evaporator and the structural design of a distributor to address uneven distribution of gas–liquid refrigerants in multiparallel evaporators under variable operating conditions. Herein, we proposed the shunting concept of “flow pattern setting and critical distribution” and designed a theoretical model for a rectifying nozzle-type critical distributor (RNCD). We built the experimental bench of a multiparallel evaporator using an RNCD and compared the cooling time of each cold storage, the superheat of each branch pipe, the cooling capacity of each cold storage, and the pressure drop of the distributor. Further, we investigated the shunting performance of the RNCD. The experimental results showed that the evaporation temperatures were −32 °C to −24 °C when cold storage temperatures of −22 °C to −14 °C, respectively. The unevenness of superheat at the outlet of each evaporator branch pipe of the RNCD was 0.0884–0.1275, while the unevenness associated with the refrigeration capacity of each cold storage was 0.1424–0.1714. This study provides basic research information to solve the problem of uneven refrigerant distribution when considering gas–liquid two-phase flow in multiparallel evaporators under variable operating conditions.

Despeckling is an essential task in polarimetric synthetic aperture radar (PolSAR) image processing. Most of the existing filters developed for multitemporal synthetic aperture radar (SAR) images make use of either real or complex information. Real information refers to amplitude or intensity values, whereas complex information refers to complex covariance matrix (CCM) derived from either interferometric SAR (InSAR) or PolSAR data. The InSAR CCM is formed using images of the same polarimetric channel but acquired at different dates, and the PolSAR CCM contains information acquired simultaneously in different polarimetric channels. Therefore, these despeckling methods may present good performance in some applications and scenes but fail in other cases, due to differences in input sources. In order to achieve a more robust result in all cases, we develop a method for multitemporal polarimetric SAR data filtering based on tensor decomposition, which aims at improving the identification of homogeneous pixels for spatially adaptive filtering. The key element of this approach consists of exploiting tensor theory to construct a new CCM that contains both polarimetric and interferometric information, as well as multitemporal information for each pixel. The effectiveness of the proposed method and its performance are evaluated with simulated and real SAR data in comparison with several established methods.

Oleic acid (OA) is a kind of monounsaturated omega-3 fatty acid that abounds in plants and animals which can induce apoptosis and has broad-spectrum inhibitory activity against a variety of tumor cell lines. However, OA is quite insoluble and thus inconvenient to be efficiently delivered in vivo. In this work, OA was fabricated into nanoparticles to generate OA elastic nanoparticles (OA-ENPs) with a particle size of 185.6 nm and good stability in various physiological media. OA-ENPs alone achieved a high tumor inhibition rate of 60.3% without significant side effect. More surprisingly, the resultant OA-ENPs displayed dose-dependent tumor targetability. Low dose of OA-ENPs (10 mg/kg) mainly distributed in the liver after intravenous injection, while high dose of OA-ENPs mainly distributed in tumor. At the high dose of 90 mg/kg, OA-ENPs accumulation in tumor reached nearly twice as that in the liver. Here we provide a simple but effective way to achieve excellent tumor targetability without the need of any surface modification of nanoparticles.

Abstract Background Circular RNAs (circRNAs) act as gene expression regulators and are involved in cancer progression. However, their functions have not been sufficiently investigated in nasopharyngeal carcinoma (NPC). Methods The expression profiles of circRNAs in NPC cells within different metastatic potential were reanalyzed. Quantitative reverse transcription PCR and in situ hybridization were used to detect the expression level of circPVT1 in NPC cells and tissue samples. The association of expression level of circPVT1 with clinical properties of NPC patients was evaluated. Then, the effects of circPVT1 expression on NPC metastasis were investigated by in vitro and in vivo functional experiments. RNA immunoprecipitation, pull-down assay and western blotting were performed to confirm the interaction between circPVT1 and β-TrCP in NPC cells. Co-immunoprecipitation and western blotting were performed to confirm the interaction between β-TrCP and c-Myc in NPC cells. Results We find that circPVT1, a circular RNA, is significantly upregulated in NPC cells and tissue specimens. In vitro and in vivo experiments showed that circPVT1 promotes the invasion and metastasis of NPC cells. Mechanistically, circPVT1 inhibits proteasomal degradation of c-Myc by binding to β-TrCP, an E3 ubiquiting ligase. Stablization of c-Myc by circPVT1 alters the cytoskeleton remodeling and cell adhesion in NPC, which ultimately promotes the invasion and metastasis of NPC cells. Furthermore, c-Myc transcriptionally upregulates the expression of SRSF1, an RNA splicing factor, and recruits SRSF1 to enhance the biosynthesis of circPVT1 through coupling transcription with splicing, which forms a positive feedback for circPVT1 production. Conclusions Our results revealed the important role of circPVT1 in the progression of NPC through the β-TrCP/c-Myc/SRSF1 positive feedback loop, and circPVT1 may serve as a prognostic biomarker or therapeutic target in patients with NPC.

The development of chemopreventive strategies with the ability to prevent the progression of lung lesions to malignant cancers would reduce the mortality and morbidity resulting from this deadly disease. Delivery of microRNA (miRNA) by inhalation is a novel method for lung cancer prevention. In this study, we investigated the combined efficacy of aerosolized miR-138-5p and miR-200c miRNA mimics in lung cancer prevention. Combination of the two miRNAs inhibited Benzo(a)pyrene (B((a))P)-induced lung adenomas and N-nitroso-tris-chloroethylurea (NTCU)-induced lung squamous cell carcinomas with no detectable side effects. Using single-cell RNA sequencing (scRNA-seq) and imaging mass cytometry (IMC), we found that both miRNAs inhibited programmed cell death ligand 1 (PD-L1) expression. Our flow cytometry results showed that aerosolized delivery of combined miRNAs increased CD4+ and CD8+ T cells and reduced the expression of programmed cell death protein 1 (PD-1) and T-regulatory cells. Our results demonstrated that the delivery of aerosolized microRNAs targeting PD-L1 can be highly effective in preventing lung cancer development and progression in mice.

There is an urgent demand for continuous detection and monitoring of active slopes in wide reservoir areas, as reservoir impoundments may activate unstable slopes. Although satellite time-series InSAR has been widely used in mapping active landslides, the tedious artificial interpretation of InSAR results limits the efficiency and reliability of landslides detection. We propose a set of procedures for deformation monitoring and continuous automatic landslide identification in wide reservoir areas. The local time window estimation can extract the nonlinear deformation in the time series InSAR signal, thus enhancing the deformation field. Spatial adaptive clustering enables the effective extraction of unstable slopes. The abnormal deformation trends of landslides can be updated through the continuous identification of new results and comparison with historical results. The procedure is used to continuously detect unstable slopes in the Lianghekou reservoir area before and after the impoundment. Combining the ascending and descending SAR data of Sentinel-1, 109 historically active landslides were found and 18 new landslides were activated within one year after the first water storage. The distribution and types of unstable slopes were analyzed, followed by two case studies. The first case shows the different effects of the two-stage water storage on slope deformations, indicating rising water levels' critical role in landslide activation. The second case shows the consolidation settlement of the embankment dam and the influence of water storage on slope deformation monitoring. The results demonstrate the effectiveness of the InSAR automatic landslide identification and this study provides a technical reference for similar reservoir area.

Fruit color is one of the most important traits of jujube (Ziziphus jujuba Mill.). However, the differences in the pigments of different varieties of Jujube are not well studied. In addition, the genes responsible for fruit color and their underlying molecular mechanisms remain unclear. In this study, two jujube varieties, namely “Fengmiguan” (FMG) and “Tailihong” (TLH), were considered. The metabolites from jujube fruits were investigated using ultra-high-performance liquid chromatography/tandem mass spectrometry. Transcriptome was used to screen anthocyanin regulatory genes. The gene function was confirmed by overexpression and transient expression experiments. The gene expression was analyzed by quantitative reverse transcription polymerase chain reaction analyses and subcellular localization. Yeast-two-hybrid and bimolecular fluorescence complementation were used to screen and identify the interacting protein. These cultivars differed in color owing to their respective anthocyanin accumulation patterns. Three and seven types of anthocyanins were found in FMG and TLH, respectively, which played a key role in the process of fruit coloration. ZjFAS2 positively regulates anthocyanin accumulation. The expression profile of ZjFAS2 exhibited its different expression trends in different tissues and varieties. Subcellular localization experiments showed that ZjFAS2 was localized to the nucleus and membrane. A total of 36 interacting proteins were identified, and the possibility of ZjFAS2 interacting with ZjSHV3 to regulate jujube fruit coloration was studied. Herein, we investigated the role of anthocyanins in the different coloring patterns of the jujube fruits and provided a foundation for elucidating the molecular mechanism underlying jujube fruit coloration.

Abstract The electrochemical reduction reaction of carbon dioxide (CO2RR) is considered to be an effective way to realize carbon neutrality. As a type of intensively studied materials, covalent organic frameworks (COFs) with a tunable pore structure and various functional groups are promising catalysts for CO2RR. Herein, COF synthesized by 2,6‐diaminoanthraquinone and 2,4,6‐triformylphloroglucinol is employed to assist the synthesis of electrocatalysts from Cu single atoms (SAs) to nanoclusters by controlling the electrodeposition. A tandem catalyst for CO2‐to‐CH4 conversion is thus achieved by the Cu nanoclusters dispersed among the isolated Cu SAs in the COF network. It is proposed that CO2 is first reduced to CO over the atomically isolated Cu SAs, followed by diffusion onto the neighboring Cu nanoclusters for further reduction into CH4. In addition, mechanistic analysis suggests that the coordinated K+ ions on the COF network promote the activation of CO2 and the adsorption of reaction intermediates, thus realizing the suppressed hydrogen evolution reaction and selective production of CH4. This study presents a new insight of COFs for the confined synthesis of a tunable SA to nanocluster electrocatalysts, disclosing the great potential of COFs in electrocatalysis.

Ruan Hua Tang (RHT) and Ruan Hua Fang (RHF) are two Chinese herbal mixtures that have been used in clinical cancer treatment for decades. This study validated our hypothesis that RHT and RHF can inhibit lung tumor development in the mouse model of Benzo(a)pyrene-induced lung tumorigenesis. An RHT oral solution was diluted to 9% and 18% in water. RHF was mixed into the diet at 15% and 30% of total food in the final doses. Two weeks after injecting BP into mice, we administered RHT and RHF for eighteen weeks. We found that 9% and 18% RHT reduced tumor multiplicity by 36.05% and 38.81% (both p < 0.05) and the tumor load by 27.13% and 55.94% (p < 0.05); 15% and 30% RHF inhibited tumor multiplicity by 12.75% and 39.84% (p < 0.01) and the tumor load by 18.38% and 61.68% (p < 0.05). Ki67 expressions in the 9% and 18% RHT groups were 19.55% and 11.51%, significantly lower than in the control (33.64%). The Ki67 levels in the 15% and 30% RHF groups were 15.56% and 14.04%, significantly lower than in the control (27.86%). Caspase 3 expressions in the 9% and 18% RHT groups were 5.24% and 7.32%, significantly higher than in the control (2.39%). Caspase 3 levels in the 15% and 30% RHF groups were 6.53% and 4.74%, significantly higher than in the control (2.07%). The bio-absorption was confirmed via a pharmacokinetic test. This study showed that RHT and RHF are safe and can inhibit lung tumor development, with anti-proliferative and pro-apoptotic effects.

Returning extraterrestrial samples to Earth has become essential for future deep space exploration. Achieving a comprehensive evaluation of the physical and chemical properties of samples with minimal damage is key to analyzing extraterrestrial samples in the future, as well as to the future sampling and returning of heterogeneous solid samples. This article aims to reconstruct the three-dimensional internal structure of high-contrast objects, select sections of interest through internal structure and detail features, and then analyze the physical and chemical properties of the samples based on laser spectroscopy technology. This paper proposes a strategy based on Raman mapping and X-ray phase-contrast imaging technology to reconstruct the three-dimensional internal structure of a heterogeneous solid sample and detect the substance composition of the region of interest. This study takes meteorite samples as an example and uses X-ray phase-contrast imaging technology to distinguish and reconstruct the spatial distribution of different components in the meteorite, providing a three-dimensional visualization reference with a high spatial resolution for the spatial positioning of the region of interest. Raman spectroscopy, in combination with LIBS, was used to further identify the meteorite as pallasite and to achieve the spectral image fusion of high spatial and high spectral resolutions. The experimental results show that the unknown meteorite’s three-dimensional structure and its components’ spatial distribution can be evaluated based on Raman mapping combined with X-ray phase-contrast imaging technology. This article provides a highly valuable analytical strategy by which to analyze samples returned from deep space exploration.

Meteorites are an essential reference for human exploration of the universe and its cosmic evolution and an essential research object for searching for extraterrestrial life. Ways to quickly identify and screen suspected meteorite samples have become the foundation and prerequisite for research on high-value meteorite samples. Therefore, this paper proposes a Raman mapping-assisted micro-laser induced breakdown spectroscopy (micro-LIBS) technology for field detection of suspected meteorite material composition without sample pre-processing, with a high detection speed and cost-effectiveness, to realize the detection of element composition and molecular structure. Raman mapping carries out multispectral imaging with high spectral resolution of the region of interest. The fusion of Raman mapping and optical microscopy images can provide mineral categories and spatial distribution characteristics in regions of interest. A quantitative analysis model for Fe, Mg, and Na elements was constructed based on the multidimensional scaling–back propagation neural network (MDS-BPNN) algorithm. The determination coefficient of the model test set was better than 0.997, and the root mean square error was better than 0.65. The content of Fe, Mg, and Na elements in the meteorite was preliminarily evaluated, providing a reference for further analysis of element information in spectral image fusion data. The Raman–LIBS combined technology has significant application potential in rapidly evaluating suspected meteorite samples. Without high-end precision instruments or field research, this technology can provide scientists with significant reference value atomic and molecular spectral information. At the same time, this technology can be extended to other petrology research. We offer a fast, efficient, cost-effective, and reliable analysis scheme for reference.

Epstein-Barr virus (EBV)-encoded latent genes are reported to regulate PD-L1 expression to promote immune escape. Here, the authors show that EBV-encoded miRNAs EBV-miR-BART11 and EBV-miR-BART17-3p upregulate PD-L1 expression in nasopharyngeal carcinoma and gastric cancer by targeting FOXP1 and PBRM1.

Abstract Background Circular RNAs play an important role in tumor genesis and progression, but they have not been sufficiently studied in patients with nasopharyngeal carcinoma (NPC). Methods The circular RNA, circCAMSAP1, was screened in NPC cells by RNA sequencing analysis. The expression of circCAMSAP1 in NPC tissues was examined by real-time quantitative polymerase chain reaction (RT-qPCR) and in situ hybridization. Wound-healing, transwell, MTT and flow cytometry assays, and nude mouse tumor models were used to explore the effect of circCAMSAP1 on proliferation and metastasis of NPC in vitro or in vivo. The downstream proteins regulated by circCAMSAP1 were screened using mass spectrometry. The interaction between circCAMSAP1 and the SERPINH1 mRNA was identified using the circular RNA immunoprecipitation method and the luciferase reporter assay. The interaction between SERPINH1 and transcription factor c-Myc was verified through Co-immunoprecipitation (Co-IP) and immunofluorescence. The effect of c-Myc on the generation of circCAMSAP1 was examined through RT-qPCR and chromatin immunoprecipitation. Finally, the splicing factors that promote the production of circCAMSAP1 were explored by RT-qPCR and RNA immunoprecipitation (RIP). Results We found that circCAMSAP1 was highly expressed in NPC tissues and promoted NPC proliferation and metastasis. Additionally, circCAMSAP1 promoted SERPINH1 expression through improved SERPINH1 mRNA stability by binding to the 3′-untranslated region (3’UTR) of SERPINH1. Highly expressed SERPINH1 reduced the ubiquitination-degradation rate of c-Myc, causing increased tumorigenesis. Meanwhile, c-Myc, cooperating with splicing factor 10 (SRSF10), could also promote CAMSAP1 pre-mRNA transcription and back-splicing, forming a positive feedback of circCAMSAP1 production, resulting in the proliferation and metastasis of NPC. Conclusions Our findings revealed that circCAMSAP1 promotes NPC proliferation and metastasis by binding to the 3’UTR of SERPINH1, suggesting that the positive feedback of circCAMSAP1-SERPINH1-c-Myc may serve as a prognostic biomarker or therapeutic target in patients with NPC.

Epidermal growth factor receptor (EGFR) mutations occur in about 50% of lung adenocarcinomas in Asia and about 15% in the US. EGFR mutation-specific inhibitors have been developed and made significant contributions to controlling EGFR mutated non-small cell lung cancer. However, resistance frequently develops within 1 to 2 years due to acquired mutations. No effective approaches that target mutant EGFR have been developed to treat relapse following tyrosine kinase inhibitor (TKI) treatment. Vaccination against mutant EGFR is one area of active exploration. In this study, we identified immunogenic epitopes for the common EGFR mutations in humans and formulated a multi-peptide vaccine (Emut Vax) targeting the EGFR L858R, T790M, and Del19 mutations. The efficacy of the Emut Vax was evaluated in both syngeneic and genetic engineered EGFR mutation-driven murine lung tumor models with prophylactic settings, where the vaccinations were given before the onset of the tumor induction. The multi-peptide Emut Vax effectively prevented the onset of EGFR mutation-driven lung tumorigenesis in both syngeneic and genetically engineered mouse models (GEMMs). Flow cytometry and single-cell RNA sequencing were conducted to investigate the impact of Emut Vax on immune modulation. Emut Vax significantly enhanced Th1 responses in the tumor microenvironment and decreased suppressive Tregs to enhance anti-tumor efficacy. Our results show that multi-peptide Emut Vax is effective in preventing common EGFR mutation-driven lung tumorigenesis, and the vaccine elicits broad immune responses that are not limited to anti-tumor Th1 response.

Abstract Lung cancer is the leading cause of cancer death worldwide. Vaccination against EGFR can be one of the venues to prevent lung cancer. Blocking glutamine metabolism has been shown to improve anticancer immunity. Here, the authors report that JHU083, an orally active glutamine antagonist prodrug designed to be preferentially activated in the tumor microenvironment, has potent anticancer effects on EGFR‐driven mouse lung tumorigenesis. Lung tumor development is significantly suppressed when treatment with JHU083 is combined with an EGFR peptide vaccine (EVax) than either single treatment. Flow cytometry and single‐cell RNA sequencing of the lung tumors reveal that JHU083 increases CD8+ T cell and CD4+ Th1 cell infiltration, while EVax elicits robust Th1 cell‐mediated immune responses and protects mice against EGFRL858R mutation‐driven lung tumorigenesis. JHU083 treatment decreases immune suppressive cells, including both monocytic‐ and granulocytic‐myeloid‐derived suppressor cells, regulatory T cells, and pro‐tumor CD4+ Th17 cells in mouse models. Interestingly, Th1 cells are found to robustly upregulate oxidative metabolism and adopt a highly activated and memory‐like phenotype upon glutamine inhibition. These results suggest that JHU083 is highly effective against EGFR‐driven lung tumorigenesis and promotes an adaptive T cell‐mediated tumor‐specific immune response that enhances the efficacy of EVax.

The stream sediment (SS) records evolution information of the water system structure and sedimentary environment in specific regions during different geological periods, which is of great significance for studying the ancient planetary environment and the law of water system changes. Based on the SS of different geographical environments on Earth, remote laser-induced breakdown spectroscopy (remote-LIBS) technology combined with the multidimensional scaling-back propagation neural network (MDS-BPNN) algorithm was used to conduct an in-depth analysis of remote qualitative and quantitative detection of the elemental composition and content of SS. The results show that the detection system based on remote LIBS combined with an artificial neural network algorithm can achieve an ideal quantitative analysis of major and trace elements. The coefficients of determination (R2) of the test set for major elements is greater than 0.9996, and the root mean square error (RMSE) is less than 0.7325. The coefficients of determination (R2) of the test set for trace elements is greater than 0.9837, and the root mean square error is less than 42.21. In addition, for the application scenario of exploring extraterrestrial life, biominerals represented by stromatolite phosphorite (SP) are easy to form sand and enter into SS under weathering. Therefore, this paper discusses the feasibility of using remote-LIBS technology to detect and identify such minerals under the disappearance of SPs’ macro- and micro-characteristics. From our research, we can find that remote-LIBS technology is the preferred candidate for discovering dust-covered biominerals. In geological environments rich in water system sedimentary rocks, such as Mars’ ancient riverbeds, LIBS technology is crucial for deciphering the “life signals” hidden in the Martian sand.

This paper evaluates the potential application of Raman baselines in characterizing organic deposition. Taking the layered sediments (Stromatolite) formed by the growth of early life on the Earth as the research object, Raman spectroscopy is an essential means to detect deep-space extraterrestrial life. Fluorescence is the main factor that interferes with Raman spectroscopy detection, which will cause the enhancement of the Raman baseline and annihilate Raman information. The paper aims to evaluate fluorescence contained in the Raman baseline and characterize organic sedimentary structure using the Raman baseline. This study achieves spectral image fusion combined with mapping technology to obtain high spatial and spectral resolution fusion images. To clarify that the fluorescence of organic matter deposition is the main factor causing Raman baseline enhancement, 5041 Raman spectra were obtained in the scanning area of 710 μm × 710 μm, and the correlation mechanism between the gray level of the light-dark layer of the detection point and the Raman baseline was compared. The spatial distribution of carbonate minerals and organic precipitations was detected by combining mapping technology. In addition, based on the BI-IHS algorithm, the spectral image fusion of Raman fluorescence mapping and reflection micrograph, polarization micrograph, and orthogonal polarization micrograph are realized, respectively. A fusion image with high spectral resolution and high spatial resolution is obtained. The results show that the Raman baseline can be used as helpful information to characterize stromatolite organic sedimentary structure.

Abstract Background Circular RNAs (circRNAs) are widely expressed in human cells and are closely associated with cancer development. However, they have rarely been investigated in the context of nasopharyngeal carcinoma (NPC). Methods We screened a new circRNA, circRNF13, in NPC cells using next-generation sequencing of mRNA. Reverse transcription polymerase chain reaction and RNA fluorescence in situ hybridization were used to detect circRNF13 expression in 12 non-tumor nasopharyngeal epithelial (NPE) tissues and 36 NPC samples. Cell proliferation was detected using MTT and flow cytometry assays, and colony formation capability was detected using colony formation assays. Cell migration and invasion were analyzed using wound-healing and Transwell assays, respectively. Cell glycolysis was analyzed using the Seahorse glycolytic stress test. Glucose transporter type 1 (GLUT1) ubiquitination and SUMOylation modifications were analyzed using co-immunoprecipitation and western blotting. CircRNF13 and Small Ubiquitin-like Modifier 2 (SUMO2) interactions were analyzed using RNA pull-down and luciferase reporter assays. Finally, to test whether circRNF13 inhibited NPC proliferation and metastasis in vivo, we used a xenograft nude mouse model generated by means of subcutaneous or tail vein injection. Results We found that circRNF13 was stably expressed at low levels in NPC clinical tissues and NPC cells. In vitro and in vivo experiments showed that circRNF13 inhibited NPC proliferation and metastasis. Moreover, circRNF13 activated the SUMO2 protein by binding to the 3′- Untranslated Region (3′-UTR) of the SUMO2 gene and prolonging the half-life of SUMO2 mRNA. Upregulation of SUMO2 promotes GLUT1 degradation through SUMOylation and ubiquitination of GLUT1, which regulates the AMPK-mTOR pathway by inhibiting glycolysis, ultimately resulting in the proliferation and metastasis of NPC. Conclusions Our results revealed that a novel circRNF13 plays an important role in the development of NPC through the circRNF13-SUMO2-GLUT1 axis. This study implies that circRNF13 mediates glycolysis in NPC by binding to SUMO2 and provides an important theoretical basis for further elucidating the pathogenesis of NPC and targeted therapy.

When combined with a MHCII-directed KRAS peptide vaccine, the authors reported that CA170 suppressed lung tumor development by increasing CD8+ T cell and decreasing MDSC and Treg infiltration, while the Kras vaccine induced CD4+ effector T cell expansion

Abstract Atovaquone, an FDA‐approved drug for malaria, is known to inhibit mitochondrial electron transport. A recently synthesized mitochondria‐targeted atovaquone increased mitochondrial accumulation and antitumor activity in vitro. Using an in situ vaccination approach, local injection of mitochondria‐targeted atovaquone into primary tumors triggered potent T cell immune responses locally and in distant tumor sites. Mitochondria‐targeted atovaquone treatment led to significant reductions of both granulocytic myeloid‐derived suppressor cells and regulatory T cells in the tumor microenvironment. Mitochondria‐targeted atovaquone treatment blocks the expression of genes involved in oxidative phosphorylation and glycolysis in granulocytic‐myeloid‐derived suppressor cells and regulatory T cells, which may lead to death of granulocytic‐myeloid‐derived suppressor cells and regulatory T cells. Mitochondria‐targeted atovaquone inhibits expression of genes for mitochondrial complex components, oxidative phosphorylation, and glycolysis in both granulocytic‐myeloid‐derived suppressor cells and regulatory T cells. The resulting decreases in intratumoral granulocytic‐myeloid‐derived suppressor cells and regulatory T cells could facilitate the observed increase in tumor‐infiltrating CD4+ T cells. Mitochondria‐targeted atovaquone also improves the anti‐tumor activity of PD‐1 blockade immunotherapy. The results implicate granulocytic‐myeloid‐derived suppressor cells and regulatory T cells as novel targets of mitochondria‐targeted atovaquone that facilitate its antitumor efficacy.

Atmospheric propagation delay correction is the key to improving the accuracy of deformation measurement of satellite interferometric synthetic aperture radar (InSAR). The empirical phase-elevation models and external data-based models present uneven performances of atmospheric delay correction for InSAR deformation monitoring. In this study, based on our previous fusion of delays predicted by multiple weather models (FDWM), we propose a new approach of adaptive fusion of multi-source tropospheric delay (AFMTD) estimates derived from multiple models over wide areas, i.e., ERA5, GACOS, WRF, MERRA2, NARR, MODIS, Linear model, and Powerlaw model. The spatially varying scaling algorithm is employed to refine the tropospheric delays predicted by the weather models. Meanwhile, we adopt a multiple-window strategy to cope with the spatially lateral variation of tropospheric delays. The AFMTD not only improves the spatial heterogeneity of tropospheric delay, but also adaptively combines multiple models to achieve a more reliable delay estimation. This AFMTD method is incorporated into the StaMPS-SBAS procedure. We compared the AFMTD with other single models using ENVISAT ASAR and Sentinel-1 datasets over Los Angeles of Southern California. The result of ASAR first demonstrates the effectiveness and reliability of the AFMTD method by referring to the assumed ground truth of simultaneous MERIS observations. The results of Sentinel-1 data show that over 95% of unwrapped interferograms have the minimum root-mean-square values after AFMTD correction for both descending and ascending tracks. The validation against GPS observation presents that the RMSEs of InSAR displacement time series after AFMTD correction decreases at more than 90% of 125 GPS stations. The average reductions of RMSE are 35.79% and 36.28% for descending and ascending data, respectively, and the maximum improvement is more than 70%. Overall, the proposed AFMTD method outperforms any single model for InSAR tropospheric delay correction and provides an open framework to fuse multi-source tropospheric delay estimates.

Abstract Background Magnolia extract (ME) is known to inhibit cancer growth and metastasis in several cell types in vitro and in animal models. However, there is no detailed study on the preventive efficacy of ME for oral cancer, and the key components in ME and their exact mechanisms of action are not clear. The overall goal of this study is to characterize ME preclinically as a potent oral cancer chemopreventive agent and to determine the key components and their molecular mechanism(s) that underlie its chemopreventive efficacy. Methods The antitumor efficacy of ME in oral cancer was investigated in a 4-nitroquinoline-1-oxide (4NQO)-induced mouse model and in two oral cancer orthotopic models. The effects of ME on mitochondrial electron transport chain activity and ROS production in mouse oral tumors was also investigated. Results ME did not cause detectable side effects indicating that it is a promising and safe chemopreventive agent for oral cancer. Three major key active compounds in ME (honokiol, magnolol and 4-O-methylhonokiol) contribute to its chemopreventive effects. ME inhibits mitochondrial respiration at complex I of the electron transport chain, oxidizes peroxiredoxins, activates AMPK, and inhibits STAT3 phosphorylation, resulting in inhibition of the growth and proliferation of oral cancer cells. Conclusion Our data using highly relevant preclinical oral cancer models, which share histopathological features seen in human oral carcinogenesis, suggest a novel signaling and regulatory role for mitochondria-generated superoxide and hydrogen peroxide in suppressing oral cancer cell proliferation, progression, and metastasis. Hydroxy genkwanin (HGK), a flavonoid compound from natural resources, showed good inhibition against the growth of breast tumor cells. However, the poor solubility restricted the further study and the in vivo drug delivery of HGK. We prepared HGK nanosuspensions by antisolvent precipitation method and investigated their characterization, stability, hemolysis probability, release behavior in vitro, antitumor activity in vitro and in vivo, and preliminary safety through acute toxicity experiments. The resultant HGK nanosuspensions (HGK-NSps) showed an average diameter of (261.1 ± 4.8 nm), a narrow particle size distribution (PDI of 0.12 ± 0.01), spherical morphology, high drug-loading content (39.9 ± 2.3%, w/w), and good stability in various physiological media. HGK-NSps was safe for intravenous injection at low concentration and HGK was slowly released from the obtained nanosuspensions. HGK-NSps showed stronger cytotoxicity than free HGK against many tumor cells in vitro. Especially against MCF-7 cells, the IC50 value was decreased to 1.0 μg/mL, 5-fold lower than the HGK solution. In the in vivo antitumor activity study HGK-NSps (40 mg/kg) displayed a similar therapeutic effect to that of the paclitaxel injection (8 mg/kg). The preliminary acute toxicity test showed that even at the highest dose of 360 mg/kg (iv), HGK-NSps had 100% of mice survival and all the mice were in a good state, suggesting a maximum tolerated dose more than 360 mg/kg. The effective antitumor effect and good tolerance showed HGK-NSps were likely to become a safe and effective antitumor drug for the treatment of breast cancer in the future.

Due to their various biological activities that are beneficial to human health and antitumor effect, flavonoid compounds have attracted much attention in recent years. Hydrous icaritin (HICT) was such a flavonoid that can inhibit the growth of breast cancer and cancer stem cells. In order to overcome the insolubility problem, HICT was fabricated into nanorods (NRs) through anti-solvent precipitation in this paper using D-α tocopherol acid polyethylene glycol succinate and sodium oleate as a co-stabilizer meanwhile using the mixture of ethanol and acetone (1:2, v/v) as the organic solvent. The obtained HICT NRs showed an average particle size 222.0 nm with a small polydispersity index value of 0.124 and a high zeta potential of – 49.5 mV. HICT NRs could maintain similar particle size in various physiological medium and could be directly lyophilized without the addition of any cytoprotectants and then reconstituted into a colloidal system of similar size. The resultant HICT NRs had a high drug loading content of 55.6% and released HICT in a steady and constant pattern. MTT assay indicated NRs enhanced HICT’s antitumor activity to ninefold against MCF-7 breast carcinoma cells. In vivo studies demonstrated oral administration free HICT had almost no tumor inhibitory effect while HICT NRs showed a tumor inhibition rate of 47.8%. When intravenously injected, HICT NRs displayed similar therapeutic efficacy to paclitaxel injections (70.4% vs. 74.5%, TIR). This may be partly due to the high accumulation of the injected HICT NRs in tumor ranking only second to that in the liver but much higher than in other organs. These results demonstrated that HICT NRs could be a promising antitumor agent for the treatment of breast cancer in clinic.

Abstract Disulfiram (DSF) has been considered as “Repurposing drug” in cancer therapy in recent years based on its good antitumor efficacy. DSF is traditionally used as an oral drug in the treatment of alcoholism. To overcome its rapid degradation and instability, DSF nanosuspensions (DSF/SPC-NSps) were prepared using soybean lecithin (SPC) as a stabilizer of high drug-loaded content (44.36 ± 1.09%). Comprehensive characterization of the nanosuspensions was performed, and cell cytotoxicity, in vivo antitumor efficacy and biodistribution were studied. DSF/SPC-NSps, having a spherical appearance with particle size of 155 nm, could remain very stable in different physiological media, and sustained release. The in vitro MTT assay indicated that the cytotoxicity of DSF/SPC-NSps was enhanced remarkably compared to free DSF against the 4T1 cell line. Abstract MicroRNAs are potential candidates for lung cancer prevention and therapy. A major limitation is the lack of an efficient delivery system to directly deliver miRNA to cancer cells while limiting systemic exposure. The delivery of miRNA via inhalation is a potential strategy for lung cancer prevention in high‐risk individuals. In this study, the authors investigate the efficacy of aerosolized let‐7b miRNA treatment in lung cancer prevention. Let‐7b shows significant inhibition of B[a]P‐induced lung adenoma with no detectable side effects. Single‐cell RNA sequencing of tumor‐infiltrating T cells from primary tumors reveals that Let‐7b post‐transcriptionally suppresses PD‐L1 and PD‐1 expression in the tumor microenvironment, suggesting that let‐7b miRNAs may promote antitumor immunity in vivo. Let‐7b treatment decreases the expression of PD‐1 in CD8+ T cells and reduces PD‐L1 expression in lung tumor cells. The results suggest that this aerosolized let‐7b mimic is a promising approach for lung cancer prevention, and that the in vivo tumor inhibitory effects of let‐7b are mediated, at least in part, by immune‐promoting effects via downregulating PD‐L1 in tumors and/or PD‐1 on CD8+ T cells. These changes potentiate antitumor CD8+ T cell immune responses, and ultimately lead to tumor inhibition.

As the longest water transfer route in the world, the South-North Water Diversion Project (SNWDP) plays a key role in mitigating the serious water shortage problem in the arid north China, and thus the routine stability monitoring of this mega infrastructure becomes a critical issue to ensure continuous fresh water supply. Compared with traditional geodetic tools such as leveling and GNSS that can only measure deformation at discrete points along the canal, modern satellite Synthetic Aperture Radar Interferometry (InSAR) that can capture subtle ground deformation over wide areas is much more suitable to accomplish this challenging task. In this study, we carried out multi-scale InSAR analyses with multi-track Sentinel-1 data stacks to monitor deformation along the Middle Route canal of SNWDP. A coarse-resolution InSAR analysis was first carried out to map regional land subsidence in north China, and the result suggested that the Middle Route canal bypassed the huge subsidence funnel in the southeast part of Hebei province and exhibited overall stability. Afterwards, the results of relatively high-resolution InSAR analyses performed within the 5 km buffering zones along the entire canal revealed that a few sections were affected by significant embankment deformation or land subsidence. In particular, the high-filled canal sections are more prone to deform, such as the cases in Ye County and Jiaozuo City in Henan province. The validation of InSAR deformation measurements with synchronous leveling in Jiaozuo City indicated that a measurement accuracy of 6 mm was achieved. Another big problem identified is the severe uneven land subsidence along the Tianjin Branch Route, which may pose great threats upon the long-term safety of local water transfer operation, and must be handled properly to avoid further degradation.

Abstract Herein, we constructed urchin‐like TiO2 hollow nanospheres (HNSs) by hydrothermal targeted etching, and used it as a substrate to load semi‐metal 1T′‐MoS2 nanosheets as effective nitrogen reduction reaction (NRR) electrocatalysts. 1T′‐MoS2/TiO2 HNSs composites display outstanding NRR activity with the highest NH3 yield of 29.62 μg h−1 mg−1cat. at −0.75 V versus RHE. Besides, the highest FE of 24.9% is obtained at −0.65 V. The remarkable NRR performance is attributed to the high conductivity of 1T′‐MoS2 and the urchin‐like structure of TiO2 hollow nanospheres. DFT calculations display that the 1T′‐MoS2 in 1T′‐MoS2/TiO2 makes the activation and further reduction of *N2 more thermodynamically favorable than pristine TiO2, contributing to better NRR catalytic activities. 15N isotopic labeling experiment reveals that N in produced NH3 comes from N2 of electrolyte.

Background: No effective approaches to target mutant Kras have yet been developed. Immunoprevention using KRAS-specific antigenic peptides to trigger T cells capable of targeting tumor cells relies heavily on lipid metabolism. To facilitate better TCR/peptide/MHC interactions that result in better cancer preventive efficacy, we combined KVax with avasimibe, a specific ACAT1 inhibitor, tested their anti-cancer efficacy in mouse lung cancer models, where Kras mutation was induced before vaccination. Methods: Control of tumor growth utilizing a multi-peptide Kras vaccine was tested in combination with avasimibe in a syngeneic lung cancer mouse model and a genetically engineered mouse model (GEMM). Activation of immune responses after administration of Kras vaccine and avasimibe was also assessed by flow cytometry, ELISpot and IHC. Findings: We found that Kras vaccine combined with avasimibe significantly decreased the presence of regulatory T cells in the tumor microenvironment and facilitated CD8+ T cell infiltration in tumor sites. Avasimibe also enhanced the efficacy of Kras vaccines target mutant Kras. Whereas the Kras vaccine significantly increased antigen-specific intracellular IFN-γ and granzyme B levels in CD8+ T cells, avasimibe significantly increased the number of tumor-infiltrating CD8+ T cells. Additionally, modulation of cholesterol metabolism was found to specifically impact in T cells, and not in cancer cells. Interpretation: Avasimibe complements the efficacy of a multi-peptide Kras vaccine in controlling lung cancer development and growth. Abstract Genetic instability of tumor cells often leads to the occurrence of a large number of mutations, and expression of non-synonymous mutations can produce tumor-specific antigens called neoantigens. Neoantigens are highly immunogenic as they are not expressed in normal tissues. They can activate CD4+ and CD8+ T cells to generate immune response and have the potential to become new targets of tumor immunotherapy. The development of bioinformatics technology has accelerated the identification of neoantigens. The combination of different algorithms to identify and predict the affinity of neoantigens to major histocompatibility complexes (MHCs) or the immunogenicity of neoantigens is mainly based on the whole-exome sequencing technology. Tumor vaccines targeting neoantigens mainly include nucleic acid, dendritic cell (DC)-based, tumor cell, and synthetic long peptide (SLP) vaccines. The combination with immune checkpoint inhibition therapy or radiotherapy and chemotherapy might achieve better therapeutic effects. Currently, several clinical trials have demonstrated the safety and efficacy of these vaccines. Further development of sequencing technologies and bioinformatics algorithms, as well as an improvement in our understanding of the mechanisms underlying tumor development, will expand the application of neoantigen vaccines in the future.

Abstract The immune system plays important roles in tumor development. According to the immune-editing theory, immune escape is the key to tumor survival, and exploring the mechanisms of tumor immune escape can provide a new basis for the treatment of tumors. In this review, we describe the mechanisms of natural killer group 2D (NKG2D) receptor and NKG2D ligand (NKG2DL) in tumor immune responses. Natural killer (NK) cells are important cytotoxic cells in the immune system, and the activated NKG2D receptor on the NK cell surface can bind to NKG2DL expressed in tumor cells, enabling NK cells to activate and kill tumor cells. However, tumors can escape the immune clearance mediated by NKG2D receptor/NKG2DL through various mechanisms. The expression of NKG2D receptor on NK cells can be regulated by cells, molecules, and hypoxia in the tumor microenvironment. Tumor cells regulate the expression of NKG2DL at the level of transcription, translation, and post-translation and thereby escape recognition by NK cells. In particular, viruses and hormones have special mechanisms to affect the expression of NKG2D receptor and NKG2DL. Therefore, NKG2D\NKG2DL may have applications as targets for more effective antitumor therapy.

Summary: Immunotherapy with monoclonal antibodies targeting immune checkpoint molecules, including programmed death-1 (PD-1), PD ligand-1 (PD-L1), and cytotoxic T-lymphocyte-associated antigen (CTLA)-4, has become prominent in the treatment of many types of cancer. However, a significant number of patients treated with immune checkpoint inhibitors (ICIs) develop immune-related adverse events (irAEs). irAEs can affect any organ system, and although most are clinically manageable, irAEs can result in mortality or long-term morbidity. Factors that can predict irAEs remain elusive. Understanding the etiology of ICI-induced irAEs and ways to limit these adverse events are needed. In this review, we provide basic science and clinical insights on the mechanisms responsible for ICI efficacy and ICI-induced irAEs. We further provide insights into approaches that may uncouple irAEs from the ability of ICIs to kill tumor cells.

Summary: Although PD-1-blocking immunotherapies demonstrate significant therapeutic promise, a subset of the patients could develop hyperprogressive disease (HPD) with accelerated tumor growth after anti-PD1 immunotherapy. To elucidate the underlying mechanisms, we compared the mutational and transcriptional landscapes between the pre- and post-therapy tumors of two patients developing HPD after anti-PD-1 immunotherapy. In post-therapy HPD tumors, somatic mutations were found in known cancer genes, including tumor suppressor genes such as TSC2 and VHL, along with transcriptional upregulation of oncogenic pathways, including IGF-1, ERK/MAPK, PI3K/AKT, and TGF-β. We found that post-therapy HPD tumors were less immunogenic than pre-therapy tumors, concurrent with an increased presence of ILC3 cells, a subset of innate lymphoid cells. We also developed a gene expression signature predictive of HPD. Summary: We synthesized a mitochondria-targeted honokiol (Mito-HNK) that facilitates its mitochondrial accumulation; this dramatically increases its potency and efficacy against highly metastatic lung cancer lines in vitro, and in orthotopic lung tumor xenografts and brain metastases in vivo. Mito-HNK is >100-fold more potent than HNK in inhibiting cell proliferation, inhibiting mitochondrial complex ǀ, stimulating reactive oxygen species generation, oxidizing mitochondrial peroxiredoxin-3, and suppressing the phosphorylation of mitoSTAT3. Within lung cancer brain metastases in mice, Mito-HNK induced the mediators of cell death and decreased the pathways that support invasion and proliferation. In contrast, in the non-malignant stroma, Mito-HNK suppressed pathways that support metastatic lesions, including those involved in inflammation and angiogenesis. Mito-HNK showed no toxicity and targets the metabolic vulnerabilities of primary and metastatic lung cancers. The deficiency of available silicon (Si) incurred by year-round agricultural and horticultural practices highlights the significance of Si fertilization for soil replenishment. This study focuses on a novel and economical route for the synthesis of Si fertilizer via the calcination method using talc and calcium carbonate (CaCO3) as starting materials. The molar ratio of talc to CaCO3 of 1:2.0, calcination temperature of 1150 °C and calcination time of 120 min were identified as the optimal conditions to maximize the available Si content of the prepared Si fertilizer. X-ray diffraction (XRD) and Fourier-transform infrared spectroscopy (FTIR) characterizations elucidate the principles of the calcination temperature-dependent microstructure evolution of Si fertilizers, and the akermanite Ca2Mg(Si2O7) and merwinite Ca3Mg(SiO4)2 were identified as the primary silicates products. The results of release and solubility experiments suggest the content of available metallic element and slow-release property of the Si fertilizer obtained at the optimum preparation condition (Si-OPC). The surface morphology and properties of Si-OPC were illuminated by the results of scanning electron microscope (SEM), surface area and nitrogen adsorption analysis. The acceleration action of CaCO3 in the decomposition process of talc was demonstrated by the thermogravimetry-differential scanning calorimetry (TG-DSC) test. The pot experiment corroborates that 5 g kg−1 soil Si-OPC application sufficed to facilitate the pakchoi growth by providing nutrient elements. This evidence indicates the prepared Si fertilizer as a promising candidate for Si-deficient soil replenishment.

With the continuous expansion of the high-speed railway network in China, long-span railway bridges carrying multiple tracks demand reliable and fast testing procedures and techniques. Bridge dynamic behavior analysis is a critical process in ensuring safe operation of structures. In this study, we present some experimental results of the vibration monitoring of a four-track high-speed railway bridge with a metro–track on each side: the Nanjing–Dashengguan high-speed railway bridge (NDHRB). The results were obtained using a terrestrial microwave radar interferometer named IBIS-S. The radar measurements were interpreted with the support of lidar point clouds. The results of the bridge dynamic response under different loading conditions, including high-speed trains, metro and wind were compared with the existing bridge structure health monitoring (SHM) system, underlining the high spatial (0.5 m) and temporal resolutions (50 Hz–200 Hz) of this technique for railway bridge dynamic monitoring. The detailed results can help engineers capturing the maximum train-induced bridge displacement. The bridge was also monitored by the radar from a lateral position with respect to the bridge longitudinal direction. This allowed us to have a more exhaustive description of the bridge dynamic behavior. The different effects induced by the passage of trains through different tracks and directions were distinguished. In addition, the space deformation map of the wide bridge deck under the eccentric load of trains, especially along the lateral direction (30 m), can help evaluating the running stability of high-speed trains.
