Purpose Degenerative mitral stenosis (DMS) is associated with a poor prognosis. Although mean transmitral gradient (TMG) has shown a good correlation with outcome, little is known about the association between other echocardiographic parameters and prognosis in patients with DMS. The current study aimed to evaluate the prognostic value of left atrial volume index (LAVI) in patients with DMS. Methods A total of 155 patients with DMS (72[63-80] years, 67% female) were included. The population was divided according to LAVI: normal-sized LAVI (LAVI 34 ml/m2). Results Patients with enlarged LAVI had a higher left ventricular mass index (120[96-146] vs. 91[70-112] g/m2 p < 0.001), as well as a higher prevalence of significant mitral regurgitation and severe aortic stenosis (23% vs. 10% p = 0.046 and 38% vs. 15% p=0.001, respectively) compared to patients with normal-sized LAVI. During a median follow-up of 25 months, 56 (36%) patients died. Patients with enlarged LAVI had worse prognosis compared to patients with normal-sized LAVI (p = 0.026). In multivariable Cox regression model, an enlarged LAVI was independently associated with all-cause mortality (HR 2.009, 95% CI 1.040 to 3.880, P = 0.038). Conclusion An enlarged LAVI (> 34 ml/m2) is significantly associated with excess mortality in patients with DMS. After adjusting for potential confounders, an enlarged LAVI was the only parameter that remained independently associated with prognosis. Inspired by the biosystems, the artificial smart membrane to control the mass transport and molecular conversion has attracted increasing attention in the fields of membrane separation, desalination, nanofiltration, healthcare and environmental remediation. However, the trade-off limitations in polymeric membranes greatly hinder the development of smart membranes with high permeability and manipulability. Recently, inspired by the unique physical/chemical properties of two-dimensional (2D) materials, 2D materials-based smart membranes (2DSMs) with the ability of intelligent regulation under different stimuli are highly suitable for membrane applications. According to the desired properties, the 2DSMs with abundant functional groups can be designed through chemical modification to change the original properties and obtain tunable interlayer spacings under different external conditions. In this review, we summarize the recent progress on artificial smart membranes based on 2D materials. The design concept and fabrication strategy of 2DSMs are first introduced. Following that, the developed 2DSMs are introduced and classified by the type of responsive stimuli, including pH, magnetic field, electric field, light and temperature. Then, the 2DSMs exhibiting unique performances as membrane separation, pressure sensors, blue energy harvesting, photoelectrochemical sensors and biomimetic devices are presented. Finally, the perspectives and challenges in the developments of 2DSMs are discussed

Sodium-glucose cotransporter 2 (SGLT2) inhibitors are a class of anti-hyperglycemic drugs that has been steadily increasing in popularity due to its cardiovascular and renal benefits. Dual SGLT1/SGLT2 (SGLT1/2) inhibitors have potentially augmented anti-hyperglycemic action due to additional SGLT1 inhibition. This network meta-analysis aimed to compare the treatment effect across various outcomes between pure SGLT2 inhibitors and combined SGLT1/2 inhibitors in patients with diabetes.Four electronic databases (PubMed, Embase, Cochrane, and Scopus) were searched for randomized controlled trials published from inception to 15th January 2022. Frequentist network meta-analysis was conducted to summarize the treatment effects reported in individual trials, stratified by type 1 (T1DM) and type 2 diabetes mellitus (T2DM). This meta-analysis was registered on PROSPERO (CRD42020222031).Our meta-analysis included 111 articles, comprising a combined cohort of 103,922 patients. SGLT2 inhibitors (dapagliflozin, empagliflozin, canagliflozin, ipragliflozin, ertugliflozin, and luseogliflozin) and SGLT1/2 inhibitors (licogliflozin and sotagliflozin) were compared. Frequentist network meta-analysis demonstrated that in T2DM patients, SGLT1/2 inhibitors led to a decreased hazard rate of myocardial infarction (hazard ratio [HR] 0.74, 95% confidence interval [CI] 0.56-0.98) and stroke (HR 0.65, 95% CI 0.47-0.92) compared with SGLT2 inhibitors. SGLT2 inhibitors achieved a greater hemoglobin A1c (HbA1c) reduction than SGLT1/2 inhibitors (0.16%, 95% CI 0.06-0.26). In patients with T2DM, the risk of diarrhea (risk ratio [RR] 1.42, 95% CI 1.07-1.88) and severe hypoglycemia (RR 5.89, 95% CI 1.41-24.57) were found to be higher with SGLT1/2 inhibitor use compared with SGLT2 inhibitor use. No differences were observed for cardiovascular, metabolic, and safety outcomes between SGLT1/2 inhibitors and SGLT2 inhibitors in patients with T1DM.In patients with T2DM, compared with pure SGLT2 inhibitors, combined SGLT1/2 inhibitors demonstrated a lower risk of myocardial infarction and of stroke, but were associated with a higher risk of diarrhea and severe hypoglycemia.

Supplementary Data from Human Leukocyte Antigen-G Protein Expression Is an Unfavorable Prognostic Predictor of Hepatocellular Carcinoma following Curative Resection

Infective endocarditis (IE) confers significant mortality and morbidity in patients with underlying mitral stenosis (MS), with both diseased native valves and after valvular procedures.We examined the clinical and echocardiographic parameters in association with the development of IE on patients with significant MS.A total of 478 consecutive patients with index echocardiographic diagnosis of significant mitral stenosis (mitral valve area1.5 cmIE was observed in 4.4% (n = 21) of the cohort. These patients were younger (45.3 ± 14.6 vs. 53.2 ± 15.5 years, p = 0.022) at the time of MS diagnosis. NT-proBNP was higher in patients who developed IE (13529 ± 12230 vs. 4381 ± 5875 pg/ml, p0.001), with larger left atrial diameter (54.4 ± 10.1 vs. 49.9 ± 9.5 mm, p = 0.040) and elevated pulmonary artery systolic pressure (PASP, 62.3 ± 17.2 vs. 47.2 ± 16.5 mmHg, p = 0.026). There was no significant difference in terms of MS severity, other concomitant valvulopathies, or etiology (rheumatic or degenerative) of MS. Patients who smoked had higher incidence of IE (33.3% vs. 14.2%, log-rank 7.27, p = 0.007). After adjusting for age, valve procedure and PASP, patients who smoked remained at elevated risk of IE (adjusted hazards ratio 2.99, 95% confidence interval 1.18-7.56, p = 0.021).IE occurs in a proportion of patients with MS. Smoking, younger age of diagnosis of MS as well as dilated left atria with elevated PASP may be associated with an elevated risk of this complication.

Natural polypeptides, a kind of molecular polymer with obvious biological activity, are widely existing in nature. They participate in various physiological activities of living organisms and play an important role in promoting human health. They are also widely applied in medicine, food, and cosmetic industries. By searching literature from Pubmed, Google Scholar, Web of Science, Springer Link and Elsevier, this work presents an overview of the preparation methods, the relationship between structure and function, and the application of natural polypeptides. The preparation methods mainly include solvent extraction, enzymatic decomposition, microbiological fermentation, chemical synthesis, genetic engineering recombination, and using cell free system. Natural polypeptide's physiological function mainly includes antioxidative, antibacterial, antihypertensive. This review could provide scientific basis for the research and development of natural polypeptide.

Cable’s fundamental frequency (CFF) is an important characteristic of the working state of long-span cable-stayed bridges. The change in the bridge’s temperature field will influence CFF by altering the cable’s tension and the cables’ sags. An accurate regression model between the temperature-induced variation of CFF and the real-time changing temperature field should be established. Then, the reference value of the temperature-induced variation of CFF can be obtained after inputting the real-time temperature data. In this study, an intelligent real-time prediction model for CFF is proposed based on the full-bridge temperature field, including the average temperature of the main beam, the vertical temperature difference of the main beam, and the temperature of the cable tower. Besides, a machine learning method named the long short-term memory (LSTM) network is exploited to ensure the nonlinear fitting performance of the model, and a paradigm for optimal hyperparameter selection and training strategy selection is provided. To verify the superiority of the LSTM-based model, the output accuracy of the linear regression, BP network, and LSTM network was tested and compared using the monitoring data collected from cable sensors in the main span and side span, which provides an important basis for the intelligent maintenance and sustainable operation of the bridge cables.

The coronavirus disease 2019 (COVID-19) pandemic, caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has resulted in a global health emergency. In addition to common respiratory symptoms, some patients with COVID-19 infections may experience a range of extra-pulmonary manifestations, such as digestive system involvement. Patients with COVID-19 have been reported to suffer from acute mesenteric ischemia (AMI) that is associated with disease-related severity and mortality. However, in the context of COVID-19, the exact cause of AMI has yet to be clearly defined. This review provides a comprehensive overview of the available data and elucidates the possible underlying mechanisms linking COVID-19 to AMI, in addition to highlighting therapeutic approaches for clinicians. Finally, given the severe global impact of COVID-19, we emphasize the importance of coordinated vaccination programs.

Traditional diagnostic strategies for infectious disease detection require benchtop instruments that are inappropriate for point-of-care testing (POCT). Emerging microfluidics, a highly miniaturized, automatic, and integrated technology, are a potential substitute for traditional methods in performing rapid, low-cost, accurate, and on-site diagnoses. Molecular diagnostics are widely used in microfluidic devices as the most effective approaches for pathogen detection. This review summarizes the latest advances in microfluidics-based molecular diagnostics for infectious diseases from academic perspectives and industrial outlooks. First, we introduce the typical on-chip nucleic acid processes, including sample preprocessing, amplification, and signal read-out. Then, four categories of microfluidic platforms are compared with respect to features, merits, and demerits. We further discuss application of the digital assay in absolute nucleic acid quantification. Both the classic and recent microfluidics-based commercial molecular diagnostic devices are summarized as proof of the current market status. Finally, we propose future directions for microfluidics-based infectious disease diagnosis.

The reaction of β-hydroxybutenyl radicals with O2 and subsequent reactions are regarded as crucial steps in the low-temperature oxidation of 1,3-butadiene. However, the reaction network is not complete and previous studies failed to include accurate rate constants. In this study, the reaction network of β-hydroxybutenyl radicals with O2 is supplemented, as well as the temperature- and pressure-dependent rate constants which are investigated by high-level quantum chemical calculation, combining with the transition state theory (TST), the variational transition state theory (VTST) and Rice–Ramsperger–Kassel–Marcus/master equation method (RRKM/ME). The results of quantum chemical calculations indicate that Waddington reaction is the dominant channel, while the intramolecular addition reactions of the radical center atoms to the double bonds are found to be important non-alkyl-analogue reactions. Moreover, some of the reactions are investigated in the present study as well, whose pressure-dependent kinetics have not been reported so far. The calculations indicate that addition reaction of β-hydroxybutenyl with O2 is pressure independent when the temperature is below 600 K, while above 600 K, the effect of pressure is increasingly significant. Furthermore, the formation of β-hydroxybutenylperoxy is a dominant channel when the pressure is above 1 atm, while below 0.1 atm, other channels begin to take place with the increase in temperature.

Alkali cations (A+ = Na+, K+, Rb+, Cs+) were coordinated with 1,2,3-hexamethylcucurbit[5]uril (1,2,3-HMeQ[5]) using different structure directing agents. Single crystal X-ray diffraction revealed the formation of heterometallic molecular capsule-like complexes in the presence of [CdCl4]2− anions as a structure directing agent. One portal of the 1,2,3-HMeQ[5] molecule is covered by a Cd2+ cation, while the other portal is covered by a A+ cation, and a chloride anion occupies the cavity of the Q[5] molecule. In contrast, in the presence of p-nitrophenol, a molecular bowl-like complex is formed, in which only one portal is covered by an A+ cation, and a water molecule occupies the cavity of the Q[5] molecule. The comparative study of these complexes suggest that the structure directing agents play important roles in the self-assembly of coordination complexes.

Alkaline anions, include CO32-, HCO3-, Al(OH)(4)(-), OH-, continuously released from bauxite residue (BR), will cause a potential disastrous impact on surrounding environment. The composition variation of alkaline anions, alkaline phase transformation pathway, and micro-morphological transition characteristics during the gypsum addition were investigated in an attempt to understand alkalinity stabilization behavior. Results demonstrated that alkaline anions stabilization degree in leachates can reach approximately 96.29%, whilst pH and alkalinity were reduced from 10.47 to 8.15, 47.39 mmol/L to 2 mmol/L, respectively. During the alkalinity stabilization, chemical regulation behavior plays significant role in driving the co-precipitation reaction among the critical alkaline anions (CO32-, HCO3-, Al(OH)(4)(-), OH-), with calcium carbonate (CaCO3)) being the most prevalent among the transformed alkaline phases. In addition, XRD and SEM-EDX analyses of the solid phase revealed that physical immobilization behavior would also influence the stability of soluble alkali and chemical bonded alkali due to released Ca2+ from gypsum which aggregated the clay particles and stabilized them into coarse particles with a blocky structure. These findings will be beneficial for effectively regulating strong alkalinity of BR.

Bauxite residue is a highly alkaline material generated from the production of alumina in which bauxite is dissolved in caustic soda. Approximately 4.4 billion tons of bauxite residues are either stockpiled or landfilled, creating environmental risks either from the generation of dust or migration of filtrates. High alkalinity is the critical factor restricting complete utilization of bauxite residues, whilst the application of alkaline regulation agents is costly and difficult to apply widely. For now, current industrial wastes, such as waste acid, ammonia nitrogen wastewater, waste gypsum and biomass, have become major problems restricting the development of the social economy. Regulation of bauxite residues alkalinity by industrial waste was proposed to achieve ‘waste control by waste’ with good economic and ecological benefits. This review will focus on the origin and transformation of alkalinity in bauxite residues using typical industrial waste. It will propose key research directions with an emphasis on alkaline regulation by industrial waste, whilst also providing a scientific reference point for their potential use as amendments to enhance soil formation and establish vegetation on bauxite residue disposal areas (BRDAs) following large-scale disposal.

Aiming at alkaline problem of bauxite residue, this work focused variation of alkaline characteristics in bauxite residue through phosphogypsum treatment. The results demonstrated that the pH of bauxite residue reduced from initial 10.83 to 8.70 when 1.50 wt% phosphogypsum was added for 91 d. The removal rates of free alkali and exchangeable sodium were 97.94% and 75.87%, respectively. Meanwhile, significant positive correlations (P OH–> AlO2–>HCO3–. In addition, alkaline phase decreased from 52.81% to 48.58% and gypsum stably presented in bauxite residue which continuously provided Ca2+ to inhibit dissolution of combined alkali. Furthermore, phosphogypsum promoted formation of macroaggregate structure, increased Ca2+, decreased Na+ and Al3+ on the surface of bauxite residue significantly, ultimately promoting soil formation in bauxite residue.

Patients with preexisting kidney disease or acute kidney injury had poorer outcomes in coronavirus disease 2019 (COVID-19) illness. Lymphopenia was associated with more severe illness. Risk stratification with simple laboratory tests may help appropriate site patients in a cost-effective manner and ease the burden on healthcare systems. We examined a ratio of serum creatinine level to absolute lymphocyte count at presentation (creatinine-lymphocyte ratio, CLR) in predicting outcomes in hospitalized patients with COVID-19. We analyzed 553 consecutive polymerase chain reaction-positive SARS-COV-2 hospitalized patients. Patients with end-stage kidney disease were excluded. Serum creatinine and full blood count (FBC) examination were obtained within the first day of admission. We examined the utility of CLR in predicting adverse clinical outcomes (requiring intensive care, mechanical ventilation, acute kidney injury requiring renal replacement therapy or death). An optimized cutoff of CLR 77 was derived for predicting adverse outcomes (72.2% sensitivity, and 83.9% specificity). Ninety-seven patients (17.5%) fell within this cut off. These patients were older and more likely to have chronic medical conditions. A higher proportion of these patients had adverse outcomes (13.4% vs 1.1%, P .001). On receiver operating curve analyses, CLR predicted patients who had adverse outcomes well (area under curve [AUC] = 0.82, 95%CI 0.72-0.92), which was comparable to other laboratory tests like serum ferritin, C-reactive protein and lactate dehydrogenase. Elevated CLR on admission, which may be determined by relatively simple laboratory tests, was able to reasonably discriminate patients who had experienced adverse outcomes during their hospital stay. This may be a simple and cost-effective means of risk stratification and triage.

Algal bloom (AB) is a serious water environment problem in China, but it is still unclear how AB affects microorganisms with special functions (such as nitrogen-, phosphorus-, sulfur- and iron-related bacteria) at different AB stages. Samples of water and sediment were taken in the AB region and free algal bloom (FAB) region of Chaohu Lake at different AB stages, and the numbers of N-, P-, S- and Fe-related bacteria were determined by culture-dependent methods. Results showed that during the whole experiment, numbers of N-, P-, S-, and Fe-related bacteria in the AB region were significantly higher than those in the FAB region, especially at the AB outbreak stage. In the FAB region, numbers of above bacteria showed little variation with time. In the AB region, however, numbers of N-, S-, and Fe-related bacteria evidently changed as ‘Λ’-type with AB process. They presented an upward trend at the AB formation stage and the initial stage of AB outbreak, and then showed a downward trend. Numbers of P-related bacteria also showed a similar trend as the above bacteria, but they kept high level for long time at the AB outbreak stage. This study indicated that AB produced different effects on microorganisms with special functions at different AB stages.

Background To explore the kinetic changes in virology, specific antibody response and imaging during the clinical course of COVID-19. Methods This observational study enrolled 20 patients with COVID-19, who were hospitalized between January 20–April 6, 2020, in the two COVID-19 designated hospitals of Zhoushan, Zhejiang and Rushan, Shandong, China, The laboratory findings, imaging, serum response to viral infection, and viral RNA level in the throat and stool samples were assessed from onset to recovery phase in patients with COVID-19. Results SARS-COV-2 RNA was positive as early as day four. It remained positive until day 55 post-onset in the sputum-throat swabs and became negative in most cases (55%) within 14 days after onset. Lymphocytopenia occurred in 40% (8/20) of patients during the peak infection period and returned to normal at week five. The most severe inflammation in the lungs appeared in week 2 or 3 after onset, and this was completely absorbed between week 6 and 8 in 85.7% of patients. All patients had detectable antibodies to the receptor binding domain (RBD), and 95% of these patients had IgG to viral N proteins. The antibody titer peaked at week four. Anti-S IgM was positive in 7 of 20 patients after week three. Conclusions All COVID-19 patients in this study were self-limiting and recovered well though it may take as long as 6–8 weeks. Our findings on the kinetic changes in imaging, serum response to viral infection and viral RNA level may help understand pathogenesis and define clinical course of COVID-19.

Diabetic peripheral neuropathy (DPN) is one of the most common chronic complications of diabetes. Symptoms of DPN mainly include spontaneous intractable pain that is diffuse and continuous and can last from several weeks to several months. DPN is associated with a high mortality rate and poor prognosis. Its pathogenesis is not fully understood, and clinical treatment is focused on relieving its clinical symptoms, as well as improving blood sugar control and cardiovascular risk factors. DPN and its clinically effective treatments need to be studied. This study discusses the treatment methods and pathogenesis of DPN, summarizes the related research progress, and attempts to provide a reference for DPN research.

The nano-friction phenomenon in a one-dimensional Frenkel–Kontorova (FK) model under Gaussian colored noise is investigated by using the molecular dynamic simulation method. The role of colored noise is analyzed through the inclusion of a stochastic force via a Langevin molecular dynamics method. Via the stochastic Runge–Kutta algorithm, the relationship between different parameter values of the Gaussian colored noise (the noise intensity and the correlation time) and the nano-friction phenomena such as hysteresis, the maximum static friction force is separately studied here. Similar results are obtained from the two geometrically opposed ideal cases: incommensurate and commensurate interfaces. It was found that the noise strongly influences the hysteresis and maximum static friction force and with an appropriate external driving force, the introduction of noise can accelerate the motion of the system, making the atoms escape from the substrate potential well more easily. Interestingly, suitable correlation time and noise intensity give rise to super-lubricity. It is noteworthy that the difference between the two circumstances lies in the fact that the effect of the noise is much stronger on triggering the motion of the FK model for the commensurate interface than that for the incommensurate interface.

This study focused on leaching behavior of alkaline anion and sodium in bauxite residue through ammonium chloride treatment. The results showed that the pH of bauxite residue decreased from 10.49 to 8.93, total alkaline anion ( HCO 3 − , CO 3 2 − , OH−, A 1 O 2 − ) concentration reduced from 38.89 to 25.50 mmol/L, leaching rate of soluble sodium was 80.86% with ammonium chloride addition of 0.75%, liquid/solid (L/S) ratio of 3 (mL/g), temperature of 30 °C and reaction time of 18 h; L/S ratio was the main factor affecting the removal of alkaline anion and the leaching of sodium. Furthermore, ammonium chloride promoted the dissolution of diaspore and changed the micro/morphological characteristics with the increase of massive structure. The findings of this work will contribute to achieve soil-formation of bauxite residue.

High-mobility group box 1 (HMGB1) is a highly conserved DNA-binding nuclear protein that facilitates gene transcription and the DNA repair response. However, HMGB1 may be released by necrotic cells as well as activated monocytes and macrophages following stimulation with lipopolysaccharide (LPS), interleukin-1β (IL-1β), or tumor necrosis factor-α (TNF-α). Extracellular HMGB1 plays a critical role in the pathogenesis of acute lung injury (ALI) through activating the nuclear transcription factor κB (NF-κB) P65 pathway, thus, it may be a promising therapeutic target in shock-induced ALI. Paeonol (Pae) is the main active component of Paeonia suffruticosa, which has been used to inhibit the inflammatory response in traditional Chinese medicine. We have proven that Pae inhibits the expression, relocation and secretion of HMGB1 in vitro. However, the role of Pae in the HMGB1-NF-κB pathway remains unknown. We herein investigated the role of Pae in LPS-induced ALI rats. In this study, LPS induced a marked decrease in the mean arterial pressure (MAP) and survival rate (only 25% after 72 h), and induced severe pathological changes in the lung tissue of rats, which was accompanied by elevated expression of HMGB1 and its downstream protein NF-κB P65. Treatment with Pae significantly improved the survival rate (>60%) and MAP, and attenuated the pathological damage to the lung tissue in ALI rats. Western blotting revealed that Pae also inhibited the total expression of HMGB1, NF-κB P65 and TNF-α in the lung tissue of ALI rats. Moreover, Pae increased the expression of HMGB1 in the nucleus, inhibited the production of HMGB1 in the cytoplasm, and decreased the expression of P65 both in the nucleus and cytoplasm of lung tissue cells in LPS-induced ALI rats. The results were in agreement with those observed in the in vitro experiment. These findings indicate that Pae may be a potential treatment for ALI through its repression of the HMGB1-NF-κB P65 signaling pathway.

Bauxite residue is a highly alkaline waste containing soluble alkaline anions, which can cause environmental concerns. The optimal leaching conditions, distribution of alkaline anions, types of pivotal alkaline anions and their dissolution behaviors were investigated based on the combination of single factors-orthogonal experiments and leaching stage experiment. Using a two-stage leaching, 86% of the soluble alkaline anions ( CO 3 2 − , HCO 3 − , Al ( OH ) 4 − , OH − ) were leached with a L/S ratio of 2 mL/g, at 30 °C, over 23 h. During the first stage of leaching, approximately 88% of alkaline anions were leached from the dissolution of free alkali (NaOH, carbonate, bicarbonate, NaAl(OH)4) with the rest originating from the dissolution of alkaline minerals (calcite, cancrinite and hydrogarnet). Supernatant alkalinity was 69.78 mmol/L with CO 3 2 − accounting for 75%. Furthermore, carbonate leaching was controlled by solid film diffusion using the Stumm Model with an apparent activation energy of 10.24 kJ/mol.

Hydrogen abstraction reactions play important roles in combustion process of fuel. In the present work, we carried out a comprehensive study on hydrogen abstraction from quadricyclane by seven radicals (H, O(3P), OH, CH3, HO2, C2H5, and CH3O). Geometry optimization and frequency calculations of all species are performed employing BH&HLYP/6-311++G(d,p) and single point energies are computed at CCSD(T)/CBS(D-T) level. High-pressure-limit rate constants for 21 elementary reactions are determined varying from 500 to 2500 K according to the conventional transition-state theory and represented with the modified Arrhenius form. Base on barrier heights, OH radical has the highest reactivity among the seven radicals and the H atom located at the secondary site is the easiest to be abstracted. The apparent discrepancies of rate parameters between our calculated results and the values taken from literatures indicate that present quantum chemical calculations are valuable and contribute to the future construction of combustion mechanism of quadricyclane.

The main causes for deterioration of carbon blocks are due to the penetration and dissolution of hot metal against pores and surfaces. The active national standard GB/T 24201-2009 can only judge the hot metal dissolution, but the penetration of hot metal during test has been neglected. On the basis of national standard, a new measurement and characterization method is proposed. The study shows that with the help of back-scattered electron detect technology combined with statistical analysis, the penetration depth of hot metal could be quantitative evaluated, which could provide a reference for optimize the production of blast furnace refractories.

Under harmful environmental conditions, stress granules (SGs), macromolecular aggregates that are associated with cell survival and death, are produced in the eukaryotic cytoplasm. However, whether and how microRNAs (miRNAs/miRs) modulate SG formation induced by acute ischemic stroke has not been investigated. In the present study, a rat model of middle cerebral artery occlusion (MCAO) was utilized and miRNA array profiling and reverse transcription-quantitative polymerase chain reaction were performed. The results revealed that miR-335 was downregulated during acute ischemic stroke, which was concomitant with reduced SG formation, enhanced apoptosis levels and increased Rho associated protein kinase 2 (ROCK2) expression. In the MCAO rat and serum-free cell models, miR-335 treatment upregulated SG formation, alleviated the ischemia-induced infarction, and decreased ROCK2 protein expression and apoptosis levels. By contrast, when compared with miR-335 treatment, the inhibition of miR-335 resulted in reduced SG formation and higher ROCK2 expression and apoptosis levels. Target prediction analysis and luciferase 3′-untranslated region reporter assay identified ROCK2 as the direct target of miR-335. Furthermore, ROCK2 silencing enhanced SG formation and attenuated the level of apoptosis in the serum-free cell model. In addition, ROCK2 silencing markedly inhibited the effect of miR-335 on SG formation and apoptosis levels. Unexpectedly, the phosphorylation of T-cell intracellular antigen-1 was significantly inhibited by miR-335 in the MCAO rat model, which provides a reasonable explanation for the promotional effect of miR-335 on SG formation by specifically targeting ROCK2. In conclusion, these results demonstrate that miR-335 promotes SG formation and inhibits apoptosis by reducing ROCK2 expression in acute ischemic stroke, which provides a possible therapeutic target for brain injury.
