Cherries (subg. Cerasus) belong to Prunus s.l. (Rosaceae) and are widely distributed in temperate and subtropical regions of the Northern Hemisphere. Given its ornamental and edible functions, it is a hugely preferred germplasm resource for human societies. However, the interspecific phylogenetic relationships, divergence times, and routes of this subgenus remain unclear. To reveal its enigmatic and intricate evolutionary history, we newly sequenced 12 subg. Cerasus species, reconstructed the plastome phylogeny with 24 previously published subg. Cerasus species. This study supports that subg. Cerasus split into five clades, among which P. mahaleb is the basal group of subg. Cerasus ; P. campanulata is genetically distantly related to P. cerasoides and is not recommended for treatment as a variety of the latter; P. subhirtella is involved in the speciation of P. yedoensis. Molecular dating and ancestral area reconstructions infer that subg. Cerasus originated in the Eastern Mediterranean–Central Asia before 11.57 Mya (95% HPD = 5.34–19.1 Mya) and subsequently spread into the Qinghai-Tibetan Plateau (QTP) and adjacent regions. The first dispersal event of modern subg. Cerasus was facilitated by 'out-of-QTP', colonizing North America across the Bering Land Bridge and re-dispersing into Europe from the Late Miocene to Pliocene and forming the initial distribution pattern of extant subg. Cerasus in the Pliocene; The end of the Quaternary glacial period witnessed a second dispersal event, and core subg. In this paper, the habitat of Cerasus clarofolia was investigated and analysed, under natural conditions the growth of Cerasus clarofolia is influenced by low temperature of winter. The seeds of Cerasus clarofolia need low-temperature to germination. The seedling with 4 leaves grew well after introduction, the survival rate reached 79.7%. Planting with clear water was found easier to take roots, the roots were come from the lenticels, under diffent environmental conditions, the lenticels formed morphological, physiological differences are the important factors take roots for Cerasus.

Cerasus dielsiana (Rosaceae) is one of the most characteristic wild flowering cherry species endemic to subtropical China, with high conservation value from ornamental, economic, and ecological perspectives. To verify its spatial genetic structure and phylogeographic history, evaluation of nuclear (ITS) sequence variation in 203 individuals from 25 wild populations across its natural distribution ranges was conducted. High levels of ribotypes and nuclear diversity (Hd = 0.879 ± 0.012, π = 3.56 ± 0.16) were detected at the species level. Spatial analysis of molecular variation (SAMOVA), median-joining network and phylogenetic Neighbour-joining tree assigned the populations into four genetically distinct groups. Meanwhile, high levels of genetic differentiation among populations and strong phylogeographic patterns (Nst = 0.837 > Gst = 0.585; P < 0.05) were revealed based on the analysis of molecular variance (AMOVA). Results of neutrality tests, mismatch distribution analyses, Mantel test, barrier analyses, together with ecological niche modelling (ENM) simulation and BEAST analysis suggest a long-term demographic history (95% HPD: 3.64–8.83 Mya) of C. dielsiana, and at least three potential in situ glacial refugia as well as recent demographic expansion in eastern region have been detected. Collectively, we highlight that the combined effect of natural selection and random genetic drift under complex topography and palaeoclimatic oscillations since the Last Glacial Maximum (LGM) in subtropical China might have been the main driver to the species' allopatric differentiation. Prunus jamasakura is a species of Prunus native to eastern Asia. We determined the first complete chloroplast genome of Prunus jamasakura using genome skimming approach. The cp genome was 157,905 bp long, with a large single-copy region (LSC) of 85,910 bp and a small single-copy region (SSC) of 19,123 bp separated by a pair of inverted repeats (IRs) of 26,436 bp. It encodes 129 genes, including 84 protein-coding genes, 37 tRNA genes, and 8 ribosomal RNA genes. We also reconstructed the phylogeny of Prunus sensu lato using maximum likelihood (ML) method, including our data and previously reported cp genomes of related taxa. The phylogenetic analysis indicated that P. jamasakura is closely related with Prunus speciosa. [ABSTRACT FROM AUTHOR]
