Abstract Microsatellite‐stable colorectal cancer (MSS‐CRC) exhibits resistance to programmed cell death protein‐1 (PD‐1) therapy. Improving the infiltration and tumor recognition of cytotoxic T‐lymphocytes (CTLs) is a promising strategy, but it encounters huge challenges from drug delivery and mechanisms aspects. Here, a zeolitic imidazolate framework (ZIF) coated with apoptotic body membranes derived from MSS‐CRC cells is engineered for the co‐delivery of ginsenoside Rg1 (Rg1) and atractylenolide‐I (Att) to MSS‐CRC, named as Ab@Rg1/Att‐ZIF. This system is selectively engulfed by Ly‐6C+ monocytes during blood circulation and utilizes a “hitchhiking” mechanism to migrate toward the core of MSS‐CRC. Ab@Rg1/Att‐ZIF undergoes rapid disassembly in the tumor, released Rg1 promotes the processing and transportation of tumor antigens in dendritic cells (DCs), enhancing their maturation. Meanwhile, Att enhances the activity of the 26S proteasome complex in tumor cells, leading to increased expression of major histocompatibility complex class‐I (MHC‐I). These coordinated actions enhance the infiltration and recognition of CTLs in the center of MSS‐CRC, significantly improving the tumor inhibition of PD‐1 treatment from ≈5% to ≈69%. This innovative design, involving inflammation‐guided precise drug co‐delivery and a rational combination, achieves synergistic engineering of the tumor microenvironment, providing a novel strategy for successful PD‐1 treatment of MSS‐CRC.

Abstract Vortices in fluids and gases have piqued the human interest for centuries. Development of classical-wave physics and quantum mechanics highlighted wave vortices characterized by phase singularities and topological charges. In particular, vortex beams have found numerous applications in modern optics and other areas. Recently, optical spatiotemporal vortex states exhibiting the phase singularity both in space and time have been described. Here, we report the topologically robust generation of acoustic spatiotemporal vortex pulses. We utilize an acoustic meta-grating with broken mirror symmetry which exhibits a topological phase transition with a pair of phase singularities with opposite topological charges emerging in the momentum-frequency domain. We show that these vortices are topologically robust against structural perturbations of the meta-grating and can be employed for the generation of spatiotemporal vortex pulses. Our work paves the way for studies and applications of spatiotemporal structured waves in acoustics and other wave systems.

We propose and experimentally demonstrate the first example of Chern fractal insulators in an acoustic Sierpinski lattice. Through introducing fractality into the well-known Haldane model, we find that the topological phase diagram is significantly squeezed by about 0.5 times, compared with that of the original Haldane model. The energy spectra for different generations of a fractal lattice exhibits a hierarchy of self-similar energy bands in the form of a devil's staircase. With local acoustic measurements, we fabricate the sonic sample based on the theoretical model and experimentally confirm the fractality-induced squeezing and observe the one-way edge states that are protected by a robust mobility gap. Our work demonstrates the fundamental interplay between the fractality and topological Haldane insulator, and may provide new directions for the advanced control of sound waves.
