2. Developmental deficits, synapse and dendritic abnormalities in a Clcn4 KO autism mice model: endophenotypic target for ASD

Abstract Autism spectrum disorder (ASD) is linked to ion channel dysfunction, including chloride voltage-gated channel-4 (CLCN4). We generated Clcn4 knockout (KO) mice by deleting exon 5 of chromosome 7 in the C57BL/6 mice. Clcn4 KO exhibited reduced social interaction and increased repetitive behaviors assessed using three-chamber and marble burying tests. Surprisingly, these symptoms were improved by Risperidone treatment, a drug commonly used to treat ASD. RNA sequencing data from mouse neural progenitor cells (mNPCs) showed that the genes regulating trans-synaptic signaling, transmembrane transport, and neuronal projection development were significantly decreased in Clcn4 knockdown (KD) cells compared to wild type (WT). Moreover, Risperidone treatment increased the genes related to the ion transmembrane transport, membrane potential, and neuron projection development in Clcn4 KD. Abnormalities in synaptic plasticity and dendritic spine formation were also observed in Clcn4 KO compared to WT. We observed that phosphorylation of SYNAPSIN, PSD95, ERK and CREB, as well as the expression of CDK5, were reduced in the brains of Clcn4 KO mice. In Clcn4 KO cortical neurons, the phosphorylation of SYNAPSIN and PSD95 expressions also decreased compared to WT, indicating disrupted synaptic function. Additionally, Sholl analysis revealed a reduction in dendritic branching and neuronal projection length in both mouse and human CLCN4 KD neurons. Finally, the decreased phosphorylation of SYNAPSIN and expression of PSD95 along with dendrite abnormalities were restored after Risperidone treatment. These data suggest that dendritic outgrowth and synapse remodeling may serve as endophenotypic targets for drug efficacy in ASD.

Obesitas pada remaja sering dikaitkan dengan permasalah psikologis, seperti stres, anxiety, depresi bahkan trauma dan PTSD. Obesitas dapat menjadi salah satu faktor yang beresiko bagi perkembangan social anxiety siswa. Social anxiety merupakan salah satu jenis gangguan kecemasan yang bersifat khusus. Pada gangguan ini penderitanya akan merasakan kekhawatiran yang berlebihan ketika berada di sekitar orang banyak. Beberapa gejala fisik yang dapat diamati adalah munculnya keringat dan gemetar saat berinteraksi dengan orang banyak. Sehingga siswa dengan gangguan ini cenderung untuk menghindari pertemuan dan kegiatan yang melibatkan orang lain. Oleh karena itu penting bagi siswa yang mengalami obesitas untuk mendapatkan bantuan psikologis yang sesuai. Salah satu bantuan yang dapat diberikan adalah dengan menggunakan teknik Ifdil Perceptual Light Technique (IPLT). Teknik IPLT merupakan sebuah alternatif pendekatan brief untuk mereduksi berbagai macam permasalahan psikologis, salah satunya kecemasan. Intervensi menggunakan IPLT dilakukan melalui modifikasi sensorik dan persepsi individu melalui spektrum cahaya. IPLT menggunakan kombinasi teknologi, bahasa pemrograman, dan beberapa pendekatan sebelumnya, yang bertujuan untuk mengurangi stes, kecemasan, trauma, fobia dan gangguan psikologis lainnya dalam waktu yang relatif singkat yaitu kurang dari sepuluh menit Penelitian ini bertujuan untuk menguji IPLT dalam mengurangi social anxiety siswa obesitas. Penelitian ini menggunakan desain subjek tunggal terhadap satu subjek yang mengalami soial anxiety.Desain penelitian menggunakan desain A-B single subject penelitian. Instrumen yang digunakan adalah observasi, wawancara, teknik scaling dan menggunakan instrumen social anxiety. Hasil penelitian menunjukkan bahwa responden memiliki tingkat social anxiety tinggi sebelum intervensi. Namun setelah diberikan intervensi melalui IPLT tingkat social anxiety menurun. Hasilnya menunjukkan bahwa IPLT efektif untuk mengurangi social anxiety pada siswa obesitas.

Understanding history is more complex than just understanding past events. One way that can be done to foster an understanding of history is by developing historical literacy skills. Students' historical literacy skills can be strengthened by utilizing applications in the history learning process integrated with digital technology. This study aims to explain the utilization or potential of electronic modules based on the heyzine flipbook application in developing students' historical literacy skills in history learning. By using the integrative review type literature review method, researchers describe the use of electronic modules based on the heyzine flipbook application through a number of features contained in the application, which consist of link features, videos, images, audio, and so on. In the heyzine flipbook application, there are various features that make it easy for users to develop interactive teaching materials. These features are in the interactions column which focuses on editing the module to be created. The features are link, image, video, audio and web features. Through this article, the researcher aims to provide a solution for utilizing electronic modules based on the heyzine flipbook application as one of the media that can be utilized in the history learning process so that history is not considered a field of study that emphasizes the memorization process alone and is able to develop students' historical literacy skills.

Background: Mutations in the RAS/MAPK pathway are a frequent driver of cancer, with oncogenic RAS or RAF mutations occurring in >30% of all cancers. First generation BRAF inhibitors are approved for use for tumors with Class I BRAF mutations (V600X). However, these drugs are not efficacious in RAF dimer mutant and RAS mutant cancers due to paradoxical activation of RAF dimers. Herein, we describe DCC-3084, a potent and selective investigational Switch Control inhibitor of BRAF and CRAF kinase dimers that targets Class I, II and III BRAF mutations, BRAF fusions, and BRAF/CRAF heterodimers. DCC-3084 combines with inhibitors of additional nodes in the MAPK pathway to potentially target a large unmet medical need in RAS and RAF mutant cancers. Methods: Inhibition of RAF kinases, including off-rate analysis, was measured using recombinant enzymes. X-ray crystallography was used for structure-based drug design. Cellular proliferation was measured using resazurin to monitor cell viability. Synergy in cells was measured using BLISS scores and curve shift analysis. Inhibition of ERK or RSK phosphorylation was measured by AlphaLISA or ELISA. Pharmacokinetics (PK) in the plasma, brain and CSF compartments were measured following oral dosing in Wistar rats. RAF and RAS mutant mouse xenograft models were used to assess PK, pharmacodynamics (PD), and efficacy. Results: DCC-3084 is a potent and selective Switch Control inhibitor of RAF dimers that was designed to target Class I, II, III BRAF mutants, BRAF fusions, and BRAF/CRAF heterodimers. DCC-3084 inhibits BRAF and CRAF, exhibiting slow off-rates (t1/2 >20 hr). Potent single-agent inhibition of MAPK pathway signaling and cellular proliferation was observed in a wide range of Class I, II, III BRAF and BRAF fusion altered cell lines. Synergy was observed in combination with inhibitors of other nodes in the RAS/MAPK pathway in RAS mutant cell lines. DCC-3084 was demonstrated to be CNS penetrable and exhibited dose dependent oral exposure with robust inhibition of the RAS/MAPK pathway in PK/PD models. DCC-3084 accumulated in tumor tissue relative to plasma, further demonstrating a favorable pharmaceutical profile. Oral treatment of DCC-3084 as a single agent resulted in tumor regression in BRAF mutant and KRAS Q61K mutant mouse xenograft models and tumor growth inhibition in KRAS G12C/D mutant models. Additionally, DCC-3084 in combination with a MEKi resulted in tumor regression in KRAS mutant models. Conclusions: The Switch Control inhibitor DCC-3084 broadly inhibits Class I, II and III BRAF mutations, BRAF fusions, and BRAF/CRAF heterodimers leading to tumor regression in preclinical models. The overall preclinical profile of DCC-3084 supports IND-enabling activities towards clinical development in a key area of unmet medical need in RAS and RAF mutant cancers. Citation Format: Stacie L. Introduction: Gastrointestinal stromal tumors (GISTs) are typically driven by primary mutations in KIT exons 9 or 11. Heterogeneous drug-resistant secondary mutations arise in patients treated with FDA approved KIT inhibitors, including imatinib and sunitinib. Drug resistant secondary mutations are found at multiple regions in the ATP pocket (encoded by exons 13 and 14) or activation switch (encoded by exons 17 and 18) of KIT kinase. In addition, multiple drug-resistant clones can arise within a tumor or in metastatic tumor sites. An inhibitor that can broadly and potently inhibit the spectrum of KIT mutations is highly sought. Ripretinib has been FDA approved as a 4th line treatment for GIST and has broad activity against KIT mutations, including clinical potency in patients with mutations in KIT exons 11, 17, or 18. DCC-3009 was designed as a next generation KIT inhibitor that broadly and potently inhibits primary KIT mutations in exons 9 and 11 and secondary drug-resistant mutations across exons 13, 14, 17, and 18. DCC-3009 is a potent and selective inhibitor in enzyme and cell-based assays, and has demonstrated efficacy in xenograft models driven by drug resistant KIT mutations. Methods: DCC-3009 was tested for inhibition of KIT mutants using standard enzyme and cell-based assays. Levels of phosphorylated KIT were determined by Western blot or ELISA. Proliferation was measured using the fluorescent dye resazurin. KIT mutant xenograft or patient-derived xenograft models were performed at Crown Biosciences or Labcorp, AAALAC accredited facilities, with the approval of Animal Care and Use Committees. Results: In BaF3 cells transfected with KIT mutants, DCC-3009 was shown to potently inhibit the spectrum of known primary and secondary drug-resistant mutations in GIST. The pan-mutant KIT profile of DCC-3009 was shown in vitro to be superior to 2nd and 3rd line standard of care therapies sunitinib and regorafenib. DCC-3009 was selective for KIT when screened against a large panel of kinases. DCC-3009 has optimized pharmaceutical properties for oral administration. In pharmacokinetic/pharmacodynamic studies DCC-3009 achieved sufficient free drug levels to significantly inhibit drug-resistant KIT mutants for 12 hr post dose. In xenograft studies, treatment with DCC-3009 twice daily led to tumor regression in drug-resistant models with KIT exon 9/13, 11/13 or 11/17 mutations. Conclusions: DCC-3009 is a pan-exon mutant KIT inhibitor exhibiting high potency in KIT mutants in pre-clinical models spanning exons 9, 11, 13, 14, 17 and 18. In vivo, DCC-3009 exhibited efficacy in drug-resistant models with KIT exon 9/13, 11/13 or 11/17 mutations. Based on this profile, DCC-3009 has entered formal preclinical development. 9. The inhibition of colon cancer development by black rice bran on BALB/C Mice

Black rice bran (BRB) is well-known for its high antioxidant activity and its pivotal role in preventing colon cancer. The present study aims to investigate the effects of BRB administration on BALB/C mice induced with azoxymethane (AOM) and dextran sodium sulphate (DSS). The 24 mice were divided into three groups: the group induced by colon cancer (C+), the group induced by cancer and given the BRB diet (C+BRB), and the normal group (C−). Both the C− and C+ groups were given a standard AIN-93 M diet containing cellulose fiber. After 16 weeks, the mice were anesthetized, and the colonic tissue was identified for nodule distribution, histopathological observation, and mRNA expression analysis of proliferating cell nuclear antigen (PCNA), caspase-3, and caspase-8 genes using qRT PCR technique. Nodule distribution in the C+BRB group showed a significant decrease compared to the C+ group, with 1.65 ± 0.71 nodule/cm2 vs. 5.73 ± 2.93 nodule/cm2. Then, the colon weight was significantly decreased in the C+BRB group, at 0.19 ± 0.04 (g) compared to 0.25 ± 0.03 (g) in the C+ group. Also, the BRB diet in the C+BRB group significantly decreased PCNA mRNA expression compared to the C+ group, with values of 0.58 ± 0.09-fold change vs. 5.22 ± 0.80-fold change. Conversely, increased the mRNA expression of caspase-3 (0.91 ± 0.20-fold change vs. 0.36 ± 0.15-fold change) and caspase-8 (0.51 ± 0.18-fold change vs. 0.13 ± 0.31-fold change). In conclusion, administration of BRB inhibited the rate of cancer development by suppressing cancer cell proliferation and inducing apoptosis.

UMKM merupakan salah satu sektor pendukung perekonomian di Indonesia. UMKM Batik Madura memberikan kontribusi yang cukup tinggi terhadap penyerapan tanaga kerja dan peningkatan ekonomi masyarakat daerah. Hal ini terbukti pada penyerapan tenaga kerja UMKM di Kabupaten Bangkalan Madura sebesar 210.003 dan Sampang sebesar 264.569. Permasalahan penelitian ini berkaitan dengan banyaknya UMKM Madura, sehingga menyulitkan Dinas Koperasi dalam menjalankan kebijakan pemerintah dalam memberikan pelatihan, bantuan pengembangan UMKM dan pendampingan. Tujuan penelitian adalah mengelompokan UMKM Batik Madura menjadi beberapa kluster menggunakan metode K-Means dan Sillhoutte Coefficient. Metode K-Means dapat melakukan pengelompokan berdasarkan data yang sama atau mempunyai similarity yang tinggi. Data UMKM akan dilakukan preprosesing terlebih dahulu untuk mengatasi data yang kosong dan normalisasi. Metode Sillhoutte Coefficient (SC) digunakan untuk menentukan jumlah kluster yang paling optimal. Pengelompokan UMKM Batik ini berdasarkan perpektif balance scorecard yaitu bisnis internal, keuangan, learning and growth dan pelanggan. Hasil cluster yang paling optimal adalah K=3. Nilai SC adalah sebesar 0,275, dengan 9 fitur dan SC = 0,403 dengan 5 fitur, artinya dengan metode seleksi fitur information gain terjadi peningkatan snilai SC sebesar 0,128. Prosentase hasil pengelompokan adalah cluster 1 sebesar 15 %, cluster 2: 25 % dan cluster 3 : 60 %. Kategori pemetaan cluster 1 adalah sangat baik, cluster 2 baik dan cluster 3 sedang.

The background of this research is that Science in Indonesia develops through institutions, not ideas, people, or instruments. This research aims to describe the institutionalization of scientific institutions in the Dutch East Indies. The historical method analyzes various resources to aid the reconstruction of the Dutch East Indies’s science institutionalization process. This research uses archives from ANRI (National Archives of the Republic of Indonesia) regarding Bataviaasch Genootschap van Wetenschappen (The Batavia Society of Arts and Science). Archives from other scientific institutions, such as Bogor and Cibodas Botanical Gardens, are also used to illustrate the institutionalization process. The findings of this research include studies on the process and the necessity to pay attention to elements such as rules, norms, cultural benefits, roles, and material resources. Institutionalizing science and technology in the Dutch East Indies underwent at least three stages: the emergence of amateur scientists from Europe, the efforts to build scientific groups, and the establishment of science institutions. This research concludes that the first two stages were outside the bureaucratic channels of the colonial government. However, in the third stage, scientists needed help to escape the role of the colonial bureaucracy in developing these scientific institutions. Bataviaasch Genootschap van Wetenschappen, founded in 1778, was a private organization studying commerce, agriculture, and humanities. It played a significant role in encouraging various other research institutions

Permasalahan secara umum terjadi pada anak-anak remaja, menjadi sebuah karakter yang perlu dikembangkan melalui lembaga pendidikan sekolah. Pengembangan yang berfokus kepada karakter profil pelajar pancasila elemen kreatifitas dan bergotong royong, harapannya peserta didik menjadi manusia berkarakter, sehingga bisa menjadi manusia beradab. Kenyataannya kendala pemahaman dalam mengembangkannya baik dari guru dan juga peserta didik, serta peran dari orang tua juga menentukan karakternya. Project based learning menuntut dalam penerapannya membuat suatu proyek yang harus dikerjakan, kemudian menghasilkan karya tulisan, gambar dll. Tujuan dalam penelitian ini yaitu memperoleh deskripsi terkait Project based learning untuk mengembangkan karakter profil pelajar pancasila elemen kreatifitas dan bergotong royong pada pembelajaran sejarah. Desain penelitian didalam penelitian ini yaitu studi literatur, yaitu sumbernya dari berbagai karya tulis artikel yang ada kaitannya dengan tema pembahasan. Project based learning merupakan metode belajar mengajar sebagai solusi untuk diterapkan, dan bisa untuk mengembangkan karakter para peserta didik agar mempunyai kualitas moral yang positif. Melalui Project based learning bisa mengembangkan karakter profil pelajar pancasila diambil dari beberapa elemen misalnya elemen kreatifitas dan bergotong royong di mata pembelajaran sejarah yang bisa untuk mengajarkan sejarah dengan benar, artinya bahwa guru mampu memilih tema permasalahan dan mampu memilih untuk menerapkan metode serta pendekatan pembelajaran kontekstual agar mencapai tujuan dengan menghubungkan materi di buku teks dengan lingkungan sekitarnya.

Modeling and linguistic representation in the form Interval Type-2 Fuzzy have better accuracy than Type-1 Fuzzy. The type-2 fuzzy set involves more uncertainty than the type-1 fuzzy set. The degree of fuzzy membership is used to explain uncertainty and ambiguity in the real world. This study presents the type-2 Fuzzy Analytic Network Process (ANP) method to determine the weight of each attribute based on the level of interest and the extension method of type-2 Fuzzy TOPSIS to handle problems based on the value of the fuzzy type-2 attribute. Decision-making is based on the assessment of several experts called Multi-Criteria Group Decision Making (MCGDM), using type-2 Fuzzy geometric mean aggregation function. The membership function in this research is type-2 fuzzy based on the trapezoid. The contribution is a hybrid method Type-2 Fuzzy TOPSIS with Fuzzy Type-2 ANP group-based with new metric intervals on fuzzy type-2 for decision making. The results are a hybrid type-2 FANP and FTOPSIS decision-making model to support the best decision-making. Based on a comparison of the accuracy of trapezoid model 1, model 2, and model 3, the best accuracy result is model 3, which is 84%. The research benefits by presenting a hybrid Type-2 Fuzzy TOPSIS and ANP method that improves decision-making accuracy and better handling uncertainty and ambiguity than Type-1 Fuzzy systems.

EdLink merupakan salah satu platform mobile berbasis Android yang telah dirancang khusus untuk mendukung pengalaman belajar di dunia pendidikan. Platform ini menawarkan berbagai fitur konten pembelajaran interaktif, tugas online, dan diskusi. EdLink telah menjadi salah satu aplikasi yang banyak diminati dan digunakan di berbagai instansi pendidikan, termasuk perguruan tinggi. Aplikasi EdLink saat ini memiliki rating 3,7 di Google Play Store. Banyak pengguna mengeluhkan berbagai aspek, seperti fitur yang kurang lengkap, pelayanan, dan kinerja. Dalam penelitian ini, digunakan analisis sentimen berbasis aspek untuk mengevaluasi aplikasi. Data ulasan yang digunakan adalah seluruh ulasan aplikasi EdLink di Google Play Store dari versi 1.1.6 hingga 4.7.8, dengan jumlah 2014 ulasan. Penelitian ini dilakukan dengan pemodelan topik Latent Dirichlet Allocation (LDA) dengan tujuan untuk menentukan aspek serta analisis sentimen menggunakan Support Vector Machine (SVM) dengan pendekatan Lexicon Based. Penelitian ini menghasilkan tiga aspek utama, yaitu Application Usability, Reliability, dan Performance Efficiency. Berdasarkan model LDA menghasilkan skor koherensi tertinggi 0,487 dan berdasarkan distribusi jumlah topik dan skor koherensi tertinggi dari num topic 1-10 adalah 3. Kemudian, labeling Lexicon Based menghasilkan 418 jumlah ulasan positif dan 1.223 ulasan negatif. Selanjutnya, klasifikasi SVM dengan rasio pembagian data 90:10 menghasilkan akurasi tertinggi 85,45% kemudian dilakukan resampling dengan hasil akurasi tertinggi 90,00% menggunakan SMOTE. Berdasarkan aspek dan sentimen dihasilkan 319 ulasan negatif dan 127 ulasan positif untuk aspek Usability, 482 ulasan negatif, dan 120 ulasan positif untuk aspek Reliability, serta 422 ulasan negatif dan 171 ulasan positif untuk aspek Performance Efficiency.

Membrane-associated intercellular adhesion molecule-1 (mICAM-1; CD54) is constitutively expressed on the surface of type I alveolar epithelial cells (AEC). Soluble ICAM-1 (sICAM-1) may be produced by proteolytic cleavage of mICAM-1 or by alternative splicing of ICAM-1 mRNA. In contrast to inducible expression seen in most cell types, sICAM-1 is constitutively released by type I AEC and is present in normal alveolar lining fluid. Therefore, we compared the mechanism of sICAM-1 production in primary cultures of two closely juxtaposed cells in the alveolar wall, AEC and pulmonary microvascular endothelial cells (PVEC). AEC, but not PVEC, demonstrated high-level baseline expression of sICAM-1. Stimulation of AEC with TNF[alpha] or LPS resulted in minimal increase in AEC sICAM-1, whereas PVEC sICAM-1 was briskly induced in response to these signals. AEC sICAM-1 shedding was significantly reduced by treatment with a serine protease inhibitor, but not by cysteine, metalloprotease, or aspartic protease inhibitors. In contrast, none of these inhibitors effected sICAM-1 expression in PVEC. RT-PCR, followed by gel analysis of total RNA, suggests that alternatively spliced fragments are present in both cell types. However, a 16-mer oligopeptide corresponding to the juxtamembrane region of mICAM-1 completely abrogated sICAM-1 shedding in AEC but reduced stimulated PVEC sICAM-1 release by only 20%. Based on these data, we conclude that the predominant mechanism of sICAM-1 production likely differs in the two cell types from opposite sides of the alveolar wall. regulation; mouse; cell culture; CD54

Purpose of this study was to identify barriers and challenges associated with reading and writing learning as well as the process of learning to read and write through mother tongue. Qualitative method descriptive instruments used interviews, observation, documentation. The results of the study, the first step in learning to read the mother tongue is to introduce letters of the alphabet, string words, read aloud, read sentences, read self-identity, and distinguish letters of the alphabet. Write alphabets, words, sentences, write self-identity, write the difference between capital and lowercase letters. Through asking and answering and interacting with friends, students understand what the teacher teaches, difficulty understanding letters, intellectual and environmental factors.

Reflection technique is one of the tools used by counselors in understanding the characteristics and conditions of clients. In this case the reflection technique is used to help problems related to self-awareness. The level of self-awareness of each individual varies, ranging from high levels to low levels. But generally what we most often encounter are individuals with low levels of self-awareness and this can also be related to the sense of responsibility of the individual himself, thus if the level of self-awareness is low then the sense of responsibility he has is also low. For this reason, the reflection technique was chosen by the author in understanding the client's characteristics. This reflection technique is one of the counseling techniques that involves thoughts, feelings and experiences for that it will be easier for researchers to get results. If the researcher is able to understand what the client is feeling, a warm relationship will also be established between the parties involved. Clients will feel comfortable to be invited for interviews and discussions of what is going on in him now. That is the importance of the counselor and client to establish a close relationship and even friendship with one another.

Ekspor minyak kelapa sawit merupakan salah satu komoditas utama dalam struktur ekspor Indonesia. Produksi minyak sawit Indonesia meningkat pesat dari 721.127 ton pada tahun 1996 menjadi 49.710.345 ton pada tahun 2021. Ekspor minyak sawit juga meningkat dari USD 825.415 pada tahun 1996 menjadi USD 26.665.128 pada tahun 2021. Penelitian ini menganlisis pola perdagangan minyak kelapa sawit Indonesia dan daya saing ekspor minyak kelapa sawit Indonesia di dunia. Intra Industri Trade Index (IIT) digunakan sebagai indikator untuk menganlisis pola perdagangan minyak kelapa sawit Indonesia terhadap 10 negara utama mitra dagang Indonesia, khususnya komoditi HS 1511 dan turunannya kode HS 151110. Daya saing minyak kelapa sawit Indonesia dianalisis dengan Reveal Symmetric Comparative Advantage (RSCA) dan Trade Intensity Index (TII). Hasil penelitian menunjukkan bahwa pola perdagangan minyak kelapa sawit Indonesia tidak terintegrasi di antara 10 mitra dagang Indonesia. Hal ini menunjukkan bahwa Indonesia hanya mengekspor dan tidak mengimpor komoditi HS 1511 dan 151110 dari 10 negara tersebut. Indonesia memiliki keunggulan komperatif dalam ekspor minyak kelapa sawit dunia. Mengingat pentingnya Indonesia dalam perdagangan dunia, indeks intensitas perdagangan menunjukkan bahwa arus perdagangan bilateral Indonesia ke 10 negara lebih besar dari yang diharapkan.

This study aims to examine bullying tendencies among students in Indonesia. A sample of 689 male and female students was selected using a simple random sampling technique. Data were collected through the Bullying Behavior Tendency Scale (BBTS), which consisted of 45 questions using a Likert scale, and were analyzed descriptively. The results showed that the bullying tendencies were in a low category. However, it was discovered that 32.08, 4.06, and 0.87% of students were in the moderate, high, and very high categories, respectively. Additionally, it was observed that there are significant differences in bullying behavior between male and female students. These results provide a basis for understanding the role of school counseling services in reducing bullying behavior which is prevalent and can cause trauma. Therefore, it is essential to implement preventive programs to address the tendencies of some students engaging in this act at school.

The purpose of this study examines how the integration of the value of happiness in Al Farabi's perspective is created through the process of Islamic counseling so that a good quality of life can be achieved. Al Farabi defines a happy quality of life obtained through a correct understanding of the purpose of a life lived, namely eternal happiness in the hereafter by showing good moral behavior. Islamic counseling is an effort to help humans achieve happiness in life. The research method is in the form of a literature study through content analysis in the literature that is relevant to the research objectives. The results of the study found that the stages of the Islamic counseling process that can be passed to help achieve the value of happiness from Al Farabi's perspective are the stages of understanding the meaning of the value of happiness from clients, identifying problems that interfere with the client's life happiness, the stage of developing solutions or the stage of coaching clients, and the evaluation stage. This stage is supported by techniques for understanding the meaning of life, techniques for dealing with stress and anxiety in life, techniques for maintaining the quality of social relations because humans are social beings, and techniques for hoping and praying that every problem has a solution following Islamic teachings and spirituality techniques.

Electronic wallet atau e-wallet adalah sebuah layanan virtual yang memungkinkan penggunanya menyimpan dana dalam betuk uang digital untuk dapat digunakan sebagai pengganti pembayaran secara cash. Layanan ini sangat mengandalkan jumlah pengguna dan keaktifan mereka dalam memanfaatkan fitur transaksi online. Usability yang baik adalah adalah satu kunci dalam mendapatkan pengguna baru atau mempertahankan pengguna lama untuk selalu menggunakan e-wallet sebagai pilihan utama dalam transaksi online. Oleh karena itu, penelitian ini bertujuan untuk melakukan pengukuran kualitas layanan 5 e-wallet yang populer di Indonesia (DANA, OVO, GOPAY, SHOPEEPAY dan LINKAJA) dari aspek usability. Penelitian ini menggunakan model PACMAD untuk menentukan tingkat usability aplikasi e-wallet yang terdiri dari 7 kriteria pengukuran antara lain: effectiveness, efficiency, satisfaction, learnability, memorability, errors dan cognitive load. Dengan menggunakan Fuzzy-AHP, masing-masing kriteria tersebut kemudian ditententukan bobotnya berdasarkan pendapat para ahli di bidang usability. Untuk mengukur nilai usability, sejumlah 50 pengguna e-wallet diundang sebagai responden pada penelitian ini. Data hasil penilaian responden selanjutnya dihitung dengan sebuah aplikasi simulasi Multi Criteria Decision Making (MCDM) yang sebelumnya telah dibangun pada penelitian ini dengan menggunakan algoritma TOPSIS. Berdasarkan hasil ujicoba yang dilakukan, penelitian ini menunjukkan bahwa aspek/indikator usability yang paling berpengaruh pada sebuah aplikasi e-wallet adalah satisfaction dengan bobot 0.214 berdasarkan proses pengambilan keputusan dengan metode Fuzzy-AHP yang menggunakan 3 pakar dengan penentuan konsistensi rasio kurang dari sama dengan 0.1 atau 10%. Berdasarkan perhitungan TOPSIS, OVO memiliki nilai usability terbaik dengan nilai preferensi sebesar 0.765816.

One of them is a marketing firm that uses a green marketing strategy. This will almost probably alter purchasers' motivation to consume a product repeatedly. The goal of this study is to learn about and investigate the concurrent and partial effects of green marketing, which comprises green product, green price, green promotion, and green location, on repurchasing items from the Avoskin brand in Surabaya. 100 respondents were sampled. Multiple linear regression analysis was utilized to process this data, which was supported by the SPSS 25.0 software. The study's findings indicate that green marketing has a favorable and significant impact on the simultaneous repurchase of Avoskin products in Surabaya. The results of this study are transference is a projection of feelings from the client to the counselor, this projection of feelings occurs because of the blurring of ethical standards or boundaries set by the counselor in the counseling process, for that we need several strategies to overcome them.

This research uses a type of research and development using the ADDIE procedural model. The module material was obtained from an analysis of students' cyberbullying behaviour needs; then, a module feasibility test was carried out by 2 material experts and 2 media experts. Usability test by 3 guidance and counselling teachers. The methods used to collect data are observation, interviews and questionnaires. The instrument used to collect data was a questionnaire. The techniques used to analyze the data are descriptive analysis and non-parametric analysis. The results of this research are that the digital cognitive behaviour modification module to reduce cyberbullying behaviour is in the very appropriate category from experts and has received results. The practicality test results of the digital module are in a very good class. It was concluded that the digital module prepared was suitable for implementation or use by guidance and counselling teachers and counsellors. The implication of this research is that the digital module developed helps counselors and guidance and counseling teachers in providing services to their clients.

Jagung (Zea mays ssp. mays) adalah tanaman pangan ketiga terbesar setelah gandum dan beras, dan di Indonesia menempati posisi kedua setelah padi. Jagung dapat ditanam di daerah dengan suhu tinggi dan rendah serta curah hujan dan irigasi yang cukup. Namun jagung sangat rentan terhadap penyakit selama siklus hidupnya, yang dapat menurunkan kualitas dan kuantitasnya. Di Sumenep, Jagung dapat dikatakan sebagai bahan pangan pokok untuk sebagian masyarakat pedesaan atau pelosok. Penyebab terjadinya serangan pada tanaman jagung adalah ketidaktahuan petani dalam pencegahan dan penanganannya sehingga menyebabkan produksi jagung mengalami penurunan. Dinas pertanian kabupaten Sumenep juga belum mempunyai sistem untuk klasifikasi hama dan penyakit jagung. Tujuan penelitian ini adalah klasifikasi penyakit dan hama tanaman jagung menggunakan metode naïve bayes dengan information gain. Naïve Bayes digunakan untuk mengolah nilai-nilai probabilitas setiap gejala, dan nilai persentase dari setiap hama dan penyakit. Information Gain untuk menyeleksi bobot gejala yang paling berpengaruh dalam menentukan hama dan penyakit jagung. Hasil uji coba, akurasi naïve bayes dengan information gain dapat meningkatkan akurasi rata-rata sebesar 3,17 % dibanding klasifikasi tanpa seleksi fitur. Akurasi terbaik diperoleh dengan metode information gain dan naïve bayes sebanyak 15 fitur dari 47 fitur dengan akurasi sebesar 98,47 %. Penelitian ini merekomendasikan 15 fitur, dengan 3 fitur terbesar adalah tidak berbuah, daun berklorosis sebagian atau seluruh daun dan adanya bekas gigitan pada batang.

The operational efficacy of lane departure warning systems (LDWS) in autonomous vehicles is critically influenced by the retro-reflectivity of road markings, which varies with environmental wear and weather conditions. This study investigated how changes in road marking retro-reflectivity, due to factors such as weather and physical wear, impact the performance of LDWS. The study was conducted at the Yeoncheon SOC Demonstration Research Center, where various weather scenarios, including rainfall and transitions between day and night lighting, were simulated. We applied controlled wear to white, yellow, and blue road markings and measured their retro-reflectivity at multiple stages of degradation. Our methods included rigorous testing of the LDWS’s recognition rates under these diverse environmental conditions. Our results showed that higher retro-reflectivity levels significantly improve the detection capability of LDWS, particularly in adverse weather conditions. Additionally, the study led to the development of a simulation framework for analyzing the cost-effectiveness of road marking maintenance strategies. This framework aims to align maintenance costs with the safety requirements of autonomous vehicles. The findings highlight the need for revising current road marking guidelines to accommodate the advanced sensor-based needs of autonomous driving systems. By enhancing retro-reflectivity standards, the study suggests a path towards optimizing road safety in the age of autonomous vehicles.

The development of the plantation sub-sector with a marketing mix strategy approach is a comprehensive and sustainable solution for the development of human and economic resources in rural areas. Marketing performance is used to determine the level of marketing success. The purpose of this study was to analyze the effect of marketing mix strategy consisting of differentiation of agricultural products, functionalization of elasticity of agricultural products, added value and resonance on the marketing performance of smallholder rubber farmers in Gunung Toar District, Kuantan Singingi Regency. The population in this study were smallholder rubber farmers with a sample of 34 farmers who were determined by the simple random sampling method. The data analysis method used is multiple linear regression using SPSS software. The results showed that the marketing mix strategy consisting of agricultural product differentiation (X1) and agricultural product elasticity functionalization (X2) had a significant effect, and added value (X3) and resonance (X4) had no significant effect on marketing performance. smallholder rubber farmers in Kab. In the medical world, diagnosis means finding a hypothesis from the observed symptoms, not much different from that understanding, diagnosis in counseling aims to make a temporary hypothesis about the problems experienced by the client. This can be observed through the behavior, expressions, client explanations of the problems experienced. With the hypothesis, the counselor can more easily explore the client so as to find the right technique/method in alleviating the client's problems and directing the client's independence

This research aims to identify the effects of the administration of a black rice bran diet on colorectal cancer in dextran sodium sulfate and azoxymethane-induced BALB/c mice. The research was conducted on three groups consisting of eight Balb/c mice: two groups were fed with carcinogens, and the third group, referred to as the normal group, was supplied with Isotonic NaCl 0.9% intraperitoneally. One group fed with carcinogens was supplied a standard AIN 1993 M diet modified with black rice bran as a substitute of fibre source, while the other two mice groups were fed the standard diet (AIN-93M) containing cellulose fibre. At the 17th week, all mice were euthanized; their colonic sections were taken for histopathological evaluation, and cecum for short-chain fatty acids concentration, total lactic acid bacteria, pH and β-glucuronidase activity evaluations. The results show an increase in the total lactic acid bacteria and short-chain fatty acids in the mice group fed with rice bran. Consequently, pH value and β-glucuronidase activity had decreased. Histopathological evaluation of mucosal tissue exhibited inhibition of the tumor growth rate in the mice groups fed rice bran compared to the group supplied with the standard diet. Furthermore, the proliferating cell nuclear antigen expression had decreased significantly, while expression of caspase-8 and caspase-3 had increased notably, in the group fed with a rice bran diet. These results suggest that black rice bran can effectively inhibit colon carcinogenesis. The potential of black rice bran as a source of fibre has not been studied in detail regarding the inhibition mechanism of colorectal cancer cells; further investigation in this field could provide valuable information about new strategies to prevent colorectal cancer. This strand of research is very important to developing preventive methods against cancer and promoting the concept of healthy products, including functional foods.

The success of customer value can affect the marketing performance of rubber. The research was conducted in Kuantan Mudik District, Kuantan Singingi Regency, Riau. The purpose of this study was to determine the effect of customer value on emotional value, social value, quality value, and price value on smallholder rubber farmers. The population in this study were 205 farmers, the sample was taken 15% of the population of 31 farmers selected by simple random sampling technique. Analysis of the data used is multiple linear regression analysis with SPSS application. The results of the study explain sig.t value for X1 is 0.003, sig.t value of X2 is 0.014, sig. t value of X3 is 0.021 which is smaller than the 0.05 level of significance. This means that X1, X2, and X3 partially have a significant effect marketing performance. While sig t value of X4 is 0.216 which is greater than the significant level, means price value partially has no significant effect marketing performance. f-test results sig.f value of 0.000 is smaller than the significant level of 0.05, meaning that the customer value seen from the emotional, social, quality, and price values simultaneously has a significant effect on increasing marketing performance.

This study aims to analyze the role of customer value as an intervening variable/mediation of the influence of marketing strategy on the marketing performance of smallholder rubber farmers in Sentajo Raya District, Kuantan Singingi Regency. Sampling in this study was 15% of the population as many as 36 smallholder rubber farmer respondents with each sample must be taken proportionally according to the population in two villages, namely Jalur Patah Village and Parit Teratak Air Hitam Village in Sentajo Raya District. The sampling technique was done by random sampling. The data analysis tool used in this research is SmartPLS 3.0 Sobel test and VAF calculation. The results of this study indicate that the customer value variable cannot mediate the relationship between market strategy and marketing performance. While the customer value variable can mediate the relationship of marketing mix strategy to marketing performance where the role of the mediating effect of customer value is partial mediation of 0.540 or 54.0%.

Metode yang digunakan adalah kegiatan pelatihan yang mencakup penyuluhan tentang jenis dan potensi protein nabati termasuk tempe kedelai dan jamur merang serta pelatihan pembuatan sosis analog (tiruan) dengan bahan baku tempe dan jamur merang. This study aims to analyze the effect of market strategy on marketing performance in smallholder rubber in Kuantan Tengah District, Kuntang Singingi Regency, Riau. These market strategies include market shaping strategies, customer engagement strategies, and technology leadership strategies. The population in this study found 225 people from 1 Gapoktan and 9 farmer groups in Kuantan Tengah District. The research sample was 15% of the population, namely 34 people who were selected by random sampling method. Data analysis used descriptive analysis and multiple linear regression models. The results of the analysis show that the respondent's level of achievement reaches 80,82% for market strategy and 74,22% for marketing performance, both of which are in the good level category. The value of sig.t for X1 is 0.861, the value of sig.t for X2 is 0,342, the value of sig.t for X3 is 0,585 is greater than the significance level (α), then the formation of market strategy, customer involvement, and technology leadership partially have no significant effect. significant to marketing performance. The sig.F value of 0,728 is higher than the significance level (α), so the formation of market strategy, customer involvement, and technology leadership simultaneously do not have a significant effect on marketing performance.

The high number of stunting children in Indonesia is still a severe problem that can impact the quality of human resources. The foster grandparent's program is one of the ways that servants do to overcome these problems. This service activity aims to increase the productive community's knowledge about stunting and improve the nutrition of children under five in Sengon Village. This service is aimed at 110 respondents who participate in Posbindu in Sengon Village, Bendungan District, Trenggalek Regency, as a foster grandparents program. The activity begins with providing education about stunting to increase public knowledge. The instruments used were leaflets, booklets about stunting and power points delivered using an LCD and projector screen. After education is given, the knowledge of the elderly will be measured again. Furthermore, the elderly will be allowed to apply their knowledge to educate their families and surroundings who have children under five with poor nutritional status and stunting. The toddler's weight will be measured again one month later. The data will be analyzed using the Wilcoxon Sign Rank Test. The service results showed that the foster grandparent's program affected the knowledge of the elderly about stunting and under-five nutrition with p=0.00. So it is recommended that the foster grandparent's program continue to be carried out so that the nutrition of children under five is increased and the number of stunting children under five decreases.

Membrane-associated intercellular adhesion molecule-1 (mICAM-1; CD54) is constitutively expressed on the surface of type I alveolar epithelial cells (AEC). Soluble ICAM-1 (sICAM-1) may be produced by proteolytic cleavage of mICAM-1 or by alternative splicing of ICAM-1 mRNA. In contrast to inducible expression seen in most cell types, sICAM-1 is constitutively released by type I AEC and is present in normal alveolar lining fluid. Therefore, we compared the mechanism of sICAM-1 production in primary cultures of two closely juxtaposed cells in the alveolar wall, AEC and pulmonary microvascular endothelial cells (PVEC). AEC, but not PVEC, demonstrated high-level baseline expression of sICAM-1. Stimulation of AEC with TNFα or LPS resulted in minimal increase in AEC sICAM-1, whereas PVEC sICAM-1 was briskly induced in response to these signals. AEC sICAM-1 shedding was significantly reduced by treatment with a serine protease inhibitor, but not by cysteine, metalloprotease, or aspartic protease inhibitors. In contrast, none of these inhibitors effected sICAM-1 expression in PVEC. RT-PCR, followed by gel analysis of total RNA, suggests that alternatively spliced fragments are present in both cell types. However, a 16-mer oligopeptide corresponding to the juxtamembrane region of mICAM-1 completely abrogated sICAM-1 shedding in AEC but reduced stimulated PVEC sICAM-1 release by only 20%. Based on these data, we conclude that the predominant mechanism of sICAM-1 production likely differs in the two cell types from opposite sides of the alveolar wall.

Abstract This conceptual review seeks to reframe the view of academic integrity as something to be enforced to an academic skill that needs to be developed. The authors highlight how practices within academia create an environment where feelings of inadequacy thrive, leading to behaviours of unintentional academic misconduct. Importantly, this review includes practical suggestions to help educators and higher education institutions support doctoral students’ academic integrity skills. In particular, the authors highlight the importance of explicit academic integrity instruction, support for the development of academic literacy skills, and changes in supervisory practices that encourage student and supervisor reflexivity. Therefore, this review argues that, through the use of these practical strategies, academia can become a space where a culture of academic integrity can flourish.

Penelitian ini bertujuan untuk mendeskripsikan efektivitas pendekatan rational emotive behavior threapy dalam mengatasi perilaku bullying siswa. Rancangan penelitian ini menggunakan pendekatan kuantitatif eksperimen. Sampel dalam penelitian ini adalah siswa yang perilaku bullying sebanyak 20 orang. 10 orang untuk pelaksanaan kelompok eksperimen dan 10 orang untuk pelaksanaan kelompok kontrol. Instrument penelitian ini menggunakan skala perilaku bullying siswa yang telah memenuhi syarat instrumen penelitian, kemudian data di analisis dengan menggunakan uji Wilcoxon Signed Rank Test dan Uji Kolmogorov Smirnov 2 Independent Samples. Hasil penelitian menunjukkan bahwa: 1) Terdapat perbedaan yang signifikan perilaku bullying peserta didik kelompok eksperimen sebelum dan sesudah mengikuti pendekatan rational emotive behavior therapy format kelompok. 2) Terdapat perbedaan yang signifikan perilaku bullying peserta didik kelompok kontrol sebelum dan sesudah mengikuti layanan bimbingan kelompok tanpa menggunakan pendekatan rational emotive behavior therapy. 3) Terdapat perbedaan yang signifikan perilaku bullying peserta didik kelompok eksperimen yang diberikan pendekatan rational emotive behavior therapy format kelompok dengan peserta didik kelompok kontrol yang diberikan layanan konseling kelompok tanpa pendekatan rational emotive behavior therapy. kelompok eksperimen dan kontrol sama-sama mengalami penurunan, akan tetapi rata-rata penurunan kelompok eksperimen lebih besar pada kelompok kontrol.

Kesulitan belajar menulis nama lain dari disgrafia. Disgrafia menunjuk adanya suatu ketidakmampuan peserta didik mengingat cara membuat huruf atau simbol-simbol yang berbentuk angka dalam matematika. Disgrafia sering dikaitkan dengan dysleksia (kesulitan membaca) karena masih dalam kategori kesulitan belajar. Gangguan menulis atau kesulitan belajar menulis terdapat beberapa faktor diantaranya yaitu, terganggunya motorik pada anak, perilaku ketika menulis, persepsi, ingatan, penggunaan tangan yang dominan, kemampuan cross modal, serta kemampuan memahami instruksi.Adanya program bimbingan dan konseling ini guru BK dapat memberikan solusi-solusi yang telah diuraian di atas terhadap keslitan belajar yang di alami siswa terkhusus pada siswa yang kesulitan dalam hal menulis.

Dalam Pembelajaran kepribadian ada sebagian kepribadian yang mau dibesarkan oleh pemerintah serta satuan Pembelajaran, antara lain kepribadian/karakter religious, kejujuran, toleransi, disiplin, kerja keras, kreatif, mandiri, demokratis, rasa mau ketahui, semangat kebangsaan, cinta tanah air, menghargai prestasi, bersahabat/ komunikatif, cinta damai, peduli social serta bertanggung jawab. Sehingga nantinya interaksi yang hendak dijalani tiap hari oleh siswa membentuk diri siswa yang berkarakter peduli sosial. Terpaut dengan aktivitas pembelajaran kepribadian/karakter di sekolah konselor sekolah harus memfasilitasi pengembangan serta penumbuhan kepribadian/karakter dan tanpa mengabaikan kemampuan hard skills lebih lanjut yang dibutuhkan dalam ekspedisi hidup pembuatan kepribadian/karakter di Sekolah. 50. Factors Affecting the Utilization of Hospital Management Information System in PKU Muhammadiyah Bantul General Hospital

Hospitals are important institution in organizing health system in Indonesia. Because of its important role, it is crucial to ensure the data credibility. Therefore, protected by law, hospitals obliged to maintain their system information. This study conducted adopting Technology Acceptance Model (TAM) variable. This study was held in Bantul Regional General Hospital using cross sectional approach. Population of this study is all staff who are identified to be involved in the use of hospital management information system in PKU Muhammadiyah Bantul General Hospital as much as 325 staff. Sample were taken using proportionate stratified random sampling as much as 85 staff. Research data obtained by distributing questionnaires and analyzed using chi square and Mann Whitney test. Univariate analysis showed not good perceived benefit (58,8%), good perceived ease of use (52,9%) and not good attitude of using (69,4%). Bivariate analysis showed variables affecting utilization of hospitals management information system are perceived benefit (p=0,001), perceived ease of use (p=0,018) and attitude of using (p=0,021). Multivariate analysis showed the most affecting variables towards utilization of hospitals management information system are perceived benefit (p=0,008) and perceived of ease use (p=0,049).
