Abstract To date, only one polymer can self-grow to an extended length beyond its original size at room temperature without external stimuli or energy input. This breakthrough paves the way for significant advancements in untethered autonomous soft robotics, eliminating the need for the energy input or external stimuli required by all existing soft robotics systems. However, only freshly prepared samples in an initial state can self-grow, while non-fresh ones cannot. The necessity of synthesizing from monomers for each use imposes significant limitations on practical applications. Here, we propose a strategy to rejuvenate non-fresh samples to their initial state for on-demand self-growth through the synergistic effects of solvents and dynamic covalent bonds during swelling. The solvent used for swelling physically transforms the non-fresh LCEs from the liquid crystal phase to the isotropic phase. Simultaneously, the introduction of the transesterification catalyst through swelling facilitates topological rearrangements through exchange reactions of dynamic covalent bonds. The rejuvenation process can also erase growth history, be repeated several times, and be regulated by selective swelling. This strategy provides a post-modulation method for the rejuvenation and reuse of self-growing LCEs, promising to offer high-performance materials for cutting-edge soft growing robotics.

Highlights A novel cascade strategy is proposed to construct Pd-NiCo2O4 electrocatalyst. First time discovery that Ni incorporation together with Pd loading is able to balance the competitive adsorption of OH– and 5-hydroxymethylfurfural. Pd–NiCo2O4 promotes both the indirect and direct synergistic oxidation process. Pd–NiCo2O4 catalyst exhibits extraordinary current density and excellent Faradaic Efficiency at a low potential.

Abstract Due to the rapid development of stem cell technology, there have been tremendous advances in molecular biological and pathological research, cell therapy as well as organoid technologies over the past decades. Advances in genome editing technology, particularly the discovery of clustered regularly interspaced short palindromic repeats (CRISPR) and CRISPR-related protein 9 (Cas9), have further facilitated the rapid development of stem cell researches. The CRISPR-Cas9 technology now goes beyond creating single gene editing to enable the inhibition or activation of endogenous gene loci by fusing inhibitory (CRISPRi) or activating (CRISPRa) domains with deactivated Cas9 proteins (dCas9). These tools have been utilized in genome-scale CRISPRi/a screen to recognize hereditary modifiers that are synergistic or opposing to malady mutations in an orderly and fair manner, thereby identifying illness mechanisms and discovering novel restorative targets to accelerate medicinal discovery investigation. However, the application of this technique is still relatively rare in stem cell research. There are numerous specialized challenges in applying large-scale useful genomics approaches to differentiated stem cell populations. Here, we present the first comprehensive review on CRISPR-based functional genomics screening in the field of stem cells, as well as practical considerations implemented in a range of scenarios, and exploration of the insights of CRISPR-based screen into cell fates, disease mechanisms and cell treatments in stem cell models. This review will broadly benefit scientists, engineers and medical practitioners in the areas of stem cell research.

Local oxidative stress in diseases or injury severely hinders cell homeostasis and organ regeneration. Antioxidant therapy is an effective strategy for oxidative stress treatment. Biomaterials with good biocompatibility and reactive oxygen species (ROS) scavenging ability are good choices for antioxidant therapeutics. However, there are few natural biomaterials that are identified with both biocompatibility and strong antioxidant activity. Here, we show, for the first time, that silk fibroin (SF) is a strong antioxidant, which can eliminate ROS in both cells and zebrafish. We further demonstrate that the β-sheet structures turn into a random coiled structure when SF is treated with hydrogen peroxide. The content of β-sheet structures can be increased by heating, thus enhancing the antioxidation properties of SF. Therefore, SF can serve as a good antioxidant biomaterial for therapeutics, and its β-sheet structure-based antioxidation mechanism provides a novel theoretical basis, which could be a new cue for more antioxidant biomaterial discovery and identification.

The treatment of internal hemorrhage remains challenging due to the current limited antibacterial capability, hemostatic efficacy, and biocompatibility of hemostatic materials. The TEMPO-oxidized cellulose nanofibers/collagen/chitosan (TCNF/COL/CS) hemostatic aerogel was developed in this work by physically encasing COL in a sandwich structure and electrostatically self-assembling polyanionic TCNF with polycationic CS. In vitro coagulation experiments revealed the favorable procoagulant properties of TCNF/COL/CS along with high adhesion to erythrocytes and platelets. TCNF/COL/CS significantly increased the hemostatic efficacy by 59.8 % and decreased blood loss by 62.2 % in the liver injury model when compared to Surgicel®, the most frequently used hemostatic material. Furthermore, it demonstrated outstanding biodegradability both in vitro and in vivo, and a substantial increase in resistance (96.8 % against E. coli and 95.4 % against S. aureus) compared to TCNF. The significant hemostatic and biodegradable characteristics of TCNF/COL/CS can be ascribed to its interconnected porous structure, increased porosity, and efficient water absorption, along with the synergistic effect of the three constituents. The TCNF/COL/CS aerogel shows significant potential to control internal bleeding. A novel plant-derived nanocellulose composite aerogel has been described here for the first time; it has outstanding antibacterial characteristics, higher biocompatibility, and outstanding hemostatic characteristics in vivo.

Abstract Gelatin was widely used as scaffold materials in 3D bio-printing due to its excellent bioactivity and availability and especially that their arginine–glycine–aspartic acid (RGD) sequences could efficiently promote cell adhesion and proliferation. In this study, an electroactive and 3D bio-printable hydrogel was prepared through a two-step chemical cross-linking process. Specifically, residual free amino groups of methacrylated gelatin (GelMA) were cross-linked with the aldehyde groups of dibenzaldehyde-terminated telechelic polyethylene glycol (DF-PEG) via Schiff base bonds, forming a gel at 37 °C. During the subsequent 3D bio-printing process, GelMA underwent UV curing, forming a secondary cross-linked network to the mechanical strength and stability of the printed structure. The uniform dispersion of carbon nanotubes (CNTs) in the GelMA/DF-PEG composite hydrogel significantly increased its conductivity. The optimized GelMA/DF-PEG composite hydrogel, i.e., 30% GelMA and 25% DF-PEG (G30D25-CNTs), exhibited superior bio-printability. When the content of CNTs was above 4%, the conductivity of G30D25-CNTs hydrogel exceeded 10–2 S/m, which satisfied the needs of cells for micro-current stimulation. Furthermore, the pore microstructures, swelling behavior, degradation ability and cell toxicity of G30D25-CNTs electroactive hydrogels were thoroughly evaluated. Thus, the G30D25-CNTs hydrogel with 4% MWCNTs could be considered for further application in electrical stimulation of tissue regeneration such as muscle and cardiac nerve tissue repair.

Vibration signals are essential data for the health monitoring of structures and the cyber–physical system. However, commercial vibration sensors are generally installed on struts/surfaces to gather raw data, which affects the result or causes the detachment problem. This study uses a facile 3D printing method to fabricate a smart vibration absorber with an interior multifunctional multimaterial elastic lattice (MMEL). These lattices, which work as vibration absorbers, have been demonstrated to possess the functions of self‐powered sensing and energy harvesting via the triboelectric effect. The triangular geometry parameter, materials type, etc. have been investigated to illustrate their basic mechanical and triboelectric properties and their coupled influence. Further, the results of the shock test show that MMEL can decrease the peak force from approximately 625 to 90 N and convey the Voc impulse signal simultaneously. The vibration signal has been collected through the MMEL to detect the vibration frequency and charge a watch simultaneously, demonstrating the feasibility and practical potential of the MMEL. The research provides a new method for constructing a multifunctional vibration absorber for applications.

Nano-assemblies have wide applications in biomedicine, functional coatings, Pickering emulsifiers, hydrogels, and so forth. The preparation of assemblies mainly utilizes the polymerization-induced self-assembly (PISA) method, which can produce high-concentration nanoscale assemblies in one step. However, the initiation processes of most reported PISA are limited to thermal initiation. Here, we reported two green and efficient methods for synthesizing nano-assemblies with various morphologies using ultrasound (20 kHz)/ microwave (500 W) assisted aqueous-phase RAFT-PISA in 3 h and 1 h. Cyclodextrin (CD) and styrene (St) nucleating monomer were complexed in a 1:1 ratio. Then, using Poly (ethylene glycol) methyl ether as the macromolecular reversible addition-fragmentation chain transfer (RAFT) agent (PEG-CTA) to control the CD/St complexes, the conversion rate of St monomer was respectively 27 %–60 %, 20 %–30 % within 3 h and 1 h under ultrasonics/microwave assisted PISA. Results showed that the morphologies of the assemblies are not only related to the length of PS block, but also to the assistance types and the remaining monomer concentration. The results showed that only PEG45-b-PS90 and PEG45-b-PS241 assemblies prepared by ultrasonics assisted PISA form evolved lamellaes and vesicles (100 nm), which break through the limitation of kinetic freezing. But the ultrasonic reaction on morphology of assemblies is not all favourable. For one thing, it can promote the movement of particles; for another, it makes reverse morphology transformation and sphere is preferred morphology. Therefore, the main reason of morphology evolution is the remaining monomer concentration of PEG45-b-PS90 and PEG45-b-PS241 assemblies reaches to 55 %–65 %, which promoting the segment movement. The results showed that the morphology of the assemblies prepared by microwave assisted PISA changed from spherical micelles to short rods, and finally to vesicles (120–140 nm) as the length of hydrophobic PS block increases. The kinetic freezing problem was solved in microwave-assisted PISA due to the action of microwaves and more remaining monomer concentration. Both them can boost particles movement.

Osteoporosis, closely related to age, is characterized by impaired bone mass and bone strength, damage of bone microstructure, resulting in a higher risk of fractures with minimal trauma, especially in the hip and spine. Quercetin (Que), a natural compound, has previously demonstrated its potential anti-osteoporosis effects via promoting bone formation and anti-bone resorption. However, Que is poor water solubility, lack of bone targeting, and requires high concentration to exert its therapeutic effects. In this work, a pH-responsive and bone-targeted nanomedicine HCCP-Que-PEG-ALN (HQPA) was developed via using hexachlorocyclotriphosphazene (HCCP) as the cross-linkage, Que, alendronate (ALN), and amino-polyethylene glycol (NH2-PEG) as other building blocks. We demonstrated that HQPA can form spherical nanoparticles in aqueous solution with average size of 158.3 ± 59.8 nm with high drug loading capacity of Que (31.4 w/w%) and obvious bone-targeting capability. in vitro experimental results indicated that HQPA NPs have dual anti-osteoporosis effects of promoting osteoblasts and inhibiting osteoclasts. Furthermore, HQPA NPs can effectively reduce the bone loss in OVX mice with low toxicity to major organs. We believe that this method should be a general and useful route for fabrication of bone-targeted natural-compounds-based nanomedicines for treatment different bone-related diseases.

Abstract The utilization of biochar derived from biomass residue to enhance anaerobic digestion (AD) for bioenergy recovery offers a sustainable approach to advance sustainable energy and mitigate climate change. However, conducting comprehensive research on the optimal conditions for AD experiments with biochar addition poses a challenge due to diverse experimental objectives. Machine learning (ML) has demonstrated its effectiveness in addressing this issue. Therefore, it is essential to provide an overview of current ML-optimized energy recovery processes for biochar-enhanced AD in order to facilitate a more systematic utilization of ML tools. This review comprehensively examines the material and energy flow of biochar preparation and its impact on AD is comprehension reviewed to optimize biochar-enhanced bioenergy recovery from a production process perspective. Specifically, it summarizes the application of the ML techniques, based on artificial intelligence, for predicting biochar yield and properties of biomass residues, as well as their utilization in AD. Overall, this review offers a comprehensive analysis to address the current challenges in biochar utilization and sustainable energy recovery. In future research, it is crucial to tackle the challenges that hinder the implementation of biochar in pilot-scale reactors. It is recommended to further investigate the correlation between the physicochemical properties of biochar and the bioenergy recovery process. Additionally, enhancing the role of ML throughout the entire biochar-enhanced bioenergy recovery process holds promise for achieving economically and environmentally optimized bioenergy recovery efficiency. This study primarily investigates the effect of fly ash (FA) content on the mechanical properties and hydration performance of alkali-activated ground granulated blast furnace slag cement (AAGC) and compares the related properties with ordinary Portland cement (OPC). Additionally, we examined the hydration products; performed thermal analysis, MIP, and SEM; and determined chemically bound water and pH values of AAGC. The compressive strength of AAGC showed a retrogression phenomenon from 3 to 28 days, with the 14-day and 28-day compressive strengths of AAGC being higher than those of OPC. The AAGC with 20% FA content exhibited the highest 28-day compressive strength (75 MPa). The hydration heat release rate curve of OPC and AAGC was divided into the initial induction period, induction period, acceleration period, deceleration period, and steady period. As FA content increased, the 28-day pore volume of AAGC increased, while pH values and chemically bound water decreased. SEM images of AAGC with low FA content showed more microcracks.

Although oily wastewater treatment realized by superwetting materials has attracted heightened attention in recent years, how to treat enormous-volume emulsion wastewater is still a tough problem, which is ascribed to the emulsion accumulation. Herein, to address this problem, a material is presented by subtly integrating chemical demulsification and 3D inner-outer asymmetric wettability to a sponge substrate, and thus wettability gradient-driven oil directional transport for achieving unprecedented enormous-volume emulsion wastewater treatment is realized based on a “demulsification-transport” mechanism. The maximum treatment volume realized by the sponge is as large as 3 L (2.08 × 104 L per cubic meter of the sponge) in one cycle, which is about 100 times of the reported materials. Besides, owing to the large pore size of the sponge, 9000 L m2 h−1 (LMH) separation flux and 99.5% separation efficiency are realized simultaneously, which overcomes the trade-off dilemma. Such a 3D inner-outer asymmetric sponge displaying unprecedented advantage in the treatment volume can promote the development of the oily wastewater treatment field, as well as expand the application prospects of superwetting materials, especially in continuous water treatment.
