Despite the demonstrated effectiveness of nano-materials for drug delivery to the brain, a comprehensive understanding of their transport processes across the blood brain barrier (BBB) remains undefined. This multidisciplinary study aimed to gain an insight into the transport processes across BBB, focusing on the transcytosis of liposomes and the impact of liposomal pH-sensitivity. Glutathione-PEGylated pH-sensitive (GSH-PEG-pSL) and non pH-sensitive liposomes (GSH-PEG-L) were fluorescently labelled with rhodamine-DOPE and calcein, both impermeable to biomembranes. Following exposure to brain microvascular endothelial cells (hBMECs), the key functional component of the BBB, intracellular trafficking were evaluated by confocal live-cell imaging. The exocytosed liposomes, including naturally-occurring extracellular vesicles (EVs), were collected using differential centrifugation and and characterised regarding the EV yield, morphology and EVs origin using nanoparticle tracking analysis, transmission electron microscopy and flow cytometry. The transcytosis of liposomes through a verified BBB model comprising of hBMECs monolayer was also quantified. GSH-PEG-L was initially retained in the endo-lysosomes before exocytosed while packed in EVs of different sizes (1 ​μm) while GSH-PEG-pSL underwent endosome escape with less degree of exocytosis with more fluorescence remaining in the cytoplasm. Compared with the untreated, hBMECs treated with GSH-PEG-L increased the yield of nano-EV and medium-EV by 7.9-fold and 4.6-fold, respectively. Conversely, GSH-pSL-treated cells produced 2.9-fold more nano-EVs but 2-fold less medium-EVs than the control cells. These vesicles were CD144-positive confirming their endothelial cell-origin. GSH-PEG-L demonstrated 2-fold higher efficiencies than GSH-PEG-pSL to cross the in vitro BBB model via exocytosis. Taken together, GSH-PEG-L might utilize EV secretion pathway to achieve transcytosis across brain endothelial cells of the BBB while liposomal pH-sensitivity favors cytoplasmic delivery.

Antiphospholipid antibodies (aPL) are autoantibodies that cause pregnancy disorders by a poorly defined mechanism that involves the placenta. The human placenta is covered by a single multinucleated cell, the syncytiotrophoblast, which extrudes vast numbers of extracellular vesicles (EVs) into the maternal blood. Extracellular vesicles are tiny packages of cellular material used by cells for remote signalling. In normal pregnancy, placental EVs assist maternal adaptations to pregnancy. We have previously shown that aPL alter the cargo of placental EVs, increasing the load of danger signals. These changes in EV cargo may explain how aPL contribute to the increased risk of recurrent miscarriage, preeclampsia and stillbirths observed in aPL-affected pregnancies. An additional possibility, that aPL alters the targeting of placental EVs to maternal organs to cause maternal maladaptation to pregnancy was investigated in this study.

Small extracellular vesicles (sEVs) have emerged as attractive drug delivery systems. However, the intracellular release of their cargoes is restricted. This study aimed to develop an efficient approach to re-engineer sEVs by hybridisation with pH-sensitive liposomes (PSLs) and investigate their endosome escape potential. Microbial fuel cells (MFCs) degrade organic contaminants in wastewater while simultaneously producing electricity, but must be stacked to yield adequate voltage and current. This study examined the evolution of the chemical oxygen demand (COD) removal rate and efficiency in two identical individual MFCs (i-MFCs) in series- and parallel-connected stacks (sc- and pc-MFCs, respectively) under batch and continuous operation. The stack voltage and current increased in the respective series and parallel connections of the two i-MFCs (MFC unit 1 and MFC unit 2). Voltage reversal was observed in the sc- MFC below an external load of 100 Ω. Regardless of occurrence of the voltage reversal, organic reduction between i-MFCs and sc-MFCs showed no significant difference (gap of < 9% and < 6% in COD removal rate and efficiency, respectively); additionally, organic removals between the two individual MFCs in series indicated differences less than 9% of COD removal rate and 5% of COD removal efficiency in batch mode. Continuous operation also yielded similar organic removals as the MFCs in individual and series connection (voltage reversal occurred) mode, even over 8 days operation. Parallel connection yielded identical organic removals and currents in the two individual MFCs of the pc-MFC, even though the two separate i-MFCs showed different organic removal rates and current productions. This study provides the guide for the application of stacked MFCs for power source and efficient organic pollutant removal in wastewater treatment process.

This study evaluated the optimal supply water temperature and flow rate in response to the outdoor air temperature in order to maintain a target indoor air temperature and minimize the amount of energy required to reheat a recirculated water curtain in a plastic greenhouse. This paper proposed a multiple regression model that estimated the indoor air temperature using the outdoor air and the inlet water temperatures, as well as the water flow rate, as independent variables. We also established a multiple regression model that estimated the average amount of energy required to reheat the recirculated water curtain. To this end, the study analyzed and quantified a reasonable inlet water temperature and water flow rate in response to the outside air temperature using isothermal and iso-energy curves. The results indicated that maintaining a higher water flow rate was preferable to maintaining a higher inlet water temperature for cultivation purposes.

In this study, a hot water pipe and a blowing fan were combined for developing zone heating technology for cherry tomato. The concept of this system was that hot air was firstly made by hot water pipe in one layer plastic duct and then a blowing fan made the hot air formed in a duct discharge through a duct hole to a shoot apex or a flower cluster which was temperature-sensitive part of cherry tomato. This system mainly consisted of hot water boiler, thermal tank, heat radiation plastic duct with the function of moving up and down electrically depending on the height of shoot apex. Developed system was applied to the cherry tomato greenhouse located in Jangam Chungcheongnamdo from Dec. 28, 2015 to Feb. 16, 2016 and compared with conventional entire space heating system of cherry tomato greenhouse and looked into cumulative yield for the estimate of growing state and energy saving rate from the conventional consumed energy. The result showed that cumulative yield was 3% higher and consumed energy was 32% lower than those of control greenhouse. The average temperature of shoot apex zone was 0.4~1.1℃ higher and the average relative humidity of shoot apex zone was 2,2~2.3% lower than those of entire space during night time in a shoot apex zone heating greenhouse and the average temperature of shoot apex zone was 0.7~1.4℃ lower and the relative humidity of shoot apex zone was 2.9~8.3% higher than those of entire space during night time in a entire space heating greenhouse.

This study was carried out to provide a valuable reference which could reduce heating loss of air-inflated double PO film. Therefore, this study was aimed to choose the best air space thickness and injected air temperature. The characteristics of heat transmission variation at experimental materials were measured and analysed in the laboratory. The experiment was conducted of two layers of PO film, each 0.15 mm tick, sandwiching 110, 175, 225 mm of inflated air with 1 m sides. Environmental control lab was constantly controlled with and experiment chamber was constantly changed with 0, 5, 10, . The analysis of heat transfer showed that heat transmission does not have a direct correlation with Air Space thickness and injected air temperature. But when inside and outside temperature difference of chamber was great, supply of outside air to Air Space had an advantage at reduction of heating load. It was required to examine accurate analysis at a real greenhouse.

We have previously shown that adenosine amine congener (ADAC), a selective A1adenosine receptor agonist, can ameliorate noise- and cisplatin-induced cochlear injury. Here we demonstrate the dose-dependent rescue effects of ADAC on noise-induced cochlear injury in a rat model and establish the time window for treatment.Methods. ADAC (25–300 μg/kg) was administered intraperitoneally to Wistar rats (8–10 weeks old) at intervals (6–72 hours) after exposure to traumatic noise (8–16 kHz, 110 dB sound pressure level, 2 hours). Hearing sensitivity was assessed using auditory brainstem responses (ABR) before and 12 days after noise exposure. Pharmacokinetic studies investigated ADAC concentrations in plasma after systemic (intravenous) administration.Results. ADAC was most effective in the first 24 hours after noise exposure at doses>50 μg/kg, providing up to 21 dB protection (averaged across 8–28 kHz). Pharmacokinetic studies demonstrated a short (5 min) half-life of ADAC in plasma after intravenous administration without detection of degradation products.Conclusion. Our data show that ADAC mitigates noise-induced hearing loss in a dose- and time-dependent manner, but further studies are required to establish its translation as a clinical otological treatment.
