Abstract

The large-scale storage and stable supply of oil products are essential for national energy security and economic development. As the economy expands and energy demands rise, centralized storage and supply systems become increasingly vital for ensuring the efficiency and reliability of oil product distribution. However, large oil storage depots present substantial safety risks. In the event of fires, explosions, or other accidents, emergency response efforts face stringent demands and challenges. To enhance the emergency response capabilities of oil storage and transportation bases (OSTBs), this paper proposes an innovative approach that integrates the improved analytic hierarchy process (IAHP) with the Entropy Weight Method (EMW) to determine the combined weights of various indices. This approach reduces the subjective bias associated with the traditional analytic hierarchy process (AHP). The emergency response capabilities of OSTBs are subsequently evaluated through fuzzy comprehensive analysis. An empirical study conducted on an OSTB in the Zhoushan archipelago quantitatively assesses its emergency preparedness. The results show that the base excels in pre-incident prevention, demonstrates robust preparedness and response capabilities, and exhibits moderate recovery abilities after incidents. These findings provide a theoretical foundation for reducing the likelihood of accidents, enhancing emergency response efficiency, and mitigating the severity of consequences. 4. Cytospora pyripromotes Erwinia amylovoravirulence by providing metabolites and hyphae

Abstract

Bacterial–fungal interactions are widespread in nature. We observed that pear orchards affected by Cytospora pyri(formerly Valsa pyri) were often accompanied with Erwinia amylovora. However, the relationship of the two pathogens was unclear. The objective of this study was to determine whether the synergistic effect exists between E. amylovoraand C. pyri. We first analyzed the coexistence frequencies of E. amylovoraand C. pyriin pear trees. Virulence of the two pathogens, growth, physical interactions, amylovoran production, and expression of genes for amylovoran biosynthesis were conducted. Our results showed that E. amylovoraand C. pyricould coexist on the same lesion and caused much more severe disease. We also found that E. amylovoracould physically attach to C. pyriand the expression of amylovoran biosynthesis genes were up-regulated with fungal metabolite treatment. These results indicate that E. amylovoraand C. pyrican cooperatively interact, which provides C. pyriwith an opportunity to promote bacterial dispersal and production of virulence factor in E. amylovora.

Abstract

Meloidogyne javanica is one of the most widespread and economically important nematodes in many countries, including China. In this study, a recombinase polymerase amplification (RPA) assay was evaluated for the detection of M. javanica based on the sequences of a sequence-characterized amplified regions marker gene segment. The RPA assay specifically detected M. javanica from individual juvenile or adult female, M. javanica-induced galls, and nematodes in the soil samples. The detection limit of M. javanica RPA assay was 1 pg of purified genomic DNA, 0.01 adult female, or 0.1 second-stage juvenile, which was 10 times more sensitive than conventional PCR assay. Furthermore, combined with lateral flow dipstick (LFD), a visual detection method of LFD-RPA assay was developed, which is suitable for onsite surveys and routine diagnostics. Results indicate that the RPA assay is rapid, sensitive, and reliable for detection and molecular identification of M. javanica.

Abstract

Bipolaris sorokiniana is a fungal pathogen that caused root rot and spot blotch on wheat. A Recombinase Polymerase Amplification (RPA) assay was developed to detect B. sorokiniana specifically based on the sequences of calmodulin gene sequence. The specificity of RPA assay was tested against 19 fungal organisms associated with wheat, and the detection limit was determined to be 10 pg for B. sorokiniana pure fungal DNA. Further, the RPA assay was capable of detecting B. sorokiniana from artificially inoculated and field collected wheat tissues directly. These results indicate that the RPA assay is rapid and effective method for detection and molecular identification of B. sorokiniana.
