Abstract

The BRCA2 tumor suppressor protects genome integrity by promoting homologous recombination-based repair of DNA breaks, stability of stalled DNA replication forks and DNA damage-induced cell cycle checkpoints. BRCA2 deficient cells display the radio-resistant DNA synthesis (RDS) phenotype, however the mechanism has remained elusive. Here we show that cells without BRCA2 are unable to sufficiently restrain DNA replication fork progression after DNA damage, and the underrestrained fork progression is due primarily to Primase-Polymerase (PRIMPOL)-mediated repriming of DNA synthesis downstream of lesions, leaving behind single-stranded DNA gaps. Moreover, we find that BRCA2 associates with the essential DNA replication factor MCM10 and this association suppresses PRIMPOL-mediated repriming and ssDNA gap formation, while having no impact on the stability of stalled replication forks. Our findings establish an important function for BRCA2, provide insights into replication fork control during the DNA damage response, and may have implications in tumor suppression and therapy response.

Abstract

The dynamic behaviors of microRNA and mRNA under external stress are studied with biological experiments and mathematics models. In this study, we developed a mathematic model to describe the biological phenomenon and for the first time reported that, as responses to external stress, the expression levels of microRNA and mRNA sustained oscillation. And the period of the oscillation is much shorter than several reported transcriptional regulation negative feedback loop., Comment: There were some mistakes in the analysis in this paper's first version submitted on 22 Apr 2013. Abstract

Cancer cells maintain a high level of aerobic glycolysis (the Warburg effect), which is associated with their rapid proliferation. Many studies have reported that the suppression of glycolysis and activation of oxidative phosphorylation can repress the growth of cancer cells through regulation of key regulators. Whether Warburg effect of cancer cells could be switched by some other environmental stimulus? Herein, we report an interesting phenomenon in which cells alternated between glycolysis and mitochondrial respiration depending on the type of radiation they were exposed to. We observed enhanced glycolysis and mitochondrial respiration in HeLa cells exposed to 2-Gy X-ray and 2-Gy carbon ion radiation, respectively. This discovery may provide novel insights for tumor therapy.

17. Vibration Characteristics and Vibration Mitigation Analysis of End Cap PM Excitation Homopolar Inductor Machine

Abstract

Vibration analysis and vibration mitigation are very important to ensure the safe and stable operation of electrical machines applied in flywheel energy storage systems. In traditional integer slot synchronous machines, it is generally believed that there is only the zeroth-mode vibration caused by odd tooth harmonics. However, the homopolar inductor machine (HIM) adopts the end cap excitation structure, which makes its air-gap magnetic field contain the dc component, odd tooth harmonics and even tooth harmonics. The force density distribution on the slotted stator is analyzed by the magnetomotive force-multiplying permeance method. The results show that there are both zeroth- and pth(pole pairs)-mode vibrations caused by tooth harmonics and dc component of the air-gap flux density. Furthermore, to reduce vibration and resonance, and to improve the electromagnetic performance of HIM, it is best to use the stator skew slot, the magnetic slot wedge and the sinusoidal rotor slot at the same time. Finally, a 54-slot/6-pole end cap permanent magnet excitation homopolar inductor machine (EPE-HIM) is tested to validate the analytical and the finite-element method (FEM) results, and the variation trend of the experimental results and FEM results is the same.

Abstract

An air-core compensated pulse alternator is a compact pulsed power supply for electromagnetic launchers. The structures of these machines are designed distinct from that of traditional synchronous generators. The materials used to build rotor and stator are non-ferromagnetic instead of the traditional ferromagnetic materials. They are usually designed slotless to enhance the stiffness of coupling of windings. As a result, the method of windings inductance calculation is different from that of the traditional ones. The parameter of windings inductance of air-core compensated pulsed alternator plays an important role in the machine design and performance simulation. Therefore, an analytical method to calculate the windings inductance is needed. In this paper, two analytical methods, a 2D method and a 3D method, are used to calculate the windings inductance. As to the special figuration of these windings, a 2D analysis method based on the parallel two-conductor line electromagnetic theory and the superposition principle is employed to calculate the self-inductance and the mutual-inductance of them. Then, a 3D analytical method based on the theory of mutual-inductance between two parallel rectangular loops whose central axes are the same is presented. Finally, a finite-element method based on the software ANSYS is also used to calculate the inductance, which is used to verify the accuracy of the analytical methods.
