The purpose of this paper is to quantify the level of new-type urbanization and unravel the spatial and nonlinear effects of new-type urbanization and technological innovation on industrial carbon emissions. Although the impact of traditional urbanization levels on carbon emissions has been widely studied, there is still a huge room for optimization, and the impact of new-type urbanization on carbon emissions has not yet been clarified. Selecting 37 cities in the Yangtze River Delta as a research sample, this paper measures the new-type urbanization based on an evaluation system we build. Consequently, we assess the spatial and nonlinear effects of new-type urbanization and technological innovation on carbon emissions by the spatial Durbin model and non-parameter addictive model, respectively. The results indicate that the new-type urbanization and low-carbon city pilot policy have significant spatial spillover effects on reducing carbon dioxide emissions, while the economic growth plays a positive role in increasing carbon emission. As for nonlinear effects, there is a significant inverted "N"-shaped relationship between the level of new-type urbanization and carbon dioxide emissions, while the nexus between technological innovation and carbon emissions is an inverted "U"-shaped relationship. This paper provides a new perspective for confirming the mechanism of the new-type urbanization on carbon emissions. Meanwhile, these findings are of significance for the relevant authorities in China to develop appropriate policy in carbon dioxide emission reduction.

A CMOS-compatible, broadband, and polarization-independent edge coupler for efficient chip coupling with standard single-mode fiber is proposed. Three layers of a silicon nitride waveguide array with the same structures are used in the top oxide cladding of the chip to achieve high coupling efficiency and to simplify the mode transformation structure. Optimal total coupling loss at the wavelength of 1550 nm, − 0.49 d B for TE mode polarization and − 0.92 d B for TM mode polarization is obtained. The − 1 d B bandwidth is beyond 160 nm for TE mode polarization and ∼ 130 n m for TM mode polarization, respectively. A significant reduction in the packaging cost of silicon photonic chips is anticipated. Meanwhile, the structure holds vast potential for on-chip three-dimensional photonic integrations or fiber-to-chip, chip-to-chip optical interconnections.

Epidermal growth factor receptor (EGFR) activating mutations represent major drivers to the development of non-small cell lung cancer (NSCLC). Among the oncogenic EGFR mutations, a significant cohort, counting for approximately 4-10% of the EGFR mutation spectrum, bear EGFR exon 20ins mutations. Meanwhile, approximately 2% of NSCLC patients bear hotspot mutations in HER2. Strikingly, over 90% of the HER2 mutations occurred in NSCLC are identified as exon 20ins mutations. Despite the successful launch of 1st, 2nd, and 3rd generation of EGFR inhibitory agents in the clinic that inactivate oncogenic EGFR signaling through targeting specific EGFR mutations, de novo or acquired, none of these standard-of-care therapies is specific to EGFR exon 20ins or HER2 exon 20ins. In addition, trastuzumab and EGFR-TKIs have limited effectiveness for NSCLC patients with HER2 exon 20ins mutation. TAK-788 (mobocertinib) and JNJ6372 (amivantamab-vmjw) are the FDA approvals for NSCLC driven by EGFR exon 20ins mutations. Only T-Dxd is used as a second-line treatment for NSCLC patients with HER2 mutation. Considering the large population of lung cancer and the fact that many patients are missed in diagnosis due to the heterogeneous characteristics of EGFR and Her2 exon 20ins, there are probably more than ten thousand lung cancer patients suffering the EGFR or Her2 exon 20ins mutations. There are urgent unmet medical needs to develop target therapeutics for EGFR and Her2 exon 20ins mutations. We discovered and developed TY-4028, which is a novel, potent, and orally available inhibitor targeting EGFR and Her2 exon 20ins mutations and is currently in the IND enabling stage. In EGFR-related tumor cells and genetically engineered Ba/F3 cell lines, TY-4028 showed similar or better antitumor effects than TAK-788, and better antitumor effects than DZD9008. The B/P ratio (brain tissue AUC0-last/plasma AUC0-last) of SD rats was 1.63 and 1.04 respectively after oral administration of TY-4028 in male and female SD rats, which suggested that TY-4028 had good potential to cross Blood Brain Barrier (BBB). Preclinical studies showed a good PK profile and manageable toxicity with TY-4028. TY-4028 has remarkable efficacy in mouse models of EGFR exon 20ins and HER2 exon 20ins. The data showed that all doses of TY-4028 had significant effects, and the tumors nearly demonstrated complete regression in the PDX LU0387 model and PC9 CDX model. At the same dose, the efficacy of TY-4028 was similar to that of TAK-788, while the tolerance of TY-4028 was better than that of TAK-788. At the same dose, the efficacy of TY-4028 was better than that of DZD9008. Taken together, the data demonstrated TY-4028 has great potential to meet the unmet medical needs for NSCLC patients with EGFR exon 20ins mutation or HER2 exon 20ins mutation. #Jun Li and Chengshan Niu contributed equally to this work. *They are the correspondent authors. Proto-oncoprotein RET (rearranged during transfection) is a receptor tyrosine kinase and belongs to the cadherin superfamily. RET fusion proteins and gatekeeper mutations represent strong cancer drivers involved in the development of a variety of cancers. Although selective RET inhibitors selpercatinib (LOXO-292) and pralsetinib (BLU667) were recently marketed for patients with RET-dependent NSCLC and thyroid cancers, RET solvent front mutations G810R/S/C/V and other secondary mutations such as Y806C/N and V728A have been identified as mechanisms counting for acquired resistance to the two drugs and limits the applications of the kinase inhibitors. To provide novel RET inhibitors effective to a broader spectrum of RET fusions and mutations, we identified TY-1091 as a novel next-generation RET inhibitor through the in-house RET program and systemic screening of RET compound candidates. TY-1091 was characterized for its anti-tumor activity through in vitro and in vivo testing of a variety of RET-dependent tumor models including a panel of 17 engineered RET mutant Ba/F3 cell lines and 2 cancer cell models. The results show that TY-1091 inhibits wild type and major RET mutants (IC50, nM): RET G810S (9.5 nM), RET V804M/L/E (2.8-12.6 nM), and double mutants RET V804M/G810S (23.5 nM) and M918T/G810S (47.0 nM) through a biochemical screening against a panel of 17 engineered RET mutant Ba/F3 cell lines. Importantly, the inhibitory activity of TY-1091 is much higher than that of first-generation RET inhibitor Cabozantinib, and comparable to other second-generation compounds LOXO-292 and BLU-667. Consistent with its mode of action, TY-1091 grants extraordinary inhibition effects to tumor cell proliferation compared to its peer compounds LOXO-292 and BLU-667 (including TT (thyroid cancer, RET C634W) and LC2/ad (NSCLC, CCDC6-RET)), and the inhibition of the RET pathway activation, i.e., inhibition of RET phosphorylation (IC50 < 1 nM), SHC phosphorylation (IC50 = 4.6 nM) and ERK phosphorylation (IC50 = 9.8 nM) in Ba/F3-KIF5B-RET cells further validated the above in vitro phenotype. The therapeutic potential of TY-1091 was then further validated through mouse pharmacology in that TY-1091 demonstrated remarkable anti-tumor efficacy in a variety of xenograft models including Ba/F3 KIF5B-RET (wt), Ba/F3 KIF5B-RET (V804L), TT (thyroid cancer, RET C634W), and LC2/ad (NSCLC, CCDC6-RET). Detailed data will be presented. In summary, TY-1091 is a highly potent, orally available, and safe small molecule inhibitor to pan-RET mutations in cancer, and may attenuate SFMs-mediated resistance to existing RET therapy. Light is one of the most prestigious environmental sign, which stimulates plant metabolite consequences. Zhong Ming 6 (ZM6) is a green tea (Camellia sinensis) cultivar with highly accumulating TGGP. Here, three kinds of supplemental light wavelengths including blue light (BL,200 µmol m−2 s−1), red light(RL,200 µmol m−2 s−1), and White Light (WL/CK, 200 µmol m−2 s−1) were applied to explore their effects on the transcriptomes and metabolomics of young shoots (one bud and two leaves) in tea plants. Interestingly, artificial BL and RL significantly affect the secondary metabolites and transcriptome factors of tea plants. Here, BL extensively dominated the multiple physiological actions with secondary metabolism. In addition, RL could induce plant growth, development, and photosynthesis. Thoroughly, the identification of eight structural genes and 34 transcription factors (TFs) signiଁcantly correlated with total catechin (TC) and anthocyanin. Due to the upregulations of the CsGSTF1 gene in the flavonoid biosynthesis pathway, anthocyanin production was maximum under BL. Then, CsMYB75, bHLH-MYC, and other R2R3-MYBs were highly upregulated in BL to increase the accumulation of TC and anthocyanins in tea plants. Again, the CsMYB4 gene was highly significant and positively correlated with TC and anthocyanin accumulation under RL. We indicated that BL is more feasible due to the number of functional metabolites (gallic acid, caffeine, TC, TGGP (1, 2, 6-tri-O-galloyl-β-D-glucopyranose), and anthocyanin) being supreme. For taste, quality, and dynamic indigenous mechanism of the tea plant, RL is also suitable that increase Chlorophyll content and tea yield.

A broadband and CMOS-compatible polarization beam splitter and rotator (PSR) built on the silicon nitride-on-silicon multilayer platform is presented. The PSR is realized by cascading a polarization beam splitter and a polarization rotator, which are both subtly constructed with an asymmetrical directional coupler waveguide structure. The advantage of this device is that the function of PSR can be directly realized in the SiN layer, providing a promising solution to the polarization diversity schemes in SiN photonic circuits. The chip is expected to have high power handling capability as the light is input from the SiN waveguide. The use of silicon dioxide as the upper cladding of the device ensures its compatibility with the metal back-end-of-line process. By optimizing the structure parameters, a polarization conversion loss lower than 1 dB and cross talk larger than 27.6 dB can be obtained for TM-TE mode conversion over a wavelength range of 1450 to 1600 nm. For TE mode, the insertion loss is lower than 0.26 dB and cross talk is larger than 25.3 dB over the same wavelength range. The proposed device has good potential in diversifying the functionalities of the multilayer photonic chip with high integration density.

ObjectiveTo investigate a seasonal variation in blood pressure (BP) for patients undergoing hemodialysis (HD).MethodsIn this retrospective study, we exported all BP measurements from the information system to investigate a seasonal variation of BP. We also investigated a seasonal variation in BP for patients of different gender types, of different age groups, with diabetic nephropathy (DN), and with non-DN having HD. Multiple linear regression models were used to explore the associations between BP and climatic parameters.ResultsIn 2019, a total of 367 patients had received HD therapy in the Longwen HD unit. We included nearly 40,000 pre-dialysis BP measurements. The result of our study demonstrated a clear seasonal variation in pre-dialysis BP in general patients with HD, in male and female patients, and patients with DN and non-DN. December seemed to be a peak in the values of pre-dialysis systolic BP (SBP) and diastolic BP (DBP). The nadir values of pre-dialysis SBP and DBP were observed in June and July, respectively. A difference between peak and nadir values of BP is 3.81/2.20 mmHg in patients undergoing HD. Maximal seasonal variation in BP is 9.03/5.08 mmHg for patients with DN. A significant association of SBP and DBP with climatic parameters was found in this study. Pre-dialysis BP was inversely correlated with outdoor temperature, daytime length, and relative humidity.ConclusionA clear seasonal variation in BP is observed for patients with HD. Pre-dialysis SBP and DBP are inversely associated with outdoor temperature, daytime length, and relative humidity. The magnitude of a seasonal variation in BP increases in patients with DN.

Diabetes mellitus (DM) is a chronic metabolic disease with high morbidity and mortality. Recently, stem cell-based therapy for DM has shown considerable promise. Here, we undertook a systematic review and meta-analysis of published clinical studies to evaluate the efficacy and safety of stem cell therapy for both type 1 DM (T1DM) and type 2 DM (T2DM). The PubMed, Cochrane Central Register of Controlled Trials, EMBASE, and ClinicalTrials.gov databases were searched up to November 2018. We employed a fixed-effect model using 95% confidence intervals (CIs) when no statistically significant heterogeneity existed. Otherwise, a random-effects statistical model was used. Twenty-one studies met our inclusion criteria: ten T1DM studies including 226 patients and eleven T2DM studies including 386 patients. Stem cell therapy improved C-peptide levels (mean difference (MD), 0.41; 95% CI, 0.06 to 0.76) and glycosylated hemoglobin (HbA1c; MD, -3.46; 95% CI, -6.01 to -0.91) for T1DM patients. For T2DM patients, stem cell therapy improved C-peptide levels (MD, 0.33; 95% CI, 0.07 to 0.59), HbA1c (MD, -0.87; 95% CI, -1.37 to -0.37) and insulin requirements (MD, -35.76; 95% CI, -40.47 to -31.04). However, there was no significant change in fasting plasma glucose levels (MD, -0.52; 95% CI, -1.38 to 0.34). Subgroup analyses showed significant HbA1c and C-peptide improvements in patients with T1DM treated with bone marrow hematopoietic stem cells (BM-HSCs), while there was no significant change in the mesenchymal stem cell (MSC) group. In T2DM, HbA1c and insulin requirements decreased significantly after MSC transplantation, and insulin requirements and C-peptide levels were significantly improved after bone marrow mononuclear cell (BM-MNC) treatment. Stem cell therapy is a relatively safe and effective method for selected individuals with DM. The data showed that BM-HSCs are superior to MSCs in the treatment of T1DM. In T2DM, MSC and BM-MNC transplantation showed favorable therapeutic effects.

Nitrogen (N) uptake, as the first step of N metabolism, is a key limiting factor for plant growth. To understand the gene expression networks that control N absorption and metabolism in tea plants, we analyzed transcriptomes in the young roots of two groups of tea plants with significantly different growth rates under different N treatments (0, 0.2, and 2 mM). Using pairwise comparisons and weighted gene co-expression network analyses (WGCNA), we successfully constructed 16 co-expression modules. Among them, a specific module (turquoise) that substantially responded to the low N treatment was identified. Based on KEGG analysis, the relative genes that enriched in the "N metabolism" pathways were used to construct gene co-expression networks of N metabolism. Finally, a high-affinity ammonium (NH4+) transporter designated CsAMT1.2 was identified as a hub gene in the N metabolism network in tea plant roots and the gene expression could be highly induced by N resupply. The gene functional analysis revealed that CsAMT1.2 could make functional complementation of MEP1, MEP2, and MEP3 genes in 31019b yeast cells and improve NH4+ uptake rate in 31019b at low NH4+ level. Thus, CsAMT1.2 was a key gene controlling N uptake in tea plants and might play a vital role in promoting NH4+ uptake from the environment in tea roots. This study provided a useful foundation for improving the NUE in tea plantations.

The emergence of e-commerce has rejuvenated backward and poor remote villages. This paper presents a revelatory case study of three remote villages in China to depict how information and communication technology (ICT, specifically e-commerce) can empower remote areas to thrive. With the concept of affordance as basis, this work identifies critical actors in the development of rural e-commerce (i.e., Taobao Village) and illustrates how they use ICT. Eight deterministic affordances of the same ICT used in the evolution of rural e-commerce are specified, and three evolution paths about how ICT empowers the formation and development of rural e-commerce are depicted. This paper also describes the challenges of the current development of rural e-commerce. The findings of the study are discussed, and suggestions on how to capitalize on ICT for rural e-commerce development are offered.

Herein we describe a highly chemoselective A3-coupling/annulation of amino alcohols, formaldehyde, two kinds of aldehydes and alkynes, catalyzed by copper(II). This cascade reaction, employing readily available materials, provides a new and highly effective access to chiral N-propargyl oxazolidines with good diastereoselectivity (up to >20:1). In the case of ortho-substituted aromatic aldehydes, an intriguing steric effect is observed: a bulky group exhibits a remarkably adverse effect on the diastereoselectivity for the formation of the title molecule.

General Secretary Xi Jinping pointed out that community service “should adhere to center on people pinpoint the various needs of residents and timely provide services to community residents with precision and refinement.” In order to implement this important instruction, this article carried out a comprehensive survey of community services of Heping District. The results show that the community service highlights the new characteristics of “personalized demand, increasing demand for day care for the elderly, and the pursuit of a higher quality of life.” Based on the systematic analysis of the new characteristics, management countermeasures such as “adhering to the demand-oriented model of community service,” “improving the process management mechanism,” and “establishing an immediate response mechanism for new service demands” are proposed.

Corporate volunteering embodies the concept of social responsibility of enterprises, highlights the characteristics of enterprise support and employee contribution. Through literature research and field research, this paper further studies the current situation of corporate volunteering in China and summarizes achievements and shortcomings of corporate voluntary. In order to better play its functions and continue to play a greater role, it puts forward a core concept of corporate volunteering, and discusses its development during 13th Five-Year Plan and finally establishes innovative mechanisms from the reputation, tax and compensation.

QuEChERS pretreatment combined with gas chromatography-quadrupole time-of-flight mass spectrometry (GC-Q-TOF/MS) has been investigated for application in screening 244 pesticide residues in chilli. Fresh chilli samples were extracted with acetonitrile, and dried chilli samples were extracted using an acetonitrile/acetic acid (99∶1, v/v) mixture. The two extraction solvents were stored at -20 ℃. After salting out and cleaning by dispersive solid phase extraction (dSPE), heptachlor epoxide B was added as an internal standard, and the resulting residues were dissolved in 1.00 mL acetone. The dissolved sample solution was loaded onto an HP-5MS UI column (30 m×0.25 mm, 0.25 μm) and eluted by GC-Q-TOF/MS with a programmable temperature vaporizer and splitless injection in the full-scan mode. The compensation effects of the analytical protectant (AP) and matrix-matched calibration method on the matrix effect were established. AP could be used in the fresh chilli matrix to compensate for matrix effects, but it was not effective in the dried chilli matrix. The matrix-matched calibration method was effective in both matrices, which was selected for the quantification of pesticide residues in the samples. Because of the existence of the isomers of one compound and the same characteristic ions of different compounds, analyte detection was based on a flexible retention time deviation of ±0.25 min and accurate mass deviation of ±20×10 -6. Screening was performed by the software in the automatic matching mode. Compound identification and quantitation were based on a database and calibration curve established with reference materials. Suspicious samples were subjected to manual analysis. Quantitative analysis of 244 pesticide residues in fresh chilli and 222 pesticide residues in dried chilli was performed. The results showed that the developed database and method can provide a reference for the high-throughput screening and quantitation of fresh and dried chilli. Different levels of pesticides were added to the blank chilli samples, and the addition level corresponding to a signal-to-noise ratio (S/N) of 10 was used as the limit of quantification (LOQ). The LOQs of 44 pesticides with a maximum residue limit (MRL) ≤0.05 mg/kg in fresh chilli did not exceed 0.010 mg/kg. The linear ranges of these 44 pesticides were 0.01-1.00 mg/L. At spiked levels of the LOQ and 2.5 times the LOQ, the ratios of the 44 pesticides with recoveries of 60% to 120% were 88.64% and 100%, respectively. The LOQs of 200 pesticides with MRLs ≥0.05 mg/kg or without MRLs in fresh chilli did not exceed 0.025 mg/kg. The linear ranges of these 200 pesticides were 0.05-1.00. At spiked levels of the LOQ, twice the LOQ, and 10 times the LOQ, the ratios of the 200 pesticides with recoveries of 60% to 120% were 49.50%, 87.00%, and 89.50%, respectively. The linear correlation coefficients (r 2) of the 244 pesticides in fresh chilli were greater than 0.99. The LOQs of 222 pesticides in dried chilli were less than 0.15 mg/kg, and the linear ranges were 0.04-1.00 mg/L. The ratios of these 222 pesticides with r 2 greater than 0.99 was 95.46%. At spiked levels of the LOQ, twice the LOQ and 10 times the LOQ in dried chilli, the ratio of the 222 pesticides with recoveries of 60% to 120% were 72.52%, 73.42%, and 81.53%, respectively. The established screening and confirmation method was used to analyze 12 fresh chilli samples and 14 dried chilli samples. Eight pesticides were found in nine fresh chilli samples and three dried chilli samples, all of which were confirmed to be positive after manual identification. The concentrations of these pesticides were lower than the MRLs required by GB 2763-2019: National Food Safety Standard Maximum Residue Limits for Pesticides in Food. The results demonstrate that the established method is rapid, easy to execute, efficient, and reliable. It can be used for the high-throughput screening and quantitation of pesticide residues in fresh and dried chilli.

The rapid development of blockchain application technology promotes continuous exploration in the field of computer application science. Although it is still in the initial stage of development, the technical features of blockchain technology such as decentralization, identity verification, tamper resistance, data integrity, and security are regarded as excellent solutions to today's computer security technical problems. In this paper, we will analyze and compare blockchain data storage and cloud data processing technologies, focusing on the concept and technology of blockchain distributed data storage technology, and analyze and summarize the key issues. The results of this paper will provide a useful reference for the application and research of blockchain technology in cloud storage security.

The collection of digital information by governments, companies and individuals creates tremendous opportunities for knowledge and information-based decision-making. Driven by mutual benefit and laws and regulations, there is a need for data exchange and publication between all parties. However, data in its original form usually contains sensitive information about individuals and publishing such data would violate personal privacy. Privacy Protection Data Distribution (PPDP) provides methods and tools to release useful information while protecting data privacy. In recent years, PPDP has received extensive attention from the research community, and many solutions have been proposed for different data release scenarios. How to ensure the availability of data under the premise of protecting user privacy is the core problem to be solved in this field. This paper studies the existing achievements of privacy protection data release anonymity technology, focusing on the existing anonymity technology in three aspects of high-dimensional, high-deficiency, and complex relational data, and analyzes and summarizes them.

Industrialization in production is developing rapidly, and a large number of non-linear, impact and volatility loads are increasing in the power system, seriously affecting the power quality of the grid. The analysis and monitoring of power quality parameters can achieve the goal of improving power quality. This paper first introduces the influence of harmonics in power quality, and then deduces the acceleration principle of FFT algorithm for DFT acceleration algorithm. In MATLAB software, the FFT algorithm is used to verify the analog signal. Finally, experimental data shows that accurate harmonic parameters can be obtained by the FFT algorithm.

With the benefit fierce competition in the steel industry market in recent years, double cold reduction products have been developed towards strength improvement and thickness reduction. The traditional cold-rolling lubrication process with a fixed flow rate and concentration cannot solve the problems which cover uncontrollable shape of strip product and the excessive consumption of lubricating oil. Moreover,based on the analysis of the traditional direct application lubrication system of double cold reduction, a set of design scheme suitable for the emulsion pipeline direct mixing lubrication system of double cold reduction unit is proposed.The design complete the selection of key components which include the static mixer and atomization nozzle selection, pump and oil pump design selection, pipeline design selection, flow type selection, pressure gauge selection, electronic control cabinet design selection and other aspects. Equipment of the emulsion pipeline direct mixing lubrication system of double cold reduction has been developed. Comparing with characteristics of the traditional direct aplication lubrication system, the emulsion pipeline direct mixing lubrication system was better applied to the production practice of a 1220 double cold reduction mill. The consumption of ton of steel was reduced by 9.6%. The rolling energy consumption and oil consumption comprehensive costs decreased by 10.7%, and the strip steel section thickness difference was reduced by 19.3%. In addition, the plate shape quality defect rate decreased by 25.6 %, otherwise creating a large economic benefit for the unit and promoting the application value.

The gut is hypothesized to be the "motor" of critical illness and plays an important role in the development of sepsis. Berberine (BBR) is an alkaloid compound extracted from herbs, which has anti-inflammatory, anti-oxidative effects and can be used in intestinal infectious diseases and inflammatory bowel disease (IBD). BBR could promote differentiation of Treg cells which play a key role in maintaining intestinal immune homeostasis. However, its effect on sepsis-induced intestinal injury remains poorly understood. This study investigated the effect of BBR on cecal ligation and puncture (CLP)-induced intestinal injury and explained the underlying mechanism. These results showed that BBR treatment decreased the mortality of septic mice, alleviated intestinal injury and reduced serum endotoxin level; at the same time, BBR had a protective effect on CLP-induced lung and liver apoptosis. Meanwhile, BBR treatment increased the proportion of Treg cells and CTLA-4 in Treg cells. Treg cells from BBR treatment mice could decrease the pro-inflammatory response by inhibiting the activation of macrophages, thus exerting a protective effect on CLP-induced intestinal injury, and CTLA-4 mediated cell-cell contact pathway is required for this protective effect.
