OBJECTIVES Migraine is a headache disorder affecting approximately 12% of the population, predominantly female individuals. Migraine has been associated with vascular events such as stroke and cardiovascular disease. The close connection between these vascular disorders and atherosclerosis is well known. Carotid artery intima-media thickness (CAIMT) is a marker for detection of subclinical atherosclerosis. The present study is an analysis of the presence of subclinical atherosclerosis in migraine patients. METHODS CAIMT was evaluated in 25 female migraine patients and 27 female controls using innovative ultrasound (US) radiofrequency (RF) data technology. Hexane, ethyl acetate, methanol, and aqueous extracts of Ranunculus marginatus d'Urv. var. trachycarpus (Fisch. & Mey.) Azn. and R. sprunerianus Boiss. were tested in vitro for their antioxidant and antibacterial activities. Antioxidant activity of the extracts was determined by DPPH radical scavenging and Trolox equivalent antioxidant capacity assays. Methanol extracts showed the highest antioxidant activity in both assays. The total phenolics in the extracts were determined colorimetrically by using the Folin-Ciocalteau reagent. The total flavonoid content of the extracts was evaluated by a spectrophotometric method. The results obtained in the antioxidant activity tests were in positive correlation with the total phenolic and flavonoid contents of the extracts. An antibacterial activity analysis was carried out using paper disk diffusion and micro-well dilution techniques. All of the extracts displayed antibacterial activity against the tested bacteria in the disk diffusion method. The minimal inhibitory concentrations (MICs) of all the extracts of both Ranunculus species were found to be between 128 and 256 µg/mL.

Kanser, tüm dünyada ve ülkemizde öldürücülüğü oldukça yüksek ve sık rastlanılan bir hastalık haline gelmiştir. Kanser tedavilerinde kullanılan ilaçlar, sağlıklı hücrelere zarar vermeden hızla büyüme gösteren kanser hücrelerini yok etmeyi amaçlamaktadır. Her gün gelişmekte olan teknoloji ve bilim sayesinde kanser ilaçlarının daha az yan etkili ve tümör hücrelerinde daha yüksek başarı oranları bulunan alternatifleri aranmaktadır. 5-Florourasil (5-FU) meme, mide, pankreas, baş ve boyun gibi solid tümörlerin sistemik tedavisinde kullanılan ilk basamak ilaçlardan biridir. Triazol bileşiklerinin etkili biyoaktiviteleri ve kullanım kolaylıkları ile 5- FU‘nun kullanımındaki kısıtlamalar ve zorluklar düşünüldüğünde yeni oluşturulacak 5-FU şeker türevi içeren triazol bileşiklerinin kansere karşı istenilen nitelikleri göstermesi beklenmektedir. Çok geniş biyolojik etkinliğe sahip triazol içeren bileşikler olması nedeniyle bu güne kadar bir çok antikanser molekül tasarlanmış ve kullanılmıştır. Bu tez çalışmasında, yeni sentezlenen 5-FU içeren yeni triazol bileşiklerin sitotoksik, genotoksik, anti-anjiyogenik ve apoptotik özelliklerinin araştırılması ve bu bileşiklerin biyolojik etkinlik ve toksisite yönünden değerlendirilmesi amaçlanmıştır. Çalışma kapsamında 5-FU içeren yeni triazol bileşiklerinin, in vitro sitotoksik etkisinin belirlenmesi için MTT testi, apoptozun belirlenmesi için Flow Sitometri analizleri, genotoksik potansiyelin belirlenmesi için AMES testi ve anjiyogenik/antianjiyogenik etkiyi gözlemlemek için HET-CAM testi yapılmıştır. MTT testi sonrasında IC50 değerleri hesaplanan bileşiklerin pozitif kontrole göre daha düşük sitotoksik etkiye sahip olduğu ortaya konmuştur. Bu test sonucunda elde edilen değerler ile bileşiklerin 1xIC50 ve 2xIC50 değerleri hesaplanarak apoptotik aktivitesine bakılmış, sonuçlarda apoptoz yanı sıra ağırlıklı olarak nekroz tespit edilmiştir. AMES testi sonucunda, maksimum konstrasyon olan 5000 μg/mL de bile bileşiklerde herhangi bir genotoksik etki gözlenmezken, HET-CAM testi ile tüm konsantrasyon (4-2-1-0,5-0,25 μg/mL) denemelerinde bileşiklerin anti-anjiyogenik etki gösterdikleri belirlenmiştir. Sonuç olarak bu maddelerin daha az toksik ve aynı zamanda daha etkili oldukları yapılan in vitro çalışmalarla bulunmuştur. Daha ileri sonuçlar in vivo denemelerle desteklenmelidir., Cancer has become a very common disease with a high lethality the whole world also in our land. Drugs used in cancer treatments aim to destroy rapidly growing cancer cells without harming healthy cells. Thanks to developing science and technology are looking for new drug alternatives that have less side effects and higher success rates in tumor cells. 5-Fluorouracil (5-FU) which is one of the first-line drugs used in general treatment of especially solid tumors such as, head, stomach, pancreas, breast and neck. Considering the effective bioactivities and ease of use of triazole compounds and the limitations and difficulties in the use of 5-FU, it is expected that the newly formed triazole compounds containing 5-FU sugar derivatives will show the desired properties against cancer. The aim of the thesis is to investigate the cytotoxic, genotoxic, apoptotic and anti-angiogenic properties of newly synthesized triazole compounds containing 5-FU and to evaluate these compounds in terms of toxicity and biological effect. Within the scope of these study in new triazole compounds containing 5-FU, MTT test to determine the in vitro cytotoxic effect, Flow Cytometry analyzes to determine apoptosis, AMES test to determine genotoxic potential and HET-CAM test to observe angiogenic/antiangiogenic effects were performed. It was revealed that the compounds whose IC50 values were calculated after the MTT test had a lower cytotoxic effect than the positive control. The 1xIC50 and 2xIC50 values of the compounds were calculated with the values obtained in consequence of tests and the apoptotic activity was examined and apoptosis as well as predominantly necrosis were detected in the results. Conclusion of the AMES test, no genotoxic effect was observed in the compounds even at the maximum concentration of 5000 μg/mL effects were determined. It was determined that the compounds showed anti-angiogenic effects in all concentration (4-2-1-0.5-0.25 μg/mL) assays with the HET-CAM test. As a result, it has become found by in vitro assays which substances are less toxic and at the same time more effective. More precise results should be supported by in vivo assays.

Fitalatlar, temel olarak çok çeşitli ürünler ve uygulamalara esneklik, şeffaflık, dayanıklılık ve uzun ömür katması için plastikleştirici ajanlar olarak kullanılan fitalik asit esterleridir. Başlıca tibbi cihazlar, kozmetik ürünler, emülsifiye edici ajanlar, oyuncaklar ve yapıştırıcılarda kullanılmaktadırlar. Fitalatlar plastik madde ile kovalent olarak bağlanmadığından kolaylıkla çevreye yayılabilirler. Dimetil İzofitalat (DMIP) parfüm fiksatifi ve polyester reçineleri yapmak için plastikleştirici olarak kullanılmaktadır. DMIP'nin toksisitesi ile ilgili herhangi bir bilgi bulunmamaktadır. Bu çalışmada DMIP'nin in vitro genotoksik ve sitotoksik etkilerinin belirlenmesinin ardından, ex-vivo denemeler uygulanmış daha sonra in vivo akut ve subakut toksisite testi uygulanarak toksik ve nörotoksik etkilerinin incelenmesi amaçlanmıştır. Tek doz akut toksisite denemesi sonucunda hesaplanan LD50 değeri dikkate alınarak, 28 günlük tekrarlı doz toksisite denemesinde DMIP, 28 gün boyunca haftada 5 gün olacak şekilde aynı saatte farelere oral olarak uygulanmış ve deneme sonunda biyokimyasal ve histolojik parametreler incelenmiştir. Sonuç olarak bu tez çalışması ile elde edilmiş olan tüm toksisite verileri doğrultusunda, DMIP'nin sitotoksisite ve akut doz denemelerinde toksik olmadığı ancak AMES testi sonucu ile genotoksik olduğu, ex-vivo ve subakut toksisite denemelerine göre toksik olduğu ilk kez ortaya konmuştur., Phthalates are esters of phthalic acid and are mainly used as plasticizers in a wide variety of products and applications which are added to plastics to increase their flexibility, transparency, durability, and longevity. They are mainly used in medical devices, cosmetics, emulsifying agents, toys and adhesives. Phthalates are not covalently bound to the plastic material and can easily leach into the environment. Dimethyl Isophthalate (DMIP) is used as a perfume fixative and as a plasticizer to make polyester resins. There is no information on DMIP toxicity. In this study, in vitro genotoxic and cytotoxic effects of DMIP were determined, ex-vivo assays were performed and then toxic and neurotoxic effects were investigated by in vivo acute and subacute toxicity tests. After single dose acute toxicity test LD50 value is calculated and with this value we start to 28 days subacute toxicity test with DMIP which is orally administered to mice 28 days, 5 days per week at the same time each day and at the end of the experiment biochemical and histological parameters were examined. In conclusion, with the toxicity data obtained from this study it is for the first time revealed that DMIP has not been found toxic in cytotoxicity and acute dose assays but it has been found to be a genotoxic substance that by AMES test results and a toxic substance in ex-vivo and subacute toxicity assays.

Bu çalışma, aspir olarak bilinen Carthamus tinctorius L. bitki tohumlarından elde edilen aspir yağı (SFO) ve içeriğinde yer alan oleik asit (OLA) ve konjugatif linoleik asit (CLA) 10E-12Z (trans10, cis12) olarak adlandırılan türevlerinin in vitro pro-apoptotik ve mutajenik etkinliğini ortaya koymak, terapötik etkilerini belirlemek ve moleküler etki mekanizmalarını saptamak amacıyla hazırlanmıştır. SFO, OLA ve CLA'nın kanser hücre hatları U-87-MG, A549, MCF-7, Caco-2, HeLa ve PC-3 maruziyeti sonucunda, her bir maddenin sitotoksisitesi ve mutajenik etkinliği belirlenmiştir. SFO'nun sitotoksik konsantrasyonunun Caco-2, HeLa ve A549 hücrelerini hangi apoptotik mekanizma ile öldürüldüğü, gen ekspresyon düzeyinde saptanmıştır. Analiz sonunda elde edilen veriler, aspir yağının kanser tedavisinde alternatif bir farmasötik olarak güvenli kullanılabilmesi için yapılması gereken çalışmaların ilk adımını oluşturmuştur.

Bu çalışmada Aliağa Körfezi'ndeki kirliliğin canlılara etkisini belirlemek amacıyla Aliağa ve Foça istasyonlarından önemli bir sucul biyomonitor organizma olan Mytilus galloprovincialis örneklemesi yapılmıştır. Disekte edilen solungaç ve hepatopankreas dokularında ağır metal maruziyeti, biyokimyasal ve sitogenetik biyomarkerlar incelenmiş ve apoptotik süreç izlenmiştir. Analiz sonunda elde edilen verilerle Aliağa'daki kirliliğin canlıların üzerinde yarattığı oksidatif stres belirlenmiştir. Bununla beraber apoptoz mekanizmalarını da indüklediği ortaya konmuştur In this study, in order to determine the effects of pollution on biota in the Aliaga Bay, Mytilus galloprovincialis, an important aquatic biomonitoring organisms samplings are done from Aliaga and Foca stations.Heavy metal exposure, biochemical and cytogenetic biomarkers were examined and apoptotic process was monitored in dissected gill and hepatopancreas tissues. Fitalatlar çok yayın olarak kullanılan plastikleştiricilerin büyük bir sınıfını oluşturmaktadır. Birçok fitalat sınıfının erkeklerde gelişimin eşeysel farlılaşma periyodunda androjen üretimini baskılayarak fetal endokrin toksisitesi ve postnatal reprodüktif malformasyonlar oluşturduğu bilinmektedir. Fitalatlar, östrojenik özelliklere sahip birçok kimyasal sınıfından yalnızca biridir. Bu çalışmada ise, daha önce hiçbir toksikolojik verisi ortaya konmamış bir fitalat olan butil siklohekzil fitalatın (BCP) erkek reprodüktif sistemi üzerindeki toksik etkileri ortaya konmuştur.BCP' nin farklı konsantrasyonları (100 mg/ kg, 200 mg/ kg and 400 mg/ kg) ve kontrol grubu (0.1 ml % 0.9 serum fizyolojik/fare), oral olarak 28 gün boyunca ferkek farelere uygulanmıştır. Çalışmamız sonunda, testis ve cauda epididimis örnekleri alınmıştır. BCP' nin farklı dozları uygulanan farelerin final total vücut ağırlıkları, kontrole oranla ve başlangıç ağırlıklarına oranla herhangi değişiklik göstermemiştir. Yine tüm uygulama gruplarında bulunan fareler, testis ve cauda epididimis ağırlıkları bakımından ve rölatif organ ağırlığı bakımından kontrole göre ve kendi aralarında herhangi bir önemli farklılık göstermemiştir. Testislerde yapılan histopatolojik incelemelerde, BCP, tubuli seminiferi contorti' lerde doz artışına bağlı olarak şekil bozukluğu, intersitisyumda ödem, defektif tubul ve spermatojenik hücre serilerinde dejenerasyona sebep olmuştur. Ayrıca, cauda epididimisde doz artışına bağlı olarak sperm sayısında ve canlılığında azalma kaydedilmiştir. Bununla birlikte BCP testislerde lipit peroksidasyonuna sebep olmamaktadır. Sonuç olarak Butil siklohekzil fitalatın erkek reprodüktif sistem üzerinde toksik etkilere sebep olduğu belirlenmiştir.Anahtar sözcükler: Butil siklohekzil fitalat, erkek fare, testis, sperm morfolojisi Phthalates constitute a large class of plasticizer compounds that are widely used for many consumer product applications. More members of the phthalate class of compounds are known to induce male fetal endocrine toxicity and postnatal reproductive malformations by disrupting androgen production during the sexual differentiation period of development. Phthalates are just one of the many classes of chemicals that have been implicated as having estrogenic properties. In this project, the determination of toxic effects on male reproductive system of Buthyl cyclohexil phthalates (BCP) which has not been studied previously were aimed.Three different concentrations of the BCP (100 mg/ kg, 200 mg/ kg and 400 mg/ kg) and a control group (0.1 ml % 0.9 saline per mouse) were designed for the experiment. The BCP was orally applied for 28 day. At the end of experiment, testes and cauda epididymis samples were taken from mice. Body weights of animals were showed no significant differences between treatment groups and control. There were no significant differences in the relative weights of testes and cauda epididymis in any of the experiment groups.In histological examinations, BCP caused a dose-dependent increase on histopathological changes in testes of mice such as degenerations in tubuli seminifer contorties, edema in some area of intersitisium, defective tubule and degenerations in spermatogenetic cell lines. Also, in cauda epididymides as increasing doses there are significant decreases in sperm count and motility. But, BCP caused no lipit peroxidation on testes.We can conclude that, Buthyl cyclohexil phthalates has toxic effects on male reproductive system.Keywords: Butyl cyclohexyl phthalate, mal emice, testes, sperm morphology 64

Anemi, eritrosit sayı ve/veya hemoglobin düzeyinin sağlıklı kişilerdeki normal değerlerin altına inmesidir. Demir eksikliği anemisi; vücuda giren demirin hemoglobin yapımı için yetersiz kalması şeklinde tanımlanabilir. Demir eksikliği anemisi özellikle gelişmekte olan ülkeler başta olmak üzere tüm dünyada en sık rastlanan hematolojik problemdir ve sadece hematolojik sistem üzerine değil, tüm sistemler üzerine etkileri bulunmaktadır. Aneminin hücresel fonksiyonlardaki biyokimyasal değişiklikler, büyüme, psikomotor gelişme, davranış, zihinsel gelişme, immün sistem, fiziksel kapasite, gastrointestinal sistem, termoregülasyon sistem üzerine etkileri önemlidir. Çalışma bölgesinde kayıtlı bebeklerin hematokrit değerlerinden yararlanarak, birçok hastalıkla birlikte ilerleyen aneminin sıklığını saptamak, birinci basamak sağlık hizmetlerinde anemi taraması sonucunda elde edilecek kazanımları görmek için bu çalışma planlanmıştır. Çalışmamıza 1998-2001 yılları arasında Kemalpaşa bölgesinde doğan 1775 bebek dahil edilmiştir. Bu bebeklerden 602'sinin hemogramı incelenmiştir. Bu bebeklerin %52.5'sı erkek, %47.5'i kızdır. %42.5, bebekte anemi görülmüş olup, %38 bebek anemi sınırında, %19.5 bebekte ise anemi görülmemiştir. Yaş gruplarına göre anemi görülme oranı 12-24 ay arasında, 6-12 aya göre daha yüksektir. Cinsiyetlere bakıldığında erkek bebeklerin % 39.5'inde; kız bebeklerin 45.8'inde anemi vardır. Sonuç olarak, yaptığımız bu retrospektif çalışmada anemi toplumumuzda çok sık rastlanan bir hastalıktır. Demir eksikliği anemisinin bebeklerde rastlanma oranı yüksek olarak saptanmıştır. Basit ve ucuz yöntemlerle bu hastalığın tanısı konabilmektedir ve başlanacak bir demir replasmanı ile bu hastalık önlenebilir.
