33. Influence of stress phytohormones on the stability of callus and potato tuber to the pattern of phytophtorosis.

Abstract

We studied the effect of signaling molecules – stress phytohormones of salicylic (SA) and jasmonic (JA) acids, on the resistance of calluses and potato tubers to late blight pathogen – oomycete Phytophthora infestans (Mont.) De Bary and the pathogen's ability to synthesize and secrete proteolytic enzymes that cause it properties. The introduction of SA and JA into the callus cultivation medium inhibited the growth of P. infestans and altered the mycelium morphology. Microscopic observations showed that infection of potato tubers led to the appearance of dead (necrotic) cells. So, 48 hours after inoculation, there was 24.6 ± 1.1% of dead cells in the tubers of the susceptible cultivar, 9.8 ± 0.9% in the tubers of the stable cultivar, and only 17.2 ± 0.7 and 16.3 ± 0.8% in the susceptible cultivars pretreated with SA and JA. It is shown that oomycete P. infestans produces mainly serine proteases with a maximum level of activity at neutral and slightly alkaline pH, and a temperature optimum of 50 °C. Based on the analysis of substrate specificity, they were assigned to trypsin-like proteases. The treatment with SA and JA significantly increased the transcriptional activity of the serine protease inhibitor gene in both healthy and late blighted potato tubers. The data obtained suggest a wide adaptive plasticity of the late blight pathogen, as well as prospects for the development and use of new antimicrobial biological products in combination with stress phytohormones to protect potatoes from P. infestans. Abstract

The effect of chitooligosaccharides (ChOSs) with an acetylation degree (AD) of 65% and 30% on the formation of defense reactions of potato plants (Solanum tuberosum L.) against Phytophthora infestans (Mont.) De Bary is studied. The content of hydrogen peroxide, the activities of peroxidases, allene oxide synthase, and hydrolase inhibitors, and the transcriptional rate of pathogenesis-related (PR) proteins are studied. ChOSs with an AD of 65% reduced the degree of late blight symptoms on the leaves of potato plants via the stimulation of H2O2 accumulation and the transcriptional activity of genes encoding peroxidase (M21334), chitinase (U49969.1), and inhibitors of amylase (XM006351484) and proteinase (JX683427). It is found that ChOSs with an AD of 30% induced the activity of anionic isoperoxidases with a pI of ~3.5-4.5 in infected plants. The mechanisms of potato resistance to Phytophthora infestans formed under the influence of different ChOSs are discussed. [ABSTRACT FROM AUTHOR]

The authors conclude that effective reforms regarding the system of Russian higher education, mentioned in the new Federal state educational standards and international documents, signed by Russia suppose that the changes in of the psycho-pedagogical basis of education occur as well.

Abstract

These data demonstrate the successful development of protective responses in treated with metabolites plants that is an effect of induced resistance of the plant upon contact with the pathogen elicitors.

Abstract

We studied the effect of two differing for aggressive strains of the fungus S. nodorum on the activity and expression of the oxidoreductases in the leaves of T. aestivum L. It is shown that the expression of oxalate oxidase was higher in plants infected with highly aggressive strains, and expression of the peroxidase higher when infected with strain weak aggressive. Abstract

The article examines the problem of digital education in high school in terms of its psychological risks and effects. The aim of the work is to identify the psychological effects of the transformation of higher education in the digital age and to determine the priority trends of psychological support in the digital educational process. In the theoretical aspect of this study the analysis of the problem of psychological risks and challenges of digital education in high school is presented on the basis of domestic and foreign experience of transition to a remote format of education. Methods of theoretical analysis of modern sources on this problem were applied, in particular, cognitive and personal-developing approach to professional and professional-pedagogical education. The author refers to the separate articles on digital education in high school. The work analyzes the sociopsychological aspects of digital education reflecting the person-oriented system risks, communicative competence risks, individual psychological security risks and a new type of students in the digital age. The article reveals only some of the psychological effects of digitalization discussed in studies on this issue. As a result, the priority trends of digital education psychological effects in high school were identified.
