Objective and Method: A new series of benzothiazole-piperazine derivatives was synthesized and a complete chemical characterization of the novel compounds was provided. In vitro cytotoxic activities were screened against colorectal (HCT-116), breast (MCF-7) and hepatocellular (Huh7) cancer cell lines by Sulforhodamine B assay. Result and Discussion: All compounds showed cytotoxic activity against hepatocellular (Huh7) and breast (MCF-7) cancer cell lines. Dihalo substituted benzylpiperazine derivatives (2a, 2e) had the highest cytotoxic activities in all the tested cell lines. In addition, acetylcholinesterase (AChE) and butyrylcholinesterase (BuChE) inhibitory activities of synthesized compounds were investigated by in vitro Ellman’s method. Compound 2j led to moderate and selective inhibition against AChE. Docking study was utilized to understand the binding mode of compound 2j in comparision with donepezil on AChE. The other tested compounds showed weak or no inhibition against AChE as promising anticancer agents. © 2017 Bentham Science Publishers. The aim of this study was to evaluate the plasma concentration of insulin like growth factor-2 (IGF-2) and its expression in tumor tissue of dogs with malignant mammary tumors. Fifteen dogs were used in the study. Eight dogs with mammary tumors constituted group 1, and 7 healthy dogs constituted group 2. Diestrus phase in dogs was determined with vaginal cytological examination. Metastasis was evaluated by ultrasonographic examination of abdomen and thorax radiography. Blood samples were collected from the dogs, plasma was separated and IGF-2 levels were measured with ELISA method. Tumors in group 1 were classified according to Clinical Grading System defined by World Health Organization and were removed by using complete unilateral mastectomy technique. The histological malignancy grading of tumors was made according tubule formation, nuclear pleomorphism, and number of mitotic cells by using Notthingham method. In histopathological examination, 4 carcinosarcoma, 1 intraductal papillary carcinoma, 1 carcinoma, 1 complex type carcinoma, and 1 mix type carcinoma were detected. In immunohistochemical investigation, IGF-2 expression was detected in mammary acinar epithelial cells, myoepithelial cells, and some vascular smooth muscle cells but not in ductal epithelial cells in tumor tissue. /0000-0002-0636-4214 WOS: 000268128600005 PubMed: 25903514 This randomized study was designed to establish the efficacy of platelet-rich plasma (PRP) gel and topical estradiol alone, or in combination, on healing of full-thickness wounds created on the trunk of rabbits. Fifteen New Zealand white female rabbits and 60 wounds were studied. Four 2.25-cm(2) full-thickness wounds were created using a template and treatments including a sterile saline solution that were assigned randomly to the wounds. Wounds were bandaged, dressed intermittently, and assessed by wound measurements and a collection, of samples at 7, 14, and 21 days to evaluate healing. Variables of interest were hydroxyproline concentration and scored gross and microscopic morphologic characteristics reflective of wound healing. PRP gel + estradiol-treated wounds completely healed faster than wounds given other treatments. Granulation tissue growth to the skin level was faster (P < 0.05) in the PRP gel + estradiol group than the other groups. Only the PRP gel + estradiol group had a significantly lower collagen level than the sterile saline solutions on day 7 (P < 0.05). The PRP gel + estradiol group and estradiol group led to an increase in angiogenesis on day 14 (P < 0.05). Complete epithelization was observed only in the PRP gel + estradiol group compared with the other groups on day 7. PRP gel + estradiol-treatment enhanced healing in full-thickness wounds by reducing the contraction rate with a trend toward accelerating both epithelial migration and the angiogenic response.

YILDIZ, Kader/0000-0001-5802-6156; /0000-0002-0636-4214 WOS: 000251604000001 An Anatolian wild sheep (Ovis orientalis anatolica) from a wild ruminant farm near Kirikkale, Turkey, was necropsied. Parasitic nodules were situated beneath the pleura of the caudodorsal lobes of the lungs. First stage larvae of Cystocaulus ocreatus were detected both in bronchial fluid and in lung paraffin sections. Clinical and parasitological examinations of the other animals (4 dwarf goats and 3 wild sheep reared on the same farm) were also performed. First stage larvae of C. ocreatus, and eggs of Nematodirus spp. and Strongyloides spp. were observed in coproscopic examination. Anthelmintic treatment, Rabenzole (R) (rafoxanide and thiabendazole, Topkim), was administered to the animals infected by parasites. At the end of the treatment the combination of rafoxanide and thiabendazole was found to be markedly effective for the treatment of lungworm infection and no parasitic eggs were detected in the faeces of any of the treated animals.

The present study was conducted to immunolocalize 3?-hydroxysteroid dehydrogenase (3?-HSD), an enzyme metabolizing pregnenolone to progesterone in the choroid plexus of the lateral ventricle in sheep, as well as to measure progesterone concentration in cerebrospinal fluid (CSF) and plasma using radioimmunoassay (RIA). Akkaraman breed rams (n = 16) and ewes (n = 16) were utilized in the study. 3?-HSD was immunolocalized in choroid epithelial cells of the choroid plexus with an apparent cytoplasmic immunoreactivity. Progesterone was detected in CSF with no significant differences between the ewes (0.76 ± 0.14 ng/mL) and rams (0.74 ± 0.13 ng/mL) (p > 0.05). However, the plasma progesterone concentration in the ewes (0.27 ± 0.04 ng/mL) was significantly higher than that of the rams (0.11 ± 0.02 ng/mL) (p < 0.001). Consequently, CSF in sheep contains progesterone in significant levels. As evidenced by 3?-HSD immunoreactivity, choroid epithelial cells may be a site of progesterone synthesis in sheep.

/0000-0002-0636-4214 WOS: 000227165300003 The present study investigated the immunolocalization of CD 14 and the bacterial antigen of Pasteurella multocida serotype A :3 in normal and pneumonic rabbit lungs. To induce pneumonia, P. multocida serotype A :3 (4 x 10(6) CFU/ml) was inoculated intra-tracheally to twelve New Zealand White rabbits (Oryctolagus cuniculus) at age of 8-10 weeks while four rabbits received physiologic saline with the same route. Nine infected rabbits died with clinical sings of the disease between the 3(rd) and 13(th) days post inoculation. On day 14, the remaining three infected and control rabbits were euthanatized. The histopathological evaluation of the lung samples collected from nine infected rabbits with clinical symptoms revealed severe cases of fibrinopurulent and necrotic pneumonia and fibrinopurulent pleuritis. The bacterial antigen was immunolocalized in macrophages and degenerating leukocytes around the necrotic tissue and in exudates occupying the lumens of bronchia, bronchioles, and alveoli. CD14 immunoreactivity was present in alveolar macrophages, neutrophiles, alveolar epithelial cell surfaces, and lumens of the capillaries of the pneumonic lungs. In three rabbits, inoculated but presenting no clinical symptoms and no histopathological lesions, the bacterial antigen was found in the alveolar wall; however, like in the controls, CD14 immunoreactivity was present only in the lumen of capillaries. Double labelling immunohistochemistry indicated that CD14 was closely associated with the bacterial antigen in inflamed regions of the lung. Results suggested that interaction of the bacterial antigens and CD14 may have a critical role in lung pathologies resulting from P. multocida serotype A :3 infection.

/0000-0002-0636-4214 WOS: 000223398200003 This study was focused on serum and cerebrospinal fluid (CSF) biochemical changes in experimental Listeria monocytogenes infection in rabbits. Total of 20 New Zealand White rabbits were used. 14 New Zealand White rabbits were inoculated with Listeria monocytogenes 1/2a (L.m. 1/2a) by intracerebral way and 6 rabbits served as controls. Ten hours after the inoculation of the bacteria the rabbits developed symptoms of meningitis, with stiffness of the neck and fever. The existence and the severity of the inflammatory reaction were controlled by microscopic observation. The presence of bacteria into the cytoplasm of neutrophils and macrophages localised in inflammatory areas was confirmed by immunohistochemical analysis using rabbit polyclonal antibodies. We have analysed in serum and cerebrospinal fluid (CSF) the concentrations of glucose, total protein, and the enzymatic activities of aspartate aminotransferase (ASAT), alanine aminotransferase (ALAT) and lactate dehydrogenase (LDH). After intracerebral inoculation of bacteria the glucose concentrations in the serum and CSF decreased to 428.69 +/- 5.50 mg/l and 309.56 +/- 3.93 mg/l (mean +/- SE) respectively compared to control values of 975.33 +/- 84.34 mg/l and 689.22 +/- 65.13 mg/l (p less than or equal to 0.001). Total protein contents, the activities ofASAT, ALAT and LDH in the serum of infected animals increased to 98.92 +/- 1.97 g/l, 327.87 +/- 22.18 U/l, 292.39 +/- 4.66 U/l, 153.28 +/- 3.04 U/l respectively compared to 64.42 +/- 1.14 g/l, 67.91 +/- 7.79 U/l, 74.25 +/- 1.79 U/l, 49.35 +/- 1.20 U/l in control rabbits. In CSF, increases in total protein concentrations and in enzyme activities were also evidenced in inoculated rabbits. Moreover, serum and CSF LDH activities were correlated with brain injury (p < 0.05).

Bozkurt, Ayhan/0000-0002-5794-709X; Bilge, S.Sirri/0000-0003-2878-6968 WOS: 000296672200006 In this study, synthesis of a series of 3,6-diaryl-1,2,4-triazolo[3,4-b]-1,3,4-thiadiazines by the condensation of appropriate 5-(3,4-dichlorophenyl)-2-(aroylmethyl)thio-1,3,4-oxadiazoles and hydrazine hydrate in acetic acid is described. The structures of newly synthesized compounds were confirmed by IR, H-1-NMR and elemental analysis. All the compounds were evaluated for anti-inflammatory activity by determining their ability to provide protection against carrageenan-induced edema in rat paw. In addition, ulcerogenic activities of the compounds were determined. The anti-inflammatory data scoring showed that most of the compounds exhibited anti-inflamatory activity profile at 100 mg/kg.

/0000-0002-0636-4214 WOS: 000221704400004 The present study examined the distribution of Listeria monocytogenes 1/2a (L.m. 1/2a) and the antigen CD14 by using immunohistochemical methods in the brain of healthy and experimentally infected rabbits with L.m. 1/2a. The ten New Zealand White rabbits were inoculated by intracerebral route, and 4 rabbits were used for control. Only the brains of the animals were removed during necropsy, and they were analysed by conventional histological techniques. By Haematoxylin and Eosin staining all L.m. 1/2a inoculated rabbits presented meningitis or meningoencephalitis. In healthy rabbits, the immunohistochemical staining of brains for CD14 was very slight and seen only in the lumen of the vessels, whereas, in infected rabbits the CD14 immunoreactivity was more prominent especially in inflammation areas of parenchyma and meninges. Moreover, the CD14 positive immunostainings were seen massively in L.m. positive inflamed areas. The present study provides the immunohistochemical expression of CD14 and L.monocytogenes antigen in the brain and a positive correlation was obtained between CD14 and this infectious agent.

Estrogens have been implicated in neurophysiology and neuropathology. The present study investigated the presence of aromatase immuno reactivity in the temporal, parietal and occipital cortices, olfactory bulb, cerebellum, and choroid plexus of the normal dog. Aromatase immunoreactivity was localized exclusively in neurons in the cortices and olfactory bulb. Immunoreactivity was also present in a small number of astrocytes in the substantia alba of the cerebellum. In the cortical regions, immunoreactive neurons, morphologically identified as pyramidal cells, were found throughout Layer 11 down to Layer VI, but not all pyramidal neurons were immunoreactive. In the olfactory bulb, immunoreactive neurons were mainly observed in mitral cells and inner granular cell layers. In the cerebellum, immunoreactivity was present in neurons of the deep cerebellar nuclei and in some neurons of the molecular and granular cell layers. Immunoreactivity was also present in endothelial cells of the subarachnoid vessels and those adjacent to ventricles in the cortex. The presence of well defined cytoplasmic aromatase immunoreactivity in neurons, some astrocytes, and endothelial cells suggests estrogen involvement in CNS physiology and function in the dog. The presence of aromatase in ependymal cells lining cerebral ventricles and choroid epithelial cells suggests that these cells may be partially responsible for estrogen concentration in the cerebrospinal fluid.

To evaluate the effects of three types of honey (chestnut, blossom, and rhododendron) on the healing of full-thickness wounds. Methods. Twenty-four (24) New Zealand White female rabbits were used. Four 1.5 cm x 1.5 cm full-thickness skin wounds were created on the back of each animal and treated with pure honey or sterile saline, respectively. Wounds were assessed by wound measurements and collection of samples at 7, 14, and 21 days post wounding to evaluate the healing process. Variables of interest were hydroxyproline concentration and gross and microscopic morphological characteristics reflective of wound healing. Wounds of the honey-treated groups healed much faster than the control group. Results. On day 7, the formation of granulation tissue, epithelization, angiogenesis, and fibroplasia levels increased in the honey-treated groups (P < 0.05). A statistical difference between the honeys was not detected. Conclusion. The present results suggest that honey accelerates the inflammatory reaction and initiates healing early on in the treatment process.
