Oral cerrahi girişimleri sonrası kanama komplikasyonlarını kontrol altına almak için ticari olarak mevcut pek çok hemostatik ajan kullanılmaktadır. Bu çalışmada, üç farklı lokal hemostatik ajanın erken dönem doku iyileşmesi üzerindeki etkileri histolojik ve sitolojik olarak değerlendirildi. Çalışmaya yarı gömük üçüncü molar diş çekimi yapılan ve çalışma kriterlerine uygun bulunan 41 gönüllü hasta dahil edildi. İşlem sonrası kullanılan hemostatik ajana göre hastalar; oral cerrahi operasyonu sonrası herhangi bir ajan kullanılmayan grup,grup I (kontrol, n=10); ile sırasıyla jelatin, kollajen ve kitosan esaslı lokal hemostatik ajan kullanılan gruplar, grup II (n=11), grup III (n=10), grup IV (n=10) olarak dört gruba ayrıldı. Cerrahi işlem sırasında ve 8. günde doku örnekleri alındı. Histolojik inceleme için her gruptaki hastalara ait dokulardan alınan 5 μm kalınlığındaki kesitlere Masson’s trichrom boyaması yapılarak ışık mikroskopisi altında incelendi. Kesitler doku harabiyeti vazokonjesyon, lökosit infiltrasyonu ve kollajen lif yapısı kriterleri kullanılanarak değerlendirildi. Sitolojik inceleme için dokulardan alınan sürüntü örnekleri May-Grünward ile boyandı. Çalışmamızda kullanılan lokal hemostatik ajanların klinik etkinlikleri yeterli bulundu, kitozan esaslı lokal hemostatik ajanın diğerlerine göre doku iyileşmesini sitolojik ve histolojik olarak daha olumlu yönde etkilediği saptandı. Klinik yararları göz önüne alındığında birçok cerrahi alanda kullanımı yaygın olan bu ajanların doku iyileşme potansiyeline etkilerinin artırılması için konuyu farklı yönlerden inceleyen çalışmaların gerekli olduğu kanısına varıldı.Anahtar kelimeler: Hemostatik ajanlar, jelatin sünger, kollajen, kitozan

Konu: Monoklonal antikorlar (mAb’lar), modifiye edilmiş proteinlerdir ve düzensiz olan sinyal iletim yolunun belirli bir bölümünü hedefleyerek insan vücudunda gerçekleşen immünolojik süreçlere müdahale ederler. Kanser tedavisi için saf mAb’lara ihtiyaç vardır. Amaç: Bu çalışmanın amacı, mAb’ların saflaştırmasını Protein A kromatografisi, katyon değişim/anyon değişim kromatografisi veya çoklu mod kromatografi ile gerçekleştirmektir. Yöntem: Protein A rezini ile olan saflaştırma, mAb’ın Fc bölgesi ile kolon matriksine immobilize edilmiş Protein A ligandları arasındaki spesifik bağlanma ile gerçekleştirildi. Çoklu mod kromatografi rezinleri kullanılarak yapılan saflaştırmada ise iyonik ve hidrofobik etkileşimleri aynı anda gerçekleştirebilecek gruplara sahip olması nedeniyle saflaştırma rezini ile mAb arasındaki çoklu bağlanma etkileşimleri ile gerçekleştirildi. Bulgular: Protein A saflaştırma basamağı ile elde edilen mAb ürününün farklı bir saflaştırma basamağı ile de elde edilebileceği gösterildi. Her iki saflaştırma basamağı sonrası elde edilen ürünlerin HPLC yöntemleri ile kalite profillerinin birbirine benzer olduğu saptandı. Sonuç: İki farklı saflaştırma yöntemi ile benzer mAb ürününün elde edilebileceği gösterildi. Protein A rezini ile gerçekleştirilen saflaştırma basamağının %Verim değeri diğer saflaştırma basamağına göre bir miktar daha fazlaydı. Bunun sebebi, Protein A rezininin IgG’nin Fc bölgesine spesifik bağlanma özelliği göstermesinden dolayı idi. Objective: The aims of the present study were to investigate the proteomic profile of nifedipine induced overgrown gingiva and compare with non-overgrown gingival tissues obtained from the same patients. Methods: Seven subjects under nifedipine medication for at least 6 months and diagnosed as nifedipine induced gingival overgrowth (NIGO) participated in the study. Periodontal clinical parameters were recorded. Gingival tissue samples were harvested from overgrown (GO+ Group, n=7) and non-overgrown regions (GO- Group, n=7) of the same patients. Proteomics was performed using Liquid Chromatography-Tandem Mass Spectrometry (LC-MS/MS) technique. The identified proteins were further classified according to their molecular functions, biological processes and cellular component distribution for functional gene ontology analysis using a web-based bioinformatics tool. Metanol alkollerin en basit üyesidir. Ağızdan, soluma yoluyla veya deri yoluyla çok küçük miktarlarda alınsa dahi ciddi zehirlenmelere neden olabilir. Bu çalışmada, taze sıkılmış ve ticari portakal suyunda metanol konsantrasyonları gaz kromatografi yöntemi ile belirlenmiş ve farklı saklama koşullarına bağlı olarak alkol oluşumu izlenmiştir. 0, 1, 7, 14, 30 ve 90. günlerde oda sıcaklığında, buzdolabında ve derin dondurucuda saklanan taze sıkılmış portakal suyundaki metanol değerleri 4-12 mg/dL civarındaydı. Ancak ticari portakal suyunda metanol her koşulda tespit edilememiştir. Bunun nedeni, ticari portakal suyunda bulunan alkol fermantasyonunu önleyen katkı maddeleri olabilir. Bu çalışmada taze portakal suyunda tespit edilen metanol değerleri, kandaki minimum öldürücü metanol konsantrasyonunun (40 mg/dL) altındaydı. Toksik limit göz önüne alındığında, arka arkaya çok miktarda taze portakal suyu tüketmek uygun olmayabilir. Depolama koşullarına göre ticari portakal suyunda metanol oluşmaması elbette iyidir. The aim of our study was to examine the effects of exercise and calorie_x000D_ restriction on various tissue damage and antioxidant parameters_x000D_ in the brain and testis of rats in a natural aging model. For this_x000D_ purpose, male Sprague-Dawley rats were the control group (C),_x000D_ the elderly (A), the elderly with calorie restriction (ACR), the elderly_x000D_ who were exercised (AE) and the elderly who were exercised with_x000D_ calorie restriction (ACRE), they were divided into 5 groups. The control_x000D_ group was composed of three-month-old animals. The other_x000D_ group consisted of 15-month-old rats. Exercise and calorie restriction_x000D_ were applied for 6 weeks. At the end of the experiment, lipid_x000D_ peroxidation (LPO), nitric oxide (NO), glutathione (GSH) levels and_x000D_ superoxide dismutase (SOD), catalase (CAT), glutathione-S-transferase_x000D_ (GST) and tissue factor (TF) were determined in brain and_x000D_ testicular tissues homogenates. As a result of the study, the A_x000D_ group’s brain and testis LPO, NO levels and TF activity increased,_x000D_ GSH levels and SOD, CAT and GST activities decreased, when compared_x000D_ to the C group. As a result of our study, an increase in oxidant_x000D_ damage was observed with TF activity in the brain and testis in the_x000D_ natural aging model, and positive effects of exercise and calorie_x000D_ restriction on the antioxidant levels in the brain were determined,_x000D_ especially in aging. Objective: The aim of this in vitro study was to investigate the fluoride-releasing/recharging and antimicrobial effects of restorative materialsused in pediatric dentistry.Material and Method: High-viscosity and resin-modified glass-ionomercement, glass carbomer, compomer, giomer and composite weretested. Dental materials’ fluoride-release was measured in distilled water(DW) and artificial saliva (AS) for eight weeks using a fluoride-specificelectrode. In the development of diabetic complications, oxidative stresscaused by hyperglycemia plays an important role. Deuterium depletionhas protective effect against cellular damage and tumorsuppressor effect in cancer cell lines. The purpose of this studywas to examine and evaluate the effect of deuterium depletion onerythrocyte oxidant and antioxidant system parameters in experimentaldiabetes induced by streptozotocin (STZ) in rats. On the30th or 40th day after STZ injection, blood samples were taken andblood glucose levels were measured. Erythrocyte hemolysateswere also prepared. Lipid peroxidation and glutathione levels andglutathione-S-transferase, superoxide dismutase and catalase enzymeactivities of erythrocytes were determined. At the 30 or 40day periods, deuterium depleted water (DDW) consumption haddifferently affected oxidant-antioxidant parameters of erythrocytes.The clear effect of deuterium depletion in drinking waterfrom 150 ppm to 85 ppm was observed in glutathione levels andit increased erythrocyte glutathione levels in both the control anddiabetic groups. Amaç: Aritmi tedavisinde kullanılan amiodaronun çeşitli organlarda yan etkilere neden olduğu gösterilmiştir. Beyaz lahana (Brassica oleracea L. var. capitata) yaygın olarak tüketilen bir sebzedir. Amiodaron maruziyeti ve beyaz lahana tüketiminin kalp dokusu üzerindeki etkileri araştırılmamıştır. Beyaz lahana ekstraktının amiodaron uygulanan sıçanların kalp dokuları üzerindeki etkilerini araştırmayı amaçladık. Gereç ve Yöntem: Sıçanlara yedi gün boyunca gavajla beyaz lahana ekstresi ve amiodaron verildi. Beyaz lahana ekstresinin uygulanmasından 1 saat sonra amiodaron uygulandı. Sekizinci günde, hayvanların kalpleri alındı ve homojenize edildi. Lipit peroksidasyon (LPO), indirgenmiş glutatyon (GSH), siyalik asit (SA) düzeyleri ile glutatyon-S-transferaz (GST), katalaz, süperoksit dismutaz (SOD) ve doku faktörü (TF) aktiviteleri belirlendi. Bulgular: Amiodaron verilen grupta GSH, CAT, SOD aktiviteleri azalırken LPO ve SA seviyeleri arttı. Beyaz lahana, LPO ve SA miktarını azaltarak amiodaron zararını önledi. Ayrıca GSH düzeyleri ile SOD, CAT ve GST aktivitelerini artırarak antioksidan savunma sistemini güçlendirdi. Sonuç: Beyaz lahana tüketiminin, amiodaronun kalp dokusunda olumsuz etkilerini önlemek için alternatif bir yaklaşım olabileceği ileri sürülebilir., Objective: Amiodarone is used in the treatment of arrhythmia and has been shown to cause side effects in various organs. White cabbage (Brassica oleracea L. var. capitata) is a widely consumed vegetable. The effects of exposure to amiodarone and white cabbage consumption on heart tissue has not previously been investigated. We aimed to investigate the effects of white cabbage extract on amiodarone given to rats’ cardiac tissues. Material and Method: Rats were given white cabbage extract and amiodarone by gavage for seven days. Amiodarone was applied 1 hour after administration of the white cabbage extract. On the eighth day, the hearts of the animals were taken and homogenized. The levels of lipid peroxidation (LPO), reduced glutathione (GSH), sialic acid (SA) and activities of glutathione-S-transferase (GST), catalase, superoxide dismutase (SOD) and tissue factor (TF) were determined. Results: The GSH level, CAT and SOD activities decreased, while the LPO and SA levels increased in the amiodarone treated group. The White cabbage extract prevented any damage of the amiodarone by decreasing the amount of LPO and SA and also strengthened the antioxidant defence system by increasing the amount of the GSH and SOD activities and the CAT and GST activities. Conclusion: It is suggested that the consumption of white cabbage could be an alternative approach to the prevention of the adverse effects of amiodarone on heart tissue.

Objectives: Nowadays in pediatric dentistry, a raise in oral-dental health studies in twins, had been observed with the increasing twin births. The aim of this study was to investigate the differences in terms of oral-dental health between twin children and singleton children.Materials and Methods: A total of 180(105girls, 75 boys)children,120 of them twins and 60 of them singleton, aged 4 to 14 years,were included to the study.All children were healthy.A questionnaire,with 30 items, including the educational level of the parents, age, gender and brushing habits of the children was applied.The df-t /df-s index for primary teeth and DMF-T /DMF-S index ​​for permanent teeth were recorded.Streptococcus Mutans(SM) values ​​of all children were recorded with the help of chairside kits.Results: In-vitro fertilization(IVF) birth rate was 28.3% in the whole group.Furthermore, 33.3%of the children were monozygotic twins,66.6% of them were dizygotic twins.There were no statistically significant relationship between the DMFT-dft values and daily teeth brushing of children(p>0.05).A statistically significant correlation was found between mothers’ education levels and DMFT values ​​of children(p>0.05).DMFT values ​​of children whose mothers’ were university graduates were found to be significantly lower than those of primary and high school graduates (p>0.05).In the monozygotic group; in SM positive cases,the level of dft-dfs was statistically significantly higher than those of SM negative cases(p

Thromboplastin is a membrane bound glycoprotein and it isoverexpressed in many cancer cells. Chemotherapy induce somealterations in either saliva composition or function leading to oralcomplications. The aim of this study was to investigate the salivarythromboplastic activity of children with cancer and to comparewith healthy children. Saliva samples were taken from 15 childrenwho were diagnosed with lymphoid and solid tumor and from 15healthy children. In saliva samples thromboplastic activity andtotal protein levels were determined and SDS polyacrylamide gelelectrophoresis was also carried out with these saliva samples. Forcytological examination, imprint samples were prepared byapplying a drop of saliva on a slide. Salivary thromboplasticactivity of children with cancer (CC) was significantly decreasedwhen compared with the healthy children (HC). pH of the saliva inCC was significantly decreased. Salivary flow rate did notsignificantly change in CC. Saliva total protein level wassignificantly decreased in CC when compared to HC. In SDSPAGE electrophoresis, some protein bands decreased and someprotein bands increased when the two group is compared. Incytologic evaluation of saliva samples, keratinized and dysplasticcells increased in the imprint samples of CC. It should be notedthat children receiving chemotherapy encounter major problemsin oral and dental health, therefore they need special preventiveprograms for their oral and dental health.

Objectives: To examine the histologic structure of major salivary glands in high-fat diet induced hyperlipidemic rats. Materials and Methods: Sixteen Wistar Albino rats were grouped as control group (CG) and hyperlipidemic group (HG). CG was fed a standard chow and HG was fed a high-fat diet. At the end of three months, blood samples were obtained for lipid analyses. For histological analysis, major salivary glands were evaluted under light microscopy. Results: In HG; the number of intracellular lipid droplets (ILD) in the major salivary glands significantly increased while the diameter of ILD significantly increased in the parotid and sublingual glands (p< 0.01). The cell types mainly affected were serous cells in the parotid and submandibular, serous demilunar cells and, to a lesser extent, mucous cells in the sublingual gland. Serum levels of all lipids were significantly correlated with the lipid numbers in the parotid and the sublingual glands. In the submandibular gland, only serum LDL and cholesterol levels were found to be significantly correlated with the lipid numbers (p, Amaç: Yağ içeriği zengin diyet ile beslenerek hiperlipidemi oluşturulan sıçanlarda, büyük tükürük bezlerinin histolojik yapısını incelemektir. Bulgular: Hiperlipidemik gruba ait büyük tükürük bezlerinde hücre içi lipit damlacıklarının sayısında anlamlı bir artış görülürken, sublingual ve parotis bezinin hücre içi lipit damlacıklarının çapında anlamlı derecede artış tespit edildi (p < 0.01). Parotis ve submandibular bezlerde esas olarak seröz hücreler, sublingual bezde ise seröz yarımlar ve daha az derecede müköz hücreler hiperlipidemiden etkilendi. Tüm serum lipit düzeyleri ile sublingual ve parotis bezde biriken lipit damlacıkları sayısı arasında istatistiksel olarak anlamlı korelasyon saptandı. Submandibular bezde ise, sadece LDL ve kolesterol düzeyleri ile lipit damlacıkları sayısı arasında anlamlı bir korelasyon tespit edildi (p < 0.05). Sonuç: Yüksek yağ içeren diyet, parotis ve sublingual bezlerde yapısal bir değişikliğe neden olmaksızın, hücre içi lipit birikimine neden olmaktadır. Submandibular bez ise intraselüler lipit birikimine karşı kısmi bir direnç göstermektedir.

Purpose: Aim of this study is to evaluation and comparision of the effect of these two new generation haemostatic agents on short term soft tissue healing of warfarin treated rats biochemically. Method: 12 of 24 Wistar Albino rats were treated with warfarin systemicly (Warfarin group) and the other rats were selected as the control groupfor the trial. All rats had 3 incisions on dorsal dermal tissue applied 40 mg of Celox, 25 µl of Ankaferd Blood Stopper or no haemostatic agent. Antioxidant parameters, gluthatione and lipid peroxidation were determined from the tissue samples taken on the day of surgery, postoperative 4th and postoperative 8th day. Result: Both haemostatic agents positively affected the haemostasis for intraoperative and postoperative bleeding and could be used safely; Warfarin treatment and local haemostatic agents decreased the antioxidant effect; Ankaferd Blood Stopper’s effect to healing potential is better than the other agent under clinical observations. Conclusion: Effect mechanisms of Ankaferd Blood Stopper and Celox needs to be enlightened and different aspects of the topic should be investigated for development of potential benefits.Key words: Warfarin, Chitosan, Ankaferd Blood Stopper, Gluthatione, Lipidperoxidation, Prothrombine time, Amaç: Çalışmamızın amacı, varfarin verilen deney hayvanlarında iki yeni nesil farklı lokal hemostatik ajanın kısa dönemde yumuşak doku iyileşmesi üzerine olan etkisinin biyokimyasal olarak incelenmesi ve karşılaştırılmasıdır.Yöntem: 24 adet Wistar cinsi albino sıçanların 12 tanesine sistemik olarak varfarin verilmiş (Varfarin grubu), diğer sıçanlar ise kontrol grubu olarak kullanılmıştır. 3 adet bistüri yarası oluşturularak birine 40 mg kitozan granülleri (Celox®), diğerine 25 µl kurutulmuş folklorik bitki ekstresi (Ankaferd Blood Stopper®) uygulanırken diğer bir tanesine de herhangi bir hemostatik ajan uygulanmamıştır. Cerrahi işlemin yapıldığı gün, postoperatif 4. gün ve postoperatif 8. günde alınan doku örneklerinde antioksidan parametrelerden glutatyon ve oksidan parametrelerden lipid peroksidasyon tayini yapılmıştır. Bulgular: Kullanılan lokal hemostatik ajanların intraoperatif ve postoperatif kanama kontrolünde çok iyi olduğu ve güvenle kullanılabileceği; varfarin uygulaması ve lokal hemostatik ajanların antioksidan etkiyi zayıflattığı; ABS’nin klinik gözlemler neticesinde iyileşme potansiyeline etkisinin diğer ajana kıyasla daha fazla olduğu tesbit edilmiştir. Sonuç: Ankaferd Blood Stopper ve Celox’un etki mekanizmalarının tam olarak aydınlatılması ve potansiyel yararlarının geliştirilmesi için konunun farklı yönleriyle de ele alınması gerekmektedir.Anahtar Kelimeler : Varfarin, kitozan, Ankaferd Blood Stopper, glutatyon, lipidperoksidasyon, protrombin zamanı
