Abstract Invasive pests reduce biodiversity and ecosystem service functions, thereby leading to economic and also agricultural losses. Banana skipper (Erionota torus Evans), red palm weevil (Rhynchophorus ferrugineus), and coconut caterpillar (Opisina arenosella Walker) are invasive insect pests in the palm-growing regions and they have had serious consequences for the planting of bananas (Musa nana), palms (Trachycarpus fortune) and coconut (Cocos nucifera). Based on screened occurrence data, the present research utilized Maximum Entropy model (Maxent) to simulate the distribution dynamics of these three invasive insects in China, under current and future climate (2050s, 2070s, 2090s) in two shared socio-economic pathways (SSPs: 126 and 585) of the newly released coupled model intercomparison project phase6 (CMIP6). The results show that: (1) Under current and future climate conditions, all model groups exhibited an AUC value exceeding 0.92, which shows that the model prediction results are very good;(2) The suitable habitat area of E. torus Evans remains relatively stable with some expansion in the SSP126 of 2090s and some contraction in the SSP585 of 2090s. The suitable habitat area of R. ferrugineus showed an overall contraction, with substantial contraction in the SSP585 of 2090s.The suitable habitat area of O. arenosella has an overall expansion, with the most pronounced expansion in the SSP585 of 2070s; (3) The current centroid of suitable habitats for R. ferrugineus and E. torus Evans is located in Guangxi Province and wholely shift toward the south direction under future climate. The centroid of suitable habitats for O. arenosella is currently located in the northeastern maritime area of Hainan Province and will shift toward the north direction under future climate; (4) Temperature, precipitation and Human disturbance factors (Population density and Human influence index) were crucial variables for describing the distribution of the three species. For E. torus Evans in particular, percentage contributions of Population density was up to 31.4, which is only 0.1 different from ranked first Bio19 (Precipitation of the coldest quarter). The dynamics of habitats of these three species and the correlating driver factors proposed in this work provide essential insights into future spatial management of the three invasive insects in China. Our work is necessary and timely in identifying newly areas at high risk of expansion of the three invasive insects in the future, then suggesting strategic control measures to prevent their spread, and finally providing scientific evidence for the early prevention and rapid response to the three invasive insects.

Biodiversity is fundamental to human well-being and economic development. The Yangtze River, the largest river in China, faces biodiversity loss due to habitat degradation, climate change, and other anthropogenic threats. However, the long-term changes in the region's biodiversity remain poorly understood. Here, we constructed an optimized living planet index (LPIO) by combining Partial Least Squares Structural Equation Modeling and Random Forest Modeling. Using data from a monitoring network of 536 sites, we observed an increasing trend in terrestrial bird diversity and functional complexity across the entire watershed from 2011 to 2020. Our findings indicate that a large-scale ecological restoration program has contributed to increases in terrestrial and aquatic bird diversity in the Yangtze River Basin. In contrast, bird diversity in the downstream area has decreased by 2.83%, largely due to a rapid decline in wetland birds. The degradation of wetland habitats and insufficient conservation measures have negatively impacted bird diversity in the downstream region. This suggests that although there have been significant improvements in terrestrial bird diversity, more effective wetland restoration is necessary for biodiversity conservation. We recommend optimizing the national large-scale biodiversity monitoring network and increasing the number of upstream monitoring sites.

Abstract The Francois' langur (Trachypithecus francoisi) is a rare primate species indicated as endangered and distributed in karst areas in northern Vietnam and southwestern China. However, research limited to specific nature reserves or sites has hampered holistic conservation management. A comprehensive map of the potential distribution for the Francois' langur is essential to advance conservation efforts and ensure coordinated management across regions. Here, we used 82 occurrence records of Francois' langur surveyed in Guangxi, Guizhou, and Chongqing from 2017 to 2020, along with 12 environmental variables, to build the potential habitat model under current and future climate (2030, 2050, 2070, and 2090s) using maximum entropy models (MaxEnt). Our results indicated that (1) precipitation‐ and temperature‐associated bioclimatic variables contributed the most to the distribution of Francois' langur. Vegetation, water sources, and anthropogenic variables also affected its distribution; (2) a total of 144,207.44 km2 of potential suitable habitat across the entire range in China was estimated by the current model. Moderate‐ and high‐suitability habitats accounted for only 23.76% (34,265.96 km2) of the predicted suitable habitat and were mainly distributed in southwest Guangxi, east of Chongqing, and the border between Guizhou and Chongqing; (3) the suitable habitats of Francois' langur will contract considerably under future climate change, and the habitat centroid will move in the southeast direction with a shifting distance of approximately 2.84 km/year from current to 2100. The habitat prediction of Francois' langur and the main drivers proposed in this study could provide essential insights for the future conservation of this endangered species. The existing distribution areas should be monitored and protected, but conservation beyond existing habitats should also be a focus of effort, especially in future expansion areas. This would ensure effective and timely protection under climate change and anthropogenic pressures.

Rana dabieshanensis is a species of frog within the family Ranidae. In this study, we assembled a complete mitochondrial genome (mito-genome) for R. dabieshanensis by high-throughput sequencing technology. It is 18,291 bp and includes 13 protein-coding genes, 22 tRNA genes, two rRNA genes and one control region. The nucleotide composition is A: 27.0%, T: 28.6%, C: 29.3% and G: 15.2%. Two overlaps among the 13 protein-coding genes were found: ATP8/ATP6, ND4L/ND4. The study of phylogenetic analysis based on complete mitochondrial genome showed that there was close genetic relationship between R. dabieshanensis and R. omeimontis and it is useful for systematic analyses of genus Rana.

Abstract Invasive alien plants pose a significant threat to biodiversity and the agricultural economy. The invasive weed (Ammannia coccinea) competes with rice in paddy fields, potentially threatening rice production. Despite the crucial need to estimate the global geographical distribution and ecological niche dynamics of A. coccinea for effective early warning, control strategies, and global rice security, relevant research remains scarce. This study utilized the Biomod2 platform, which integrates multiple single models into ensemble model, incorporating environmental and species data to analyze the distribution range shifts of A. coccinea under current and future climate scenarios. It also quantified and analyzed shifts in the species’ ecological niche across these climate scenarios. The results indicated that the potential suitable areas for A. coccinea were mainly in Southern North America, northern and south-eastern South America, south-western Europe, the Middle East, central Africa, western Asia, south-eastern Asia, with a gradual increase in mid-high suitability habitat over time and radiation levels. While the overall ecological niche of A. coccinea remains stable, minor shifts are expected under future conditions. Temperature, precipitation, and the human impact index were the key factors influencing the future distribution of A. coccinea. Climate change contributes to the expansion of A. coccinea's highly suitable areas and shifts its ecological niche. Organizations efforts should focus on preventing the spread of A. coccinea in regions where its potential distribution overlaps with key rice production areas. The findings of this study provide critical insights into the global distribution and ecological niche dynamics of A. coccinea, aiding in the development of early warning and control strategies to mitigate its impact on biodiversity, agriculture, and particularly rice production under future climate scenarios.

Abstract Background Podocyte injury causes proteinuria and accelerates glomerular sclerosis during diabetic kidney disease (DKD). Disruptor of telomeric silencing 1-like (DOT1L), an evolutionarily conserved histone methyltransferase, has been reported in preventing kidney fibrosis in chronic kidney disease models. However, whether DOT1L exerts beneficial effects in diabetes induced podocyte injury and the underlying molecular mechanisms need further exploration. Methods The expression of DOT1L was confirmed by Western blotting in MPC-5 cells and cortex of kidney from db/db mice, as well as immunofluorescence staining in human renal biopsy samples. The effect of DOT1L on podocyte injury was obtained using MPC-5 cells and db/db mice. The potential target genes regulated by DOT1L was measured by RNA-sequencing. Then, a series of molecular biological experiments was performed to investigate the regulation of PLCL1 by DOT1L in MCP-5 cells and db/db mice. Lipid accumulation was assessed by UPLC-MS/MS analysis and Oil Red O staining. Results DOT1L expression was significantly declined in high glucose (HG)-treated MPC-5 cells, podocyte regions of kidney tissues from db/db mice and human renal biopsy samples. Subsequent investigations revealed that upregulation of DOT1L ameliorated HG-induced cell apoptosis in MPC-5 cells as well as primary podocytes. Furthermore, podocyte-specific DOT1L overexpression inhibited diabetic podocyte injury in db/db mice. Mechanistically, we revealed that DOT1L upregulated phospholipase C-like 1 (PLCL1) expression by mediating H3K79me2 at its promoter and PLCL1 silencing suppressed the protective role of DOT1L on podocyte injury. Moreover, DOT1L improved diabetes induced abnormal fatty acid metabolism in podocytes and PLCL1 knockdown reversed its protective effects. Conclusions Taken together, our results indicate that DOT1L protects podocyte injury via PLCL1-mediated fatty acid metabolism and provides new insights into the therapeutic target of DKD.

Abstract Background Assessing dietary phenylalanine (Phe) tolerance is crucial for managing hyperphenylalaninemia (HPA) in children. However, traditionally, adjusting the diet requires significant time from clinicians and parents. This study aims to investigate the development of a machine-learning model that predicts a range of dietary Phe intake tolerance for children with HPA over 10 years following diagnosis. Methods In this multicenter retrospective observational study, we collected the genotypes of phenylalanine hydroxylase (PAH), metabolic profiles at screening and diagnosis, and blood Phe concentrations corresponding to dietary Phe intake from over 10 years of follow-up data for 204 children with HPA. To incorporate genetic information, allelic phenotype value (APV) was input for 2965 missense variants in the PAH gene using a predicted APV (pAPV) model. This model was trained on known pheno-genotype relationships from the BioPKU database, utilizing 31 features. Subsequently, a multiclass classification model was constructed and trained on a dataset featuring metabolic data, genetic data, and follow-up data from 3177 events. The final model was fine-tuned using tenfold validation and validated against three independent datasets. Results The pAPV model achieved a good predictive performance with root mean squared error (RMSE) of 1.53 and 2.38 on the training and test datasets, respectively. The variants that cause amino acid changes in the region of 200–300 of PAH tend to exhibit lower pAPV. The final model achieved a sensitivity range of 0.77 to 0.91 and a specificity range of 0.8 to 1 across all validation datasets. Additional assessment metrics including positive predictive value (0.68–1), negative predictive values (0.8–0.98), F1 score (0.71–0.92), and balanced accuracy (0.8–0.92) demonstrated the robust performance of our model. Conclusions Our model integrates metabolic and genetic information to accurately predict age-specific Phe tolerance, aiding in the precision management of patients with HPA. This study provides a potential framework that could be applied to other inborn errors of metabolism.

Objective To understand the prevalence and influencing factors of different types of obesity among middle-aged and elderly Han and Tibetan residents in Gansu province, and to provide evidence for the prevention and control of obesity. Methods Data of 18 114 middle-aged and elderly adults aged 35 to 75 years old with complete body measurement information were analyzed. The data were collected in the baseline survey of "Gansu Province′s Urban and Rural Natural Population Cohort Establishment and Tumor Follow-up Study" of the "Regional Ethnic Cohort Study in Northwest China" conducted from November 2018 to May 2019. Body mass index (BMI), waist circumference (WC), body fat percentage (BFP) and visceral fat index (VFI) were used to assess the prevalence of general obesity, central obesity, body fat obesity, and visceral obesity, respectively. Multivariable unconditional logistic regression analyses were used to explore the influencing factors of different types of obesity. Results Among the 18 114 middle-aged and elderly adults enrolled in Gansu Province, there were 16 999 Han (93.8%) and 1 115 Tibetan (6.2%) ones. The prevalence rates of general obesity, central obesity, body fat obesity, and visceral obesity among middle-aged and elderly Han and Tibetan residents were 12.5% and 9.0%, 31.4% and 33.9%, 50.7% and 41.9%, 40.0% and 27.7%, respectively. The prevalence rates of general obesity, body fat obesity, and visceral obesity among Han adults were higher than those among Tibetan residents (all P < 0.05). The results of multivariable unconditional logistic regression analysis showed that being female was a risk factor for body fat obesity among middle-aged and elderly Han residents and for general obesity, central obesity, and body fat obesity among middle-aged and elderly Tibetan residents in Gansu province, but was a protective factor for general obesity, central obesity, visceral obesity among middle-aged and elderly Han residents and for visceral obesity among middle-aged and elderly Tibetan residents in Gansu province. Age ≥ 45 years old was a risk factor for general obesity, central obesity, body fat obesity, and visceral obesity among middle-aged and elderly Han residents and for body fat obesity among middle-aged and elderly Tibetan residents in Gansu province. Primary school education and above was a protective factor for central obesity among middle-aged and elderly Han residents in Gansu province. Being married was a risk factor for general obesity and visceral obesity among middle-aged and elderly Han residents in Gansu province. Being a non-farmer was a risk factor for general obesity and central obesity among middle-aged and elderly Han residents in Gansu province. Per capita annual household income ≥ 4 000 Yuan was a risk factor for visceral obesity among middle-aged and elderly Han residents in Gansu province. Smoking was a protective factor for general obesity, body fat obesity, and visceral obesity among middle-aged and elderly Han residents in Gansu province. Drinking was a risk factor for central obesity among middle-aged and elderly Tibetan residents in Gansu province. ConclusionThe prevalence rates of general obesity, body fat obesity, and visceral obesity were higher among middle-aged and elderly Han residents than those among middle-aged and elderly Tibetan residents in Gansu province, and gender was a common influencing factor for the four types of obesity among middle-aged and elderly Han and Tibetan residents in Gansu province.

Introduction: Reverse genetic studies conducted in the plant with a complex or polyploidy genome enriched with large gene families (like wheat) often meet challenges in identifying the key candidate genes related to important traits and prioritizing the genes for functional experiments. Objective: To overcome the above-mentioned challenges of reverse genetics, this work aims to establish an efficient multi-species strategy for genome-wide gene identification and prioritization of the key candidate genes. Methods: We established the integrative gene duplication and genome-wide analysis (iGG analysis) as a strategy for pinpointing key candidate genes deserving functional research. The iGG captures the evolution, and the expansion/contraction of large gene families across phylogeny-related species and integrates spatial–temporal expression information for gene function inference. Transgenic approaches were also employed to functional validation. Results: As a proof-of-concept for the iGG analysis, we took the wheat calcineurin B-like protein-interacting protein kinases (CIPKs) family as an example. We identified CIPKs from seven monocot species, established the orthologous relationship of CIPKs between rice and wheat, and characterized Triticeae-specific CIPK duplicates (e.g., CIPK4 and CIPK17). Integrated with our analysis of CBLs and CBL-CIPK interaction, we revealed that divergent expressions of TaCBLs and TaCIPKs could play an important role in keeping the stoichiometric balance of CBL-CIPK. Furthermore, we validated the function of TaCIPK17-A2 in the regulation of drought tolerance by using transgenic approaches. Overexpression of TaCIPK17 enhanced antioxidant capacity and improved drought tolerance in wheat. Conclusion: The iGG analysis leverages evolutionary and comparative genomics of crops with large genomes to rapidly highlight the duplicated genes potentially associated with speciation, domestication and/or particular traits that deserve reverse-genetic functional studies. Through the identification of Triticeae-specific TaCIPK17 duplicates and functional validation, we demonstrated the effectiveness of the iGG analysis and provided a new target gene for improving drought tolerance in wheat.

Abstract Background Saline lakes are home to various archaea that play special and crucial roles in the global biogeochemical cycle. The Qinghai-Tibet Plateau hosts a large number of lakes with diverse salinity ranging from 0.1 to over 400 g/L, harboring complex and diverse archaea. To the best of our knowledge, the formation mechanisms and potential ecological roles of archaea in Qinghai-Tibetan Plateau saline lakes remain largely unknown. Results Using High-throughput Illumina sequencing, we uncovered the vastly distinct archaea communities between two typical saline lakes with significant salinity differences on the Qinghai Tibet Plateau (Qinghai saline lake and Chaka hypersaline lake) and suggested archaea played different important roles in methanogenesis-related and nitrate reduction-related functions of these two lakes, respectively. Rather than the individual effect of salinity, the composite effect of salinity with diverse environmental parameters (e.g., temperature, chlorophyll a, total nitrogen, and total phosphorus) dominated the explanation of the variations in archaeal community structure in different habitats. Based on the network analysis, we further found the correlations between dominant archaeal OTUs were tight but significantly different between the two habitats, implying that archaeal interactions may also largely determine the shape of archaeal communities. Conclusion The present study improved our understanding of the structure and function of archaea in different saline lakes on the Qinghai-Tibet Plateau and provided a new perspective on the mechanisms underlying shaping their communities.

Abstract Background This study was designed to assess the associations between emerging cardiometabolic indices—the atherogenic index of plasma (AIP), the stress hyperglycemia ratio (SHR), the triglyceride-glucose (TyG) index, and the homeostasis model assessment of insulin resistance (HOMA-IR)—and the incidence of diabetic kidney disease (DKD) in type 2 diabetes (T2D) patients. Methods We consecutively enrolled 4351 T2D patients. The AIP, SHR, TyG index, and HOMA-IR were calculated from baseline parameters. Abstract Background A major obstacle faced by families with rare diseases is obtaining a genetic diagnosis. The average "diagnostic odyssey" lasts over five years and causal variants are identified in under 50%, even when capturing variants genome-wide. To aid in the interpretation and prioritization of the vast number of variants detected, computational methods are proliferating. Knowing which tools are most effective remains unclear. To evaluate the performance of computational methods, and to encourage innovation in method development, we designed a Critical Assessment of Genome Interpretation (CAGI) community challenge to place variant prioritization models head-to-head in a real-life clinical diagnostic setting. Methods We utilized genome sequencing (GS) data from families sequenced in the Rare Genomes Project (RGP), a direct-to-participant research study on the utility of GS for rare disease diagnosis and gene discovery. Challenge predictors were provided with a dataset of variant calls and phenotype terms from 175 RGP individuals (65 families), including 35 solved training set families with causal variants specified, and 30 unlabeled test set families (14 solved, 16 unsolved). We tasked teams to identify causal variants in as many families as possible. Predictors submitted variant predictions with estimated probability of causal relationship (EPCR) values. Model performance was determined by two metrics, a weighted score based on the rank position of causal variants, and the maximum F-measure, based on precision and recall of causal variants across all EPCR values. Results Sixteen teams submitted predictions from 52 models, some with manual review incorporated. Top performers recalled causal variants in up to 13 of 14 solved families within the top 5 ranked variants. Newly discovered diagnostic variants were returned to two previously unsolved families following confirmatory RNA sequencing, and two novel disease gene candidates were entered into Matchmaker Exchange. In one example, RNA sequencing demonstrated aberrant splicing due to a deep intronic indel in ASNS, identified in trans with a frameshift variant in an unsolved proband with phenotypes consistent with asparagine synthetase deficiency. Conclusions Model methodology and performance was highly variable. Models weighing call quality, allele frequency, predicted deleteriousness, segregation, and phenotype were effective in identifying causal variants, and models open to phenotype expansion and non-coding variants were able to capture more difficult diagnoses and discover new diagnoses. Overall, computational models can significantly aid variant prioritization. For use in diagnostics, detailed review and conservative assessment of prioritized variants against established criteria is needed.

Accurate monitoring of fertilizer concentration and variable irrigation components is crucial for achieving precision irrigation through variable-rate fertigation. However, technological and cost constraints pose challenges in effectively managing mixed variable-rate fertigation. This study addresses these challenges by integrating machine learning (ML) with easily monitored physical parameters, such as electrical conductivity (EC), pH, and temperature, to predict fertilizer solution components and concentrations in mixed variable-rate fertigation. Cubic spline interpolation (CSI) was used to enhance the training dataset. Six ML algorithms—Multivariate Linear Regression (MLR), Support Vector Machines (SVM), K-Nearest Neighbors (KNN), Extremely Randomized Trees (ETs), Multilayer Perceptron (MLP), and Extreme Gradient Boosting (XGB)—were used to develop prediction models, with their performance evaluated using the coefficient of determination (R2), root mean square error (RMSE), and Akaike information criterion (AIC). The Extended Fourier Amplitude Sensitivity Test (EFAST) assessed the sensitivity of the ML models to physical parameters. All of the ML models, except for MLR, particularly SVM, demonstrated superior performance in predicting fertilizer solution components’ concentrations, with R2 values between 0.989 and 0.997 and RMSE values between 0.089 and 0.210. The CSI significantly enhanced model performance, resulting in larger R2 values and smaller RMSE values. The AIC results and sensitivity analysis confirmed the exceptional performance of SVM, emphasizing its suitability for predicting fertilizer components using easily measured physical parameters. The developed ML models offer valuable insights for decision-makers managing irrigation and fertilization in mixed variable-rate fertigation.

Abstract Gravity heat pipe system has previously been proven as an environmental and efficient technology in exploiting hot dry rock. However, it is unclear what geothermal reservoir siting is more favorable for the system’s heat extraction performance. Herein, we analyzed the influences of geothermal field siting (rock property and reservoir environment) on heat extraction performance of gravity heat pipe systems through a 3D thermal–hydraulic coupled model. It is found that rock properties have huge influences on heat compensation, heat extraction ratio and heat compensation ratio. Low rock density, low rock specific heat capacity and high thermal conductivity could increase heat compensation, heat extraction ratio and heat compensation ratio. It is also found that geothermal reservoir environment affects the heat extraction rate seriously. High initial temperatures and low temperature gradients increase heat extraction rates. Geothermal reservoir pressure affects the heat extraction performance slightly, and low initial pressures increase heat extraction rate. The study results would provide suggestions on deep geothermal exploitation locations.

Background/Objectives: Hemodialysis-induced myocardial stunning (HIMS) is a frequent complication in patients undergoing maintenance hemodialysis, characterized by transient left ventricular dysfunction due to ischemic episodes. Mitochondrial dysfunction and fluctuations in key ions such as potassium (K+) and calcium (Ca2+) are implicated in the pathogenesis of HIMS. This study aims to investigate the role of mitochondrial dysfunction and the protective potential of mitochondrial ATP-sensitive potassium channels (mitoKATP) in mitigating HIMS. Methods: A 5/6 nephrectomy rat model was established to mimic chronic kidney disease and the subsequent HIMS. The effects of mitoKATP channel modulators were evaluated by administering diazoxide (DZX), a mitoKATP opener, and 5-hydroxydecanoate (5-HD), a mitoKATP blocker, before hemodialysis. Mitochondrial function was assessed by measuring membrane potential, ATP synthase activity, and intramitochondrial Ca2+ levels. Myocardial function was evaluated using speckle tracking echocardiography. Results: Rats undergoing hemodialysis exhibited significant reductions in left ventricular strain and synchrony. DZX administration significantly improved mitochondrial function and reduced myocardial strain compared to controls. Conversely, 5-HD worsened mitochondrial swelling and disrupted myocardial function. Higher K+ and Ca2+ concentrations in the dialysate were associated with improved mitochondrial energy metabolism and myocardial strain. Conclusions: Mitochondrial dysfunction and ion imbalances during hemodialysis are key contributors to HIMS. The activation of mitoKATP channels provides mitochondrial protection and may serve as a potential therapeutic strategy to mitigate HIMS.

Abstract Background The gut microbiota plays a crucial role in regulating host metabolism and producing uremic toxins in patients with end-stage renal disease (ESRD). Our objective is to advance toward a holistic understanding of the gut ecosystem and its functional capacity in such patients, which is still lacking. Results Herein, we explore the gut microbiome of 378 hemodialytic ESRD patients and 290 healthy volunteers from two independent cohorts via deep metagenomic sequencing and metagenome-assembled-genome-based characterization of their feces. Our findings reveal fundamental alterations in the ESRD microbiome, characterized by a panel of 348 differentially abundant species, including ESRD-elevated representatives of Blautia spp., Dorea spp., and Eggerthellaceae, and ESRD-depleted Prevotella and Roseburia species. Through functional annotation of the ESRD-associated species, we uncover various taxon-specific functions linked to the disease, such as antimicrobial resistance, aromatic compound degradation, and biosynthesis of small bioactive molecules. Additionally, we show that the gut microbial composition can be utilized to predict serum uremic toxin concentrations, and based on this, we identify the key toxin-contributing species. Furthermore, our investigation extended to 47 additional non-dialyzed chronic kidney disease (CKD) patients, revealing a significant correlation between the abundance of ESRD-associated microbial signatures and CKD progression. Conclusion This study delineates the taxonomic and functional landscapes and biomarkers of the ESRD microbiome. Understanding the role of gut microbiota in ESRD could open new avenues for therapeutic interventions and personalized treatment approaches in patients with this condition.

For ultrasonic welding (USW) of fiber reinforced thermoplastic composites without energy director (ED), the special contact surface and interphase structures due to the addition of fibers have strong relation to the weld quality. In this study, through the servo-driven USW experiments and the corresponding mini-anisotropy modeling for PA6 and CF/PA6 composites welding without ED, the fusion phenomena and weldability of the materials are addressed. The results show that as compared to pure PA6, CF/PA6 has the higher loss modulus and lower specific capacity, leading to more viscoelastic heat generation during USW. As the CF/PA6 surface has the higher roughness, the more significant local stress and energy concentrations occur around the initial contact area, leading to the increased welding area; while more pores are observed within the joints of composites due to the poor liquidity and air trap from larger contact gap. Due to the interphase structure between CF and PA6, the high strain and high temperature occur around the fibers, where pores form due to thermal decomposition.

Dominance hierarchies are common in social mammals, especially primates. The formation of social hierarchies is conducive to solving the problem of the allocation of scarce resources among individuals. From August 2015 to July 2016, we observed a wild, provisioned Yunnan snub-nosed monkey (Rhinopithecus bieti) group at Xiangguqing in Baimaxueshan National Nature Reserve, Yunnan Province, China. Aggressive and submissive behaviors were used to investigate dominance hierarchies between female individuals in the same one-male unit (OMU), and the grooming reciprocity index was used to detect reciprocal relationships between these females within the OMU. The results showed that loose social hierarchies exist among the females in each OMU, and more dominant individuals have higher grooming incomes. BackgroundThe global burden of digestive diseases has increased in recent years. The study aims to comprehend the trend of incidence and death rates related to digestive diseases in China from 2000 to 2020.MethodsThe study collected data on digestive diseases and their causes, such as incidence rates, death rates, Years of Life Lost, Years Lived with Disability, Disability-Adjusted Life Years and estimated annual percentage change from the 2019 Global Burden of Disease website and the Chinese Health and Wellness Statistical Yearbook spanning. And we employed the age-period-cohort model to analyze the influence of age, period, and birth cohort on the trend of death rates associated with digestive diseases.ResultsIn contrast to the global burden of digestive disease, China experienced increases in the age-standardized incidence for inflammatory bowel disease, gallbladder and biliary diseases, as well as appendicitis from 2000 to 2019. The corresponding estimated annual percentage change for these diseases were 2.06, 1.74, and 0.99. Females showed a significantly higher incidence of digestive diseases, while males experienced a higher death rate. Moreover, individuals in the age groups under 5 years and over 60 years exhibited higher death rates than those in other age groups.ConclusionThe findings underscore the ongoing importance of digestive diseases as a substantial public health issue in China. Reducing the disease burden of IBD in China necessitates healthcare systems to enhance their infrastructure and personnel readiness, ensuring an equitable, affordable, and accessible distribution of care for IBD patients. To reduce the occurrence and mortality rates of digestive diseases in China, the government should promote the adoption of early screening policies for individuals under the 5 year and those above the 60 year. These policies should be accompanied by customized preventive measures.

Objective: We aimed to investigate the clinical and genetic risk factors associated with neonatal severe unconjugated hyperbilirubinemia.Methods: This was a retrospective, 1:1 matched, case–control study. We included 614 neonates diagnosed with severe unconjugated hyperbilirubinemia (serum total bilirubin level ≥425 μmol/L or serum total bilirubin concentration that met exchange transfusion criteria) from the China Neonatal Genomes Project in Children’s Hospital of Fudan University. Clinical exome sequencing data were analyzed using a data analysis pipeline of Children’s Hospital of Fudan University. The factors associated with severe unconjugated hyperbilirubinemia were assessed using univariable and multivariable logistic regression analyses. Interaction analyses were examined between clinical and genetic risk factors.Results: ABO/Rh incompatibility hemolysis (odds ratio [OR] 3.36, 95% confidence interval [CI] 2.32–4.86), extravascular hemorrhage (OR 2.95, 95% CI 2.24–3.89), weight loss (OR 5.46, 95% CI 2.88–10.36), exclusive breastmilk feeding (OR 3.56, 95% CI 2.71–4.68), and the homozygous mutant of UGT1A1 211G>A (OR 2.35, 95% CI 1.54–3.59) were all identified as factors significantly associated with severe unconjugated hyperbilirubinemia. The presence of UGT1A1 211G>A mildly increased the risk of severe unconjugated hyperbilirubinemia caused by ABO/Rh incompatibility hemolysis (OR 3.98, 95% CI 2.19–7.23), although the effect is not statistically significant.Conclusion: ABO/Rh incompatibility hemolysis, extravascular hemorrhage, weight loss, exclusive breastmilk feeding, and the homozygous mutant of UGT1A1 211G>A were found to be risk factors for severe unconjugated hyperbilirubinemia. Clinical factors remain the most crucial and preventable determinants in managing severe unconjugated hyperbilirubinemia, with a minimal genetic contribution. The establishment of preconception care practices and the reinforcement of screening for the aforementioned risk factors are essential steps for preventing severe unconjugated hyperbilirubinemia.

Abstract As a globally important staple crop, wheat seeds provide us with nutrients and proteins. The trend of healthy dietary has become popular recently, emphasizing the consumption of whole-grain wheat products and the dietary benefits. However, the dynamic changes in nutritional profiles of different wheat seed regions (i.e., the embryo, endosperm and outer layers) during developmental stages and the molecular regulation have not been well studied. Here, we provide this multi-omic resource of wheat seeds and describe the generation, technical assessment and preliminary analyses. This resource includes a time-series RNA-seq dataset of the embryo, endosperm and outer layers of wheat seeds and their corresponding metabolomic dataset, covering the middle and late stages of seed development. Our RNA-seq experiments profile the expression of 63,708 genes, while the metabolomic data includes the abundance of 984 metabolites. We believe that this was the first reported transcriptome and metabolome dataset of wheat seeds that helps understand the molecular regulation of the deposition of beneficial nutrients and hence improvements for nutritional and processing quality traits.

The regolith of the Chang'E-4 landing site was disturbed by its engine exhaust. To explore the interaction between the engine exhaust and the regolith, it was necessary to identify the exhaust-disturbed region. This article focuses on determining the extent of the disturbed region by using lunar reconnaissance orbiter camera narrow angle camera (LROC NAC) images. For this purpose, the tools of temporal-ratio images, phase-ratio images, reflectance profiles, and reflectance isoline graphs are employed. The reflectance profiles and isoline graphs derived from the temporal-ratio images reveal the reflectance changes before and after landing. Compared with the reflectance profiles, isoline graphs further include the spatial information of isolines, thus more robust to noise. Based on the magnitudes of changed reflectance around the lander, the engine exhaust-disturbed region was further divided into the focus disturbed region (FDR) and the diffuse disturbed region (DDR). The final estimated spatial extent along the north–south and east–west directions of the FDR were ∼9.6 and ∼10.8 m, and those of the DDR were ∼75 and ∼80 m. As compared with the estimated spatial extent of the Chang'E-3 landing site, the DDR of the Chang'E-4 landing site was larger, but the FDR was smaller. We attributed this to geological and topography factors. The reflectance changes between the FDR and the undisturbed region increased by ∼10±1%. This indicates similar processes causing the variations in the regolith properties, likely including the smoothing of the surface from microscopic to macroscopic by destroying fine-grained regolith components, or changing of the surface maturity.

The current study investigated the effects of lactic acid bacteria fermentation on the protein gelation and digestive characteristics from four pulses, namely, chickpea, red kidney bean, lentil and pea. Protein extracted from the four pulses was named CP, RKP, LP and PP. Protein gels were prepared by using Lactobacillus plantarum B1-6, and the least gelation concentration (LGC), texture characteristics, microstructure, and color were investigated. The gels were subsequently subjected to an in vitro dynamic gastrointestinal digestion, and the protein digestive characteristics and the protective effect on vitality of lactic acid bacteria were studied. The results showed that the LGC of CP, RKP, LP and PP gels was 1.6% (w/v), 1.6% (w/v), 1.4% (w/v) and 1.2% (w/v), respectively. Summary: Adherens junctions between tubular epithelial cells are disrupted in renal ischemia/reperfusion (I/R) injury. Syndecan-1 (SDC-1) is involved in maintaining cell morphology. We aimed to study the role of SDC-1 shedding induced by renal I/R in the destruction of intracellular adherens junctions. We found that SDC-1 shedding was increased while the expression of E-cadherin was decreased. This observation was accompanied by the activation of STAT3 in the kidneys. Inhibiting the shedding of SDC-1 induced by I/R could alleviate this effect. Mild renal I/R could induce more severe renal injury, lower E-cadherin expression, damaged cell junctions, and activated STAT3 in knockout mice with the tubule-specific deletion of SDC-1 mice. The results in vitro were consistent with those in vivo. Inhibiting the shedding of SDC-1 could alleviate the decreased expression of E-cadherin and damage of cell adherens junctions through inhibiting the activation of STAT3 during ischemic acute kidney injury.

As the additive manufacturing provides an advanced method for fabricating polymeric composite components in thermal engineering applications, the thermal contact conductance (TCC) between the contact composites can become an important problem. In order to understand the interfacial heat transfer mechanism between thermoplastic composites, TCC between additively manufactured carbon fiber reinforced polyamide 12 (CF/PA12) composites was experimentally studied with steady-state measurements in this work. By considering the material surface microstructure, thermal and mechanical properties, the effects of interface temperature, contact pressure, surface morphology, carbon fiber (CF) content and orientation in composites on TCC were analyzed. The results show that the TCC is dependent on the actual contact area and the thermal conduction path inside the material. In general, high surface roughness decreases the TCC because of the reduction of contact area; while it is the opposite of temperature and pressure. The CF distribution affects the internal thermal conduction path and surface roughness, leading to the variations in TCC.

Abstract Brachial plexus root avulsion (BPRA) is frequently caused by high-energy trauma including traffic accident and birth trauma, which will induces massive motoneurons (MNs) death as well as loss of motor and sensory function in the upper limb. The death of MNs is attributed to energy deficiency, neuroinflammation and oxidative stress at the injured ventral horn of spinal cord triggered by BPRA injury. It has been reported which aldose reductase (AR), an endogenous enzyme that catalyzes fructose synthesis, positively correlates with the poor prognosis following cerebral ischemic injury, diabetic retinopathy and diabetic peripheral neuropathy. However, the role of AR in BPRA remains unknown. Herein, we used a mouse model and found that in the spinal cord of BPRA mice, the upregulation of AR correlated significantly with (1) an inactivated SIRT1–AMPK–mTOR pathway and disrupted autophagy; (2) increased byproducts accumulation of lipid peroxidation metabolism and neuroinflammation; and (3) increased MNs death. Furthermore, our results demonstrated the role of AR in BPRA injury whereby the absence of AR (AR knockout mice, AR−/−) prevented the hyper-neuroinflammation and disrupted autophagy as well as motor neuron death caused by BPRA injury. Finally, we further demonstrate that AR inhibitor epalrestat is neuroprotective against BPRA injury by increasing autophagy level, alleviating neuroinflammation and rescuing MNs death in mice. Collectively, our data demonstrate that the AR upregulation in the spinal cord is an important factor contributing to autophagy disruption, neuroinflammation and MNs death following brachial plexus roots avulsion in mice. Our study also provides a promising therapy drug to assist re-implantation surgery for the treatment of BPRA.

Partial root-zone drying (PRD) is an effective water-saving irrigation strategy that improves stress tolerance and facilitates efficient water use in several crops. It has long been considered that abscisic acid (ABA)-dependent drought resistance may be involved during partial root-zone drying. However, the molecular mechanisms underlying PRD-mediated stress tolerance remain unclear. It’s hypothesized that other mechanisms might contribute to PRD-mediated drought tolerance. Here, rice seedlings were used as a research model and the complex transcriptomic and metabolic reprogramming processes were revealed during PRD, with several key genes involved in osmotic stress tolerance identified by using a combination of physiological, transcriptome, and metabolome analyses. Our results demonstrated that PRD induces transcriptomic alteration mainly in the roots but not in the leaves and adjusts several amino-acid and phytohormone metabolic pathways to maintain the balance between growth and stress response compared to the polyethylene glycol (PEG)-treated roots. Integrated analysis of the transcriptome and metabolome associated the co-expression modules with PRD-induced metabolic reprogramming. Several genes encoding the key transcription factors (TFs) were identified in these co-expression modules, highlighting several key TFs, including TCP19, WRI1a, ABF1, ABF2, DERF1, and TZF7, involved in nitrogen metabolism, lipid metabolism, ABA signaling, ethylene signaling, and stress regulation. Thus, our work presents the first evidence that molecular mechanisms other than ABA-mediated drought resistance are involved in PRD-mediated stress tolerance. Overall, our results provide new insights into PRD-mediated osmotic stress tolerance, clarify the molecular regulation induced by PRD, and identify genes useful for further improving water-use efficiency and/or stress tolerance in rice.

A servo-driven ultrasonic welding (USW) offers a good control for joining carbon fiber reinforced thermoplastic composites (CFRTP) with a constant plunging speed, a dynamic applied force and adjusting power in real time. This study focuses on the process and strength analysis during servo-driven USW of CF/PA6 composites without energy director (ED) to obtain the correlation between the operation parameters and joint strength. The results show that a servo-driven welder benefits in USW without ED due to the initial intimate contact between workpieces without horn retraction. It is also found that a typical vibration process can be divided into four stages following the heat evolution and characteristic temperature of CFRTP, i.e., “initial stage”, “soften stage”, “melting stage” and “steady stage”. High energy and amplitude bring enough total energy and high power to melt materials; while a low trigger force and a slow plunging speed can mitigate the excessive squeeze-out with a result of the increase in weld strength. Thus, as the melted material remained in the weld region decides the bonding efficiency, the desired joints can be obtained under the enough welding energy (≥400 J) and amplitude (≥80%), not too high welding force (≤800 N) and plunging speed (≤0.8 mm/s). Meanwhile, weld quality can be online inspected and judged based on welding force evolution, and the measured displacement relative to the set value. This work contributes to the optimal parametric selection for USW of CFRTP plates without ED applying a servo-driven welder.

Abstract The composition of microbial communities varies considerably across ecological environments, particularly in extreme environments, where unique microorganisms are typically used as the indicators of environmental conditions. However, the ecological reasons for the differences in microbial communities remain largely unknown. Herein, we analyzed taxonomic and functional community profiles via high-throughput sequencing to determine the alkaline saline soil bacterial and archaeal communities in the Qarhan Salt Lake area in the Qinghai–Tibet Plateau. The results showed that Betaproteobacteria (Proteobacteria) and Halobacteria (Euryarchaeota) were the most abundant in the soils of this area, which are common in high salinity environments. Accordingly, microbes that can adapt to local extremes typically have unique metabolic pathways and functions, such as chemoheterotrophy, aerobic chemoheterotrophy, nitrogen fixation, ureolysis, nitrate reduction, fermentation, dark hydrogen oxidation, and methanogenesis. Methanogenesis pathways include hydrogenotrophic methanogenesis, CO2 reduction with H2, and formate methanogenesis. Thus, prokaryotic microorganisms in high salinity environments are indispensable in nitrogen and carbon cycling via particular metabolic pathways.

Affected by dip angle and thickness of strata and the tunneling method, soft rock tunnel has obvious characteristics of large deformation, long deformation time and difficult support. Based on a case study of Gelong the deformation and failure mechanism of surrounding rock, stress characteristics of supporting structural and control method of large section highway tunnel passing through strong-medium weathered carbonaceous slate stratum are studied. This paper proposed construction method based on strengthening the longitudinal stiffness of supporting structure and increasing the integrity of surrounding rock, The results showed that the deformation of surrounding rock and the stress of supporting structure increased rapidly in the early stage of construction. The cumulative deformation of vault settlement and horizontal convergence reached 116.9 mm and 97.9 mm, respectively, accounting for 73.53% and 76.62% of the total deformation. The proportion of surrounding rock pressure shared by the primary support and the secondary lining was about 8.9:1.1. This shows that the initial support after comprehensive reinforcement has a strong supporting capacity, and effectively reduces the secondary lining pressure of the tunnel, which plays a vital role in the long-term service of the secondary lining.

In this paper, the tunnel temperature (air, structure, and rock) and wall heat flow were monitored in-situ for 63 days in cold and warm terms. The in-situ monitoring results reveal that the traffic-induced thermal effect (TTE) is one of the main factors inducing tunnel temperature variation. The tunnel wall temperature change rate caused by traffic-induced thermal effect is 20.53–43.18% in the cold term. A simplified analytical solution for the tunnel wall temperature is proposed based on Green's function and verified by the monitoring data in this study. The influence coefficients of traffic flow formed by different vehicle types on the tunnel wall temperature in the cold and warm terms are obtained using an optimized machine learning method. The impact ratio is LDV:HDV: HDT = 6:1:1 in the cold (that of LDT is minimal); LDV:HDV:LDT: HDT = 1:8:4:10 in the warm. The assumed tunnel heat absorption ratio (THAR) ranges from 0.19 to 2.05% in the cold term and −0.22 to 0.52% in the warm. The thermal loss degree in tunnel lining is lower than that of ignoring the traffic-induced thermal effect in the cold term. TTE increases THAR by about 1.45% and partly delays the heat dissipation of the tunnel wall. Furthermore, a dynamic network model with vehicles as moving heat sources is established to investigate the thermal effect of different vehicle types. The study results provide reference to new ideas and calculation solutions for designing cold-region highway tunnels and implementing new energy utilization systems.

Wheat is one of the most important food crops in the world and is considered one of the top targets in crop biotechnology. With the high-quality reference genomes of wheat and its relative species and the recent burst of genomic resources in Triticeae, demands to perform gene functional studies in wheat and genetic improvement have been rapidly increasing, requiring that production of transgenic wheat should become a routine technique. While established for more than 20 years, the particle bombardment-mediated wheat transformation has not become routine yet, with only a handful of labs being proficient in this technique. This could be due to, at least partly, the low transformation efficiency and the technical difficulties. Here, we describe the current version of this method through adaptation and optimization. We report the detailed protocol of producing transgenic wheat by the particle gun, including several critical steps, from the selection of appropriate explants (i.e., immature scutella), the preparation of DNA-coated gold particles, and several established strategies of tissue culture. More importantly, with over 20 years of experience in wheat transformation in our lab, we share the many technical details and recommendations and emphasize that the particle bombardment-mediated approach has fewer limitations in genotype dependency and vector construction when compared with the Agrobacterium-mediated methods. The particle bombardment-mediated method has been successful for over 30 wheat genotypes, from the tetraploid durum wheat to the hexaploid common wheat, from modern elite varieties to landraces. In conclusion, the particle bombardment-mediated wheat transformation has demonstrated its potential and wide applications, and the full set of protocol, experience, and successful reports in many wheat genotypes described here will further its impacts, making it a routine and robust technique in crop research labs worldwide.

Background Myocardial infarction (MI) is characterized by the emergence of dead or dying cardiomyocytes and excessive immune cell infiltration after coronary vessel occlusion. However, the complex transcriptional profile, pathways, cellular interactome, and transcriptional regulators of immune subpopulations after MI remain elusive. Methods and Results Here, male C57BL/6 mice were subjected to MI surgery and monitored for 1 day and 7 days, or sham surgery for 7 days, then cardiac CD45‐positive immune cells were collected for single‐cell RNA sequencing to determine immune heterogeneity. A total of 30 135 CD45+ immune cells were partitioned into macrophages, monocytes, neutrophils, dendritic cells, and T or B cells for further analysis. We showed that macrophages enriched for Olr1 and differentially expressed Gpnmb represented 2 crucial ischemia‐associated macrophages with distinct proinflammatory and prophagocytic capabilities. In contrast to the proinflammatory subset of macrophages enriched for Olr1, Gpnmb‐positive macrophages exhibited higher phagocytosis and fatty acid oxidation preference, which could be abolished by etomoxir treatment. In addition to macrophages, MI triggered prompt recruitment of neutrophils into murine hearts, which constituted the sequential cell‐fate from naïve S100a4‐positive, to activated Sell‐high, to aging Icam1‐high neutrophils. In silico tools predicted that the excessively expanded neutrophils at 1 day were attributed to chemokine C‐C motif ligand/chemokine C‐X‐C motif ligand pathways, whereas CD80/inducible T‐cell costimulator (ICOS) signaling was responsible for the immunosuppressive response at day 7 after MI. Finally, the Fos/AP‐1 (activator protein 1) regulon was identified as the critical regulator of proinflammatory responses, which was significantly activated in patients with dilated cardiomyopathy and ischemic cardiomyopathy. We showed the enriched Fos/AP‐1 target gene loci in genome‐wide association study signals for coronary artery diseases and MI. Targeting Fos/AP‐1 with the selective inhibitor T5224 blunted leukocyte infiltration and alleviated cardiac dysfunction in the preclinical murine MI model. Conclusions Taken together, this single‐cell RNA sequencing data lay the groundwork for the understanding of immune cell heterogeneity and dynamics in murine ischemic hearts. Moreover, Fos/AP‐1 inhibition mitigates inflammatory responses and cardiac dysfunction, which might provide potential therapeutic benefits for heart failure intervention after MI.

Tannic acid (TA) has been recently considered as a new dough additive for improving the bread-making quality of wheat. However, the effects of TA supplementation on the sensory quality parameters (color, crumb grain structure, and sensory properties) of bread have not been studied. Further, the potential of TA supplementation in bread-making quality improvement has not been evaluated by using commercial flour. In the present study, three commercial wheat flours (namely, XL, QZG, and QZZ) with different gluten qualities were used to evaluate the effects of TA supplementation (in concentrations of 0.1% and 0.3%, respectively). TA supplementation did not change the proximate composition of the breads but increased the volumes and specific volumes of XL and QZG breads. TA supplementation enhanced antioxidant activities, with 0.3% TA significantly increasing the antioxidant capacities of bread made from all three flour samples by approximately four-fold (FRAP method)/three-fold (ABTS method). Positive effects of TA on the reduction in crumb hardness, gumminess, and chewiness were observed in the XL bread, as determined by the texture profile analysis. For the analyses on visual and sensory attributes, our results suggest that TA did not affect the crust color, but only slightly reduced the L* (lightness) and b* (yellowness) values of the crumb and increased the a* (redness) value. TA supplementation also increased the porosity, total cell area, and mean cell area. Satisfactorily, the sensory evaluation results demonstrate that TA-supplemented breads did not exhibit negative sensory attributes when compared to the non-TA-added breads; rather, the attributes were even increased. In summary, TA-supplemented breads generally had not only better baking quality attributes and enhanced antioxidant activities, but, more importantly, presented high consumer acceptance in multiple commercial flour samples. Our results support the commercial potential of TA to be used as a dough improver.

Abstract The meteorological data such as temperature of the upper atmosphere is ssential for accurate weather forecasting. The Universal Rawinsonde Observation Program (RAOB) establishes an extensive radiosonde network worldwide to observe atmospheric meteorological data from the surface to the low stratosphere. The RAOB data data has very high accuracy but can offer a very limited spatial coverage. Meanwhile, ERA-Interim reanalysis data is widely available but with low-quality. We propose a 4D spatiotemporal statistical model which can make effective inferences from ERA-Interim reanalysis data to RAOB data. Finally, we can obtain a huge amount of RAOB data with high-quality and can offer a very wide spatial coverage. In empirical research, we collected data from 200 launch sites around the world in January 2015. The 4D spatiotemporal statistical model successfully analyzed the observation gaps at different pressure levels.

Craniosynostosis is a premature fusion of cranial sutures, resulting in abnormally shaped skull and brain development disorder. The description of craniosynostosis in patients with BCL11B mutations is rare. Here, we firstly report a 25-month-old Chinese boy with a novel frameshift variant in BCL11B gene. The patient was identified c.2346_2361del by whole-exome sequencing and was confirmed to be de novo by parental Sanger sequencing. This patient presented clinical phenotype of craniosynostosis as well as global developmental delay. He had a small mouth, thin upper lip, arched eyebrows, a long philtrum, midfacial hypoplasia and craniosynostosis. Brain MRI showed brain extracerebral interval and myelination changes, and brain CT with 3D reconstruction showed multi-craniosynostosis. Our study expands the clinical phenotypes of patients with BCL11B gene mutation, and our findings may help guide clinical treatment and family genetic counseling.

Acute kidney injury (AKI) is associated with high risk of mortality, post-disease renal fibrosis, kidney dysfunction and renal failure. Renal macrophages play a key role in the pathogenesis (M1 subpopulation), healing and remodeling (M2 subpopulation) in AKI and, thus, have been a promising target for clinical treatment of AKI. Here, in a mouse renal ischemia/reperfusion injury (IRI) model for AKI, we showed that renal macrophages could be further classified into Clec7a+ M1 macrophages, Clec7a- M1 macrophages, Clec7a+ M2 macrophages and Clec7a- M2 macrophages, representing distinct macrophage populations with different functionality. Interestingly, Clec7a+ M1 macrophages exhibited potent pro-inflammatory and phagocytic effects compared to Clec7a- M1 macrophages, while Clec7a- M2 macrophages exhibited better proliferating and migrating potential, which is critical for their role in tissue repairing after injury. These data from mice were further strengthened by bioinformatics analyses using published database. In vivo, combined expression of Clec7a in M1 macrophages and depletion of Clec7a in M2 macrophages significantly improved the renal function after IRI-AKI. Together, our data suggest that Clec7a is crucial for the fine regulation of macrophage phenotype during AKI and could be a novel target for boosting clinical therapy.

Abstract Aims Prostaglandin E2 (PGE2) is involved in the development of cardiac hypertrophy. However, whether PGE2 regulates the chronic kidney disease‐associated cardiac hypertrophy and the tentative mechanism remains to be elucidated. Methods and results We explored the effect of PGE2 receptor inhibitors on cardiac hypertrophy in vitro and in a 5/6 nephrectomy (5/6NT) rat model using quantitative reverse transcription polymerase chain reaction, western blotting, enzyme‐linked immunosorbent assay, immunohistochemical staining, and immunofluorescence staining assays. The result showed that EP2 and EP4 receptors were both up‐regulated in the PGE2‐treated cardiomyocyte cells. PGE2 treatment enhanced active β‐catenin (non‐phosphorylated) signalling through mediating EP2 and EP4 receptors. Interestingly, inhibition of EP2 receptor suppressed PGE2‐induced cardiomyocyte hypertrophy and cardiac fibrosis‐related proteins in vitro. In the 5/6NT rat model, the increased secretion PGE2 was identified in the 5/6NT rat model for 2 weeks (P = 0.0251). EP2 receptor inhibitor administration significantly improved the cardiac function and fibrosis in 5/6NT rats. Conclusions Our study demonstrated that inhibition of EP2 receptor could improve PGE2‐induced cardiac hypertrophy in 5/6NT rats. The exploration of these mechanisms may contribute to the optimization of therapy in chronic kidney disease accompanied cardiac hypertrophy in clinic.

Hemodialysis (HD) is the main treatment modality for patients with end-stage kidney disease. Cardiovascular diseases (CVD) are highly prevalent in HD patients and are the leading cause of death in this population, with the mortality from CVD approximately 20 times higher than that of the general population. Traditional and non-traditional cardiovascular risk factors accelerate progression of CVD and exacerbate the prognosis in HD patients. This review provides a brief overview of the characteristics of CVD in HD patients, and a description of advances in its management.

Abstract Background DKC1, the dyskerin encoding gene, functions in telomerase activity and telomere maintenance. DKC1 mutations cause a multisystem disease, dyskeratosis congenita (DC), which is associated with immunodeficiency and bone marrow failure. Methods In this research, we reported a novel intronic mutation of DKC1 causing dyskerin functional loss in a Chinese family. Whole exome sequence (WES) of the proband and validation by sanger sequencing help us identify a pathogenic DKC1 mutation. Minigene splicing assays were performed to evaluate functional change of DKC1. Results A pathogenic DKC1 intronic mutation(c.84 + 7A > G) was identified in the proband, which was inherited from heterozygous mother and not reported before. We detected the novel transcript with a 7 bp intron retention through minigene splicing assay. The newly spliced transcript is so short that would be degraded by nonsense‐mediated mRNA decay in vitro and we infer that the novel DKC1 mutation would influences normal physiological function of dyskerin. Conclusions Our study identified a novel intronic mutation, which expands the spectrum of pathogenic DKC1 gene mutations and can be used in molecular diagnosis. The mutant allele was transmitted to the next generation with high frequency in the family and causes still birth or early death.

Abstract Plant roots and soil prokaryotes primarily interact with each other in the rhizosphere. Changes in the rhizosphere prokaryotic structure are influenced by several factors. In this study, the community structure of the Potentilla anserina L. rhizosphere prokaryotes was identified and evaluated by high-throughput sequencing technology in different continuous cropping fields and developmental stages of the plant. In total, 2 archaeal (Euryarchaeota and Thaumarchaeota) and 26 bacterial phyla were identified in the P. anserina rhizosphere. The bacterial community was mainly composed of Acidobacteria, Actinobacteria, Bacteroidetes, Chloroflexi, Gemmatimonadetes, Planctomycetes, Proteobacteria, and Verrucomicrobia. Moreover, the prokaryotic community structure of the rhizosphere varied significantly during plant development. Our results provide new insights into the dynamics of the P. anserina rhizosphere prokaryotic community and may provide useful information for enhancing the growth and development of P. anserina through artificial control of the soil prokaryotes.

Background Intradialytic hypotension (IDH) is a common complication in maintaining hemodialysis (MHD) patients. Immune activation might be part of the mechanisms. However, the association between pro-inflammatory cytokines and blood pressure (BP) has not been deeply explored. So we aim to evaluate the potential role of pro-inflammatory cytokines in IDH. Methods MHD patients starting hemodialysis before January 2016 were enrolled in our retrospective study. Patients' characteristics, laboratory results, and intradialytic BP were collected. IDH was defined as nadir systolic BP ≤ 90 mmHg during hemodialysis. The definition of IDH group was that those who suffered from more than one hypotensive event during one month after the enrollment (10% of dialysis treatments). Spearman correlation analysis and logistic regression were employed to explore the relationship between pro-inflammatory cytokines and IDH. Results Among 390 patients, 72 were identified with IDH (18.5%). High levels of serum tumor necrosis factor-α (TNF-α) and interleukin-1β (IL-1β) were observed in the IDH group (p
