The detection of antimitochondrial antibodies (AMA) is the specific diagnostic marker for primary biliary cholangitis. Indeed, it is the most specific autoantibody in clinical autoimmunity, with a high titer directed response to the inner lipoyl domain of PDC-E2. The current international reference for AMA detection is based upon sera samples of PBC patients. In rheumatic diseases, i.e. rheumatoid arthritis, great efforts are placed at development of international standards. In this study, we report the development of a monoclonal chimeric IgG1 antibody as a reference for AMA testing. A monoclonal 4G6 antibody was constructed from a murine monoclonal antibody specific for the inner lipoyl domain (ILD) of PDC-E2, by combining the variable region with the constant region of human IgG1. The 4G6 antibody recognizes all AMA epitopes containing the ILD of PDC-E2, including the classical BPO recombinant antigen in all currently available diagnostic methods. The binding affinity of the 4G6 antibody to PDC-E2 and BPO antigen reaches KD value of 7.22 × 10−11 M and 4.55 × 10−11 M, which is sufficient to use as a quantitative reference for all AMA tests. The unlimited availability of the 4G6 antibody makes it a promising candidate for use as an AMA reference or assay calibrator for the international community.

Wildfires cause great losses to the ecological environment, economy, and people’s safety and belongings. As a result, it is crucial to establish wildfire susceptibility models and delineate fire risk levels. It has been proven that the use of remote sensing data, such as meteorological and topographical data, can effectively predict and evaluate wildfire susceptibility. Accordingly, this paper converts meteorological and topographical data into fire-influencing factor raster maps for wildfire susceptibility prediction. The continuous convolutional neural network (CCNN for short) based on coordinate attention (CA for short) can aggregate different location information into channels of the network so as to enhance the feature expression ability; moreover, for different patches with different resolutions, the improved CCNN model does not need to change the structural parameters of the network, which improves the flexibility of the network application in different forest areas. In order to reduce the annotation of training samples, we adopt an active learning method to learn positive features by selecting high-confidence samples, which contributes to enhancing the discriminative ability of the network. We use fire probabilities output from the model to evaluate fire risk levels and generate the fire susceptibility map. Taking Chongqing Municipality in China as an example, the experimental results show that the CA-based CCNN model has a better classification performance; the accuracy reaches 91.7%, and AUC reaches 0.9487, which is 5.1% and 2.09% higher than the optimal comparative method, respectively. Furthermore, if an accuracy of about 86% is desired, our method only requires 50% of labeled samples and thus saves about 20% and 40% of the labeling efforts compared to the other two methods, respectively. Ultimately, the proposed model achieves the balance of high prediction accuracy and low annotation cost and is more helpful in classifying fire high warning zones and fire-free zones.

Fresh-cut potatoes (Solanum tuberosum L.) are susceptible to browning and microbial contamination during storage. In this study, the effects of trans-2-hexenal (E2H), ascorbic acid (VC), dimethyl dicarbonate (DMDC), and the combined treatment of E2H, VC, and DMDC on quality deterioration in fresh-cut potatoes were investigated. The response surface methodology (RSM) demonstrated that E2H, VC, and DMDC concentrations of 0.010%, 0.65%, and 240 mg/L, respectively, were the optimum conditions for fresh-cut potato preservation. Further analysis showed that the combined treatment of E2H, VC, and DMDC was the most effective method of reducing quality deterioration in potatoes compared to the control and individual treatments. Furthermore, the combined treatment of E2H, VC, and DMDC could decrease the accumulation of reactive oxygen species (ROS) via improving antioxidant enzyme activities. Meanwhile, energy-metabolism-related enzyme activities and glutamate decarboxylase (GAD) activity were enhanced, while γ-aminobutyric acid transaminase (GABA-T) activity was reduced via the combined treatment of E2H, VC, and DMDC, which contributed to maintaining high energy levels and GABA content in potatoes. These findings suggested that the combined treatment of E2H, VC, and DMDC could protect membrane integrity through enhancing antioxidant capacity, energy levels, and GABA content to maintain quality in fresh-cut potatoes.

To study the effect of chlorine dioxide fumigation on the preservation of postharvest broad beans, broad beans were fumigated with different concentrations of ClO2 (0, 15, 30 and 60 μL/L), stored at room temperature and evaluated for color difference, firmness, mass loss percentage, browning index and decay index after 0, 2, 4, 6 and 8 days. The results showed that compared to the control group, ClO2, at all concentrations tested, could inhibit the increase in mass loss percentage, browning index and decay index, and delay the decline in firmness, especially at a concentration of 30 μL/L. Furthermore, 30 μL/L ClO2 fumigation significantly reduced the respiratory intensity, inhibited chlorophyll degradation and malondialdehyde accumulation in broad beans during the 8-day storage period, suppressed the increase in superoxide anion generation rate and the accumulation of hydrogen peroxide, and maintained higher activities of superoxide dismutase (SOD) and catalase (CAT), suggesting that ClO2 fumigation could alleviate oxidative damage to the cell membrane. In addition, ClO2 treatment repressed the activities of polyphenol oxidase (PPO) and peroxidase (POD), maintained the activity of phenylalanine aminolyase (PAL) and the content of total phenols, and delayed the occurrence of browning. Moreover, ClO2 treatment increased the contents of vitamin C, soluble protein and starch, and inhibited the increase of the reducing sugar content in broad beans (P < 0.05), thus maintaining better quality of broad beans. In conclusion, 30 μL/L ClO2 fumigation effectively maintained the edible value of broad beans and improved the storage quality while enhancing the antioxidant capacity.

PurposeOral health plays an important role in overall health. But there is scarce information available on oral health behavior and oral health service utilization among cancer patients. This study aimed to evaluate oral health behavior and oral health service utilization among different population groups of cancer patients in China.MethodsA multicenter cross-sectional study in three tertiary hospitals was conducted to explore the oral health behaviors and oral health service utilization of 162 cancer patients in China.ResultsWe investigated a total of 162 cancer patients, 81 from urban and rural areas, respectively. The participant’s ages ranged from 18 and 82 years, mean age was 44.62 years (SD = 15.72). Overall, cancer patients have poor oral health behaviors and limited oral health service utilization. There were statistically significant differences (p < 0.05) between urban and rural cancer patients in terms of oral health behaviors, including brushing methods, the use of fluoride toothpaste, the use of dental floss, dental caries, and bleeding gums while brushing teeth. As for oral health service utilization, there were significant differences (p < 0.05) between urban and rural cancer patients on regular dental cleaning, the reasons for visiting a dental clinic, and whether they took the initiative to learn about oral health.ConclusionThe study findings suggest that cancer patients had poor oral health behaviors and limited oral health service utilization, and rural patients perform poorer than their urban counterparts. Oral health education should be provided to cancer patients to improve their oral health behaviors and oral health service utilization.

Backgroud/Objectives: Ferula spp. is an essential crop in Central Asia with pronounced economic benefits governed by its flowering process. However, the mechanisms of the flowering phenotype remain unclear. Methods: In this study, using F. sinkiangensis as a model plant, we integrated transcriptome, proteome, and metabolome analyses to compare the multilayer differences in leaves and roots of plants with flowering and unflowering phenotypes. Results: We found that several variations in the transcriptome, proteome, and metabolome were closely associated with flowering. The Photosynthesis and Phenylpropanoid biosynthesis pathways in plants with the flowering phenotype were more active. Additionally, three flowering genes, named FL2–FL4, were upregulated in the leaves of flowering plants. Notably, six transcription factors were potentially responsible for regulating the expression of FL2–FL4 in the leaves to mediate flowering process of F. sinkiangensis. Moreover, genes relevant to Photosynthesis and Phenylpropanoid biosynthesis were also involved in regulating the expression of FL2–FL4 in flowering plants. Conclusions: The active regulation network together with Photosynthesis and Phenylpropanoid biosynthesis were essential for inducing the expression of flowering-related genes in leaves to promote the flowering process of F. sinkiangensis. Ferula is a genus of flowering plants known for its edible and medicinal properties. Since ancient times, many species of Ferula have been used in traditional medicine to treat various health issues across countries, such as digestive disorders, respiratory problems, and even as a remedy for headaches and toothaches. In addition, they are also used as a flavoring agent in various cuisines. As the main active ingredients in Ferula, sesquiterpenes and their derivatives, especially sesquiterpene coumarins, sesquiterpene phenylpropanoids, and sesquiterpene chromones, have attracted the attention of scientists due to the diversity of their chemical structures, as well as their extensive and promising biological properties, such as antioxidative, anti-inflammatory, antibacterial properties. However, there has not been a comprehensive review of sesquiterpenes and their derivatives from this plant. This review aims to provide an overview of the chemical structures, biosynthetic pathways, and biological properties of sesquiterpenes and sesquiterpene derivatives from Ferula, which may help guide future research directions and possible application methods for this valuable edible and medicinal plant.

The present study sought to examine how sea buckthorn (SBT) oil impacts mitochondrial and overall functionality in Aspergillus flavus. In order to assess the effect of SBT oil, it was used to treat mycelia across a range of concentrations, after which mitochondrial structures were imaged via transmission electron microscopy (TEM). In order to explore the functional impact of this treatment, we additionally examined reactive oxygen species (ROS) production, malondialdehyde (MDA) levels, enzyme activity, and shifts in mitochondrial membrane potential (ΔΨm) following treatment. We found that SBT oil induced mitochondrial damage in A. flavus in a dose-dependent manner, resulting in altered succinate dehydrogenase (SDH) and adenosine triphosphatase (ATPase) activity, reduced ΔΨm levels, and markedly elevated MDA and ROS levels. Together, these findings suggest that SBT oil can mediate antifungal activity against A. flavus through a mechanism associated with its ability to disrupt the tricarboxylic acid (TCA) cycle and mitochondrial potential, leading to MDA and ROS accumulation.

Ferula sinkiangensis K. M. Shen (Apiaceae) is distributed in arid desert areas of Xinjiang, and its resin is a traditional Chinese medicine to treat gastrointestinal digestive diseases. To explore bioactive components from F. sinkiangensis, three new lignans and thirteen known components were isolated. The structural elucidation of the components was established utilizing spectroscopic analyses together with ECD calculations. Griess reaction results indicated new compounds 1 and 2 significantly decreased NO production in LPS-stimulated RAW 264.7 macrophages, and ELISA results indicated that they effectively attenuated LPS-induced inflammation by inhibiting TNF-α, IL-1β, and IL-6 expressions. The in silico approach confirmed that compound 1 docked into the receptors with strong binding energies of −5.84~−10.79 kcal/mol. In addition, compound 6 inhibited the proliferation of AGS gastric cancer cells with IC50 values of 15.2 μM by suppressing the cell migration and invasion. This study disclosed that F. sinkiangensis might be a promising potential resource for bioactive components.

BackgroundRoutine administration of adjuvant chemotherapy for stage IB non-small cell lung cancer (NSCLC) remains controversial. To our knowledge, no available studies have assessed the outcomes of chemotherapy in patients with stage IB NSCLC who had prior malignancies.MethodsPatients with pathological stage IB NSCLC with previous malignancies who underwent surgery between 2004 and 2015 were identified from the Surveillance, Epidemiology, and End Results (SEER) database. The patients were categorized into chemotherapy and observation group based on whether they received adjuvant chemotherapy. Propensity score matching was performed to reduce confounding bias, and Kaplan–Meier curves and log-rank tests were used to compare overall survival (OS) and cancer-specific survival (CSS) between the two groups. Subgroup analyses of the matched cohorts were then conducted to evaluate the relationship between clinical features and chemotherapy.ResultsA total of 894 eligible patients were identified; 90 (10.1%) patients received postoperative chemotherapy. Patients who received adjuvant chemotherapy obtained obvious OS benefits compared with those who received observation alone (HR = 0.68, 95% CI: 0.48–0.97, P = 0.031). In addition, the 5-year OS rate and median OS time in the chemotherapy group were higher and longer, respectively. Although chemotherapy offered no obvious benefits for CSS (HR = 0.80, 95% CI: 0.57–1.14, P = 0.35), patients who received chemotherapy showed a better 5-year CSS rate. On subgroup analyses, a chemotherapy advantage was observed in advanced age (≥65 years, HR = 0.62, 95% CI: 0.38–0.99, P = 0.045). The same chemotherapy advantages were observed in patients diagnosed with higher histological grades (poorly differentiated to undifferentiated) (HR = 0.56, 95% CI: 0.33–0.96, P = 0.033) and tumor sizes >3.1–4 cm (HR = 0.57, 95% CI: 0.37–0.87, P = 0.010). Interestingly, NSCLC patients with previous malignancies originating from the kidney and bladder (HR = 0.34, 95% CI: 0.12–0.99, P = 0.049) showed a chemotherapy advantage. The same chemotherapy advantages were observed in patients diagnosed with NSCLC within 3 to 5 years after prior cancers (HR = 0.39, 95% CI: 0.16–0.98, P = 0.044) and with localized SEER stage of prior cancers (HR = 0.49, 95% CI: 0.29–0.86, P = 0.012).ConclusionThese findings indicate that adjuvant chemotherapy may improve long-term outcomes for stage IB NSCLC patients with previous malignancies. It is recommended that physicians consider the clinical features of previous cancers when making adjuvant chemotherapy decisions for these patients.

There are a large number of earthquake networks under the management of cities and counties，which have made historical contributions to reducing earthquake disasters and promoting the development of earthquake prevention and disaster reduction. In recent years，the development of earthquake networks managed by cities and counties lags behind the development level of the same industry，exposing problems such as unclear positioning，low efficiency and cognitive deviation. Taking Yangzhou Earthquake Networks Center as an example，the advantages and disadvantages before and after the institutional reform in 2019 are summarized and compared. The objectives and tasks of the modernization development of earthquake prevention and disaster reduction are combined. In order to promote the development of work，this paper puts forward suggestions from four aspects：self-orientation，work implementation，coordination of multiple relationships，scientific understanding of reform and development.

BackgroundBladder carcinoma (BLCA) is a common malignant tumor with high morbidity and mortality in the urinary system. Pyroptosis is a pattern of programmed cell death that is closely associated with progression of tumors. Therefore, it is significant to probe the expression of pyroptosis-related genes (PRGs) in BLCA.MethodsThe differentially expressed genes in normal and BLCA tissues were first obtained from the Cancer Genome Atlas (TCGA) database analysis, as well as PRGs from the National Center for Biotechnology Information (NCBI) database, intersecting to obtain differentially expressed pyroptosis-related genes (DEPRGs) in BLCA. With the construction of a prognostic model of pyroptosis by regression analysis, we derived and validated key genes, which were ascertained as a separate prognostic marker by individual prognostic and clinical relevance analysis. In addition, we gained six immune cells from the Tumor Immune Evaluation Resource (TIMER) website and analyzed the relationship between pyroptosis prognostic genes and immune infiltration.ResultOur results revealed that 31 DEPRGs were available by comparing normal and BLCA tissues with |log2 (fold change, FC)| > 0.5 and FDR

Hop (Humulus lupulus) is a perennial plant with commercial values. Here, we reported the complete chloroplast genome for a local hop cultivar (Humulus lupulus cv. ‘Fubei-1’) from Xinjiang, China. The chloroplast genome is 153,614 bp long with an A + T-biased base composition, and contains a total of 113 gene species, including 79 protein-coding, 30 tRNA, and four rRNA gene species. Nineteen gene species are duplicated, and 18 gene species harbor one or two introns. Phylogenetic analysis revealed close relatedness among the three hop cultivars (‘Saazer’, ‘Hallertauer’, and ‘Fubei-1’).

In public use of a large display, it is a usual phenomenon that multiple users individually participate in respective tasks on a common device. Previous studies have categorized such activity as independent interaction that involves little group engagement. However, by investigating how users approach, participate in, and interact with large displays, we found that parallel use is affected by group factors such as group size and between-user relationship. To gain a thorough understanding of individual and group behaviors, as well as parallel interaction task performance, one 70-inch display-based information searching task and experiment was conducted, in which a mobile eye movement tracking headset and a motion sensing RGB-depth sensor were simultaneously applied. The results showed that (1) a larger group size had a negative influence on the group users’ concentration on the task, perceived usability, and user experience; (2) a close relationship between users contributed to occasional collaborations, which was found to improve the users’ task completion time efficiency and their satisfaction on the large display user experience. This study proves that an integrated application of eye movement tracking and motion sensing is capable of understanding individual and group users’ behaviors simultaneously, and thus is a valid and reliable scheme in monitoring public activities that can be widely used in public large display systems.

Ferula sinkiangensis K. M. Shen is a valuable traditional Chinese medicine historically used to treat stomachache and rheumatoid arthritis. The chloroplast genome of Ferula genus plant has not been previously reported. This study reported the complete chloroplast genome sequence of F. sinkiangensis based on high-throughput sequencing. The genome was 166,583 bp in length, containing a small single-copy (SSC) region of 17,595 bp and a large single-copy (LSC) region of 85,242 bp, separated by two inverted repeats (IRs) of 31,873 bp, each. The genome contained 114 unique genes, including 80 protein-coding genes (PCGs), four rRNA genes, and 30 tRNA genes. In addition, 17 genes contained one or two introns, including nine PCG genes with a single intron, two PCG genes harboring two introns, and six tRNA genes harboring a single intron. In this study, F. sinkiangensis K. M. had the closest genetic relationship with Torilis scabra and clustered with the Umbelliferae family species.

The wildlife re-identification recognition methods based on the camera trap were used to identify different individuals of the same species using the fur, stripes, facial features and other features of the animal body surfaces in the images, which is an important way to count the individual number of a species. Re-identification of wild animals can provide solid technical support for the in-depth study of the number of individuals and living conditions of rare wild animals, as well as provide accurate and timely data support for population ecology and conservation biology research. However, due to the difficulty of recording the shy wild animals and distinguishing the similar fur of different individuals, only a few papers have focused on the re-identification recognition of wild animals. In order to fill this gap, we improved the locally aware transformer (LA transformer) network structure for the re-identification recognition of wild terrestrial animals. First of all, at the stage of feature extraction, we replaced the self-attention module of the LA transformer with a cross-attention block (CAB) in order to calculate the inner-patch attention and cross-patch attention, so that we could efficiently capture the global information of the animal body’s surface and local feature differences of fur, colors, textures, or faces. Then, the locally aware network of the LA transformer was used to fuse the local and global features. Finally, the classification layer of the network realized wildlife individual recognition. In order to evaluate the performance of the model, we tested it on a dataset of Amur tiger torsos and the face datasets of six different species, including lions, golden monkeys, meerkats, red pandas, tigers, and chimpanzees. The experimental results showed that our wildlife re-identification model has good generalization ability and is superior to the existing methods in mAP (mean average precision), and obtained comparable results in the metrics Rank 1 and Rank 5.

Abstract Background Accumulating evidence suggests that serum vascular endothelial growth factor (VEGF) in many neurobiological processes potentially contributes to the pathophysiology of psychiatric disorders, particularly cognitive decline. The purpose of this study was to explore the differences in neurocognition, social cognition and VEGF among remitted first-episode schizophrenic patients, non-remitters and normal control subjects. Moreover, we investigated the association between serum VEGF levels and cognitive functions. Method 65 remission (RS) and 45 nonremission patients (NRS) after first-episode schizophrenia, as well as 58 healthy controls (HC) were enrolled in this study. Social cognition was assessed using the Chinese Facial Emotion Test (CFET); neurocognition was measured with a test battery consisting of Hopkins Verbal Learning Test-Revised, Verbal Fluency Test, Trail Making Tests, Digit Span Tests (DST) and Stroop Tests. Blood samples were collected for VEGF measurements. Data was analyzed with SPSS 22.0 (Chicago, IL, USA). Results On nearly all neurocognitive tests (except for DST), RS performed significantly worse than HC but better than NRS (P RS > NRS). Conclusion Compared to HC, RS demonstrated poorer neurocognitive but intact social cognition functioning. These results indicate that VEGF levels decreased gradually with the severity of cognitive impairment in schizophrenia. VEGF may be involved in the pathological mechanism of cognitive performance in RS.

Although low-temperature storage could maintain the quality of fruits and vegetables, it may also result in chilling injury (CI) in cold-sensitive produce, such as cucumbers. This can seriously affect their quality.” The antioxidant capacity, energy metabolism and proline metabolism of cucumbers treated with hydrogen sulfide (H2S) were studied in this assay. The outcomes displayed that H2S treatment effectively reduced CI and delayed the increase in electrolyte leakage (EL) and malondialdehyde (MDA) content. In addition, the H2S-treated cucumber fruit exhibited higher L* and hue angle values, as well as nutrients such as ascorbic acid (AsA). The H2S-treated fruit showed lower levels of reactive oxygen species (ROS) and higher antioxidant enzyme activities. Meanwhile, H2S treatment also increased the activities of the essential enzymes involved in energy metabolism, including cytochrome C oxidase (CCO), succinate dehydrogenase (SDH), H+-ATPase and Ca2+-ATPase, which improved the energy supply. H2S induced higher ornithine δ-aminotransferase (OAT) and Δ-1-pyrroline-5-carboxylate synthetase (P5CS) activities, and reduced proline dehydrogenase (PDH) activity, promoting the accumulation of proline. These results indicated that H2S could alleviate CI in the cucumber fruit by modulating antioxidant capacity, energy metabolism and proline metabolism, thereby extending the shelf life of postharvest cucumbers.

Camera traps are widely used in wildlife research, conservation, and management, and abundant images are acquired every day. Efficient real-time instance segmentation networks can help ecologists label and study wild animals. However, existing deep convolutional neural networks require a large number of annotations and labels, which makes them unsuitable for small datasets. In this paper, we propose a two-stage method for the instance segmentation of wildlife, including object detection and contour approximation. In the object detection stage, we use FSOD (few-shot object detection) to recognize animal species and detect the initial bounding boxes of animals. In the case of a small wildlife dataset, this method may improve the generalization ability of the wild animal species recognition and even identify new species that only have a small number of training samples. In the second stage, deep snake is used as the contour approximation model for the instance segmentation of wild mammals. The initial bounding boxes generated in the first stage are input to deep snake to approximate the contours of the animal bodies. The model fuses the advantages of detecting new species and real-time instance segmentation. The experimental results show that the proposed method is more suitable for wild animal instance segmentation, in comparison with pixel-wise segmentation methods. In particular, the proposed method shows a better performance when facing challenging images.

Pasteurella multocida can invade and translocate through endothelial cells and result in vascular-system infection, which can cause severe economic losses in the poultry industry. Antibacterial therapy (especially florfenicol) plays an important part in controlling P. multocida infection. To preserve the effect of florfenicol, in vivo pharmacokinetic/pharmacodynamic (PK/PD) modeling of florfenicol against three P. multocida strains in duck was established. Then, the efficacy of the currently marketed dose, a rational dosage regimen for populations, and the PK/PD cutoff were predicted through Monte Carlo simulations (MCSs). The area under the concentration–time curve from 0 to 24 h/minimum inhibitory concentration (AUC0–24 h/MIC) was the optimal PK/PD parameter. The PK/PD surrogate values of florfenicol against P. multocida were similar using different organs as the PD target, but varied in different strains. For the florfenicol-sensitive strain 0825Y1, when the AUC0–24 h/MIC reached 117.54 and 108.19, florfenicol showed a bactericidal effect in the liver and lung, respectively. For the florfenicol-sensitive strain 0901J1, the corresponding value was 78.39 and 54.30, respectively. For the florfenicol-resistant strain JY160110, florfenicol could attain a maximum effect of 1 – log10 reduction in bacteria in the liver and lung when the AUC0–24 h/MIC reached 2.03 and 2.06, respectively. The PK/PD-based prediction for the population dose indicated a poor effect for the low end of the currently marketed dose (40 mg/kg body weight per day), but a robust effect for the high end of the currently marketed dose (60 mg/kg body weight per day) with a target attainment rate of 92.79% and 81.44% against P. multocida in mainland China and worldwide, respectively. The recommended dose optimized by MCSs was 52 mg/kg body weight in mainland China. The PK/PD cutoff of florfenicol against P. multocida at the low end and high end of the current daily dose (40 and 60 mg/kg body weight) and predicted daily dose in mainland China (52 mg/kg body weight) was 0.25, 4, and 0.5 μg/ml, respectively. These results suggested that more than one strain should be involved for PK/PD modeling and contributed to rational use of florfenicol in populations. We also provided fundamental data for determination of florfenicol breakpoints in poultry.
