Abstract Advancements in human-engineered heart tissue have enhanced the understanding of cardiac cellular alteration. Nevertheless, a human model simulating pathological remodeling following myocardial infarction for therapeutic development remains essential. Here we develop an engineered model of myocardial repair that replicates the phased remodeling process, including hypoxic stress, fibrosis, and electrophysiological dysfunction. Transcriptomic analysis identifies nine critical signaling pathways related to cellular fate transitions, leading to the evaluation of seventeen modulators for their therapeutic potential in a mini-repair model. A scoring system quantitatively evaluates the restoration of abnormal electrophysiology, demonstrating that the phased combination of TGFβ inhibitor SB431542, Rho kinase inhibitor Y27632, and WNT activator CHIR99021 yields enhanced functional restoration compared to single factor treatments in both engineered and mouse myocardial infarction model. This engineered heart tissue repair model effectively captures the phased remodeling following myocardial infarction, providing a crucial platform for discovering therapeutic targets for ischemic heart disease.

The Yangtze River is the mother river of China. To promote the aquatic ecosystem protection of the great river, the Project of Yangtze Fisheries Resources and Environment Investigation (2017–2021) supported by the Ministry of Agriculture and Rural Affairs, P. R. China carried out by 24 institutes and universities that located in the Yangtze River basin surveys the status of (1) fish species composition and spatial distribution, (2) current fish abundance, (3) endangered fishes, (4) Yangtze finless porpoise, (5) aquatic eco-environments, (6) water-level fluctuation areas, (7) capture fisheries and recreational fisheries of the Yangtze River mainstream and 10 of its main tributaries, including Yalong River, Heng River, Min River (including Dadu River), Chishui River, Tuo River, Jialing River, Wu River, Han River, Dongting Lake and Poyang Lake. The results showed that there were 443 fish species (belonging to 163 genus, 37 families, and 18 orders) before 2017, but only 323 fish species (including 15 new recorded exotic species) were recorded in the project of 2017–2021. Among them, Cyprinus carpio, Carassius auratus, Hypophthalmichthys molitrix, Pelteobagrus fulvidraco, Coilia brachygnathus, Silurus asotus, Saurogobio dabryi, Ctenopharyngodon idellus, Pelteobagrus nitidus, Hemiculter leucisculus, Siniperca chuatsi, Coreius heterodon, Culter alburnus, Parabramis pekinensis, and Aristichthys nobilis were the dominant species across the whole Yangtze River system. It is estimated that there were 886 million individuals weighing 124.8 million kg, merely equivalent to 27.3% of the resources in 1950s, 30.9% of the resources in 1960s, or 58.7% of the resources in 1980s. In the new list of protected fishes that recorded in the Yangtze River system, only 15 of 29 were collected in this project of 2017–2021. Psephurus gladius has been affirmed to be Extinct by International Union for Conservation of Nature (IUCN). The wild individuals of Tenualosa reevesii and Luciobrama macrocephalus have disappeared for many years and maybe have been extinct already. Acipenser dabryanus has been affirmed to be Extinct in the Wild by IUCN. The natural propagations of A. sinensis, Myxocyprinus asiaticus and Trachidermus fasciatus have been interrupted for many years. The populations of Yangtze finless porpoise in the Yangtze River mainstream, Dongting Lake and Poyang Lake have steadily rising sizes and expanding distributions in 2017–2021. Parts of them migrate from one region to another with the seasons, which would result in the fluctuation of Yangtze finless porpoise population within some regions. The conventional indicators of water quality in the Yangtze River system were good and conformed to the water quality criteria of fishery in 2017–2021. In the last 40 years, the maximum surface water area in the Yangtze River basin extended to approximately 63,360 km2, the minimum surface water area covered approximately 26,396 km2, and the seasonal water-level fluctuation areas occupied approximately 36,964 km2. Compared with 1984–2000 period, the 2001–2020 period witnessed an overall decreasing trend in the frequency of surface water occurrence within about 25,869 km2 of aquatic areas. From 1984 to 2000 period to 2001–2020 period, permanent surface water has decreased by nearly 8,750 km2. In 2017, the fishermen were mainly 40–60 years old and their educational levels were mainly lower than junior high school. In 2017, most anglers were older than 40 and used hand rod and/or sea rod. Their average catch of each time was mainly less than 1 kg. Results suggested that the fishing ban in key waters of the Yangtze River basin is not only an ecological project of aquatic ecosystem conservation, but also a livelihood project for the fisherman to embrace industrial transformation and improve their living conditions. After the implementation of the fishing ban, the fisheries resources would gradually recover, and the Yangtze finless porpoise population size would also see a steady increase. However, the endangered species would remain threatened for a long time. The degraded waterbodies and water-level fluctuation areas would be the key restrictive factors for future aquatic ecosystem recovery in the Yangtze River basin. Since this survey was carried out before the fishing ban, the current results could provide a baseline for future evaluation of the effect of the Yangtze River fishing ban.

In this study, the dynamic response of a semi-buried steel vessel under a near-surface explosion shock load was investigated through full-scale model tests. A three-dimensional precise numerical simulation model of the semi-buried steel vessel structure was established, and the dynamic response and damage consequences under different load conditions were analyzed. The influence of the thickness of corrugated steel on the dynamic damage and antiknock properties of the structure was studied. The results show that the weak part of the rectangular steel vessel structure is mainly located at the connection between the blast-facing panel and the beam column under near-surface explosion conditions. The peak stress and displacement of the blasting surface decrease with the increase in the horizontal angle of the incident wave, whilst/while increase with the height of the explosion. Under the same explosive yield, the main failure mode of the structure is the tearing of the plate and beam under plastic deformation. Increasing the thickness of the corrugated steel can significantly restrain the plastic deformation and acceleration of the plate. When the thickness is increased from 2 mm to 6 mm, the antiknock performance is significantly improved. However, when the thickness is increased from 6 mm to 8 mm, the improvement in antiknock performance slows down. Increasing the height of the enclosing soil can also effectively improve the explosion-proof performance of the structure, with full burial providing the best effect. The research results provide a theoretical basis for the application of rectangular steel vessel structures in the field of protection engineering.

In the past decade, research has demonstrated that viral miRNAs encoded by a number of viral genomes, particularly by most of the herpesvirus including Marek's disease virus (MDV), play important regulatory roles in viral infection, replication, and regulation of tumorigenesis. As macrovesicles in cells, exosomes can deliver viral miRNAs and exert gene regulatory functions. Whether the exosomes play a role in the replication, pathogenesis/tumorigenesis of avian herpesviruses such as oncogenic Marek's disease virus (MDV) remains unclear. Herein we extracted and identified the exosomes from MDV-transformed T cell line MSB-1 and demonstrated high abundance of MDV-1 miRNA expression. Using dual luciferase-based reporter assay, we also demonstrated that the exosomes derived from MSB-1 can deliver functional miRNA successfully into primary chicken embryo fibroblasts. These findings provide new insights into the role of exosomes and the mechanisms of how virus-encoded miRNA function in MDV latency/activation switching, viral replication, pathogenesis and/or tumorigenesis.

Abstract Collagen-based biomaterials (CBB) are highly esteemed by researchers in materials science and biomedicine due to their extensive applications across various biomedical disciplines. In recent years, owing to advancements in developmental biology techniques, this superior biomaterial has seen increasing utilization in 3D in vitro tissue culture. Three-dimensional cell cultures, often referred to as organoids, have emerged in response to technological advancements in biomaterials and the growing need in the field of medical research. They serve as important models for simulating normal physiological activities in vivo, addressing limitations in experimental material sources, and resolving ethical issues. In this review, we discuss the material characteristics of CBBs commonly used for organoid culture, integrating aspects such as Matrigel and decellularized ECM as culture matrices. We also analyzed the development prospects and directions of various materials in the context of biology, clinical medicine, and particularly reproductive medicine. Currently, despite the FDA approval and clinical research incorporating numerous CBBs, existing challenges in multiple studies indicate a significant unmet need in the development of key tissue models for both medical research and clinical applications. In summary, CBBs are swiftly broadening their applicability in the realms of organoid nature and medical research, serving as a versatile and high-performing material for 3D in vitro tissue culture. The Mengshan granitoid complex is located in the central part of Jiangnan Orogen and belongs to the western part of Jiangxi Province, where several phases of granitic magmatism record the crustal evolution of the late-Indosinian. However, its petrogenesis remains uncertain, largely due to controversies over its origin, evolutionary process and tectonic setting during intrusion. In this study, the lithological features and contact relationships observed in the systematic field geological investigations indicate that the late magmatic phases of the Mengshan granitoid complex are mainly composed of medium–fine-grained biotite monzogranite and fine-grained monzogranite, both of which developed primary fine-grained muscovite. Petrological, petrographic, geochemical and zircon U-Pb-Hf isotopic studies were further carried out on these rocks. Zircon U-Pb isotopic analyses suggest that the medium–fine-grained biotite monzogranite mainly formed at 220.7 ± 1.0 Ma to 218.0 ± 0.8 Ma, and that the fine-grained monzogranite formed at 211.5 ± 2.9 Ma to 212.9 ± 1.0 Ma. Whole-rock geochemical analysis results suggest that these rocks are rich in Cs, Rb, and U, and deficient in Ba, Sr, and Ti, and that they have properties characteristic of rocks with high silica, low P content and high K calc-alkali. Mineralogical and geochemical analysis results suggest that they are S-type granites. The εHf(t) values of the early-stage medium–fine-grained biotite monzogranite and late-stage fine-grained monzogranite range from –4.7 to 0.3 and from –3.2 to 0.7, respectively. Geochemical and isotopic data suggest that these granitoids were derived from the partial melting of Proterozoic continent basement rocks, and that minor mantle materials were involved during their generation. The presence of the early Mesozoic Mengshan granitoid complex reflects a reduplicated far-field converge effect of the collision of the North China and South China blocks and the subduction of the Palaeo-Pacific plate into the South China block. The thickening of the Earth’s crust facilitated crustal delamination, underplating of mantle-derived magma, and crustal heating, triggering intense partial melting of the lithosphere and magma enrichment.

The South China Plate is an important part of the Rodinia supercontinent in the Neoproterozoic. The Rizhao area, located on the northeastern margin of the South China Plate, records multiple periods of magmatism, among which Neoproterozoic granitic gneiss is of great significance to the tectonic evolution of the South China Block. In this study, systematic petrology, geochemistry, isotopic chronology, and zircon Hf isotopic analyses were carried out on gneisses samples of biotite alkali feldspar granitic and biotite monzogranitic compositions in the Rizhao area. Geochemical analyses suggest that these granitic rocks belong to the sub-alkaline series and have high potassium contents. They are enriched in large-ion lithophile elements K, Rb, and Ba; depleted in high field strength elements P, Nb, and Ti; enriched in light rare earth elements and moderately depleted in heavy rare earth elements; and have weak to moderate negative Eu anomalies and weak negative Ce anomalies. These rocks are post-orogenic A-type granites. LA-MC-ICP-MS U-Pb dating of zircons from two biotite alkali-feldspar granitic gneiss samples yielded weighted mean ages of 785 ± 8 Ma (MSWD = 3.0) and 784 ± 6Ma (MSWD = 1.5), respectively, and a biotite monzogranitic gneiss sample yielded a weighted mean age of 789 ± 6 Ma (MSWD = 2.3). Lu-Hf isotopic analyses on zircon grains from the two types of Neoproterozoic-aged gneisses yielded negative εHf(t) values ranging from −19.3 to −8.8 and from −18.3 to −10.4, respectively, and the corresponding two-stage Hf model age ranges are 2848–3776 Ma and 2983–3682 Ma, respectively. These granites are the product of Neoproterozoic magmatic activity and are mainly derived from the partial melting of Archean continental crust. Combining the geochemical characteristics and zircon U-Pb-Lu-Hf isotopic analyses, these A-type granitic gneisses appear to have formed in an intracontinental rift extension environment during the initial break-up of the Rodinia supercontinent, as part of the supercontinent break-up process at the northeastern margin of the South China Block.

Abstract Introduction Mindfulness-based stress reduction (MBSR) has demonstrated its effectiveness in reducing pain-related stress in adults with chronic pain. However, the implementation of MBSR needs modifications across cultures. This pilot study reports the findings of a randomized controlled trial that investigated the effects of a culturally adaptive MBSR program on self-report and neuroimaging outcomes for chronic pain adults in China. Methods Sixty-seven participants were randomly assigned to the treatment group (n = 40) or the treatment-as-usual group (n = 27) group at a ratio of 1.5:1. Participants completed self-report measures of pain severity, pain interference, depression, perceived stress, pain catastrophizing, mindfulness, and resilience at baseline assessment (T1), post-treatment (T2), and 3-month follow-up (T3) assessments. Functional magnetic resonance imaging (fMRI) scanning was also performed at T1 and T3 assessments. Results For the intention-to-treat sample, the results of the mixed-effect model indicated that Group × Time interaction was significant for pain catastrophizing only (F (2, 130) = 3.51, p = 0.033). Compared with the control group, those in the MBSR group reported greater reductions in pain catastrophizing at T2 (d = − 0.60), though this effect was not maintained at T3 (d = − 0.05). Additionally, the results of completer analyses found significant Group × Time interactions for pain interference (F (2, 88) = 4.40, p = 0.015) and perceived stress (F (2, 88) = 3.13, p = 0.048), but not for other measures. Finally, both groups exhibited decreased regional homogeneity (ReHo) in the frontal lobe, while increased ReHo in the cerebellum anterior lobe was unique to the MBSR group. Conclusions The present findings suggest that the minor modified MBSR program improves certain pain-related outcomes for Chinese adults with chronic pain. Future studies with larger samples of Chinese chronic pain patients are needed to detect the small-to-moderate benefit of MBSR on fMRI and/or other objective methods.

In this work, a series of thermosensitive ionic liquid functionalized polymers, PNx(IL)y, with controllable morphology and particle size were prepared by free radical polymerization. Then, using the polymer PN64(IL)8 with uniform morphology as a templating agent, the ZnO composite photocatalytic materials doped with rare earth metal Ce were prepared in combination with a microwave-assisted and templated hydrothermal reaction method. Series different Ce-doping amount photocatalytic materials ZnO-Ce-x‰ were characterized by XRD, SEM, TEM, XPS, and other methods. The results demonstrated that the templated materials PN64(IL)8 can prepare ZnO-Ce-2‰ with uniform petaloid ambulacra shape, good distribution of elements, and excellent photocatalytic performance. Photocatalytic degradation experiments of methyl orange (MO) showed that when the Ce-doping amount is only 2‰, the degradation rate of organic dyes can reach 96.5% by reacting the photocatalytic materials in water for 1 h. In addition, this kind of photocatalyst can be used for the degradation of high-concentration MO, as well as being easily recovered and effectively reused by simple filtration. Therefore, the structure of this kind of photocatalyst is controllable in the preparation process with an extremely low Ce-doping amount compared with current reports, and it has a good application prospect in the field of wastewater treatment technology.

Avian leukosis viruses (ALVs) include a group of avian retroviruses primarily associated with neoplastic diseases in poultry, commonly referred to as avian leukosis. Belonging to different subgroups based on their envelope properties, ALV subgroups A, B, and J (ALV-A, ALV-B, and ALV-J) are the most widespread in poultry populations. Early identification and removal of virus-shedding birds from infected flocks are essential for the ALVs’ eradication. Therefore, the development of rapid, accurate, simple-to-use, and cost effective on-site diagnostic methods for the detection of ALV subgroups is very important. Cas13a, an RNA-guided RNA endonuclease that cleaves target single-stranded RNA, also exhibits non-specific endonuclease activity on any bystander RNA in close proximity. The distinct trans-cleavage activity of Cas13 has been exploited in the molecular diagnosis of multiple pathogens including several viruses. Here, we describe the development and application of a highly sensitive Cas13a-based molecular test for the specific detection of proviral DNA of ALV-A, B, and J subgroups. Prokaryotically expressed LwaCas13a, purified through ion exchange and size-exclusion chromatography, was combined with recombinase polymerase amplification (RPA) and T7 transcription to establish the SHERLOCK (specific high-sensitivity enzymatic reporter unlocking) molecular detection system for the detection of proviral DNA of ALV-A/B/J subgroups. This novel method that needs less sample input with a short turnaround time is based on isothermal detection at 37 °C with a color-based lateral flow readout. The detection limit of the assay for ALV-A/B/J subgroups was 50 copies with no cross reactivity with ALV-C/D/E subgroups and other avian oncogenic viruses such as reticuloendotheliosis virus (REV) and Marek’s disease virus (MDV). The development and evaluation of a highly sensitive and specific visual method of detection of ALV-A/B/J nucleic acids using CRISPR-Cas13a described here will help in ALV detection in eradication programs.

The fat deposition of different adipose tissues is widely recognized as correlated, with distinct effects on meat quality traits and reproductive performance in poultry. In this study, we utilized lipidomics and transcriptomics analyses to investigate the heterogeneity and regulators of intramuscular fat (IMF), abdominal fat (AF), and subcutaneous fat (SF) in geese. Lipidomic profiling revealed 165, 129, and 77 differential lipid molecules (DLMs) between AF vs. IMF, SF vs. IMF, and SF vs. AF, respectively, with 47 common DLMs identified between AF vs. IMF and SF vs. IMF. Transcriptomic analysis identified 3369, 5758, and 131 differentially expressed genes (DEGs) between AF vs. IMF, SF vs. IMF, and SF vs. AF, respectively, with 2510 common DEGs identified between AF vs. IMF and SF vs. IMF. The KEGG results indicate that DLMs were predominantly enriched in glycerophospholipid and glycerolipid metabolism pathways, while DEGs were primarily enriched in metabolic pathways. Pearson correlation analysis identified FABP4, LPL, PLCB1, DSE, and PDE5A as potential factors influencing fat deposition. This study elucidates the heterogeneity and regulatory factors of different adipose tissues in geese, offering new insights for targeted improvements in goose meat quality and production efficiency.

ObjectiveIncreasing evidence indicated that schizophrenia and obesity are associated with altered mitochondrial and immune function. In this study, we investigated the levels of CRP (C-reactive protein) and mitochondrial lymphocytes in chronically treated schizophrenia patients with atypical antipsychotic medications and further explored the relationship between mitochondrial lymphocyte and weight gain as well as cognitive function in these patients.MethodsWe evaluated the mitochondrial lymphocyte count of 97 patients (53 overweight, 44 non-overweight) and 100 healthy controls using mitochondrial fluorescence staining and flow cytometry (NovoCyte, Agilent Technologies, US). The serum CRP was measured by high-sensitivity enzyme-linked immunosorbent assay (ELISA). Clinical symptoms and cognitive function of the patients were assessed using the Positive and Negative Syndrome Scale (PANSS) and the Repeatable Battery for the Assessment of Neuropsychological Status (RBANS).ResultsThe results showed that mitochondrial lymphocyte counts of CD3+ T, CD3+CD4+ T, and CD3+CD8+ T cells in schizophrenia patients were higher than in the control group (p < 0.05). Additionally, overweight patients had significantly higher mitochondrial lymphocyte counts of CD3+ T and CD3+CD4+ T cells compared to schizophrenia patients with normal weight. Stratified analysis by gender revealed that there was a statistically significant difference in CD3+CD4+ mitochondrial lymphocyte count in male patients (p = 0.014) and a marginal trend toward significance in female patients (p = 0.058). Furthermore, the mitochondrial lymphocyte counts of CD3+ T and CD3+CD4+ T cells, as well as CRP levels, were positively correlated with BMI in schizophrenia patients, but the mitochondrial lymphocyte counts of CD3+CD4+ T cells were negatively correlated with the language scale in the RBANS.ConclusionOur study results provide evidence for the association between altered mitochondrial T lymphocyte and weight gain as well as cognitive impairment in schizophrenia patients treated with atypical antipsychotic medications.

Female fertility decline is an accumulative consequence caused by complex factors, among them, the disruption of the immune profile in female reproduction stands out as a crucial contributor. Presently, the effects of immune microenvironment (IME) on the female reproductive process have attracted increasing attentions for their dynamic but precisive roles. Immunocytes including macrophages, dendritic cells, T cells, B cells and neutrophils, with diverse subpopulations as well as high plasticity functioned dynamically in the process of female reproduction through indirect intercellular communication via specific cytokine release transduced by molecular signal networks or direct cell-cell contact to maintain the stability of the reproductive process have been unveiled. The immune profile of female reproduction in each stage has also been meticulously unveiled. Especially, the application of single-cell sequencing (scRNA-seq) technology in this process reveals the distribution map of immune cells, which gives a novel insight for the homeostasis of IME and provides a research direction for better exploring the role of immune cells in female reproduction. Here, we provide an all-encompassing overview of the latest advancements in immune modulation within the context of the female reproductive process. Our approach involves structuring our summary in accordance with the physiological sequence encompassing gonadogenesis, folliculogenesis within the ovaries, ovulation through the fallopian tubes, and the subsequent stages of embryo implantation and development within the uterus. Our overarching objective is to construct a comprehensive portrayal of the immune microenvironment (IME), thereby accentuating the pivotal role played by immune cells in governing the intricate female reproductive journey. Additionally, we emphasize the pressing need for heightened attention directed towards strategies that focus on immune interventions within the female reproductive process, with the ultimate aim of enhancing female fertility.

As the global digital transformation process accelerates, how SMEs can successfully achieve digital transformation is currently an important research question in the field of strategic management and a real-life dilemma that entrepreneurs need to address. This study classifies business model innovation into market-driven business model innovation and driving-market business model innovation based on market orientation theory. The relationship between digital ambidextrous capabilities, business model innovation and transformation performance is explored according to the paradigm of ‘capability-behaviour-performance’. This study used SEM and fsQCA to analyze 289 questionnaires collected from middle and senior managers of Chinese SMEs. The results show that digital exploitation capability is positively associated with market-driven business model innovation, while digital exploration capability is positively associated with driving-market business model innovation. In particular, business model innovation plays a fully mediating role in the process of digital ambidextrous capabilities enhancing the transformation performance. Our findings shed new lights on the current debate surrounding the digital transformation of SMEs and will be instructive for both academics and business managers.

The excessive exploitation of groundwater not only destroys the dynamic balance between coastal aquifer and seawater but also causes a series of geological and environmental problems. Groundwater level prediction provides an efficient way to solve these intractable ecological problems. Although several hydrological numerical models have been employed to conduct prediction, no study has accurately predicted the groundwater level change under the consideration of groundwater exploitation, especially in coastal aquifers. This is due to the characteristics of spatially and temporally complex hydrological processes. This study proposes a novel data-driven method based on the combination of time series analysis and a machine learning method for accurately predicting the variation of groundwater level in a coastal aquifer under the influence of groundwater exploitation. The partial autocorrelation function and continuous wavelet coherence were used to analyze the monitoring data of groundwater level at three wells, which indicated that the historical monitored data and the dataset of precipitation could be considered as the input variables to construct the hydrological model. Then, three models based on the different inputs were constructed, namely, the LSTM, PACF-LSTM, and PACF-WC-LSTM models. The performances of the three models were compared by the calculation of four error metrics. The results showed that the performance of the PACF-LSTM and PACF-WC-LSTM models was better than that of the LSTM model and that the PACF-WC-LSTM model achieved the best prediction performance. Accurately predicting the variation of groundwater level provides the basis for managing groundwater resources and preserving the ecological environment.

To explore the population structures and dynamics of Rhododendron shrub communities at different stages of succession in northwest Guizhou, China, this study examined the populations of Rhododendron annae and Rhododendron irroratum shrub with two different stages. A space-for-time substitution was employed to establish the diameter class/height structures, static life tables, and survival/mortality rate/disappearance rate curves of both Rhododendron populations with different orders of succession. Their structural and quantitative dynamics were analyzed, and their development trends were predicted. The results showed that, quantitatively, the populations of R. annae and R. irroratum in the two Rhododendron communities with different orders of succession were dominated by age classes one, two, and three as well as height classes i, ii, and iii. The number of Rhododendron plants at the three age classes and the three height classes accounted for 97.61–100% of the total. The quantitative dynamic indices of R. annae and R. irroratum were both greater than 0, with and without considering external interference. In terms of age class and height structures, both Rhododendron populations were expanding populations, presenting “inverted-J-shaped” and irregular pyramid patterns. There was a sufficient number of young individuals, but few or no old individuals. Both survival curves of the populations of R. annae and R. irroratum in the two Rhododendron communities with different orders of succession belonged to the Deevy-II type. In the late stage of succession, the mortality curves and disappearance curves of both Rhododendron populations in these communities presented a trend of increasing first and then decreasing with increasing age class. This result indicates that at each age class, R. annae and R. irroratum showed a trend of gradual increase after two, four, and six years. In brief, the populations of R. annae and R. irroratum have rich reserves of seedlings and saplings, but high mortality and disappearance rates. In this context, it is necessary to reduce human interference and implement targeted conservation measures to promote the natural renewal of Rhododendron populations.

Objectives: Cognitive impairment in schizophrenia patients with auditory hallucinations is more prominent compared to those without. Our study aimed to investigate the cognitive improvement effects of 10 Hz repetitive transcranial magnetic stimulation (rTMS) over the left dorsolateral prefrontal cortex (DLPFC) in schizophrenia with auditory hallucinations. Methods: A total of 60 schizophrenic patients with auditory hallucinations in this study were randomly assigned to sham or active group. Both groups received 10 Hz or sham rTMS targeted in left DLPFC for 20 sessions. The Positive and Negative Syndrome Scale (PANSS), the Auditory Hallucination Rating Scale (AHRS), the Repeatable Battery for the Assessment of Neuropsychological Status (RBANS), and the Udvalg for Kliniske Under-sogelser (UKU) side effect scale were used to measure psychiatric symptoms, auditory hallucinations, cognition, and side reaction, respectively. Results: Our results indicated that the active group experienced greater improvements in RBANS-total score (P = 0.043) and immediate memory subscale score (P = 0.001). Tree base sprouting is the main reproduction and expansion mode of Rhododendron plants. By leveraging the plot survey method, the species composition, community renewal, and species diversity in three Rhododendron shrub communities in control, and before and after clearcutting (CK, D3yr, and D6yr, respectively) were studied. Moreover, the dissimilarity of Rhododendron communities in CK, D3yr and D6yr were analyzed. The results showed that there were 26 plant species belonging to 14 families and 22 genera, in 3 communities in total, with 19 species of shrub plants and 7 species of herbaceous plants. The number of species increased from 13 in CK to 23 in D3yr and then decreased to 20 in D6yr. The height and coverage of D3yr and D6yr reached 39.3% and 58.9% of that of CK, respectively. The relative height of CK, D3yr, and D6yr was 43.79%, 65.4%, and 58.54%, respectively. The coverage of D3yr and D6yr reached 60.8% and 114.70% of that of CK, respectively. The relative coverage of CK, D3yr, and D6yr was 19.05%, 83.24%, and 77.32%, respectively. The important value of Rhododendron plants in the communities increased from 0.42 in CK to 0.74 in D3yr, and then decreased to 0.67 in D6yr. The α diversity in the shrub layer of D3yr and D6yr were generally lower than those of CK except Pielou evenness index. The β diversity indicates that the similarity between CK and D3yr was lower, that between CK and D6yr was moderate, and that between D3yr and D6yr was higher. The sprouting height and coverage of Rhododendron plants was significantly correlated with age and sprouting time. The sprouting ability of Rhododendron plants increased first and then decreased with age, while the sprouting ability of Rhododendron plants with age of 10–12 years was the strongest. Clearcutting measures can improve the dominance of Rhododendron plants in the communities, promote the sprouting and renewal of Rhododendron population, and accelerate the succession rate of communities.

Optimal fertilization is an important measure for managing cultivated grasslands, and a necessary means for maintaining the nutrient balance, yield, and quality of grassland ecosystems. This study aimed to explore the effects of organic fertilizers on the production performance and nutritional quality of cultivated grasslands in karst areas. Two types of monocultured cultivated grasslands (i.e., Medicago sativa and Dactylis glomerata) were employed as the research objects, and a randomized block design was adopted to investigate the effects of five fertilization gradients on the forage height, coverage, yield, and nutritional quality of the cultivated grasslands. According to the results, the plant height, coverage, and yield of M. sativa first presented an increasing trend, then decreased with increasing fertilization gradient, with a peak at 20–30 t/hm2 fertilization gradient. The height, coverage, and yield of D. glomerata increased gradually with increase in fertilization gradient, and peaked at 40 t/hm2. Meanwhile, the crude protein (CP) and ether extract (EE) contents of both grassland types displayed first presented an increasing trend, then decreased with increasing fertilization gradient, and peaked at 10–30 t/hm2 fertilization gradient. The neutral detergent fiber (NDF) and acidic detergent fiber (ADF) contents of M. sativa presented “N-shaped” and “M-shaped” change trends with increasing fertilization gradient, while those of D. glomerata showed “V-shaped” and “M-shaped” change trends, reaching minimum values under fertilization gradients of 30 and 20 t/hm2, respectively. Year, fertilization, and year × fertilization (Y×F) significantly affected the plant heights, coverages, dry/fresh weight ratios, and yields of M. sativa and D. glomerata. The contribution of coverage to the subordinate function of M. sativa was greatest at a fertilization gradient of 20 t/hm2. Meanwhile, the subordinate function values of the height and coverage of D. glomerata increased gradually with increasing fertilization gradient, but the difference in the subordinate function value of height was only 0.09%–0.18% under the fertilization gradient of 20–40 t/hm2. Evaluation of forage nutrition revealed 10–30 t/hm2 and 20–30 t/hm2 as the optimal organic fertilizer application rates for M. sativa and D. glomerata, respectively.

The escalating issues of worldwide energy scarcity and environmental contamination have brought geothermal resources into the spotlight as a sustainable and eco-friendly energy alternative. The circum-Wugongshan geothermal belt has abundant geothermal resources at a medium-low temperature, offering significant potential for development and utilization. In this study, samples of geothermal groundwater, cold spring water, and surface water were collected from the western Wugongshan area. Hydrochemical and thermodynamic methods were used to estimate the reservoir temperature and analyze its mechanism of origin. The results of these analyses indicated that, in terms of hydrochemical characteristics, most geothermal groundwater samples were Na-HCO3 and Na-SO4, while cold spring and surface water samples were Na-HCO3 and Ca-HCO3, respectively. Quartz and multicomponent geothermometers provided the most reliable estimations of reservoir temperatures, ranging from 64.8°C to 93.4°C. The circulation depth of geothermal water was found to range from 1729.3 m to 2,292.5 m. A mixing model indicated that shallow cold water was blended at a rate of 62.1%–78.8%. The δD and δ18O values validated the conclusion that the geothermal water originates from atmospheric precipitation, with recharge elevations varying between 503.1 m and 1,375.6 m. Based on the above analysis, a conceptual model is proposed to illustrate the mechanism of geothermal groundwater genesis.

Abstract Background Joint Awareness is thought to be closely linked to Quality of Life (QoL) for patients undergoing Total Knee Arthroplasty (TKA), yet to date there have been no longitudinal studies to explore how Joint Awareness actually affects QoL. The purpose of this study was therefore to examine the development of Joint Awareness and QoL after TKA as well as the dynamic impact of Joint Awareness on QoL. Methods A total of 342 patients were followed up at 3 months (T1), 6 months (T2), and 12 months (T3) after TKA. Joint Awareness was evaluated using the Forgotten Joint Score-12 (FJS-12), and QoL was measured by SF-36. Abstract Background Linalool is a monoterpenoid, also a vital silvichemical with commercial applications in cosmetics, flavoring ingredients, and medicines. Regulation of mevalonate (MVA) pathway metabolic flux is a common strategy to engineer Saccharomyces cerevisiae for efficient linalool production. However, metabolic regulation of the MVA pathway is complex and involves competition for central carbon metabolism, resulting in limited contents of target metabolites. Results In this study, first, a truncated linalool synthase (t26AaLS1) from Actinidia arguta was selected for the production of linalool in S. cerevisiae. To simplify the complexity of the metabolic regulation of the MVA pathway and increase the flux of isopentenyl pyrophosphate (IPP) and dimethylallyl pyrophosphate (DMAPP), we introduced the two-step isopentenyl utilization pathway (IUP) into S. cerevisiae, which could produce large amounts of IPP/DMAPP. Further, the S. cerevisiae IDI1 (ecoding isopentenyl diphosphate delta-isomerase) and ERG20 F96W−N127W (encoding farnesyl diphosphate synthase) genes were integrated into the yeast genome, combined with the strategies of copy number variation of the t26AaLS1 and ERG20 F96W−N127W genes to increase the metabolic flux of the downstream IPP, as well as optimization of isoprenol and prenol concentrations, resulting in a 4.8-fold increase in the linalool titer. Eventually, under the optimization of carbon sources and Mg2+ addition, a maximum linalool titer of 142.88 mg/L was obtained in the two-phase extractive shake flask fermentation. Conclusions The results show that the efficient synthesis of linalool in S. cerevisiae could be achieved through a two-step pathway, gene expression adjustment, and optimization of culture conditions. The study may provide a valuable reference for the other monoterpenoid production in S. cerevisiae.

Abstract Endometriosis is a common, estrogen‐dependent chronic gynecological disease that endangers the reproductive system and systemic metabolism of patients. We aimed to investigate the differences in metabolic profiles in the follicular fluid between infertile patients with endometriosis and controls. A total of 25 infertile patients with endometriosis and 25 infertile controls who were similar in age, BMI, fertilization method and ovulation induction treatment were recruited in this study. Metabolomics analysis of follicular fluid was performed by two methods of high‐performance liquid chromatography tandem mass spectrometry. There were 36 upregulated and 17 downregulated metabolites in the follicular fluid of patients in the endometriosis group. KEGG pathway analysis revealed that these metabolites were enriched in phenylalanine, tyrosine and tryptophan biosynthesis, aminoacyl‐tRNA biosynthesis, phenylalanine metabolism and pyrimidine metabolism pathways. A biomarker panel consisting of 20 metabolites was constructed by random forest, with an accuracy of 0.946 and an AUC of 0.988. This study characterizes differences in follicular fluid metabolites and associated pathway profiles in infertile patients with endometriosis. These findings can provide a better comprehensive understanding of the disease and a new direction for the study of oocyte quality, as well as potential metabolic markers for the prognosis of endometriosis.

Forage quality is a key property of grassland ecosystems. In this study, grassland forage qualities were measured at 373 sampling sites throughout Guizhou Province in the karst mountain region of Southwest China, and the factors affecting it were explored. The forage quality level of most plant species was categorized into four levels: (1) preferred forage species; (2) desirable forage species; (3) consumed but undesirable forage species; and (4) non-consumable or toxic forage species. High temperature and precipitation appeared to facilitate the growth of preferred forage species, but limited the growth of other plants. Increasing soil pH had a positive impact on the number and biomass of preferred forage plants, but a negative influence on other plants, especially non-consumable or toxic plants. Both GDP and population density had a positive correlation with the number and biomass of preferred forage species, while such correlations for other levels of forage species tended to be negative. Grazing could lead to a decrease in the preferred forage species. Therefore, it is suggested that by focusing on soil improvement in grassland and maintaining an appropriate grazing intensity, global warming and rapid economic growth in Guizhou Province will likely contribute to increase the forage quality of karst grasslands in Southwest China.

Several studies have investigated the causal effect of different plasma lipid and apolipoprotein parameters on the occurrence and progression of endometriosis. However, the results of these studies remained controversial. Herein, this study aims to analyze the causal associations of genetically predicted levels of different blood lipid and apolipoprotein traits with the risk of endometriosis using a two-sample Mendelian randomization (MR) approach. Methods: The single-nucleotide polymorphisms (SNPs) of exposures which indicate the genetic variants were used as instrumental variables (IVs). We assessed the correlation between each blood lipid and apolipoprotein parameter and the occurrence of endometriosis from various cohorts of European ancestry, and the SNPs of them were obtained from the genome-wide association study (GWAS) from the IEU GWAS database. The causal effects were estimated through the inverse-variance weighted method (IVW). The sensitivity examinations of each result after the MR analysis were performed to detect if any heterogeneity and pleiotropy existed. Besides, sub-analyses were performed to provide more detailed information about the actual associations. Results: Our results demonstrated that cholesterol in chylomicron (CM) and extremely large very low-density lipoprotein (VLDL) was causally linked to higher odds of endometriosis, while high-density lipoprotein (HDL) cholesterol showed the opposite correlation. No evidence was detected on the causal relations of low-density lipoprotein (LDL) cholesterol, VLDL cholesterol, triglyceride (TG), apolipoprotein A1 and apolipoprotein B with endometriosis. Neither pleiotropy nor heterogeneity was found in our study. In addition, subgroup analyses about subtypes of endometriosis suggested the causal association of several serum lipids with peritoneal endometriosis. Conclusion: Our results suggested that dysregulated lipid metabolism is causally associated with the occurrence of endometriosis, especially peritoneal endometriosis. Our findings provided genetic insights on potential novel strategies in the primary prevention and treatment of endometriosis.

No evidence of bidirectional association was detected on the other serum inflammatory biomarkers with endometriosis. Neither pleiotropy nor heterogeneity was found in our study. Conclusion: Our results suggested that some dysregulated circulating biomarkers of immunity and inflammation is causally associated with the occurrence of endometriosis, and at the same time endometriosis may lead to the abnormality of some of these factors. Our findings provided genetic insights on potential novel strategies in the primary prevention, diagnosis and treatment of endometriosis.

The properties of the Caledonian orogeny along the transition belt of the Yangtze and Cathaysia blocks have received much attention in recent years. The widespread Early Paleozoic granites provide critical geological clues for unraveling the tectonic evolution and geodynamic processes of the South China Continent (SCC). Here we present new zircon U–Pb chronology, whole-rock major and trace elements, in situ Hf isotopes for Paleozoic granites, i.e., the Wugongshan granites in the northwest Jiangxi province, and aim to explore the magmatism and properties of the Caledonian orogeny involved in their formation. Our new data show that the Wugongshan granites were emplaced during the Early Silurian Period (442–438 Ma). The Paleozoic Wugongshan granites belong to S-type muscovite-bearing peraluminous granites (MPG) and show a single origin. The Wugongshan granites exhibit negative εHf(t) values (−11.56 to −6.19) and TDM2 model ages of 2148–1809 Ma, indicating their derivation from an ancient crustal source, through partial melting of ancient crustal material. The Wugongshan granitic magmatism is probably being generated in an extensional environment related to an intracontinental orogeny setting. It is inferred that the Paleozoic tectonic–magmatic event in the Wugongshan area was associated with the oceanic–continental convergence of the Paleo-Tethys Ocean. The Wugongshan granites highlight the intracontinental magmatism in the Early Paleozoic orogeny in the SCC.

The Wugongshan area is rich in medium–low temperature convective geothermal resources, among which there are more than 10 geothermal fields in Wentang, Wanlongshan, Wenjia, Hongjiang, etc. There are few basic geothermal geological studies in the geothermal fields and their peripheral areas; thus far, no systematic research work into the thermophysical parameters has been carried out. In this paper, 85 rock samples were collected from the surface and boreholes covering the strata and magmatic rocks in the study area. The results show that the average radioheat generation rate, the average thermal conductivity, and the average specific heat are 0.24–5.49 (μW/m3), 1.995–4.390 (W/mK), and 1.318–4.829 (MJ/m3K), respectively. The average thermal diffusivity ranges from 1.115 to 1.611 × 10-6 m2/s. The highest radioheat generation rate is Jurassic granite, and the lowest is quartz vein. The largest thermal conductivity and specific heat is the siliceous quartzite, and the smallest is the quartz vein. The highest thermal diffusivity is Cambrian metamorphic mica schist, and the lowest is siliceous quartzite. The radioactive heat generation rate, thermal conductivity, specific heat, and thermal diffusivity are closely related to the chemical composition, mineral composition, rock fabric, porosity, water content, and temperature and pressure conditions of rocks in the whole area. There is a linear relationship between thermal conductivity (K) and thermal diffusivity (κ), and the correlation equation is K = −0.3144k + 3.2172. Combined with the characteristics of thermophysical parameters, the genetic theory of deep crust heat generation + structural heat accumulation + siliceous quartzite heat conduction + granite heat preservation is preliminarily proposed.

Abstract Extracellular matrix (ECM) is characterized as widespread, abundant, and pluripotent. Among ECM members, collagen is widely accepted as one of the most prominent components for its essential structural property that can provide a scaffold for other components of ECM and the rich biological functions, which has been extensively used in tissue engineering. Emerging evidence has shown that the balance of ECM degradation and remodeling is vital to regulations of maternal–fetal interface including menstrual cycling, decidualization, embryo implantation and pregnancy maintenance. Moreover, disorders in these events may eventually lead to failure of pregnancy. Although the improvement of assisted conception and embryo culture technologies bring hope to many infertile couples, some unfavorable outcomes, such as recurrent implantation failure (RIF), recurrent pregnancy loss (RPL) or recurrent miscarriage (RM), keep troubling the clinicians and patients. Recently, in vitro three-dimensional (3D) model mimicking the microenvironment of the maternal–fetal interface is developed to investigate the physiological and pathological conditions of conception and pregnancy. The progress of this technology is based on clarifying the role of ECM in the endometrium and the interaction between endometrium and conceptus. Focusing on collagen, the present review summarized the degradation and regulation of ECM and its role in normal menstruation, endometrium receptivity and unsatisfying events occurring in infertility treatments, as well as the application in therapeutic approaches to improve pregnancy outcomes. More investigations about ECM focusing on the maternal–fetal interface interaction with mesenchymal stem cells or local immunoregulation may inspire new thoughts and advancements in the clinical application of infertility treatments. Fowl cholera is caused by the bacterium Pasteurella multocida, a highly transmissible avian ailment with significant global implications, leading to substantial economic repercussions. The control of fowl cholera outbreaks primarily relies on vaccination using traditional vaccines that are still in use today despite their many limitations. In this research, we describe the development of a genetically engineered herpesvirus of turkeys (HVT) that carries the OmpH gene from P. multocida integrated into UL 45/46 intergenic region using CRISPR/Cas9-NHEJ and Cre-Lox system editing. The integration and expression of the foreign cassettes were confirmed using polymerase chain reaction (PCR), indirect immunofluorescence assays, and Western blot assays. The novel recombinant virus (rHVT-OmpH) demonstrated stable integration of the OmpH gene even after 15 consecutive in vitro passages, along with similar in vitro growth kinetics as the parent HVT virus. The protective efficacy of the rHVT-OmpH vaccine was evaluated in vaccinated ducks by examining the levels of P. multocida OmpH-specific antibodies in serum samples using ELISA. Groups of ducks that received the rHVT-OmpH vaccine or the rOmpH protein with Montanide™ (SEPPIC, Paris, France) adjuvant exhibited high levels of antibodies, in contrast to the negative control groups that received the parental HVT or PBS. The recombinant rHVT-OmpH vaccine also provided complete protection against exposure to virulent P. multocida X-73 seven days post-vaccination. This outcome not only demonstrates that the HVT vector possesses many characteristics of an ideal recombinant viral vaccine vector for protecting non-chicken hosts, such as ducks, but also represents significant research progress in identifying a modern, effective vaccine candidate for combatting ancient infectious diseases.

Abstract Collagen, the main component of mammal skin, has been traditionally used in leather manufacturing for thousands of years due to its diverse physicochemical properties. Collagen is the most abundant protein in mammals and the main component of the extracellular matrix (ECM). The properties of collagen also make it an ideal building block for the engineering of materials for a range of biomedical applications. Reproductive medicine, especially human fertility preservation strategies and reproductive organ regeneration, has attracted significant attention in recent years as it is key in resolving the growing social concern over aging populations worldwide. Collagen-based biomaterials such as collagen hydrogels, decellularized ECM (dECM), and bioengineering techniques including collagen-based 3D bioprinting have facilitated the engineering of reproductive tissues. This review summarizes the recent progress in applying collagen-based biomaterials in reproductive. Furthermore, we discuss the prospects of collagen-based materials for engineering artificial reproductive tissues, hormone replacement therapy, and reproductive organ reconstruction, aiming to inspire new thoughts and advancements in engineered reproductive tissues research. Studies have shown that fetal immune cell activation may result from potential exposure to microbes, although the presence of microbes in fetus has been a controversial topic. Here, we combined metagenomic and virome techniques to investigate the presence of bacteria and viruses in fetal tissues (small intestine, cecum, and rectum). We found that the fetal gut is not a sterile environment and has a low abundance but metabolically rich microbiome. Specifically, Proteobacteria and Actinobacteria were the dominant bacteria phyla of fetal gut. In total, 700 species viruses were detected, and Human betaherpesvirus 5 was the most abundant eukaryotic viruses. Especially, we first identified Methanobrevibacter smithii in fetal gut. Through the comparison with adults’ gut microbiota we found that Firmicutes and Bacteroidetes gradually became the main force of gut microbiota during the process of growth and development. Interestingly, 6 antibiotic resistance genes were shared by the fetus and adults. Our results indicate the presence of microbes in the fetal gut and demonstrate the diversity of bacteria, archaea and viruses, which provide support for the studies related to early fetal immunity. This study further explores the specific composition of viruses in the fetal gut and the similarities between fetal and adults’ gut microbiota, which is valuable for understanding human fetal immunity development during gestation.

Since Chinese government work report in 2010 put forward the content of strengthening the construction of smart grids, the construction of smart grids has become one of the important contents of the national development strategy. In the in-depth study of smart grids, the issue of security defense systems and disaster prevention frameworks has always been Research hotspots by scholars at home and abroad, but most of my country’s existing security defenses are aimed at large-scale interconnected systems such as transmission grids, and there is a lack of research on disaster prevention of distribution networks. Therefore, in this research context, this article starts from the perspective of multi-source information fusion, takes into account the uncertainty of distribution network information and other factors, constructs a distribution network disaster response resilience index system and a distribution network resilience assessment model, and uses accidents at the same time. The scenario optimization method improves the disaster warning capability of the distribution network, and provides a scientific basis for the distribution network to take active defense measures. Then, a coordinated defense evolution strategy for the distribution network considering the optimization of accident scenarios was established. This strategy takes limited resources as the constraint, takes the minimum load loss as the objective function, and considers the economics of strategy conversion. Power supply equipment output, load transfer, and switching linkage mechanism between the three load is established, forming islands of security, and network transfer, emergency coordination of the three islands of the distribution network evolution strategy of active defense. This strategy looks for a directional evolution path from real-time accident scenarios to defense strategies from both horizontal and vertical perspectives, and realizes the coordinated control of the distribution network’s active defense strategy locally and globally. Finally, this paper takes the ice disaster climate as an example for simulation verification to prove the comprehensiveness of the risk assessment model and the rationality of the security defense method, and prove that the defense strategy can meet the online adaptive evolution requirements of the active defense of the distribution network, while taking into account The local autonomy and global optimal coordinated control of the active defense of the distribution network make up for the shortcomings of the current active defense strategy and are of great significance to further improve the security defense level of the active distribution network.

BackgroundEpidemiological observational studies have investigated the relationship between rheumatoid arthritis(RA) and pre-eclampsia, but no consistent conclusions were obtained due to various limitations. Hence, we conducted a two-sample mendelian randomization analysis to evaluate the potential causal effect of RA on pre-eclampsia.MethodsSummary-level statistics for RA were derived from a large-scale meta-analysis of datasets of genome-wide association studies(GWAS) which involved 14,361 cases and 43,923 controls. Moreover, summary statistics for pre-eclampsia or eclampsia were sourced from the Finn biobank which contained 3,903 cases and 114,735 controls. Abnormal spermatozoa can not only reduce the fertilization rate, but also prolong the natural conception time and even increase the risk of spontaneous miscarriage. Diabetes mellitus (DM) has become a major global health problem, and its incidence continues to rise, while affecting an increasing number of men in their reproductive years. Type 2 Diabetes Mellitus (T2DM), accounting for about 85-95% of DM, is closely related to the development of sperm. However, the exact association between T2DM and abnormal spermatozoa remains unclear. Herein, we designed a Two-sample Mendelian randomization (MR) study to explore the causal association between T2DM and abnormal spermatozoa risk in European population data which come from the GWAS summary datasets. We selected 9 single nucleotide polymorphisms (SNPs) of T2DM (exposure data) as instrumental variables (IVs), and then retrieved the suitable abnormal spermatozoa genome-wide association study (GWAS) data of European from Ieu Open GWAS Project database which includes 915 cases and 209,006 control as the outcome data. Our results indicate that strict T2DM might not result in a higher risk of abnormal spermatozoa genetically in Europeans (OR: 1.017, 95% confidence interval (CI): 0.771-1.342, p=0.902). Our findings demonstrate that only T2DM may not explain the relatively higher risk of abnormal spermatozoa in men with it in Europeans. In subsequent studies, more comprehensive and larger samples need to be studied to reveal the relationship and potential mechanism between T2DM and abnormal spermatozoa.

Uterine fibroids (UFs), the most common benign gynecological tumor, can bring severe negative impacts on a woman’s life quality. Vitamin D, is thought to play an important role in regulating cell proliferation and differentiation. In recent years, several studies suggested that higher level of vitamin D has a negative effect on the occurrence of UFs, but the results of studies on the relationship between them are conflicting and further evidence needs to be studied. Here in, we used a two-sample Mendelian Randomization (2SMR) study to explore the causal relationship between genetically predicted vitamin D levels and the risk of UFs. The exposure data comes from a genome-wide association study (GWAS) summary dataset consisting of 441,291 individuals, which includes datasets from United Kingdom Biobank, FinnGen Biobank and the corresponding consortia. Single-nucleotide polymorphisms (SNPs) associated with vitamin D at a significant level of p < 5 × 10−8 and low linkage disequilibrium (LD) level (r2 < 0.01) were selected. The outcome data comes from a GWAS dataset of IEU analysis of United Kingdom Biobank phenotypes consisting of 7,122 UFs cases and 455,811 controls. Our inverse-variance weight (IVW) analysis results support the causal association of genetically predicted vitamin D with the risk of UFs (OR = 0.995,95% CI = 0.990-0.999, p = 0.024). In addition, heterogeneity and pleiotropy were not observed in statistical models. In summary, our results indicate that elevated serum vitamin D levels are in strong relationship with reduction of the risk of UFs, which indicates that the clinical treatment of UFs may have a new and excellent option.

A chitosan/poly(vinyl alcohol)-stabilized copper nanoparticle (CP@Cu NPs) was used as a heterogeneous catalyst for the borylation of α, β-unsaturated ketones, MBH alcohols, and MBH esters in mild conditions. This catalyst not only demonstrated remarkable efficiency in synthesizing organoboron compounds but also still maintained excellent reactivity and stability even after seven recycled uses of the catalyst. This methodology provides a gentle and efficient approach to synthesize the organoboron compounds by efficiently constructing carbon–boron bonds.

Rhodamine B (RhB) in dyes is widely used in various industries, but it poses a great threat to the natural environment and human health. In this work, a series of thermosensitive polymer materials, PNxDy, with controllable morphology and particle size were prepared by free radical polymerization using N-isopropylacrylamide and N,N-dimethylacrylamide as monomers. Then, by using PNxDy as a template, bimetallic Mn- and Co-doped MCM-41 molecular sieves with good morphology and properties were prepared by the microwave-assisted hydrothermal method. The effects of a series of thermosensitive templates on the morphology and properties of the Mn-Co-MCM-41 molecular sieve were investigated. The results demonstrated that the Mn-Co-MCM-41 by PN100D4 as a templating agent showed the best mesoporous ordering and the most regular material morphology with 2 nm nanoparticles. In addition, the molecular sieve with the best structure was selected for the RhB degradation experiments. The Mn-Co-MCM-41 with PN100D4 as the template showed regular morphology and uniform pore channels. It was applied as a catalyst for the degradation of RhB by potassium monopersulfate (PMS). The degradation rate of RhB could reach 98% with a 20 min reaction by Mn-Co-MCM-41 (PN100D4). Meanwhile, the degradation rate could be maintained at 91% after being reused six times. The bimetallic-doped Mn-Co-MCM-41 molecular sieves prepared using the thermosensitive material PN100D4 as a template have good catalytic performance and can be effectively reused.

Abstract Pregnant women are generally more susceptible to viral infection. Although the impact of SARS-CoV-2 in pregnancy remains to be determined, evidence indicates that the risk factors for severe COVID-19 are similar in pregnancy to the general population. Here we systemically analyzed the clinical characteristics of pregnant and non-pregnant female COVID-19 patients who were hospitalized during the same period and found that pregnant patients developed marked lymphopenia and higher inflammation evident by higher C-reactive protein and IL-6. To elucidate the pathways that might contribute to immunopathology or protective immunity against COVID-19 during pregnancy, we applied single-cell mRNA sequencing to profile peripheral blood mononuclear cells from four pregnant and six non-pregnant female patients after recovery along with four pregnant and three non-pregnant healthy donors. We found normal clonal expansion of T cells in the pregnant patients, heightened activation and chemotaxis in NK, NKT, and MAIT cells, and differential interferon responses in the monocyte compartment. Our data present a unique feature in both innate and adaptive immune responses in pregnant patients recovered from COVID-19.

Three-dimensional porous polycaprolactone (PCL) bone scaffolds prepared by selective laser sintering (SLS) have demonstrated great potential in the repair of non-load-bearing bone defects. The microgeometry and surface roughness of PCL scaffolds during the SLS process may change the biocompatibility and bioactivity of the scaffolds. However, in addition to the widely concerned mechanical properties and structural accuracy of scaffolds, there is still a lack of systematic research on how SLS process parameters affect the surface roughness of PCL scaffolds and the relationship between roughness and biocompatibility of scaffolds. In this study, we use the energy density model (EDM) combined with the thermodynamic properties of PCL powder to calculate the energy density range (Ed1–Ed3) suitable for PCL sintering. Five PCL scaffolds with different laser powers and scanning speeds were prepared; their dimensional accuracy, mechanical strength, and surface properties were comprehensively evaluated, and the bioactivities were compared through the attachment and proliferation of MC3T3-E1 cells on the scaffolds. It was found that the high energy density (Ed3) reduced the shape fidelity related to pore size and porosity, and the dense and smooth surface of the scaffolds showed poor cytocompatibility, while the low energy density (Ed1) resulted in weak mechanical properties, but the rough surface caused by incomplete sintered PCL particles facilitated the cell adhesion and proliferation. Therefore, the surface roughness and related biocompatibility of PCL bone scaffolds should be considered in energy-density-guided SLS parameter optimization.

In recent years, many studies have indicated that vitamin C might be negatively associated with the risk of cancer, but the actual relationship between vitamin C and cancer remains ambivalent. Therefore, we utilized a two-sample Mendelian randomization (MR) study to explore the causal associations of genetically predicted vitamin C with the risk of a variety of cancers. Single-nucleotide polymorphisms (SNPs) associated with vitamin C at a significance level of p < 5 × 10–8 and with a low level of linkage disequilibrium (LD) (r2 < 0.01) were selected from a genome-wide association study (GWAS) meta-analysis of plasmid concentration of vitamin C consisting of 52,018 individuals. The data of the GWAS outcomes were obtained from United Kingdom Biobank, FinnGen Biobank and the datasets of corresponding consortia. In the inverse-variance weight (IVW) method, our results did not support the causal association of genetically predicted vitamin C with the risk of overall cancer and 14 specific types of cancer. Similar results were observed in sensitivity analyses where the weighted median and MR-Egger methods were adopted, and heterogeneity and pleiotropy were not observed in statistical models. Therefore, our study suggested that vitamin C was not causally associated with the risk of cancer. Further studies are warranted to discover the potential protective and therapeutic effects of vitamin C on cancer, and its underlying mechanisms.

Abstract With the increasing demand for antimicrobial agents and the spread of antibiotic resistance in pathogens, the exploitation of plant oils to partly replace antibiotic emerges as an important source of fine chemicals, functional food utility and pharmaceutical industries. This work introduces a novel catalytic method of plant oils hydroxylation by Fe(III) citrate monohydrate (Fe3+-cit.)/Na2S2O8 catalyst. Methyl (9Z,12Z)-octadecadienoate (ML) was selected as an example of vegetable oils hydroxylation to its hydroxy-conjugated derivatives (CHML) in the presence of a new complex of Fe(II)-species. Methyl 9,12-di-hydroxyoctadecanoate 1, methyl-9-hydroxyoctadecanoate 2 and methyl (10E,12E)-octadecanoate 3 mixtures is produced under optimized condition with oxygen balloon. The specific hydroxylation activity was lower in the case of using Na2S2O8 alone as a catalyst. A chemical reaction has shown the main process converted of plantoils hydroxylation and (+ 16 Da) of OH- attached at the methyl linoleate (ML-OH). HPLC and MALDI-ToF-mass spectrometry were employed for determining the obtained products. It was found that adding oxidizing agents (Na2S2O8) to Fe3+ in the MeCN mixture with H2O would generate the new complex of Fe(II)-species, which improves the C-H activation. Hence, the present study demonstrated a new functional method for better usage of vegetable oils.Producing conjugated hydroxy-fatty acids/esters with better antipathogenic properties. CHML used in food industry, It has a potential pathway to food safety and packaging process with good advantages, fundamental to microbial resistance. Lastly, our findings showed that biological monitoring of CHML-minimum inhibitory concentration (MIC) inhibited growth of various gram-positive and gram-negative bacteria in vitro study. The produced CHML profiles were comparable to the corresponding to previousstudies and showed improved the inhibition efficiency over the respective kanamycin derivatives.

The Ailaoshan orogenic belt is one of the most significant orogenic belts in the southeastern margin of the Qinghai–Tibet Plateau. The widely developed magmatic rocks in this belt preserve the multi-stage tectonic evolution records of the South China Plate. As an important response to the Rodinia breakup tectonic event, the study of Neoproterozoic magmatic rocks in the area is of great significance for reconstructing the Neoproterozoic tectonic process of the Ailaoshan orogenic belt and the tectonic evolution of the South China Plate. Petrology, geochemistry, zircon U-Pb, and Lu-Hf isotopes of the Daping pluton in the Ailaoshan orogenic belt are studied in this paper. The Daping pluton is mainly divided into gabbros and granites. Gabbros and granites belong to the sub-alkaline series, which are relatively enriched in large ion lithophilic elements and depleted in high-field strength elements. The ΣREE contents of the gabbro are low with enrichment in LREEs and depletion in HREEs, and the degree of differentiation of light and heavy rare earth is low, with positive δEu and weak negative δCe anomalies. The ΣREE contents of the granite are low with enrichment in LREEs and depletion in HREEs, and the degree of light and heavy rare earth differentiation is high, with medium–weak negative δEu and weak positive δCe anomalies, suggesting an A2-type granite with A1-A2 transition characteristics. The weighted average age of the gabbro is 816.1 ± 4.1 Ma (MSWD = 0.11), with zircon εHf(t) values of −7.5–5.5. The magma source is a mixture of an ancient crust source and a new mantle source; the weighted average age of the syenogranite is 783.7 ± 8.1 Ma (MSWD = 1.4), with zircon εHf(t) values of −4.3–0.4. The magma source is mainly ancient crustal material (Ailaoshan Group), mixed with a small amount of mantle-derived material; the weighted average age of the monzogranite is 754.8 ± 6.1 Ma (MSWD = 3.0), with positive zircon εHf(t) values of 1.65–10.36. The magma source is a mixture of a large number of mantle-derived materials and a small number of crust-derived materials (Ailaoshan Group). The Daping pluton was formed in the transitional tectonic environment from post-collision to intraplate continental margin rift, corresponding to the Rodinia breakup process in the western margin of the South China Plate.

N-methylation of amines and imines with formic acid as C1 source by easily prepared Pt NPs as heterogeneous catalyst is reported here. Primary, secondary amines and imines were successfully methylated in good to excellent yields under mild conditions. Both aromatic and aliphatic amines could be applied as substrates. Meanwhile, the heterogeneous Pt NPs were capable to be recylcled and reused for 7 times without significant decrease of reactivity. This protocol provided an indirect utilization pathway of CO2 by using formic acid as C1 source.

Abstract Background Fasciola gigantica infection threatens the health of both humans and animals in the world. The excretory/secretory products (ESPs) of this fluke has been reported to impair the activation and maturation of immune cells. We have previously shown the influence of F. gigantica ESPs (FgESPs) on the maturation of buffalo dendritic cells (DCs). However, the underlying mechanisms remain unclear. The objective of this study was to investigate the potency of FgESPs in shifting the differentiation and immune functions of buffalo DCs. Methods Buffalo DCs were incubated with FgESPs directly or further co-cultured with lymphocytes in vitro. qRT-PCR was employed to determine the gene expression profile of DCs or the mixed cells, and an ELISA was used to measure cytokine levels in the supernatants. Hoechst and Giemsa staining assays, transmission electron microscopy, caspase-3/7 activity test and histone methylation test were performed to determine DC phenotyping, apoptosis and methylation. To investigate the mechanism involved with DNA methylation, a Co-IP assay and immunofluorescent staining assay were performed to observe if there was any direct interaction between FgESPs and DNMT1/TET1 in buffalo DCs, while RNAi technology was employed to knockdown DNMT1 and TET1 in order to evaluate any different influence of FgESPs on DCs when these genes were absent. Results qRT-PCR and ELISA data together demonstrated the upregulation of DC2 and Th2/Treg markers in DCs alone and DCs with a mixed lymphocyte reaction (MLR), suggesting a bias of DC2 that potentially directed Th2 differentiation in vitro. DC apoptosis was also found and evidenced morphologically and biochemically, which might be a source of tolerogenic DCs that led to Treg differentiation. In addition, FgESPs induced methylation level changes of histones H3K4 and H3K9, which correlate with DNA methylation. Co-IP and immunofluorescent subcellular localization assays showed no direct interaction between the FgESPs and DNMT1/TET1 in buffalo DCs. The productions of IL-6 and IL-12 were found separately altered by the knockdown of DNMT1 and TET1 in DCs after FgESPs treatment. Conclusions FgESPs may induce the DC2 phenotype or the apoptosis of buffalo DCs to induce the downstream Th2/Treg response of T cells, possibly through a DNMT1- or TET1-dependent manner(s).

Abstract Backgrounds Major depressive disorder is an ordinary mental disorder, and suicide is considered to be a major concern among patients with MDD. Previous studies focused on the relationship between suicide attempts and metabolism in elderly patients with MDD, while ignore the young people. The aim of this study is to find the potential relationship between suicide attempts and metabolism in young patients with MDD to find a way to prevent and ultimately reduce suicide in young patients with MDD. Methods Cross-sectional design was employed in the study.740 patients aged between 18 and 45 years old with MDD had been consecutively recruited in this study between 2011 and 2017, 128 of whom had suicide attempts. Their serum samples used to monitor fasting blood glucose, serum lipids as well as socio-demographic characteristics were collected. Besides, some clinical scales were also employed to measure symptoms of anxiety, depression and other conditions.
