AIM: To investigate the mechanism of action of Qingxuan Runmu Yin(QRY)in the treatment of dry eye(DE)based on transcriptomics and network pharmacology, and to validate the efficacy and key targets of QRY through a animal model of DE.METHODS:RNA-sequencing(RNA-seq)technology was used to detect differentially expressed genes(DEGs)between mice in the DE group and mice in the normal control group, the active ingredients and potential targets of QRY were screened through database, and gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analysis were carried out after overlapping the results and obtaining key targets. Additionally, “drug-component-target signaling pathways” network was built and the protein-protein interaction(PPI)was analyzed. Mice were examined for Schirmer I test(SⅠt), tear film breakup time(BUT), and corneal fluorescein staining(FL)every 7 d from the beginning of the animal experiments. Hematoxylin-eosin staining(HE)was performed to observe pathologic changes in mouse corneal tissues. Enzyme linked immunosorbent assay(ELISA), Western blot and quantitative real-time polymerase chain reaction(qRT-PCR)were performed to verify the mRNA and protein expression levels of the core targets in mouse corneal tissues.RESULTS:Totally 2 234 DEGs, 233 active ingredients and 457 related targets of QRY were collected, with a total of 64 key targets obtained. GO function and KEGG pathway results showed that QRY was closely related to inflammatory mediators, and 19 core targets such as interleukin-1β(IL-1β)were screened by PPI network construction; SⅠt, BUT and FL results in the QRY group were statistically significantly different compared with the model group(all P

IntroductionThe rapid development of urbanization has brought about changes in residents’ living environment and behavior, leading to health challenges such as hypertension. An improvement in the built-up environment in the community could contribute to the construction of a healthy city, promote the active life of the residents, and prevent and relieve hypertension. However, there is little research on the relationship between the built environment of the community and hypertension. This cross-sectional study aims to evaluate the relationship between communities’ built environment, health behavior, and hypertension grade of residents in Yuhui District of Bengbu City.MethodsThis study is based on data from the 2022 Health Survey of Residents in 21 communities. To investigate the impact of the community’s built environment on residents’ hypertension and the underlying mechanisms, regression and structural equation modeling were employed.Results and discussionThe results show that the built environment of urban communities has a significant impact on the residents’ hypertension. The presence of high densities of supermarkets, convenience stores, parks and plazas, but low densities of clinics and hospitals, has been identified as a significant risk factor for the development of high blood pressure among the residents. Nevertheless, the adoption of healthy behaviors, including regular walking, physical activity, and a diet rich in fruit and vegetables, can play an important role in reducing the risk of hypertension. The findings of this study show that enhancements to the built environment in urban neighborhoods could contribute to a reduction in the prevalence of hypertension among residents. Furthermore, the implementation of efficacious health interventions in urban settings would facilitate the alteration of residents’ health behaviors and enhance their overall health status.

BackgroundChronic insomnia is characterized by a prolonged and recurrent course. The efficacy of repeated transcranial magnetic stimulation （rTMS） as a physical therapy method to improve sleep quality remains inadequately supported by evidence， particularly regarding its relationship with personality traits.ObjectiveTo explore the efficacy and influencing factors of rTMS in the treatment of chronic insomnia， and to provide insights into its therapeutic potential.MethodA total of 46 patients who met the diagnostic criteria for chronic insomnia according to the Diagnostic and Statistical Manual of Mental Disorders， fifth edition （DSM-5）， and were treated at the Third Hospital of Mianyang from September 2022 to September 2023 were selected. Prior to treatment， participants underwent assessments using the Eysenck Personality Questionnaire （EPQ）， Hamilton Depression Scale-17 item （HAMD-17） and Hamilton Anxiety Scale （HAMA）. The Pittsburgh Sleep Quality Index （PSQI） was used to assess sleep quality before treatment， at the end of the second week of treatment and one week post-treatment.ResultsAt the end of the second week of treatment， patients exhibited significantly improved total PSQI score and subscale scores related to subjective sleep quality， sleep latency， sleep duration， sleep efficiency， sleep disturbance and daytime dysfunction （t=4.755~13.361， P

The rapid development of modern aerospace and energy industries requires increasingly high service performance of metallic materials. Laves phase intermetallic compounds with the chemical formula AB2-type have attracted much attention due to their high melting point, excellent high-temperature strength, and oxidation resistance, and are regarded as one of the promising candidates for a new generation of high-temperature structural applications. Although it can be identified that Laves phase alloy research has been an active research field in the last 20 years in terms of the trend of literature volume growth, there is still a lack of systematic descriptions and comprehensive overviews of the latest frontiers and hotspots in the field. In this article, a quantitative and visual knowledge mapping of the articles published in the field of Laves phase research in the last 20 years has been carried out based on the bibliometric analysis methodology of Cite Space software. Quantitative analysis of high-frequency keywords to understand the basic development of Laves phase and the evolution of the main research hotspots in each period of time identified the hotspots of common concern seven themes: Laves phase, electronic structure, precipitation, Inconel 718 alloy, phase diagram, high-entropy alloys, and TiAl-based alloys. The analysis of the emergent keywords shows that electronic structure, high-entropy alloys, total energy calculations, and laser cladding can represent the research frontiers of Laves phases. This study provides systematic and valuable reference information to gain insight into the current state of Laves phase research and the evolution of the theme, which will be useful for further research.

BackgroundInsomnia disorder has become a common disease in the current society. Cognitive Behavior Therapy for Insomnia （CBTI） is one of the non-drug treatment methods for insomnia disorder， but relevant studies of its effect on sleep quality and cognitive function of patients with insomnia disorder are limited.ObjectiveTo explore the effects of CBTI on sleep quality and cognitive function in patients with insomnia disorder， so as to provide references for non-drug treatment of insomnia disorder.MethodsA total of 47 patients with insomnia disorder were recruited as the study subjects. They all met the diagnostic criteria of the Diagnostic and Statistical Manual of Mental Disorders， fifth edition （DSM-5） and have visited Sichuan Mental Health Center from January 2021 to October 2022. The patients underwent CBTI for 6 weeks. Before the treatment， depression and anxiety symptoms were assessed using Hamilton Depression Scale-24 item （HAMD-24） and Hamilton Anxiety Scale （HAMA）. Sleep status and cognitive function were assessed using Pittsburgh Sleep Quality Index （PSQI） and Montreal Cognitive Assessment （MoCA） before and 6 weeks after the treatment. Spearman correlation analysis was used to examine the correlation between the reduction of PSQI score and the increase of MoCA score after treatment.ResultsAfter the 6-week treatment， the factor scores and total score of PSQI across 6 subscales （the sleep quality， sleep onset time， sleep time， sleep efficiency， sleep disorder and daytime dysfunction） were lower than those before the treatment， and the score differences were of statistical significance （t=5.569~15.290， P

Objective: To explore the correlation between insomnia and language ability in patients with schizophrenia. Methods: 120 patients with schizophrenia admitted to our hospital from June 2020 to January 2022 were enrolled as the research objects. According to the total score of the Pittsburgh Sleep Index (PSQI), they were divided into the insomnia group (PSQI total score > 10 points) and the non-insomnia group (PSQI total score ≤ 10 points). To compare the difference in verbal fluency scale between insomnia and non-insomnia groups and to understand the correlation between insomnia symptoms and language ability in patients with schizophrenia. Results: There were no significant differences in age, gender, and years of education between the insomnia group and the non-insomnia group (P > 0.05). Studies have found that the phosphatase actin regulatory factor 1 expression can be related to stroke, but it remains unclear whether changes in phosphatase actin regulatory factor 1 expression also play a role in traumatic brain injury. In this study we found that, in a mouse model of traumatic brain injury induced by controlled cortical impact, phosphatase actin regulatory factor 1 expression is increased in endothelial cells, neurons, astrocytes, and microglia. When we overexpressed phosphatase actin regulatory factor 1 by injection an adeno-associated virus vector into the contused area in the traumatic brain injury mice, the water content of the brain tissue increased. However, when phosphatase actin regulatory factor 1 was knocked down, the water content decreased. We also found that inhibiting phosphatase actin regulatory factor 1 expression regulated the nuclear factor kappa B signaling pathway, decreased blood-brain barrier permeability, reduced aquaporin 4 and intercellular adhesion molecule 1 expression, inhibited neuroinflammation, and neuronal apoptosis, thereby improving neurological function. The findings from this study indicate that phosphatase actin regulatory factor 1 may be a potential therapeutic target for traumatic brain injury.

Sirtuin 2 (SIRT2) inhibition or Sirt2 knockout in animal models protects against the development of neurodegenerative diseases and cerebral ischemia. However, the role of SIRT2 in traumatic brain injury (TBI) remains unclear. In this study, we found that knockout of Sirt2 in a mouse model of TBI reduced brain edema, attenuated disruption of the blood-brain barrier, decreased expression of the nucleotide-binding oligomerization domain-like receptor protein 3 (NLRP3) inflammasome, reduced the activity of the effector caspase-1, reduced neuroinflammation and neuronal pyroptosis, and improved neurological function. Knockout of Sirt2 in a mechanical stretch injury cell model in vitro also decreased expression of the NLRP3 inflammasome and pyroptosis. Our findings suggest that knockout of Sirt2 is neuroprotective against TBI; therefore, Sirt2 could be a novel target for TBI treatment.

Objective To explore the effect of Toll-like receptor 3 (TLR3) activation on the expression of neuronal disrupted-in-schizophrenia 1 (DISC1). Methods C57BL/6J mouse hippocampal neurons were incubated with polyI:C(1 μg/mL) or/and mIFNα(100 IU/mL) for 24 hrs. DISC1 mRNA and protein were detected by quantitive RT-PCR, immuno-cytochemistry staining and Western blot. The expression of STAT1, phospho-STAT1, p38 MAPK, phospho-p38 MAPK, GSK-3β, and phospho-GSK-3β was also detected by Western blot. Results Comparing with control group, the mRNA and protein expression of DISC1 in polyI:C group and IFNα group didn't show any changes, but significantly decreased with the co-stimulation by polyI:C and IFNα (P＜0.05). In addition, the phosphorylation of STAT1 was significantly increased in both IFNα group and co-stimulation group(P＜0.05) and the phosphorylation of p38 MAPK was elevated in polyI:C group and co-stimulation group (P＜0.05).The expression of GSK-3β and phospho-GSK-3β had no changes. Conclusions The combined stimulations of IFNα and TLR3 activation may inhibit the expression of DISC1 potentially through JAK/STAT1 and p38 MAPK signaling pathways.

Abstract Background Microglia-mediated neuroinflammatory response following traumatic brain injury (TBI) is considered as a vital secondary injury factor, which drives trauma-induced neurodegeneration and is lack of efficient treatment. ACT001, a sesquiterpene lactone derivative, is reportedly involved in alleviation of inflammatory response. However, little is known regarding its function in regulating innate immune response of central nervous system (CNS) after TBI. This study aimed to investigate the role and underlying mechanism of ACT001 in TBI. Methods Controlled cortical impact (CCI) models were used to establish model of TBI. Cresyl violet staining, evans blue extravasation, neurobehavioral function assessments, immunofluorescence and transmission electron microscopy were used to evaluate therapeutic effects of ACT001 in vivo. Microglial depletion was induced by administering mice with colony stimulating factor 1 receptor (CSF1R) inhibitor, PLX5622. Cell-cell interaction models were established as co-culture system to simulate TBI conditions in vitro. Cytotoxic effect of ACT001 on cell viability was assessed by cell counting kit-8 and activation of microglia cells were induced by Lipopolysaccharides (LPS). Pro-inflammatory cytokines expression was determined by Real-time PCR and nitric oxide production. Apoptotic cells were detected by TUNEL and flow cytometry assays. Tube formation was performed to evaluate cellular angiogenic ability. ELISA and western blot experiments were used to determine proteins expression. Pull-down assay was used to analyze proteins that bound ACT001. Results ACT001 relieved the extent of blood-brain barrier integrity damage and alleviated motor function deficits after TBI via reducing trauma-induced activation of microglia cells. Delayed depletion of microglia with PLX5622 hindered therapeutic effect of ACT001. Furthermore, ACT001 alleviated LPS-induced activation in mouse and rat primary microglia cells. Besides, ACT001 was effective in suppressing LPS-induced pro-inflammatory cytokines production in BV2 cells, resulting in reduction of neuronal apoptosis in HT22 cells and improvement of tube formation in bEnd.3 cells. Mechanism by which ACT001 functioned was related to AKT/NFκB/NLRP3 pathway. ACT001 restrained NFκB nuclear translocation in microglia cells through inhibiting AKT phosphorylation, resulting in decrease of NLRP3 inflammasome activation, and finally down-regulated microglial neuroinflammatory response. Conclusions Our study indicated that ACT001 played critical role in microglia-mediated neuroinflammatory response and might be a novel potential chemotherapeutic drug for TBI. Video Abstract

This paper aims to analyze the clinical efficacy of cognitive behavioral therapy for insomnia （CBTI） in chronic insomnia treatment， with a view to providing a theoretical basis for promoting the clinical use of CBTI and providing a reference for the non-pharmacological treatment of chronic insomnia patients in China. Chronic insomnia is a common disorder， and CBTI has emerged as the first approach to treating chronic insomnia， so this review focuses on the current status， clinical efficacy， adverse effects， existed problems and management strategies of CBTI for chronic insomnia treatment from the perspectives of physiological mechanisms and neuroimaging.

Urolithin A (UA) is a natural metabolite produced from polyphenolics in foods such as pomegranates, berries, and nuts. UA is neuroprotective against Parkinson’s disease, Alzheimer’s disease, and cerebral hemorrhage. However, its effect against traumatic brain injury remains unknown. In this study, we established adult C57BL/6J mouse models of traumatic brain injury by controlled cortical impact and then intraperitoneally administered UA. We found that UA greatly reduced brain edema; increased the expression of tight junction proteins in injured cortex; increased the immunopositivity of two neuronal autophagy markers, microtubule-associated protein 1A/B light chain 3A/B (LC3) and p62; downregulated protein kinase B (Akt) and mammalian target of rapamycin (mTOR), two regulators of the phosphatidylinositol 3-kinase (PI3K)/Akt/mTOR signaling pathway; decreased the phosphorylation levels of inhibitor of NFκB (IκB) kinase alpha (IKKα) and nuclear factor kappa B (NFκB), two regulators of the neuroinflammation-related Akt/IKK/NFκB signaling pathway; reduced blood-brain barrier permeability and neuronal apoptosis in injured cortex; and improved mouse neurological function. These findings suggest that UA may be a candidate drug for the treatment of traumatic brain injury, and its neuroprotective effects may be mediated by inhibition of the PI3K/Akt/mTOR and Akt/IKK/NFκB signaling pathways, thus reducing neuroinflammation and enhancing autophagy.

Based on the first-principles method of density functional theory, the microscopic mechanism of the effect of addition of alloying element Ru content on the stability and elastic properties of Laves phase TaCr2 was investigated by parameters such as formation enthalpy, electronic structure, and elastic constants. The addition of Ru atoms tends to preferentially occupy the lattice sites of Cr. With the increase in the Ru content, the alloying ability of Ta8Cr16−n Run (n = 0–6) becomes progressively weaker, the stability gradually decreases, whereas the Poisson’s ratio grows. The bonding peak appears to drop and widen, weakening the bonding strength of Ta–Cr atoms, rendering the shear deformation to be performed easily, thereby improving toughness. When the Ru content rises to 20.83 at%, the bulk modulus, shear modulus, Young’s modulus, and Poisson’s ratio of the alloy attain the maximum value, the brittleness diminishes to the most extent, the resistance to elastic deformation is the strongest, as well at the optimum fracture toughness.

ObjectiveTo establish a method for the determination of several kinds of pyrethroids and organochlorine pesticides in Chinese oolong tea samples based on liquid-liquid extraction-solid phase extraction-gas chromatography-negative chemical ionization-mass spectrometry (GC-NCI-MS).MethodsOolong tea samples were crushed and screened, soaked in boiling water, extracted with acetone-n-hexane, purified by Carb-NH2 column, and then identified and quantified by GC-NCI-MS.ResultsAll the 18 kinds of pyrethroids and 9 kinds of organochlorine pesticides showed good linear relationships in range of 0.006-1.00 μg/mL and 0.006-0.50 μg/mL respectively, and the correlation coefficients were above 0.996. The recoveries at different spiked levels for all target compounds in blank matrices were 60.1%-119.8%, and the relative standard deviation (RSD) were 1.5%-10.4% (n=6). The limits of detection and quantification of the method were 0.5-4.8 μg/kg and 1.7-16.0 μg/kg, respectively.ConclusionThe method was suitable for rapid screening and analysis of 18 pyrethroids and 9 organochlorine pesticides in Chinese oolong tea with the advantage of accuracy, rapidity and high sensitivity.

Abstract Presented are the synthesis, characterizations, and reactive surface modification (RSM) of a novel nine atomic layered V4C3T x MXene. With the advantages of the multilayered V4C3T x MXene that can simultaneously support the RSM reaction and keep the inner skeleton stable, a series of amorphous Ni/Fe/V‐ternary oxide hydroxides thin layer can be successfully modified on the surface of the V4C3T x MXene (denoted as MOOH @V4C3T x, M = Ni, Fe, and V) without disrupting its original structure. Attributed to the in situ reconstruction of highly active oxide hydroxide layer, the nanohybrids exhibited an enhanced oxygen evolution reaction (OER) activity and excellent long‐time stability over 70 hours. In particular, a current density of 10 mA cm−2 can be reached by the nanohybrid with the optimized Ni/Fe ratio at an overpotential (η) as low as 275.2 mV, which is comparable to most of the state‐of‐the‐art OER catalysts and better than other MXene‐based derivatives. Demonstrated by the tunable physicochemical properties and excellent structural stability of these nanohybrids, we may envision the promising role of the M4X3‐based MXenes as substrates for a wide range of energy conversion and storage materials.

This paper summarizes the rapid detection methods of edible vegetable oil components from the aspects of analysis principle and application technology, such as simulation sensory analysis, spectral analysis, electromagnetic spectrum analysis and biochip technology. The application results of these techniques in the component mixing identification of edible vegetable were further analyzed. It was showed that the integrated online rapid detection technology of multi-channel, multi parameter and multi capacity will be the developing direction of component analysis of edible vegetable oil and will lay a technical foundation for further improving the quality supervision of edible vegetable oil and safeguarding the interests of consumers in China.

Allergic rhinitis (AR) is a common disease that causes severe inflammation and even disabilities. Previous studies have reported baicalein to have an anti-inflammatory effect. However, the pharmacological action of baicalein on anaphylaxis has not been clarified yet. This study assessed the in vivo protective effect of baicalein post-treatment in an ameliorating ovalbumin (OVA)-sensitized AR rat model. Baicalein attenuated histological alterations, aberrant tissue repair and inflammation after OVA-induced AR. Baicalein reduced the frequency of nasal/ear rubs and sneezes in rats, and inhibited generation of several inflammatory cytokines (TNF-α, IL-1β, and IL-6) in both blood and nasal lavage of rats. Infiltrations of eosinophils, lymphocyte, and neutrophils were decreased in baicalein-administered rats. Furthermore, baicalein inhibited the expression of STAT3 phosphorylation in the nasal mucosa. In summary, baicalein attenuated OVA-induced AR and inflammation, which suggests it as a promising therapeutic agent for the alleviation of AR-associated inflammation and pathology.

Traumatic brain injury (TBI) could highly induce coagulopathy through breaking the dynamic balance between coagulation and fibrinolysis systems, which may be a major contributor to the progressive secondary injury cascade that occurs after TBI. Phosphatase and tensin homolog deleted on chromosome 10 (PTEN) inhibition is reported to exert neuroprotection in TBI, making it a potential regulatory target involved in TBI-induced coagulation disorder. PTEN level is controlled in a major way by E3 ligase-mediated degradation through the ubiquitin–proteasome system. The C terminus of Hsc70-interacting protein (CHIP) has been shown to regulate proteasomal degradation and ubiquitination level of PTEN. In the present study, CHIP was overexpressed and knocked down in mouse brain microvascular endothelial cells (bEnd.3) and tissues during the early phase of TBI. In vitro cell proliferation, cell apoptosis, migration capacity, and invasion capacity were determined. The changes of procoagulant and apoptosis molecules after TBI were also detected as well as the micrangium density and blood–brain barrier permeability after in vivo TBI. In vitro results demonstrated that CHIP overexpression facilitated bEnd.3 cell proliferation, migration, and invasion and downregulated cell apoptosis and the expressions of procoagulant molecules through promoting PTEN ubiquitination in a simulated TBI model with stretch-induced injury treatment. In vivo experiments also demonstrated that CHIP overexpression suppressed post-TBI apoptosis and procoagulant protein expressions, as well as increased microvessel density, reduced hemorrhagic injury, and blood–brain barrier permeability. These findings suggested that the upregulation of CHIP may attenuate apoptosis and procoagulant activity, facilitate brain repair, and thus exerts neuroprotective effects in TBI.

Traumatic brain injury (TBI) is a major cause of disability and mortality worldwide, in part resulting from secondary apoptosis of neurons in peri-contusion areas. Serpina3k, a serine protease inhibitor, has been shown to inhibit apoptosis in injury models. In this study, we investigated the anti-apoptotic function of serpina3k in vivo using a mouse model of TBI, as well as the underlying neuroprotective mechanism in vitro using the SH-SY5Y human neuroblastoma cell line. TBI was induced in adult male C57BL/6 mice using controlled cortical impact. Serpina3k protein was intravenously administered at a concentration of 0.5 mg/kg twice daily for up to 14 days. SH-SY5Y cells were subjected to biaxial stretch injury and then treated with different concentrations of serpina3k. We found that endogenous serpina3k protein levels were elevated in peri-contusion areas of the mouse brain following TBI. Serpina3k-treated mice had fewer apoptotic neurons, lower levels of oxidative stress, and showed greater recovery of neurological deficits relative to vehicle-treated mice. Meanwhile, in the SH-SY5Y cell injury model, serpina3k at an optimal concentration (150 nM) inhibited the generation of intracellular reactive oxygen species, abrogated changes of the mitochondrial membrane potential, and reduced the phospho-extracellular regulated protein kinases (p-ERK)/ERK, phospho-P38 (p-P38)/P38, B cell lymphoma (Bcl)-2-associated X protein/Bcl-2, and cleaved caspase-3/caspase-3 ratios, thereby reducing the apoptosis rate. These results demonstrate that serpina3k exerts a neuroprotective function following TBI and thus has therapeutic potential.

The after-school program is a crucial initiative for implementing the Double Reduction policy; however, prior research has not provided conclusive evidence on whether extended school hours contribute to students' cognitive and non-cognitive development or on which types of after-school services are more beneficial for student development. This study analyzed 37 after-school programs from 18 publications using meta-analytic techniques, and the results indicated that participation in after-school programs had positive effects on student cognitive and non-cognitive development despite the small effect size (d = 0.327, p = 0.000). The decomposition of the effects of after-school programs revealed that they had modestly positive effects on academic achievement (d = 0.369) and social-emotional competence (d = 0.220). In addition, the analysis of moderating variables revealed that socioeconomic status, educational phase, number of after-school service days per week, sample size, and testing instrument all influenced the after-school program effects. This study concludes, based on the results of the meta-analysis, that there should be a balanced consideration of the development of student cognitive and non-cognitive abilities in planning after-school service, a substantial variety of activities in afterschool programs, a flexible adoption of diverse after-school programs, and a reasonable participation frequency in after-school service.

Based on the continuum hypothesis, problem of compressing wave was studied analytically. By exploring the temperature distribution, the propagation velocity, and the thickness of the transition region, the developing process of compressing wave turning into shock wave was revealed, and the following conclusions were reached: (1) the governing equations of compressing wave can be turned into two autonomous equations, and the phase diagram can be used as an effective tool for the analysis of the compressing wave; (2) the solution of compressing wave was composed of two parts: the inner solution and the outer solution; when Pr=3/4, the analytical inner solution can be obtained; (3) as the disturbance velocity increases, the thickness of compressing wave will decrease, and the propagation velocity of the compressing wave will increase, and the compressing wave will become shock wave when the disturbance velocity is large enough; (4) velocity and temperature across compressing wave change monotonically and continuously, but the entropy generation changing tendency is closely related to Pr; therefore, the inner solution reveals the mechanism of irreversibility happening in compressing waves.
