Abstract

The lack of periosteum in bone allografts is considered a major cause of bone graft failure; however, the currently used artificial periosteum has failed to proactively adapt to its chronological role in the repair process. This study designed an adaptive periosteal system with dual‐response mechanisms to enable interactive angiogenic/osteogenic stimulation customized for inflammatory and anabolic stages of bone healing. Utilizing the enzymatic reactions and photodynamic properties of gold nanorod‐porous silicon, angiogenic peptides (QK) are released in response to matrix metalloproteinase 2 (MMP‐2), whereas bone‐forming peptides (BFP) are triggered by timed near‐infrared (NIR) irradiation, providing a portfolio of growth factors that match the dynamics of bone healing. The incorporation of such nanocarriers into a poly(N‐isopropyl acrylamide)/methacrylic acid gel (PNIPAM/GelMA) successfully formed a bionic periosteum that actively adapted to the function of the periosteum in different scenarios, resulting in more than doubled its ability to promote angiogenesis, stem cell migration, and osteogenesis compared to the controls. Furthermore, over 80% of the defects are covered with a new scab after the timed near‐infrared irradiation of large bone defects implanted with the system. Abstract

Chlorogenic acid (CGA), one of the most abundant polyphenols in the diet, has been reported to have potent anti-inflammatory properties. However, the effect of CGA on lipopolysaccharide (LPS)-induced mice mastitis has not been investigated. The purpose of the present study was to elucidate whether CGA could ameliorate the inflammation response in LPS-induced mice mastitis and to clarify the possible mechanism. The mouse model of mastitis was induced by injection of LPS through the duct of mammary gland. CGA was administered intraperitoneally with the dose of 12.5, 25, and 50mg/kg respectively 1h before and 12h after induction of LPS. In this study, the effect of CGA on LPS-induced mice mastitis was assessed through histopathological examination, ELISA assay, and western blot analysis. The results showed that CGA significantly reduced TNF-α, IL-1β, and IL-6 production compared with LPS group. Besides, western blot analysis showed that CGA could inhibit the expression of TLR4 and the phosphorylation of NF-κB and IκB induced by LPS. These results suggested that anti-inflammatory effects of CGA against LPS-induced mastitis may be due to its ability to inhibit TLR4-mediated NF-κB signaling pathway. Therefore, CGA may be a potent therapeutic reagent for the prevention of the immunopathology encountered during Escherichia coli elicited mastitis.

Abstract

Selenium, in the form of selenoproteins, plays a pivotal role in anti-inflammatory processes and antioxidant defense system. The aim of this study was to examine the effects of selenium on lipopolysaccharide (LPS)-induced inflammatory responses in bovine mammary epithelial cells (bMEC) and to investigate the potential mechanism. bMEC viability was measured by MTT assay. TNF-α, IL-1β, cyclooxygenase-2 (COX-2), and inducible nitric oxide synthase (iNOS) messenger RNA (mRNA) expressions were evaluated by quantitative real-time polymerase chain reaction (qRT-PCR). The activation of nuclear factor-kappa B (NF-κB) was determined by Western blotting. The results showed that the mRNA expressions of these inflammatory factors were significantly inhibited by selenium in a dose-dependent manner. At protein levels, Western blot analysis demonstrated that selenium dose-dependently decreased NF-κB p65 translocating from the cytoplasm to the nucleus. Taken together, these results suggest that the anti-inflammatory property of selenium in LPS-stimulated primary bMEC may be attributed to the downregulation of NF-κB activation.
