Abstract Introduction Alveolar echinococcosis (AE), caused by the larval forms of Echinococcus multilocularis, is a zoonotic disease affecting the liver, lungs, lymph nodes, kidneys, brain, bones, thyroid, and other organs. Diagnosing AE in a non-endemic area is usually challenging. With the rapid development and increasing application of sequencing techniques in recent years, metagenomic next-generation sequencing (mNGS) has become a powerful tool for diagnosing rare infectious diseases. Case Presentation A 45-year-old woman was admitted to the hospital for the presence of pulmonary shadows for more than 3 months. The lung computed tomography (CT) at a local hospital revealed scattered solid and quasi-circular nodules in the left upper lobe, left lower lobe, right middle lobe, and right lower lobe. The largest nodule was located in the dorsal part of the right lung, measuring 2.0 × 1.7 × 1.5 cm. Moreover, abdominal CT revealed one space-occupying lesion each in the left and right lobes. The pathological analysis of the lung biopsy specimen revealed infiltration of lymphocytes, plasma cells, and eosinophils in the alveolar wall and interstitial area. No pathogenic bacteria were observed in the sputum smear and culture tests. There were no parasite eggs in the stool. The mNGS of the lung puncture tissue revealed 6156 sequence reads matching E. multilocularis; thus, the condition was diagnosed as AE. Albendazole 400 mg was administered twice daily, and the patient was stable during follow-up. Conclusion This case emphasizes the role of mNGS in diagnosing AE. As a novel, sensitive, and accurate diagnostic method, mNGS could be an attractive approach for facilitating early diagnosis and prompt treatment of infectious diseases, especially when the infection was caused by rare pathogens.

Multimodal sentiment analysis faces a number of challenges, including modality missing, modality heterogeneity gap, incomplete datasets, etc. Previous studies usually adopt schemes like meta-learning or multi-layer structures. Nevertheless, these methods lack interpretability for the interaction between modalities. In this paper, we constructed a new dataset, SM-MSD, for sentiment analysis in social media (SAS) that differs significantly from conventional corpora, comprising 10K instances of diverse data from Twitter, encompassing text, emoticons, emojis, and text embedded in images. This dataset aims to reflect authentic social scenarios and various emotional expressions, and provides a meaningful and challenging evaluation benchmark for multimodal sentiment analysis in specific contexts. Furthermore, we propose a multi-task framework based on heterogeneous graph neural networks (H-GNNs) and contrastive learning. For the first time, heterogeneous graph neural networks are applied to multimodal sentiment analysis tasks. In the case of additional labeling data, it guides the emotion prediction of the missing mode. We conduct extensive experiments on multiple datasets to verify the effectiveness of the proposed scheme. Experimental results demonstrate that our proposed scheme surpasses state-of-the-art methods by 1.7% and 0 in accuracy and 1.54% and 4.9% in F1-score on the MOSI and MOSEI datasets, respectively, and exhibits robustness to modality missing scenarios.

Background and aim: Insulin-like growth factor-1 may be involved in the epithelial-to-mesenchymal transition process. It can mitigate adverse effects when interacting with insulin-like growth factor binding protein 3. This study aimed to explore alterations in the expression of these two factors in the colonic tissue of mice with ulcerative colitis. Method: This study utilized animal models. Mice were randomly allocated into three distinct groups. Disease activity index assessment was performed first, followed by histological grading of colitis. Protein and mRNA expression levels were determined using Western blotting and RT-qPCR. Immunohistochemical detection was used to determine histochemistry scores. Pearson correlation and SPSS 25.0 software were used for data analysis. Results: The findings indicated a reduction in the expression of the two investigated factors as well as in epithelial-to-mesenchymal transition epithelial markers during inflammation, while the expression of noninflammatory factors increased. These effects were notably amplified following treatment. Interestingly, the changes in epithelial-to-mesenchymal transition-inducing factors and mesenchymal markers contradicted this trend. Pearson correlation analysis revealed a correlation between molecular indicators of change and epithelial-to-mesenchymal transition. Conclusion: Insulin-like growth factor-1 and insulin-like growth factor binding protein 3 may play a protective role in the development and progression of ulcerative colitis, potentially through their inhibition of the epithelial-to-mesenchymal transition. These factors hold promise as targets for the clinical diagnosis and treatment of ulcerative colitis.

Abstract Determining the precise course of bacterial infection requires abundant in vivo real‐time data. Synchronous monitoring of the bacterial load, temperature, and immune response can satisfy the shortage of real‐time in vivo data. Here, we conducted a study in the joint‐infected mouse model to synchronously monitor the bacterial load, temperature, and immune response using the second near‐infrared (NIR‐II) fluorescence imaging, infrared thermography, and immune response analysis for 2 weeks. Staphylococcus aureus (S. aureus) was proved successfully labeled with glucose‐conjugated quantum dots in vitro and in subcutaneous‐infected model. The bacterial load indicated by NIR‐II fluorescence imaging underwent a sharp drop at 1 day postinfection. At the same time, the temperature gap detected through infrared thermography synchronously brought by infection reached lowest value. Meanwhile, the flow cytometry analysis demonstrated that immune response including macrophage, neutrophil, B lymphocyte, and T lymphocyte increased to the peak at 1 day postinfection. Moreover, both M1 macrophage and M2 macrophage in the blood have an obvious change at ~ 1 day postinfection, and the change was opposite. In summary, this study not only obtained real‐time and long‐time in vivo data on the bacterial load, temperature gap, and immune response in the mice model of S. aureus infection, but also found that 1 day postinfection was the key time point during immune response against S. aureus infection. Our study will contribute to synchronously and precisely studying the complicated complex dynamic relationship after bacterial infection at the animal level.

Abstract Background Paclitaxel, a tubulin-binding agent, is a Food and Drug Administration-approved first-line drug for the treatment of non-small cell lung cancer (NSCLC), for both squamous and non-squamous cell lung carcinoma, with paclitaxel/carboplatin + bevacizumab a common chemotherapy regimen for stage IV non-squamous NSCLC; however, primary or acquired resistance to paclitaxel is gradually increasing, leading to treatment failure. Methods Our results show that Ras-related C3 botulinum toxin substrate 3 (RAC3) is overexpressed in cultured paclitaxel-resistant cells and that RAC3 expression levels are negatively correlated with sensitivity of lung adenocarcinoma cells to paclitaxel. Pulsatilla saponin D could inhibit RAC3 expression, and we hypothesize that it may block paclitaxel resistance. Further, we found that treatment with paclitaxel combined with Pulsatilla saponin D, can overcome lung adenocarcinoma cell resistance to paclitaxel alone in cell culture and mouse xenograft models.

Remote sensing images play a critical role in urban planning, land resources, and environmental monitoring. Land cover classification is one of the straightforward applications of remote sensing. However, the anomalous remote sensing data challenges the reliability of land cover classification results. Deep learning has been widely used in remote sensing image analysis, but it remains sensitive to anomalous data. To address this issue, we reevaluate a land cover classification map in high-noise scenarios with anomalous data and propose a novel network architecture to solve the problem. A new network architecture is proposed to solve this problem. Our proposed network architecture focuses on decoupling the extraction of global information and local information. Through three global–local feature fusion modules, we output features emphasizing global information, features emphasizing local information, and consistency evaluation scores, respectively. A specially designed decoder integrates these three features. Our method performs better compared to mainstream models on the public datasets the Wuhan high-definition landscape dataset with obvious anomaly data, with a mean intersection over union (MIoU) of 63.58% and a mean pixel accuracy (Mpa) of 74.32%. Compared to the suboptimal method, our method improves MIoU by 1.29% and Mpa by 3.05%.

Abstract Full activation of the NLRP3 inflammasome needs two sequential signals: a priming signal, followed by a second, assembly signal. Several studies have shown that the two signals trigger post-translational modification (PTM) of NLRP3, affecting activity of the inflammasome, however, the PTMs induced by the second signal are less well characterized. Here, we show that the assembly signal involves acetylation of NLRP3 at lysine 24, which is important for the oligomerization and the actual assembly of NLRP3 without affecting its recruitment to dispersed trans-Golgi network (dTGN). Accordingly, NLRP3 inflammasome activation is impaired in NLRP3-K24R knock-in mice. We identify KAT5 as an acetyltransferase able to acetylate NLRP3. KAT5 deficiency in myeloid cells and pharmacological inhibition of KAT5 enzymatic activity reduce activation of the NLRP3 inflammasome, both in vitro and in vivo. Thus, our study reveals a key mechanism for the oligomerization and full activation of NLRP3 and lays down the proof of principle for therapeutic targeting of the KAT5-NLRP3 axis.

Abstract The reconstruction models of hourly 10‐m wind speeds were developed for each of 2384 stations over China using stepwise regression, random forest and XGBoost machine learning approaches based on hourly observed and ERA5 reanalysis data from 2005 to 2021. The reconstruction procedures applied observed hourly data to reduce the systematic biases of reanalysis data sets. Furthermore, the procedures employed the past dynamically consistent states of the atmosphere simulated by ERA5 reanalysis techniques to reduce/remove wind‐observed data impacts of long‐term non‐meteorological condition changes over time (e.g., urbanization around weather stations), which provide homogenous hourly wind speed data sets from 1959 to 2021. The systematic errors of the models' simulations derived from three approaches are similarly small, with almost two orders of magnitude smaller than ERA5 original data sets. The systematic errors of reconstructed data sets derived from stepwise regression are similar to its simulation; however, the biases from two machine learning methods are even much greater than ERA5 original data sets. This result implies that machine learning methods are not suitable for such typical time‐series predictions using the previous‐hour wind speed as a predictor to reconstruct wind speed data for the next hour. Therefore, stepwise regression was selected to reconstruct hourly wind speed data sets, which have much better quality than ERA5 reanalysis data with the median correctness increased by >50% and the median rRMSE decreased by 25%–50%. Consequently, the reconstructed wind speed data sets have great potential to be useful for more precisely assessing the characteristics/trends of wind energy resources in the past 60 years over China.

Conventional spherical nucleic acid enzymes (SNAzymes), made with gold nanoparticle (AuNPs) cores and DNA shells, are widely applied in bioanalysis owing to their excellent physicochemical properties. Albeit important, the crowded catalytic units (such as G-quadruplex, G4) on the limited AuNPs surface inevitably influence their catalytic activities. Herin, a hybridization chain reaction (HCR) is employed as a means to expand the quantity and spaces of G4 enzymes for their catalytic ability enhancement. Through systematic investigations, we found that when an incomplete G4 sequence was linked at the sticky ends of the hairpins with split modes (3:1 and 2:2), this would significantly decrease the HCR hybridization capability due to increased steric hindrance. In contrast, the HCR hybridization capability was remarkably enhanced after the complete G4 sequence was directly modified at the non-sticky end of the hairpins, ascribed to the steric hindrance avoided. Accordingly, the improved SNAzymes using HCR were applied for the determination of AFB1 in food samples as a proof-of-concept, which exhibited outstanding performance (detection limit, 0.08 ng/mL). Importantly, our strategy provided a new insight for the catalytic activity improvement in SNAzymes using G4 as a signaling molecule.

Tetracycline (TC) is widely used in the livestock industry, but undigested TC is excreted with livestock waste and accumulates in the environment. In this study, swine manure-derived biochar (SBC) was modified with KMnO4 (MnOx-SBC), and used to remove TC. SEM-EDS, FTIR, XPS and elemental analysis all indicated that ultrafine MnOx particles were attached to the biochar surface. The surface properties and composition of the oxygen-containing functional groups were enhanced by KMnO4 modification. Batch sorption experiments showed that MnOx-SBC's TC-adsorption capacity was 105.9 mg·g−1, 46.4% higher than SBC's. The TC-adsorption onto MnOx-SBC agreed well with the pseudo-second-order model and Freundlich isotherm. A new platform is proposed for reusing swine manure while solving the livestock industry's antibiotic pollution risk by ‘treating waste with waste’. HIGHLIGHTS TC adsorption capacity of swine manure derived biochar enhanced after KMnO4 modification.; Initial solution pH, temperature and initial solution concentration affected TC removal.; Pseudo-second-order and Freundlich models can well fit the adsorption behavior.; Adsorption mechanism included pore-filling, surface complexation, π-π interactions and H-bonding.;

IntroductionChimeric antigen receptor (CAR) T cell therapy has achieved unprecedented efficacy recently. However, the factors related to responses and durable remission are elusive. This study was to investigate the impact of pre-lymphodepletion (pre-LD) absolute lymphocyte count (ALC) on CAR T cell therapy outcomes.MethodsWe conducted a retrospective study of 84 patients with relapsed/refractory diffuse large B-cell lymphoma (R/R DLBCL) who underwent CAR T cell treatment at the Affiliated Hospital of Xuzhou Medical University between March 1,2016 and December 31, 2021. The enrolled patients were divided into high group and low group according to the optimal cutoff value of pre-LD ALC. The Kaplan-Meier analyses was used to calculate survival curves. The Cox proportional hazards model was used for univariate and multivariate analysis to assess the prognostic factors.ResultsThe ROC showed that the optimal cutoff value of pre-LD ALC was 1.05 x 109/L. The overall response (defined as partial response or complete response) rate was significantly higher in patients with a high pre-LD ALC (75% versus 52.08%; P=0.032). Patients with a low pre-LD ALC had significantly inferior overall survival (OS) and progression-free survival (PFS) compared with those having a high pre-LD ALC (median OS, 9.6 months versus 45.17 months [P=0.008]; median PFS, 4.07 months versus 45.17 months [P= 0.030]). Meanwhile, low pre-LD ALC is an independent risk factor for PFS and OS.DiscussionThe data suggested that pre-LD ALC may serve as a helpful indicator to predict the outcomes of CAR T cell therapy in patients with R/R DLBCL.

Green and low cost CO2 and CaO were used to stimulate amine emulsions to reveal the responsive behavior of amine emulsions. On this basis, oil-based drilling fluids responsive to CO2 and CaO were formulated and their properties were evaluated. The results showed that the amine emulsions inversed from water-in-oil state to oil-in-water state readily and their rheological behavior underwent transitions of decreasing, rising again and decreasing again via induction by CO2. These CO2 responsive behaviors could be reversed by CaO. Oil-based drilling fluids prepared based on the amine emulsions with oil-water volume ratios of 50:50 to 70:30, densities of 1.4–2.0 g/cm3 had good rheological and filtration properties at 160 °C; and be readily cleaned up using CO2 bubbling. The useless solid phase with low density could be removed efficiently via reducing the viscosity of emulsion by CO2 and the residual liquid phase could be restored to the original state by CaO and reused to prepare drilling fluid. The mechanisms analysis indicated that CO2/CaO induced the reversible conversion between amine emulsifiers and their salts, which enabled the reversible regulation of both the hydrophilic-lipophilic balance of amine emulsifiers and the emulsion particles' size and finally caused the controllable-reversion of the form and rheology of amine emulsion.

Molybdenum disulfide nanomaterials (MoS2 NMs) have shown significant role as photocatalysts, lubricating agents and sterilant due to their remarkable physicochemical properties. Because of the increasing demand for MoS2 NMs in numerous industrial domains, greater occupational exposure and subsequent NMs release into environment would be unavoidable. However, much efforts have been made to uncover the biological effects of NMs at unrealistic high concentration or acute duration, placing constraints on setting the realistic occupational exposure thresholds with confidence. In order to fill the current knowledge gap, this study aimed to evaluate the nanotoxicity of MoS2 NMs with or without surface defects under the more realistic exposure mode. Noteworthily, the artificial sweat transformed-occupational exposure-cytotoxicity investigation of MoS2 NMs was established as the main studied line. And the high cellular internalization and augmented oxidative stress triggered by surface defect could be recognized as the main factors for triggering serious cellular damage. Moreover, the HaCaTs exhibited loss of cell membrane integrity, dysfunction of mitochondria, disorder of endoplasmic reticulum and damages of nuclei after chronic exposure, compared with acute exposure. The study provided closely realistic exposure scenarios for NMs which exhibited significant difference from acute toxic investigation, enriching understanding towards real environmental safety of NMs.

Volatile organic compounds (VOCs), particularly monoaromatic hydrocarbon compounds (MACHs), pose a potential risk to the atmospheric environment and human health. Therefore, the progressive development of efficient detection methodologies is a pertinent need, which is still a challenge at present. In this study, we present a rapid and sensitive method to detect trace amounts of MACHs using a bifunctional SERS composite substrate. We prepared an Au/SiO2 enhanced layer and a porous Cu(OH)2 adsorption layer via microfluidic-assisted gas-liquid interface self-assembly. The composite substrate effectively monitored changes in benzaldehyde using time-varying SERS spectra, and track-specifically identified various VOCs such as benzene, xylene, styrene, and nitrobenzene. In general, the substrate exhibited a rapid response time of 20 s to gaseous benzaldehyde, with a minimum detection concentration of less than 500 ppt. Further experimental assessments revealed an optimum Cu(OH)2 thickness of the surrounding adsorption layer of 150 nm, which can achieve an efficient SERS response to MACHs. Furthermore, the recoverable and reusable property of the composite substrate highlights its practicality. This study presents a straightforward and efficient approach for detecting trace gaseous VOCs using SERS, with significant implications in the designing of SERS substrates for detecting other VOCs.

BackgroundThere is an increasing interest in preoperative strength training for promoting post-operative rehabilitation, but the effectiveness of preoperative strength training for clinical outcomes after total knee arthroplasty (TKA) remains controversial.ObjectiveThis study aims to systematically evaluate the effect of preoperative strength training on clinical outcomes before and after TKA.MethodsWe systematically searched PubMed, Cochrane Library, Web of Science, and EMBASE databases from the inception to November 17, 2021. The meta-analysis was performed to evaluate the effects of preoperative strength training on clinical outcomes before and after TKA.ResultsSeven randomized controlled trials (RCTs) were included (n = 306). Immediately before TKA, the pooled results showed significant improvements in pain, knee function, functional ability, stiffness, and physical function in the strength training group compared with the control group, but not in strength (quadriceps), ROM, and WOMAC (total). Compared with the control group, the results indicated strength training had a statistically significant improvement in post-operative knee function, ROM, and functional ability at less than 1 month and 3 months, and had a statistically significant improvement in post-operative strength (quadriceps), stiffness, and WOMAC (total) at 3 months, and had a statistically significant improvement in post-operative pain at 6 months. However, the results indicated strength training had no statistically significant improvement in post-operative strength (quadriceps) at less than 1 month, 6, and 12 months, had no statistically significant improvement in post-operative pain at less than 1 month, 3, and 12 months, had no statistically significant improvement in post-operative knee function at 6 and 12 months, and had no statistically significant improvement in post-operative physical function at 3 months.ConclusionsPreoperative strength training may be beneficial to early rehabilitation after TKA, but the long-term efficacy needs to be further determined. At the same time, more caution should be exercised when interpreting the clinical efficacy of preoperative strength training for TKA.

Biochar is a carbon-rich stable substance, defined as charred organic matter, produced during biomass thermochemical decomposition, and its application is currently considered as a mean of enhancing soil productivity, which is an important requirement for increasing crop yields whereas, simultaneously, it improves the quality of contaminated soil and water. However, depending on pedoclimatic conditions, its applicability exhibits negative aspects as well. It can also support biofuel production, therefore helping in reducing the demand for fossil fuels. Biochar is providing ecosystem services such as immobilization and transformation of contaminants and mitigation of climate change by sequestering carbon and reducing the release of greenhouse gases such as nitrous oxide and methane. It can further reduce waste as it could be produced from everything that contains biomass thereby assisting in waste management. Due to such wide-ranging applications, this review was conceptualized to emphasize the importance of biochar as an alternative to classic products used for energy, environmental and agricultural purposes. Based on the detailed information on the factors impacting biochar properties, the benefits and limitations of biochar, and the potential application guidelines for growers, this work aimed to help in partial achievement of multiple environmental goals and a practical recommendation to growers although its large-scale application is still controversial.

Abstract Background Overactive bladder is defined as “urgency, with or without urge incontinence, usually with frequency and nocturia”. Electroacupuncture may be a safe and an effective alternative therapy for overactive bladder, but the evidence is limited. Methods We will conduct a three-arm, non-inferiority, multi-centre randomized controlled clinical trial. A total of 420 patients with moderate and severe overactive bladder will be randomly assigned to one of three groups: the electroacupuncture group (N = 140), sham electroacupuncture group (N = 140), and solifenacin group (N = 140). The primary outcome will be the change in the overactive bladder symptom score from baseline to the end of the 12-week treatment. The secondary outcomes will include the proportion of participants with a decrease in the overactive bladder symptom score ≥ 3 at weeks 4, 8, 12, 20, and 32; the change in average 24 h values of urination, nocturnal urination, urgency incontinence and urgency episodes from baseline to weeks 4, 8, 12, 20 and 32, and so forth. The adverse events will be recorded. Statistical analysis will include covariance analysis, nonparametric tests and descriptive statistics. Discussion This study will answer the question of whether electroacupuncture is effective and non-inferior to solifenacin for improving the symptoms of overactive bladder patients. Trial registration Chinese clinical trial registry ( ChiCTR1800019928 ).

Abstract This study was designed to evaluate ERK5 expression in lung cancer and malignant melanoma progression and to ascertain the involvement of ERK5 signaling in lung cancer and melanoma. We show that ERK5 expression is abundant in human lung cancer samples, and elevated ERK5 expression in lung cancer was linked to the acquisition of increased metastatic and invasive potential. Importantly, we observed a significant correlation between ERK5 activity and FAK expression and its phosphorylation at the Ser910 site. Mechanistically, ERK5 increased the expression of the transcription factor USF1, which could transcriptionally upregulate FAK expression, resulting in FAK signaling activation to promote cell migration. We also provided evidence that the phosphorylation of FAK at Ser910 was due to ERK5 but not ERK1/2, and we then suggested a role for Ser910 in the control of cell motility. In addition, ERK5 had targets in addition to FAK that regulate epithelial-to-mesenchymal transition and cell motility in cancer cells. Taken together, our findings uncover a cancer metastasis-promoting role for ERK5 and provide the rationale for targeting ERK5 as a potential therapeutic approach.

Background: Clinical incidences of chronic low back pain among the elderly are increasing. However, studies have not fully elucidated on changes in biomechanical properties of paravertebral muscles in patients with unilateral chronic low back pain. We evaluated the changes in biomechanical properties of painful and non-painful paravertebral muscles in elderly patients with unilateral chronic low back pain.Methods: Biomechanical properties of paravertebral muscles, including muscle tone and stiffness, in elderly patients with unilateral chronic low back pain were measured using MyotonPRO. Lumbar Lordosis and Sacral Slope were measured by magnetic resonance imaging. Cross-sectional areas of paravertebral muscles were evaluated using ImageJ software version 1.53. Chronic low back pain severity was assessed by Visual Analogue Scale (VAS) and Oswestry Disability Index (ODI) scores. The correlations between VAS scores, ODI scores, Lumbar Lordosis, Sacral Slope, cross-sectional areas (painful side), disease duration, and biomechanical properties of paravertebral muscles in the painful side were analyzed.Results: A total of 60 elderly patients with unilateral chronic low back pain were enrolled in this study. The muscle tone and stiffness of paravertebral muscles on the painful side were significantly higher than those on the non-painful side (p < .05). Cross-sectional areas of paravertebral muscles on the painful side at the L3 level were smaller than those of the non-painful side (p < .05). The VAS scores and ODI scores were significantly positively correlated with muscle tone and stiffness of paravertebral muscles on the painful side (p < .05 and p < .01, respectively). There were no significant correlations between disease duration, cross-sectional areas (painful side), Lumbar Lordosis, or Sacral Slope and muscle tone and stiffness of paravertebral muscles on the painful side (p > .05).Conclusion: In elderly patients with unilateral chronic low back pain, muscle tone and stiffness of paravertebral muscles on the painful side are higher than for those on the non-painful side. The asymmetry of biomechanical properties of paravertebral muscles is associated with severity of chronic low back pain.

Abstract Land application of biochar as a soil amendment can be an effective strategy for reducing nutrient leaching while providing simultaneous benefits to soil health. However, biochar effects on soil nitrogen (N) and phosphorus (P) dynamics in biochar‐amended soils are poorly understood, particularly in Florida sandy soils receiving fertilizers. This study evaluated the effects of different biochar materials (particle size of 0.1 mm produced from pine (Pinus spp.) wood vs. ∼0.5–2 mm produced from fescue (Festuca arundinaceaSchreb.) grass, herein referred as “powder” and “coarse” biochar, respectively) and their application methods (surface vs. incorporated) on N and P leaching from soils amended with inorganic fertilizer or biosolids. Biochar materials were either mixed throughout columns of soil or surface applied at a 1% (w/w) rate. Columns were periodically leached over 6 mo to attain 138 ml (equivalent to ∼1 pore volume) of drainage in each event (total of ∼16 pore volume of leachate). Biochar materials exhibited modest affinity to retain ammonium (NH4+) but showed no ability to sorb nitrate (NO3–). Powder biochar incorporated throughout the soil reduced cumulative NH4–N, NO3–N, and total N mass leached (average of 25% reduction) relative to control (no biochar) treatments. Biochar materials acted as P sources and were ineffective in reducing leachate P. Biosolids generally resulted in significant lower risk of N and P leaching than inorganic fertilizer. Results suggested biochar ability to retain N and P in soils varies with biochar source, application method, and nutrient source. Biochar type and application method affected N and P dynamics in biosolids or inorganic fertilizer‐amended soils, but understanding the mechanisms requires additional study.

Volatile organic compounds (VOCs) in indoor environments have typical features of multiple components, high concentration, and long duration. The development of gas sensors with high sensitivity to multiple VOCs is of great significance to protect human health. Herein, we proposed a sensitive ZnO/WO3 composite chemi-resistive sensor facilely fabricated via a sacrificial template approach. Based on the transferable properties of self-assembled monolayer colloidal crystal (MCC) templates, two-dimensional honeycomb-like ordered porous ZnO/WO3 sensing matrixes were constructed in situ on commercial ceramic tube substrates with curved and rough surfaces. The nanocomposite thin films are about 250 nm in thickness with large-scale structural consistency and integrity, which facilitates characteristic responses with highly sensitivity and reliability. Furthermore, the nanocomposite sensor shows simultaneous responses to multiple VOCs that commonly exist in daily life with an obvious suppression sensing for traditional flammable gases. Particularly, a detection limit of 0.1 ppm with a second-level response/recovery time can be achieved, which is beneficial for real-time air quality assessments. We proposed a heterojunction-induced sensing enhancement mechanism for the ZnO/WO3 nanocomposite film in which the formation of abundant heterojunctions between ZnO and WO3 NPs significantly increases the thickness of the electron depletion layer in the bulk film and improves the formation of active oxygen species on the surface, which is conducive to enhanced responses for reducing VOC gases. This work not only provides a simple approach for the fabrication of high-performance gas sensors but also opens an achievable avenue for air quality assessment based on VOC concentration detection.

In past years, lithium-ion batteries (LIBs) can be found in every aspect of life, and batteries, as energy storage systems (ESSs), need to offer electric vehicles (EVs) more competition to be accepted in markets for automobiles. Thick electrode design can reduce the use of non-active materials in batteries to improve the energy density of the batteries and reduce the cost of the batteries. However, thick electrodes are limited by their weak mechanical stability and poor electrochemical performance; these limitations could be classified as the critical cracking thickness (CCT) and the limited penetration depth (LPD). The understanding of the CCT and the LPD have been proposed and the recent works on breaking the CCT and improving the LPD are listed in this article. By comprising these attempts, some thick electrodes could not offer higher mass loading or higher accessible areal capacity that would defeat the purpose.

This study investigates the impact of assimilating conventional weather observations on the wind forecast over the nearshore region of the East China Sea. Multi-level wind measurements in the boundary layer from five masts near the coast were used to verify the numerical model forecasts. Four numerical experiments with a rapid update cycle were performed to forecast the wind field over the masts. The observation shows that the characteristics of the wind field are distinct between the onshore and offshore masts. The numerical forecasts were able to reproduce the main features of the observed wind field both onshore and offshore. However, the wind forecasts of the offshore masts showed larger BIAS and MAE than those onshore. The forecast skill was shown to be sensitive to different weather events and the choice of control variables in the assimilation. The use of new momentum control variables allows a smaller observation-minus-analysis field compared with the traditional control variables, and the resultant wind forecast showed significant improvements. Further tuning of the new control variable scheme showed little improvement of the wind forecast which demonstrates the importance of maintaining the balance between large-scale and small-scale fields in the analysis. The larger forecast error at the offshore masts was likely due to the distribution of conventional observations and the uncertainties in representing the marine boundary layer in numerical models.

Adenylate kinase 2 (AK2) is a wide-spread and highly conserved protein kinase whose main function is to catalyze the exchange of nucleotide phosphate groups. In this study, we showed that AK2 regulated tumor cell metastasis in lung adenocarcinoma. Positive expression of AK2 is related to lung adenocarcinoma progression and poor survival of patients. Knockdown or knockout of AK2 inhibited, while overexpression of AK2 promoted, human lung adenocarcinoma cell migration and invasion ability. Differential proteomics results showed that AK2 might be closely related to epithelial-mesenchymal transition (EMT). Further research indicated that AK2 regulated EMT occurrence through the Smad-dependent classical signaling pathways as measured by western blot and qPCR assays. Additionally, in vivo experiments showed that AK2-knockout in human lung tumor cells reduced their EMT-like features and formed fewer metastatic nodules both in liver and in lung tissues. In conclusion, we uncover a cancer metastasis-promoting role for AK2 and provide a rationale for targeting AK2 as a potential therapeutic approach for lung cancer.

An increasing number of inflammatory pathologies is associated with IL-1 production downstream of caspases 1 and 11. Here the authors show that graft-versus-host-disease (GvHD) is diminished in mice with genetic or pharmacological ablation of caspase-11, and provide mechanistic insights into the signals leading to caspase-11 activation in GvHD.

The multireceptor tyrosine kinase inhibitor sorafenib is a Food and Drug Administration-approved first-line drug for the treatment of advanced liver cancer that can reportedly extend overall survival in patients with advanced hepatocellular carcinoma (HCC). Primary and acquired resistance to sorafenib are gradually increasing however, leading to failure of HCC treatment with sorafenib. It is therefore crucial to study the potential mechanism of sorafenib resistance. The results of the current study indicate that neurite outgrowth inhibitor protein B receptor (NgBR) is overexpressed in cultured sorafenib-resistant cells, and that its expression is negatively correlated with the sensitivity of liver cancer cells to sorafenib. Artesunate can inhibit the expression of NgBR, and it may block sorafenib resistance. Herein we report that sorafenib treatment in combination with artesunate overcomes HCC resistance to sorafenib alone in a cell culture model.

The liver fluke Clonorchis sinensis inhabits the bile ducts, where bile concentration disparities across the fluke cell membrane can cause bile intoxication. Sodium-bile acid co-transporter (SBAT) plays a crucial role in bile acid recycling. The process by which SBAT imports bile acids is electrically coupled to sodium ion co-transportation. Here, we report that the SBAT of C. sinensis (CsSBAT) is involved in bile acid transportation. CsSBAT cDNA encoded a putative polypeptide of 546 amino acid residues. Furthermore, CsSBAT consisted of ten putative transmembrane domains, and its 3D structure was predicted to form panel and core domains. The CsSBAT had one bile acid- and three Na+-binding sites, enabling coordination of a symport process. CsSBAT was mainly localized in the mesenchymal tissue throughout the fluke body and sparsely localized in the basement of the tegument, intestinal epithelium, and excretory bladder wall. Bile acid permeated into the adult flukes in a short time and remained at a low concentration level. Bile acid accumulated inside the mesenchymal tissue when CsSBAT was inhibited using polyacrylic acid-tetradeoxycholic acid conjugate. The accumulated bile acid deteriorated the C. sinensis adults leading to death. CsSBAT silencing shortened the lifespan of the fluke when it was placed into bile. Taken together, we propose that CsSBAT transports bile acids in the mesenchymal tissue and coordinate with outward transporters to maintain bile acid homeostasis of C. sinensis adults, contributing to C. sinensis survival in the bile environment.

The sustainable development of the marine economic system to accurately grasp the dynamic evolution characteristics and main driving factors of the vulnerability of the marine economic system so as to effectively identify the problems in the operation of the marine economy is extremely important. Based on the establishment of a three-level assessment index system for the vulnerability of the marine economic system, the vulnerability of the marine economic system of Jiangsu Province in 2008–2016 was evaluated by using the comprehensive index model and the catastrophe progression model, and the results were compared based on the unified evaluation criteria. The main influencing factors were analyzed by principal component analysis and Pearson correlation analysis. It’s found that 2014, 2015, and 2012 are in a general state, 2009 and 2010 are in a poor state, and the vulnerability state from 2008 to 2011 is lower than that in 2012. The overall volatility is small indicating a stable system. Among them, the scale of marine industry and profit and loss of marine resources are the main components of marine economic system vulnerability having the highest correlation with the vulnerability of marine economic system. This study enriches the theoretical method and empirical research on the vulnerability of the marine economic system and has practical significance in allowing for timely targeted control measures to affect the vulnerability of the marine economic system.

Background: Aspirin has recently emerged as an anticancer drug, but its therapeutic effect on lung cancer has been rarely reported, and the mechanism of action is still unclear. Long-term use of celecoxib in large doses causes serious side effects, and it is necessary to explore better ways to achieve curative effects. In this study, we evaluated the synergistic anticancer effects of celecoxib and aspirin in non-small cell lung cancer (NSCLC) cells. Methods: In vitro , we evaluated the combined effects of celecoxib (40 μM) and aspirin (8 mM) on cell apoptosis, cell cycle distribution, cell proliferation, cell migration and signaling pathways. Furthermore, the effect of aspirin (100 mg/kg body weight) and celecoxib (50 mg/kg body weight) on the growth of xenograft tumors was explored in vivo . Results: Our data suggest that cancer sensitivity to combined therapy using low concentrations of celecoxib and aspirin was higher than that of celecoxib or aspirin alone. Further research showed that the anti-tumor effect of celecoxib combined with aspirin was mainly produced by activating caspase-9/caspase-3, arresting cell cycle and inhibiting the ERK-MAPK signaling pathway. In addition, celecoxib alone or in combination with aspirin inhibited the migration and invasion of NSCLC cells by inhibiting MMP-9 and MMP-2 activity levels. Moreover, we identified GRP78 as a target protein of aspirin in NSCLC cells. Aspirin induced an endoplasmic reticulum stress response by inhibiting GRP78 activity. Furthermore, combination therapy also exhibited a better inhibitory effect on tumor growth in vivo . Conclusions: Our study provides a rationale for further detailed preclinical and potential clinical studies of the combination of celecoxib and aspirin for NSCLC therapy.

Celecoxib has potential as an effective antineoplastic agent, but it may exhibit side effects. Given the glucose-addicted properties of tumor cells, metformin is recognized for its inhibitory effect on oxidative phosphorylation. In the present study, we aimed to combine low dose of celecoxib with metformin to alleviate the side effects of nonsteroidal anti-inflammatory drugs (NSAIDs) and overcome potential drug resistance. We found that celecoxib combined with metformin obviously suppressed cell migration and proliferation and induced cell apoptosis. Most importantly, in vivo experiments revealed the superior antitumor efficacy of combination treatment with a low dosage of celecoxib (25 mg/kg/day) without apparent toxicity. Further study of the underlying mechanism revealed that the two drugs in combination caused ROS aggregation in NSCLC cells, leading to DNA double-strand breaks and increased expression of the tumor suppressor factor p53. Elevated p53 subsequently caused cell cycle arrest and cell proliferation inhibition. The presence of metformin also sensitized NSCLC cells to celecoxib-induced apoptosis by activating caspase-9, -8, -3, and -7, upregulating the pro-apoptotic proteins Bad and Bax, and downregulating the antiapoptotic proteins Bcl-xl and Bcl-2. Moreover, the superior anticancer effect of combined therapy was also due to suppression of Raf-MEK-ERK cascades and PI3K-AKT signaling, which is conducive to overcoming drug resistance. In addition, either celecoxib alone or in combination with metformin suppressed NSCLC cell migration and invasion by inhibiting FAK, N-cadherin, and matrix metalloproteinase-9 activities. Together, our study provided a rational combination strategy with a low dosage of celecoxib and metformin for preclinical cancer application.

The interactive effects of straw-derived biochar and bio-based polymer-coated urea (BPCU) was examined with a pot experiment conducted in 2014 and 2015. Using a split-plot design, the main plot factor was the form of straw use and the sub-plot factor was the type of N fertilizer. The soil inorganic nitrogen (N), organic carbon and lint yield of biochar treatments were significantly higher than for straw treatments. Meanwhile, the BPCU treatments enhanced nitrogen use efficiency (NUE) and yield over urea treatments. Biochar combined with BPCU resulted in the highest lint yield, 14.3–108.2% increasing over the other treatments, with NUE 27.1–63.5% increased. We attributed this superior performance to the interactive effects between BPCU’s controlled supply of N according to cotton’s N requirements and biochar’s functionalities in enhancing soil quality. Thus, the application of biochar and BPCU is a sustainable strategy to improve soil quality and increase cotton yield.

Introduction: A novel analytical method using fast gas chromatography combined with surface acoustic wave sensor (GC-SAW) was developed for rapid determination of the pharmacological volatiles of turmeric (Curcuma longa L.). Methods: The volatile compounds in 20 turmeric samples, collected from different parts and different origins, were assessed by the fast GC-SAW. In addition, gas chromatography–mass spectrometry (GC-MS) was employed to confirm the chemical composition of the main volatiles. The digital fingerprint of turmeric was established and analysed by principal component analysis and cluster analysis. Results: Curcumene (9.1%), β-sesquiphellandrene (5.1%) and ar-turmerone (69.63%) were confirmed as the main pharmacological volatiles of turmeric. The content of ar-turmerone in lateral rhizome turmeric was significantly higher than that of top rhizome and ungrouped turmeric. The contents of curcumene and β-sesquiphellandrene in top rhizome turmeric were higher than those in lateral and ungrouped turmeric. The 20 turmeric samples were divided into four categories, which reflected the quality characteristics of the turmeric from different parts and origins. Conclusion: The GC-SAW method can rapidly and accurately detect pharmacologically volatiles of turmeric, and it can be used in the quality control of turmeric.

Abstract Background Multidrug resistance-associated protein 4 (MRP4) is a member of the C subfamily of the ABC family of ATP-binding cassette (ABC) transporters. MRP4 regulates ATP-dependent efflux of various organic anionic substrates and bile acids out of cells. Since Clonorchis sinensis lives in host’s bile duct, accumulation of bile juice can be toxic to the worm’s tissues and cells. Therefore, C. sinensis needs bile transporters to reduce accumulation of bile acids within its body. Results We cloned MRP4 (CsMRP4) from C. sinensis and obtained a cDNA encoding an open reading frame of 1469 amino acids. Phylogenetic analysis revealed that CsMRP4 belonged to the MRP/SUR/CFTR subfamily. A tertiary structure of CsMRP4 was generated by homology modeling based on multiple structures of MRP1 and P-glycoprotein. CsMRP4 had two membrane-spanning domains (MSD1 & 2) and two nucleotide-binding domains (NBD1 & 2) as common structural folds. Docking simulation with nine bile acids showed that CsMRP4 transports bile acids through the inner cavity. Moreover, it was found that CsMRP4 mRNA was more abundant in the metacercariae than in the adults. Mouse immune serum, generated against the CsMRP4-NBD1 (24.9 kDa) fragment, localized CsMRP4 mainly in mesenchymal tissues and oral and ventral suckers of the metacercariae and the adults. Conclusions Our findings shed new light on MRPs and their homologs and provide a platform for further structural and functional investigations on the bile transporters and parasites’ survival.

The reliability of MyotonPRO that can monitor the mechanical properties of tissues is still unclear. This study aimed to analyze the within-day inter-operator and between-day intra-operator reliability of MyotonPRO for assessing tone and stiffness of quadriceps femoris and patellar tendon at different knee angles. The tone and stiffness of healthy participants (15 males and 15 females, aged 24.7±1.6 years) in the supine and resting position were measured using the MyotonPRO device. The measurements were quantified at 0°, 30°, 60°, and 90° of knee flexion. The intraclass correlation coefficient (ICC), standard error of measurement (SEM), and minimal detectable change (MDC) were calculated and a Bland-Altman analysis was conducted to estimate reliability. The results indicated excellent inter-operator reliability (ICC > 0.78) and good to excellent intra-operator reliability (ICC > 0.41). The inter-operator SEM measurements ranged between 0.1-0.9 Hz and 3.8-37.9 N/m, and intra-operator SEM ranged between 0.5-1.3 Hz and 7.9-52.0 N/m. The inter-operator MDC ranged between 0.3-2.5 Hz and 10.5-105.1 N/m, and intra-operator SEM ranged between 1.1-3.3 Hz and 21.9-144.1 N/m. The agreement of inter-operator was better than that of intra-operator. The study concluded that MyotonPRO is a reliable device to detect the tone and stiffness of quadriceps femoris and patellar tendon.

Clonorchis sinensis is a liver fluke that can dwell in the bile ducts of mammals. Bile acid transporters function to maintain the homeostasis of bile acids in C. sinensis, as they induce physiological changes or have harmful effects on C. sinensis survival. The organic solute transporter (OST) transports mainly bile acid and belongs to the SLC51 subfamily of solute carrier transporters. OST plays a critical role in the recirculation of bile acids in higher animals. In this study, we cloned full-length cDNA of the 480-amino acid OST from C. sinensis (CsOST). Genomic analysis revealed 11 exons and nine introns. The CsOST protein had a 'Solute_trans_a' domain with 67% homology to Schistosoma japonicum OST. For further analysis, the CsOST protein sequence was split into the ordered domain (CsOST-N) at the N-terminus and disordered domain (CsOST-C) at the C-terminus. The tertiary structure of each domain was built using a threading-based method and determined by manual comparison. In a phylogenetic tree, the CsOST-N domain belonged to the OSTα and CsOST-C to the OSTβ clade. These two domains were more highly conserved with the OST α- and β-subunits at the structure level than at sequence level. These findings suggested that CsOST comprised the OST α- and β-subunits. CsOST was localized in the oral and ventral suckers and in the mesenchymal tissues abundant around the intestine, vitelline glands, uterus, and testes. This study provides fundamental data for the further understanding of homologues in other flukes.

The utilization of advanced remote sensing methods to monitor the coastal wetlands is essential for conservation and sustainable development. With multiple polarimetric channels, the polarimetric synthetic aperture radar (PolSAR) is increasingly employed in land cover classification and information extraction, as it has more scattering information than regular SAR images. Polarimetric decomposition is often used to extract scattering information from polarimetric SAR. However, distinguishing all land cover types using only one polarimetric decomposition in complex ecological environments such as coastal wetlands is not easy, and thus integration of multiple decomposition algorithms is an effective means of land cover classification. More than 20 decompositions were used in this research to extract polarimetric scattering features. Furthermore, a new algorithm combining random forest (RF) with sequential forward selection (SFS) was applied, in which the importance values of all polarimetric features can be evaluated quantitatively, and the polarimetric feature set can be optimized. The experiments were conducted in the Jiangsu coastal wetlands, which are located in eastern China. This research demonstrated that the classification accuracies were improved relative to regular decision tree methods, and the process of polarimetric scattering feature set optimization was intuitive. Furthermore, the scattering matrix elements and scattering features derived from H / α , Yamaguchi3, VanZyl3, and Krogager decompositions were determined to be very supportive of land cover identification in the Jiangsu coastal wetlands.

A theoretical model is established to describe the response mechanism of surface acoustic wave (SAW) gas sensors based on physical adsorption on the detector surface. Wohljent’s method is utilized to describe the relationship of sensor output (frequency shift of SAW oscillator) and the mass loaded on the detector surface. The Brunauer-Emmett-Teller (BET) formula and its improved form are introduced to depict the adsorption behavior of gas on the detector surface. By combining the two methods, we obtain a theoretical model for the response mechanism of SAW gas sensors. By using a commercial SAW gas chromatography (GC) analyzer, an experiment is performed to measure the frequency shifts caused by different concentration of dimethyl methylphosphonate (DMMP). The parameters in the model are given by fitting the experimental results and the theoretical curve agrees well with the experimental data.

To better understand the regulatory mechanism of phenolics and ascorbic acid accumulation as well as antioxidant activities in mung beans during legume development, the gene expression profiles of 25 key-coding genes in ascorbic acid and phenolics metabolic pathways were analyzed. As well as the dynamitic changes of ascorbic acid, phenolic profiles and antioxidant activities with legume development were studied. The results indicated that gene expression profiles were closely related to the ascorbic acid and phenolics accumulation regularity during legume development. VrVTC2 and VrGME played important roles for ascorbic acid accumulation from 8 to 17 days after flowering (DAF). VrPAL and VrCHS exhibited positive correlations with daidzein and glycitin accumulation, and VrIFS had a strong positive correlation with glycitin biosynthesis. Antioxidant activities dramatically increased during mung bean maturing, which were significantly related to ascorbic acid and phenolics accumulation. Eight days after flowering was the essential stage for ascorbic acid and phenolics biosynthesis in mung beans.

To investigate if the role of Cav-1 in GDM-induced macrosomia is through regulating AMPK signaling pathway in placenta.We used diagnostic criteria of gestational diabetes mellitus (GDM) and macrosomia to separate and compare placental protein and mRNA levels from GDM with macrosomia group (GDMM), GDM with normal birth weight group (GDMN) and normal glucose tolerance (NGT) with normal birth weight group (CON). Western blotting was performed to examine differentially expressed proteins of caveolin-1 (Cav-1) and Adenosine monophosphate-activated protein kinase (AMPK) signaling pathway related proteins, including phosphorylated-AMPKα(Thr172), AMPKα, phosphorylated-Acetyl-CoA carboxylase(Ser79) (p-ACC(Ser79)), ACC and glucose transporter 1 (GLUT1) in placenta between the three groups. The mRNA levels of Cav-1, AMPKα, ACC and GLUT1 in placenta were measured by real time-PCR.In the GDMM placenta group, both protein and mRNA levels of Cav-1 were down-regulated, while GLUT1 was up-regulated; the phosphorylation and mRNA levels of ACC and AMPKα were decreased, but total ACC protein levels were increased compared to both the GDMN (p
