Abstract

The problem of the possibility of stimulating a sense of ownership by such a form of socio-cultural impact as cinema is posed. The theoretical foundations of the hypothesis about the possibility of such stimulation are given. In particular, the main aspects of the emotional content of the horror film: victim; aggressor; quasi-messianic idea of the aggressor — largely correspond to the aspects of the sense of ownership: thrift; expansion; self-confidence. The results of a study in which a horror film changed the attitude to the category “Mine” are presented. Ставится проблема возможности стимулирования чувства собственности таким социально-культурным фактором, как кино. Приводятся теоретические основания гипотезы о возможности такого стимулирования. В частности, состав эмоционально значимых образов фильма ужасов: жертва; агрессор; квази-мессианская (псевдоморальная) идея агрессора — подобны аспектам чувства собственности: бережливость; экспансия; самоуверенность. Приведены результаты исследования, в котором фильм ужасов изменял отношение к категории «Мое».

Abstract

The global coronavirus disease 2019 (COVID-19) pandemic has mobilized efforts to develop vaccines and antibody-based therapeutics, including convalescent-phase plasma therapy, that inhibit viral entry by inducing or transferring neutralizing antibodies (nAbs) against the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) spike glycoprotein (CoV2-S). However, rigorous efficacy testing requires extensive screening with live virus under onerous biosafety level 3 (BSL3) conditions, which limits high-throughput screening of patient and vaccine sera. Myriad BSL2-compatible surrogate virus neutralization assays (VNAs) have been developed to overcome this barrier. Yet, there is marked variability between VNAs and how their results are presented, making intergroup comparisons difficult. To address these limitations, we developed a standardized VNA using CoV2-S pseudotyped particles (CoV2pp) based on vesicular stomatitis virus bearing the Renilla luciferase gene in place of its G glyco-protein (VSVDG); this assay can be robustly produced at scale and generate accurate neutralizing titers within 18 h postinfection. Our standardized CoV2pp VNA showed a strong positive correlation with CoV2-S enzyme-linked immunosorbent assay (ELISA) results and live-virus neutralizations in confirmed convalescent-patient sera. Three independent groups subsequently validated our standardized CoV2pp VNA (n . 120). Our data (i) show that absolute 50% inhibitory concentration (absIC50), absIC80, and absIC90 values can be legitimately compared across diverse cohorts, (ii) highlight the substantial but consistent variability in neutralization potency across these cohorts, and (iii) support the use of the absIC80 as a more meaningful metric for assessing the neutralization potency of a vaccine or convalescent-phase sera. Lastly, we used our CoV2pp in a screen to identify ultrapermissive 293T clones that stably express ACE2 or ACE2 plus TMPRSS2. When these are used in combination with our CoV2pp, we can produce CoV2pp sufficient for 150,000 standardized VNAs/week. IMPORTANCE Vaccines and antibody-based therapeutics like convalescent-phase plasma therapy are premised upon inducing or transferring neutralizing antibodies that inhibit SARS-CoV-2 entry into cells. Virus neutralization assays (VNAs) for measuring neutralizing antibody titers (NATs) are an essential part of determining vaccine or therapeutic efficacy. However, such efficacy testing is limited by the inherent dangers of working with the live virus, which requires specialized high-level biocontainment facilities. We there-fore developed a standardized replication-defective pseudotyped particle system that mimics the entry of live SARS-CoV-2. This tool allows for the safe and efficient measurement of NATs, determination of other forms of entry inhibition, and thorough investigation of virus entry mechanisms. Four independent labs across the globe validated our standardized VNA using diverse cohorts. We argue that a standardized and scalable assay is necessary for meaningful comparisons of the myriad of vaccines and antibody-based therapeutics becoming available. Abstract

The multinational Arabidopsis research community is highly collaborative and over the past thirty years these activities have been documented by the Multinational Arabidopsis Steering Committee (MASC). 16. Sustainable Availability Provision in Distributed Cloud Services

Abstract

The article is an extension of this paper 1 . It describes methods for dealing with reliability and fault tolerance issues in cloud-based datacenters. These methods mainly focus on the elimination of a single point of failure within any component of the cloud infrastructure, availability of infrastructure and accessibility of cloud services. Methods for providing the availability of hardware, software and network components are also presented. The analysis of the actual accessibility of cloud services and the mapping of a cloud-based datacenter infrastructure with different levels of reliability to the Tier Classification System2 is described. Non-compliance of the actual accessibility with the level of High Availability for cloud web services is unraveled.

17. SMART-ITEM: IoT-Enabled Smart Living

Abstract

The main goal of this proposed project is to harness the emerging IoT technology to empower elderly population to self-manage their own health, stay active, healthy, and independent as long as possible within a smart and secured living environment. An integrated open-sourced IoT ecosystem will be developed. It will encompass the entire data lifecycle which involves the following processes: data acquisition, data transportation; data integration, processing, manipulation and computation; visualisation; data intelligence and exploitation; data sharing; data storage. This innovative cloud-based IoT ecosystem will provide a one-stop shop for integrated smart IoT-enabled services to support older people (greater or equal to 65 years old) who live alone at home (or care homes). Another innovation of this system is the design and implementation of an integrated IoT gateway for wellbeing wearable and home automation system sensors with varying communication protocols. The SMART-ITEM system and services will appropriately address the following (i) smart health and care; (ii) smart quality of life; (iii) SMART-ITEM social community. The development of the system will be based on the User Centred Design methodology so as to ensure active user engagement throughout the entire project lifecycle and necessary standards as well as compliances will be adhered to (e.g. security, trust and privacy) in order to enhance user acceptance.

Abstract

Предлагается авторская позиция рассмотрения теории З. Фрейда как разновидности интенционалистских моделей психики. Выявлен «психотехнический» характер значительной части положений фрейдизма. Представлен анализ взаимосвязи метода психоанализа и особенностей представлений фрейдизма о сознании и бессознательном. Предлагается объяснение пансексуализма в психоанализе как следствия определенных исходных постулатов. Сопоставлен психоаналитический и религиозный взгляд на проблему отношений личности со своим Творцом. In the article the author's point of view on the theory of Freud as a kind of intention models of the psyche. Revealed "psychotechnical" nature of much of the provisions of Freudianism. The analysis of the relationship of the method of psychoanalysis and Freudian ideas about the characteristics of conscious and unconscious. 49. fhy3-1 retains inductive responses of phytochrome A.

Abstract

The fhy3 mutation of Arabidopsis impairs phytochrome A (phyA)-mediated inhibition of hypocotyl growth without affecting the levels of phyA measured spectrophotometrically or immunochemically. We investigated whether the fhy3-1 mutation has similar effects on very low fluence responses (VLFR) and high irradiance responses (HIR) of phyA. When exposed to hourly pulses of far-red light, etiolated seedlings of the wild type or of the fhy3-1 mutant showed similar inhibition of hypocotyl growth, unfolding of the cotyledons, anthocyanin synthesis, and greening upon transfer to white light. In the wild type, continuous far-red light was significantly more effective than hourly far-red pulses (at equal total fluence). In the fhy3-1 mutant, hourly pulses were as effective as continuous far-red light, i.e. the failure of reciprocity typical of HIR was not observed. Germination was similarly promoted by continuous or pulsed far-red in wild-type and fhy3-1 seeds. Thus, for hypocotyl growth, cotyledon unfolding, greening, and seed germination, the fhy3-1 mutant retains VLFR but is severely impaired in HIR. These data are consistent with the idea that VLFR and HIR involve divergent signaling pathways of phyA.
