Abstract: Water-based bio-sourced resists are promising candidates as alternatives for deep ultraviolet (DUV) lithography by replacing current photoresists issued from petro-chemistry for microelectronics application. Chitosan films produced from seafood industry wastes enable patterning processes free of organic solvent and alkali-based developers, by substitution with water. After demonstrating high-resolution patterning at lab-scale after transfer into silica 10 mm wafer, we investigate here the industrial pre-transfer chitosan-based photoresist on the 300 mm pilot line scale at CEA-Leti for 193 nm DUV lithography.

Abstract: Hybrid nanoparticles composed of an efficient nonlinear optical core and a gold shell can enhance and tune the nonlinear optical emission thanks to the plasmonic effect. However the influence of an incomplete gold shell, i.e., isolated gold nano-islands, is still not well studied. Here LiNbO3 (LN) core nanoparticles of 45 nm were coated with various densities of gold nano-seeds (AuSeeds). As both LN and AuSeeds bear negative surface charge, a positively-charged polymer was first coated onto LN. The number of polymer chains per LN was evaluated at 1210 by XPS and confirmed by fluorescence titration. Then, the surface coverage percentage of AuSeeds onto LN was estimated to a maximum of 30% using ICP-AES. The addition of AuSeeds was also accompanied with surface charge reversal, the negative charge increasing with the higher amount of AuSeeds. Finally, the first hyperpolarizability decreased with the increase of AuSeeds density while depolarization values for Au-seeded LN were close to the one of bare LN, showing a predominance of the second harmonic volumic contribution.

Abstract: Phase-pure, highly crystalline sub-50 nm LiNbO3 nanocrystals were prepared from a non-aqueous solvothermal process for 72 h at 230 °C and a commercial precursor solution of mixed lithium niobium ethoxide in its parent alcohol. A systematic variation of the reaction medium composition with the addition of different amounts of co-solvent including butanol, 1,3-propanediol, 1,4-butanediol, and 1,5-pentanediol resulted in the formation of nanocrystals of adjustable mean size and shape anisotropy, as demonstrated from XRD measurements and TEM imaging. Colloidal stability of ethanol- and water-based suspensions was evaluated from dynamic light scattering (DLS)/zeta potential studies and correlated with FTIR data. Thanks to the evolution in the nanocrystal size and shape distribution we observed, as well as to the available literature on the alkoxide chemistry, the reaction pathways and growth mechanisms were finally discussed with a special attention on the monomer formation rate, leading to the nucleation step. The polar, non-perovskite crystalline structure of LiNbO3 was also evidenced to play a major role in the nanocrystal shape anisotropy.

Abstract: Single-step orthogonal chemical functionalization procedures have been developed with patterned gold on silica surfaces. Different combinations of a silane and a thiol were simultaneously deposited on a gold/silica heterogeneous substrate. The orthogonality of the functionalization (i.e., selective grafting of the thiol on the gold areas and the silane on the silica) was demonstrated by X-ray photoelectron spectroscopy (XPS) as well as time-of-flight secondary ion mass spectrometry (ToF–SIMS) mapping. The orthogonal functionalization was used to immobilize proteins onto gold nanostructures on a silica substrate, as demonstrated by atomic force microscopy (AFM). These results are especially promising in the development of future biosensors where the selective anchoring of target molecules onto nanostructured transducers (e.g., nanoplasmonic biosensors) is a major challenge.

Abstract: The Gram negative bacterium Pseudomonas aeruginosa (PA) is an opportunistic bacterium that causes severe and chronic infection of immune-depressed patients. It has the ability to form a biofilm that gives a selective advantage to the bacteria with respect to antibiotherapy and host defenses. Herein, we have focused on the tetrameric soluble lectin which is involved in bacterium adherence to host cells, biofilm formation, and cytotoxicity. It binds to l-fucose, d-mannose and glycan exposing terminal fucose or mannose. Using a competitive assay on microarray, 156 oligosaccharides and polysaccharides issued from fermentation or from the biomass were screened toward their affinity to LecB. Next, the five best ligands (Lewisa, Lewisb, Lewisx, siayl-Lewisx and 3-fucosyllactose) were derivatized with a propargyl aglycon allowing the synthesis of 25 trivalent, 25 tetravalent and 5 monovalent constructions thanks to copper catalyzed azide alkyne cycloaddition. The 55 clusters were immobilized by DNA Directed immobilization leading to the fabrication of a glycocluster microarray. Their binding to LecB was studied. Multivalency improved the binding to LecB. The binding structure relationship of the clusters is mainly influenced by the carbohydrate residues. Molecular simulations indicated that the simultaneous contact of both binding sites of monomer A and D seems to be energetically possible.

Abstract: This review paper reports the applications of magnetic microbeads in biosensors and bio-bar code assays. Affinity biosensors are presented through different types of transducing systems: electrochemical, piezo electric or magnetic ones, applied to immunodetection and genodetection. Enzymatic biosensors are based on biofunctionalization through magnetic microbeads of a transducer, more often amperometric, potentiometric or conductimetric. The bio-bar code assays relie on a sandwich structure based on specific biological interaction of a magnetic microbead and a nanoparticle with a defined biological molecule. The magnetic particle allows the separation of the reacted target molecules from unreacted ones. The nanoparticles aim at the amplification and the detection of the target molecule. The bio-bar code assays allow the detection at very low concentration of biological molecules, similar to PCR sensitivity.

Abstract: International audience; Desorption ionization on silicon mass spectrometry (DIOS-MS) enables the high throughput analysis of low molecular weight biomolecules. However, the detection of metabolites biomarkers from complex fluids, like plasma, is subordinated to sample pre-treatments which limit clinical applications. Herein, we show that porous silicon chemically modified using monolayers of n-propyldimethylmetoxysilane molecules, can be a good candidate for the fingerprinting of Lysophosphatidylcholine (LysoPC) in plasma, without any pre-treatment of the sample, for DIOS-MS-based diagnosis like sepsis diagnosis. Results are correlated to the location of LysoPC molecules inside/outside the pores, determined by time of flight-secondary ions mass spectrometry profiling, and to their physico-chemical properties.

Abstract: In the context of the COVID-19 outbreak since December 2019, antigenic tests are widely used, for diagnosis purposes, to detect the SARS-CoV-2 spike protein in nasopharyngeal fluid through its interactions with specific antibodies. However, the SARS-CoV-2 spike protein is subject to rapid mutations yielding more and more variants that might lose their affinity toward the currently used antibodies. The virus entry into the host cell involves interactions between the angiotensin-converting enzyme 2 (ACE2) and the SARS-CoV-2 spike protein receptor-binding domain. Consequently, ACE2 could be a target with limited mutation escaping possibilities. However, as the enzyme has not evolved to recognize the virus, its affinity with the spike protein receptor-binding domain is lower than that with specific antibodies. The present molecular dynamics simulations study suggests that the adsorption of the ACE2 on specific silane monolayers could increase its affinity toward the spike protein receptor-binding domain. Indeed, silane monolayers, combining silane molecules with short alkyl chains and positively charged head groups and silane molecules without charged head groups, could adsorb the ACE2 while maintaining its bioactivity (orientation compatible with the spike protein trapping, low conformational changes) and increasing its interactions with the spike protein receptor-binding domain (number of hydrogen bonds and electrostatic interactions) to lead to an increase by 20% both in the binding free energy and in the enzyme /receptor-binding domain rupture force. This work could help develop biosensing tools efficient toward any variants of the SARS-CoV-2 spike protein.

Abstract: Understanding the organization of the hydration layer at functionalized silica surfaces is relevant for a large range of biosensing applications or surface phenomena such as biomolecule adsorption. Silane monolayers are widely used to functionalize silica surfaces. Using molecular dynamics simulations, we have investigated the role of silane molecule head-group charge, alkyl chain length, and surface coverage in the structural organization and dynamic properties of Na+ ions, Cl- ions, and water molecules at the interface. The silane molecules studied are 3-aminopropyldimethylethoxysilane, n-propyldimethylmethoxysilane, octadecyldimethylmethoxysilane, and (dimethylamino)dimethylsilylundecanoate. Our results suggest that the distribution of interfacial ions is sensitive to the 2D dispersion of the silane-charged head groups. Also, while charged silane monolayers show a strong orientation of interfacial water molecules, which leads to a rupture in the hydrogen bond network and disturbs their tetrahedral organization, the arrangement of water molecules at the interface with uncharged silane monolayers seems to be related to the surface roughness and to alkyl chain length. In line with these results, the diffusion of ions and water molecules is higher at the CH3 long monolayer interface than at the CH3 short monolayer interface and at the charged monolayer interfaces. Also, whatever the silane molecules studied, bulk properties are recovered around 0.7 nm above the interface. The interfacial water organization is known to impact biomolecule adsorption. Therefore, these results could further help in optimizing the functionalization layers to capture analytes.

Abstract: Macropatterned and micropatterned gold/silicon dioxide/titanium tungsten (Au/SiO2/TiW) substrates were orthogonally functionalized: three different molecules (monovalent silane, thiol, and phosphonic acid) were used to specifically form organolayers on Au, SiO2, or TiW areas of patterned substrates. The orthogonality of the functionalization (i.e., selective grafting of thiol on Au, phosphonic acid on TiW, and silane on SiO2) was assessed by X-ray photoelectron spectroscopy (XPS), time-of-flight secondary ion mass spectrometry (ToF-SIMS), Fourier transform infrared spectroscopy (FTIR), and contact angle measurements. These results are especially promising for the selective anchoring of targets (e.g., biomolecules, nanoparticles, nanowires, nanotubes, or other nano-objects) onto patterned zones of multimaterial substrates, such as nanosensors or other nanodevices.

Abstract: International audience; The development of environmentally friendly materials and processes is a major issue that concerns all industrial sectors, including microelectronics. The aim of this study is to demonstrate the possibility of using chitosan-based photoresists for microelectronic applications on silicon by 193 nm photolithography. The photopatterning of chitosan films is demonstrated and analyzed by different spectroscopy and microscopy techniques. In particular, it is shown that 193 nm irradiation allows one to induce chain breaks that modify the solubility of chitosan in an aqueous developing solution, without denaturing the chitosan macromolecule chains. This mechanism allows one to obtain patterns, and it is shown that these patterns can be transferred by physical etching into silica. It is also demonstrated that the formulated resins are compatible with industrial spin-coating and exposure tools, which opens very interesting perspectives for these chitosan-based positive resins in a microelectronic context.

Abstract: Hybrid nanoparticles composed of an efficient nonlinear optical core and a gold shell can enhance and tune the nonlinear optical emission thanks to the plasmonic effect. However the influence of an incomplete gold shell, i.e., isolated gold nano-islands, is still not well studied. Here LiNbO3 (LN) core nanoparticles of 45 nm were coated with various densities of gold nano-seeds (AuSeeds). As both LN and AuSeeds bear negative surface charge, a positively-charged polymer was first coated onto LN. The number of polymer chains per LN was evaluated at 1210 by XPS and confirmed by fluorescence titration. Then, the surface coverage percentage of AuSeeds onto LN was estimated to a maximum of 30% using ICP-AES. The addition of AuSeeds was also accompanied with surface charge reversal, the negative charge increasing with the higher amount of AuSeeds. Finally, the first hyperpolarizability decreased with the increase of AuSeeds density while depolarization values for Au-seeded LN were close to the one of bare LN, showing a predominance of the second harmonic volumic contribution. ispartof: NANOMATERIALS vol:11 issue:4 ispartof: location:Switzerland status: published

Abstract: International audience; Due to the ability of Pseudomonas aeruginosa (PA) to develop antibiotic resistances, alternative therapeutic strategies have been proposed. Among others, carbohydrate multivalent molecules targeting lectin‐based virulent factors have been widely reported in particular those targeting LecA. LecA is a tetravalent galactose specific lectin involved in biofilm formation and cell internalization. Herein, we report the synthesis of 36 galactoclusters built from galactosides with aromatic and non‐aromatic aglycons and with an additional chain. The chains were either neutral or positively charged. Only the galactoclusters with naphthyl or tyrosine aglycon showed a moderate increase of binding for the positively charged 3‐dimethylammonium propyl chain. In contrast, the non‐aromatic galactoclusters display typically poorer binding properties towards LecA. The introduction of these side chains led to improved affinities up to becoming comparable to the high‐affinity aromatic galactoclusters.

Abstract: International audience; Surface chemical functionalization is used in analytical tools to immobilize biomolecules that will capture a specific analyte and also to reduce the nonspecific adsorption. Silane monolayers are widely used to functionalize silica surfaces. Their interfacial properties are linked to the silane organization. Here, we study, by molecular dynamics simulations, the effects of silane molecule head-group charge, alkyl chain length, and surface coverage on the structure of silane monolayers. Four molecules are investigated: 3-aminopropyldimethylethoxysilane, n-propyldimethylmethoxysilane, octadecyldimethylmethoxysilane, and tert-butyl-11-(dimethylamino(dimethyl)silyl)undecanoate. The results suggest that, while long alkyl chains straighten out and adopt a more organized structure as surface coverage increases, the tilt angle of short chains is independent of surface coverage. Furthermore, in the case of long alkyl chains, a charged head-group seems to reduce the tilt angle to surface coverage dependence. The simulated alkyl chain tilt angles were qualitatively validated by infrared spectroscopy and X-ray photoelectron spectroscopy. Also, a hexagonal packing is observed in all of the monolayers but is more defined as surface coverage increases. The nematic order parameter suggests that this packing is governed by the parallel orientation of the first C–C bonds near the surface. So, even short alkyl chains, with a large tilt angle distribution, present a hexagonal packing.

20. Deciphering multivalent glycocluster–lectin interactions through AFM characterization of the self-assembled nanostructures

Abstract: International audience; Pseudomonas aeruginosa is a human opportunistic pathogen responsible for lung infections in cystic fibrosis patients. The emergence of resistant strains and its ability to form a biofilm seems to give a selective advantage to the bacterium and thus new therapeutic approaches are needed. To infect lung, the bacterium uses several virulence factors, like LecA lectins. These proteins are involved in bacterial adhesion thanks to their specific interaction with carbohydrates of the host epithelial cells. The tetrameric LecA lectin specifically binds galactose residues. A new therapeutic approach is based on the development of highly affine synthetic glycoclusters able to selectively link with LecA to interfere the natural carbohydrate-LecA interaction. In this study, we combined Atomic Force Microscopy imaging and Molecular Dynamics simulations to visualize and understand the arrangements formed by LecA and five different glycoclusters. Our glycoclusters are small scaffolds characterized by a core and four branches, which terminate by a galactose residue. Depending on the nature of the core and the branches, the glycocluster-lectin interaction can be modulated and the affinity increased. We show that glycocluster-LecA arrangements highly depend on the glycocluster architecture: the core influences the rigidity of the geometry and the directionality of the branches, whereas the nature of the branch determines the compactness of the structure and the ease of the binding.

Abstract: International audience; A current evolution of nanobiosensors stresses the interest of multi-material nanopatterned surfaces, for enhancing sensing performances. Besides, titanium tungsten (TiW) is mastered routinely implemented in nanoelectronic devices, in a reproducible way and at industrial production scales. Such material may be envisioned for being used in (bio)chemical nanoelectronic sensors, but the surface functionalization of such material has still to be studied. In the present article, orthogonal chemical functionalization of patterned Au on titanium tungsten (TiW) substrates have been explored for the first time to our knowledge. Surface functionalizations were assessed by X-ray photoelectron spectroscopy (XPS), polarization modulation infrared reflection-absorption spectroscopy (PMIRRAS) and time-of-flight secondary ion mass spectrometry (ToF-SIMS) imaging. Au/TiW patterned substrates were functionalized with mercapto-undecamine. Thanks to the orthogonality of thiol/Au versus phosphonic acid/TiW reactions, only Au features were modified leading to the amine derivatized surface. It allowed localizing carboxy-functionalized nanoparticles by electrostatic interaction on Au with a selectivity above 10 compared to TiW.

Abstract: International audience; This work presents a new and simple procedure for the shape selective purification of gold nanorods from a mixture of rods and spheres. Previously reported methods were time-consuming and revealed several drawbacks such as low yields and difficulty to recover the purified nanoparticles. Additionally, they were mostly applied to high aspect ratio (AR) nanorods. Our process is based on only simple and short centrifugation steps in order to precipitate specifically gold nanospheres. Samples containing low AR nanorods (AR < 6) were selected to perform the purification process. The supernatant content was followed by UV-Visible absorption spectroscopy after each centrifugation step. Then, transmission electron microscopy (TEM) allowed extract the purification efficiency thanks to shape analyses performed on more than 1000 nanoparticles. These results showed that our centrifugation process was applied successfully to three sizes of nanorods (2.4, 3.7, and 5.3). High purification yields of 72 and 78% were attained for AR = 3.7 and AR = 5.3 nanorods, respectively.

Abstract: International audience; This work focused on the introduction of electrochemical moieties (electroactive molecules) onto the surface of magnetic nanoparticles (NPs) based on carbodiimide coupling chemistry and the influence of magnetic attrac- tion at the working electrode surface on their electrochemical behaviour. The magnetic NPs were first func- tionalized with polyamidoamine (PAMAM) dendrimer generation 0 and then with ferrocenecarboxylic acid (Fc) as electroactive molecule. Colorimetric titration, zeta potential measurements and infrared spectroscopy proved the effective conjugation of PAMAM and Fc onto NPs. The conjugation of Fc onto the NPs was also evidenced by voltammetry as it resulted in a half-peak potential shift from 0.328 V for free Fc to 0.590 V after coupling. A transition from a reversible to an irreversible behaviour was also observed. Finally, an external magnetic field was applied on Fc-conjugated NPs to attract them onto the working electrode of a microchip electrochemical device, and the resulting modifications of the electrochemical behaviour were studied. Compared to Fc dispersed in solution, the current peak height of Fc-conjugated NPs was 2-fold higher under magnetic attraction. The mechanism of electrochemical reaction was investigated and was found to be irreversible. Surprisingly, the reaction was diffusion-controlled upon attraction.

Abstract: Preparation of stable BiFeO3 nanocrystal suspensions through a simple, low-cost precipitation technique is described. Amorphous precursors are first precipitated from metal nitrate salts in highly basic KOH solutions, and a short high-temperature annealing step is then performed to induce crystallization. Nanoparticles are characterized by X-ray diffraction (XRD), TEM, DLS and ζ-potential measurements, and the synthesis conditions optimized after a systematic variation of the KOH concentration within the range of 1–12 M. The presence of residual impurities (mainly Bi25FeO39 and Bi2Fe4O9) quantified from XRD and mean nanocrystal size is found to be strongly influenced by the initial KOH solution content. A concentration at about 3–4 M is optimal in terms of BiFeO3 phase-purity and nanocrystal size. Stability of aqueous dispersions of the amorphous precursors and of the purest crystallized nanoparticles is also characterized between pH = 2 and pH = 13. After preparation of stable, almost phase-pure BiFeO3 nanocrystal suspensions, second and third harmonic scattering (SHS and THS) at excitation wavelengths of 1064 nm and 1250 nm are reported from nonlinear optical scattering measurements and compared with other recently published literature values.

25. The anti-adhesive effect of glycoclusters on Pseudomonas aeruginosa bacteria adhesion to epithelial cells studied by AFM single cell force spectroscopy

Abstract: International audience; The human opportunistic pathogen Pseudomonas aeruginosa (PA) is responsible for chronic infections of the respiratory epithelium in cystic fibrosis patients. PA takes advantage of an arsenal of virulence factors to infect and colonize human lungs. Among them, the lectin LecA favours epithelium invasion by interacting with host cell globotriaosylceramide (Gb3). A new therapeutic approach is based on the development of synthetic multivalent molecules (glycoclusters) targeting LecA with a higher affinity than its natural ligand. Atomic force microscopy-single cell force spectroscopy has been used to study the effect of glycoclusters on the bacteria–cell interaction. Glycoclusters have been shown to affect the detachment work and detachment force of the bacteria–cell interaction. The specificity and the efficiency of the glycocluster in targeting the lectin and destabilizing the PA–epithelial cell adhesion are demonstrated and discussed.

Abstract: International audience; We report the use of the Second Harmonic Generation response from a riboflavin doped chitosan film as a characterization method of the film morphology. This film is of particular interest in the development of new and bio-sourced material for eco-friendly UV lithography. The method allows us to determine how riboflavin is distributed as a function of film depth in the sample. This possibility is of importance in order to have a better understanding of the riboflavin influence in chitosan films during the lithography process. On the contrary, linear optical techniques provide no information beyond the mere confirmation of the riboflavin presence.

Abstract: We report on the influence of silver-silica core-shell nanoparticles (Ag@SiO2 NPs) and bare silver nanoparticles (Ag NPs) on the photoelectric current of plasmonic inverted organic solar cells. The synthesized Ag@SiO2 NPs are deposited on a zinc oxide layer. The optimized plasmonic devices concern the Ag@SiO2 NPs with 5 nm shell thickness and present the highest short-circuit current density of 13.44 mA/cm2, enhanced by 12% compared to the reference device. Such enhancements can mainly be attributed to the localized surface plasmon resonance and to the light scattering effect originating from Ag NPs and Ag@SiO2 NPs. Finite-difference time-domain simulation reveals that a thin shell thickness facilitates the extension of a strong localized enhanced electromagnetic field in the active layer, as confirmed by measured and simulated optical absorption and photoluminescence measurements. Furthermore, AFM images present a well-distributed Ag@SiO2 NPs array on the ZnO films, which contributes to such an improvement.

Abstract: Oligo ethylene glycol layers are widely used in biosensor applications, mainly for their anti-fouling abilities. Such surfaces are often characterized by X-ray photoelectron spectroscopy (XPS) as this method allows a precise determination of the surface chemical composition. We show herein that X-rays used during XPS characterization quickly and significantly degrade oligo ethylene glycol immobilized onto silica substrates. It is therefore necessary to introduce a correcting factor to assess the true (i.e. without degradation) corresponding ether contribution in the XPS spectrum of such organic layers. Eventually, fluorescence scans show the loss of anti-fouling properties of the degraded surface, leading to greater amounts of adsorbed (fluorescently labeled) protein. Abstract: International audience; Biopolymers represent natural, renewable and abundant materials. Their use is steadily growing in various areas (food, health, building …) but, in lithography, despite some works, resists, solvents and developers are still oil-based and hazardous chemicals. In this work, we replaced synthetic resist by chitosan, a natural, abundant and hydrophilic polysaccharide. High resolution sub-micron patterns were obtained through chitosan films as water developable, chemically unmodified, positive tone mask resist for an eco-friendly electron beam and deep-UV (193 nm) lithography process. Sub-micron patterns were also successfully obtained using a 248 nm photomasker thanks to the addition of bio-sourced photoactivator, riboflavin. Patterns were then transferred by plasma etching into silica even for high resolution patterns.

This article may be used for non-commercial purposes in accordance with Wiley Terms and Conditions for Use of Self-Archived Versions.; International audience; The efficient conjugation of ferrocenecarboxylic acid (Fc), a usual electroactive probe, onto COOH‐modified magnetic nanoparticles (NPs) is reported. Amine crosslinkers were used to couple COOH groups from NPs and from Fc. The coupling efficiencies using different generations of polyamidoamine (PAMAM) dendrimer and ethylenediamine (EDA) were compared through zeta potential measurements, square‐wave voltammetry (SWV) and NH2 titration. Highest PAMAM generations led to NH2‐modified NPs with increased zeta potential and NH2 density compared to lowest ones. Moreover, no aggregation was observed after crosslinker grafting. SWV was applied to characterize Fc‐modified NPs. PAMAM generations 0–3 improved the electrochemical signal in comparison to EDA. However, increasing PAMAM generations led to NPs aggregation. Quantitative results supported the fact that PAMAM generation 0 was the best compromise for large electrochemical signal without NPs aggregation.

Abstract: International audience; The urokinase-type plasminogen activator (uPA) and PA inhibitor 1 (PAI-1) play important roles in breast cancer metastasis through cell migration and invasion. They are clinically applicable prognostic and predictive markers. High levels of uPA and PAI-1 are associated with high risk of recurrence and adjuvant chemotherapy provides substantial benefit for this breast cancer population. The current sole validated method for quantifying uPA level in breast tumour tissue is ELISA assay. It requires 50-300 mg of fresh or frozen tissue, which is the main limitation for routine use. In this study, we evaluated the performances of customized antibody microarray to quantify uPA concentration from reduced extraction solution of breast tumour tissue and compared it with standard ELISA kit. We firstly optimized the elaboration of customized antibody microarray in order to sensitively detect and quantify uPA standard solutions. In the best conditions, we analysed uPA concentration in 16 cytosolic extracts from breast tumour tissue. Results showed that our customized antibody microarray could correctly quantify uPA concentration while consuming 100 times less volume of tumour tissue extraction solution than ELISA. Our antibody microarray is a powerful and promising tool for the miniaturization of the immunoassay quantification of uPA from breast tumour tissue extracts.

Abstract: Current chemicals used in electron beam (e-beam) lithography generate safety and waste management issues. To replace them, chitosan, a natural and abundant polymer soluble in water based solutions, was assessed as a positive and water developable resist for a two-layer e-beam lithography and as a mask for transfer by etching in silica. Fifty nanometer line patterns were successfully obtained in a chitosan film by e-beam lithography at doses between 160 and 300 μC cm−2, then, transferred into a silica layer by CHF3 plasma reactive ion etching with respect of the feature dimensions.

Abstract: Mannose-centered galactoclusters specific for lectin I of Pseudomonas aeruginosa (LecA) were synthesised by a combination of phosphoramidite chemistry and metal-free thiol click chemistry (i.e., thiol addition to acrylamide or nucleophilic displacement of bromine in a bromoacetamide group by a thiol function). These thiol click reactions were performed with microwave assistance in the presence of Et3N and with use of a reducing agent to avoid disulfide formation. Nine tetravalent galactoclusters containing different linkers (aliphatic, oligoethyleneglycol or aromatic) were synthesised with a DNA tag. Their binding to LecA was monitored in a DNA-based glycoarray and compared with that of a galactocluster synthesised by copper-catalyzed azide–alkyne cycloaddition. The results indicated stronger binding of all galactoclusters relative to the monovalent galactoside but slightly weaker binding than that shown by the galactocluster incorporating a triazole ring, due to a favourable interaction of the latter with proline 51 of LecA.

Abstract: International audience; A library of 24 new mannose-centered tetragalactoclusters with four different linkers (di- and triethyleneglycol with phosphodiester or phosphorothioate linkages) and six different aromatic aglycons (O-phenyl, S-phenyl, O-benzyl, S-benzyl, O-biphenyl and O-naphthyl) was synthesized. Their interactions with LecA were evaluated on a DNA Directed Immobilization (DDI) based glycocluster array allowing the determination of their IC50 against lactose and the evaluation of their dissociation constant (Kd). Finally, the docking simulations confirm the experimental results and demonstrated that the better affinity of O-biphenyl- and O-naphthyl-galactoside is due to a double interaction between the aromatic ring and the histidine 50 and proline 51 of LecA.

Abstract: Nanoparticles composed of a silver core coated with a silica shell (Ag@SiO 2 ) were prepared. A dye, fluorescein isothiocyanate (FITC), was further encapsulated during the growth of a second silica shell onto Ag@SiO 2 nanoparticles. The proximity of silver nanoparticles led to a 4-fold maximal enhancement in the fluorescence of FITC when the first silica shell thickness was set at 5 nm. After amino-functionalization of Ag@SiO 2 -FITC nanoparticles, iron oxide nanoparticles were bonded to their surface. The magnetic and metal-enhanced fluorescence properties appeared simultaneously when Ag@SiO 2 -FITC-Fe 3 O 4 hybrid nanoparticles were dispersed in a solution.

Abstract: New therapeutics strategy against cystic fibrosis seeks to prevent the adhesion of the bacterium Pseudomonas aeruginosa (PA) on the epithelial cells in the lungs. One of the factors that induces the adhesion is the interaction between natural glycocluster present on the cells and lectins such as the PA lectin LecA (PA-IL) present on the bacterium. By introducing synthetic glycoclusters with a great affinity with the lectin PA-IL, the adhesion can be prevented. In this study, we characterized, by atomic force microscopy, the interaction between a tetra-galactosylated glycocluster and the PA-IL lectin for high concentration of lectins (2.5 μM).We showed that the strong lectin/lectin interaction is reduced even for low concentration of glycoclusters (1 for 20 000 lectins). We assumed that it is due to the tensioactive behavior of the glycoclusters. It was shown that the arrangement of the created complexes induced different structures evolving from one-dimensional elongated aggregates to two-dimensional compact islands when increasing the glycocluster concentration. This evolution can be interpreted as the predominance of the glycocluster/lectin interaction. Copyright © 2013 John Wiley & Sons, Ltd.

Abstract: International audience; Lectin A (LecA) from Pseudomonas aeruginosa is an established virulence factor. Glycoclusters that target LecA and are able to compete with human glycoconjugates present on epithelial cells are promising candidates to treat P. aeruginosa infection. A family of 32 glycodendrimers of generation 0 and 1 based on a bifurcated bis-galactoside motif have been designed to interact with LecA. The influences both of the central multivalent core and of the aglycon of these glycodendrimers on their affinity toward LecA have been evaluated by use of a microarray technique, both qualitatively for rapid screening of the binding properties and also quantitatively (Kd ). This has led to high-affinity LecA ligands with Kd values in the low nanomolar range (Kd =22 nm for the best one).

Abstract: International audience; Thanks to their specificity and stability in the sera, autoantibodies (AAbs) against tumor-associated antigens (TAAs) are very attractive biomarkers for the development of less invasive serological tests for the diagnosis and prognosis of cancer. Heat shock proteins (HSP) belong to TAAs and they are over-expressed in various human cancers. Elevated HSP can stimulate the immune system to produce anti-HSP antibodies. So far, AAbs against HSP have been identified in the circulation of various cancer patients. Here we will review current literature on the use of anti-HSP antibodies for cancer diagnosis and prognosis. The challenges as well as future directions of AAbs identification in oncology are also discussed.

Abstract: International audience; Anti‐infectious strategies against pathogen infections can be achieved through antiadhesive strategies by using multivalent ligands of bacterial virulence factors. LecA and LecB are lectins of Pseudomonas aeruginosa implicated in biofilm formation. A series of 27 LecA‐targeting glycoclusters have been synthesized. Nine aromatic galactose aglycons were investigated with three different linker arms that connect the central mannopyranoside core. A low‐nanomolar (Kd=19 nm, microarray) ligand with a tyrosine‐based linker arm could be identified in a structure–activity relationship study. Molecular modeling of the glycoclusters bound to the lectin tetramer was also used to rationalize the binding properties observed.

Abstract: pH was used as the main driving parameter for specifically immobilizing silicon nanowires onto Si3N4 microsquares at the surface of a SiO2 substrate. Different pH values of the coating aqueous solution enabled to experimentally distribute nanowires between silicon nitride and silicon dioxide: at pH 3 nanowires were mainly anchored on Si3N4; they were evenly distributed between SiO2 and Si3N4 at pH 2.8; and they were mainly anchored on SiO2 at pH 2. A theoretical model based on DLVO theory and surface protonation/deprotonation equilibria was used to study how, in adequate pH conditions, Si nanowires could be anchored onto specific regions of a patterned Si3N4/SiO2 surface. Instead of using capillary forces, or hydrophilic/hydrophobic contrast between the two types of materials, the specificity of immobilization could rely on surface electric charge contrasts between Si3N4 and SiO2. This simple and generic method could be used for addressing a large diversity of nano-objects onto patterned substrates.

Abstract: International audience; Homo- and heterofunctionalized glycoclusters with galactose and/or fucose residues targeting both PA-IL and PA-IIL lectins of Pseudomonas aeruginosa were synthesized using "Click" chemistry and DNA chemistry. Their binding to lectins (separately or in a mixture) was studied using a DNA Directed Immobilization carbohydrate microarray. Homoglycoclusters bind selectively to their lectin while the heteroglycocluster binds simultaneously both lectins with a slight lower affinity.

Abstract: International audience; Pseudomonas aeruginosa (PA) is a Gram negative opportunistic pathogen and is the major pathogen encounter in the cystic fibrosis (CF) lung airways. It often leads to chronic respiratory infection despite aggressive antibiotic therapy due to the emergence of resistant strains and to the formation of biofilm. The lectin PA-IIL (LecB) is a fucose-specific lectin from PA suspected to be involved in host recognition/adhesion and in biofilm formation. Thus, it can be foreseen as a potential therapeutic target. Herein, 16 fucosylated glycoclusters with antenna-like, linear, or crownlike spatial arrangements were synthesized using a combination of DNA solid-phase synthesis and alkyne azide 1,3-dipolar cycloaddition (CuAAC). Their binding properties toward PA-IIL were then evaluated based on DNA directed immobilization (DDI) carbohydrate microarray. Our results suggested that the antenna-like scaffold was preferred to linear or crown-like glycoclusters. Among the crown-like carbohydrate centered fucosylated glycoclusters, mannose-based core was better than glucose- and galactose-based ones. The influence of the linker arm was also evaluated, and long linkers between fucoses and the core led to a slight better binding than the short ones.

Abstract: Silicon carbide (SiC) nanoparticles were coated with gold nanorods to prepare nonlinear plasmonic nanohybrids. Their structural properties were evaluated by Transmission Electron Microscopy. Nonlinear optical properties of the two components (second harmonic generation for SiC nanoparticles, two-photon luminescence for gold nanorods) were simultaneously found into nanohybrids and spatially colocalized. Photothermal effect was observed for nanohybrids. The cytotoxicity of nanoparticles, nanorods and nanohybrids towards healthy and cancer cell lines was then evaluated. SiC nanoparticles and nanohybrids showed no cytotoxicity for healthy cells while gold nanorods were found to exhibit an increasing toxicity with increasing concentrations, which was related to the presence of CTAB surfactant on their surface. Finally, nanohybrids exhibit toxicity for cancer cells and this effect was increased under laser irradiation at 808 nm, which matches the plasmon resonance of gold nanorods.

Abstract: International audience; Carbohydrate and glycoconjugates play a major role in key biological events such as cell-cell recognition, pathogenesis and inflammation[1]. As a consequence, there is a need to understand the structural parameters governing the recognition of carbohydrate by their receptors. Technologies for rapid monitoring and evaluating such interactions are of great importance to provide deep insights relevant to carbohydrate involving biological events. However, most conventional approaches are cumbersome and material and time consuming. Carbohydrate microarray technology is a promising approach for probing carbohydrate/protein interactions because it permits the simultaneous screening of a number of biological interactions with only minute amounts of material. Herein, we develop a strategy to probe different glycoconjugates (different spatial configuration, linker, geometry...) toward proteins in one time on a DNA Directed Immobilization (DDI) oligosaccharide biochip.

Abstract: International audience; We present a label-free biosensor capable to map local potential by photogenerating charge carriers in a field effect device. It consists in an amplitude-modulated laser beam scanning through an electrolyte/insulator/semiconductor structure at very low illumination around the equilibrium state of semiconductor. A proof of principle is demonstrated by depositing a self-assembled monolayer on the dielectric and by showing a clear photopotential change between naked and covered areas.
