During plant growth and development, the YABBY gene plays a crucial role in the morphological structure, hormone signaling, stress resistance, crop breeding, and agricultural production of plant lateral organs, leaves, flowers, and fruits. Astragalus mongholicus is a perennial herbaceous plant in the legume family, widely used worldwide due to its high medicinal and edible value. However, there have been no reports of the YABBY gene family in A. mongholicus. This study used bioinformatics methods, combined with databases and analysis websites, to systematically analyze the AmYABBY gene family in the entire genome of A. mongholicus and verified its expression patterns in different tissues of A. mongholicus through transcriptome data and qRT-PCR experiments. A total of seven AmYABBY genes were identified, which can be divided into five subfamilies and distributed on three chromosomes. Two pairs of AmYABBY genes may be involved in fragment duplication on three chromosomes. All AmYABBY proteins have a zinc finger YABBY domain, and members of the same group have similar motif composition and intron – exon structure. In the promoter region of the genes, light-responsive and MeJa-response cis-elements are dominant. AmYABBY is highly expressed in stems and leaves, especially AmYABBY1, AmYABBY2, and AmYABBY3, which play important roles in the growth and development of stems and leaves. The AmYABBY gene family regulates the growth and development of A. mongholicus. In summary, this study provides a theoretical basis for in-depth research on the function of the AmYABBY gene and new insights into the molecular response mechanism of the growth and development of the traditional Chinese medicine A. mongholicus.

To study the buckling behavior and the design method of the load-carrying capacity of cold-formed thin-walled lipped angle, axial compression tests were carried out on 32 lipped angle columns with different sections and slenderness ratios which were manufactured with high strength zinc-coated grades G450 structural steel sheets. The experimental results showed that the long column with a small width-to-thickness ratio displayed global flexural-torsional buckling, local buckling was found for the short column with a large width-to-thickness ratio. Other specimens demonstrated interactive buckling with local buckling and flexural-torsional buckling. The specimens with torsional buckling all showed post-torsional buckling strength. Then ABAQUS finite element software was used to simulate the lipped angle specimens. The simulation results were in good agreement with the test results about buckling modes and ultimate strength which indicated that the established finite element analysis model was reasonable and feasible. Therefore, the influences of the slenderness ratio, width-thickness ratio, and width ratio of the leg to lip on the ultimate strength of cold-formed thin-walled lipped angle were analyzed using finite element software. The analysis showed that the ultimate capacity of the angle decreased greatly with the increase of the slenderness ratio. The ultimate capacity increased in the range of width-to-thickness ratios of 15–30 when the angles had the same lengths. The increasing of the width of the lips increased the ultimate capacity, but the increase of the ultimate capacity of the angles would lower when local buckling occurred at the lips. Finally, based on the test results, the modified effective width method and direct strength method were proposed to calculate the capacity of the cold-formed thin-walled lipped angle under axial compression. The comparison between the predicted results with the test and finite element analysis showed that the proposed methods were accurate and feasible.

Under shut-in conditions in deepwater drilling, the gas invading the bottomhole ascends along the wellbore and accumulates at the wellhead, forming a high-pressure trap, challenging wellbore pressure prediction and control. The accurate prediction of bottomhole pressure is essential for well control during shut-in conditions. In this study, a new bottomhole pressure prediction model that considers wellbore storage effects was developed to address gas invasion issues during shut-in conditions in deepwater drilling. This model incorporates factors such as the wellbore elasticity, fluid compressibility, and drilling fluid filtration loss. The calculated values show good agreement with experimental values, with the average absolute and relative errors of 2.095 × 10−2 MPa and 3.71%, respectively. Meanwhile, the results indicate that the bottomhole pressure initially increases logarithmically over time and then transitions to a linear increase, and the residual flow and gas ascent significantly influence the bottomhole pressure. Finally, the effects of various parameters on the bottomhole pressure were evaluated. Larger initial pressure differential, exposed thickness, and formation permeability accelerate the increase in bottomhole pressure during residual flow stage, while smaller filter cake permeability and drilling fluid viscosity quicken its increase during gas ascent stage. Drilling fluid density affects the initial pressure and the residual flow duration. The findings of this study would provide theoretical support for well control operations in deepwater drilling.

Background and Objectives: The Chansu injection (CSI), a sterile aqueous solution derived from Chansu, is applied in clinical settings to support antitumor and anti-radiation treatments. CSI’s principal active components, bufadienolides (≥90%), demonstrate potential effects on pancreatic cancer (PDAC), but their underlying mechanisms remain unclear. This study aimed to elucidate the antitumor effects and pathways associated with CSI in PDAC. Methods: Network pharmacology and bioinformatics analyses explored CSI’s mechanisms against PDAC. MTT, colony-formation, and migration assays evaluated CSI’s impact on proliferation and migration in PANC-1 and MIA PACA-2 cells, both as a single agent and in combination with erlotinib (EGFR inhibitor). Cell cycle analysis employed flow cytometry. Animal experiments were performed on tumor-bearing mice, with targets and pathways assessed via molecular docking and western blotting. Results: CSI treatment suppressed PDAC cell proliferation and migration by inducing G2/M phase arrest. Network pharmacology, bioinformatics, and molecular docking indicated that CSI’s anti-PDAC effects may involve EGFR pathway modulation, with CSI lowering p-EGFR/KRAS/p-ERK1/2 pathway expressions in PDAC cells. Additionally, sustained KRAS activation in mediating erlotinib resistance in PDAC and CSI potentiated erlotinib’s antitumor effects through enhanced KRAS and p-ERK1/2 inhibition. CSI also enhanced erlotinib’s efficacy in tumor-bearing mice without causing detectable toxicity in renal, cardiac, or hepatic tissues at therapeutic doses. Conclusions: CSI as an adjuvant used in antitumor and anti-radiation therapies enhanced erlotinib’s antitumor effects through modulation of the KRAS pathway. CSI and erlotinib’s synergistic interaction represents a promising approach for addressing erlotinib resistance in PDAC treatment.

Constructing fast electron transfer pathways and abundant electro-active sites is an effective strategy to improve the oxygen evolution reaction (OER) performance of catalysts. Herein, structural engineering and dual-phase engineering were employed to construct a NiS2 nanoparticle-encapsulated MOF configured with a pseudo-neuronal structure (NiS2/MOF/HT). It was found that the pseudo-neuronal structure, constructed with a carbon nanohorn (CNH) and carbon nanotube (CNT), provided fast electron transfer pathways and abundant exposed active sites. Moreover, the NiS2/MOF/HT composite obtained via partial vulcanization not only inherited the pseudo-neuronal structure but also prevented the aggregation and growth of NiS2 particles. NiS2/MOF composites provide various active sites. With the combination of the promotion of electronic transfer and enrichment of electro-active sites (NiS2, MOF), NiS2/MOF/HT showed excellent performance, whose overpotential at 25 mA cm−2 was reduced by 19.5% compared with MOF/HT.

As oil and gas exploration gradually advances into deep waters, the combined effects of various types of gas kick and the accurate calculation of the gas-kick volume have gained increasing attention. This study focused on gas kicks from permeable gas-bearing formations, considering the mass transfer of gas in the filtration region of the drilling fluids and revealed the mechanisms of seepage-driven and diffusion-driven gas kicks. Based on seepage mechanics and diffusion theory, a comprehensive model for calculating gas-kick volume was established, considering the synergistic effect of gas-concentration-diffusion and negative-differential-pressure, as well as mass transfer in both the filtrate zone and the filter-cake zone. The new model showed high calculation accuracy. The sensitivity analysis showed that both the seepage-driven and diffusion-driven gas-kick volumes in the wellbore increased with increasing formation porosity and open-hole length, while the thickness of the filter cake had a strong inhibitory effect on both. Additionally, a “seepage–diffusion ratio” was introduced to reveal the gas-kick evolution pattern under a seepage–diffusion mechanism. Under specific case conditions, when the seepage–diffusion ratio was less than approximately 1%, diffusion-driven gas kick contributed more than seepage-driven gas kick; when the seepage–diffusion ratio exceeded 1%, seepage-driven gas kick contributed more than diffusion-driven gas kick. The research can provide crucial parameters for wellbore multiphase flow calculation and wellbore pressure prediction.

An experimental study of cold-formed thin-walled steel unequal-leg angles (CFTWS-ULAs) under axially oriented pressure is presented in this paper. Firstly, the initial imperfections and material properties of the angle specimens were measured in detail. The angle specimens were tested under fixed-ended conditions. The results of the experiments showed that the angle specimens with small slenderness ratios were susceptible to local buckling, the angle specimens whose legs had high slenderness ratios and low width–thickness ratios were found to easily suffer from the occurrence of flexural buckling, and the angle specimens whose legs had high width–thickness ratios were found to easily suffer from the occurrence of interactive buckling between local buckling and flexural buckling. The finite element analysis of the ULAs was conducted using ABAQUS6.14 finite element software by creating a model. The buckling modes and ultimate bearing capacities of the test specimens were compared, and the finite element analysis verified that the established model built using the finite element is credible and subsequent parametric analysis was performed. The slenderness ratio had the most significant impact on the ultimate bearing capacities of the unequal-leg angles, as indicated by the analysis results. When the width–thickness ratio and the width ratio of the legs fell within a specific range, the ultimate bearing capacities of the unequal-leg angles increased as the width–thickness ratio and the width ratio of the legs increased. Finally, the comparison results showed that the design strengths predicted by the specifications were very conservative, because the local buckling and torsional buckling were calculated at the same time. Therefore, a recommendation was proposed that the calculation of the load-carrying capacity of an unequal-leg angle should ignore torsional buckling.

Abstract Background Pseudoaneurysm of the mitral-aortic intervalvular fibrosa (P-MAIVF) is an uncommon but potentially life-threatening condition. The most common pathogenic factors of P-MAIVF are infective endocarditis and surgical valve operation. Here, we report a rare case of P-MAIVF which occurred one year after percutaneous transluminal coronary angioplasty (PTCA). Case presentation A 31-year-old man developed a P-MAIVF one year after PTCA. Transthoracic echocardiography (TTE) revealed a pseudoaneurysm between the aortic root and the left atrium. Three-dimensional transesophageal echocardiography (3D-TEE) clearly demonstrated the orifice of the pseudoaneurysm. This case was initially diagnosed by ultrasound, and the prognosis was good after surgical repair. Conclusions We report a rare case of P-MAIVF that occurred one year after PTCA.

Abstract Introduction Despite numerous observational studies reporting a positive correlation between polyunsaturated fatty acids (PUFAs) and the risk of sepsis and mortality, the causation of such an association has yet to be firmly established. Thus, our study aimed to undertake the Mendelian randomization (MR) approach to scrutinize the potential causalities of PUFAs with sepsis and mortality risk. Methods We conducted the MR investigation using genome-wide association study (GWAS) summary statistics of PUFAs [including omega-3 fatty acids (omega-3), omega-6 fatty acids (omega-6), the ratio of omega-6 to omega-3 fatty acids (omega-6:3), docosahexaenoic acid (DHA), linoleic acid (LA)], sepsis, and sepsis mortality. We utilized the GWAS summary data from the UK Biobank. To establish reliable causality, we employed the inverse-variance weighted (IVW) method as the primary analytical approach, together with four additional MR methods. In addition, we performed heterogeneity and horizontal pleiotropy assessments using Cochrane’s Q test and MR-Egger intercept test, respectively. Finally, we performed a series of sensitivity analyses to enhance the precision and veracity of our findings. Results The IVW method showed that genetically predicted omega-3 [odd ratio (OR) 0.914, 95% confidence interval (CI) 0.845–0.987, P = 0.023] and DHA (OR 0.893, 95% CI 0.815–0.979, P = 0.015) were suggestively linked to a decreased risk of sepsis. Furthermore, genetically predicted DHA (OR 0.819, 95% CI 0.681–0.986, P = 0.035) was suggestively associated with a reduced risk of sepsis-related death. Conversely, the omega-6:3 ratio (OR 1.177, 95% CI 1.011–1.371, P = 0.036) was suggestively linked to an increased risk of sepsis-induced mortality. On the basis of the MR-Egger intercept assessment, it appears that our MR examination was not influenced by any horizontal pleiotropy (all P > 0.05). Moreover, the reliability of the estimated causal association was confirmed by the sensitivity analyses. Conclusion Our study supported the casual effect between PUFAs and susceptibility to sepsis and sepsis-related death. Our findings underline the importance of specific PUFAs levels, particularly for individuals with a genetic susceptibility to sepsis. Further research is needed to confirm these findings and investigate the underlying mechanisms.

This research presents a new, eco-friendly, and swift method combining solid-phase extraction and hydrophobic deep eutectic solvents (DES) with high-performance liquid chromatography (SPE-DES-HPLC) for extracting and quantifying catechin and epicatechin in Shanxi aged vinegar (SAV). The parameters, such as the elution solvent type, the XAD-2 macroporous resin dosage, the DES ratio, the DES volume, the adsorption time, and the desorption time, were optimized via a one-way experiment. A central composite design using the Box–Behnken methodology was employed to investigate the effects of various factors, including 17 experimental runs and the construction of three-dimensional response surface plots to identify the optimal conditions. The results show that the optimal conditions were an HDES (tetraethylammonium chloride and octanoic acid) ratio of 1:3, an XAD-2 macroporous resin dosage of 188 mg, and an adsorption time of 11 min. Under these optimal conditions, the coefficients of determination of the method were greater than or equal to 0.9917, the precision was less than 5%, and the recoveries ranged from 98.8% to 118.8%. The environmentally friendly nature of the analytical process and sample preparation was assessed via the Analytical Eco-Scale and AGREE, demonstrating that this method is a practical and eco-friendly alternative to conventional determination techniques. In summary, this innovative approach offers a solid foundation for the assessment of flavanol compounds present in SAV samples.

Abstract Background Axial involvement in children with enthesitis-related arthritis (ERA) has characteristics that differ from those of peripheral involvement. This study characterized their clinical characteristics and treatment. Methods Patients with ERA at the Children’s Hospital of Nanjing Medical University between January 2018 and December 2020 were included. The ERA cohort was divided into two based on the presence or absence of axial joint involvement. Demographic characteristics, clinical features, and treatments were described and compared. Results In total, 105 children with ERA were enrolled (axial ERA, n = 57; peripheral ERA, n = 48). The age at disease onset of the axial group tended to be higher (11.93 ± 1.72 vs. 11.09 ± 1.91 years) and the diagnosis delay was bigger in patients with axial ERA (10.26 ± 11.66 months vs. 5.13 ± 7.92 months). The inflammatory marker levels were significantly higher in patients with axial. There were no differences in HLA-B27 positivity between the groups (34 [59.65%] vs. 28 [58.33%], P > 0.05). Hip involvement was more frequent in the axial group (52.63% vs 27.08%; X2 = 7.033). A total of 38 (66.67%) and 10 (20.83%) patients with axial and peripheral ERA, respectively, were treated with biological disease-modifying anti-rheumatic drugs (DMARDs) at diagnosis. The administration of biologics increased gradually in the axial ERA group, peaking at 18 months and decreasing thereafter, whereas that in the peripheral ERA group peaked at 6 months and began to decline thereafter. Conclusions Axial ERA is a persistent active disease and requires a more aggressive treatment. Classification and early recognition of axial involvement may help with timely diagnosis and appropriate management.

Betula pendula purple rain is a variety of Betula pendula that is native to Europe and has important ornamental and economic value. In this study, we sequenced the complete chloroplast genome of B. pendula purple rain. This genome had a typical quadripartite structure with 160,552 bases, including a large single copy (LSC) region of 89,433 bases, a small single copy (SCC) region of 19,007 bases and two inverted repeat (IR) regions of 26,056 bases. The GC content of the chloroplast genome was 36% and contained 124 genes, including 79 protein-coding genes, 8 rRNA genes and 37 tRNA genes. The maximum likelihood phylogenetic analysis of reported chloroplast genomes showed that B. pendula purple rain was most closely related to Betula occidentalis and Betula platyphylla.

Abstract Kinectin 1 antisense RNA 1 (KTN1-AS1), a long non-coding RNA (lncRNA), has been proved to have tumor-promoting properties and its expression is enhanced in several human tumors. However, the role of KTN1-AS1 in the pathogenesis of esophageal squamous cell carcinoma (ESCC) remains unknown. This study aimed to investigate the expression status, functional roles, and molecular mechanisms of KTN1-AS1 in the development of ESCC. Considerable upregulation of KTN1-AS1 was confirmed in esophageal cancer cells and ESCC tissues and its expression was associated with TNM stage, pathological differentiation, and lymph node metastasis. SOX2 directly activated transcription of KTN1-AS1, and overexpression of KTN1-AS1 facilitated ESCC cells proliferation and invasion in vitro and in vivo. Furthermore, KTN1-AS1 could bind to retinoblastoma binding protein 4 (RBBP4) in the nucleus and enhanced its binding with histone deacetylase 1 (HDAC1), thereby activating the epithelial–mesenchymal transition (EMT) process through downregulating E-cadherin expression at the epigenetic level. In conclusion, KTN1-AS1, induced by SOX2, acts as a tumor-promoting gene and may serve as a potential therapeutic and prognostic biomarker for ESCC.

During deepwater drilling, testing, production, or hydrate mining, the circulating medium in the wellbore may contain solid particles, such as rock chips and sand, in addition to drilling fluids, gas, and water. In the high-pressure, low-temperature conditions of deep water, gas intrusion can easily combine with free water in the drilling fluid to form hydrates, increasing the drilling risk. Therefore, understanding the formation patterns of hydrates in drilling fluids is of significant importance for the prevention and control of hydrates. This study utilized a small-scale high-pressure reactor to investigate the impact of the stirring rate, NaCl, and methanol additives, as well as the sand content on the hydrate formation process and gas consumption. The results indicate that the hydrate formation process can be divided into an induction stage, a rapid formation stage, and a slow formation stage. The induction stage and rapid formation stage durations are significantly reduced under stirring conditions. In NaCl and methanol solutions, hydrate formation is inhibited, with the induction stage duration increasing with higher concentrations of NaCl and methanol. There was no apparent rapid formation stage observed. The final gas consumption decreases substantially with increasing concentrations of NaCl and methanol, reaching no significant hydrate formation at a 20% concentration. The sand content has a significant impact on the slow formation stage, with the final gas consumption increasing within a certain range (in this work, at a sand content of 20%), and being notably higher than in the pure water system under the same conditions.

ObjectiveIn children with enthesitis-related arthritis (ERA), the hip and sacroiliac joint function might be impaired if not properly treated. We sought to evaluate the effectiveness of anti-tumor necrosis factor-α (TNF-α) therapy using the inflammatory indicators, Juvenile Arthritis Disease Activity Score 27 (JADAS27) and magnetic resonance imaging (MRI).MethodsWe conducted a single-center retrospective study of 134 patients with ERA. We evaluated the effect of anti-TNF therapy on the inflammatory indicators, active joint count, MRI quantitative score, and JADAS27 over 18 months. We used the Spondyloarthritis Research Consortium of Canada (SPARCC) and the Hip Inflammation MRI Scoring System (HIMRISS) scoring systems for hip and sacroiliac joints scoring.ResultsThe average age of onset of children with ERA was 11.62 ± 1.95 years, and they were treated with disease-modifying antirheumatic drugs (DMARDs) combined with biologics (n = 87, 64.93%). There were no differences in HLA-B27 positivity between the biologics and non-biologics treatment groups [66 (49.25%) vs. 68 (50.75%), P > 0.05]. Children who received anti-TNF (71 received etanercept, 13 adalimumab, 2 golimumab, and 1 infliximab) therapy improved significantly. Children with ERA used DMARDs and biologics at baseline (Group A) were followed up to 18 months, and their active joint count (4.29 ± 1.99 vs. 0.76 ± 1.33, P = 0.000), JADAS27 (13.70 ± 4.80 vs. 4.53 ± 4.52, P = 0.000) and MRI quantitative scores (P = 0.001) were significantly lower than those at baseline. Some of the patients (n = 13, 9.70%) were treated with DMARDs at the onset of the disease, but did not show significant improvement (Group B). Abstract Pro-tumoral and immunosuppressive M2-like tumor-associated macrophages (TAMs) contribute to tumor progression, recurrence and distal metastasis. However, current TAMs-modulating therapeutic strategies often encounter challenges including insufficient immune activation, weak antigen presentation ability and unsatisfactory antitumor immune performance. Herein, cyclic RGD peptide functionalized and manganese doped eumelanin-like nanocomposites (RMnMels) are reported for combined hyperthermia-immunotherapy against PC3 prostate cancer. The RMnMels could promote M2-to-M1 macrophage repolarization via scavenging multiple reactive oxygen species and remodeling the immunosuppressive tumor microenvironment. Following near-infrared light irradiation, RMnMels-mediated thermal ablation not only could destroy tumor cells directly, but also elicit the release of damage associated molecular patterns and tumor-associated antigens, provoking robust tumor immunogenicity and strong antitumor immune responses. The results showed that RMnMels could effectively scavenge reactive oxygen species and promote M2-to-M1 macrophage repolarization both in vitro and in vivo. Synergistically enhanced anti-tumor therapeutic efficacy was achieved following single administration of RMnMels plus single round of laser irradiation, evidenced by decreased primary tumor sizes and decreased number of distant liver metastatic nodules. The as-developed RMnMels may represent a simple and high-performance therapeutic nanoplatform for immunomodulation and enhanced antitumor immune responses. Graphical Abstract

The current NRC dietary selenium (Se) requirement (0.15 mg/kg) of broilers is primarily based on growth performance data reported in 1986. Our study aimed to determine optimal dietary Se levels of broilers fed a practical corn-soybean meal diet for the full expression of selenoproteins in various tissues. A total of 384 one-d-old male broilers (n = 8 replicates/diet) were fed a basal corn-soybean meal diet or the basal diet supplemented with 0.1, 0.2, 0.3, 0.4 or 0.5 mg Se/kg in the form of Na2SeO3 for 21 d. BackgroundAnthracycline-induced cardiotoxicity is well-known as a side effect of chemotherapy. Currently, clinical imaging techniques are not capable to detect doxorubicin (DOX)-induced cardiotoxicity before a functional decline. The purpose of this study was to evaluate whether myocardial contrast echocardiography (MCE) can dynamically monitor the cardiac changes in the early stage in the DOX-induced rat model of cardiotoxicity.MethodsA weekly injection of 2.5 mg/kg of DOX was used to generate a rat model of cardiotoxicity. All groups underwent ultrasonic examinations including standard echocardiography, 2D speckle tracking echocardiography (2D-STE), and MCE. Then all rats were sacrificed immediately for histopathological evaluation.ResultsA total of eight control rats and 32 DOX-treated rats were included in the study and grouped according to their treatment period. Decreased quantitative parameters of myocardial blood flow (MBF) (control vs. group 1: 133.31 ± 20.23 dB/s vs. 103.35 ± 21.60 dB/s, P = 0.048) and β (control vs. group 2: 11.17 ± 1.48/s vs. 7.15 ± 1.23/s, P < 0.001) were observed after 2 and 4 weeks of treatment, respectively, while left ventricular global strain (control vs. group 3: −23.67 ± 3.92% vs. −16.01 ± 3.40%, P = 0.002) decreased after 6 weeks of treatment and left ventricular ejection fraction (LVEF) (control vs. group 4: 82.41 ± 3.20% vs. 70.89 ± 9.30%, P = 0.008) decreased after 8 weeks of treatment. The main histopathological features are increased myocardial vacuolization and interstitial fibrosis and decreased myocardial microvessel density.ConclusionCompared with standard echocardiography and 2D-STE, MCE can accurately and non-invasively detect changes in early myocardial perfusion, demonstrating the clinical potential of continuous and dynamic monitoring of DOX-induced cardiotoxicity.

BackgroundArtificial insemination (AI) is an effective reproductive technique to improve the performance of cashmere goats and prevent the spread of diseases, and the quality of the semen determines the success of AI. The potential of Moringa oleifera leaf powder (MOLP) and Moringa oleifera leaf ethanolic extract (MOLE) to improve semen quality has been reported, but the underlying mechanisms remain unclear. For the purpose, 18 mature male cashmere goats were randomly assigned into three groups: the control (CON), MOLP, and MOLE groups. The CON group received distilled water orally; the MOLP group was orally treated with 200 mg/kg body weight (BW) MOLP; and the MOLE group was orally treated with 40 mg/kg BW MOLE.ResultsResults showed that MOLE contained long-chain fatty acids and flavonoids. Treatment with MOLP and MOLE increased the activities of the serum catalase, superoxide dismutase, and glutathione peroxidase (P < 0.05), enhanced the total antioxidant capacity (P < 0.05), and reduced the serum malondialdehyde level (P < 0.05). At the same time, MOLE increased the contents of serum gonadotropin releasing hormone and testosterone (P < 0.05). Moreover, MOLE significantly increased sperm concentration, motility, and viability (P < 0.05). Meanwhile, MOLE raised the Chao1 index (P < 0.05) and altered the composition of the rumen microbiota; it also raised the relative abundance of Treponema (P < 0.05) and Fibrobacter (P < 0.05) and reduced the relative abundance of Prevotella (P < 0.1). Correlation analysis revealed the genus Prevotella was significantly negatively correlated with sperm concentration, as well as sperm motility and viability. Furthermore, MOLE significantly increased the rumen levels of the steroid hormones testosterone and dehydroepiandrosterone (P < 0.05), as well as the polyunsaturated fatty acids (PUFAs) alpha-Linolenic acid, gamma-Linolenic acid, docosapentaenoic acid, and 9-S-Hydroperoxylinoleicacid (P < 0.05).ConclusionsOral MOLE supplementation can improve semen quality by increasing the antioxidant capacity and altering the rumen microbiota and metabolites of cashmere goats. Moreover, the MOLP supplementation could enhance the antioxidant capacity of cashmere goats.

Directly applying brain signals to operate a mobile manned platform, such as a vehicle, may help people with neuromuscular disorders regain their driving ability. In this paper, we developed a novel electroencephalogram (EEG) signal-based driver–vehicle interface (DVI) for the continuous and asynchronous control of brain-controlled vehicles. The proposed DVI consists of the user interface, the command decoding algorithm, and the control model. The user interface is designed to present the control commands and induce the corresponding brain patterns. The command decoding algorithm is developed to decode the control command. The control model is built to convert the decoded commands to control signals. Offline experimental results show that the developed DVI can generate a motion control command with an accuracy of 83.59% and a detection time of about 2 s, while it has a recognition accuracy of 90.06% in idle states. A real-time brain-controlled simulated vehicle based on the DVI was developed and tested on a U-turn road. Experimental results show the feasibility of the DVI for continuously and asynchronously controlling a vehicle. This work not only advances the research on brain-controlled vehicles but also provides valuable insights into driver–vehicle interfaces, multimodal interaction, and intelligent vehicles.

Zinc (Zn) has been shown to attenuate the adverse effects of heat stress on broilers, but the mechanisms involving this process remain unclear. We aimed to investigate possible protective mechanisms of Zn on primary cultured hepatocytes of broiler embryos subjected to heat stress. Three experiments were conducted. In Exp. 1, hepatocytes were treated with 0, 50, 100, 200, or 400 μmol/L added Zn as inorganic Zn sulfate (iZn) for 12, 24 or 48 h. In Exp. 2, cells were exposed to 40 °C (a normal temperature [NT]) and 44 °C (a high temperature [HT]) for 1, 2, 4, 6, or 8 h. In Exp. 3, cells were preincubated with 0 or 50 μmol/L Zn as iZn or organic Zn lysine chelate (oZn) for 8 h under NT, and then incubated with the same Zn treatments under NT or HT for 4 or 6 h. The biomarkers of antioxidative status and heat stress in cells were measured. The use of cold-formed steel (CFS) channel sections with rectangular holes in the web is becoming gradually popular in building structures. However, such holes can result in sections becoming more susceptible to be distortional buckling and display lower load-carrying capacities. This paper presents a total of 44 axially-compressed tests of CFS lipped channel columns with and without rectangular web holes including different hole sizes and cross-sections. The test results show that the specimens were controlled by distortional buckling or interaction of local buckling and distortional buckling. The load-carrying capacities of specimens with rectangular holes were lower than that of specimens without hole. The load-carrying capacities of specimens were gradually decreased with the increasing of dimensions of holes. Then a nonlinear elasto-plastic finite element model (FEM) was developed and the analysis results showed good agreement with the test results. The validated FE model was used to conduct a parametric study involving 16 FEM to investigate the effects of the section, the dimension of the hole, and the number of holes on the ultimate strength of such channels. Furthermore, the formulas to predict the distortional buckling coefficient were developed for the section with holes by using the verified FEM. Finally, the tests and parametric study results were compared against the distortional buckling design strengths calculated in accordance with the developed method. The comparison results show that the proposed design method closely predict the load carrying capacity of CFS channel sections with rectangular web holes.

This research presents a novel, eco-friendly, vortex-assisted liquid–liquid microextraction (VALLME) approach, integrating hydrophobic deep eutectic solvents (DESs) with HPLC for the identification and quantification of nine specific flavonoids in Shanxi aged vinegar (SAV). The parameters of DES-VALLME, including the ratio of trioctylmethylammonium chloride to 1,4-butanediol (1:6), DES volume (150 μL), vortex duration (5 min), the concentration of NaCl (0.40 g), and centrifugation time (10 min), were optimized to achieve the maximum extraction efficiency of target substances. Under these optimal conditions, quantitative analyses performed via HPLC demonstrated a broad linear range of 0.20–50.00 μg/mL and correlation coefficients (r2) greater than 0.9944 for all nine calibration curves. The limits of detection (LOD) and limits of quantitation (LOQ) were 0.09–0.18 μg/mL and 0.30–0.60 μg/mL, respectively, ensuring high sensitivity. The relative standard deviations for intra-day and inter-day variability were within the acceptable range, 2.34–3.77% and 3.04–4.96%, respectively, demonstrating the method’s reliability. The recovery rates ranged from 85.97% to 108.11%, underscoring the method’s precision. This technique exhibited a significant enrichment effect (enrichment factor: 43 to 296) on SAV flavonoids. Notably, the eco-friendliness of this procedure was evaluated using the Analytical Eco-Scale, Green Analytical Procedure Index, and Analytical Greenness Metric. The results suggested that this technique is a viable green alternative to traditional flavonoid determination methods in SAV. In summary, this novel method provides a theoretical basis for assessing flavonoid content in SAV samples and tracing SAV products. This contribution has significant implications for enhancing analytical techniques in food chemistry and environmental science and the sustainable development of the food industry.

Abstract The sharing economy has changed people’s lives. These changes have accelerated the pace of life and improved the quality of life, but have also had some negative effects. For example, in China, in the supply chain of the takeout delivery platform, the service deliverer often violates traffic rules to shorten the delivery time. This phenomenon has aroused widespread concern in society. From the perspective of sustainable operation of platform enterprises and reducing social risks, this issue is worthy of in-depth study. First, this study analyses the optimal decision-making about pricing and service efforts, then discusses the supply chain coordination mechanism under a revenue-sharing contract, and finds that the platform’s service level is related to the efforts of deliverers and the elasticity of demand/service. However, under a traditional revenue-sharing contract, the economic returns are not enough to motivate deliverers to avoid violations of traffic regulations; When the increase of cost caused by the improvement of service level is not fully covered by the revenue sharing from the platform, they have a strong incentive to break the rules even if the optimal coordination can be achieved in the supply chain. Second, this study reconstructs the revenue-sharing contract by adding social responsibility to the objective function of the platform enterprises and designs contingent rewards to regulate behaviours of service deliverers. The results indicate that when the contingent reward factor reaches a certain level, the optimal effort of the service deliverers is constrained within a reasonable range, which can to some extent reduce the occurrence of illegal behaviour. Our findings offer fresh insights on coordination of supply chain in the sharing economy, identify new direction for future research on CSR, and provide managers of platform enterprises with suggestions for regulating the behaviour of partners and balance the economic and social benefits.

Copy number variation (CNV) has been considered to be an important source of genetic variation for important phenotypic traits of livestock. In this study, we performed whole-genome CNV detection on Suhuai (SH) (n = 23), Chinese Min Zhu (MZ) (n = 11), and Large White (LW) (n = 12) pigs based on next-generation sequencing data. The copy number variation regions (CNVRs) were annotated and analyzed, and 10,885, 10,836, and 10,917 CNVRs were detected in LW, MZ, and SH pigs, respectively. Some CNVRs have been randomly selected for verification of the variation type by real-time PCR. We found that SH and LW pigs are closely related, while MZ pigs are distantly related to the SH and LW pigs by CNVR-based genetic structure, PCA, VST, and QTL analyses. A total of 14 known genes annotated in CNVRs were unique for LW pigs. Among them, the cyclin T2 (CCNT2) is involved in cell proliferation and the cell cycle. The FA Complementation Group M (FANCM) is involved in defective DNA repair and reproductive cell development. Ten known genes annotated in 47 CNVRs were unique for MZ pigs. The genes included glycerol-3-phosphate acyltransferase 3 (GPAT3) is involved in fat synthesis and is essential to forming the glycerol triphosphate. Glutathione S-transferase mu 4 (GSTM4) gene plays an important role in detoxification. Eleven known genes annotated in 23 CNVRs were unique for SH pigs. Neuroligin 4 X-linked (NLGN4X) and Neuroligin 4 Y-linked (NLGN4Y) are involved with nerve disorders and nerve signal transmission. IgLON family member 5 (IGLON5) is related to autoimmunity and neural activities. The unique characteristics of LW, MZ, and SH pigs are related to these genes with CNV polymorphisms. These findings provide important information for the identification of candidate genes in the molecular breeding of pigs.

Abstract Background Combining ultrasound imaging with photoacoustic imaging provides tissue imaging with high contrast and resolution, thereby enabling rapid, direct measurements and the tracking of tumour growth and metastasis. Moreover, ultrasound-targeted nanobubble destruction (UTND) provides an effective way to deliver drugs, effectively increasing the content of the drug in the tumour area and reducing potential side effects, thereby successfully contributing to the treatment of tumours. Results In this study, we prepared multifunctional nanobubbles (NBs) carrying indocyanine green (ICG) and paclitaxel (PTX) (ICG-PTX NBs) and studied their applications in ultrasound imaging of prostate cancer as well as their therapeutic effects on prostate cancer when combined with UTND. ICG-PTX NBs were prepared by the mechanical oscillation method. The particle size and zeta potential of the ICG-PTX NBs were 469.5 ± 32.87 nm and − 21.70 ± 1.22 mV, respectively. The encapsulation efficiency and drug loading efficiency of ICG were 68% and 2.52%, respectively. In vitro imaging experiments showed that ICG-PTX NBs were highly amenable to multimodal imaging, including ultrasound, photoacoustic and fluorescence imaging, and the imaging effect was positively correlated with their concentration. The imaging effects of tumour xenografts also indicated that ICG-PTX NBs were of good use for multimodal imaging. Pigs are important biomedical model animals for the study of human neurological diseases. Similar to human aggressive behavior in children and adolescents, weaned pigs also show more aggressive behavior after mixing, which has negative effects on animal welfare and growth performance. The identification of functional single-nucleotide polymorphisms (SNPs) related to the aggressive behavior of pigs would provide valuable molecular markers of the aggressive behavioral trait for genetic improvement program. The Rho GTPase–activating protein 24 (ARHGAP24) gene plays an important role in regulating the process of axon guidance, which may impact the aggressive behavior of pigs. By resequencing the entire coding region, partially adjacent introns and the 5′ and 3′ flanking regions, six and four SNPs were identified in the 5′ flanking region and 5′ untranslated region (UTR) of the porcine ARHGAP24 gene, respectively. Association analyses revealed that nine SNPs were significantly associated with aggressive behavioral traits (p = < 1.00 × 10–4–4.51 × 10–2), and their haplotypes were significantly associated with aggressive behavior (p = < 1.00 × 10–4–2.99 × 10–2). The core promoter region of the ARHGAP24 gene has been identified between −670 and −1,113 bp. Furthermore, the luciferase activity of allele A of rs335052970 was significantly less than that of allele G, suggesting that the transcriptional activity of the ARHGAP24 gene was inhibited by allele A of rs335052970. It was identified that the transcription factor p53 bound to the transcription factor binding sites (TFBSs) containing allele A of rs335052970. In porcine primary neural cells, p53 binds to the target promoter region of the ARHGAP24 gene, reduces its promoter transcriptional activity, and then reduces its messenger RNA (mRNA) and protein expression. The results demonstrated that the ARHGAP24 gene had significant genetic effects on aggressive behavioral traits of pigs. Therefore, rs335052970 in the ARHGAP24 gene can be used as a molecular marker to select the less aggressive pigs.

Background The poor immunogenicity of solid tumors limits the efficacy ofanti-programmed cell death protein 1 (anti-PD1)-based immune checkpoint blockade (ICB); thus, less than 30% of patients with cancer exhibit a response. Currently, there is still a lack of effective strategies for improving tumor immunogenicity.Methods The antitumor effect of ultrasound-stimulated nanobubbles (USNBs) alone and in combination with an anti-PD1 antibody was evaluated in RM1 (prostate cancer), MC38 (colon cancer) and B16 (melanoma) xenograft mouse models. The phenotypes of antigen-presenting cells and CD8+ T cells were evaluated by flow cytometry. Damage-associated molecular pattern (DAMP) release, antigen release and tumor cell necrosis were assessed via western blot, flow cytometry, transmission electron microscopy and confocal microscopy.Results USNB promoted the infiltration and antitumor activity of CD8+ T cells. The combination of USNB and anti-PD1 blockade improved systemic antitumor immunity and resulted in an abscopal effect and long-term immune memory protection after complete tumor remission. Mechanistically, tumor-targeting USNB induced tumor cell necrosis through an ultrasound-mediated cavitation effect, which significantly increased DAMP release and tumor antigen presentation, consequently sensitizing tumors to ICB treatment.Conclusion The administration of USNB increased tumor immunogenicity by remodeling the tumor-immune microenvironment, providing a promising strategy for sensitizing poorly immunogenic solid tumors to immunotherapy in the clinic.

Resveratrol shows promizing anti-inflammatory effects in recent clinical trials, however its function in cardiovascular patients remains conflicting, suggesting there may be new mechanisms underlying its cardioprotective activity. Acute sympathetic stress induces early activation of the NLR family, pyrin domain containing 3 (NLRP3) inflammasome in cardiomyocytes as a critical step for triggering cardiac inflammation. Thus, this study explored targets of resveratrol activity involved in the inhibition of early inflammasome activation in cardiomyocytes following acute sympathetic stress. Network pharmacology was used to analyze common candidate targets in the sympathetic stress pathway, resveratrol activity, and myocardial inflammation and showed the Phosphoinositol 3—kinase (PI3K)/serine threonine protein kinase (Akt) signaling pathway and the target AKT1 may play a critical role. Molecular docking provided support for potential binding of resveratrol on AKT1. Furthermore, the effect of resveratrol on AKT1 activation was determined in cardiomyocytes. resveratrol dose-dependently inhibited AKT1 activation after activation of β-adrenoceptor. The AKT1 inhibitor A-674563 suppressed the activation of the NLRP3 inflammasome in cardiomyocytes following β-adrenoceptor activation, suggesting that AKT1 is a critical regulator molecule upstream of the NLRP3 inflammasome. Consistently, treatment with resveratrol suppressed β-adrenoceptor-mediated NLRP3 inflammasome activation in both cardiomyocytes and mouse hearts, as well as the resultant cardiac inflammation. In conclusion, resveratrol targets AKT1 to inhibit NLRP3 inflammasome activation in cardiomyocytes and cardiac inflammation following acute sympathetic stress. AKT1 is an important target of resveratrol, which should be considered as a treatment option for cardiovascular patients, especially those at risk of acute sympathetic stress.

Malignant tumors are a grave threat to human health. Esophageal squamous cell carcinoma (ESCC) is a common gastrointestinal malignant tumor. China has a high incidence of ESCC, and its morbidity and mortality are higher than the global average. Increasingly, studies have shown that long noncoding RNAs (lncRNAs) play a vital function in the occurrence and development of tumors. Although the biological function of FOXP4-AS1 has been demonstrated in various tumors, the potential molecular mechanism of FOXP4-AS1 in ESCC is still poorly understood. The expression of FOXP4 and FOXP4-AS1 was detected in ESCC by quantitative real-time PCR (qRT–PCR) or SP immunohistochemistry (IHC). shRNA was used to silence gene expression. Apoptosis, cell cycle, MTS, colony formation, invasion and migration assays were employed to explore the biological functions of FOXP4 and FOXP4-AS1. The potential molecular mechanism of FOXP4-AS1 in ESCC was determined by dual-luciferase reporter, RNA immunoprecipitation (RIP) and chromatin immunoprecipitation (ChIP). Here, we demonstrated that FOXP4-AS1 was significantly increased in ESCC tissues and cell lines, associated with lymph node metastasis and TNM staging. Cell function experiments showed that FOXP4-AS1 promoted the proliferation, invasion and migration ability of ESCC cells. The expression of FOXP4-AS1 and FOXP4 in ESCC tissues was positively correlated. Further research found that FOXP4-AS1, upregulated in ESCC, promotes FOXP4 expression by enriching MLL2 and H3K4me3 in the FOXP4 promoter through a “molecular scaffold”. Moreover, FOXP4, a transcription factor of β-catenin, promotes the transcription of β-catenin and ultimately leads to the malignant progression of ESCC. Finally, FOXP4-AS1 may be a new therapeutic target for ESCC.

In order to study the hysteretic behavior and restoring force model of cold-formed thin-walled steel (CFTWS) lipped channel beam-column, a finite element analysis (FEA) model considering the material and geometric nonlinearity and the influence of the initial geometric imperfection under cyclic loading was established by ABAQUS software. Based on the verified FEA model, the hysteretic behavior of CFTWS lipped channel beam-column members with different influencing factors was analyzed by parameterization. The effects of width-to-thickness ratio, axial compression ratio and slenderness ratio on ductility, energy dissipation capacity and stiffness degradation were studied. Based on the results of parameter analysis and the theoretical and regression analysis, the calculation formula of each characteristic point of the trilinear skeleton curve of beam-column member was proposed. Based on the hysteresis curve analysis of beam-column member, the simplified restoring force model (SRFM) of CFTWS lipped channel beam-column was established according to different hysteresis rules for the different buckling modes. The results showed that the SRFM under different buckling modes was in good agreement with the hysteretic curve of FEA, which can better indicate the hysteretic behavior of the member. The proposed SRFM was accurate and applicable, and can provide a fundament for the seismic analysis of the CFTWS lipped channel beam-column.

Objectives The aim of this study was to evaluate the performance of deep learning-based detection and classification of carotid plaque (DL-DCCP) in carotid plaque contrast-enhanced ultrasound (CEUS).Methods and analysis A prospective multicentre study was conducted to assess vulnerability in patients with carotid plaque. Data from 547 potentially eligible patients were prospectively enrolled from 10 hospitals, and 205 patients with CEUS video were finally enrolled for analysis. The area under the receiver operating characteristic curve (AUC) was used to evaluate the effectiveness of DL-DCCP and two experienced radiologists who manually examined the CEUS video (RA-CEUS) in diagnosing and classifying carotid plaque vulnerability. Sepsis is a dysregulated systemic response to infection, and no effective treatment options are available. Acacetin is a natural flavonoid found in various plants, including Sparganii rhizoma, Sargentodoxa cuneata and Patrinia scabiosifolia. Studies have revealed that acacetin potentially exerts anti-inflammatory and antioxidative effects on sepsis. In this study, we investigated the potential protective effect of acacetin on sepsis and revealed the underlying mechanisms using a network pharmacology approach coupled with experimental validation and molecular docking. First, we found that acacetin significantly suppressed pathological damage and pro-inflammatory cytokine expression in mice with LPS-induced fulminant hepatic failure and acute lung injury, and in vitro experiments further confirmed that acacetin attenuated LPS-induced M1 polarization. Then, network pharmacology screening revealed EGFR, PTGS2, SRC and ESR1 as the top four overlapping targets in a PPI network, and GO and KEGG analyses revealed the top 20 enriched biological processes and signalling pathways associated with the therapeutic effects of acacetin on sepsis. Further network pharmacological analysis indicated that gap junctions may be highly involved in the protective effects of acacetin on sepsis. Finally, molecular docking verified that acacetin bound to the active sites of the four targets predicted by network pharmacology, and in vitro experiments further confirmed that acacetin significantly inhibited the upregulation of p-src induced by LPS and attenuated LPS-induced M1 polarization through gap junctions. Taken together, our results indicate that acacetin may protect against sepsis via a mechanism involving multiple targets and pathways and that gap junctions may be highly involved in this process.

The cold-formed steel (CFS) double-lipped equal-leg angle is widely used in modular container houses and cold-formed steel buildings. To study the buckling behavior and bearing capacity design method of the cold-formed steel (CFS) double-lipped equal-leg angle under axial compression, 24 CFS double-lipped equal-leg angles with different sections and slenderness ratios the axial compression were conducted. The test results showed that the distortional buckling occurs for specimens with a small width-to-thickness ratio and small slenderness ratio. The buckling interactive with distortional and global flexural buckling was observed for the specimens with small width-to-thickness ratios and large slenderness ratios. The specimens with large width-to-thickness ratios and small slenderness ratios showed interactive buckling with local and distortion buckling. The specimens with large width-to-thickness ratios and large slenderness ratio developed interactive buckling with local, distortional, and global flexural buckling. The finite element model established by ABAQUS software was used to simulate and analyze the test. The buckling modes and the load-carrying capacities analyzed by the finite element model agreed with the test results, which showed that the developed finite element model was feasible to analyze the buckling and bearing capacity of the CFS double-lipped equal-leg angles. The experimental results were compared with those calculated by the direct strength method in the North American standard and the effective width method in the Chinese standard. The comparisons indicated that the calculated results are very conservative with maximum value 36% and 51% for direct strength method and effective width method, respectively. The coefficient of variation was 0.276 and 0.397, respectively. Finally, the modified direct strength method and the modified effective width method were proposed based on the experimental results. The comparison on the ultimate strength between test results and calculated results by using the modified method showed a good agreement. The modified method can be as a proposed desigh method for the ultimate strength of the CFS double-lipped equal-leg angles under axial compression.

Processing will improve the digestion of corn by ruminant animals. The objectives of this study were to investigate the effects of processing methods (grinding, G; steam flaking, SF; extrusion, E) on the starch properties, nutrient profiles, in vitro ruminal fermentation and molecular structure of corn. Compared with G, SF and E increased (p < 0.05) the starch content, starch gelatinization, ruminal gas production (GP, 0.5–32 h), propionic acid, starch degradability (SD), the area and height of carbohydrate peaks, and decreased (p < 0.05) starch crystallinity, content of crude protein, neutral detergent fiber and acid detergent fiber, ruminal NH3-N, the area and height of amide I and II, α-helix, and β-sheet. The total VFA (24 h, 48 h) tended to be increased by SF and E (p < 0.10). The carbohydrate peak area and height were positively (p < 0.05) correlated with GP (1–24 h) and SD. The protein molecular absorption intensity was negatively correlated with SD (p < 0.05). The change in starch properties, GP (1–24 h) and molecular structure caused by E was greater than SF (p < 0.05). These results indicated that the higher starch gelatinization and lower starch crystallinity of E corn, induced by the high temperature and pressure, enabled more fermentation and digestion in the artificial rumen. The carbohydrate and protein molecular structures were correlated with the nutritional characteristics of corn.

Visible and long-lasting estrus expression of gilts and sows effectively sends a mating signal. To reveal the roles of Long Intergenic Non-coding RNAs (lincRNAs) in estrus expression, RNA-seq was used to investigate the lincRNAs expression of follicular tissues from Large White gilts at diestrus (LD) and estrus (LE), and Chinese Mi gilts at diestrus (MD) and estrus (ME). Seventy-three differentially expressed lincRNAs (DELs) were found in all comparisons (LE vs. ME, LD vs. LE, and MD vs. ME comparisons). Eleven lincRNAs were differentially expressed in both LD vs. LE and MD vs. ME comparisons. Fifteen DELs were mapped onto the pig corpus luteum number Quantitative Trait Loci (QTL) fragments. A protein–protein interaction (PPI) network that involved estrus expression using 20 DEGs was then constructed. Interestingly, three predicted target DEGs (PTGs) (CYP19A1 of MSTRG.10910, CDK1 of MSTRG.10910 and MSTRG.23984, SCARB1 of MSTRG.1559) were observed in the PPI network. A competitive endogenous RNA (ceRNA) network including three lincRNAs, five miRNAs, and five genes was constructed. Our study provides new insight into the lincRNAs associated with estrus expression and follicular development in gilts.
