Background: Rhabdomyosarcoma (RMS) is one of the most common soft tissue sarcomas in children and adolescents, in which PAX3-FOXO1 fusion gene positive patients have very poor prognosis. PAX3-FOXO1 has been identified as an independent prognostic predictor in RMS, with no currently available targeted therapeutic intervention. The novel tyrosine kinase inhibitor anlotinib exhibits a wide range of anticancer effects in multiple types of cancers; however, there have been no relevant studies regarding its application in RMS. Materials and methods: We investigated the effects of PAX3-FOXO1 on the therapeutic efficacy of anlotinib using the CCK-8 assay, flow cytometry, invasion assay, wound healing assay, western blotting, quantitative polymerase chain reaction(qPCR), and xenograft experiments. RNA-seq and co-immunoprecipitation assays were conducted to determine the specific mechanism by which anlotinib regulates PAX3-FOXO1 expression. Results: Anlotinib effectively inhibited RMS cell proliferation and promoted apoptosis and G2/M phase arrest while impeding tumor growth in vivo. Downregulation of PAX3-FOXO1 enhances the antitumor effects of anlotinib. Anlotinib upregulates protein kinase NEK2 and increases the degradation of PAX3-FOXO1 via the ubiquitin-proteasome pathway, leading to a reduction in PAX3-FOXO1 protein levels. Conclusion: Anlotinib effectively inhibited the malignant progression of RMS and promoted degradation of the fusion protein PAX3-FOXO1. Anlotinib could be a targeted therapeutic approach to treat PAX3-FOXO1 fusion-positive RMS.

Summary: Background: Adjunctive newer antiseizure medications (ASMs) are being used in patients with treatment-resistant focal-onset seizures (FOS). An updated network meta-analysis (NMA) was necessary to compile evidence in this critical area. Methods: We systematically searched PubMed, Embase, Cochrane Library, Web of Science, and Scopus from their inception until 17 January 2024, evaluating the efficacy, tolerability, and safety of rufinamide (RUF), brivaracetam (BRV), cenobamate (CNB), eslicarbazepine (ESL), lacosamide (LCM), retigabine (RTG), and perampanel (PER) as adjunctive treatments for FOS. Efficacy outcomes included seizure response and seizure freedom. Tolerability was assessed by discontinuation due to adverse events (AEs). Safety outcomes were evaluated based on the number of patients experiencing at least one AE and serious adverse events (SAEs). This review is registered with PROSPERO (CRD42023485130). Findings: A total of 29 studies involving 11,750 participants were included. For seizure response, all ASMs were significantly superior to placebo, with RTG ranking highest, followed by CNB. Considering dosage, CNB 400 mg/d was top-ranked, followed by RTG 1200 mg/d. For seizure freedom, BRV was highest-ranked, followed by CNB, with BRV 100 mg/d leading, followed by CNB 400 mg/d. Regarding tolerability, LCM 600 mg/d had the lowest ranking, followed by CNB 400 mg/d. For the safety outcome of AEs, ESL 1200 mg/d was ranked lowest, followed by CNB 400 mg/d. Regarding SAEs, LCM 400 mg/d was ranked lowest, followed by RTG 1200 mg/d. Interpretation: ASMs at different dosages have varying efficacy and tolerability profiles. We have provided hierarchical rankings of ASMs for efficacy and safety outcomes. Our findings offer the most comprehensive evidence available to inform patients, families, physicians, guideline developers, and policymakers about the choice of ASMs in patients with treatment-resistant FOS. Vegetation is an important type of land cover. Long-term, large-scale, and high-precision vegetation monitoring is of great significance for ecological environment investigation and regional sustainable development in protected areas. This paper develops a long-term remote sensing monitoring method for vegetation by calculating the normalized difference vegetation index (NDVI) based on the Google Earth Engine (GEE) cloud platform and Landsat satellite remote sensing images. First, based on Landsat long-term satellite images and GEE, the spatiotemporal distribution map of the NDVI is accurately drawn. Subsequently, the NDVI is accurately classified, and the time trend analysis of the NDVI is conducted based on the NDVI mean trend graphs, transition matrices, etc. Then, combined with Moran’s I, high/low clusters, and other methods, the spatial pattern characteristics of the NDVI are analyzed. Finally, climate factors, terrain factors, and anthropologic factors are considered comprehensively. An analysis of the factors affecting the evolution of the NDVI is performed. Taking Zhoushan Island, China, as an example, an experiment is conducted, and the results reveal that (1) the average NDVI exhibits a decreasing trend from 1985 to 2022, decreasing from 0.53 in 1985 to 0.46 in 2022. (2) Regarding vegetation index transitions, the high NDVI areas (0.6–1) exhibit the most substantial shift toward moderately high NDVI values (0.4–0.6), covering an area of 83.10 km2. (3) There is an obvious spatial agglomeration phenomenon in the NDVI on Zhoushan Island. The high-high NDVI clusters and the significant hot spots are predominantly concentrated in the island’s interior regions, while the low-low NDVI clusters and the significant cold spots are mainly situated along the coastal areas. (4) The DEM, slope, and temperature have a greater influence among the single factors on the spatial pattern distribution of the NDVI in 2015. There are significant differences in the spatial pattern distribution of the NDVI between the temperature and DEM, temperature and slope, DEM and precipitation, slope and precipitation, aspect and population, and aspect and gross domestic product (GDP). The DEM and slope, DEM and temperature, and DEM and population are three sets of factors with a strong influence on spatial pattern interaction. This study provides data support for the scientific management of vegetation resources on Zhoushan Island and is of great significance to the sustainable development of the island region.

Neuroblastoma is the most common extracranial solid tumor in children. Patients with neuroblastoma have a poor prognosis. The development of therapy targets and the ability to predict prognosis will be enhanced through further exploration of the genetically related genes of neuroblastoma. The present investigation utilized CRISPR-Cas9 genome-wide screening based on the DepMap database to determine essential genes for neuroblastoma cells’ continued survival. WGCNA analysis was used to determine the progression-related genes, and a prognostic signature was constructed. The signature gene, NCAPG, was downregulated in neuroblastoma cells to explore its impact on various cellular processes. This research used DepMap and WGCNA to pinpoint 45 progression-related essential genes for neuroblastoma. A risk signature comprising NCAPG and MAD2L1 was established. The suppression of NCAPG prevented neuroblastoma cells from proliferating, migrating, and invading. The results of flow cytometric analysis demonstrated that NCAPG inhibition caused cell cycle arrest during the G2 and S phases and the activation of apoptosis. Additionally, NCAPG downregulation activated the p53-mediated apoptotic pathway, inducing cell apoptosis. The present work showed that NCAPG knockdown reduced neuroblastoma cell progression and may serve as a basis for further investigation into diagnostic indicators and therapy targets for neuroblastoma.

Abstract Background Circular RNA (circRNA), producing by special selective splicing, was widely expressed in the cytoplasm of eukaryotic cells as a newly non-coding RNAs. It played different roles in a variety of diseases including cancer and performed different functions. Nonetheless, reports on the specific function of circRNA in pancreatic cancer (PC) were still rarely so far. In particular, the role of circSEC24A in PC remains unclear. Methods Real-time fluorescent quantitative PCR was used to evaluate the expression level of circSEC24A in pancreatic cancer tissues and cell lines. Furthermore, we used some functional experiments, such as EDU and Transwell assays, to explore the effects of circSEC24A on the proliferation and invasiveness of pancreatic cancer. Finally, the corresponding relationship among circSEC24A, miR-606 and TGFBR2 was explored by dual luciferase reporter and other mechanism studies. Results The expression of circSEC24A in both pancreatic cancer tissues and cell lines was evidently up-regulated. Furthermore, knockdown of circSEC24A significantly inhibited the proliferative, migration and invasive capacity of pancreatic cancer cells, whereas miR-606 inhibitor obviously counteracted these effects. Further study confirmed that circSEC24A alleviated suppression on target TGFBR2 expression by directly sponging miR-606 and then influenced the tumorigenesis of pancreatic cancer. Conclusions These findings indicated that the progression of pancreatic cancer can be driven by circSEC24A influencing miR-606/TGFBR2 axis. Therefore, circSEC24A might be used as a critical biomarker influencing the early diagnosis and prognosis of pancreatic cancer.

Abstract In this paper, laser welding-brazing of TC4 Titanium (Ti) alloy and Al2O3 ceramic dissimilar material was carried. The results showed that the Ti alloy and Al2O3 were joined by melting filler metal when the laser was concentrated in the Ti alloy side of the joint. The joint with one fusion weld and one brazed weld separated by remaining unmelted Ti alloy. Laser beam offset the Ti alloy 1.5 mm, Ti alloy would not be completely melted in joint. Through heat conduction, the filler metal melted occurred at the Ti-ceramic interface. A brazed weld was formed at the Ti-ceramic interface with the main microstructure of β-CuZn + Ti2Zn3, β-CuZn and Al2Cu + β-CuZn. The joint fractured at the brazed weld with the maximum tensile strength of 169 MPa.

Abstract Background The underlying mechanism behind the tumorigenesis and progression of pancreatic cancer is not clear, and treatment failure is generally caused by early metastasis, recurrence, drug resistance and vascular invasion. Exploring novel therapeutic regimens is necessary to overcome drug resistance and improve patients outcomes. Methods Functional assays were performed to investigate the role of [6]-Paradol (6-P) in proliferation and metastasis of pancreatic cancer in vitro and in vivo. The interaction between EGFR and 6-P was tested by KEGG enrichment analysis and molecular docking analysis. qRT-PCR was performed to detect the mRNA expression of EGFR in 6-P treated groups. Involvement of the PI3K/AKT pathway was measured by western blotting. Results 6-P significantly suppressed pancreatic cancer cell proliferation and metastasis. KEGG enrichment analysis and molecular docking analysis suggested that there existed certain interaction between EGFR and 6-P. In addition, 6-P obviously decreased EGFR protein expression level but did not change the mRNA expression level of EGFR. 6-P could induce degradation of EGFR through decreasing the protein stability of EGFR and enhancing the ubiquitin-mediated proteasome-dependent degradation, 6-P-mediated EGFR degradation led to inactivation of PI3K/AKT signaling pathway. However, ectopic expression of EGFR protein resulted in resistance to 6-P-mediated inactivity of PI3K/AKT signaling and inhibition of malignant phenotype of pancreatic cancer. Inversely, erlotinib could enhance the 6-P-mediated anticancer activity. Conclusion Our data indicated that 6-P/EGFR/PI3K/AKT signaling axis might become one of the potential therapies for the treatment of pancreatic cancer.

Abstract LncRNAs play essential regulatory roles in pancreatic cancer (PC) tumorigenesis and progression. We aimed to investigate the role of lncRNA CERS6-AS1 in PC. CERS6-AS1 expression was determined in PC tissues and cell lines by PCR analysis. The roles of CERS6-AS1 on proliferation, migration, invasion, and epithelial to mesenchymal transition (EMT) were confirmed via CCK-8 assay, EDU assay, transwell assay, wound healing assay, and western blot assay. Besides, the interaction between CERS6-AS1 and their target genes was verified by luciferase report assays and RIP assays. Animal assays and clinical data analysis were performed to validate the functions in vivo. We found that lncRNA CERS6-AS1 was highly expressed in PC tissues and cells. Additionally, high expression of CERS6-AS1 was obviously associated with poor prognosis. Functional assays demonstrated that CERS6-AS1 downregulation significantly inhibited PC cell growth and migration. Moreover, CERS6-AS1 exerted as a molecular sponge for miR-217-5p (miR-217), and miR-217 was confirmed as a potential target of CERS6-AS1. Subsequently, miR-217 suppressed PC cell proliferation and metastasis by directly targeting YWHAG, which interacted with RAF1 and promoted its phosphorylation, leading to RAF1-mediated ERK signaling activation and translocation of phosphorylated ERK from the cytoplasm to the nucleus. Mechanically, CERS6-AS1 silencing significantly inhibited PC cell proliferation and metastasis via a miR-217/YWHAG/RAF1 signaling axis. CERS6-AS1 exerts as a carcinogen in PC to promote malignant features and behaves as a competitive endogenous RNA for miR-217. We identified CERS6-AS1 as a potential biomarker or therapeutic target to improve PC diagnosis and treatment outcomes.

BackgroundDelayed post-hypoxic leukoencephalopathy (DPHL) is a demyelinating syndrome that occurs days to weeks after the brain has recovered from a coma. It is caused by the period of hypoxia and is characterized by mental disorders, extrapyramidal system symptoms, and motor changes. Common causes include cardiogenic shock, severe anemia, massive blood loss, and poisoning. Poisoning, mostly resulting from intoxication with carbon monoxide and several narcotic drugs, has been reported to be a cause of DPHL. There are only a few reports of DPHL due to nitrite poisoning in literature. We report DPHL in a patient following nitrite poisoning and a review of the literature in this context.Case PresentationA 64-year-old man presented with dizziness and nausea without vomiting. He later went into a coma after consuming a spare rib soup. After blood gas analysis, we suspected nitrite poisoning combined with metabolic acidosis, hypoxemia, and electrolyte imbalance. He gradually showed neurologic recovery to premorbid baseline after intravenous administration of methylene blue (40 mg) and symptomatic treatment. Two months later, the patient's cerebral magnetic resonance imaging (MRI) showed signs that are compatible with injury, with the patient in late stages of mental decline.ConclusionNitrite poisoning can cause DPHL. There is a period of intermittent recovery between the time of poisoning and the development of DPHL, but the specific pathogenesis and treatment are still unclear.

As two important industrial manufacturing materials, ceramics and metals have their own advantages and disadvantages in terms of physical, chemical, and mechanical properties. The field of material manufacturing has witnessed efforts to develop technical processes that can properly combine these two alloys, aiming to effectively use their respective advantages. The welding technology for ceramics and metals, as a research topic with broad prospects, is comprehensively and deeply analyzed in this review. The current research progress in this field is analyzed from different process perspectives such as fusion welding, brazing welding, diffusion welding. By using appropriate intermediate materials between these two materials, the residual stress and brittle intermetallics near the interface of the transition joint can be minimised by solving the thermal expansion mismatch, reducing the bonding temperature and pressure.

In this work, Cu was used as an interlayer to prevent the formation of these brittle Ti-Fe intermetallics when joining Ti alloy to stainless steel (SS). Microstructures of the joint were analyzed by optical microscopy, scanning electron microscopy, transmission electron microscope and X-ray diffraction. The tensile strengths of the joint were also measured. The laser was focused on the SS side of the joint, which joined the SS and Cu by fusion welding. At SS-Cu interface, a weld zone was formed due to dilution of the Cu and mixing with the SS. At the Ti alloy-Cu interface, a eutectic reaction was responsible for joining. The presence of unmelted Cu interlayer ensured that crack free welds were obtained and no brittle Ti-Fe intermetallics were observed. Tensile strength of the joint can reach to 320 MPa. Laser welding-brazing of TC4 Titanium (Ti) alloy to 304 stainless steel (SS) has been applied using 38Zn-61Cu alloy as filler metal. Microstructures of the joints were studied using scanning electron microscopy (SEM), energy dispersive spectroscopy (EDS) and X-ray diffraction (XRD). Mechanical properties of the joints were evaluated by performing tensile tests. The temperature field and stress field distributed in laser welding based on SS-Ti alloy joint were dynamically simulated using the ANSYS in this study. A new welding process for SS-Ti alloy joint was introduced on the basis of the controlling the formation of Ti-Fe intermetallics in the joint. One process was one pass welding involving creation of a joint with one fusion weld and one brazed weld separated by remaining unmelted SS. When laser beam on the SS side was 1.5 mm, SS would not be completely melted in joint. Through heat conduction, the filler metal melted occurred at the SS-Ti alloy interface. A brazed weld was formed at the SS-Ti alloy interface with the main microstructure of (Fe, Zn)+Fe3Zn7, β-CuZn and β-CuZn + Ti2Zn3. The joint fractured at the brazed weld with the maximum tensile strength of 210 MPa. By comparing the simulation results with the corresponding experimental findings, the validity of the numerical model is confirmed. p53/p21 signaling plays a vital role in pancreatic cancer (PC) progression. ZWINT was shown to function as an oncoprotein in the progression of multiple cancers. However, the involvement of ZWINT and p53 activation in the progression of PC remains poorly understood. Bioinformatics and tissue array chip analyses were performed to evaluate ZWINT expression in pancreatic cancer. ZWINT mRNA and protein expression were evaluated in normoxia and hypoxia. CHIP was used to evaluate HIF1α interaction with the ZWINT promoter. CCK8, colony formation, EDU, and cell cycle analysis were used to examine PC cell proliferation. Immunoprecipitation and immunofluorescence were used to examine the interaction of ZWINT, MDM2, and p53. p53 activity was evaluated by q-PCR and luciferase assay. Protein degradation and ubiquitination assays were used to analyze the role of ZWINT in p53 ubiquitination. ZWINT was overexpressed in pancreatic cancer and induced in hypoxia. ZWINT promoted pancreatic cancer growth and cell cycle progression. Bioinformatic analysis revealed that ZWINT may regulate the p53 signal pathway. ZWINT interacts with p53 and promotes its ubiquitination and degradation. ZWINT promoted proliferation via p53/p21. Immunohistochemistry of clinical specimens revealed that that ZWINT expression was significantly negatively correlated with p53/p21. Our data showed that hypoxia regulates the expression of ZWINT, which activated p53/p21 signaling pathway to promote PC growth.

In this work, the thermogravimetry–Fourier transform infrared spectroscopy (TG–FTIR) and gas chromatography–mass spectrometry (GC–MS) techniques are used to investigate the thermal degradation behavior of polypropylene (PP) with 20 wt.% diatomite (DM). The initial decomposition temperature of these blends was 17 °C lower than that of pristine PP, and more olefin degradation products were formed during the pyrolysis process under Ar atmosphere. These results could be attributed to the catalytic effects of DM on the degradation of PP and the changes of PP chain scission pathways around the particles (more β scission happened via the secondary radical transfer). These olefins could be caught by DM through the Si–O–C bond formed during the heat–treatment around 400~500 °C. The formation of the cross–linked structure could facilitate the growth of graphene during a high–temperature graphitization process.

Surface charge polarity and density influence the immune clearance and cellular uptake of intravenously administered lipid nanoparticles (LNPs), thus determining the efficiency of their delivery to the target. Here, we modified the surface charge with ascorbyl palmitate (AsP) used as a negatively charged lipid. AsP-PC-LNPs were prepared by dispersion and ultrasonication of AsP and phosphatidylcholine (PC) composite films at various ratios. AsP inserted into the PC film with its polar head outward. The pKa for AsP was 4.34, and its ion form conferred the LNPs with negative surface charge. Zeta potentials were correlated with the amount and distribution of AsP on the LNPs surface. DSC, Raman and FTIR spectra, and molecular dynamics simulations disclosed that AsP distributed homogeneously in PC at 1–8% (w/w), and there were strong hydrogen bonds between the polar heads of AsP and PC (PO2−), which favored LNPs’ stability. But at AsP:PC > 8% (w/w), the excessive AsP changed the interaction modes between AsP and PC. The AsP–PC composite films became inhomogeneous, and their phase transition behaviors and Raman and FTIR spectra were altered. Our results clarified the mechanism of surface charge modification by AsP and provided a rational use of AsP as a charged lipid to modify LNP surface properties in targeted drug delivery systems. Furthermore, AsP–PC composites were used as phospholipid-based biological membranes to prepare paclitaxel-loaded LNPs, which had stable surface negative charge, better tumor targeting and tumor inhibitory effects.

Multigraphene was prepared via a one-pot pyrolysis method using polypropylene (PP) as the carbon source and diatomite (DM) as the catalyst. The obtained graphene had 4–6 layers and a D/G intensity ratio of 0.70 and a 2D/G intensity ratio of 0.72, indicating a high degree of graphitization. When the pyrolysis temperature was higher than 850 °C under argon, the graphene yield was greatly dependent on the DM content. The highest graphene yield of 25.86% was obtained by pyrolysis of PP with 30 wt.% DM at the temperature of 1000 °C. A catalytic effect of DM and infusible cross-linking structure formation were proposed to explain the possible mechanism of graphene growth during the pyrolysis of the DM/PP composites.

The long propagation delay in underwater acoustic channels has attracted tremendous attentions in designing Medium Access Control (MAC). The low acoustic propagation speed and wide area of the acoustic communication range led to a wide range of variations in the propagation delay. This paper identifies an important characteristic of two-scale delay variations by field test results. We carry out simulations to study the impact of delay variations on MAC, and the results suggest a slot length adaptation scheme for the handshake and slotting based MAC. We further model an absorbing Markov chain to derive the closed-form equation for the throughput of MAC with adaptive slot length. Both the analytical and simulation results show that our proposed slot length adaptation improves significantly the throughput of MAC in underwater acoustic networks. Particularly, the Slotted-FAMA with an adaptive slot length achieves more than double the throughput than the Slotted-FAMA with a fixed slot length in a network with six nodes.

The metabotropic glutamate receptors (mGluRs) are known as both synaptic receptors and taste receptors. This feature is highly similar to the Property and Flavor theory of Traditional Chinese medicine (TCM), which has the pharmacological effect and flavor. In this study, six ligand based pharmacophore (LBP) models, seven homology modeling models, and fourteen molecular docking models of mGluRs were built based on orthosteric and allosteric sites to screening potential compounds from Traditional Chinese Medicine Database (TCMD). Based on the Pharmacopoeia of the People’s Republic of China, TCMs of compounds and their flavors were traced and listed. According to the tracing result, we found that the TCMs of the compounds which bound to orthosteric sites of mGluRs are highly correlated to a sweet flavor, while the allosteric site corresponds to a bitter flavor. Meanwhile, the pharmacological effects of TCMs with highly frequent flavors were further analyzed. We found that those TCMs play a neuroprotective role through the efficiencies of detumescence, promoting blood circulation, analgesic effect, and so on. This study provides a guide for developing new neuroprotective drugs from TCMs which target mGluRs. Moreover, it is the first study to present a novel approach to discuss the association relationship between flavor and the neuroprotective mechanism of TCM based on mGluRs.

Adlay (Coix larchryma-jobi L.) was the commonly used Traditional Chinese Medicine (TCM) with high content of seed storage protein. The hydrolyzed bioactive oligopeptides of adlay have been proven to be anti-hypertensive effective components. However, the structures and anti-hypertensive mechanism of bioactive oligopeptides from adlay were not clear. To discover the definite anti-hypertensive oligopeptides from adlay, in silico proteolysis and virtual screening were implemented to obtain potential oligopeptides, which were further identified by biochemistry assay and molecular dynamics simulation. In this paper, ten sequences of adlay prolamins were collected and in silico hydrolyzed to construct the oligopeptide library with 134 oligopeptides. This library was reverse screened by anti-hypertensive pharmacophore database, which was constructed by our research team and contained ten anti-hypertensive targets. Angiotensin-I converting enzyme (ACE) was identified as the main potential target for the anti-hypertensive activity of adlay oligopeptides. Three crystal structures of ACE were utilized for docking studies and 19 oligopeptides were finally identified with potential ACE inhibitory activity. According to mapping features and evaluation indexes of pharmacophore and docking, three oligopeptides were selected for biochemistry assay. An oligopeptide sequence, NPATY (IC50 = 61.88 ± 2.77 µM), was identified as the ACE inhibitor by reverse-phase high performance liquid chromatography (RP-HPLC) assay. Molecular dynamics simulation of NPATY was further utilized to analyze interactive bonds and key residues. ALA354 was identified as a key residue of ACE inhibitors. Hydrophobic effect of VAL518 and electrostatic effects of HIS383, HIS387, HIS513 and Zn2+ were also regarded as playing a key role in inhibiting ACE activities. This study provides a research strategy to explore the pharmacological mechanism of Traditional Chinese Medicine (TCM) proteins based on in silico proteolysis and virtual screening, which could be beneficial to reveal the pharmacological action of TCM proteins and provide new lead compounds for peptides-based drug design.

The acquisition of digital regional-scale information and ecological environmental data has high requirements for structural texture, spatial resolution, and multiple parameter categories, which is challenging to achieve using satellite remote sensing. Considering the convenient, facilitative, and flexible characteristics of UAV (unmanned air vehicle) remote sensing technology, this study selects a campus as a typical research area and uses the Pegasus D2000 equipped with a D-MSPC2000 multi-spectral camera and a CAM3000 aerial camera to acquire oblique images and multi-spectral data. Using professional software, including Context Capture, ENVI, and ArcGIS, a 3D(three-dimensional) campus model, a digital orthophotomap, andmultispectral remote sensing map drawing are realized, and the geometric accuracy of typical feature selection is evaluated. Based on a quantitative remote sensing model, the campus ecological environment assessment is performed from the perspectives of vegetation and water body.
