Introduction: The increase in high-fat diet (HFD)-induced obesity and food allergy leads to an assumption that the 2 are related. This study aims to (1) systematic verification of HFD-induced obesity aggravates food allergy and (2) explore the correlation and molecular mechanisms of HFD-induced obesity promotes food allergy. Methods: Female BALB/c mice are divided into the control group (control), the ovalbumin (OVA)-sensitized group (OVA), the HFD-induced obesity group (HFD), and HFD-induced allergic obesity group (HFD + OVA). Results: In vivo data showed that HFD feed enhance clinical symptoms and intestinal mucosa villi shed on allergic mice. Moreover, we found that HFD and OVA irritation enhanced levels of mast cell degranulation and Th2 humoral response. Additionally, Western blot analysis showed the potentiation of peroxisome proliferator-activated receptor γ (PPAR γ) remarkably reduced on intestinal in HFD and OVA group, thereby inhibiting the expression of nuclear factor kappa B (NF-κB)/PPAR γ signal the phosphorylation of NF-κB P65. Conclusions: Overall, our results suggest that HFD-induced obesity is a potential risk factor for food allergy, which related to intestinal barrier destruction and inflammation through the PPAR γ/NF-κB signaling pathway.

In this study, we experimentally evaluate the ion transportation through a cone guide target, which accelerates ions up to MeV energies via target normal sheath acceleration, and transports them onto the position of imploding fuel in the fast ignition scenario of nuclear fusion. We measured the electric and magnetic fields (EM-fields) induced by return current streaming along the cone wall by proton radiography, and we report that the EM-fields are predominantly induced within a temporal window up to 30 ps after the laser injection. The magnitude of the electric field is maximized around 13 ps, reaching $4.0\times 10^{10} \mathrm {V}\ \mathrm {m}^{-1}$ , when the magnetic field is below 200 T. The present scheme provides insights on the EM-fields evaluation in the time region that is difficult to treat with simulations due to the computing resources.

In recent decades, the prevalence of food allergy has increased worldwide; however, a comprehensive estimate of the prevalence of food allergy and allergens in China is not yet available.By searching the English databases PubMed, Embase, Cochrane Library, and Chinese databases CNKI, Wanfang Data, and VIP Chinese epidemiological studies on food allergy, the probability of food allergy in China and related influencing factors were determined.The findings of this study provide up-to-date estimates of the prevalence of food allergy rates in China in terms of age, gender, and the eight major food allergens.

In this paper we study photon emission in the interaction of the laser beam with an under-dense target and the attached reflecting plasma mirror. Photons are emitted due to the inverse Compton scattering when accelerated electrons interact with a reflected part of the laser pulse. The enhancement of photon generation in this configuration lies in using the laser pulse with a steep rising edge. Such a laser pulse can be obtained by the preceding interaction of the incoming laser pulse with a thin solid-density foil. Using numerical simulations we study how such a laser pulse affects photon emission. As a result of employing a laser pulse with a steep rising edge, accelerated electrons can interact directly with the most intense part of the laser pulse that enhances photon emission. This approach increases the number of created photons and improves photon beam divergence.

We previously reported that polyploid giant cancer cells (PGCCs) exhibit cancer stem cell properties and can generate daughter cells with the epithelial-mesenchymal transition phenotype. This study investigated the role of PGCC formation in the prognostic value of neoadjuvant chemoradiation therapy (nCRT) in locally advanced rectal cancer (LARC). The morphological characteristics were observed in patients with LARC after nCRT. Colorectal cancer cell lines were treated with irradiation or chemotherapeutic drugs, and the metastasis-related proteins were detected. 304 nCRT cases and 301 paired non-nCRT cases were collected for analysis. More PGCCs and morphologic characteristics related to invasion and metastasis appeared in tumor tissue after nCRT. Irradiation or chemicals could induce the formation of PGCCs with daughter cells exhibiting strong migratory, invasive, and proliferation abilities. In patients after nCRT, pathologic complete remission, partial remission, stable disease, and progressive disease were observed in 29 (9.54%), 125 (41.12%), 138 (45.39%), and 12 (3.95%) patients, respectively. Mucinous adenocarcinomas (MCs) occurred more frequently in nCRT than in non-nCRT patients (χ2 = 29.352,P=0.001), and the prognosis in MC patients was worse than that in non-MC patients (χ2 = 24.617,P=0.001). The difference in survival time had statistical significance for 60 days (χ2 = 5.357,P=0.021) and 70 days (χ2 = 18.830,P=0.001) rest interval time. On multivariable analysis, 60 days rest interval, Duke’s stage, and recurrence and/or distant metastasis remained significant predictors of survival. In conclusion, irradiation or chemicals induce the formation of PGCCs and PGCCs produce daughter cells with strong migration and invasion abilities after a long incubation period. Appropriate rest interval (incubation period) is very important for patients with LARC who will receive nCRT.

Purpose: Most colorectal cancers (CRCs) show positive immunohistochemical (IHC) staining for CK20 and negative staining for CK7. However, in clinical settings, some CRCs show positive IHC staining for CK7, and the clinicopathological significance of this needs to be studied. This study investigated the clinicopathological significance of CK7 positivity in CRCs. Materials and Methods: A total of 178 patients with CRC were used to study the clinicopathological significance of CK7 positivity. Western blotting and immunocytochemical (ICC) staining were used to compare the expression levels of CK7 before and after CoCl2 treatment. Results: CK7 expression was associated with the location, differentiation, lymph node metastasis, and the Dukes' stage of CRCs. CK7 positive cells were mainly distributed at the edge of cancer nests, at the invasion front, as single stromal polyploid giant cancer cells (PGCCs), in tumor buds, in intravascular tumor emboli, and in a micropapillary pattern. Results of ICC staining showed that CK7 expression was almost negative in LoVo and HCT116 before CoCl2 treatment. After CoCl2 treatment, the PGCCs and their daughter cells of LoVo and HCT116 yielded positive results in CK7 ICC staining. Results of western blotting also confirmed that there was higher CK7 expression in LoVo and HCT116 after CoCl2 treatment than in the control. Conclusion: CRC cells expressing CK7 may have strong invasive and metastatic abilities. Some metastasis-related morphological characteristics in CRCs including the invasion front, micropapillary pattern, tumor emboli, and single stromal PGCCs associated with CK7 positive expression.

Electron–positron pair production via Breit–Wheeler process requires laser intensities approaching 1024 W cm−2 due to the small cross-section. Here, we propose a mechanism for brilliant γ-ray emission and dense GeV pairs creation accompanied with high-harmonic generation by using plasma mirror and an ultra short pulse with the intensity of 3 × 1023 W cm−2. The laser is reflected by the solid surface after propagating tens of microns in a near-critical density plasma and breaks into short wave packets. The intensity of the reflected high order harmonic field is enhanced by the focusing and compression effects from the deformed oscillating mirror. The radiation trapped electrons emit γ-photons while colliding with the reflected attosecond pulses. The peak intensity of the γ-ray reaches 0.74 PW with the brilliance of 2 × 1024 s−1 mm−2 mrad−2 (0.1%BW)−1 (at 58 MeV). A GeV positron beam is obtained with density of 4 × 1021 cm−3 and a particle number of 5.6 × 109. Laser technology is rapidly developing to the point where pulse power and intensity are expected to reach such levels that new physical processes will be able to be studied for the first time. Using simulation the authors theoretically investigate the generation of high brilliance gamma rays and electron-positron pairs during extreme intense laser interaction with a specific target.

The aim of this study was to investigate the effect and underlying mechanisms of short-term high-intensity sound exposure on guinea pigs to mimic the effects of non-lethal anti-riot weapons. A total of 92 male adult guinea pigs were exposed to high-intensity sound at 0 dB, 110 dB and 130 dB for 5 min. Basic clinical observation, repellent behaviour detection, peripheral blood routine examination, serum cortisol detection and hearing ability assessment were performed to analyse the functional changes after high intensity sound exposure. Meanwhile, routine haematoxylin and eosin staining, scanning electron microscopy and transmission electron microscopy were used to observe the structure of the cochlear tissue. To investigate the mechanisms underlying the tissue changes, the levels of apoptosis and caspase 3, 8 and 9 were detected using TUNEL staining and immunohistochemistry. After short-term exposure to high-intensity sound, the guinea pigs exhibited fear and agitation, increased repulsive behaviour, high serum cortisol and an increase in auditory threshold. The inner hair cells and outer hair cells exhibited degeneration. In addition, apoptosis was observed in the cochlear tissue. After short‑term exposure to high-intensity sound, the guinea pigs exhibited not only stress reactions but also impaired hearing and signs of hair cell degeneration. Apoptosis in the cochlear tissue may play an important role in the functional and structural injuries caused by high-intensity sound.

As a ligand-activated transcription factor, peroxisome proliferator-activated receptor gamma (PPAR-γ) plays a crucial role in allergic inflammation. Recently, the nuclear factor kappa B (NF-κB) pathway and PAK1 have been indicated to be associated with allergic diseases. However, the effect of PPAR-γ on NF-κB and PAK1 production in food allergies is not known. This study aims to 1) systematically validate that the activation of PPAR-γ attenuates allergic reactions and 2) elucidate the mechanism by which PPAR-γ regulates mast cell degranulation. Brown Norway rats were separated into control, ovalbumin, ovalbumin + rosiglitazone, and ovalbumin + GW9662 groups. In vivo experiments demonstrated that rosiglitazone administration markedly inhibited the clinical symptoms and the serum levels of immunoglobulins E and G1. In addition, cytokine release was regulated by activated PPAR-γ and characterized by increased levels of IFN-γ and decreased levels of IL-4, IL-5, and TNF-α. Our data showed that activated PPAR-γ has the potential to alleviate food allergies by enhancing intestinal mucosal integrity and tight junctions. Moreover, we found that PPAR-γ activation inhibited mast cell degranulation both in vivo and in vitro. Our in vitro findings also showed that the activated PPAR-γ signal could inhibit PAK1 phosphorylation and the expression of p65. Furthermore, the interaction between p65 and p-PAK1 during ovalbumin treatment was attenuated after PPAR-γ activation. Collectively, these results demonstrate that PPAR-γ is an important regulator of mast cell degranulation and the Th2-type response, which sheds new light on the importance of PPAR-γ in food allergies.

Dynamic mitigation for the tearing mode instability in the current sheet in collisionless plasma is demonstrated by applying a wobbling electron current beam. The initial small amplitude modulations imposed on the current sheet induce the electric current filamentation and the reconnection of the magnetic field lines. When the wobbling or oscillation motion is added from the electron beam having a form of a thin layer moving along the current sheet, the perturbation phase is mixed and consequently the instability growth is saturated remarkably, like in the case of the feed-forward control.

The constant-speed straight-line propagation in free space is a basic characteristic of light. Recently, several novel spatiotemporal coupling methods, for example, flying focus (or named sliding focus), are developed to control light propagation including velocity and direction. In the method of flying focus, where temporal chirp and longitudinal chromatism are combined to increase the degree of freedom for coherent control, tunable-velocities and even backward-propagation have been demonstrated. Herein, we studied the transverse and longitudinal effects of the flying focus in space and time, respectively, and found in a specific physics interval existing an unusual reciprocating propagation that was quite different from the previous result. By significantly increasing the Rayleigh length in space and the temporal chirp in time, the newly created flying focus can propagate along a longitudinal axis firstly forward, secondly backward, and lastly forward again, and the longitudinal spatial resolution for a clear reciprocation flying focus improves with increasing the temporal chirp. When this new type of light is applied in the radiation pressure experiment, a reciprocating radiation-force can be produced in space-time accordingly. This finding further extends the control of light and might enable important potential applications.

Magnetic reconnection driven by laser plasma interactions attracts great interests in the recent decades. Motivated by the rapid development of the laser technology, the ultra strong magnetic field generated by the laser-plasma accelerated electrons provides unique environment to investigate the relativistic magnetic field annihilation and reconnection. It opens a new way for understanding relativistic regimes of fast magnetic field dissipation particularly in space plasmas, where the large scale magnetic field energy is converted to the energy of the nonthermal charged particles. Here we review the recent results in relativistic magnetic reconnection based on the laser and collisionless plasma interactions. The basic mechanism and the theoretical model are discussed. Several proposed experimental setups for relativistic reconnection research are presented.

A target using a paisley pattern generates 100-kT-level magnetic fields. Laser irradiation induces local charge separation on the target, which creates surface currents along the concave surface, generating a magnetic field. For a laser intensity of [Formula: see text], the target generates a 150-kT magnetic field. We developed a simple model to describe the magnetic field as a function of the laser intensity and target radius. A double paisley configuration extends the lifetime of the magnetic field to the picosecond scale. The paisley design generates comparable results even if it is simplified. Thus, it is a robust and modular target suitable for magnetic field applications such as 100-kT magnetic field generation and magnetic reconnection.

Through an investigation in 50 cities, the authors conclude that company’s brand and service quality are key influence factors in consumer satisfaction. Company CC gets the best customer satisfaction, while BB and FF get the lowest result. Customers prefer the product integrity, ease of ordering, overall image and scope of delivery. They give the worst experience in the service characteristics.

Unusual position exchange among ions constituting a swift cluster was found in a high-energy-density plasma. In this paper, the peculiar ion behavior appears in a Si cluster, moving with a speed of ~0.2c (c: speed of light) in a solid Al plasma in the context of the cluster-ion beam driven inertial confinement fusion (ICF). The ion position exchange behavior of cluster ions may influence the cluster-ion beam ICF (CIF) target structure design. In this paper we discuss a possibility of cluster-ion beams as the energy driver in CIF. In conclusions, the present work suggests that the following two points in CIF: (1) The unusual ion position exchange of the cluster ions would not induce a remarkable change in the ion stopping range in CIF targets, and (2) the swift cluster ions may not provide a significant vicinage effect on the stopping range.

Singularities in multi-stream flows of relativistic plasmas can efficiently produce coherent high-frequency radiation, as exemplified in the concepts of the Relativistic Flying Mirror [Bulanov et al., Phys. Rev. Lett. 91, 085001 (2003)] and Burst Intensification by Singularity Emitting Radiation [Pirozhkov et al., Sci. Rep. 7, 17968 (2017)]. Direct observation of these singularities is challenging due to their extreme sharpness (tens of nanometers), relativistic velocity, and transient non-local nature. We propose to use an ultrafast (a few light cycles) optical probe for identifying relativistic plasma singularities. Our estimations and Particle-in-Cell simulations show that this diagnostic is feasible.

A dynamic mitigation is presented for sausage and kink instability growths of a z-current driven magnetized plasma column. We have proposed a dynamic mitigation method based on a phase control to smooth plasma non-uniformities and to mitigate the instability growth in perturbed plasma systems. In this paper we found that a wobbling motion of the z-current electron axis induces a phase-controlled perturbation, so that the growths of the sausage and kink instabilities are successfully mitigated. In general, plasma instabilities emerge from perturbations, and the perturbation phase is normally unknown. However, if the perturbation phase is known or actively imposed by, for example, a designed driver wobbling behavior, the instability growth would be controlled and mitigated by a superimposition of the perturbations imposed. We present the results of theoretical studies of formation and evolution of the current sheet in a colliosionless plasma during magnetic reconnection in relativistic limit. Relativistic magnetic reconnection is driven by parallel laser pulses interacting with underdense plasma target. Annihilation of laser created magnetic field of opposite polarity generates strong non-stationary electric field formed in between the region with opposite polarity magnetic field accelerating charged particles within the current sheet. This laser-plasma target configuration is discussed in regard with the laboratory modeling of charged particle acceleration and gamma flash generation in astrophysics. We present the results of 3-dimensional kinetic simulations and theoretical studies on the formation and evolution of the current sheet in a collisionless plasma during magnetic field annihilation in the ultra-relativistic limit. Annihilation of oppositively directed magnetic fields driven by two laser pulses interacting with underdense plasma target is accompanied by an electromagnetic burst generation. The induced strong non-stationary longitudinal electric field accelerates charged particles within the current sheet. Properties of the laser-plasma target configuration are discussed in the context of the laboratory modeling for charged particle acceleration and gamma flash generation in astrophysics.

The initiation of spontaneous breast cancer (SBC) in Tientsin Albino 2 (TA2) mice is related to mouse mammary tumor virus (MMTV) infection, and MMTV amplification is hormonally regulated. To explore the insertion site of MMTVLTR in TA2 mouse genome, reverse PCR and nested PCR were used to amplify the unknown sequence on both sides of the MMTV-LTRSAG gene in SBC and normal breast tissue of TA2 mice. Furthermore, the clinicopathological significance of the insertion site was evaluated in 43 samples of normal breast tissue, 46 samples of breast cystic hyperplasia, 54 samples of ductal carcinoma in situ, 142 samples of primary breast cancer and 47 samples of lymph node metastatic breast cancer by RNA in situ hybridization. We confirmed that the insertion site of the MMTV-LTRSAG gene was located between Igκv2-112 and Igκv14-111 in chromosome 6 of TA2 mouse. IGκC was localized in the stromal cells of TA2 mouse with SBC and in human breast cancer tissues. Tumor cells were negative for IGκC in RNA in situ hybridization. The positive staining index of IGκC in stromal cells was the highest in lymph node metastatic breast cancer, followed by primary breast cancer, ductal carcinoma in situ, and breast cystic hyperplasia. Furthermore, the positive staining index of IGκC was related to the expression of ER, PR, HER2 and Ki-67. Our findings showed that stromal IGκC expression was associated with the initiation of SBC in TA2 mice. IGκC may be a high-risk factor for the initiation and progression of human breast cancer.

Background Radiotherapy and cisplatin-based chemotherapy are currently considered as standard treatments employed for advanced cervical cancer (CC). However, patients with local recurrence or distant metastasis continue to have poor outcomes. EGFR overexpression correlated with chemo/radioresistance, and disease failure has been well proved in the previous studies. Hence, the aim of this study was to explore the therapeutic efficacy and underlying mechanism of the sensitization to radiation or cisplatin of icotinib hydrochloride (IH), a high-selective EGFR tyrosine kinase inhibitor (TKI), in the Hela S3 human CC cell line. Methods Cell proliferation was measured with cell counting kit-8 (CCK-8) assay. Flow cytometry analysis was performed to examine cell cycle distribution and apoptosis. The phosphorylation of EGFR and its downstream signaling molecules were measured by Western blot analysis. γ-H2AX foci and RAD51 foci in the cellular nucleus were visualized using immunofluoresence staining. Expression levels of RAD51 in the whole cells and subceullar fractions were detected to demonstrate the impact of IH on DNA repair. Results IH can significantly inhibit cell proliferation, redistribute cell cycle, enhance apoptosis and impair DNA damage response of Hela S3 cells following radiation or cisplatin treatment through suppressing the activation of the EGFR signaling pathway and attenuating the expression and function of homologous recombination (HR) protein RAD51. Conclusion This study suggests that IH is a potential sensitizer in radiotherapy and cisplatin-based chemotherapy for CC and RAD51 may serve as a prognosis biomarker for this combination treatment.

LncRNA in non-homologous end joining (NHEJ) pathway 1 (LINP1) is an lncRNA which promotes therapeutic resistance in triple-negative breast cancer (TNBC). However, the expression and function of LINP1 in cervical cancer is not yet well-understood. In this study, we evaluated the expression levels of LINP1 in tumor tissues and cell lines of cervical cancer. We found that LINP1 associates with NHEJ proteins (Ku80 and DNA-PKcs). LINP1 translocates from cytosol to nucleus in response to irradiation. In addition, LINP1 knockdown significantly increases the levels of cleaved caspase3 and PARP, leading to enhanced cell apoptosis after ionizing radiation (IR). LINP1-knockdown cells showed delayed repairs of DNA double-strand breaks (DSBs) after IR. Finally, LINP1 knockdown increases radiosensitivity of Hela S3 cells. These results suggest that LINP1 facilitates DSBs repair through NHEJ pathway and may thus serve as a prognostic marker and a potential target for the therapy of cervical cancer.

ELI-Beamlines (ELI-BL), one of the three pillars of the Extreme Light Infrastructure endeavour, will be in a unique position to perform research in high-energy-density-physics (HEDP), plasma physics and ultra-high intensity (UHI) (>1022W/cm2) laser–plasma interaction. Recently the need for HED laboratory physics was identified and the P3 (plasma physics platform) installation under construction in ELI-BL will be an answer. The ELI-BL 10 PW laser makes possible fundamental research topics from high-field physics to new extreme states of matter such as radiation-dominated ones, high-pressure quantum ones, warm dense matter (WDM) and ultra-relativistic plasmas. HEDP is of fundamental importance for research in the field of laboratory astrophysics and inertial confinement fusion (ICF). Reaching such extreme states of matter now and in the future will depend on the use of plasma optics for amplifying and focusing laser pulses. This article will present the relevant technological infrastructure being built in ELI-BL for HEDP and UHI, and gives a brief overview of some research under way in the field of UHI, laboratory astrophysics, ICF, WDM, and plasma optics. A novel regime of high frequency radiation generation via reflection at the electron density spikes in under- dense plasma is proposed. Intense driver laser pulse propagating in underdense plasma forms dense electron singularities near the front part of the bow waves, moving at relativistic velocity. By irradiating a source pulse counterpropagating to the electron density singularities, it is reflected and compressed, producing ultrashort coherent high order harmonics with frequency upshift.

Laser–plasma interaction (LPI) at intensities $10^{15}{-}10^{16}~\text{W}\cdot \text{cm}^{-2}$ is dominated by parametric instabilities which can be responsible for a significant amount of non-collisional absorption and generate large fluxes of high-energy nonthermal electrons. Such a regime is of paramount importance for inertial confinement fusion (ICF) and in particular for the shock ignition scheme. In this paper we report on an experiment carried out at the Prague Asterix Laser System (PALS) facility to investigate the extent and time history of stimulated Raman scattering (SRS) and two-plasmon decay (TPD) instabilities, driven by the interaction of an infrared laser pulse at an intensity ${\sim}1.2\times 10^{16}~\text{W}\cdot \text{cm}^{-2}$ with a ${\sim}100~\unicode[STIX]{x03BC}\text{m}$ scalelength plasma produced from irradiation of a flat plastic target. The laser pulse duration (300 ps) and the high value of plasma temperature ( ${\sim}4~\text{keV}$ ) expected from hydrodynamic simulations make these results interesting for a deeper understanding of LPI in shock ignition conditions. Experimental results show that absolute TPD/SRS, driven at a quarter of the critical density, and convective SRS, driven at lower plasma densities, are well separated in time, with absolute instabilities driven at early times of interaction and convective backward SRS emerging at the laser peak and persisting all over the tail of the pulse. Side-scattering SRS, driven at low plasma densities, is also clearly observed. Experimental results are compared to fully kinetic large-scale, two-dimensional simulations. Particle-in-cell results, beyond reproducing the framework delineated by the experimental measurements, reveal the importance of filamentation instability in ruling the onset of SRS and stimulated Brillouin scattering instabilities and confirm the crucial role of collisionless absorption in the LPI energy balance.

The sub-relativistic laser beam interaction with an underdense plasma is investigated via two-dimensional (2D) numerical simulations with respect to the laser polarization direction. Different parametric instabilities dominate the interaction depending on the propagation direction of the daughter waves in the simulation plane with respect to the laser field polarization and laser propagation direction. In the plane containing the laser electric field (p-polarization) the interaction is dominated by the two-plasmon decay instability and the beating of large amplitude electron plasma waves produces periodic ion density perturbations suppressing stimulated Raman scattering in the quarter critical density zone. A stronger absorption and heating of hot electrons is observed in the case where laser polarization is perpendicular to the simulation plane (s-polarization). Furthermore, by comparing a plane laser wave with a narrow beamlet, the effect of the initial transverse laser profile is proven to play an important role in exciting the filamentation instability, which competes with stimulated Brillouin scattering and affects the laser absorption and hot electron generation. A dedicated three-dimensional simulation indicates that a 2D simulation with p-polarization produces a more reliable results while the case of s-polarization overestimates the laser absorption and hot electron generation.

Visual object tracking is a fundamental problem, which tracks the target in the whole sequence with the given object in the first frame. Numerous tracking frameworks have been proposed. Actor-Critic Tracking (ACT) is based on RL, which is a popular framework with real-time speed. However, ACT cannot handle scale estimation effectively, since it cannot change the ratio of target size. In this paper, we aim to improve the scale estimation of ACT and propose the Action-improved ACT(AIACT) model. This work is based on the basic framework of ACT. ACT builds the ‘Actor’ deep neural network to track the target and the ‘Critic’ network as an inspection module to improve the model. We add a new element to the bounding box of ‘Actor’ model to change the aspect ratio. The experiment result shows that our model achieves better accuracy than the previous model. Our method achieves 0.646 and 0.912 score of success rate and precision rate on OTB dataset, which means AIACT model can handle the scale variation of the target. Therefore, it will benefit more applications.
