Abstract To provide a basis for further optimization of the polio sequential immunization schedule, this study evaluated the effectiveness of booster immunization with one dose of bivalent oral poliovirus vaccine (bOPV) at 48 months of age after different primary polio immunization schedules. At 48 months of age, one dose of bOPV was administered, and their poliovirus types 1–3 (PV1, PV2, and PV3, respectively)-specific neutralizing antibody levels were determined. Participants found to be negative for any type of PV-specific neutralizing antibody at 24, 36, or 48 months of age were re-vaccinated with inactivated polio vaccine (IPV). The 439 subjects who received a bOPV booster immunization at the age of 48 months had lower PV2-specific antibody levels compared with those who received IPV. One dose of IPV during basic polio immunization induced the lowest PV2-specific antibody levels. On the basis of our findings, to ensure that no less than 70% of the vaccinated have protection efficiency, we recommend the following: if basic immunization was conducted with 1IPV + 2bOPV (especially Sabin strain-based IPV), a booster immunization with IPV is recommended at 36 months of age, whereas if basic immunization was conducted with 2IPV + 1bOPV, a booster immunization with IPV is recommended at 48 months of age. A sequential immunization schedule of 2IPV + 1bOPV + 1IPV can not only maintain high levels of antibody against PV1 and PV3 but also increases immunity to PV2 and induces early intestinal mucosal immunity, with relatively good safety. Thus, this may be the best sequential immunization schedule for polio in countries or regions at high risk for polio.

Kaempferol (KPR), a flavonoid compound found in various plants and foods, has garnered attention for its anti‐inflammatory, antioxidant, and anticancer properties. In preliminary studies, KPR can modulate several signaling pathways involved in inflammation, making it a candidate for treating cholecystitis. This study aimed to explore the effects and mechanisms of KPR on lipopolysaccharide (LPS)‐induced human gallbladder epithelial cells (HGBECs). To assess the impact of KPR on HGBECs, the HGBECs were divided into control, KPR, LPS, LPS + KPR, and LPS + UDCA groups. Cell viability and cytotoxicity were evaluated by MTT assay and lactate dehydrogenase (LDH) assay, respectively, and concentrations of KPR (10–200 μM) were tested. LPS‐induced inflammatory responses in HGBECs were to create an in vitro model of cholecystitis. The key inflammatory markers (IL‐1β, IL‐6, and TNF‐α) levels were quantified using ELISA, The modulation of the MAPK/NF‐κB signaling pathway was measured by western blot using specific antibodies against pathway components (p‐IκBα, IκBα, p‐p65, p65, p‐JNK, JNK, p‐ERK, ERK, p‐p38, and p38). The cell viability and LDH levels in HGBECs were not significantly affected by 50 μM KPR, thus it was selected as the optimal KPR intervention concentration. KPR increased the viability of LPS‐induced HGBECs. Additionally, KPR inhibited the inflammatory factors level (IL‐1β, IL‐6, and TNF‐α) and protein expression (iNOS and COX‐2) in LPS‐induced HGBECs. Furthermore, KPR reversed LPS‐induced elevation of p‐IκBα/IκBα, p‐p65/p65, p‐JNK/JNK, p‐ERK/ERK, and p‐p38/p38 ratios. KPR attenuates the LPS‐induced inflammatory response in HGBECs, possibly by inhibiting MAPK/NF‐κB signaling. [ABSTRACT FROM AUTHOR]

Pollination dynamics was studied in a first-generation Platycladus orientalis (L.) Franco seed orchard with pedigree reconstruction using eight nuclear and four chloroplast SSRs. The pedigree reconstruction assigned 371 of 448 studied seeds to one of the orchard's 192 candidate male parents and showed a high level of outcrossing and pollen contamination in the orchard's seed crop. While the orchard's seed population showed greater allelic richness compared with the parental population, a few alleles present in the parental population were missing in the seed crop. Additionally, we detected no significant correlation between male reproductive energy (pollen yield) and male reproductive success; however, uneven parental contribution was also observed. Pollen management practices were recommended to ensure the maintenance of genetic diversity in the seed crops and increase in genetic gain. La dynamique de la pollinisation a ete etudiee dans un verger a graines de premiere generation de Platycladus orientalis (L.) Franco a l'aide de la reconstruction de pedigree se basant sur huit marqueurs SSRs de type nucleaire et quatre de type chloroplastique. La reconstruction de pedigree a permis d'assigner 371 des 448 graines etudiees a l'un des 192 parents males candidats du verger, tout en indiquant un niveau eleve d'exogamie et de contamination pollinique parmi la recolte de graines du verger. Alors que la population de graines du verger avait une richesse allelique plus elevee comparativement a la population parentale, certains des alleles presents dans la population parentale etaient absents des graines recoltees. De plus, une correlation significative a ete detectee entre l'energie des males consacree a la reproduction (production de pollen) et leur succes reproducteur; toutefois, des contributions parentales inegales ont aussi ete observees. Des pratiques de gestion du pollen sont recommandees afin d'assurer le maintien de la diversite genetique des recoltes de graines et d'augmenter le gain genetique. Aims/Introduction: To investigate the association between serum 25‐hydroxyvitamin D (25‐[OH]D) concentrations and metabolic syndrome (MetS) in the middle‐aged and elderly Chinese population. Methods: The present study included 2,764 participants (aged >50 years). The joint interim statement was used for the standard definition of MetS. Serum 25‐(OH)D concentrations were measured by electrochemiluminescence immunoassay. The study participants were categorized into quartiles based on serum 25‐(OH)D concentrations, and the quartiles were calculated for the differences using anova and the χ2‐test for continuous and categorical data, respectively. A logistic regression analysis model was applied to estimate the odds ratios and 95% confidence intervals (95% CI) for each quartile of serum 25‐(OH)D concentrations compared with the highest quartile. Results: Serum 25‐(OH)D levels were markedly lower in men in the MetS group than in those without MetS. We observed a negative correlation between the higher quartiles of serum 25‐(OH)D levels and the presence of MetS among men. The correlation between serum 25‐(OH)D levels and the prevalence of MetS persisted even after adjusting for potential confounders, including age, cigarette smoking status, alcohol consumption, physical activity, low‐density lipoprotein, creatinine and total serum cholesterol. Adjusted odds ratios of MetS in the second through fourth compared with the lowest quartile for serum 25‐(OH)D levels were 0.93 (95% CI 0.54–1.59), 0.89 (95% CI 0.50–1.56) and 0.48 (95% CI 0.28–0.84), respectively. Conclusions: Decreased serum 25‐(OH)D level is significantly correlated with MetS in middle‐aged men. [ABSTRACT FROM AUTHOR]
